An anti-interference colorimetric sensor array (CSA) technique was developed for the qualitative and quantitative detection of target heavy metals in corn oil. This method involves a binding mechanism that triggers changes in atomic energy levels and visible color changes. A custom-built olfactory visualization device was employed to gather spectral data, revealing distinct CSA color difference patterns. Subsequently, three pattern recognition algorithms were used to create an identification model for the target heavy metals. The results showed that the ACO-KNN (Ant Colony Optimization-K-Nearest Neighbor) model outperformed the other models, achieving accuracy rates of 90.28% and 89.58% for the calibration and prediction sets, respectively. The ACO-PLS (Partial Least Square) model was more stable with the lowest root mean square error of prediction (RMSEP), which were 0.1730 and 0.1180, respectively. A novel superparamagnetic photonic crystal DNA probe (Fe 3 O 4 @SiO 2 @amino@DNA SPC) was developed to enable rapid visual detection of Hg 2+ . This unique photonic crystal (PC) was synthesized by combining superparamagnetic nanospheres with DNA probes. The DNA probe, rich in thymine (T), detects mercury ions through base mismatch, resulting in the formation of T-Hg 2+ -T loop hairpin structures. With the binding of Hg 2+ to the probe attached to superparamagnetic nanospheres, the PC structure assembled by these nanospheres, formed by the magnetic field, was changed. This change enhanced the reflection intensity; it could be quantified using a fiber optic spectrometer and was visible to the naked eye. The Fe 3 O 4 @SiO 2 @amino@DNA SPC, specific to Hg 2+ , exhibited a reflection peak at 679 nm, which intensified with increasing Hg 2+ concentration. Background: Nanozymes have the advantages of cost effective, simple synthesis, high durability and stability, and have been widely used in various fields. However, only a few nanomaterials with multiple enzyme-like activity have been reported, and most of the currently developed nanozymes are usually used in colorimetric or fluorescence analysis depending on a single colorimetric or fluorescence signal output. In this study, a copper-based dual-ligand biomimetic nanoflower (Cu-MN) was constructed, which demonstrated potential multiple enzyme-like activity, and was applied to the multi-mode detection of 2,4-dichlorophenol (2,4-DP), epinephrine (EP), and H 2 O 2 ., Results: The laccase-like activity of Cu-MN can catalyze the conversion of 2,4-DP and EP, resulting in the formation of red and yellow-brown oxidation products with distinct UV absorption peaks at 510 nm and 485 nm, respectively. Furthermore, the fluorescence emission peak at 426 nm of Cu-MN can be dynamic quenched during substrate oxidation due to the fluorescence internal filtration effect (IFE). Therefore, a dual-mode analysis method was constructed to detect 2,4-DP and EP by fluorescence and ultraviolet colorimetry, which was successfully applied in natural lake water and rabbit plasma analysis, respectively. Furthermore, a colorimetric sensing strategy based on the peroxidase-like activity of Cu-MN was developed and successfully applied to the monitoring of H 2 O 2 in hydrogen peroxide disinfectant. Considering the high incidence of breast cancer, a sensitive and specific approach is crucial for its early diagnosis and follow-up treatment. Folate receptors (FR), which are highly expressed on the epithelial tissue such as breast cancer cells (e.g., MCF-7), have been used in cancer diagnosis and bioimaging. This study presents an innovative colorimetric and fluorescence bioimaging platform towards MCF-7 using folic acid (FA)-conjugated iron-oxide magnetite silica-based nanocomposite (Fe 3 O 4 @SiO 2 -3-aminopropyl)triethoxysilane (APTES-NH 2 )@cysteine (Cyt)-Cyt@FA). For identification of MCF-7, the polyvinylpyrrolidone (PVP)-capped-platinum (Pt) nanoparticle was utilized as a nanozyme to catalyze the reaction between 3,3',5,5'-tetramethylbenzidine (TMB) and H 2 O 2 for visual detection of MCF-7 cells. Colorimetric changes are detectable by the naked eye and spectrophotometry at the wavelength of 450 nm, with a linear range of 50-5000 cells/mL and a detection limit of 30 cells/mL. The Fe 3 O 4 @SiO 2 -APTES-NH 2 @Cyt-Cyt@FA complex was modified with rhodamine B as a fluorescence bioimaging probe to monitor FR-overexpressed MCF-7 cells. The nanocomposite is biocompatible with a toxicity threshold of about 800 µg/mL. Achieving rapid extraction and detection of analytical solutions from plant tissues to circumvent cumbersome procedures and reduce the dependence on detection instruments remains a challenge. Herein, a colorimetric microneedle integrated platform was developed for the rapid extraction and detection of plant fluids for testing purposes. Colorimetric microneedle patches (CMPs) offer a swift and effective method to swiftly extract and detect ascorbic acid (AA) within 10 min from various fruit crops like mango, nectarine, apple, pear, and kiwifruit, facilitated by a smartphone application. CMPs are constructed for the rapid and sensitive analysis of AA with good linearity amid the range 0.05-25 μM and a low limit of detection of 30 nM. The novel CMPs demonstrate significant potential as a rapid detection platform for AA in plants. CMPs offer significant advantages over traditional ultraviolet spectrophotometry, such as simplified operational procedures and accelerated extraction and detection processes. This establishes robust groundwork for conducting in situ extraction and molecular detection of diverse crops across a spectrum of application scenarios.

A light-responsive covalent-organic framework (COF) nanozyme, which integrates the advantages of the COF structure and light-stimulated nanozyme catalysis, is a class of sensing star materials with wide application prospects. However, the sensing methods based on light-responsive COF nanozymes are relatively single at present. Therefore, it is necessary to develop new sensing strategies to broaden its application in chemical sensing and achieve highly efficient detection. Here, a Cu 2+ -modified COF composite material (TpDA-Cu) was rationally designed. The addition of Cu significantly inhibits the excellent light-responsive nanozyme activity of TpDA itself. However, because of the restoration of the enzyme activity by thiram (Tr) and the oxidase mimic activity of the newly formed Cu/Tr complex, TpDA-Cu/Tr exhibits stronger light-responsive nanozyme activity. Enzyme kinetic data show that compared with TpDA, TpDA-Cu/Tr has a larger V max value, which can achieve efficient catalytic oxidation of 3,3',5,5'-tetramethylbenzidine (TMB). In addition, the strong coordination effect of Tr and TpDA-Cu also plays a key role in achieving ultrafast, sensitive, and selective colorimetric detection of Tr. This work develops a dual activity regulation strategy of light-responsive COF nanozymes based on analyte induction and provides a new perspective for the application of light-responsive COF nanozymes in the field of sensing.

