Tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE) is the major TBBPA derivative. It has been detected in different environmental samples. Previous studies show that TBBPA-DHEE caused neurotoxicity in rats. In this study, juvenile zebrafish were exposed to various concentrations of TBBPA-DHEE to ascertain the potential neurotoxicity of TBBPA-DHEE, the chemical, and its possible molecular mechanism of action. Behavioral analysis revealed that TBBPA-DHEE could significantly increase the swimming distance and speed in the 1.5 mg/L group compared to the control. In contrast, the swimming distance and speed were significantly reduced in the 0.05 and 0.3 mg/L groups, affecting learning, memory, and neurodevelopment. Similarly, TBBPA-DHEE exposure caused a concentration-dependent significant increase in the levels of excitatory neurotransmitters, namely, dopamine, norepinephrine, and epinephrine, which could be attributed to the change observed in zebrafish behavior. This demonstrates the neurotoxicity of TBBPA-DHEE on juvenile zebrafish. The concentration-dependent increase in the IBR value revealed by the IBR index reveals the noticeable neurotoxic effect of TBBPA-DHEE. Transcriptomic analysis shows that TBBPA-DHEE exposure activated the PPAR signaling pathways, resulting in a disturbance of fatty acid (FA) metabolism and changes in the transcript levels of genes involved in these pathways, which could lead to lipotoxicity and hepatotoxicity. Our findings demonstrate a distinct endocrine-disrupting response to TBBPA-DHEE exposure, possibly contributing to abnormal behavioral alterations. Background: Alternaria can infest pears to produce metabolites, which can contaminate pears and their processed products. Pear paste, one of the most important pear-based products, is popular among Chinese consumers especially for its cough relieving and phlegm removal properties. Although people are concerned about the risk of Alternaria toxins in many agro-foods and their products, little is known about the toxins in pear paste., Results: A method was developed for the determination of tenuazonic acid, alternariol, alternariol menomethyl ether, altenuene and tentoxin in pear paste by ultra-performance liquid chromatography tandem mass spectrometry with saturated sodium sulphate dissolution and acidified acetonitrile extraction. The mean recoveries of the five toxins were 75.3-113.8% with relative standard deviations of 2.8-12.2% at spiked levels of 1.0-100 μg kg -1 . Alternaria toxins were detected in 53 out of 76 samples, with a detection rate of 71.4%. Tenuazonic acid (67.1%), alternariol (35.5%), tentoxin (23.7%) and alternariol monomethyl ether (7.9%) were detected in all samples at concentrations of < limit of quantification (LOQ)-105.0 μg kg -1 , < LOQ-32.1 μg kg -1 , < LOQ-74.2 μg kg -1 and < LOQ-15.1 μg kg -1 , respectively. Altenuene was never found in pear paste samples. Tenuazonic acid, alternariol, tentoxin and alternariol menomethyl ether should be focused on due to their toxicity and detection rates., Conclusion: To the best of our knowledge, this is the first report on the detection method and residue levels of Alternaria toxins in pear paste. The proposed method and research data can provide technical support for the Chinese government to continuously monitor and control Alternaria toxins in pear paste, especially tenuazonic acid. Objectives. Researchers have shown that cleaning workers have an increased risk of asthma and rhinitis, mainly due to exposure to chemical substances present in the cleaning products they use. Among the important substances are glycol ethers, increasingly used as components in cleaning products. This study aimed to assess exposure levels of glycol ether in professional cleaning products and compare them to existing regulatory exposure limit values. Methods. Information from safety data sheets of the products is used to identify the glycol ethers present in the cleaning products and their respective concentrations. Other sources were used to obtain the relevant data required for use in the tool to generate exposure assessments. Exposure levels for various cleaning work exposure scenarios were estimated using the ConsExpo Web tool. Results. The estimated exposure values are significantly lower than the existing regulatory occupational exposure limit (OEL) values for the different glycol ethers. Conclusions . The study showed that the risk of exposure to glycol ethers by inhalation from professional cleaning products is minimal as exposure estimates were much below the regulatory OEL values.

TBBPA bis(2-hydroxyethyl) ether (TBBPA-DHEE), one of the main derivatives of TBBPA, has been widely detected in environmental samples and been discovered to be potential neurotoxic. In this study, the juvenile zebrafish were selected as the research subject to explore the neurotoxicity and its mechanism of low-dose TBBPA-DHEE exposure, and to reveal the neurotoxicity susceptibility in different sexes. Behavioral studies revealed that TBBPA-DHEE could significantly reduce the swimming velocity, maximum acceleration and cumulative duration of high-speed mobility, significantly increasing the cumulative duration of low-speed mobility and average social distance. It significantly reduced the contents of ATP, glutamate and Ca 2+ in the whole brain. The histopathological study demonstrated that TBBPA-DHEE could cause brain tissue damage in female and male juvenile zebrafish. The comprehensive data analysis indicated that female zebrafish were more susceptible to TBBPA-DHEE exposure than male zebrafish. Transcriptomic analysis showed that TBBPA-DHEE could significantly affect the expressions of behavioral and development-related genes. Furthermore, female and male juvenile zebrafish have different molecular mechanisms of neurotoxicity. For female juvenile zebrafish, the potential mechanism of neurotoxicity could be that it interfered with the feedback regulation of nerves by affecting the related genes expressions in the signaling pathways such as Ca 2+ signaling, Wnt signaling and synapses. For male juvenile zebrafish, the potential mechanism of neurotoxicity may be through affecting the expression of related genes in hormones and neuro-related genes. Endocrine-disrupting chemicals (EDCs) are now ubiquitously distributed in the environment. Tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE) pollution in environment media poses a significant threat to humans and aquatic organisms as a result of its potential neurotoxicity and endocrine-disrupting effect. The endocrine-disrupting effects of TBBPA-DHEE on aquatic organisms, however, have received limited attention. In this study, the neurotoxicity and reproductive endocrine-disruptive effect of TBBPA-DHEE was evaluated by observing the neurobehavioral changes, vitellogenin (VTG), testosterone, 17β-estradiol and gene expression levels in adult male and female zebrafish exposed to TBBPA-DHEE (0.05, 0.2 and 0.3 mg/L) for 100 days. Furthermore, transcriptomic analysis was conducted to unravel other potential neuroendocrine-disrupting mechanism. Our result showed TBBPA-DHEE significantly (p < 0.05) altered the locomotor behavior and motor coordination abilities in both sexes. Steroid hormone and VTG levels were also altered indicating the neuroendocrine-disrupting effect of TBBPA-DHEE on the hypothalamic-pituitary-gonadal-axis. A total of 1568 genes were upregulated and 542 genes downregulated in males, whereas, 1265 upregulated and 535 downregulated genes were observed in females. The KEGG enrichment analysis showed that cell cycle and p55 signaling pathways were significantly enriched due to TBBPA-DHEE exposure. These pathways and its component genes are potential target of EDCs. The significant upregulation of genes in these pathways could partly explain the neuroendocrine disrupting effect of TBBPA-DHEE. Limited information is available about prenatal exposure to per- and polyfluoroalkyl substances (PFAS) in electronic waste (e-waste) recycling sites. In this study, we determined 21 emerging PFAS and 13 legacy PFAS in 94 paired maternal and cord serum samples collected from an e-waste dismantling site in Southern China. We found 6:2 fluorotelomer sulfonate (6:2 FTSA), 6:2 chlorinated polyfluorinated ether sulfonate (6:2 Cl-PFESA), and perfluorooctanephosphonate (PFOPA) as the major emerging PFAS, regardless of matrices, at median concentrations of 2.40, 1.78, and 0.69 ng/mL, respectively, in maternal serum samples, and 2.30, 0.73, and 0.72 ng/mL, respectively, in cord serum samples. Our results provide evidence that e-waste dismantling activities contribute to human exposure to 6:2 FTSA, 6:2 Cl-PFESA, and PFOPA. The trans-placental transfer efficiencies of emerging PFAS (0.42-0.94) were higher than that of perfluorooctanesulfonic acid (0.37) and were structure-dependent. The substitution of fluorine with chlorine or hydrogen and/or hydrophilic functional groups may alter trans-placental transfer efficiencies. Multiple linear regression analysis indicated significant associations between maternal serum concentrations of emerging PFAS and maternal clinical parameters, especially liver function and erythrocyte-related biomarkers. This study provides new insights into prenatal exposure to multiple PFAS in e-waste dismantling areas and the prevalence of emerging PFAS in people living near the sites.

