RNAs, especially non-coding RNAs (ncRNAs), are crucial players in regulating cellular mechanisms due to their ability to interact with and regulate other molecules. Altered expression patterns of ncRNAs have been observed in prostate cancer (PCa), contributing to the disease’s initiation, progression, and treatment response. This study aimed to evaluate the ability of a specific set of RNAs, including long ncRNAs (lncRNAs), microRNAs (miRNAs), and mRNAs, to discriminate between PCa and the non-neoplastic condition benign prostatic hyperplasia (BPH). After selecting by literature mining the most relevant RNAs differentially expressed in biofluids from PCa patients, we evaluated their discriminatory power in samples of unfiltered urine from 50 PCa and 50 BPH patients using both real-time PCR and droplet digital PCR (ddPCR). Additionally, we also optimized a protocol for urine sample manipulation and RNA extraction. This two-way validation study allowed us to establish that miRNAs (i.e., miR-27b-3p, miR-574-3p, miR-30a-5p, and miR-125b-5p) are more efficient biomarkers for PCa compared to long RNAs (mRNAs and lncRNAs) (e.g., PCA3, PCAT18, and KLK3), as their dysregulation was consistently reported in the whole urine of patients with PCa compared to those with BPH in a statistically significant manner regardless of the quantification methodology performed. Moreover, a significant increase in diagnostic performance was observed when molecular signatures composed of different miRNAs were considered. Hence, the abovementioned circulating ncRNAs represent excellent potential non-invasive biomarkers in urine capable of effectively distinguishing individuals with PCa from those with BPH, potentially reducing cancer overdiagnosis.

Abstract In the framework of metric-affine gravity, we consider the role of the boundary term in Symmetric Teleparallel Gravity assuming f(Q, B) models where f is a smooth function of the non-metricity scalar Q and the related boundary term B. Starting from a variational approach, we derive the field equations and compare them with respect to those of f(Q) gravity in the limit of $$B\rightarrow 0$$ B → 0 . It is possible to show that $$f(Q,B)=f(Q-B)$$ f ( Q , B ) = f ( Q - B ) models are dynamically equivalent to f(R) gravity as in the case of teleparallel $$f(\tilde{B}-T)$$ f ( B ~ - T ) gravity (where $$B\ne \tilde{B}$$ B ≠ B ~ ). Furthermore, conservation laws are derived. In this perspective, considering boundary terms in f(Q) gravity represents the last ingredient towards the Extended Geometric Trinity of Gravity, where f(R), $$f(T,\tilde{B})$$ f ( T , B ~ ) , and f(Q, B) can be dealt under the same standard. In this perspective, we discuss also the Gibbons–Hawking–York boundary term of General Relativity comparing it with B in f(Q, B) gravity.

Extracellular vesicles represent a large heterogeneous class of near and long-distance intercellular communication mediators, released by both prokaryotic and eukaryotic cells. Specifically, the scientific community has shown growing interest in exosomes, which are nano-sized vesicles with an endosomal origin. Not so long ago, the physiological goal of exosome generation was largely unknown and required more investigation; at first, it was hypothesized that exosomes are able to remove excess, reject and unnecessary constituents from cells to preserve cellular homeostasis. However, thanks to recent studies, the central role of exosomes in regulating cellular communication has emerged. Exosomes act as vectors in cell–cell signaling by their cargo, proteins, lipids, and nucleic acids, and influence physiological and pathological processes. The findings on exosomes are widespread in a large spectrum of biomedical applications from diagnosis and prognosis to therapies. In this review, we describe exosome biogenesis and the current methods for their isolation and characterization, emphasizing the role of their cargo in female reproductive processes, from gametogenesis to implantation, and the potential involvement in human female disorders.