Pesticides are primarily used in agriculture to protect crops and extend their longevity. However, pesticide exposure has been linked to various acute and chronic health effects, raising significant environmental concerns. Current detection methods are often expensive and time-consuming, relying on complex instruments. Although enzyme-inhibition-based microfluidic paper-based analytical device (mPAD) platforms offer an easier alternative, they suffer from slow analyte transport and analyte adsorption issues in microchannels. Consequently, there is a need for a fast, simple, and cost-effective point-of-need platform for pesticide sensing. In this study, we present a rapid microfluidic platform for on-site pesticide residue detection. Unlike traditional mPAD platforms, our system transports pesticide samples through hollow capillary channels within seconds without adsorption of pesticides in the microchannels. While much research has focused on acetylcholinesterase inhibition on paper, this study is the first to introduce a tyrosinase inhibition-based assay on a paper platform for pesticide detection. Ziram, a representative dithiocarbamate pesticide, was detected using a colorimetric enzymatic inhibition assay. A limit of detection (LoD) of 1.5 ppm was obtained. In this study, we optimized the fast-flow device, assessed its stability and susceptibility to various interferences, and conducted real-sample tests using glove extraction to evaluate its capability in real-world settings. Spike recovery analysis revealed an extraction efficiency of 82.5% to 87.5% for leather gloves and 68.9% to 71.9% for nitrile gloves. This platform demonstrates strong selectivity against interferences, with the enzyme retaining 90% activity even after a week under the established storage protocols with room for further investigation. While primarily a proof of concept, this device shows promise as an additional tool for pesticide detection, with potential future integration into multiplexed devices.

LaNiO 3 perovskite nanoparticles, especially nanospheres (LNNS), show great promise in biomedical assays due to their peroxidase-like catalytic properties. However, LNNS-based diagnostic reagents have not been tested in nanozyme enzyme-linked immunosorbent assay (NLISA) or other enzyme-linked immunosorbent assays, and there is limited data on their synthesis. To fill this gap, it is necessary to develop a method for creating LNNS conjugates with monoclonal antibodies and to investigate the reproducibility, scalability, and applicability of these diagnostic reagents in NLISA. We have successfully developed a method for producing novel diagnostic reagents utilizing LaNiO 3 nanospheres. Our research demonstrates the application of these nanospheres in a NLISA specifically designed for the detection of C-reactive protein (CRP) in real serum samples. This method is both reproducible and scalable, allowing for the efficient production of nanospheres that are functionalized with monoclonal antibodies targeting CRP, with a mean diameter of approximately 270 nm. Based on the promising results obtained from our experiments, we have developed and optimized a sandwich-format NLISA for CRP detection. This assay achieved a lower limit of detection at 0.178 μg L -1 , with a dynamic range from 12.5 to 0.195 μg L -1 and a linear detection range extending from 0.195 to 6.25 μg L -1 , showcasing its potential for clinical applications. The new NLISA method, utilizing LaNiO 3 nanospheres in a sandwich format for the detection of CRP, significantly enhances sensitivity compared to similar use horseradish peroxidase-based ELISA. In this study for the first time, the functionalization of lanthanum nickelate nanospheres with recognition elements has been demonstrated. This advancement also sheds light on the technological challenges involved in synthesizing diagnostic reagents, identifying areas that need further exploration.

