Abstract Endoplasmic reticulum (ER)-associated degradation (ERAD) and ER-phagy are two principal degradative mechanisms for ER proteins and aggregates, respectively; however, the crosstalk between these two pathways under physiological settings remains unexplored. Using adipocytes as a model system, here we report that SEL1L-HRD1 protein complex of ERAD degrades misfolded ER proteins and limits ER-phagy and that, only when SEL1L-HRD1 ERAD is impaired, the ER becomes fragmented and cleared by ER-phagy. When both are compromised, ER fragments containing misfolded proteins spatially coalesce into a distinct architecture termed Coalescence of ER Fragments (CERFs), consisted of lipoprotein lipase (LPL, a key lipolytic enzyme and an endogenous SEL1L-HRD1 substrate) and certain ER chaperones. CERFs enlarge and become increasingly insoluble with age. Finally, we reconstitute the CERFs through LPL and BiP phase separation in vitro, a process influenced by both redox environment and C-terminal tryptophan loop of LPL. Hence, our findings demonstrate a sequence of events centered around SEL1L-HRD1 ERAD to dispose of misfolded proteins in the ER of adipocytes, highlighting the profound cellular adaptability to misfolded proteins in the ER in vivo.

The soil organic carbon (SOC) content is determined by multiple factors in terrestrial ecosystems. This study explored the SOC content and its influencing factors in multiple forests of subtropical, temperate, and transitional climate zones in Central China. The quantile regression forest algorithm (QRF) was employed to model the SOC contents along the soil depths up to 100 cm. The models had the highest explained variance score of 0.621 for the topsoil layer (0–20 cm), which decreased with soil depths. In addition to parent material, bulk density, mean tree height, NDVI and topography, climate factors, especially summer precipitation, and surface soil temperature, were identified as the most influential factors for SOC accumulation and spatial variability in these forests. SOC was positively affected by summer precipitation from zero to 100 cm. A higher surface soil temperature was beneficial only for forest SOC in the top layer. The highest SOC > 50 g kg−1 was found under the cover of mixed coniferous forests and evergreen broad-leaved forests at high elevations approximately 1000 m asl, which were dominated by high summer precipitation. At the vertical depth, SOC largely accumulated at a depth of 40 cm but decreased thereafter. Owing to the positive impact of precipitation on forest SOC and the urgent demand for increasing carbon sinks, forest management could focus on mixed evergreen broad-leaved forests and mixed coniferous forests, afforestation, and reforestation of mixed forests based on local species in vast areas.

Sleep apnea syndrome is a breathing disorder with a prevalence exceeding 20% in the overall population, and it can seriously affect health and well-being. However, this condition usually remains undetected because suitable monitoring solutions are lacking. This contribution presents an approach to facilitate apnea diagnosis using a battery-powered, wireless, miniaturized sensing system embedded in a patient’s mask. It combines a photoacoustic-based carbon dioxide detector with temperature and humidity sensors as well as embedded algorithms to automatically detect apnea episodes. The results show the feasibility of detecting apnea using an easily deployable analysis system.

Procambarus clarkii is adept at using natural shelters and caves to evade attacks from predators. However, the concealment abilities and mechanisms of P. clarkii for different types of shelters under predation pressure have not yet been reported. In this study, laboratory experiments were carried out to determine the effects of different coverages (25%, 50%, and 75%) and different combinations (I–VII) of three types of shelters (PVC pipes, water grass, and stone) on the predation rhythm, behavior, and abilities of Silurus asotus on P. clarkii. The results indicated that the predation of S. asotus on P. clarkii exhibited significant rhythmicity under shelter conditions, excluding PVC pipes, 75% stone, and combination VI. Among the three types of shelters, PVC pipes provided the strongest concealment, followed by stone and water grass. With the increase in shelter coverage, the anti-predation ability of P. clarkii continued to increase, and the optimal shade rate for water grass was 50%. In the different shelter combinations, the environmental complexity had little effect on the predation activity of S. asotus on P. clarkii. These findings demonstrated that the type and abundance of shelters in the wild environment can affect the predation rhythm and activities of S. asotus on P. clarkii.