Tetrabromobisphenol A bis (2-hydroxyethyl ether) (TBBPA-DHEE) is a derivative of Tetrabromobisphenol A (TBBPA) used as an intermediate flame retardant in engineering polymers. The mechanism of neurodevelopmental toxicity of TBBPA-DHEE remains unclear due to limited toxicological data. We performed behavioral and transcriptomic analyses to assess the neurodevelopmental effects of TBBPA-DHEE on developing zebrafish and potential toxicity mechanisms. Our result shows that exposure to TBBPA-DHEE significantly increased mortality, deformity rate, and reduction in hatch rate, hatchability, and body length relative to the DMSO control. The behavior analysis indicates that TBBPA-DHEE significantly reduced the spontaneous movement of larva compared to the control. The TSH and GH levels were significantly reduced in all the exposure groups in a concentration-dependent manner relative to the DMSO control. TBBPA-DHEE exhibited a significant reduction in locomotor activity across all the exposure groups in the light/dark locomotion test. The transcriptomic analysis result shows that 579 genes were differentially expressed. KEGG analysis shows the enrichment of complement cascade, JAK-STAT signaling pathway, cytokine-cytokine interaction, and phototransduction pathway resulting in a change in mRNA expression of their genes. These observed changes in developmental endpoints, hormonal level, and alteration in mRNA expression of component genes involved in neurodevelopmental pathways could be part of the possible mechanism of the observed toxic effects of TBBPA-DHEE exposure on zebrafish. The sediment distribution of per- and polyfluoroalkyl substances (PFAS) along a river to ocean transect was investigated. Samples were collected between September 2017 and October 2019 with targeted quantification of six legacy and replacement PFAS by LC-MS/MS. Total PFAS concentrations ranged from below the LOQ to 7.47 ng per g dry weight with PFOA, PFOS, HFPO-DA and PFMOAA the most frequently detected. Significant correlations ( p < 0.05) were found between PFOS and HFPO-DA sedimentary concentration and percent organic carbon (% OC); however, PFOA and PFMOAA were not correlated with sediment % OC. This study highlights the occurrence of the replacement PFAS in sediments for the first time. Sediment extracts were screened for 18 additional PFAS compounds by high resolution mass spectrometry. A series of perfluorinated ether carboxylic acid and perfluorinated ether sulfonic acid with either one or two acidic functional groups were detected at various locations in the upper portion of the Cape Fear River. A series of chromatographically resolved isomers (C 7 F 13 O 5 S 1 ; M-1) were detected and may be Nafion™ degradation products.

PEGylated emulsifiers have been largely used in topical formulations for skin research. They have been a continuous study focus in our group as well. According to our previous studies, severe interruptions of the skin barrier were observed with certain types of emulsifiers. To restore the skin barrier function and counteract the effects of emulsifiers, we considered topically delivering lipids into the lipid matrix of the SC. Herein, PEG-20 cetyl ether (C20) -based oil-in-water (O/W) emulsions were developed owing to the stronger interactions of C20 with skin. The lipids containing ceramides (Cers), palmitic acids (PA), and cholesterol with different ratios and combinations were merged into the base emulsion. PEG-40 stearyl ether (S40)-based emulsion was used as a reference as S40 showed negligible impact on SC lipids. The evaluations were conducted ex vivo with confocal Raman spectroscopy (CRS) regarding the SC lipid, SC thickness, and skin penetration properties. In parallel, the in vivo irritation studies were also implemented including the transepidermal-water-loss (TEWL), skin hydration, and erythema index. The results indicated less SC lipid extraction of topically delivered lipids on ex vivo porcine skin with the addition and ratio of incorporated Cers influencing the extent of formulations counteracting the skin interruption by C20. The ex vivo penetration study showed a similar trend in drug penetration depths. In regards to the in vivo studies, TEWL was demonstrated to be suitable for differentiating the impact on skin barrier properties. The in vivo observations were generally correlated with the ex vivo results. The exact findings in this research can lead us to a better selection of applied lipid components and compositions. The prevalence of childhood obesity has increased nearly ten times over the last 40 years, influenced by early life nutrients that have persistent effects on life-long metabolism. During the first six months, infants undergo accelerated adipose accumulation, but little is known regarding infant fatty acid status and its relationship to infant body composition. We tested the hypothesis that a low arachidonic to docosahexaenoic acid ratio (AA/DHA) in infant red blood cells (RBCs), a long-term indicator of fatty acid intake, would associate with more infant fat-free mass (FFM) and/or less adipose accumulation over the first 4 months of life. The fatty acid and composition of breastmilk and infant RBCs, as well as the phospholipid composition of infant RBCs, were quantified using targeted and unbiased lipid mass spectrometry from infants predominantly breastfed or predominantly formula-fed. Regardless of feeding type, FFM accumulation was inversely associated with the infant’s RBC AA/DHA ratio (p = 0.029, R2 = 0.216). Infants in the lowest AA/DHA ratio tertile had significantly greater FFM when controlling for infant sex, adiposity at 2 weeks, and feeding type (p < 0.0001). Infant RBC phospholipid analyses revealed greater peroxisome-derived ether lipids in the low AA/DHA group, primarily within the phosphatidylethanolamines. Our findings support a role for a low AA/DHA ratio in promoting FFM accrual and identify peroxisomal activity as a target of DHA in the growing infant. Both FFM abundance and peroxisomal activity may be important determinants of infant metabolism during development.

Tetrabromobisphenol A bis(2-hydroxyetyl) ether (TBBPA-DHEE) is among the main derivatives of Tetrabromobisphenol A (TBBPA). Result from previous study showed that TBBPA-DHEE can cause neurotoxicity in rat. In this study, zebrafish larvae were used for evaluation of TBBPA-DHEE-induced developmental toxicity, apoptosis, oxidative stress and the potential molecular mechanisms of action. Our result showed that TBBPA-DHEE exposure caused a significant concentration-dependent developmental toxicity endpoints like death rate, malformation rate, growth rate. TBBPA-DHEE altered locomotor and enzymes activities of larvae and caused apoptosis within the brain indicating the potential TBBPA-DHEE-induced cardiac, brain impairment in the zebrafish larvae. Our transcriptomic analysis shows that 691 genes were differentially expressed (DEGs) (539 upregulated, 152 downregulated). The KEGG and GO enrichment pathway analysis shows that the DEGs were involved in development, immunity, enzyme activity. Our study provides novel evidence on the neurodevelopmental toxicity and toxicity mechanism of TBBPA-DHEE which are vital for assessment of the environmental toxicity and risk assessment of the chemical., (Copyright © 2022 Elsevier Inc. Background/objectives: Multiple studies have examined the relationship between PFAS and semen quality, but none has explored the associations of PFAS mixture that includes emerging alternatives and branched isomers., Methods: 22 PFAS, including 10 linear legacy PFAS, 7 branched isomers, 3 short chain alternatives and 2 components of F53B [e.g., 6:2 chlorinated polyfluorinated ether sulfonate (Cl-PFESA)] were quantified in blood plasma among 740 healthy men. Five semen quality parameters (i.e., volume, count, concentration, total motility and progressive rate) were assessed. Multiple linear regression and three multiple pollutant models (i.e., adaptive elastic net regression, quantile based g-computation, and XGBoost method) were used to assess the associations of individual PFAS and PFAS mixture with semen quality and the potential interactive effects among congeners., Results: After adjusting for selected confounders, perfluorobutane sulfonate (PFBS) and perfluorohexane sulfonate (PFHxS) presented significant and negative associations with sperm count [β AENET = -0.09 (95%CI: -0.14, -0.03) for PFBS, and -0.16 (95%CI: -0.25, -0.07) for PFHxS] and sperm concentration [-0.04 (95%CI: -0.08, -0.001) for PFBS and -0.11 (95%CI: -0.17, -0.04) for PFHxS]. 6:2 Cl-PFESA showed negative associations with total motility (-2.33, 95%CI: -3.80, -0.86) and progressive rate (-1.46, 95%CI: -2.79, -0.12). But perfluoroheptanesulfonic acid (PFHpS) was positively associated with sperm count and concentration. These associations were supported by the importance assessment of these four congeners in XGBoost analyses. However, no associations were found between PFAS mixture or branched isomers and semen quality; nor were there significant interactions among PFAS congeners., Conclusions: In the current cross-sectional study, we found that two emerging PFAS replacements (i.e., 6:2 Cl-PFESA and PFBS) and PFHxS exposure were associated with reduced semen concentration, total sperm count and motility in men. Meanwhile, significant positive associations between PFHpS and sperm count and concentration were also observed. Human exposure to per- and polyfluoroalkyl substances (PFASs) has gained worldwide attention due to their widespread presence in the environment and adverse health effects, but the exposure assessment in the elderly is still lacking. This study aimed to assess exposures to 3 emerging PFASs (chlorinated polyfluoroalkyl ether sulfonic acids, Cl-PFESAs) and 15 legacy PFASs. The temporal variability of internal exposures and intake amounts of these PFASs were evaluated among a population of 76 healthy elderly adults (age: 60-69) in Jinan, China over 5 consecutive months. Fifteen PFASs were detected in whole blood with the mean total concentration (ΣPFAS) at 20.1 ng/mL (range: 5.0-135.9 ng/mL) dominated by perfluorooctanoic acid (PFOA) (9.0 ng/mL), perfluorooctanesulfonic acid (PFOS) (5.3 ng/mL), and 6:2 Cl-PFESA (1.6 ng/mL). Across the 5 month assessment period, significant variation was only observed for short-chain (C4-C7) perfluoroalkyl carboxylic acids, and their variations ranged from 53 to 334%. The median intake of PFOA and PFOS was estimated to be 1.46 and 0.92 ng/kg bw/day, respectively. Regression analysis showed that dietary ingestion, especially fish, was likely an important exposure pathway for PFASs among the elderly adults. Various pathways (e.g., dietary, water, air, and dust) should thus be considered to fully understand human exposure to PFASs.

