Background: Phytoremediation is affected by physical and chemical properties of the soil such as soil pH, moisture, and nutrient content. Soil pH is a key element influencing Cd bioavailability and can be easily adjusted in agricultural practices. The soil pH level may relate to the effectiveness of phytoremediation; however, this has not been extensively investigated. In the current study, we evaluated the effect of Cd contamination level (0.56 and 0.92 mg/kg) and soil pH (5, 6, and 7) on Cd accumulation and allocation in Artemisia argyi, a fast-growing perennial crop. Results: Our results indicated that higher soil Cd concentrations reduce A. argyi biomass, and the loss of the root mass was particularly significant. Higher soil pH decreased Cd content in stems and roots of A. argyi cultivated in moderately Cd-polluted soil, and increased Cd content in stems and roots of the plant grown in low Cd-polluted soil. Higher soil pH decreased the percentage of Cd distributed in the soluble fraction and cell walls and increased the percentage of Cd in the organelles of leaf cells for moderate soil Cd levels. The bioconcentration and translocation factor exceeded 4.0 and 1.0, respectively, across all tested treatments, indicating that A. argyi is a promising candidate for phytoremediation. Notably, the effects of soil pH on Cd accumulation and subcellular distribution in A. argyi differed between low and moderately Cd-contaminated soils. Conclusion: Adjustments to soil pH based on the degree of Cd contamination can enhance Cd extraction by A. argyi, thereby reducing the overall remediation cycle of cadmium-polluted paddy fields of South China. Riparian vegetation is crucial for maintaining terrestrial and aquatic biodiversity, but it is threatened by land-use activities. To assess the ecological impacts of riparian vegetation conversion to an oil tea (Camellia oleifera) plantation, we quantified the responses of small mammals in two natural habitats (mature forest and flood-meadow) and in Camellia forests at the community, population, and individual level. We found that the community diversity was similar between Camellia forests and mature forests, but higher than the flood-meadow. Meanwhile, the community composition differed across three habitats, with Camellia forests favoring habitat generalist species. At the population level, Camellia forests and flood-meadow had a similar population density, which were higher than mature forests. At the individual level, Rattus nitidus was less sensitive to this conversion, but the body condition index of Niviventer confucianus was higher in Camellia forests than in mature forests, and Apodemus agrarius in Camellia forests had more ectoparasite load than in the flood-meadow, indicating a species-specific response to the impacts of oil tea plantation. Our study highlights that the occurrence of habitat generalist species and high ectoparasite loads may threaten regional biodiversity and increase the risk of parasite transmission with enlarging the oil tea plantation area within riparian zones. Cytochrome b (cyt b) gene complete sequences (1143bp) of Takydromus sylvaticus were sequenced. In order to clarify the phylogenetic position of the Takydromus sylvaticus, we investigated the phylogeny of 15 Takydromus spp. distributed in East-Asia by Maximum Parsimony (MP), Bayesian Inference (BI), and Maximum Likelihood (ML) methods using DNA fragments of cyt b genes. The results supported that the Platyplacopus merged into Takydromus and negated the validity of Platyplacopus. Furthermore, the prediction of tertiary structures of cyt b exhibited the CD loop region contain two short helices forming a hairpin arrangement, namely cd1 and cd2. Thermostability analysis shows that the CD-loop region is unstable thermodynamically and may provide mobility to amino acids located at the heme, and might provide high flexibility to the top of ISP (iron-sulfur protein) and the cavity region of Qo binding site. It suggested that the two short helices of CD loop region of cyt b was a dominating portion for ISP binding site.

