SUMMARY: The diatom genus Druehlago was established to differentiate the type species from Craspedostauros and Achnanthes, on the basis of unique heteropolar, cuneate frustules and numerous lenticular plastids. The phylogenetic placement of Druehlago has, until now, relied solely on morphological evidence, suggesting a close relationship with the genus Craspedostauros. In the present study, we discovered a population of Druehlago cf. cuneata along the coast of Japan and established a cultured strain. Molecular phylogenetic analysis based on rbcL, psbC and 18S rRNA genes was conducted to explore the phylogenetic position and classification of Druehlago. Our results revealed that D. cf. cuneata belongs to the Craspedostauros clade, but with weak statistical support, which is, in turn, sister to Achnanthes. Given the lack of support for various nodes within the Craspedostauros clade, constrained trees were constructed based on several hypotheses regarding the phylogenetic position of Druehlago. The hypothesis inferring monophyly of Druehlago and Achnanthes was statistically rejected, which highlights the close relationship between Druehlago and Craspedostauros. However, our topology tests provided no further resolution regarding the phylogenetic position of Druehlago within or outside Craspedostauros. Craspedostauros E.J. Cox is a diatom genus comprising 17 taxa reported from various regions of the world. While many species of Craspedostauros are epibiontic, the taxa have variable ecological preferences. In this study we formally describe Craspedostauros nazmii sp. nov., an epilithic species discovered along the Turkish Black Sea Coast, based on light and scanning electron microscopy. Craspedostauros nazmii sp. nov. is characterized by valves that are lanceolate to narrowly lanceolate, slightly constricted near the apices with uniseriate, parallel throughout the whole valve, transapical striae and and the presence of an apical silica flap. The areolae are distributed over the valve face and the mantle. The differences and similarities between C. nazmii sp. nov. and established species of Craspedostauros are discussed. Based on shape and morphometrics, the most similar species is Craspedostauros capensis, but it is easily distinguished from C. nazmii sp. nov. by its lack of an apical silica flap. The current study focuses on the biological diversity of two strains of Antarctic diatoms (strains IMA082A and IMA088A) collected and isolated from the Ross Sea (Antarctica) during the XXXIV Italian Antarctic Expedition. Both species presented the typical morphological characters of the genus Craspedostauros: cribrate areolae, two "fore-and-aft" chloroplasts and a narrow "stauros". This classification is congruent with the molecular phylogeny based on the concatenated 18S rDNA-rbcL-psbC alignment, which showed that these algae formed a monophyletic lineage including six taxonomically accepted species of Craspedostauros. Since the study of the evolution of this genus and of others raphe-bearing diatoms with a "stauros" is particularly challenging and their phylogeny is still debated, we tested alternative tree topologies to evaluate the relationships among these taxa. The metabolic fingerprinting approach was implemented for the assessment of the chemical diversity of IMA082A and IMA088A. In conclusion, combining (1) traditional morphological features used in diatoms identification, (2) phylogenetic analyses of the small subunit rDNA (18S rDNA), rbcL and psbC genes, and (3) metabolic fingerprint, we described the strains IMA082A and IMA088A as Craspedostauros ineffabilis sp. nov. and Craspedostauros zucchellii sp. nov. as new species, respectively. Two new species of the previously monospecific diatom genus Druehlago were discovered during the ongoing studies exploring the epizoic and epiphytic diatom flora of the Southern African coasts. These species are formally described here as D. craspedostauriformis sp. nov. and D. lobbanii sp. nov. Detailed morphological examination using both light and scanning electron microscopy revealed wedge-shaped frustules with a central constriction containing multiple lenticular chloroplasts, as well as heteropolar valves with wider headpoles and biarcuate raphes, typical of Druehlago. A stauros narrower than the fascia, a feature not yet reported from Druehlago but typical of the morphologically similar genus Craspedostauros, was observed in D. craspedostauriformis. Apart from the presence of a stauros, the latter species can be easily distinguished from other Druehlago members by its large size, low stria density, externally raised raphe-sternum, and girdle bands with a crenate pars interior. Although the gross morphology of D. lobbanii strongly resembled that of D. cuneata, the two species can be distinguished based on clear differences in size and stria density. Moreover, whereas shallow septa run along the almost entire length of the girdle bands in D. cuneata, they are absent in D. lobbanii. Based on the new findings and observations, the taxonomic status of the genus Druehlago and its morphological and ecological similarity to Craspedostauros and Achnanthes are discussed and evaluated. The current study focuses on four species from the primarily marine diatom genus Craspedostauros that were observed