Volatile organic compound (VOC) emissions from pyrolysis of widely used biomass are expected to increase significantly under the carbon neutrality target. However, the dynamic emissions and evolution mechanism of biomass-VOCs remain unclear, hindered by complex reactions and offline measurements. Here, we propose a novel covariant evolution mechanism to interpret the emission heterogeneities, sequential temperature responses, and evolved correlations of both VOCs and residual functional groups (RFGs) during corn straw (CS), wood pellet (WP), and semibituminous coal (SBC) pyrolysis. An innovative combination of online thermogravimetric-Fourier transform infrared-gas chromatography/mass spectrometry and two dimensional-correlation spectroscopy was applied. The relative percentages of CS/WP-VOCs were higher than those of SBC-VOCs, and most VOCs tended to have relatively small carbon skeletons as the average carbon oxidation state increased. With the temperature increased from low to high during CS/WP pyrolysis, the primary sequential response of VOCs (acids → phenols/esters → alcohols/ethers/aldehydes/ketones → hydrocarbons/aromatics) corresponded to the RFG response (hydroxyl groups → -CH 3 /-CH 2 -/-CH groups → aliphatic ethers and conjugated ketones). Compared with the relative regularity for CS/WP responses, the gas-solid products from SBC pyrolysis exhibited complex temperature-dependent responses and high oxidation-induced variability. These insights provide favorable strategies for the online monitoring system to facilitate priority removal of coal and biomass fuels-VOCs.

Gambierdiscus spp. are epi-benthic dinoflagellates that have been associated with ciguatera poisoning. These microalgae can have complex secondary metabolite profiles including ciguatoxins, maitotoxins, and gambierones, with varying compositions and toxicities across species and strains. Given this chemical diversity there is a need to develop selective and sensitive methods for secondary metabolite profiling. In this study, we used a cultured Caribbean strain of Gambierdiscus silvae to develop sample preparation and analysis strategies for characterizing vic-diol containing secondary metabolites. A pooled cellular extract was first screened by liquid chromatography-high-resolution mass spectrometry (LC-HRMS) for ciguatoxin-related compounds, which resulted in the confirmation of gambierone (1) and a novel isomer of 44-methylgambierone (3). Treatment of the extract with periodate confirmed that the gambierones each contained one reactive vic-diol, which was exploited for the development of a selective extraction procedure using m-aminophenylboronic acid gel and the non-aqueous binding solvent chloroform. Using this non-traditional boronate affinity procedure, LC-HRMS also revealed the presence of additional sulfated polycyclic ethers in the gambierone-containing vic-diol fraction, while pigments and other contaminants were removed. The developed tools could be applied to screen collections of Gambierdiscus and other benthic algae to provide additional chemical characterization of gambierone-related compounds. Fuel oxygenates (FOs) are mainly ethers or alcohols which are added to gasoline either to boost the octane number or to make the fuel burning process more "cleaner" with increasing the oxygen content, or to obtain a combination of both effects. FOs are water soluble with high mobility in the environment which presence even at very low concentrations lower the quality of water making it unsafe or unpleasant due to their objectionable taste and/or odor. Thus, their determination at trace in environmental samples is of high importance because of their sparingly biodegradability and their biological hazards. Instruments such as gas chromatography, Fourier transform infrared spectroscopy and ion mobility spectrometry are mainly used for the determination of FOs. However, the main challenge for determination of such oxygenates relates to proper sample preparation. Dilute or complex samples often demand a specific treatment to ensure effective enrichment of FOs before their detection. The main techniques used for this purpose are purge and trap, membrane extraction, and solid phase microextraction. This review presents a comprehensive evaluation of extraction/preconcentration techniques and analytical methods for determination of FOs in environmental samples. Advantages and disadvantages of each method are discussed in details along with critical evaluation of currently available methods.

Species of the dinophyte genus Alexandrium are widely distributed and are notorious bloom formers and producers of various potent phycotoxins. The species Alexandrium taylorii is known to form recurrent and dense blooms in the Mediterranean, but its toxin production potential is poorly studied. Here we investigated toxin production potential of a Mediterranean A. taylorii clonal strain by combining state-of-the-art screening for various toxins known to be produced within Alexandrium with a sound morphological and molecular designation of the studied strain. As shown by a detailed thecal plate analysis, morphology of the A. taylorii strain AY7T from the Adriatic Sea conformed with the original species description. Moreover, newly obtained Large Subunit (LSU) and Internal Transcribed Spacers (ITS) rDNA sequences perfectly matched with the majority of other Mediterranean A. taylorii strains from the databases. Based on both ion pair chromatography coupled to post-column derivatization and fluorescence detection (LC-FLD) and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) analysis it is shown that A. taylorii AY7T does not produce paralytic shellfish toxins (PST) above a detection limit of ca. 1 fg cell -1 , and also lacks any traces of spirolides and gymnodimines. The strain caused cell lysis of protistan species due to poorly characterized lytic compounds, with a density of 185 cells mL -1 causing 50% cell lysis of cryptophyte bioassay target cells (EC 50 ). As shown here for the first time A. taylorii AY7T produced goniodomin A (GDA) at a cellular level of 11.7 pg cell -1 . This first report of goniodomin (GD) production of A. taylorii supports the close evolutionary relationship of A. taylorii to other identified GD-producing Alexandrium species. As GD have been causatively linked to fish kills, future studies of Mediterranean A. taylorii blooms should include analysis of GD and should draw attention to potential links to fish kills or other environmental damage.

