9. Expression of IGF-1 splice variants in women's reproductive system

Abstract

Studies in skeletal muscle following damage have demonstrated splicing of the insulin-like growth factor-1 gene to IGF-1Ea that is the source of mature IGF-I which is a main anabolic factor, IGF-1Eb,the function of which is not known and IGF-1Ec, known as Mechano Growth Factor (MGF). MGF increases the pool of muscle stem cells by activating them to proliferate. The aim of the thesis was to study expression of IGF-1 variants in female reproductive organs where cellular damage may lead to pathology: 1. The levator ani (LA) muscle: significant damage occurs following vaginal delivery and is a key element in pelvic floor physiology. LA samples from pre and post menopausal women were used as controls. 2. Ovarian tissue: repeated ovulations and failure of the epithelium to repair which may be a predisposing factor for ovarian cancer. 3. Myometrium: single monoclonal cellular damage may lead to abnormal proliferation of myometrium and may be a factor in the aetiology of uterine fibroids.

Abstract

The spike protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is critical for virus infection through the engagement of the human ACE2 protein1 and is a major antibody target. Here we show that chronic infection with SARS-CoV-2 leads to viral evolution and reduced sensitivity to neutralizing antibodies in an immunosuppressed individual treated with convalescent plasma, by generating whole-genome ultra-deep sequences for 23 time points that span 101 days and using in vitro techniques to characterize the mutations revealed by sequencing. There was little change in the overall structure of the viral population after two courses of remdesivir during the first 57 days. However, after convalescent plasma therapy, we observed large, dynamic shifts in the viral population, with the emergence of a dominant viral strain that contained a substitution (D796H) in the S2 subunit and a deletion (ΔH69/ΔV70) in the S1 N-terminal domain of the spike protein. As passively transferred serum antibodies diminished, viruses with the escape genotype were reduced in frequency, before returning during a final, unsuccessful course of convalescent plasma treatment. In vitro, the spike double mutant bearing both ΔH69/ΔV70 and D796H conferred modestly decreased sensitivity to convalescent plasma, while maintaining infectivity levels that were similar to the wild-type virus.The spike substitution mutant D796H appeared to be the main contributor to the decreased susceptibility to neutralizing antibodies, but this mutation resulted in an infectivity defect. The spike deletion mutant ΔH69/ΔV70 had a twofold higher level of infectivity than wild-type SARS-CoV-2, possibly compensating for the reduced infectivity of the D796H mutation. These data reveal strong selection on SARS-CoV-2 during convalescent plasma therapy, which is associated with the emergence of viral variants that show evidence of reduced susceptibility to neutralizing antibodies in immunosuppressed individuals.

15. Tool for Experimenting with Concepts of Mobile Robotics as Applied to Children's Education

Abstract

This paper describes the design and implementation of a tool for experimenting with mobile robotics concepts, primarily for use by children and teenagers, or by the general public, without previous experience in robotics. This tool helps children learn about science in an approachable and interactive way, using scientific research principles in captivating activities that help the child develop problem-solving skills within his group. This initiative seeks to spark interest in scientific and engineering careers as a response to diminished enrollment in such courses by young students. The application of this tool was tested on groups of people ranging from schoolchildren to college students with diverse backgrounds and varying levels of robotics experience and knowledge. The following performance parameters were measured: time to build the robotic platform, time to finalize challenges, and the percentage of challenges successfully accomplished. Abstract

Background/objective: Long chain polyunsaturated fatty acids have beneficial effects in preterm neurophysiological development and are semi-essential. Their levels and variation in plasma and red cells in term and preterms are better known than their intestinal absorption. In this paper the absorption of supplemented arachidonic acid (AA) and docosahexaenoic acid (DHA) is evaluated in a preterm group. Design: Four newborn randomized groups were studied. Group T comprised 11 terms on regular formula. Preterms: group P (n = 9) was on a classic preterm formula. Intervention: groups PA (n = 9) and PB (n = 13) were on the same formula but PB contained AA and DHA in similar proportion to breast milk. At 20 days a 3 day metabolic balance was taken for Ca, Pi, Mg, total fat and individual fatty acids (C8-C24, saturated unsaturated). Results: Calcium absorption was (mean [+ or -] s.d.) 51 [+ or -] 13% in terms. In preterms it was respectively 45 [+ or -] 18, 38 [+ or -] 11 and 37 [+ or -] 21%. Total fat absorption was 92.0 [+ or -] 8.0% in terms, and from 95.0 [+ or -] 2.0 to 91.0 [+ or -] 8.0% in preterms. Absorption of 8:0, 10:0 and 12:0 showed a very high and constant rate despite significant intake differences (71533 mg/kg/day). Linoleic acid and [alpha]-linolenic acid were absorbed in the three groups at around 94% regardless of a greater LA intake in group P. Details of absorption (mg/kg/day) were: for AA, intake 17 [+ or -] 7, fecal excretion 5 [+ or -] 4, net retention 12 [+ or -] 5 (75.0 [+ or -] 18%); for DHA, intake 10 [+ or -] 3, fecal excretion 3 [+ or -] 2, net retention 6 [+ or -] 4 (62.3 [+ or -] 30%). Conclusion: Intestinal absorption of fatty acids is high and is comparable in terms and preterms as regards the studied acids. Longer acids were less well absorbed. Abstract