Heat stress is an increasingly concerning topic under global warming. Heat stress can induce organisms to produce excess reactive oxygen species, which will lead to cell damage and destroy the antioxidant defense of aquatic animals. Chinese mitten crab, Eriocheir sinensis, is sensitive to the change in water temperature, and parent crabs are more vulnerable during the breeding stage. In the present study, the multi-omics responses of parent E. sinensis gills to heat stress (24 h) were determined via transcriptome and proteome. The integrative analysis revealed that heat shock protein 70 (HSP70) and glutathione s-transferase (GST) were significantly up-regulated at gene and protein levels after heat stress, indicating that HSP70 and the antioxidant system participated in the regulatory mechanism of heat stress to resist oxidative damage. Moreover, the “Relaxin signaling pathway” was also activated at gene and protein levels under 30 °C stress, which implied that relaxin may be essential and responsible for reducing the oxidative damage of gills caused by extreme heat stress. These findings provided an understanding of the regulation mechanism in E. sinensis under heat stress at gene and protein levels. The mining of key functional genes, proteins, and pathways can also provide a basis for the cultivation of new varieties resistant to oxidative stress.

The effects of salinity on behavior are far-reaching, and Eriocheir sinensis showed disparate behaviors under different salinity conditions. Female crabs were more active in saline water, especially low salinity stress, which is beneficial for female crabs to escape from the low-salinity environment quickly. Then, antennal movement indicated that antennae might be the main osmoreceptors in E. sinensis, and 65 min might be a good starting time for salinity stress to analyze osmoregulation in crabs. Interestingly, E. sinensis had obvious behavioral differences in the high and low salinity stress, and behaviors were more intense in a salinity dip from salinity 18 to salinity 0. This study analyzed the osmoregulatory process of catadromous E. sinensis in different salinity from the point of osmoregulatory organ and behavioral response. These results will provide a scientific basis for the osmoregulatory mechanism of E. sinensis, which are conducive to evaluating and analyzing the impact of saltwater intrusion in the Yangtze River estuary on resource fluctuation.

At present, the resources of anadromous fish in the world are drastically reduced and one of the solutions to promote its recovery may be to identify and protect its feeding grounds. To determine whether the feeding grounds of anadromous Coilia nasus in the Yangtze River are still in the sea area, the quantitative line analysis of Sr and Ca contents (Sr/Ca ratios) and the area distribution analysis of Sr content in the otoliths of C. nasus in three water areas (Poyang Lake, Yangtze River estuary and Shengsi Sea area) were carried out by electron probe microanalysis (EPMA) to analyze the habitat use and the migratory patterns of different estuarine C. nasus. These results showed that the Sr/Ca ratios of all otoliths fluctuated significantly. By comparison, it was found that C. nasus of Yangtze River might exist in the Shengsi Sea area, and the Shengsi Sea area might be one of the feeding grounds of migratory C. nasus. Therefore, it is urgent to carry out a comprehensive investigation on the distribution of related groups in the Shengsi Sea area and the influence of potential threat factors, such as fishing and harsh habitat, to effectively protect the resources and habitats of C. nasus in the Shengsi Sea area.

A shelter is a good habitat for aquatic organisms, which could aid in avoiding cannibalism and facilitate predation. Chinese Mitten Crab (Eriocheir sinensis) is an important aquaculture species with troglodytism and nocturnal habit. To clarify the preference for shelters at different developmental stages of E. sinensis, different shelters (mud, sand, grass and rocks) were selected for comparison. These results indicated that juvenile crabs had a significant preference for grass; button-sized crabs preferred to hide in mud; and the favorite shelters for parent crabs were rocks, followed by mud. E. sinensis in three stages all showed concealing behavior. The concealing behavior of juvenile crabs was the most significant, followed by button-sized and parent crabs. Additionally, E. sinensis held a low hiding rate at night but a high hiding rate during the daytime due to nocturnal habits. These findings will help to better understand the habits of E. sinensis and provide a reference for resource restoration, habitat construction and the restoration of E. sinensis.