Deficient ether lipid biosynthesis in rhizomelic chondrodysplasia punctata and other disorders is associated with a wide range of severe symptoms including small stature with proximal shortening of the limbs, contractures, facial dysmorphism, congenital cataracts, ichthyosis, spasticity, microcephaly, and mental disability. Mouse models are available but show less severe symptoms. In both humans and mice, it has remained elusive which of the symptoms can be attributed to lack of plasmanyl or plasmenyl ether lipids. The latter compounds, better known as plasmalogens, harbor a vinyl ether double bond conferring special chemical and physical properties. Discrimination between plasmanyl and plasmenyl ether lipids is a major analytical challenge, especially in complex lipid extracts with many isobaric species. Consequently, these lipids are often neglected also in recent lipidomic studies. Here, we present a comprehensive LC-MS/MS based approach that allows unequivocal distinction of these two lipid subclasses based on their chromatographic properties. The method was validated using a novel plasmalogen-deficient mouse model, which lacks plasmanylethanolamine desaturase and therefore cannot form plasmenyl ether lipids. We demonstrate that plasmanylethanolamine desaturase deficiency causes an accumulation of plasmanyl species, a too little studied but biologically important substance class.

The toxicity and environmental persistence of anthropogenic per- and poly-fluoroalkyl substances (PFAS) are of global concern. To address legacy PFAS concerns in the United States, industry developed numerous replacement PFAS that commonly are treated as confidential information. To investigate the distribution of PFAS in New Jersey, soils collected from across the state were subjected to nontargeted mass-spectral analyses. Ten chloroperfluoropolyether carboxylates were tentatively identified, with at least three congeners in all samples. Nine congeners are ≥(CF 2 ) 7 Distinct chemical formulas and structures, as well as geographic distribution, suggest airborne transport from an industrial source. Lighter congeners dispersed more widely than heavier congeners, with the most widely dispersed detected in an in-stock New Hampshire sample. The environmental concentration of tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE) and tetrabromobisphenol A mono(hydroxyethyl) ether (TBBPA-MHEE), as some of the main derivatives and byproducts of tetrabromobisphenol A (TBBPA), are obscure due to lacking available analytical methods. In the present study, a multiplexed competitive chemiluminescent imaging immunoassay (CLIIA) based on specific monoclonal antibody respectively against TBBPA-MHEE and TBBPA-DHEE has been developed for the simultaneous detection of the two targets. To improve the sensitivity of immunoassay, Au nanoparticles (AuNPs) were utilized as solid support for loading horseradish peroxidase, labeled to obtain the multi-enzyme particles. Under the optimized conditions, the limits of detection (LODs) for TBBPA-MHEE and TBBPA-DHEE were 1.85 ng/mL and 2.05 ng/mL, respectively. After estimating its accuracy and precision, the established method was applied for investigation of the contaminants at the inner rivers in Zhenjiang, Jiangsu Province. Our results indicated that no TBBPA-MHEE was found in all 11 water samples and two samples of TBBPA-DHEE were detected up to 6.34 ng/mL, possibly ascribed to the dense population and slow flow rate around the sampling site. The impact of crop level and leaf removal on Istrian Malvasia (Vitis vinifera L.) white wine aroma and phenolic acids composition was studied over two vine-growing seasons. Two crop levels were combined with leaf removal or untreated control in two-factorial design. Crop level showed limited impact on aroma compounds in wine and the concentration of some esters was increased by higher crop level. In contrast, leaf removal increased the concentration of several aroma compounds and especially monoterpenes and esters. The concentration of hydroxycinnamic acids in wine was enhanced only by leaf removal, while no consistent impact of the investigated factors on hydroxybenzoic acids was observed. The obtained results suggest that in cases where environmental conditions are not limiting, increasing the crop level under adequate microclimate in fruit zone has no detrimental effects on white wine aroma and hydroxycinnamic acids composition, potentially leading to economically more sustainable grape production., (Copyright © 2019 Elsevier Ltd. Brazilian red propolis has been proposed as a new source of compounds with cytotoxic activity. Red propolis is a resinous material of vegetal origin, synthesized from the bees of the Appis mellifera family, with recognized biological properties. To obtain actives of low polarity and high cytotoxic profile from red propolis, in this work, we proposed a new solvent accelerated extraction method. A complete 2 3 factorial design was carried out to evaluate the influence of the independent variables or factors (e.g., temperature, number of cycles, and extraction time) on the dependent variable or response (i.e., yield of production). The extracts were analyzed by gas chromatography coupled with mass spectrometry for the identification of chemical compounds. Gas chromatography analysis revealed the presence of hydrocarbons, alcohols, ketones, ethers, and terpenes, such as lupeol, lupenone, and lupeol acetate, in most of the obtained extracts. To evaluate the cytotoxicity profile of the obtained bioactives, the 3-(4,5-dimethyl-2-thiazole)-2,5-diphenyl-2- H -tetrazolium bromide colorimetric assay was performed in different tumor cell lines (HCT116 and PC3). The results show that the extract obtained from 70 °C and one cycle of extraction of 10 min exhibited the highest cytotoxic activity against the tested cell lines. The highest yield, however, did not indicate the highest cytotoxic activity, but the optimal extraction conditions were indeed dependent on the temperature (i.e., 70 °C)., Competing Interests: The authors declare no conflict of interest.

The spontaneous process of release of plasticizers from membranes typically used in ion-selective sensors is an effect which limits the lifetime of sensors and comes with a risk of safety hazards. We use a nanosponge approach to look at the magnitude of this problem, quantifying the resulting contents of the plasticizer in solution. This novel method takes advantage of the spontaneous partition of the plasticizer (released and present in solution) into nanoparticles loaded with a solvatochromic dye. As a result, nanoparticles are transformed into capsules. This process is coupled with the turn-on fluorescence intensity change of the dye embedded in nanostructures, proportional to analyte concentration in the ppm range, providing insight into plasticizer contents in the solution. It was found that the spontaneous release of the plasticizer is dependent on its nature as well as the presence of an ionophore and ion-exchanger. For a typical ion-selective membrane composition the leakage effect results in up to 20 ppm of 2-nitrophenyl octyl ether found in solution after 12 h contact. On the other hand, for a less polar plasticizer - bis(2-ethylhexyl) sebacate, although the presence of an ionophore and ion-exchanger also increases the amount of the compound released from the membrane, its concentration in solution does not exceed 2 ppm after 12 h. The conclusions presented herein can be important not only for designing robust sensors but also for end-user safety. The results obtained for ion-selective membranes were equal within the range of experimental errors with those obtained using a liquid chromatography coupled with mass spectrometry (LC MS) approach, confirming the high analytical potential of the nanosponge approach.

The floor polish removal (FPR) and reapplication (FPA) are important cleaning tasks in public buildings that have hard floor surfaces. Usually, the FPR and FPA are conducted once or twice a year, during the periodic cleaning of these buildings. The FPR can be performed either chemically (CFPR) or by using dry scrubber (DFPR), when the polish is ground from the floor. In this study, cleaning workers' exposure to volatile organic compounds (VOCs) and particulate matter (PM) during the FPR and FPA, and the differences in the exposures between the two FPR methods were investigated. In total, three buildings located in Central Finland were included, and total of six cleaning workers (two per building) participated in the study. In Buildings 1 and 2, the CFPR and FPA were performed and in Building 3, the DFPR was conducted. TVOC (total volatile organic compounds) concentrations in the breathing zone of the workers during the CFPR were 8,740 and 390 µg/m 3 (SD 3,290 and 180 µg/m 3 ) for Buildings 1 and 2, respectively. During the DFPR in Building 3, the average TVOC concentration was 400 µg/m 3 (SD 180 µg/m 3 , stationary sampling). The TVOC concentrations during the FPA were high, 1,640 and 2,170 µg/m 3 on average (SD 1,570 and 930 µg/m 3 ) for Buildings 1 and 2, respectively. Glycol ethers were the most prominent VOCs during the CFPR and FPA, whereas carboxylic acids were the most common during the DFPR. The inhalable dust concentrations in the workers' breathing zone were noticeably higher during the DFPR (1.55 mg/m 3 on average, SD 0.01 mg/m 3 ) than the CFPR (0.24 mg/m 3 on average, SD 0.05 mg/m 3 ). Finnish occupational exposure limit value for organic inhalable dust is 5 mg/m 3 . As the products used during the CFPR and FPA contain glycol ethers and ethanolamines that are absorbed via the skin as well, the use of skin protection is recommended. Whereas the use of FFP3 respirators and skin protection are recommended during the DFPR to prevent the PM exposure.