growing attached to numerous sea turtles and sea turtle‐associated barnacles from Croatia and South Africa. Three of the examined taxa, C. danayanus sp. nov., C. legouvelloanus sp. nov., and C. macewanii sp. nov., are described based on morphological and, whenever possible, molecular characteristics. The new taxa exhibit characters not previously observed in other members of the genus, such as the presence of more than two rows of cribrate areolae on the girdle bands, shallow perforated septa, and a complete reduction of the stauros. The fourth species, C. alatus, itself recently described from museum sea turtle specimens, is reported for the first time from loggerhead sea turtles rescued in Europe. A 3‐gene phylogenetic analysis including DNA sequence data for three sea turtle‐associated Craspedostauros species and other marine and epizoic diatom taxa indicated that Craspedostauros is monophyletic and sister to Achnanthes. This study, being based on a large number of samples and animal specimens analyzed and using different preservation and processing methods, provides new insights into the ecology and biogeography of the genus and sheds light on the level of intimacy and permanency in the host–epibiont interaction within the epizoic Craspedostauros species. Several populations of a new Craspedostauros species were observed on museum specimens of juvenile green turtle and Kemp’s ridleys collected from Long Island Beach, New York, USA. The new taxon, Craspedostauros alatus Majewska & Ashworth sp. nov., exhibits a distinctive set of morphological features typical of the genus, including cribrate areolae, a stauros narrower than the fascia, multiple doubly perforated girdle bands, and two fore and aft chloroplasts, but clearly differs from all known Craspedostauros species in possessing partially fused proximal helictoglossae forming an internal thickening with a shallow central cavity, distinctive external wing-like silica flaps near the apices, and a combination of valve dimensions and stria density not observed in other taxa. The same taxon was identified in the NCMA culture collection (strain CCMP120), isolated in 1967 from a sample collected from the equatorial upwelling zone in the Pacific Ocean. Nine samples from three sea turtle species were collected from the same location (Long Island Beach). Statistical analyses suggested that the epizoic diatom flora composition was not affected by the collection season but differed among sea turtle species. The most significant difference was observed between the samples with and without C. alatus. Samples with C. alatus were always dominated by Achnanthes elongata and Berkeleya cf. rutilans, while those lacking the new taxon were characterized by remarkably higher contributions of epizoic Poulinea species. This observation suggests that the microhabitat provided by each sea turtle differs among specimens, which may be related to the different stages of biofilm development on the host sea turtle. Several lineages of raphe-bearing diatoms possess a “stauros,” which is a transverse, usually thickened area free of pores across the center of the valve. It has been suggested that this structure has evolved several times across the raphid diatoms, but we have noticed similarities beyond the stauros between two marine genera— Craspedostauros and Staurotropis —in the structure of their pore occlusions. We have isolated, cultured and extracted DNA from several strains of both genera to infer the phylogenetic relationship between these taxa, as well as test the suggested relationship of Craspedostauros to Achnanthes and Mastogloia based on plastid morphology. DNA sequence data (nuclear-encoded rRNA SSU, plastid-encoded rbc L and psb C) suggest that, except for Mastogloia , these genera are closely-related, though not sister taxa. The DNA phylogeny also suggests that the Mastogloiales are not monophyletic, with clades containing Achnanthes and Craspedostauros sister to clades containing taxa in the Bacillariales. Using evidence from molecular and morphological data, we describe the following new taxa: Craspedostauros alyoubii and C. paradoxa from the Red Sea and Guam, respectively; Staurotropis khiyamii and S. americana from the Red Sea and the Gulf of Mexico, respectively; and Dreuhlago cuneata n. gen., n. sp. from Guam. Despite variation in their protoplast organization and wall structure, monoraphid diatoms have traditionally been grouped as a single family or order, intermediate between the araphid and biraphid diatoms. However, the predominantly marine or brackish species of Achnanthes sensu stricto share protoplast and frustule features with representatives of the Mastogloiales rather than with other monoraphid diatoms. Meanwhile, studies of morphogenesis in Achnanthes have revealed that cells become monoraphid by filling in one raphe system during valve formation, indicating that the monoraphid condition is derived rather than primitive. Evidence from light and electron microscopy is presented to support the transfer of Achnanthes to the Mastogloiales, and an emended description of the order is given. It is concluded that the Achnanthales sensu Round et al . is a paraphyletic group and that the closest relatives of the various monoraphid genera must be sought among other raphid diatoms.