Many aptamers have been generated by systematic evolution of ligands by exponential enrichment (SELEX) to recognize spike proteins of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2&ek), some of which have been engineered into dimeric and trimeric versions for enhanced affinity for diagnostic applications. However, no studies have been conducted to compare the utilities of monomeric, dimeric and trimeric aptamers in diagnostic assays with real clinical samples to answer the question of what levels of affinity an aptamer must have for accurate clinical diagnostics. Herein, we carried out a comparative study with two monomeric aptamers MSA1 and MSA5, one dimeric aptamer and two homotrimeric aptamers constructed with MSA1 and MSA5, with affinity varying by 1000-fold. Using a colorimetric sandwich assay to analyze 48 human saliva samples, we found that the trimeric aptamer assay (K d ≈10 pM) can identify the SARS-CoV-2 infection much more accurately than the dimeric aptamer assay (K d ≈100 pM) and monomeric aptamer assay (K d ≈10,000 pM). Based on the experimental data, we theoretically predict the levels of affinity an aptamer needs to possess to achieve 80-100 % sensitivity and 100 % specificity. Detecting glucose levels is crucial for diabetes patients as it enables timely and effective management, preventing complications and promoting overall health. In this endeavor, we have designed a novel, affordable point-of-care diagnostic device utilizing microfluidic principles, a smartphone camera, and established laboratory colorimetric methods for accurate glucose estimation. Our proposed microfluidic device comprises layers of adhesive poly-vinyl films stacked on a poly methyl methacrylate (PMMA) base sheet, with micro-channel contours precision-cut using a cutting printer. Employing the gold standard glucose-oxidase/peroxidase reaction on this microfluidic platform, we achieve enzymatic glucose determination. The resulting colored complex, formed by phenol and 4-aminoantipyrine in the presence of hydrogen peroxide generated during glucose oxidation, is captured at various glucose concentrations using a smartphone camera. Raw images are processed and utilized as input data for a 2-D convolutional neural network (CNN) deep learning classifier, demonstrating an impressive 95% overall accuracy against new images. The glucose predictions done by CNN are compared with ISO 15197:2013/2015 gold standard norms. Furthermore, the classifier exhibits outstanding precision, recall, and F1 score of 94%, 93%, and 93%, respectively, as validated through our study, showcasing its exceptional predictive capability. Next, a user-friendly smartphone application named "GLUCOLENS AI" was developed to capture images, perform image processing, and communicate with cloud server containing the CNN classifier. Background: Aptasensors have been extensively utilized in target detection due to their advantages of high sensitivity and fast response. However, the reliability of the detection results of the single-mode aptasensor cannot be verified in time. Developing efficient detection methods with cross-validation capability is beneficial to achieving highly reliable detection. This study aims to design a colorimetric and fluorescent dual-mode aptasensor by skillfully engineering G-quadruplex assembly and rolling circle amplification for highly reliable IFN-γ detection., Results: The complexes of anti-IFN-γ aptamers and complement sequences (cDNA) were modified on the magnetic beads. In the presence of IFN-γ, the preferential combination of aptamers with IFN-γ resulted in the release of cDNAs. The cDNAs were collected by magnetic separation and then used as primers to trigger rolling circle amplification reaction to generate enriched G-quadruplexes. The G-quadruplex could be utilized to combine with hemin to catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine for colormetric mode or to couple with the fluorogenic dye Thioflavin T for fluorescent mode. The developed dual-mode aptasensor displayed a linear range of 1-10000 pM with a detection limit of 0.406 pM for the colormetric mode and a range of 0.1-10000 pM with a detection limit of 0.037 pM for the fluorescent mode. Background: Heavy metal pollution poses a serious threat to the ecological environment. Mercury ion (Hg 2+ ) is a class of highly toxic heavy metal ions, which is bioaccumulative, difficult to breakdown, and has a significant affinity with sulfur and thiol-containing proteins, which seriously affects environmental safety and human health. Nanozyme-based sensing methods are expected to be used to detect toxic heavy metal ions. However, the application of precious metal nanozymes to develop portable sensors with simplicity, high stability, and high sensitivity has not been explored to a large extent., Results: In this paper, based on MXene's unique adsorption capacity for certain precious metal ions, PtNPs/Ti 3 C 2 T X NR composites were successfully prepared by in-situ growth of Pt nanoparticles (PtNPs) on the surface of Ti 3 C 2 T X MXene nanoribbons (Ti 3 C 2 T X NR) using the hydrothermal technique. Experimental data revealed PtNPs/Ti 3 C 2 T X NR exhibited superior peroxidase-like activity, attributed to the synergistic effect of well-dispersed ultrasmall PtNPs and electron transfer effect. Hg 2+ can significantly inhibit enzyme-like activity of PtNPs/Ti 3 C 2 T X NR due to specific capture and partial in-situ reduction of PtNPs, so a colorimetric sensor was constructed for ultra-trace detection of Hg 2+ with a linear range of 0.2 nM and 400 nM. Furthermore, using the portable detecting capabilities of smartphones and hydrogel, a smartphone-assisted hydrogel sensing platform of Hg 2+ was constructed. Notably, the two-mode sensing platforms exhibited outstanding detection performance with LOD values as low as 15 pM (colorimetric) and 26 pM (hydrogel), respectively, superior to recently reported nanozyme-based Hg 2+ sensors., Significance: Compared with other methods, the PtNPs/Ti 3 C 2 T X NR-based dual-mode sensor designed in this paper has superior sensitivity, high selectivity, simple operation and portability. Background: Sildenafil (SIL) is regarded as an illegal adulterant in functional foods. Some functional foods doped with SIL have posed significant concern about their safety risks. However, the facile colorimetric detection of SIL is rarely investigated., Results: Herein, we prepared a monodispersed spherical composite nanozyme (Fe 3 O 4 -NH 2 /GONRs), possessing excellent peroxidase-like (POD-like) and catalase-like (CAT-like) activities and strong superparamagnetic property. The enzyme-like activities of Fe 3 O 4 -NH 2 /GONRs can be selectively inhibited by SIL due to the synergistic effect of hydrogen bonds and π-π stacking between Fe 3 O 4 -NH 2 /GONRs and SIL. Leveraging this mechanism, a highly sensitive and selective colorimetric detection for SIL with a detection limit (LOD) of 0.26 ng/mL was developed. Chlorine is a common disinfectant used in water treatment. However, its reaction with organic matter can lead to the formation of harmful byproducts, such as trihalomethanes (THMs), which are potentially carcinogenic. To address this issue, the aim of this work was to enhance a colorimetric method capable of quantifying THMs in drinking water through UV/Vis Spectrophotometry, using cost-effective equipment, and validate this methodology for the first time according to established validation protocols. The method's innovation involved replacing the solvent pentane with the more common hexane, along with adjusting the heating ramp, elucidating the mechanisms involved in the process. This method involves the reaction between THMs, pyridine, and NaOH to produce a colored compound, which is then monitored through molecular absorption spectroscopy in the visible region. The method was thoroughly validated, achieving a limit of detection of 13.41 μg L -1 and a limit of quantification of 40.65 μg L -1 . Recovery assays ranged from 86.1 % to 90.7 %, demonstrating high accuracy. The quality of the linear fit for the analytical curve exceeded R 2  > 0.98. The method was applied to real samples, revealing concentrations ranging from 13.58 to 55.46 μg L -1 , all way below the legal limit in Brazil (Maximum Contaminant Levels (MCL) = 100 μg L -1 ). Copper oxide nanosheets (CuO NSs) have been successfully obtained by exploiting an effective one-step approach of sugar-blowing method followed by calcination. The nanosheets were characterized by several techniques like X-ray powder diffraction (XRD), Transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS) and Fourier transform infrared spectroscopy (FTIR). Impressively, CuO NSs display haloperoxidase (HPO) like catalytic activity which catalyses the oxidation of chloride ions by H 2 O 2 giving rise to reactive chlorine species (RCS). A sensitive and selective colorimetric sensor was then demonstrated via the oxidation of chromogenic substrate 3,3',5,5'- tetramethylbenzidine (TMB) by the novel nanoenzyme CuO NSs through the generation of RCS for H 2 O 2 and glucose detection with limit of detection of 109 nM and 21 nM in the linear ranges of 4.6 µM to 769 µM and 0.22 µM to 19.57 µM respectively. Mineral water is a natural water that originated from an underground water table, a well, or a natural spring which is considered microbiologically intact. The