Abstract

In genome mapping experiments, long DNA molecules are stretched by confining them to very narrow channels, so that the locations of sequence-specific fluorescent labels along the channel axis provide large-scale genomic information. It is difficult, however, to make the channels narrow enough so that the DNA molecule is fully stretched. In practice, its conformations may form hairpins that change the spacings between internal segments of the DNA molecule, and thus the label locations along the channel axis. Here, we describe a theory for the distribution of label spacings that explains the heavy tails observed in distributions of label spacings in genome mapping experiments. [ABSTRACT FROM AUTHOR]

Abstract

The relation between the shape of the force driving a turbulent flow and the upper bound on the dimensionless dissipation factor $\beta$ is presented. We are interested in non-trivial (more than two wave numbers) forcing functions in a three dimensional domain periodic in all directions. A comparative analysis between results given by the optimization problem and the results of Direct Numerical Simulations is performed. We report that the bound on the dissipation factor in the case of infinite Reynolds numbers have the same qualitative behavior as for the dissipation factor at finite Reynolds number. As predicted by the analysis, the dissipation factor depends strongly on the force shape. However, the optimization problem does not predict accurately the quantitative behavior. We complete our study by analyzing the mean flow profile in relation to the Stokes flow profile and the optimal multiplier profile shape for different force-shapes. We observe that in our 3D-periodic domain, the mean velocity profile and the Stokes flow profile reproduce all the characteristic features of the force-shape. 22. State of the art: gene therapy of haemophilia.

Abstract

Clinical gene therapy has been practiced for more than a quarter century and the first products are finally gaining regulatory/marketing approval. As of 2016, there have been 11 haemophilia gene therapy clinical trials of which six are currently open. Each of the ongoing phase 1/2 trials is testing a variation of a liver-directed adeno-associated viral (AAV) vector encoding either factor VIII (FVIII) or factor IX (FIX) . As summarized herein, the clinical results to date have been mixed with some perceived success and a clear recognition of the immune response to AAV as an obstacle to therapeutic success. We also attempt to highlight promising late-stage preclinical activities for AAV-FVIII where, due to inherent challenges with manufacture, delivery and transgene product biosynthesis, more technological development has been necessary to achieve results comparable to what has been observed previously for AAV-FIX. Finally, we describe the development of a stem cell-based lentiviral vector gene therapy product that has the potential to provide lifelong production of FVIII and provide a functional 'cure' for haemophilia A. Integral to this program has been the incorporation of a blood cell-specific gene expression element driving the production of a bioengineered FVIII designed for optimal efficiency. As clearly outlined herein, haemophilia remains at the forefront of the rapidly advancing clinical gene therapy field where there exists a shared expectation that transformational advances are on the horizon. Abstract

Far-infrared, near-infrared, millimeter-wave, and optical data are presented for two of the large molecular clouds which border the W5 (IC 1848) giant H II region. These data are combined to identify three B0-B2 zero-age main-sequence stars which are forming in these clouds and to study the density distribution and kinematic structure of their surrounding molecular gas. This investigation into the environment in which massive stars have formed in these clouds, when considered alongside similar studies of the W5A/S201 molecular cloud, leads to the conclusion that the expanding W5 H II region has played an important role in initiating the formation of these massive stars.

Abstract

The relation between the form of a body force driving a turbulent shear flow and the dissipation factor β = ϵℓ/U3 is investigated by means of rigorous upper bound analysis and direct numerical simulation. We consider unidirectional steady forcing functions in a three-dimensional periodic domain and observe that a rigorous infinite Reynolds number bound on β displays the same qualitative behaviour as the computationally measured dissipation factor at finite Reynolds number as the force profile is varied. We also compare the measured mean flow profiles with the Stokes flow profile for the same forcing. The mean and Stokes flow profiles are strikingly similar at the Reynolds numbers obtained in the numerical simulations, lending quantitative credence to the notion of a turbulent eddy viscosity. A new variational problem for upper bounds on the rate of energy dissipation in body-forced shear flows is formulated by including a balance parameter in the derivation from the Navier–Stokes equations. The resulting min–max problem is investigated computationally, producing new estimates that quantitatively improve previously obtained rigorous bounds.