Nanoscale localization of electromagnetic fields near metallic nanostructures underpins the fundamentals and applications of plasmonics. The unavoidable energy loss from plasmon decay, initially seen as a detriment, has now expanded the scope of plasmonic applications to exploit the generated hot carriers. However, quantitative understanding of the spatial localization of these hot carriers, akin to electromagnetic near-field maps, has been elusive. Here we spatially map hot-electron-driven reduction chemistry with 15 nm resolution as a function of time and electromagnetic field polarization for different plasmonic nanostructures. We combine experiments employing a six-electron photo-recycling process that modify the terminal group of a self-assembled monolayer on plasmonic silver nanoantennas, with theoretical predictions from first-principles calculations of non-equilibrium hot-carrier transport in these systems. The resulting localization of reactive regions, determined by hot-carrier transport from high-field regions, paves the way for improving efficiency in hot-carrier extraction science and nanoscale regio-selective surface chemistry., Quantitative understanding of the spatial localization of hot carriers has been elusive. Abstract

Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy with a dismal survival rate. Persistent activation of pancreatic stellate cells (PSCs) can perturb the biomechanical homoeostasis of the tumour microenvironment to favour cancer cell invasion. Here we report that ATRA, an active metabolite of vitamin A, restores mechanical quiescence in PSCs via a mechanism involving a retinoic acid receptor beta (RAR-β)-dependent downregulation of actomyosin (MLC-2) contractility. We show that ATRA reduces the ability of PSCs to generate high traction forces and adapt to extracellular mechanical cues (mechanosensing), as well as suppresses force-mediated extracellular matrix remodelling to inhibit local cancer cell invasion in 3D organotypic models. Our findings implicate a RAR-β/MLC-2 pathway in peritumoural stromal remodelling and mechanosensory-driven activation of PSCs, and further suggest that mechanical reprogramming of PSCs with retinoic acid derivatives might be a viable alternative to stromal ablation strategies for the treatment of PDAC., Persistent activation of pancreatic stellate cells (PSCs) can perturb the biomechanical homeostasis of the tumour microenvironment. Abstract

The interaction between nitrogen-doped graphene defects (N3V1 and N4V2 pyridinic, and N3V1 and N3V3 pyrrolic) and benzene have been investigated by applying density functional theory (DFT), together with the vdW-DF correction. We discovered that only the N3V3 pyrrolic defect is a reactive site (6π-component), forming a cycloadduct with benzene (4π-component) that has energy barriers below 154.38 kJ mol−1 (1.60 eV). The conduction and valence bands (HOMO and LUMO) for N3V3 form a degenerate pair of orbitals at the gamma point, with the same ionization potential (IP) and electron affinity (EA). Likewise, inspection of the orbital symmetries for both systems confirms that these must undergo concerted reactions based on the Woodward and Hoffmann principles of orbital symmetry, with the appropriate orbital occupancies. This is the first time that substitutionally doped graphene has been demonstrated to participate as a 6π-component for cycloaddition reactions with benzene. [ABSTRACT FROM AUTHOR]

Abstract

The NUCLEUS experiment aims for the detection of coherent elastic neutrino-nucleus scattering at a nuclear power reactor with gram-scale, ultra-low-threshold cryogenic detectors. This technology leads to a miniaturization of neutrino detectors and allows to probe physics beyond the Standard Model of particle physics. A 0.5 g NUCLEUS prototype detector, operated above ground in 2017, reached an energy threshold for nuclear recoils of below 20 eV. This sensitivity is achieved with tungsten transition edge sensors which are operating at temperatures of 15 mK and are mainly sensitive to non-thermal phonons. These small recoil energies become accessible for the first time with this technology, which allows collecting large-statistics neutrino event samples with a moderate detector mass. A first-phase cryogenic detector array with a total mass of 10 g enables a 5-sigma observation of coherent scattering within several weeks. We identified a suitable experimental site at the Chooz Nuclear Power Plant and performed muon and neutron background measurements there. The operation of a NUCLEUS cryogenic detector array at such a site requires highly efficient background suppression. NUCLEUS plans to use an innovative technique consisting of separate cryogenic anticoincidence detectors against surface backgrounds and penetrating (gamma, neutron) radiation. Abstract

This paper is endeavored to motivate the use and further research of dissipativeand passive discrete-time systems exploiting their frequency-domain characteristics. Some important features and implications of the dissipativity property in the discrete-time setting are collected. These properties are mainly referred to the stability analysis (feedback stability systems and study of the zero dynamics), the relative degree, and the preservation of passivity under feedback and parallel interconnections. Dissipativity frequency-domain properties are related to some of the most important frequency-domain stability criteria.