A competitive immunosensor was established using an electrochemical amperometric strategy for sensitive detection of tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE), an important derivative of tetrabromobisphenol A (TBBPA). In this system, the amplified electrochemical signal towards the reduction of hydrogen peroxide (H 2 O 2 ) was recorded by amperometric method. Meanwhile, the synthetized catalase functionalized AuNPs-loaded self-assembled polymer nanospheres showed an excellent electrocatalytic ability to catalyse H 2 O 2 , which was beneficial for strengthening the electrochemical signals. Under the optimized conditions, this method displayed: (i) low detection limits (0.12 ng/mL, 7 times lower than the traditional ELISA with the same antibody); (ii) satisfactory accuracy (recoveries, 78-124%; RSD, 2.1-8.3%) and good agreement with the corresponding ELISA; (iii) low sample consumption (6 μL) and low cost. A novel chemiluminescence immunoassay based on luminol-modified gold nanoclusters (AuNCs@Peps@luminol) was developed for simultaneous detection of tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE) and tetrabromobisphenol A mono(hydroxyethyl) ether (TBBPA-MHEE), an important derivative and byproduct of tetrabromobisphenol A (TBBPA), respectively. In the system, alkaline phosphatase (ALP) was labeled on the second antibody (Ab 2 ) for signal amplification. When ALP-Ab 2 was captured by antigen-primary antibody (Ab 1 ) complex, disodium phenyl phosphate (PPNa) generated massive phenol under the catalysis of ALP, markedly inhibiting the chemiluminescence intensity of AuNCs@Peps@luminol. Under the optimized conditions, the calculated detection of limit (LOD, 90% inhibition) was 0.078 μg/L for TBBPA-DHEE with a linear range of 0.23-9.32 μg/L, which was lower 9 times than that of conventional ELISA with the same antibody. An external standard of goniodomin A (GDA) was prepared from a strain of Alexandrium pseudogonyaulax originating from New Zealand and its chemical structure was confirmed by nuclear magnetic resonance (NMR) spectroscopy. Using the GDA standard, an ultra-performance liquid chromatography-tandem mass spectrometric (UPLC-MS/MS) method in selected reaction monitoring (SRM) mode was developed for separation and quantification of GDA. This method was successfully applied to planktonic field samples collected during an oceanographic expedition conducted with R/V Uthörn along the Danish west coast, Limfjord and Kattegat in June 2016. In addition, this method was used to characterize goniodomin (GD) profiles of 17 A. pseudogonyaulax strains from the coastal North Sea and from Limfjord. Highest GDA levels were found in Limfjord (up to 590 ng NT -1 m -1 ), but GDA was also detected in the North Sea appearing at the latitude of Sylt Island northwards and in Kattegat from the eastern mouth of Limfjord down to the Kiel Bight, but at lower abundances than within Limfjord. This is the first reported detection of GDA in planktonic field samples. A gas chromatography with mass spectrometry method was developed for the simultaneous determination of ten kinds of glycol ethers and their acetates in cosmetics. The samples were extracted with methanol/ethyl acetate (80:20, v/v), further treated with vortex and ultrasound, and analyzed by gas chromatography with mass spectrometry. The concentration of each analyte was calibrated by the external standard method. Under the optimal conditions, the analytes showed linear relationship in the range of 0.05-25 mg/L with determination coefficients larger than 0.9987. The limits of detection and quantification were in the range of 0.09-0.59 and 0.31-1.95 mg/kg, respectively. The average recoveries of three spiked levels were 80.2-105.4% with intra- and interday precisions of 1.1-6.3 and 1.9-6.5%, respectively. Method validation from different labs confirmed the satisfactory recoveries and precisions. Based on our produced polyclonal antibody capable of recognizing tetrabromobisphenol A bis(2-hydroxyethyl) ether (TBBPA-DHEE) and tetrabromobisphenol A mono(hydroxyethyl) ether (TBBPA-MHEE) (cross-reactivity, 100% for TBBPA DHEE; 98.7% for TBBPA MHEE), an important derivative and byproduct of tetrabromobisphenol A (TBBPA), respectively, a novel ultrasensitive competitive immunosensor was established using an electrochemical impedimetric strategy for the simultaneous detection of both chemicals. A significantly amplified electrochemical impedance spectroscopy (EIS) for quantitative target analysis was obtained through (i) the biocatalytic precipitation of 4-chloro-1-naphthol (CN) on the electrode surface triggered by horseradish peroxidase (HRP) and (ii) increased amounts of the enzyme with HRP-loaded silica nanoparticles carrying poly-brushes (SiO 2 @PAA) as labels, achieving a remarkable improvement in catalytic performance. Under the optimized conditions, the immunosensor showed satisfactory accuracy (recovery, 84.6-118%) and a good linear range (0.21- 111.31ng/mL) with a limit of detection (LOD) of 0.08ng/mL (S/N = 3) for TBBPA DHEE and TBBPA MHEE. The aim of this investigation is to clarify the types and concentrations of VOCs present in various commercial household water-based hand pump spray products used in Japan, and to estimate their average concentrations in indoor air when the spray product is used. We selected glycol and glycol ethers as the main target compounds, as these chemicals were detected at high frequencies and concentrations in a national survey of Japanese indoor air pollution. The extraction of these chemicals using graphite carbon cartridges was examined, with good recoveries and reproducibilities being obtained. Eighteen chemicals were analyzed in 54 commercial products and 8 chemicals were detected. More specifically, dipropylene glycol (DPG) was present in 44 samples (1.1 × 10 1 -1.8 × 10 4 μg/mL); propylene glycol (PG) was present in 22 samples (1.5 × 10 1 -2.9 × 10 4 μg/mL); diethylene glycol monoethyl ether (DGMEE) was found in 15 samples (trace amount-1.9 × 10 3 μg/mL); diethylene glycol (DEG) was present in 9 samples (1.0 × 10 1 -2.4 × 10 3 μg/mL); 1,3-butandiol (13BG) was found in 5 samples (trace amount-7.4 × 10 3 μg/mL); 2-ethyl-1-hexanol (2E1H) was detected in 5 samples (3.2 × 10 -1 -4.4 × 10 1 μg/mL); diethylene glycol monobutyl ether (DGMBE) was present in 4 samples (2.1 × 10 1 -7.1 × 10 1 μg/mL); and 3-methoxy-3-methylbutanol (MMB) was found in 2 samples (2.4 × 10 1 -4.7 × 10 2 μg/mL). In addition, the average concentrations of these chemicals in indoor air were estimated using their maximum concentrations observed in the spray product. The estimated average concentrations of the chemicals in indoor air were determined to range between 1.0 × 10 -2 and 1.0 mg/m 3 , with the exception of 2E1H and DGMBE. Furthermore, the estimated average concentrations of PG, 13BG, and DGMEE in indoor air were comparable to or higher than those reported in a national survey of Japanese indoor air pollution. It therefore appeared that household water-based hand pump sprays may contribute to the presence of these chemicals in indoor air. In contrast, estimated average concentrations of 2E1H in indoor air were low, its concentrations observed in a national survey of Japanese indoor air pollution are likely due to the use of plasticizers and paints.