In lacustrine wetlands connected to rivers, the changes in flood regimes caused by hydrological projects lead to changes in the community traits of dominant macrophytes and, consequently, influence the structure and function of wetland vegetation. However, community trait responses of macrophytes to the timing and duration of flood disturbance have been rarely quantified. In 2011–2019, we investigated plant species diversity, density, and biomass in three dominant macrophyte communities (Carex brevicuspis C.B. Clarke, Miscanthus sacchariflorus (Maxim.) Hackel, and Polygonum hydropiper L.) through monthly field surveys in Dongting Lake wetlands. Partial least squares regressions were used to analyze how the variations in hydrological regimes affected plant community traits. Apparent inter-annual fluctuations in plant community traits were detected during 2011–2019. The species richness and Shannon index of diversity of Miscanthus and Polygonum communities increased, whereas the Shannon index of diversity of Carex community decreased. Variation in flooding had a greater effect on Polygonum and Carex community traits than on Miscanthus community traits. Flooding disturbed all plant communities, especially when the duration and timing varied. Shorter inundation periods caused the biomass of Miscanthus community to decline, and that of Carex and Polygonum communities to increase. Earlier flood recession caused the species richness and Shannon index of diversity of Polygonum and Miscanthus community to increase, and those of Carex community to decrease. These findings imply that shorter inundation durations and earlier flood recession generated by the operation of the Three Gorges Dam have changed the macrophyte growth pattern. In species that occur over a wide range of flooding conditions, plant populations may have evolved divergent strategies as a consequence of long-term adaptation to local flooding conditions. In the present study, we investigated the effects of a flooding gradient on the growth and carbohydrate reserves of Polygonum hydropiper plants originating from low- and high-elevation habitats in the Dongting Lake wetlands. The results indicated that shoot length did not differ, whereas the total biomass and carbohydrate reserves were reduced under flooded compared to well-drained conditions for plants originating from both habitat types. However, shoot length, shoot mass, rhizome mass, and total biomass were lower in plants from low-elevation habitats than in those from high-elevation habitats in the flooded condition. Soluble sugar and starch contents in belowground biomass were higher in plants from low-elevation habitats than in those from high-elevation habitats independently of the water level. Therefore, P. hydropiper plants from low-elevation habitats exhibit a lower growth rate and more conservative energy strategy to cope with flooding in comparison with plants from high-elevation habitats. Differential strategies to cope with flooding among P. hydropiper populations are most likely a response to the flooding pressures of the habitat of origin and may potentially drive ecotype differentiation within species along flooding gradients. Belowground bud banks play a fundamental role in the population dynamics of clonal wetland macrophytes. We investigated the size and seasonal dynamics of the belowground bud banks of four dominant macrophytes along a small-scale elevational gradient in Dongting Lake wetlands, China. The belowground bud banks of all four species were found to peak in January, the coldest month of the year. Bud banks were densest for low-elevation Phalaris arundinacea L. (1049 ± 70 buds m −2 ), intermediate for mid-elevation Carex brevicuspis C.B. Clarke and Polygonum hydropiper L. (565 ± 52 buds m −2 and 484 ± 78 buds m −2 , respectively), and lowest for high-elevation Miscanthus sacchariflorus (Maxim.) Hackel (333 ± 52 buds m −2 ). Elevation, which reflects stand flooding regimes, explained 85% of the variability in bud bank size among the four species. The bud bank size of C. brevicuspis , P. hydropiper , and P. arundinacea decreased markedly after the spring sprout, thus indicating that these three species may be sensitive to the timing of disturbance. Bud bank densities for all four species were lowest just before flooding (in May), which might be a strategy to reduce maintenance costs through the flooding season (June–October). Among the four species examined, the bud bank density of P. arundinacea was highest during the growing season, which suggests that this species may be the most resilient to environmental change. Total concentrations of cadmium (Cd), lead (Pb), chrome (Cr), and nickel (Ni) in surface sediment (0–10 cm) from the East Dongting Lake wetlands were determined and the spatial distribution of heavy