Dibutyl phthalate (DBP) poses a severe threat to aquatic ecosystems, introducing hazards to both aquatic species and human health. The ecotoxic effects of DBP on aquatic organisms have not been fully investigated. This study investigates acute toxicity, oxidative damage, and antioxidant enzyme parameters in neonate and adult Daphnia magna exposed to DBP. The obtained results show comparable DBP toxic responses in neonates and adults. The median lethal concentrations (LC50) of DBP in neonates exposed for 24 and 48 h were 3.48 and 2.83 mg/L, respectively. The LC50 of adults for the same DBP exposure durations were 4.92 and 4.31 mg/L, respectively. Increased hydrogen peroxide and malondialdehyde were found in neonates and adults at both 24 and 48 h, while the total antioxidant capacity decreased. Superoxide dismutase activity increased significantly in neonates and adults exposed to 0.5 mg/L DBP, and subsequently diminished at higher DBP concentrations and prolonged exposure. Catalase and glutathione S-transferases activities both decreased markedly in neonates and adults. The changes observed were found to be time and concentration dependent. Overall, these data indicated that the acute toxic effects of DBP exposure on neonates were more pronounced than in adults, and oxidative injury may be the main mechanism of DBP toxicity. These results provide a functional link for lipid peroxidation, antioxidant capacity, and antioxidant enzyme levels in the Daphnia magna response to DBP exposure.

In this study, the complete 15,662 bp mitochondrial genome was determined from Uca borealis using next-generation sequencing technology. It consists of 13 protein-coding genes, 22 tRNAs, 2 ribosomal RNAs, and a control region. Among the 37 genes, 23 genes (9 PCGs and 14 tRNA genes) are encoded on the heavy (+) strand while the remaining 14 genes (four PCGs, eight tRNA genes and two rRNA genes) are located on the light (−) strand. The AT-skew (0.01) for the whole mitogenome is slightly positive while GC-skew (−0.25) is negative, indicating a higher occurrence of As than Ts and Cs than Gs. The phylogenetic tree was constructed using MrBayes method based on the PCGs showed the present species clustered within the Brachyura. The complete mitogenome data would be useful for further study of U. borealis.

The asphalt binders could be modified to improve road perfo rmances by using various methods, and the reinforcement mechanisms are obviously different and quietly affect the road performances. According to the composite reinforcement theory, nano-CaCO3 particles, CaCO3 whiskers, and aramid fibers were used as reinforcements to prepare modified asphalt binders, and the conventional technical performances and dynamic shear rheometer (DSR) rheological properties were measured and investigated. In addition, scanning electron microscopy (SEM) was applied to observe the interfaces between reinforcements and asphalt matrixes and the different reinforcement mechanisms were analyzed. The test results showed that the use of nano-CaCO3 particles, CaCO3 whiskers, and aramid fibers can improve the high-temperature stability of modified asphalt binders by different reinforcement mechanisms. The nano-CaCO3 particles were still effective under high-temperature conditions and could last for a long time through the principle of dispersion enhancement. The CaCO3 whiskers segregate easily and cannot be well dispersed in asphalt binders. The aramid fibers played an important role of stress transmission and fiber reinforcement in asphalt binders, and 2 mm-aramid fibers hold the best reinforcement effects. The conclusions can provide a reference for the selection and application of modifiers in the preparation of modified asphalt in laboratory and engineering projects.