The migration and accumulation of octachlorodipropyl ether (OCDPE) in soil-tea systems were investigated using a gas chromatography-electron capture detector (GC-ECD) method in young and old tea gardens. When the residual concentration of OCDPE was 100 g a.i. hm -2 in soils, the peak concentrations of OCDPE in fresh leaves of young and old tea plants were 0.365 mg/kg and 0.144 mg/kg, taking 45 days and 55 days, respectively. Equations for the accumulation curves of OCDPE in fresh leaves of young and old tea plants were C t = 0.0227e 0.0566t (R² = 0.9154) and C t = 0.0298e -0.0306t (R² = 0.7156), and were C t = 3.8435e 0.055t (R² = 0.9698) and C t = 1.5627e -0.048t (R² = 0.9634) for dissipation curves, with a half-life of 14.4 days and 12.6 days, respectively. Flame ionized detector has been such widely applied in chemical analysis since its great invention and the discovery of chem-ionization. Thanks to the excellent contribution of Sternberg and the successors in this field, effective carbon number concept (ECN) can make the analysis calibration greatly simplified especially when authentic standard substances are unavailable or in the complicated case such as petrochemical industry and biomass processing. To supplement the ECN rule in multi-ethers, this work determined relative response factors of poly(oxymethylene) dimethyl ethers experimentally, and developed a probabilistic P 1 -P 2 effective carbon number model (P-ECN) for multi-ethers compounds. showed this method could improve the precision of quantitative analysis for poly(oxymethylene) dimethyl ethers and could predict relative response factors of other ethers with similar structure to a degree. LOD for each DMMn monomer ranged between 0.4-0.7 ng, and MDL ranged between 2 and 4 ug/mL. In the practical quantitation of diluted samples at level of 10 ug/mL, the relative standard deviation was less than 5%. In this study, we isolated and identified an aggregation-sex pheromone from Monochamus saltuarius, the major insect vector of the pine wood nematode in Korea. Adult males of M. saltuarius produce 2-undecyloxy-1-ethanol, which is known as an aggregation-sex pheromone in other Monochamus species. We performed field experiments to determine the attractiveness of the pheromone and other synergists. More M. saltuarius adult beetles were attracted to traps baited with the pheromone than to unbaited traps. Ethanol and (-)-α-pinene interacted synergistically with the pheromone. Traps baited with the pheromone + (-)-α-pinene +ethanol were more attractive to M. saltuarius adults than traps baited with the pheromone, (-)-α-pinene, or ethanol alone. Ipsenol, ipsdienol, and limonene were also identified as synergists of the aggregation-sex pheromone for M. saltuarius adults. In field experiments, the proportion of females was much higher in the beetles caught in traps than among the beetles emerging from naturally-infested logs in the laboratory. Our results suggest that a combination of aggregation-sex pheromone and synergists could be very effective for monitoring and managing M. saltuarius.

Hainanmycin is a new veterinary polyether antibiotic and has few sensitive analytical method in present days. In this study, a liquid chromatography-tandem mass spectrometry (LC-MS/MS) relying on multiple reaction monitoring (MRM) detection was developed for analysis of hainanmycin in animal feed. Feed samples were extracted with ethyl acetate and purified by two steps of liquid-liquid extraction (LLE) to get rid of water solvable matrix and lipids one by one. The final simple was analyzed by LC-MS/MS. The LC mobile phase was composed of 0.1% aqueous formic acid and 0.1% formic acidified acetonitrile by gradient elution. Average recoveries ranged from 74.22% to 87.85%, as determined by spiking with 2.0 (LOQ) ∼2500μgkg -1 of hainanmycin. The inter-day and intra-day coefficient of variation was 9.21% to 11.77% and 7.67% to 13.49%, respectively. A simple and universal analytical method based on ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) for high throughput screening of 21 bisphenols, bisphenols digycidyl ethers and their derivatives in dairy products was developed. Response Surface Methodology (RSM) was used to optimize sample preparation conditions based on a quick, easy, cheap, effective, rugged and safe (QuEChERS) method. The analytes were extracted by using 15 mL acetonitrile with 1% acetic acid, and the extracts were further purified by using 190 mg of C18 and 390 mg of PSA. The extracts were analyzed by UHPLC-MS/MS with electrospray ionization (ESI) source. Linearity was assessed by using matrix-matched standard calibration and good correlation coefficients (r 2  > 0.99) were obtained. The limits of quantitation (LOQs) for the analytes ranged from 0.02 to 5 μg kg -1 . The extraction recoveries were in a range of 88.2%-108.2%. Good method reproducibility in terms of intra- and inter-day precision was observed, yielding relative standard deviations (RSDs) less than 8.9% and 9.9%, respectively. Soils, paleosols and terrestrial sediments serve as archives for studying climate change, and represent important terrestrial carbon pools. Archive functioning relies on the chronological integrity of the respective units. Incorporation of younger organic matter (OM) e.g. by plant roots and associated microorganisms into deep subsoil and underlying soil parent material may reduce reliability of paleoenvironmental records and stability of buried OM. Long-term effects of sedimentary characteristics and deep rooting on deep subsoil microbial communities remain largely unknown. We characterized fossil and living microbial communities based on molecular markers in a Central European Late Pleistocene loess-paleosol sequence containing recent and ancient roots with ages of several millenia. The molecular approach, comprising free and phospholipid fatty acids (FAs), core and intact polar glycerol dialkyl glycerol tetraethers (GDGTs), as well as 16S rRNA genes from bacterial DNA, revealed the presence of living microorganisms along the sequence, with bacterial community composition comparable to that of modern topsoils. Up to 88% redundancy between bacterial genetic fingerprint and molecular signature of fossil microorganisms suggested a time-integrated signal of the molecular markers accumulated over a time span potentially lasting from sedimentation over one or more rooting phases until today. Free FAs, core GDGTs and DNA, considered as remains of fossil microorganisms, corresponded with ancient and recent root quantities, whereas phospholipid FAs and intact polar GDGTs, presumably derived from living microorganisms, correlated only with living roots. The biogeochemical and ecological disequilibrium induced by postsedimentary rooting may entail long-term microbial processes like OM mineralization, which may continue even millenia after the lifetime of the root. Deep roots and their fossil remains have been observed in various terrestrial settings, and roots as well as associated microorganisms cause both, OM incorporation and mineralization. Surface porous polyether-ether-ketone has the ability to maintain the tensile monotonic and cyclic strength necessary for many load bearing orthopedic applications while providing a surface that facilitates bone ingrowth; however, the relevant deformation behavior of the pore architecture in response to various loading conditions is not yet fully characterized or understood. The focus of this study was to examine the compressive and wear behavior of the surface porous architecture using micro Computed Tomography (micro CT). Pore architectures of various depths (~0.5-2.5mm) and pore sizes (212-508µm) were manufactured using a melt extrusion and porogen leaching process. Compression testing revealed that the pore architecture deforms in the typical three staged linear elastic, plastic, and densification stages characteristic of porous materials. The experimental moduli and yield strengths decreased as the porosity increased but there was no difference in properties between pore sizes. The porous architecture maintained a high degree of porosity available for bone-ingrowth at all strains. A novel 7,10-diphenylfluoranthene grafted polysiloxane (DPFP) was synthesized and statically coated on a fused-silica capillary column. High column efficiency (3864 plates per m) was achieved for naphthalene at 120°C. Thermo-gravimetric analysis showed that the DPFP polymer began to decompose at 380°C. The chromatogram of polyethylene pyrolysis products indicated that the maximum allowable temperature of the DPFP column could reach 360°C. By relying on its specific π-π stacking and dipole-induced dipole interactions with aromatic solutes, the DPFP stationary phase achieved excellent resolution for substituted benzene and aromatic hydrocarbons with satisfactory peak shapes, as well as showed superiority to current stationary phases that failed to resolve some critical pairs. Many of the molecular proxies commonly used for paleoenvironmental reconstruction are focused on a limited set of glycerol ether lipids, mainly due to the lack of more comprehensive analytical methods and instrumentation able to deal with a more diverse range of species. In this study, we describe an FTICR-MS-based method for rapid, nontargeted screening of ether lipid biomarkers in recent marine sediments. This method involves simplified sample preparation and enables rapid identification of known and novel ether lipid species. Using this method, we were able to identify complete series of core glycerol dialkyl glycerol tetraethers (GDGTs with 0 to 8 alicyclic rings), including the complete resolution of GDGT-4 and the unexpected detection of GDGTs with more than 5 rings, in sediments from mesophilic marine environments (sea surface temperature, SST, of 24-25 °C). Additionally, mono- and dihydroxy-GDGT analogs (including novel species with >2 rings), as well as glycerol dialkanol diethers, GDDs (including novel species with >5 rings) were detected. Finally, we putatively identified other, previously unreported groups of glycerol ether lipid species. Adequacy of the APPI-P FTICR-MS data for the determination of commonly used GDGT-based proxy indices was demonstrated. The results of this study show great potential for the use of FTICR-MS as both a rapid method for determining existing proxy indices and, perhaps more importantly, as a tool for the early detection of possible new biomarkers and proxies that may establish novel geochemical relationships between archaeal ether lipids and key environmental-, energy-, and climate-related system variables.