metals was mapped. The results showed that the single risk indices (Eri) of heavy metals were ranked in the order of Cd > Pb > Ni > Cr. The content of Cd and Pb was gradually reduced from the east (Xiangjiang River) to the west, while the Cr and Ni content had a patchy distribution pattern in the East Dongting Lake wetlands. Cd and Pb contents were correlated with soil pH significantly, while Cr and Ni contents were correlated with soil organic carbon (SOC) and total nitrogen (TN). The origination of heavy metal (Cd, Cr, Ni, and Pb) could be divided into two groups: Cd and Pb from anthropogenic source and Cr and Ni from parent material weathering. Our results indicated that Cd posed a high risk to local ecosystem. The relatively lower pH and higher soil organic carbon (SOC) and total nitrogen (TN) in sediment may inhibit the fixation of heavy metals, which in turn increased the concentration of heavy metal in sediment. The wetland sedge Carex brevicuspis reproduces vegetatively by producing short rhizomes to form clumping ramets phalanx) and long rhizomes to form spreading ramets (guerrilla), resulting in a combined growth form. As an initial step towards understanding the adaptation of Carex growth strategies to seasonal fluctuations in wetland habitats, we investigated the density and composition of C. brevicuspis rhizome populations immediately after flooding (November), in winter (January), in spring (March), and before flooding (May) in the Dongting Lake wetlands, China. The total rhizome density peaked in winter and was lowest before flooding. A large rhizome population in winter may enable C. brevicuspis to survive the seasonal cold weather and recruit a shoot population in the spring. A small rhizome population before flooding may optimize reproductive allocations and be a strategy for enduring the long flooding season. Regardless of date, short rhizomes comprised the majority of the rhizome population (73.0% in March to 98.2% in May). This indicates that C. brevicuspis primarily uses a phalanx growth strategy to utilize locally abundant resources in wetlands. The percentage of long rhizomes in the rhizome population varies significantly between seasons (1.8% in May to 27.0% in March), indicating that growth form also changes with seasonal fluctuation of wetland habitats. Abstract: The rhizomatous sedge Carex brevicuspis can produce clumping ramets from shortened rhizomes (phalanx) and spreading ramets from elongated rhizomes (guerrilla) to form a combined clonal growth form. In this paper, changes in clonal growth and biomass allocation pattern of C. brevicuspis in response to sedimentation were studied. Four sedimentation depths (0, 3, 6, and 9cm) were applied to 48 ramets in a randomized block design. Plants were harvested after 20 weeks. With increasing sedimentation depth, the proportion of spreading ramets to total ramets increased from 19.6% in 0cm to 92.9% in 9cm sedimentation treatments, whereas that of clumping ramets decreased from 80.4% to 7.1%, indicating a change of clonal growth form from phalanx to guerrilla as a response to sedimentation. With increasing sedimentation depth, biomass allocation to shoots and roots did not change, but rhizome mass ratio increased from 2.7% in 0cm to 7.2% in 9cm sedimentation treatments, suggesting that production of long rhizomes changes biomass allocation pattern. The results show that plasticity of clonal growth forms, by which more spreading ramets are produced, is an effective strategy to avoid sedimentation stress under our experimental conditions. Species with mixed mating systems often demonstrate variable expression of breeding system characteristics and thus represent the opportunity to understand the factors and mechanisms that promote both outcrossed and selfed seed production. Here, we investigate variation in levels of herkogamy (variation in stigma-anther separation distance) in a Puerto Rican population of hummingbird pollinated Gesneria citrina Urban. There is significant variation in herkogamy levels among individuals of this species and stigma-anther separation is negatively associated with the ability to set fruits and seeds in the absence of pollinators. The variation in levels of herkogamy may represent a mechanism to ensure the production of some self-fertilized progeny in the absence of hummingbird pollinators. We also describe a novel breeding system in G. citrina, where stamens elongate over time to reach stigma height, but stamen elongation is accelerated by pollination. These results suggest that once the flowers are pollinated, stamen elongation may favor increased pollen removal and siring success, while the reduction in stigma-anther distance no longer imposes the risk of interference between male and female functions. Analysis of the human and mouse genome