While there is a persistent inverse relationship between latitude and species diversity across many taxa and ecosystems, deviations from this norm offer an opportunity to understand the conditions that contribute to large-scale diversity patterns. Marine systems, in particular, provide such an opportunity, as marine diversity does not always follow a strict latitudinal gradient, perhaps because several hypothesized drivers of the latitudinal diversity gradient are uncorrelated in marine systems. We used a large scale public monitoring dataset collected over an eight year period to examine benthic marine faunal biodiversity patterns for the continental shelf (55-183 m depth) and slope habitats (184-1280 m depth) off the US West Coast (47°20'N-32°40'N). We specifically asked whether marine biodiversity followed a strict latitudinal gradient, and if these latitudinal patterns varied across depth, in different benthic substrates, and over ecological time scales. Further, we subdivided our study area into three smaller regions to test whether coast-wide patterns of biodiversity held at regional scales, where local oceanographic processes tend to influence community structure and function. Overall, we found complex patterns of biodiversity on both the coast-wide and regional scales that differed by taxonomic group. Importantly, marine biodiversity was not always highest at low latitudes. We found that latitude, depth, substrate, and year were all important descriptors of fish and invertebrate diversity. Invertebrate richness and taxonomic diversity were highest at high latitudes and in deeper waters. Fish richness also increased with latitude, but exhibited a hump-shaped relationship with depth, increasing with depth up to the continental shelf break, ~200 m depth, and then decreasing in deeper waters. We found relationships between fish taxonomic and functional diversity and latitude, depth, substrate, and time at the regional scale, but not at the coast-wide scale, suggesting that coast-wide patterns can obscure important correlates at smaller scales. Our study provides insight into complex diversity patterns of the deep water soft substrate benthic ecosystems off the US West Coast.

With the ongoing crisis of biodiversity loss and limited resources for conservation, the concept of biodiversity hotspots has been useful in determining conservation priority areas. However, there has been limited research into how temporal variability in biodiversity may influence conservation area prioritization. To address this information gap, we present an approach to evaluate the temporal consistency of biodiversity hotspots in large marine ecosystems. Using a large scale, public monitoring dataset collected over an eight year period off the US Pacific Coast, we developed a methodological approach for avoiding biases associated with hotspot delineation. We aggregated benthic fish species data from research trawls and calculated mean hotspot thresholds for fish species richness and Shannon's diversity indices over the eight year dataset. We used a spatial frequency distribution method to assign hotspot designations to the grid cells annually. We found no areas containing consistently high biodiversity through the entire study period based on the mean thresholds, and no grid cell was designated as a hotspot for greater than 50% of the time-series. To test if our approach was sensitive to sampling effort and the geographic extent of the survey, we followed a similar routine for the northern region of the survey area. Our finding of low consistency in benthic fish biodiversity hotspots over time was upheld, regardless of biodiversity metric used, whether thresholds were calculated per year or across all years, or the spatial extent for which we calculated thresholds and identified hotspots. Our results suggest that static measures of benthic fish biodiversity off the US West Coast are insufficient for identification of hotspots and that long-term data are required to appropriately identify patterns of high temporal variability in biodiversity for these highly mobile taxa. Given that ecological communities are responding to a changing climate and other environmental perturbations, our work highlights the need for scientists and conservation managers to consider both spatial and temporal dynamics when designating biodiversity hotspots.