revenue from the bottled mineral water industry will be USD 342.40 billion in 2023, and it is expected to grow at a compound annual growth rate (CAGR) of 5.24 %. Consequently, the discrimination of original bottled mineral water from tap water is an important issue that requires designing sensors for simple and portable identification of these two types of water. In this work, we have developed a Dip-Type colorimetric paper-based sensor array with three organic dyes (Bromothymol Blue, Bromophenol Blue, and Methyl Red) followed by chemometrics' pattern recognition methods (PCA and LDA) for discrimination of original bottled mineral waters from tap waters based on differences in ion variety and ion quantity. Forty brands of mineral water and twenty-six Tap water samples from different regions of Shiraz and other Iranian cities were analyzed by this sensor array. Moreover, these experiments were performed in two consecutive years to check the versatility of the sensor with seasonal changes in waters. Rapid and accurate diagnosis of tuberculosis (TB) is of great significance to control the spread of this devastating infectious disease. In this work, a sensitive and low-cost point-of-care testing (POCT) detection platform for TB was developed based on recombinase polymerase amplification (RPA)-catalytic hairpin assembly (CHA)-assisted dual signal amplification strategy. This platform could achieve homogeneous fluorescent and visual diagnosis of TB by using CdTe quantum dots (QDs) signal reporter. In the presence of target DNA (IS1081 gene fragment), RPA amplicons blocked by short oligonucleotide strands could trigger CHA signal amplification, leading to the Ag + releasing from C-Ag + -C structure and the fluorescence quenching of CdTe QDs by the released Ag + . Furthermore, the detection performance of CdTe QDs modified by 3-mercaptopropionic acid (MPA) or thiomalic acid (TMA) (MPA-capped QDs and TMA-capped QDs) was systematically compared. Experimental results demonstrated that TMA-capped QDs exhibited better detection sensitivity due to their stronger interaction with Ag + . The limits of detection (LODs) of fluorescence and visual analysis were as low as 0.13 amol L -1 and 0.33 amol L -1 . Acetaminophen, also known as paracetamol (APAP), is a commonly used over-the-counter medication that is often used to treat headaches, toothaches, joint pain, muscle pain, and to lower body temperature. However, overdose can lead to liver damage, gastrointestinal distress, kidney damage, and cardiovascular disease. Therefore, it is very important to establish a method to quickly detect APAP. A novel "ON-OFF-ON" colorimetric and fluorescence dual-signal sensing system was constructed for the quantitative detection of APAP based on 5,10,15,20-tetrakis(4-sulphonatophenyl) porphyrin (TSPP) dual-signal probe. The absorbance and fluorescence intensity of TSPP respectively were quenched when Fe 3+ was introduced into TSPP solution. At this point, the color of the corresponding solution changed from red to green. The absorbance and fluorescence intensity of TSPP respectively were restored when APAP was added to the TSPP-Fe 3+ system. At this time, the color of the solution changed from green to colorless. Therefore, an "ON-OFF-ON" dual-signal sensing study of APAP were constructed using TSPP as the colorimetric and fluorescent probe. The proposed colorimetric sensing system had a wide linear range in the 13.12 mM ∼ 23.20 mM with 0.11 mM of limit of detection (LOD, S/N = 3). And the proposed fluorescence sensing system had a wide linear range in the 3.45 mM ∼ 12.50 mM and 41.67 mM ∼ 65.22 mM with 0.83 mM of limit of detection (LOD, S/N = 3). Unsafe food additives pose a significant threat to global health, especially in developing countries. Many existing methods rely on clean laboratories, complicated optics equipment, trained personnel and lengthy detection time, which are not suitable for onsite food safety inspections in emergency situations, peculiarly in impoverished areas. In this paper, a fast and visual onsite method is designed for the detection and quantification of additives in food safety by engineering a nanohybrid (MoS 2 /SDBS/Cu-CuFe 2 O 4 )-based catalysis. Interestingly, the nanohybrid presents peroxidase-like mimetic activity toward the substrate containing 3,3',5,5'-tetramethylbenzidine (TMB) and hydrogen peroxide (H 2 O 2 ), which are then integrated simply into a detection kit. The blue oxidated TMB in this kit can be converted completely to colorless by some bio-molecule additives in commercial food, such as glutathione (GSH), cysteine (Cys), and ascorbic acid (AA). Remarkably, this process takes just less than 2 min and the detection limits are 2.8 nM, 5.5 nM and 47 nM, respectively. These results show excellent repeatability with a statistical analysis with (*P < 0.05) over 30 tests. Next, the images of the color changes can be captured clearly using a smartphone by red-green-blue (RGB) channels, which provides an opportunity for the development of field-operation device. Additionally, our approach is applied to some targets-indicative foods, showing a recovery range between 95.8 % and 104.2 %, offering an attractive and promising pathway for future practical food safety inspection applications. Due to increasing food safety issues, developing intelligent, on-site, and visual methods for detecting fish freshness has attracted significant attention. Here, we have prepared a benzo[h]chromene derivative BCN that can visually detect 12 biogenic amines (BAs) with high sensitivity. The mechanism for recognizing cadaverine (Cad) is that the probe reacts with Cad to produce a Schiff base derivative, which alters the charge distribution within the molecule, resulting in significant colorimetric and fluorescence changes. The sensing label BCN/FPS was prepared by loading the probe BCN on filter paper, and a visual detection platform was constructed by combining it with a smartphone. By monitoring the correspondence between label color and TVB-N content, a working curve of (R + B)/(R + B + G) with TVB-N content was obtained, enabling visual evaluation of salmon freshness using only a mobile phone. In addition, based on the good solubility and processability of BCN, its application in fluorescent dyes including impregnating dyes, printing inks, coatings, and flexible films has been explored, which opens up new directions for the application of BCN. Neomycin sulfate (NEO) is a kind of aminoglycoside antibiotics. Because of its strong ototoxicity, nephrotoxicity and other side effects, its content in the body should be strictly monitored during use. In this paper, a rapid colorimetric detection method for NEO based on ultrasmall polyvinylpyrrolidone modified gold nanoparticles (PVP/Au NPs) with peroxidase-like activity was developed. Firstly, ultra small PVP/Au NPs with weak peroxidase-like activity were synthetized. When they were mixed with NEO, strong hydrogen bonds were formed between NEO and PVP, resulting in the aggregation of PVP/Au NPs, and the aggregated PVP/Au NPs showed stronger peroxidase-like activity. Therefore, rapid colorimetric detection of NEO was achieved by utilizing the enhanced peroxidase-like activity mechanism caused by the aggregation of ultra small PVP/Au NPs. The naked eye detection limit of this method is 50 nM. Within the range of 1 nM-300 nM, there was a good linear relationship between NEO concentration and the change in absorbance intensity of PVP/Au NPs-H 2 O 2 -TMB solution at 652 nm, with the regression curve of y = 0.0045x + 0.0525 (R 2  = 0.998), and the detection limit is 1 nM. In addition, this method was successfully applied to the detection of NEO in mouse serum. The accurate and sensitive detection of metallothionein (MT) is of great significance in the fields of biomedical, toxicological and environmental sciences. In this work, based on the high affinity interaction between MT and the heavy metal ions of Hg 2+ and the significant signal amplification capability of Cas12a/crRNA enzyme as well, we report a simple and highly sensitive method for visual detection of MT, a biomarker in fish for heavy metal ion-induced water bio-pollution. The target MT molecules bind Hg 2+ in the Hg 2+ - complexed hairpin DNA probes to unfold the hairpin structure into ssDNAs, which hybridize with the partial dsDNA duplexes via strand displacement to yield specific sequence-containing dsDNAs. Cas12a/crRNA recognizes these specific sequences to activate its enzyme activity to cyclically cleave the ssDNA linkers in the blue colored gold nanoparticle aggregates to transit their color into red to realize visual detection of MT. Owing to the signal amplification by Cas12a/crRNA, as low as 25 nM of MT can be visually detected with naked eye. In addition, our colorimetric detection method has high selectivity for MT against other interference proteins and can detect MT in the livers and kidneys of crucian carps bought from a local supermarket. An Fe-based metal-organic framework (Fe-MOF) nanozyme with MIL-88B(Fe) structure was facilely prepared by an ultrasonic method. Its oxidase-like activity could be specifically inhibited by methidathion, which conformed to a reversible uncompetitive inhibition mechanism. Furthermore, a specific and enzyme-free colorimetric assay of methidathion was established using Fe-MOF as the ideal recognition element.