sequences has enabled a detailed analysis of the structure and organization of the lipoxygenase genes in the respective species. Humans appear to possess six functional genes and at least three pseudogenes while mice have seven functional genes. The arrangement of the genes is quite similar between the species with most of the human lipoxygenase genes appearing on the short arm of chromosome 17 and in mice on the syntenic portion of chromosome 11. The 5-lipoxygenase gene is unique in several respects including its distinct separate chromosomal localization and its size (4–7× larger than other lipoxygenase genes). Three of the seven murine lipoxygenase genes have been disrupted by gene targeting. While the knockout mice appear outwardly normal, a number of important findings have been discovered using these mice and these will be covered in this review. [Copyright &y& Elsevier]

A baculoviral expression vector consisting of a sequence encoding a six-histidine tag apposed to the human platelet 12-lipoxygenase cDNA, under control of the polyhedrin promoter, was constructed. Recombinant 12-lipoxygenase baculoviruses were used to infect Spodoptera frugiperda insect cells (Sf9). At 54 h post-infection, maximal 12-lipoxygenase activity and protein levels were achieved: the enzyme was purified to apparent homogeneity in a single step by nickel-ion-chelation chromatography in which the (His)6-tagged 12-lipoxygenase was eluted with 100 mM imidazole. The purified enzyme metabolized arachidonic acid almost exclusively to 12-hydroperoxyeicosatetracnoic acid with little, if any. epoxyalcohol or reduction products and had a Vmax of 2–4 μmol min-1mg protein-1, Km, of 10 μM and keat of ≈250 min-1. Linoleic acid, on the other hand, was converted to (13S)-13-hydroperoxy-octadecadienoic acid at a rate which was about 2% of that obtained with arachidonic acid as substrate, but displayed the same Km. The enzyme was most active between pH 7.5–8 and activity was stimulated significantly in the presence of 0.006% Tween-20. A polyclonal antibody to the recombinant enzyme was generated and found to recognize a single 75-kDa band in platelets, human erythroleukemia cells and 12-1ipoxygenase baculoviral-infected Sf9 cells by immunoblot and immunoprecipitation methods. 12-Lipoxygenase protein represented 0.1% of the total soluble protein in platelet preparations. In immunofluorescence experiments 12-lipoxygenase was observed in the cytoplasm of infected insect cells and in the human megakaryoblastic DAMI cell line. The trade-off between allocation to sexual and clonal reproduction in clonal plants is influenced by a variety of environmental factors; however, it has rarely been examined under field conditions. In this study, we investigated the trade-off between two modes of reproduction in Carex brevicuspis C. B. Clarke across a small-scale elevational gradient (21-27 m a.s.l.) at the Dongting Lake wetlands, China. The proportion of biomass allocated to and the density of reproductive ramets were higher at low than at intermediate and high elevations. In contrast, the proportion of biomass allocated to and the density of rhizome buds were lower at low than at intermediate and high elevations. Redundancy analysis showed that sexual reproduction was positively correlated with soil moisture content, soil organic matter, total phosphorus, and pH, and negatively correlated with elevation and ramet density. Our findings suggested that allocation to sexual reproduction is favored in disturbed habitats with fertile soils, whereas allocation to vegetative propagation is favored in stable and competitive habitats. Trade-off between allocation to sexual reproduction and vegetative propagation along an elevational gradient might be a reproductive strategy of C. brevicuspis to adapt to the water level fluctuations in wetland habitats. Seedlings and vegetative ramets may contribute differentially to the recruitment of clonal populations in different growth phases, but this has rarely been investigated. In this study, we quantified the number and survivorship of seedlings and vegetative ramets monthly in juvenile and mature populations of Carex brevicuspis. During the first growing season after flooding (from October to January), 9 seedlings m−2 (13% of all established shoots) were found in juvenile populations, while no seedlings were found in mature populations. During the second growing season before flooding (from February to May), no new seedling recruits were found either in juvenile or in mature populations. All shoots of seedlings were withered during the dormant season (January and February), but 62.5% seedlings could produce vegetative ramets in the following growing season. During the dormant season, all the early emerging ramets (sprouted in October) withered, but the later emerging