This paper details the cross-section measurements of 38 fission-like events in 19F + 181Ta system at energy 115 MeV, originating from complete and/or incomplete fusion-fission processes. The recoil-catcher off-beam activation technique followed by γ-ray spectroscopy has been used to measure production cross sections of fission-like events. The measured cross-section data of fission-like events align well with Gaussian distributions indicating their production via de-excitation of equilibrated compound nucleus formed. Additionally, the study explores mass and isotopic yield distributions of fission fragments, with the presently measured isotopic yield variance which is found to be consistent with literature values for other fissioning systems. The objective of this study was to detect the changes in physicochemical and functional properties of raw and processed divergent honey varieties after adulterating with different sugar syrups. Specifically, when 25% to 55% cane syrup was added to raw Acacia honey, the HMF content increased from 46.25 to 101.6 mg/kg. In processed honey that contained 55% cane syrup, HMF content was even higher, reaching up to 402.47 mg/kg. The reported values for HMF (>80 mg/kg) content and DN (>8 DN) were above the described quality evaluation standard of honey. The results indicated that adulterating Acacia honey with up to 55% corn and corn syrup caused a reduction in diastase content to below 3.5 DN, and an increase in HMF content above 93.28 mg/kg. Similarly, adding 10% corn syrup to raw Ziziphus honey resulted in a decrease in both DPPH and ABTS+ values from 83.16% and 88.58%, respectively, to 72.83% and 76.97%. The findings in this study demonstrated that the addition of malt syrup (55%) to raw Trifolium honey resulted in an increase in the phytochemical content, with the TPC value reaching 805.15 mg GAE/100 g. However, processing Trifolium honey caused a significant decrease in TPC content to 505.15 mg GAE/100 g. This work highlights the need for more research into unexplored honey varieties and adulteration methods to improve quality evaluation standards and mitigate authenticity issues. In earlier studies from this laboratory, Xanthomonas campestris pv. glycines was found to exhibit a nutrition stress-related postexponential rapid cell death (RCD). The RCD was exhibited in protein-rich media but not in starch or other minimal media. This RCD in X. campestris pv. glycines was found to display features similar to those of the programmed cell death (PCD) of eukaryotes. Results of the present study showed that the observed RCD in this organism is both positively and negatively regulated by small molecules. The amino acids glycine and L-alanine as well as the D isomers of valine, methionine, and threonine were found to induce the synthesis of an active caspase-3-like protein that was associated with the onset of RCD. Addition of pyruvate and citrate to the culture medium induced both the synthesis of active caspase-3-like protein and RCD. Higher levels of intracellular accumulation of pyruvate and citrate were also observed under conditions favoring RCD. On the other hand, dextrin and maltose, the hydrolytic products of starch, inhibited the synthesis of the caspase-3-like protein. Addition of glucose and cyclic AMP (cAMP) to the RCD-favoring medium prevented RCD. Glucose, cAMP, caffeine (a known inhibitor of a phosphodiesterase that breaks down cAMP), and forskolin (from the herb Coleusforskholii, known to activate the enzyme adenylate cyclase that forms cAMP) inhibited the caspase enzyme activity in vivo and consequently the RCD process. The addition of glucose and other inhihitors of RCD enhanced intracellular cAMP accumulation. This is the first report demonstrating the involvement of small molecules in the regulation of nutrition stress-related stationary-phase rapid cell death in X. campestris pv. glycines, which is programmed.