Olfaction plays a vital role in the survival and reproduction of shrimps. However, very little information is available about Procambarus clarkii, an important aquaculture species. Thus, we conducted the antennules transcriptome of Procambarus clarkii and identified 26 olfactory-related genes, including nine ionotropic receptors, two ionotropic glutamate receptors, ten variant ionotropic glutamate receptors, four cytochrome P450s, and one carboxylesterase. Our study suggested that ionotropic receptors were the main odorant receptors in Procambarus clarkii. Additionally, the key genes that are responsible for olfactory transduction in Procambarus clarkii were identified in the cAMP-mediated olfactory transduction pathway, including Cyclic nucleotide-gated cation channel beta-1, Adenylyl cyclase III, Calcium/calmodulin-dependent protein kinase II and Na+/Ca2+ exchanger. As a whole, our study laid a solid foundation for further functional elucidations of olfactory molecular mechanism in Procambarus clarkii, and provided further insight for a better understanding of olfaction molecular mechanism in crustaceans. ABSTRACTObjectives Multiple myeloma (MM) is a malignant disease characterized by a single clonal proliferation of B cell-derived plasma cells in the bone marrow. It is the second most common haematologic malignancy in adults. The objective of this retrospective study is to evaluate the diagnostic and prognostic value of haematologic parameters in MM.Methods The difference of NLR/ALB ratio (NAR) and NLR/HDL-C ratio (NHR) between the 151 newly diagnosed MM patients and 153 healthy controls was compared. According to NAR and NHR cutoff values obtained from the ROC curve, MM patients were divided into low group and high group. The differences in hematological parameters and survival time between the two groups were compared. Accurate estimates of forest site productivity are essential for environmental planning and forest management. In this study, we developed a new productivity index, hereafter termed basal area potential productivity index (BAPP), to estimate site productivity for irregular and complex forests characterized by multi-aged, multi-species, and multi-layer stands. We presented the biological relevance of BAPP with its computational details. We also compared BAPP against basal area realized productivity (BARP) in order to verify the practicability and reliability of BAPP. Time-series data of the national forest inventory on 1912 permanent sample plots that were located in two main forest types and consisted of oak-dominated mixed forests and other broadleaf forests in northeast China were used to demonstrate the application of BAPP. The results showed that the value of BAPP for each sample plot was larger than or equal to the corresponding BARP value for each forest type. For appropriately managed stands with relatively better site conditions, the values of both BARP and BAPP were almost identical. The values of the difference between BAPP and BARP could therefore be used to effectively assess forest site productivity. Climate change is expected to substantially affect forest site productivity. However, its effects may vary depending on the climate scenario, region and tree species. We chose Larix olgensis in northeast China to investigate the responses of forest site productivity to regional climate change using a generalized additive model (GAM). Based on site index data and climate variables from 335 townships across the Jilin Province, we developed a climate-sensitive forest site index model, which accounted for 72.9% of the variation in the site index at the referred age of 20 (SI20). Our results indicated that climatic and geographic factors significantly affect forest site productivity. The geographic location, mean annual temperature, mean annual precipitation and mean temperature differential were found to be statistically significant explanatory variables. We predict that the change of mean SI20 would vary from 0.3 m to -0.8 m (2.2% to -5.9%) by 2050 and from 0.5 m to -1.6 m (3.7% to -11.8%) by 2070 under three BC-RCP scenarios with rising temperature and increasing precipitation. Our study suggests that climate is an important factor affecting forest site productivity. Future climate changes could affect the forest site productivity both positively and negatively for Larix olgensis in northeast China. The relationship between climate and forest site productivity has strong implications for adaptive forest management and needs to be considered in forest management planning under future climate change. Allometric models of internodes are an important component of Functional-Structural Plant Models (FSPMs), which represent the shape of internodes in tree architecture and help our understanding of resource allocation in organisms. Constant allometry is always assumed in these models. In this paper, multilevel nonlinear mixed-effect models were used to characterize the variability of internode allometry, describing the relationship between the last internode length and biomass of Pinus tabulaeformis Carr. trees within the GreenLab framework. We demonstrated that there is significant variability in allometric relationships at the tree and different-order branch levels, and the variability decreases among levels from trees to first-order branches and, subsequently, to second-order branches. The variability was partially explained by the random effects of site characteristics, stand age, density, and topological position of the internode. Tree- and branch-level-specific allometric models are recommended because they produce unbiased and accurate internode length estimates. Based on meteorological data from 1978 to 2007, we investigated the annual, seasonal, and fixed hourly variations of Shanghai urban heat island (UHI) in urban and suburban areas and spatial distribution of UHI. The results demonstrated a remarkable interannual increase of UHI intensity in Shanghai. The UHI was the strongest in autumn and the weakest in summer, as a consequence of the prevailing weather conditions. Similar to previous studies of other cities, the UHI in Shanghai was stronger in the nighttime than in the daytime. The SW–NE cross section followed the general cross section of the typical UHI described by Oke () who defines its characteristic parts as “cliff”, “plateau”, and “peak”. Analysis of the association of UHI and urbanization indicated that the UHI increased with the expansion of population and rapid increase of gross domestic product. The continuous increase of power consumption and area of paved road and decrease of area of cropland caused the growth of UHI intensity. Green land had a positive effect on mitigation of heat island based on an inversed U-shaped curve with UHI intensity. [ABSTRACT FROM AUTHOR]