Creatinine, an important biomarker for renal functions, is often conventionally estimated using the gold standard Jaffe reaction from blood, which involves the usage of a spectrophotometric measurement, thus restricting its utilization scope in point-of-care settings. Here, we report the development of a method for the single-step quantification of creatinine from whole blood using a paper-based microfluidic device. Our platform uses Whatman filter paper integrated with an LF1 membrane. The on-chip separation of blood plasma is achieved through the LF1 membrane, while the Whatman component of the device contains the embedded reagents for the Jaffe reaction. The combination of two different grades of paper enables a single-step quantification of creatinine as the separated blood plasma traverses to the reaction zone through capillary imbibition. Colorimetric readouts were quantified using an RGB sensor instead of a smartphone, which is highly platform dependent and incurs a relatively higher cost compared to the other components in typical point-of care (POC) devices. Our sensor was integrated within a 3D box, thereby making the detection virtually instrument free and perfectly suited for POC settings. The limit of detection (LOD) of our device was 0.219 mg dL -1 , which falls within the lower range of physiological values. The coefficient of determination ( R 2 ) for the linearity and median accuracy were 0.978 and 94.047%, respectively. The relative standard deviation (RSD) for precision measurements remained below 5% for the developed protocol. Furthermore, we validated the performance of our device with 35 clinical samples in laboratory settings against the gold standard measurements. Our Bland-Altman plot as well as t -test and chi-square test results clearly confirmed the validity of our device within a 95% confidence interval.