Raw leafy greens are commonly associated with global foodborne outbreaks due to pathogenic contaminants. In the current study, greens, such as spinach (Spinacia oleracea L.), coriander (Coriandrum sativum L.) and mint (Mentha spicata L.) showed presence of coliforms (including E. coli) along with other aerobic microbes, yeast and molds. These vegetables mostly consumed in raw or culinary purpose, and hence increase the chances of food borne illnesses. Moreover, the leafy greens are perishable. In this context, we optimized a combination process including radiation treatment to achieve hygienization and shelf life extension of these leafy green vegetables. The combination treatment comprising potable water wash, chlorination (NaOCl-200 ppm) followed by irradiation (2 kGy using electron beam or gamma) was developed, and the processed samples showed keeping quality up to 15 days at 4-6°C, whereas control samples spoiled within two days. The treatment resulted in coliform count below detection level while retaining the nutritional, phenolic content and organoleptic qualities. Thus, the combination treatment could ensure safety, keeping quality enhancement of perishable leafy greens and to control global food outbreaks. Grape extracts of different cultivars (Flame seedless, Kishmish chorni, Red globe and Thompson seedless) were investigated for in vitro antioxidant activity by ABTS assay, and compared protective efficacy against radiation-induced DNA damage. Seed extract showed the highest scavenging activity, followed by skin extract. Among different cultivars, 'flame seedless' skin extract showed higher scavenging activity followed by 'Kishmish chorni' skin extract. Grape extracts significantly prevented radiation-induced plasmid DNA damage. Super-coiled pBR 322 plasmid DNA (~93%) is completely converted to open circular (~97%) and linear (~2%) form at a dose of 150 Gy γ-radiation. Pretreatment with different grape extracts showed various degree of protection against radiation-induced DNA damage. Pretreatment with 1.6 µg grape skin extract of 'Thompson seedless' cultivar or grape flesh extract of any tested cultivar diminished the DNA strand breaks, and there was an increase in the super coiled form of DNA against 150 Gy of γ-radiation. However, pretreated pBR 322 DNA with the skin of 'Kishmish chorni' cultivars or seed of 'red globe' grape cultivars remained static during electrophoresis and confined in the groove on exposure to 150 Gy γ-radiation treatment. Co-treatment with the skin of red globe cultivar also partially confined plasmid DNA in the groove. The same trend was observed when plasmid DNA was exposed to 1.2 kGy γ- radiation. Our investigation revealed that anthocyanin present in grape skin was probably involved in radio protective activities through the formation of co-pigmentation with DNA. [ABSTRACT FROM AUTHOR]

Abstract: BACKGROUND: Profound apprehension towards safety of irradiated food has remained a major cause behind tardy acceptance of this technology although it has immense socio‐economic potential. Generation of in‐depth scientific evidence will help to refute these apprehensions. With this prospective, the present study was undertaken where safety of various irradiated (Dmin up to 25 kGy) foods was evaluated through long‐term exposure studies in models including human lymphoblast TK6 cell line (100 generations) and Escherichia coli MG1655 cells (exclusive sub‐culturing in irradiated food medium for 3000 generations). Additionally, the Ames test, micronucleus test, comet assay, DNA sequencing and restriction profiling of phagemid DNA from E. coli cells sub‐cultured in irradiated food medium were also performed. RESULTS: No induced mutagenesis was observed in these cells during long‐term sub‐culturing in various irradiated food medium. Also no change was observed in profiles of comet, micronucleus, restriction digestion, random amplification of polymorphic DNA as well as DNA sequences. The latter also ruled out the possibility of any silent mutation. CONCLUSION: Findings of the current study thus provided credible molecular evidence supporting the safety of irradiated foods. Summary: Fenugreek microgreen and mint leaf displayed antidiabetic potential during in vitro assays including cell line‐based analysis. Aqueous fenugreek microgreen extract (FME) (2 mg mL−1) inhibited α‐amylase by 70%. It also enhanced glucose uptake in L6 cells by 25% at 10 mg mL−1 which further improved to 44% in the presence of insulin. On the other hand, fresh mint leaf extract (MLE) inhibited α‐glucosidase up to 90% and increased glucose uptake by 15% in HepG2 cells. Besides, both these extracts also inhibited nonenzymatic glycation of protein. FME and MLE were also found to have high levels of total phenolics, flavonoids and antioxidants which could play a possible role in the observed antidiabetic activity. As blood glucose levels in diabetes can be regulated by inhibition of enzymes regulating carbohydrate metabolism and improving glucose uptake in cells, the findings indicate a complimentary therapeutic role of these two dietary herbs in diabetes. Purpose:Current study was aimed to enhance the confidence of consumers as well as entrepreneurs towards food irradiation program. Materials and methods:In this work, safety of high dose (25 kGy) irradiated meat samples (HDIMS) was ascertained by scoring mutation frequency through a long-term sub-culturing study inEscherichia coliMG1655 cells (ATCC 700926) up to 1500 generations (at 1%), 250 generations (at 5% and 10%) and human lymphoblast thymidine kinase heterozygote (TK6) cell line (ATCC CRL-8015) [at two gene loci,tk−/+(thymidine kinase) andhprt+(Hypoxanthine Phosphoribosyltransferase)] up to 156 generations using goat meat sample. Also these samples were assayed at further radiation doses of 10, 45 and 70 kGy at 2% concentration (in cell line), and 1% (inE. coli). Study was also performed with other meat samples such as chicken, fishes (pomfret and rohu) and shrimps by carrying out limited long-term sub-culturing trials in human lymphoblast cell line. Mutation analysis was also carried out using a novel DPAR (Differential loss of Plasmid Antibiotic Resistance) assay followed by sequencing oftcR(tetracycline resistance) gene of pBR322 plasmid isolated fromE. colicells grown for 1500 generations on HDIMS medium and RAPD (Random Amplified Polymorphic DNA) analysis of the genome. Results and conclusion:None of the assays exhibited any induced mutation when analyzed at regular time intervals. RAPD analysis also did not indicate any change in its nucleotide sequence, ruling out the occurrence of any silent mutation.