The idea that proteins are the main determining factors in the functioning of cells and organisms, and their dysfunctions are the first cause of pathologies, has been predominant in biology and biomedicine until recently. This protein-centered view was too simplistic and failed to explain the physiological and pathological complexity of the cell. About 80% of the human genome is dynamically and pervasively transcribed, mostly as non-protein-coding RNAs (ncRNAs), which competitively interact with each other and with coding RNAs generating a complex RNA network regulating RNA processing, stability, and translation and, accordingly, fine-tuning the gene expression of the cells. Qualitative and quantitative dysregulations of RNA-RNA interaction networks are strongly involved in the onset and progression of many pathologies, including cancers and degenerative diseases. This review will summarize the RNA species involved in the competitive endogenous RNA network, their mechanisms of action, and involvement in pathological phenotypes. Moreover, it will give an overview of the most advanced experimental and computational methods to dissect and rebuild RNA networks.

Abstract We discuss equivalent representations of gravity in the framework of metric-affine geometries pointing out basic concepts from where these theories stem out. In particular, we take into account tetrads and spin connection to describe the so called Geometric Trinity of Gravity. Specifically, we consider General Relativity, constructed upon the metric tensor and based on the curvature R; Teleparallel Equivalent of General Relativity, formulated in terms of torsion T and relying on tetrads and spin connection; Symmetric Teleparallel Equivalent of General Relativity, built up on non-metricity Q, constructed from metric tensor and affine connection. General Relativity is formulated as a geometric theory of gravity based on metric, whereas teleparallel approaches configure as gauge theories, where gauge choices permit not only to simplify calculations, but also to give deep insight into the basic concepts of gravitational field. In particular, we point out how foundation principles of General Relativity (i.e. the Equivalence Principle and the General Covariance) can be seen from the teleparallel point of view. These theories are dynamically equivalent and this feature can be demonstrated under three different standards: (1) the variational method; (2) the field equations; (3) the solutions. Regarding the second point, we provide a procedure starting from the (generalised) second Bianchi identity and then deriving the field equations. Referring to the third point, we compare spherically symmetric solutions in vacuum recovering the Schwarzschild metric and the Birkhoff theorem in all the approaches. It is worth stressing that, in extending the approaches to f(R), f(T), and f(Q) gravities respectively, the dynamical equivalence is lost opening the discussion on the different number of degrees of freedom intervening in the various representations of gravitational theories.

The management of waste cooking oil (WCO) often poses significant challenges. The improper disposal of WCO results in negative environmental impacts and economic losses. However, from a circular economy perspective, WCO can be recycled and used as a sustainable feedstock for numerous industrial products, replacing virgin vegetable oils. This approach enables the recovery of resources while simultaneously addressing the problem of WCO disposal. By employing a multi-criteria decision analysis (MCDA) approach, the study assesses three alternative recycling pathways for WCO used as a feedstock in the production of (A1) biodiesel, (A2) biolubricant, and (A3) biosurfactant. The aim is to identify the optimal alternative, taking into account environmental, economic, and technical factors. The procedure involved a team of chemical engineers working in the WCO recycling sector who were selected as decision makers. The ‘priority scale’ combined with the Paired Comparison Technique was employed as a weighting method to evaluate the selected criteria. The results revealed that the decision makers considered environmental sustainability as the most crucial evaluation criterion, followed by the economic criterion. In contrast, the aspect of process management was deemed less significant. Among the compared alternatives, utilizing WCO as a feedstock for biosurfactant production was assessed as the optimal WCO recycling solution. This alternative not only demonstrated the lowest coefficient variation but was also deemed the most favourable option. Biolubricant production was determined to be the second-best alternative. The adopted MCDA approach proved to be a reliable and effective tool, enabling the clear identification of the preferred WCO recycling alternative among those assessed. This was achieved through the utilization of the decision makers’ expertise and knowledge.