Abstract

The occurrence of tiger shark Galeocerdo cuvier in the Atlantic Ocean was assessed using at-sea observer data from multiple pelagic longline fisheries. Geographic positions of 2764 G. cuvier recorded between 1992 and 2013 and covering a wide area of the Atlantic Ocean were compared with the currently accepted distribution ranges of the species. Most records fell outside those ranges in both the Southern and Northern Hemispheres, which strongly suggests that the distribution range of G. cuvier in the open ocean is considerably larger than previously described [ABSTRACT FROM AUTHOR]

Abstract

Swallowing depends on physiological variables that have a decisive influence on the swallowing capacity and on the tracheal stress distribution. Prosthetic implantation modifies these values and the overall performance of the trachea. The objective of this work was to develop a decision support system based on experimental, numerical and statistical approaches, with clinical verification, to help the thoracic surgeon in deciding the position and appropriate dimensions of a Dumon prosthesis for a specific patient in an optimal time and with sufficient robustness. A code for mesh adaptation to any tracheal geometry was implemented and used to develop a robust experimental design, based on the Taguchi's method and the analysis of variance. This design was able to establish the main swallowing influencing factors. The equations to fit the stress and the vertical displacement distributions were obtained. The resulting fitted values were compared to those calculated directly by the finite element method (FEM). Finally, a checking and clinical validation of the statistical study were made, by studying two cases of real patients. The vertical displacements and principal stress distribution obtained for the specific tracheal model were in agreement with those calculated by FE simulations with a maximum absolute error of 1.2 mm and 0.17 MPa, respectively. It was concluded that the resulting decision support tool provides a fast, accurate and simple tool for the thoracic surgeon to predict the stress state of the trachea and the reduction in the ability to swallow after implantation. Abstract

To gain further insight into the immunopathogenesis of bovine tuberculosis (b TB), the cytokine and chemokine expression of cattle experimentally infected with Mycobacterium bovis was analysed in TB granulomas, using immunohistochemistry ( IHC) and laser capture microdissection ( LCM) followed by q PCR. Immunohistochemistry was conducted for cell types using labelling for CD68, CD3, CD4, CD8, WC1 and CD79a and for the cytokines IFN-γ, TNF-α and TGF-β as well as inducible form of nitric oxide synthase (i NOS). q PCR was conducted for m RNA expression of IFN-γ, TNF-α, TGF-β, IL-17 A, IL-22, IL-2, granzyme A and the chemokines CXCL9 and CXCL10. Early stages of granuloma were primarily comprised of epithelioid MΦs expressing high levels of IFN-γ and i NOS, with significantly upregulated expression of CXCL9 and CXCL10 when compared with control tissue. These chemokines displayed a trend of decreasing m RNA expression as lesion progressed, suggesting a higher level of importance during the early stages of the immune response to mycobacterial infection. IL-22 levels showed a strong trend of decrease through granuloma development, and IL-17 A was shown to be upregulated, supporting its investigation as a potential biomarker of b TB. The use of LCM and q PCR may prove especially useful for the study of IL-17 A as previous attempts to analyse its expression using IHC and in situ hybridization proved unsuccessful. 50. The psychology of LUTS: ICI-RS 2011.

Abstract

Introduction There is undoubtedly a link between lower urinary tract and psychological symptoms. The association is likely to be multifactorial, but whether psychological symptoms are causal or coincidental remains unknown. Methods This paper is a result of a think tank session conducted at the ICI-RS meeting 2011 in Bristol and a PubMed search on the subject of psychology and LUTS. Results Recent population studies have shown that LUTS are prevalent and that psychological morbidity is related to severity of LUTS. Evidence suggests that the psychological status of patients plays a significant role in their symptoms perception. Animal studies have shown the importance of programming and emotional stress on subsequent psychological problems. The link between early events in childhood and resulting LUTS in later life remains uncertain. The possibility that for some patients urinary symptoms could be part of a somatizing process requires further consideration. The possibility that patients' symptoms could be part of a somatization process needs further consideration. Neuroendocrine and neuroimaging studies suggest pathways that may link psychological and lower urinary tract functioning from early life, opening new avenues to better understand the treatment of both symptom groups. Conclusion Whilst psychological morbidity will result from any distressing chronic medical condition the possibility of causation, and or maintenance of LUTS through psychological causes requires further research. It is important to recognize that amongst our younger and older patients suffering with LUTS a significant psychological component exists.