The rapid classification of crude baijiu is pivotal for its industrialization and automated development. In this study, a colorimetric sensor array employing peroxidase nanase (Por-CoMoO 4 ) was developed to detect reducing substances and crude baijiu. The peroxidase-like activity of CoMoO 4 was significantly enhanced by porphyrin (Por), exhibiting a K m value of 0.044 mM and V max of 19.37 × 10 -8 for TMB substrate. Peroxidase activity varies at different pH levels. Organic and crude baijiu scavenge free radicals, thereby inhibiting oxTMB formation and yielding distinctive fingerprint profiles. Using linear discriminant analysis, 14 types of small molecules and 16 varieties of Luzhou-flavor crude baijiu were identified within specific concentration ranges. The method achieved 100% accuracy in distinguishing baijiu samples sourced from different distilleries, offering a straightforward, rapid, and effective approach to differentiate crude baijiu during alcoholic beverage production.

Cr(VI) is one of the most typical heavy metal contaminants and rapid detection of Cr(VI) is highly important in food control and public health. Herein, a core-shell Au@Pt nanozyme-based colorimetric assay was developed for the rapid and sensitive detection of Cr(VI). The monodispersed Au@Pt core-shell nanoparticles exhibited high peroxidase-mimicking activity and can catalyze colorless TMB into blue-colored oxidized oxTMB. After the addition of Cr(VI), the oxTMB molecules can be reduced into colorless TMB. The ultrathin Pt shell can prevent the Pt component from aggregation, thus improving the catalytic activity of Au@Pt nanozyme. These Au@Pt nanozyme-based Cr(VI) assays exhibited high sensitivity and selectivity and displayed satisfactory recoveries in practical samples. Our work highlights opportunities for the development of core-shell nanozymes with extensive applications in food safety, biomedicine, and environmental monitoring.

Background: Brucellosis is a major worldwide zoonotic disease that is caused by Brucella spp. and threatens the health of communities. Novel methods for rapid detection of Brucella bacteria are beneficial and necessary in preventing infection and subsequent economic losses. Constructing biosensors with nanoparticles is a promising approach for identification of pathogenic bacteria in a short time. This study aimed to introduce a new detection method of Brucella cells using a biosensor, based on gold nanoparticles and a specific aptamer, via a colorimetric reaction. In this work, gold nanoparticles (GNPs) were synthesized and attached to the aptamer through electrostatic bonding. The binding of aptamer to gold nanoparticles was confirmed by Uv/vis spectrophotometry, FT-IR, transmission electron microscope (TEM) and zeta sizer (DLS)., Results: In the presence of the bacterial cells, aptamers were bound to their targets, and the surfaces of the nanoparticles were depleted from aptamers resulting in intensified peroxidation activity of GNPs, and with the addition of 3, 3', 5, 5'-tetramethylbenzidine (TMB), the color of the solution was changed from red to purple, which indicated the presence of Brucella. The sensitivity of the aptasensor was investigated using different concentrations of Brucella cells and its specificity was confirmed against several species of bacteria. Listeria monocytogenes is a major foodborne pathogen affecting developing, and developed countries. The analysis of food contact surfaces in food industries is key for better controlling this pathogen. The current study focused on the development, optimization, and evaluation of a rapid and simple method for the detection of L. monocytogenes on stainless steel surfaces, suitable for decentralized setups, taking advantage of Loop-mediated isothermal amplification (LAMP). This was accomplished using a general pre-enrichment broth (TSB), with a simple DNA extraction based on a chelating resin, and final isothermal amplification. Two different detection strategies were tested, real-time fluorescence and naked-eye colorimetric, which were evaluated after 5, 7, and 24 h of pre-enrichment. Regardless the detection chemistry selected, after 5-7 h of pre-enrichment, 10 3 -10 4  CFU/cm 2 were needed to obtain a positive result, while after 24 h, it was possible to detect concentrations below 10 CFU/cm 2 . Within each given time, all the performance parameters calculated, relative sensitivity, specificity, and accuracy, reached values higher than 80-90%; likewise, a Cohen's k of concordance with a culture-based approach higher than 0.8. Overall, the most sensitive assay can be performed in roughly 25 h. The liquid-liquid interface offers a confined space to control the growth of nanomaterials. In this study, Fe(II) (water phase) induced Meso-tetra (4-carboxyphenyl) porphyrin (H 2 TCPP) (CHCl 3 , organic phase) into nanoaggregates (Fe-TCPP) in the liquid-liquid interface. By tuning the ratio of DMF in organic solvents, Fe(II) induced H 2 TCPP into two nanoaggregates (Fe-TCPP-1 and Fe-TCPP-2) with different morphologies via coordination interaction occurring at the water-CHCl 3 interface. Interestingly, the Fe-TCPP nanoaggregates possess dual enzyme-like activity (peroxidase-like and oxidase-like activity). In particular, both Fe-TCPP-1 and Fe-TCPP-2 demonstrate a peroxidase-/oxidase-like activity under visible light irradiation that is higher than that in the dark. Comparatively, Fe-TCPP-2 exhibits enhanced peroxide-like (POD) activity together with oxidase-like (OXD) activity compared with that of Fe-TCPP-1 under the corresponding similar conditions. The excellent enzyme mimic activity of Fe-TCPP nanozymes is ascribed to the generated hydroxyl radicals (·OH) and superoxide anions (O 2 •- ). Remarkably, the catalytic activity of Fe-TCPP-2 remains more than 90% even in the higher temperature range of 35-40 °C, which is significant for biological detection under physiological conditions. Based on the outstanding dual enzyme-like activity of Fe-TCPP-2, a colorimetric sensing platform for methimazole (an antithyroid medicine) has been developed, demonstrating a linear detection range of 10-100 μM and a detection limit of 4.44 μM.

A benzothiazole-azo based sensor (BTAN) was developed for rapid and on-site detection of arginine. The sensor's selectivity in a semi-aqueous medium was thoroughly investigated, focusing on the colorimetric response to arginine in the presence of 11 different amino acids. Notably, the limit of detection (LOD) for arginine was determined to be 0.7 μM. The underlying sensing mechanism was addressed using 1 H-NMR and UV-vis spectroscopy. BTAN exhibited significant changes in both absorption as well as emission spectra exclusively in the presence of arginine. Furthermore, the arginine sensing capability was extended to the solid state by immobilizing BTAN into a starch-PVA hydrogel matrix as well as paper strips. The hydrogel film of BTAN enabled effective on-site sensing of arginine in a 100% aqueous medium. Moreover, the practicability of the sensor was demonstrated by detecting arginine in human blood samples.

Dual-mode-based sensors have drawn a lot of interest due to their high accuracy and sensitivity compared to single-response systems. A simple electrochemical and colorimetric dual-mode sensor based on enzyme-linked immunosorbent assay (ELISA), without complex electrode surface modification, was developed for accurate and sensitive detection of carcinoembryonic antigen (CEA). The target CEA is recognized by an antibody coupled to horseradish peroxidase (HRP), which then oxidizes the substrate 3,3',5,5'-tetramethylbenzidine (TMB) to generate both a colorimetric and an electrochemical signal. A paper-based analysis device (μPAD) with dual-mode homogeneous sensing microfluidic was created; three paper-based detection areas for colorimetric testing, and a two-electrode embedded detection area for electrochemical testing. When applying colorimetric analysis technology, the linear range of CEA detection is 0.6-40 ng mL -1 , and the limit of detection (LOD) is 0.2 ng mL -1 . The linear range is 0.1-40 ng mL -1 and the LOD is 0.03 ng mL -1 by applying electrochemical analysis. The visibility and intuitiveness of colorimetry provide a reference for higher sensitivity and quick response of the electrochemical method. A smartphone application (APP) was also developed to realize the dual extraction of colorimetric signals. The colorimetric detection system based on ELISA can provide a new path for the development of electrochemical sensing and makes it have inherent self-verification and self-correction functions and is expected to provide more reliable and accurate detection results.