Abstract Background Despite availability of clinical guidelines, underdiagnosis, undertreatment, and poor adherence are still significant concerns in allergic rhinitis (AR) therapeutic management. We investigated clinical practice patterns and prescribing behavior of Italian healthcare professionals (HCPs) specialized in AR. Methods One-hundred allergologists, 100 ear, nose and throat (ENT) specialists, and 150 general practitioners (GPs) were recruited. The survey assessed: socio-demographic, work experience, monthly caseload, prescription drivers. Next, HCPs were invited to retrospectively recover patients’ clinical data to investigate: AR clinical characteristics, therapy management, prescription patterns, patient adherence. Descriptive statistics, Chi square, One-Way analysis of variance, and Two-Way Analysis of Variance were performed. Results Allergologists visited more AR patients (31% of monthly caseload) than ENTs (21%, p 7). Conclusions This survey describes the therapeutic approach adopted by Italian physicians to cope with AR and shows that HCPs underestimated AR severity and had a non-realistic perception of patients’ adherence. These findings suggest that further efforts are required to improve AR clinical management in Italy.

Advanced maternal age impairs reproductive performance, influencing the quantity and the quality of oocytes. Mitochondria dysfunction seems to play a decisive role in conditioning the quality of the female gamete. Different in vitro and in vivo studies, demonstrated the antioxidant and anti-inflammatory activities of Resveratrol and its ability to improve mitochondria function even if the exact mechanism of action has not yet been demonstrated in human oocytes. In this paper, by retrospective analysis, we evaluated follicular fluid (FF) miRNome modification in aged women with a poor ovarian reserve receiving a resveratrol-based supplement the three months before the in vitro Fertilization (IVF) cycle. We found 13 differentially expressed microRNAs (miRNAs) in women treated with resveratrol and specifically miR-125b-5p, miR-132-3p, miR-19a-3p, miR-30a-5p and miR-660-5p, regulating mitochondrial proteins, are able to control metabolism and mitochondrial biogenesis. MiRNA expression differences, observed after resveratrol treatment in FF from women with a poor prognosis for IVF, demonstrated that resveratrol may act on mitomiRNAs to improve follicular microenvironment by transcriptomic and proteomic modifications in granulosa cells.

In the last few years, microRNA-mediated regulation has been shown to be important in viral infections. In fact, viral microRNAs can alter cell physiology and act on the immune system; moreover, cellular microRNAs can regulate the virus cycle, influencing positively or negatively viral replication. Accordingly, microRNAs can represent diagnostic and prognostic biomarkers of infectious processes and a promising approach for designing targeted therapies. In the past 18 months, the COVID-19 infection from SARS-CoV-2 has engaged many researchers in the search for diagnostic and prognostic markers and the development of therapies. Although some research suggests that the SARS-CoV-2 genome can produce microRNAs and that host microRNAs may be involved in the cellular response to the virus, to date, not enough evidence has been provided. In this paper, using a focused bioinformatic approach exploring the SARS-CoV-2 genome, we propose that SARS-CoV-2 is able to produce microRNAs sharing a strong sequence homology with the human ones and also that human microRNAs may target viral RNA regulating the virus life cycle inside human cells. Interestingly, all viral miRNA sequences and some human miRNA target sites are conserved in more recent SARS-CoV-2 variants of concern (VOCs). Even if experimental evidence will be needed, in silico analysis represents a valuable source of information useful to understand the sophisticated molecular mechanisms of disease and to sustain biomedical applications.

Due to the serious problem of plastic pollution in aquatic environment, many people reject plastic packaging in favour of glass containers which are considered more sustainable. To avoid misjudgements, the sustainability assessment of packaging alternatives should be carried out with a life cycle thinking approach. In this regard, the study presents a comparative Life Cycle Assessment (LCA) of two alternative packaging systems for drinking water: reusable glass bottles and polyethylene (PET) bottles. The case study was performed considering the real data of an Italian mineral water company that bottles and distributes both natural and sparkling water. The environmental impacts of the two packaging systems were estimated with the ReCiPe 2016 (H) evaluation method adopting both midpoint and endpoint approaches. The results showed that the PET bottle is the most sustainable alternative for natural water for many impact categories; while, in the case of sparkling water, the environmental impacts of the two packaging systems are similar and the most environmentally sound solution can vary depending on the impact category. The following are the most significant aspects of the analysis: (1) the number of reuses of a single glass bottle; (2) the distribution distance. Their variation can determine which packaging is the most sustainable. Therefore, a life cycle assessment approach is needed for each specific case.