ones (sprouted in November and December) survived in both mature and juvenile populations. These results indicated that seedling recruitment was only apparent in juvenile populations of C. brevicuspis. The genetic diversity in mature C. brevicuspis populations may be established in juvenile populations by seedling recruitment, and sustained in mature populations by vegetative reproduction. Abstract Dongting Lake is the second largest freshwater lake in China and is one of the globally important wintering sites for migratory waterbirds in the East Asian–Australasian Flyway. Crucial sites and environmental variables for wintering migratory waterbirds are of great concern in the Dongting Lake wetlands. In this research, based on annual (2003/2004–2016/2017) waterbird and habitat census data, we recognized the crucial sites for waterbirds during wintering seasons by comparing the difference of waterbird populations at the community, foraging guild and species levels in different natural wetlands within East Dongting Lake, and then identified the crucial environmental variables affecting waterbird distributions by analyzing the relationship between waterbird populations and the environmental variables, including vegetation area, mudflat area, water area with the depth of 0–20 cm, water area with the depth of 20–50 cm, water area with the depth of 50–100 cm, water area with the depth >100 cm, growth status of vegetation (Min, Mean and Max NDVI), and the human disturbance. Results indicated that five natural wetlands, i.e., Daxiaoxi, Chunfeng, Baihu lakes, Dingzi dyke and Tanjiaweizi, were recognized as the crucial wintering sites for migratory waterbirds in the East Dongting Lake. Among the ten selected environmental variables, water areas with the depth of 0–20 cm, 20–50 cm and >100 cm, human disturbance, Min and Mean NDVIs were identified as the crucial environmental variables overall. Waterbirds at different levels exhibited significant linear relationship with certain environmental variables, with the exception of Bean goose and Lesser White-fronted goose at the species level, which showed Gaussian distribution with changes in mean NDVI. The crucial environmental variables appeared to be foraging guild- and species-specific. These findings provide significant information for managers to understand the differences of wetlands and waterbird populations within East Dongting Lake, and to make more targeted conservation efforts. Sediment accretion (burial) and nutrient enrichment may exert a synergistic influence on the growth and distribution of macrophytes in floodplain wetlands; however, this phenomenon has rarely been examined. In this study, we investigated the effects of sediment accretion and nutrient enrichment on the growth and vegetative propagation of Phalaris arundinacea within a Carex thunbergii stand (one P. arundinacea ramet within 25 C . thunbergii ramets) using a factorial sediment burial (0, 3, and 6 cm) and nutrient addition (low, medium, and high) experimental design. High sediment burial (6 cm) without nutrient addition decreased the aboveground and total biomass of C. thunbergii but did not affect P. arundinacea , indicating that P. arundinacea is more tolerant to sediment burial than C. thunbergii . Moderate sediment burial (3 cm) with nutrient addition enhanced the aboveground and total biomass of P. arundinacea but did not affect C. thunbergii , indicating that P. arundinacea may gain a growth advantage over C. thunbergii under moderate sedimentation. High sediment burial with nutrient addition increased the number of rhizomes and ramets produced by P. arundinacea but did not affect C. thunbergii , indicating that the relative abundance of P. arundinacea may increase within the Carex community under high sedimentation conditions. Based on these results, it can be concluded that an increased sedimentation rate facilitates the invasion of P. arundinacea into Carex grasslands, and this invasion is further enhanced by nutrient enrichment. Both seed and bud banks play important roles in the recruitment and maintenance of macrophyte communities; however, few studies have investigated them simultaneously. We investigated the abundance, species composition, and seasonal patterns of seed and bud banks in two dominant macrophyte communities, Carex and Miscanthus , in the Dongting Lake wetlands. The seed densities of both communities were lower from November (after flooding) to March and increased dramatically before flooding (in May). The bud densities of the two dominant communities peaked in the coldest month of the year (January), decreased markedly in March, and were the lowest before flooding. The seed banks of the two macrophyte communities were mainly composed of annual species and a few perennial species, whereas the bud banks