Single-atom nanozymes (SAzymes) as a class of highly active nanozymes with the advantages of high atom utilization, high catalytic activity and stability have attracted great attention. In this work, Fe-N-C SAzymes with exceptional oxidase (OXD)-like activity were achieved utilizing polyvinylpyrrolidone (PVP) as a template. The Fe-N-C SAzymes with remarkable OXD-like activity could oxidize TMB to blue oxTMB, but 8-hydroxyquinoline (8-HQ) as a metal chelator is capable of discoloring oxTMB. Thus, the addition of 8-HQ decolorized the solution. However, upon the introduction of Cr(VI) ions, 8-HQ preferentially chelated with the Cr(VI) ions, reversing the inhibition of the color reaction and restoring the blue color. Based on this phenomenon, we constructed a novel paper-based analytical device (PAD) that exhibited a linear range of 5-1000 μM and an LOD of 1.2 μM. Importantly, the PAD used in this study shows the merits of simplicity, low preparation costs, and rapid reaction times. When combined with smartphone RGB analysis, it enables the simultaneous analysis of eight different Cr(VI) concentrations without the need for large-scale instrumentation. Moreover, the proposed PAD displays high selectivity, accuracy and utility in testing actual short-necked clam samples. This work not only provides a simple and cost-effective method to detect Cr(VI) but also makes a contribution to rapid food testing.

In this work, a mitochondria-targeted NIR ratiometric and colorimetric fluorescent probe 1 was tactfully designed and synthesized by a novel design strategy of modifying chromene to pyridine for the first time. 1 exhibited a maximum absorption peak at 508 nm and a maximum fluorescence emission peak at 650 nm. Under the stimulus of biothiols (cysteine (Cys), homocysteine (Hcy), and glutathione (GSH)), the maximum absorption and fluorescence emission peaks of 1 blue-shifted to 448 and 541 nm, respectively, along with color changes from red to yellow under visible light and from red to green under a 365 nm ultraviolet (UV) lamp, which can be ascribed to the click reaction of biothiols with the α,β-unsaturated ketone of the chromene moiety with pyran ring-opening, phenol formation, and 1,6-elimination of the p -hydroxybenzyl moiety. 1 detected biothiols (Cys, GSH, and Hcy) with high sensitivity (LODs of 29, 23, and 16 nM for Cys, GSH, and Hcy, respectively), excellent selectivity, and fast response. Moreover, 1 can target mitochondria and image the fluctuation of intracellular biothiols by dual-emission channels.

Protein-nanoparticle interactions play a crucial role in both biomedical applications and the biosafety assessment of nanomaterials. Here, we found that nanobodies can induce citrate-capped gold nanoparticles (AuNPs) to aggregate into large clusters. Subsequently, we explored the mechanism behind this aggregation and proposed the "gold nucleation mechanism" to explain this phenomenon. Building on this observation, we developed a one-step label-free colorimetric method based on nanobody-induced AuNP aggregation. When nanobodies bind to target bacteria, spatial hindrance occurs, preventing further AuNPs aggregation. This alteration in surface plasmon resonance properties results in visible color changes. As an example, we present a simple and sensitive "mix-and-read" chromogenic immunosensor for Cronobacter sakazakii ( C. sakazakii ). The experiment can be completed within 20 min, with a visual detection limit of 10 3 CFU/mL and a quantitative detection limit of 136 CFU/mL. Importantly, our method exhibits no cross-reactivity with other bacterial species. This strategy harnesses the excellent properties of nanobodies and the optical characteristics of AuNPs for direct and rapid detection of foodborne pathogen.

Mercury ion (Hg 2+ ) is considered a harmful neurotoxin, and real-time monitoring of Hg 2+ concentrations in environmental and biological samples is critical. Fluorescent probes are a rapidly emerging visualization tool owing to their simple design and good selectivity. Herein, a novel fluorescence (FL) probe 2-(4-((6-((quinolin-8-yloxy)methyl)pyridin-2-yl)methyl)piperazin-1-yl)anthracene-9,10-dione (QPPA) is designed using piperazine as a linker between the anthraquinone group, which serves as a fluorophore, and N 4 O as the Hg 2+ ligand. The probe exhibits FL "turn-on" sensing of Hg 2+ because the complex inhibits the photo-induced electron transfer (PET) process. Moreover, QPPA can overcome the invasion by other possible cations, resulting in a clear color change from orange to colorless with the addition Hg 2+ . The chelation of QPPA with Hg 2+ in a 1:1 ratio. Subsequently, the theoretically determined binding sites of the ligand to Hg 2+ are validated through 1 H NMR titration. The in situQPPA-Hg 2+ complex can be subjected to Hg 2+ extraction following the introduction of S 2- owing to its robust binding capacity. The exceptional limit of detection values for Hg 2+ and S 2- are obtained as 63.0 and 79.1 nM (S/N = 3), respectively. As a crucial endogenous reactive oxygen species, hypochlorous acid (HClO) plays an indispensable role in numerous physiological and pathological processes. Additionally, it serves as a biomarker closely associated with inflammation and liver injury. The utilization of near-infrared fluorescence probes has surged in recent years for live biological imaging, owing to their minimal tissue damage and potent tissue penetration capabilities. In this work, a novel near-infrared fluorescence probe MB-HPD was synthesized to sensitively detect HClO. Probe MB-HPD exhibits remarkable selectivity, high sensitivity (14.3 nM), and rapid response towards HClO (20 s). Probe MB-HPD has demonstrated successful application in the imaging of HClO within cells and zebrafish. Remarkably, it has proven to be effective for detecting HClO within environmental samples, as well as imaging HClO in mice models of arthritis and APAP-induced liver injury. In this article, we present the first demonstration of a distance-based paper analytical device (dPAD) for uric acid quantification in human urine samples with instrument-free readout and user-friendliness for the rapid diagnosis and prognosis of various related diseases. By employing gold nanoparticles (AuNPs) as a peroxidase-like nanozyme, our proposed technique eliminates the utilization of horseradish peroxidase (HRP), making the device cost-effective and stable. In our dPAD, uric acid in the sample is oxidized by the uricase enzyme and subsequently catalysed with AuNPs in the sample zone, generating hydroxyl radicals (˙OH). Then, the produced ˙OH reacts with 3,3'-diaminobenzidine (DAB) to form poly DAB (oxDAB), resulting in a coloured distance signal in the detection zone of the dPAD. The variation of the distance of the observed red-brown colour is directly proportional to the uric acid concentration. Our sensor exhibited a linear range from 0.50 to 6.0 mmol L -1 ( R 2 = 0.9922) with a detection limit (LOD) of 0.25 mmol L -1 , covering the clinical range of uric acid in urine. Hence, there is no need for additional sample preparation or dilution. Additionally, this assay is highly selective, with no interferences. We also found that this approach could accurately and precisely determine uric acid in human control samples with the recovery ranging from 99.37 to 100.35 with the highest RSD of 4.05%. Our method is comparable with the use of a commercially available uric acid sensor at a 95% confidence interval. Consequently, the developed dPAD offers numerous advantages such as cost-effectiveness, simplicity, and ease of operation with unskilled individuals. Furthermore, this concept can be applied for extensive biosensing applications in monitoring other biomarkers as an alternative analytical point-of-care (POC) device.