Recent scientific scrutiny and concerns over exposure, toxicity, and risk have led to international regulatory efforts resulting in the reduction or elimination of certain perfluorinated compounds from various products and waste streams. Some manufacturers have started producing shorter chain per- and polyfluorinated compounds to try to reduce the potential for bioaccumulation in humans and wildlife. Some of these new compounds contain central ether oxygens or other minor modifications of traditional perfluorinated structures. At present, there has been very limited information published on these "replacement chemistries" in the peer-reviewed literature. In this study we used a time-of-flight mass spectrometry detector (LC-ESI-TOFMS) to identify fluorinated compounds in natural waters collected from locations with historical perfluorinated compound contamination. Our workflow for discovery of chemicals included sequential sampling of surface water for identification of potential sources, nontargeted TOFMS analysis, molecular feature extraction (MFE) of samples, and evaluation of features unique to the sample with source inputs. Specifically, compounds were tentatively identified by (1) accurate mass determination of parent and/or related adducts and fragments from in-source collision-induced dissociation (CID), (2) in-depth evaluation of in-source adducts formed during analysis, and (3) confirmation with authentic standards when available. We observed groups of compounds in homologous series that differed by multiples of CF2 (m/z 49.9968) or CF2O (m/z 65.9917). Compounds in each series were chromatographically separated and had comparable fragments and adducts produced during analysis. We detected 12 novel perfluoroalkyl ether carboxylic and sulfonic acids in surface water in North Carolina, USA using this approach. A key piece of evidence was the discovery of accurate mass in-source n-mer formation (H(+) and Na(+)) differing by m/z 21.9819, corresponding to the mass difference between the protonated and sodiated dimers.

The dissipation behavior of octachlorodipropyl ether (OCDPE) residues in fresh tea shoots and in tea prepared under field conditions was investigated, and the transfer of residues from brewed tea to tea infusion was determined. OCDPE levels in tea shoots, prepared tea, tea infusion, and spent tea leaves were determined using a sensitive and simple method. The dissipation of OCDPE is fairly slow in tea shoots and prepared tea, with half-life values of 5.10 and 5.46 days, respectively. The degradation rates of OCDPE residues in tea processing were 23.9-43.1 %. The terminal residues of OCDPE in tea shoots and prepared tea samples after 20 and 30 days of OCDPE application were higher than 0.01 mg/kg. However, OCDPE's transfer rates from brewed tea to tea infusion were only 6.0-14.8 %. Further studies on risk assessment of OCDPE residue in tea on the basis of the relationship of OCDPE in prepared tea and infusion are warranted.