were composed of only dominant perennials. Furthermore, the perennial species present in bud banks did not occur in seed banks. Among the soil variables, the bud densities of both plant communities were negatively associated with soil bulk density, whereas the seed density of the Miscanthus community was positively associated with soil bulk density. Our results suggest that seed and bud banks are complementary in the potential recruitment of macrophyte communities; that is, bud banks regulate the demography of dominant perennials, and seed banks contribute to the recruitment and dispersal of annual species. The growth rate hypothesis (GRH) states that a negative correlation exists between the growth rate and N:P and C:P ratios, because fast-growing organisms need relatively more phosphorus-rich RNA to support their high rates of protein synthesis. However, it is still uncertain whether the GRH is applicable in freshwater wetlands. Several studies have shown that water level and sediment type are key factors influencing plant growth and plant C:N:P characteristics in freshwater wetlands. Thus, this study aimed to elucidate the influence of these factors on plant growth and test the GRH under varying water levels and sediment conditions. We designed a controlled experiment at three water levels and under three sediment types using the two dominant plants ( Carex brevicuspis and Polygonum hydropiper ) in the East Dongting Lake wetland, and we further investigated the relative growth rate (RGR); concentrations of total carbon (TC), total nitrogen (TN), and total phosphorus (TP); and plant stoichiometry (ratios of C:N, C:P, and N:P) in the aboveground and belowground parts and whole plants in both species. Results demonstrated that the RGR and TC of both species decreased significantly with decreasing sediment nutrient supply and increasing water level. However, TN and TP of both species were markedly higher at high water levels than at low water levels; furthermore, these were significantly higher on clay than on the other two sediment types at each water level. The C:N and C:P ratios of both species decreased with increasing sediment nutrient supply and water level, whereas N:P decreased in both species with increasing sediment nutrient supply. The aboveground part of C. brevicuspis as well as the aboveground part and whole plant of P. hydropiper were negatively correlated with N:P, which is consistent with the GRH. However, the relationship between the belowground RGR and N:P of these species was inconsistent with GRH. Therefore, the water level and sediment type and their interaction significantly influenced plant RGR and C:N:P characteristics. Despite the predominant role of bud banks in the regeneration of clonal macrophyte populations, few studies have examined the way in which clonal macrophytes adjust the demographic features of bud banks to regulate population dynamics in response to defoliation in wetlands. We investigated the density and composition of bud banks under repeated defoliation in the wetland sedge Carex brevicuspis C. B. Clarke in the Dongting Lake wetlands, China. The density and biomass of rhizome buds and shoots did not decrease significantly in response to repeated defoliation over two consecutive years. The composition of bud banks, which consisted of long and short rhizome buds, also did not change significantly in response to repeated defoliation. Nevertheless, the ramet height and the shoot, root, and rhizome mass of C. brevicuspis declined significantly under repeated defoliation. Our findings suggest that bud banks are a conservative reproductive strategy that enables C. brevicuspis to tolerate a certain amount of defoliation. The maintenance of large bud banks after repeated defoliation may enable C. brevicuspis populations to regenerate and persist in disturbed habitats. However, bud bank density of C. brevicuspis might decline in the long term because the amount of carbon stored in rhizome buds and plants is reduced by frequent defoliation.

Aim: To study the effect of 5-lipoxygenase inhibitors on the expression of intercellular adhesion molecule-1 (ICAM-1) in melanoma cells., Methods: ICAM-1 protein of human melanoma cell a375 was detected by enzyme-linked immunosorbent, flow cytometry and Western blot analysis. Level of ICAM-1 mRNA in a375 was evaluated by Northern blot analysis. Adhesion of a375 to endothelial cell EC304 was analyzed by isotopic tracing., Results: 5-Lipoxygenase inhibitors nordihydroguaiaretic acid, AA861 and MK886, could suppress the expression of ICAM-1 protein as well as of its mRNA in a375 cells and reduce the adhesion of a375 to EC304., Conclusion: 5-Lipoxygenase inhibitors can inhibit the expression of ICAM-1 in human melanoma cells and may be valuable for treatment of melanoma metastasis.