We report a water-soluble fluorescence and colorimetric copper probe (LysoBC1); this system can also serve for lysosome labeling and for the dynamic tracking of Cu 2+ in living cells. The sensing mechanism takes advantage of the synergic action by the following three components: i) a lysosome targeting unit, ii) the spirolactam ring-opening for the selective copper chelation and iii) the metal-mediated hydrolysis of the rhodamine moiety for fluorescence enhancement. In aqueous environment the molecule acts as a fluorescent reversible pH sensor and as colorimetric probe for Cu 2+ at physiological pH; the hydrolysis of the copper targeting unit resulted in a 50-fold increase of the fluorescence intensity. Existing permanganates (MnO 4 - ) sensing methods suffered from the poor selectivity, restricted solubility, and/or less desirable signaling mechanisms (e.g., "turn-off" processes), resulting in the false positive result. In this work, an effective "turn-on" fluorescence method for MnO 4 - detection in aqueous media was proposed via yellow fluorescence carbon dots (Y-CDs) with the maximum emission peak of 553 nm. Upon the introduction of MnO 4 - , the fluorescence of Y-CDs increased significantly due to the enhanced surface passivation degree and intramolecular charge transfer. In the range of 0.1-10 μM, the fluorescence ratio (F/F 0 ) is proportional to the MnO 4 - ions concentration, realizing the "turn-on" fluorescence sensing of MnO 4 - . On the other hand, utilizing the significant color change of Y-CDs by MnO 4 - ions, the colorimetric analysis can also be established for MnO 4 - assay. Imbalances in acetylcholine levels within the human body readily precipitate neurological disorders. Hence, establishing a highly efficient and sensitive acetylcholine detection platform is of paramount importance. Palladium-based nanoparticles have high catalytic performance, which is of profoundly important in the development of nanozyme technology. Herein, we focused on extracting Leonurus cardiaca L. polysaccharide (LCLP) from Leonurus cardiaca L., which possesses an average molecular weight of 11,910 Da. Meanwhile, it has certain reducing power. Leonurus cardiaca L. polysaccharide-stabilized palladium nanoparticles (Pd n -LCLP NPs) were prepared. Pd n -LCLP NPs exhibited remarkable peroxidase-like properties due to their ability to decompose H 2 O 2 into OH. In addition, Pd n -LCLP NPs were combined with the chromogenic substrate 3,3',5,5'-tetramethylbenzidine to form a colorimetric detection system for the detection of acetylcholine. The linear detection range and the limit of detection were 10 μM-200 μM and 1.02 μM (S/N = 3), respectively. The widespread application of organophosphorus pesticides (OPs) has inflicted significant damage on human well-being and food security. Hence, it is imperative to develop a friendly and accessible biosensor for the detection of OPs. Herein, apple polysaccharide (AP) stabilized palladium nanoparticles (AP-Pd n NPs) with a particle size of 2.75-5.95 nm were prepared using AP as a stabilizer and reducing agent. AP-Pd 30 NPs exhibited good peroxidase-like activity and effectively decomposed H 2 O 2 to ·OH, which catalyzed the 3,3',5,5'-tetramethylbenzidine system to become blue. The catalytic kinetics of AP-Pd 30 NPs conformed to the typical Michelis-Menten equation. Furthermore, OPs directly inhibited the peroxidase-like activity of AP-Pd 30 NPs. Thus, a highly effective colorimetric biosensor was developed for the detection of OPs. The detection range of the biosensor was 0.050 μg/L - 200 mg/L, and the limit of detection was extremely low to 0.010 μg/L. Compared with other nanomaterials, the detection platform based on AP-Pd 30 NPs can effectively detect organophosphorus pesticides without coupling natural enzymes；this method is more economical and practical. A nanocomposite of tannic acid and cellulose nanowhiskers (CNW)-reinforced polysulfone (PSF) was used to develop a metallochromic nanofibrous membrane sensor for iron(III) in aqueous media. Tannic acid was used as an active detecting probe, whereas the CNW@PSF composite was employed as a hosting material. Cellulose nanowhiskers (7-12 nm) were obtained from microcrystalline cellulose (MCC). According to the coloration parameters, a bathochromic shift from colorless (415 nm) to purple (561 nm) occurs when ferric cations bind to the phenolic hydroxyls of the tannic acid probe. The concentration of ferric was found to be directly correlated to the extent of the color change, demonstrating a detection limit of 0.1-250 ppm. This could be attributed to the creation of a coordinative complex between ferric ions and phenolic tannic acid. The generated nanofibers were inspected by energy-dispersive X-ray (EDX) and scanning electron microscopy (SEM). The electrospun nanofibrous membrane showed an average diameter between 75 and 150 nm. The tannic acid-containing nanofibers are remarkably reusable and simple. The tannic acid-encapsulated polysulfone nanofibrous membrane was used to detect various metal ions, demonstrating a high selectivity for Fe 3+ . This study developed a dual-readout system utilizing fluorescence and colorimetry based on iron oxide quantum dots (IO-QDs) for detecting tetracycline (TCy). IO-QDs were synthesized within 6 h using L-leucine as a surface modifier, achieving a more efficient route. Upon interaction with TCy, IO-QDs exhibited a significant decrease in fluorescence response and observable color changes, while fluorescence lifetime remained consistent regardless of TCy presence. Moreover, IO-QDs' fluorescence response remained stable across various temperatures. The Förster resonance energy transfer distance of less than 2 nm and a quenching constant of 2.90 × 10 12  M -1 s -1 indicated static quenching in the presence of TCy. Additionally, significant changes in observed and corrected fluorescence efficiency suggested the involvement of the inner filter effect in the fluorescence quenching of IO-QDs. The synthesized IO-QDs were then utilized for selective and rapid fluorescence-based TCy detection, showing a linear range of 0.5 to 80 μM. Simultaneously, a colorimetric method for TCy detection was established, demonstrating a good linear relationship within the range of 0.5 to 20 μM. The detection limits for TCy were determined as 0.539 and 0.329 μM using fluorescence and colorimetric approaches, respectively. The colloidal gold nanoparticle (AuNP)-based colorimetric lateral flow assay (LFA) is one of the most promising analytical tools for point-of-care disease diagnosis. However, the low sensitivity and insufficient accuracy still limit its clinical application. In this work, a machine learning (ML)-optimized colorimetric LFA with ultrasound enrichment is developed to achieve the sensitive and accurate detection of tau proteins for early screening of Alzheimer's disease (AD). The LFA device is integrated with a portable ultrasonic actuator to rapidly enrich microparticles using ultrasound, which is essential for sample pre-enrichment to improve the sensitivity, followed by ML algorithms to classify and predict the enhanced colorimetric signals. The results of the undiluted serum sample testing show that the protocol enables efficient classification and accurate quantification of the AD biomarker tau protein concentration with an average classification accuracy of 98.11% and an average prediction accuracy of 99.99%, achieving a limit of detection (LOD) as sensitive as 10.30 pg mL -1 . Further point-of-care testing (POCT) of human plasma samples demonstrates the potential use of LFA in clinical trials. Lu'an Gua Pian (LAGP) tea is one of the most famous green teas in China. The quality of green tea is related to its picking periods, especially the green tea before Qingming Festival (usually April 6th) is highly praised as precious in the market. In this work, a simple and cheap indicator displacement colorimetric sensor array combined with smartphone was developed to rapidly identify LAGP picked during different picking periods. First, the chemical component contents of LAGP picked before and after Qingming Festival were analyzed. Second, a well-designed colorimetric sensor array was proposed based on the tea component contents differences. Finally, machine learning was used to process the array data taken by a smartphone. By comparison, the accuracy of the best model for the prediction set was 97%. Meanwhile, the multi-channel advantages of the sensing array were demonstrated by an ablation experiment. MicroRNAs (miRNAs) play an important role in the process of heart failure (HF) and are emerging biomarkers that can be used for the auxiliary diagnosis of HF. However, it is very challenging to accurately analyze the expression levels of trace miRNAs in complex clinical samples. Here, we developed an enzyme-free colorimetric sensor for the ultrasensitive detection of miRNA-423-5p (HF-associated miRNA) based on three-dimensional DNA walkers constructed from functional nucleic acids and gold nanoparticles (AuNPs). DNAzyme with cleavage activity was specifically activated by miRNA-423-5p to sustainably cleave the substrate, thereby releasing the trigger sequence to initiate the subsequent mismatched catalytic hairpin assembly (MCHA) cycle. Then, as the MCHA cycle proceeded to continuously expose the G-quadruplex (GQ) sequence, the sequence bound with hemin to form a large amount of GQ/hemin DNAzyme on the surface of the AuNPs, which rapidly catalyzed the chromogenic oxidation of 3,3',5,5'-tetramethylbenzidine to yield an amplified colorimetric signal readout. The colorimetric sensor exhibited an ultralow detection limit (32 fM), showed excellent specificity and performed well in serum samples. The sensor was applied to detect miRNA-423-5p in clinical plasma samples from healthy individuals and HF patients, and the results revealed its good clinical application in HF diagnosis. Current methods for therapeutic drug monitoring (TDM) have a long turnaround time as they involve collecting patients' blood samples followed by transferring the samples to medical laboratories where sample processing and analysis are performed. To enable real-time and minimally invasive TDM, a microneedle (MN) biosensor to monitor the levels of two important antibiotics, vancomycin (VAN) and gentamicin (GEN) is developed. The MN biosensor is composed of a hydrogel MN (HMN), and an aptamer-functionalized flexible (Flex) electrode, named HMN-Flex. The HMN extracts dermal interstitial fluid (ISF) and transfers it to the Flex electrode where sensing of the target antibiotics happens. The HMN-Flex performance is validated ex vivo using skin models as well as in vivo in live rat animal models. Data is leveraged from the HMN-Flex system to construct pharmacokinetic profiles for VAN and GEN and compare these profiles with conventional blood-based measurements. Additionally, to track pH and monitor patient's response during antibiotic treatment, an HMN is developed that employs a colorimetric method to detect changes in the pH, named HMN-pH assay, whose performance has been validated both in vitro and in vivo. Neospora caninum (N. caninum) is a protozoan parasite that poses a serious risk to livestock by infecting various domestic and wild animals. Loop-Mediated Isothermal Amplification (LAMP) offers a cost-effective, highly sensitive, and specific method for detecting protozoan parasites. This study aims to develop a precise, rapid, and visually assessable colorimetric LAMP method, improving on traditional techniques. We employed a rigorous screening process to identify the optimal primer set for this experiment. Subsequently, we fine-tuned the LAMP reaction at 65 °C for 40 min with 270 μmol/L neutral red. We then confirmed the specificity of primers for N. caninum through experimental validation. The LAMP method demonstrated a lower detection limit compared to traditional Polymerase Chain Reaction (PCR) techniques. While LAMP offers clear advantages, the prevalence of DNA detected in 89 sheep serum and 59 bovine serum samples using the nested PCR method was 3.37 % (3/89) and 1.69 % (1/59), respectively. In contrast, when the LAMP method was employed, the prevalence of detected DNA rose to 5.61 % (5/89) for sheep and 3.38 % (2 /59) for bovine. A comparison of two molecular assays using the intragroup correlation coefficient (ICC) resulted in a value of 0.999 (95 % CI: 0.993-0.996, p < 0.001), indicating the LAMP method is in the "better" range according to James Lee's categorization. An intelligent pH response indicator film is an easy-to-use device for the real-time monitoring of meat freshness during transport and storage. Therefore, a novel pH-sensitive anthocyanin indicator film composed of polyvinyl alcohol-blueberry anthocyanin (BA)-2-hydroxypropyltrimethyl ammonium chloride chitosan (HACC) called PAH-2.0 with 1.2 mg/mL HACC to monitor meat freshness using HACC as the colorimetric enhancer has been developed. BA and HACC were mixed and immobilized in the polyvinyl alcohol matrix by hydrogen bonds, as confirmed via Fourier-transform infrared spectroscopy and X-ray diffraction. The inclusion of HACC improved the color stability and antioxidant and antibacterial properties of the PAH-2.0 film. When applied to pork for freshness monitoring at 4 °C, three freshness stages, including fresh, sub-fresh, and spoiled, could be clearly distinguished based on the color variations of the PAH-2.0 film. The distinct hierarchical color change from purple to blue-violet and finally to grayish-blue was highly correlated with the indicators of pork freshness: pH values, total volatile basic nitrogen, and total viable count. Nanoceria have demonstrated a wide array of catalytic activity similar to natural enzymes, holding considerable significance in the colorimetric detection of alkaline phosphatase (ALP), which is a biomarker of various biological disorders. However, the issues of physiological stability and formation of protein corona, which are strongly related to their surface chemistry, limit their practical application. In this work, CeO 2 nanoparticles characterized by enhanced dimensional uniformity and specific surface area were synthesized, followed by encapsulation with various polymers to further increase catalytic activity and physiological stability. Notably, the CeO 2 nanoparticles encapsulated within each polymer exhibited improved catalytic characteristics, with PAA-capped CeO 2 exhibiting the highest performance. We further demonstrated that the PAA-CeO 2 obtained with enhanced catalytic activity was attributed to an increase in surface negative charge. PAA-CeO 2 enabled the quantitative assessment of AA activity within a wide concentration range of 10 to 60 μM, with a detection limit of 0.111 μM. Similarly, it allowed for the evaluation of alkaline phosphatase activity throughout a broad range of 10 to 80 U/L, with a detection limit of 0.12 U/L.