Countercurrent chromatography (CCC) is a technique, which uses two immiscible liquid phases for a separation process in a long and hollow tube. The technique allows the separation of high amounts of sample (50mg to several grams) with a low consumption of solvents. In this study, we fractionated 50mg technical octabromodiphenyl ether (DE-79) and analyzed the fractions by gas chromatography with mass spectrometry (GC/MS) and proton nuclear magnetic resonance ((1)H NMR) spectroscopy. CCC separations were performed with n-hexane/acetonitrile as solvent system in tail-to-head (i.e. the upper phase is mobile) mode. Twelve CCC fractions were studied for the PBDE composition. CCC elution of PBDE congeners was dependent both on the degree of bromination and substitution pattern. Higher brominated congeners eluted faster than lower brominated congeners and isomers with vicinal hydrogen atoms eluted last. In addition to several known PBDE congeners in DE-79, we were able to unequivocally identify BDE 195 in DE-79 and we could verify the presence of BDE 184. Finally, we also established the online hyphenation of CCC with (1)H NMR. The use of deuterated solvents could be avoided by using n-hexane/acetonitrile as two-phase system. The bivalve mollusc, Crassostrea virginica, is frequently exposed to blooms of Karenia brevis along the west coast of Florida during periods of spawning and early larval development. A continuous 4-day exposure of gametes and 2-4 cell stage embryos of C. virginica to whole-cell and culture filtrate of K. brevis at 500 and 5000 cells mL(-1), was followed by a 4-day 'recovery' period. Larval growth, percent of normal, abnormal and dead larvae, and the presence of food in the larval gut were measured throughout the exposure period. Results suggest that negative effects mainly occur during embryogenesis and early development. Damage to feeding apparatus/gut may occur during embryonic development or exposure to toxins may act as a feeding deterrent on non-toxic algae. Following 2-h in vitro exposure of gametes, differences in oocyte and sperm cell parameters were investigated using flow cytometry. The reduced sperm viability in the whole-cell 5000 cells mL(-1) treatment suggests the involvement of extracellular brevetoxins (PbTx) and perhaps other harmful, uncharacterized compounds associated with the K. brevis cell membrane. The cumulative effects of reduced sperm viability, fertilization success, embryonic and larval survival, and the near-annual exposure to blooms of K. brevis could cause significant bottlenecks on oyster recruitment., (Copyright © 2015 Elsevier Ltd. Multidimensional gas chromatography is an appropriate tool for the non-targeted and comprehensive characterisation of complex samples generated from combustion processes. Particulate matter (PM) emission is composed of a large number of compounds, including condensed semi-volatile organic compounds (SVOCs). However, the complex amount of information gained from such comprehensive techniques is associated with difficult and time-consuming data analysis. Because of this obstacle, two-dimensional gas chromatography still receives relatively little use in aerosol science [1-4]. To remedy this problem, advanced scripting algorithms based on knowledge-based rules (KBRs) were developed in-house and applied to GCxGC-TOFMS data. Previously reported KBRs and newer findings were considered for the development of these algorithms. The novelty of the presented advanced scripting tools is a notably selective search criterion for data screening, which is primarily based on fragmentation patterns and the presence of specific fragments. Combined with "classical" approaches based on retention times, a fast, accurate and automated data evaluation method was developed, which was evaluated qualitatively and quantitatively for type 1 and type 2 errors. The method's applicability was further tested for PM filter samples obtained from ship fuel combustion. Major substance classes, including polycyclic aromatic hydrocarbons (PAH), alkanes, benzenes, esters and ethers, can be targeted. This approach allows the classification of approximately 75% of the peaks of interest within real PM samples. This paper presents, on the one hand, the study of the influence of the pH of the medium on the determination of bisphenol F (BPF), bisphenol A (BPA) and their corresponding diglycidyl ethers (BFDGE and BADGE, respectively) by GC-MS after a solid-phase extraction step, using BPA-d16 as internal standard and Parallel Factor Analysis (PARAFAC) decomposition as a multi-way tool for the unequivocal identification and quantification of the four analytes. As the structure of both BFDGE and BADGE has two 2,3-epoxypropoxy groups that can undergo an acid- or base-catalyzed ring-opening via nucleophilic substitution reactions, several samples spiked with the four analytes were set to different pH values between 2 and 12. The best results were obtained in the pH region 8-10, being 9 the most suitable value. Coelution of interferents was overcome using the PARAFAC decomposition; otherwise, the presence of some analytes could not have been ensured according to the regulations currently in force. Secondly, the release of BPA from polycarbonate glasses into food simulant D1 (ethanol 50% (v/v)) over time was studied through seven migration tests and the differences found in this migration process with the incubation temperature (50 and 70°C) were evaluated. A nonlinear regression was used to fit the experimental data following an exponential relation between the concentration of BPA transferred from every glass and the respective migration test. None of the quantities of BPA released exceeded the specific migration limit of 0.6mgkg(-1) laid down for this compound in the Commission Regulation (EU) No 10/2011, so the compliance of the glasses evaluated was ensured. The average recovery percentages of the four analytes at a fortification level of 800ngL(-1) ranged from 50.14 to 92.75%. The in vitro potential of sensitizers and related compounds (SRCs) originating from impurities in waste paper in activating the human aryl hydrocarbon receptor (AhR) α was assessed using yeast reporter gene as well as cytochrome P450 (CYP)1A1 and ethoxyresorufin O-deethylase (EROD) assays. In the yeast assay, eight compounds exhibited agonist activity, and their activity relative to β-naphthoflavone (BNF) ranged from 1.4 × 10(-4) to 8.3 × 10(-2), with the highest activity observed for benzyl 2-naphthyl ether (BNE). In the EROD assay, six compounds caused a more significant induction of CYP1A-dependent activity than did the vehicle control at 50 μM (p<0.01), and their induction levels were 5.1- to 11-fold more potent; 1,2-bis(3-methylphenoxy)ethane (BME) was the most effective inducer. The water from the waste paper recycling area was fractioned using solid-phase extraction (SPE) combined with a C18 disk and florisil cartridge. In gas chromatography-mass spectrometry (GC-MS) analysis, SRCs were detected in the first fraction, at a total concentration of 5.5 μg/L. This fraction also activated AhR, and its activity, expressed as a BNF equivalent value, was 0.42 nM in the yeast assay. The contribution ratio of active compounds accounted for up to 34% and 4.4% observed activity of the fraction and total samples, respectively. Recently, iso-diabolic acid (13,16-dimethyl octacosanedioic acid) has been identified as a major membrane-spanning lipid of subdivisions 1 and 3 of the Acidobacteria, a highly diverse phylum within the Bacteria. This finding pointed to the Acidobacteria as a potential source for the bacterial glycerol dialkyl glycerol tetraethers that occur ubiquitously in peat, soil, lakes, and hot springs. Here, we examined the lipid composition of seven phylogenetically divergent strains of subdivision 4 of the Acidobacteria, a bacterial group that is commonly encountered in soil. Acid hydrolysis of total cell material released iso-diabolic acid derivatives in substantial quantities (11 to 48% of all fatty acids). In contrast to subdivisions 1 and 3 of the Acidobacteria, 6 out of the 7 species of subdivision 4 (excepting "Candidatus Chloracidobacterium thermophilum") contained iso-diabolic acid ether bound to a glycerol in larger fractional abundance than iso-diabolic acid itself. This is in agreement with the analysis of intact polar lipids (IPLs) by high-performance liquid chromatography-mass spectrometry (HPLC-MS), which showed the dominance of mixed ether-ester glycerides. iso-Diabolic acid-containing IPLs were not identified, because these IPLs are not released with a Bligh-Dyer extraction, as observed before when studying lipid compositions of subdivisions 1 and 3 of the Acidobacteria. The presence of ether bonds in the membrane lipids does not seem to be an adaptation to temperature, because the five mesophilic isolates contained a larger amount of ether lipids than the thermophile "Ca. Chloracidobacterium thermophilum." Rationale: Free sterols are neutral molecules that are difficult to analyze by MALDI or ESI and their molecular ions easily fragment. In order to increase their ionization efficiency and selectivity, sterols were derivatized by different reagents., Methods: Selected sterols were converted into their corresponding picolinyl esters, N-methylpyridyl ethers and sulphated esters. The derivatives were optimized for MALDI-TOFMS analysis through proper selection of the matrix. MALDI-TOF/TOF experiments were carried out to study the fragmentation pathways of the derivatives and their use in structural elucidation. Lipid extracts from mussels were used as test samples for MALDI analysis of sterols in biological samples also analyzed by GC/MS for comparison., Results: Sterol picolinyl esters were identified as sodiated adducts [M+Na](+) and the signal significantly enhanced after addition of sodium acetate (20 mM). Sterol N-methylpyridyl ethers were easily detected as [M](+) while sulphated sterols were best detected as [M-H](-). The ester bonds of picolinyl and sulphated esters easily cleaved in MS/MS resulting in diagnostic derivative fragments at m/z 146.03 and 96.89, respectively. Cleavage of the ether bond of N-methylpyridyl ethers gave a diagnostic fragment ion at m/z 110.04. Sterol profiles in mussels obtained by MALDI-TOFMS were in close agreement with those obtained by GC/MS. Two sterols (cholesterol and β-sitosterol) were selected for quantification as their sulphated and picolinyl esters. Calibration curves gave excellent correlation coefficients., Conclusions: Suitable matrices for picolinyl esters are DHB and THAP, for N-methylpyridyl ethers THAP, and for sulphated esters p-nitroaniline and dithranol. Using cholesterol, the limits of detection (LODs) for sulphated esters were 0.2 µg/mL and for picolinyl esters, 1.5 µg/mL. Smog chamber/gas chromatography techniques are used to investigate the atmospheric degradation of fluroxene, an anesthetic, through oxidation with OH and Cl radicals at 298 K and under atmospheric pressure of N2 or air. The measured rate constants (k) are: k(fluroxene+OH(.) )=(2.96±0.61)×10(-11) and k(fluroxene+Cl(.) )=(1.62±0.19)×10(-10) cm(3)  molecule(-1)  s(-1) . The only product detected after the oxidation of fluroxene with OH radicals is 2,2,2-trifluoroethyl formate (79 % and 83 % molar yield in the absence and presence of NOx, respectively). However, after oxidation with Cl radicals, the detected products are 2,2,2-trifluoroethyl formate (78 %), 2,2,2-trifluoroethyl-1-chloroacetate (5 %), and chloroacetaldehyde (4 %), in the absence of NOx, and 2,2,2-trifluoroethyl formate (93 %), 2,2,2-trifluoroethyl-1-chloroacetate (6 %), and chloroacetaldehyde (5 %), in the presence of NOx. The results indicate that, both in the absence and presence of NOx, the main fate of fluroxene is the addition of the oxidant to the double bond and, once the alkoxy radical is formed, the main decomposition pathway is by means of degradation. Moreover, it is expected that 2,2,2-trifluoroethyl formate is the only oxidation product able to actively contribute to climate change. To successfully assess the contribution of fluroxene to global warming, we measure the infrared spectra of fluroxene and 2,2,2-trifluoroethyl formate, and calculate the radiative efficiencies (REs) to be 0.27 and 0.28 W m(-2)  ppbv(-1) , respectively. In addition, the cumulative effect owing to the formation of 2,2,2-trifluoroethyl formate is investigated, and the direct, indirect, and net global-warming potentials are calculated by using the REs and lifetimes of fluroxene and 2,2,2-trifluoroethyl formate., (Copyright © 2013 WILEY-VCH Verlag GmbH & Co. The behavior of organophosphates and ethers during riverbank filtration and groundwater flow was assessed to determine their suitability as organic tracers. Four sampling campaigns were conducted at the Oderbruch polder, Germany to establish the presence of chlorinated flame retardants (TCEP, TCPP, TDCP), non-chlorinated plasticizers (TBEP, TiBP, TnBP), and hydrophilic ethers (1,4-dioxane, monoglyme, diglyme, triglyme, tetraglyme) in the Oder River, main drainage ditch, and anoxic aquifer. Selected parameters were measured in order to determine the hydro-chemical composition of both, river water and groundwater. The results of the study confirm that organophosphates (OPs) are more readily attenuated during bank filtration compared to ethers. Both in the river and the groundwater, TCPP was the most abundant OP with concentrations in the main drainage ditch ranging between 105 and 958 ng L(-1). 1,4-dioxane, triglyme, and tetraglyme demonstrated persistent behavior during bank filtration and in the anoxic groundwater. In the drainage ditch concentrations of 1,4-dioxane, triglyme, and tetraglyme ranged between 1090 and 1467 ng L(-1), 37 and 149 ng L(-1), and 496 and 1403 ng L(-1), respectively. A positive correlation was found for the inorganic tracer chloride with 1,4-dioxane and tetraglyme. These results confirm the possible application of these ethers as environmental organic tracers. Both inorganic and organic compounds showed temporal variability in the surface- and groundwater. Discharge of the river water, concentrations of analytes at the time of infiltration and attenuation were identified as factors influencing the variable amounts of the analytes in the surface and groundwater. These findings are also of great importance for the production of drinking water via bank filtration and natural and artificial groundwater recharge as the physicochemical properties of ethers create challenges in their removal., (Copyright © 2013 Elsevier B.V.
