18. Sleep Matters in Chronotype and Mental Health Association: Evidence from the UK and Germany.

Abstract

Background: There is considerable evidence supporting the elevated risk of mental health problems in individuals with evening chronotype relative to those with morning or intermediate chronotypes. Recent data, however, suggest that this risk may be explained, at least partially, by poor sleep quality. Methods: This study aimed to further clarify the roles of chronotype and sleep quality in mental health outcomes (depression, anxiety, stress) in young individuals (18–40 years) living in the UK (n = 185) or Germany (n = 209). Results: Consistent with our recent observations in a comparable North Indian sample, we found that poor quality of sleep had significantly positive associations with adverse mental health outcomes both in the UK and Germany-based samples. Significant associations between evening chronotype and poor mental health were also evident, but these associations were fully mediated by poor quality of sleep in both samples. Conclusions: These observations suggest that efforts to identify sleep disruption in a timely manner and promotion of good sleep may prevent mental health problems, especially in individuals with evening chronotype and other known risks for mental disorders. 44. Smooth pursuit and antisaccade eye movements as endophenotypes in schizophrenia spectrum research

Abstract

Smooth pursuit eye movement (SPEM) and antisaccade deficits have been proposed as schizophrenia spectrum endophenotypes. An endophenotype is a behavioural or biological deficit thought to represent, more closely than the disease phenotype, the effects of an underlying disease gene. Oculomotor endophenotypes possess phenotypic homogeneity, well-understood neural correlates and objective assessment and may thus be used as phenotypes in linkage studies. This thesis investigated a number of issues concerning the reliability and validity of the SPEM and antisaccade tasks as schizophrenia spectrum endophenotypes (and two tasks thought to be unimpaired in the schizophrenia spectrum, visual fixation and prosaccades). The schizophrenia spectrum encompasses not only people with schizophrenia but any population with an increased frequency of schizophrenia-related phenotypes or genotypes, such as schizotypal individuals or first-degree relatives of schizophrenia patients. A valid endophenotype should thus be detected in these populations. Study I investigated reliability, namely internal consistency and temporal stability, of eye movements in healthy individuals. Study II utilised first-episode psychosis patients and healthy controls, aiming to detect behavioural oculomotor deficits in the absence of secondary confounds that may be encountered in chronic schizophrenia. Study III assessed performance in siblings discordant for schizophrenia. Study IV explored the relationship between psychometric schizotypy and oculomotor performance. Study V examined possible state effects of procyclidine, an anticholinergic compound often administered to schizophrenia patients, on performance in a patient group. The results generally confirmed the validity of the SPEM and antisaccade deficits as schizophrenia spectrum endophenotypes: Oculomotor performance was mostly stable both within and between assessments. SPEM and antisaccade impairments were observed in first­episode psychosis patients and schizophrenia patients and their healthy siblings. Antisaccade, but not SPEM, impairments were associated with high levels of schizotypy. State effects of procyclidine on SPEM and antisaccade performance were observed, suggesting the need to consider the influence of pharmacological treatment in future patient studies. These findings suggest that SPEM and antisaccade deficits may be studied profitably as endophenotypes in schizophrenia spectrum research.

Abstract

Temporal discounting, the tendency to devalue future rewards as a function of delay until receipt, is influenced by time framing. Specifically, discount rates are shallower when the time at which the reward is received is presented as a date (date condition; e.g., June 8, 2023) rather than in delay units (delay condition; e.g., 30 days), which is commonly referred to as the date/delay effect. However, the cognitive and neural mechanisms of this effect are not well understood. Here, we examined the date/delay effect by analysing combined fMRI and eye‐tracking data of N = 31 participants completing a temporal discounting task in both a delay and a date condition. The results confirmed the date/delay effect and revealed that the date condition led to higher fixation durations on time attributes and to higher activity in precuneus/PCC and angular gyrus, that is, areas previously associated with episodic thinking. Additionally, participants made more comparative eye movements in the date compared to the delay condition. A lower date/delay effect was associated with higher prefrontal activity in the date > delay contrast, suggesting that higher control or arithmetic operations may reduce the date/delay effect. Our findings are in line with hypotheses positing that the date condition is associated with differential time estimation and the use of more comparative as opposed to integrative choice strategies. Specifically, higher activity in memory‐related brain areas suggests that the date condition leads to higher perceived proximity of delayed rewards, while higher frontal activity (middle/superior frontal gyrus, posterior medial frontal cortex, cingulate) in participants with a lower date/delay effect suggests that the effect is particularly pronounced in participants avoiding complex arithmetic operations in the date condition.
