Abstract: Differential cross sections have been extracted from exclusive and kinematically complete high-statistics measurements of quasifree polarized n⃗p scattering performed in the energy region of the d*(2380) dibaryon resonance covering the range of beam energies Tn=0.98−1.29 GeV (s=2.32−2.44 GeV). The experiment was carried out with the WASA-at-COSY setup having a polarized deuteron beam impinged on the hydrogen pellet target and utilizing the quasifree process dp→np+pspectator. In this way the np differential cross section σ(Θ) was measured over a large angular range. The obtained angular distributions complement the corresponding analyzing power Ay(Θ) measurements published previously. A SAID partial-wave analysis incorporating the new data strengthens the finding of a resonance pole in the coupled D33−G33 waves. more...

Abstract: We report on the experimental search for the bound state of an eta meson and He-3 nucleus performed using the WASA-at-COSY detector setup. In order to search for the eta-mesic nucleus decay, the pd -> (3)He2(gamma) and pd -> (3)He6 gamma channels have been analysed. These reactions manifest the direct decay of the ti meson bound in a He-3 nucleus. This non-mesonic decay channel has been considered for the first time. When taking into account only statistical errors, the obtained excitation functions reveal a slight indication for a possible bound state signal corresponding to a He-3-eta nucleus width Gamma above 20 MeV and binding energy B-s between 0 and 15 MeV. However, the determined cross sections are consistent with zero in the range of the systematic uncertainty. Therefore, as final result we estimate only the upper limit for the cross section of the eta-mesic He-3 nucleus formation followed by the eta meson decay which varies between 2 nb and 15 nb depending on possible bound state parameters. more...

Abstract: The Horizon 2020 Project EuPRAXIA (European Plasma Research Accelerator with eXcellence In Applications) is producing a conceptual design report for a highly compact and cost-effective European facility with multi-GeV electron beams accelerated using plasmas. EuPRAXIA will be set up as a distributed Open Innovation platform with two construction sites, one with a focus on beam-driven plasma acceleration (PWFA) and another site with a focus on laser-driven plasma acceleration (LWFA). Abstract: This letter reports a first quantitative analysis of the contribution of higher partial waves in the charge symmetry breaking reaction dd -> He-4 pi(0) using the WASA-at-COSY detector setup at an excess energy of Q = 60 MeV. The determined differential cross section can be parametrized as d sigma/d Omega = a + b cos(2) theta*, where theta* is the production angle of the pion in the center-of-mass coordinate system, and the results for the parameters are a = (1.55 +/- 0.46(stat)(-0.8)(+0.32)(syst)) pb/sr and b = (13.1 +/- 2.1(stat)(-2.7)(+1.0)(syst)) pb/sr. The data are compatible with vanishing p-waves and a sizable d-wave contribution. This finding should strongly constrain the contribution of the A isobar to the dd -> He-4 pi(0) reaction and is, therefore, crucial for a quantitative understanding of quark mass effects in nuclear production reactions. more...

Abstract: We report on the search for the rare decay eta -> pi(0)e(+)e(-) which is of interest to study C violation in the electromagnetic interaction which would indicate contributions from physics beyond the Standard Model, since the allowed decay via a two-photon intermediate state is strongly suppressed. The experiment has been performed using the WASA-at-COSY installation, located at the COSY accelerator of the Forschungszentrum Julich, Germany. In total 3 x 10(7) events of the reaction pd -> He-3 eta have been recorded at an excess energy of Q = 59.8MeV. Based on this data set the C parity violating decay eta -> pi(0)gamma* -> pi(0)e(+)e(-) via a single-photon intermediate state has been searched for, resulting in new upper limits of Gamma(eta -> pi(0)e(+) e(-))/Gamma(eta -> pi(+) pi(-) pi(0)) < 3.28 x 10(-5) and Gamma(eta -> pi(0)e(+) e(-))/Gamma(eta -> all)< 7.5 x 10(-6) (CL=90%), respectively. Abstract: New data on the production of single neutral pions in the pd -> He-3 pi(0) reaction are presented. For fifteen proton beam momenta between p(p) = 1.60GeV/c and p(p) = 1.74 GeV/c, differential cross sections are determined over a large fraction of the backward hemisphere. Since the only previous systematic measurements of single-pion production at these energies were made in collinear kinematics, the present work constitutes a significant extension of the current knowledge on this reaction. Even this far above the production threshold, significant changes are found in the behaviour of the angular distributions over small intervals in beam momentum. more...

Abstract: Exclusive measurements of the quasi-free pn -> pp pi(-) and pp -> pp pi(0) reactions have been performed by means of pd collisions at T-p= 1.2 GeV using the WASA detector setup at COSY. Total and differential cross sections have been obtained covering the energy region T-p= 0.95-1.3 GeV (root s = 2.3-2.46 GeV), which includes the regions of triangle(1232), N*(1440) and d*(2380) resonance excitations. From these measurements the isoscalar single-pion production has been extracted, for which data existed so far only below T-p = 1 GeV. We observe a substantial increase of this cross section around 1 GeV, which can be related to the Roper resonance N*(1440), the strength of which shows up isolated from the triangle resonance in the isoscalar (N pi)(I=0) invariant-mass spectrum. No evidence for a decay of the dibaryon resonance d*(2380) into the isoscalar (NN pi)(I=0) channel is found. An upper limit of 180 mu b (90% CL.) corresponding to a branching ratio of 9% has been deduced. more...

Abstract: Using the production reactions pd -> He-3 omega and pp -> pp omega, the Dalitz plot distribution for the omega -> pi(+)pi(-)pi(0)decay is studied with the WASA detector at COSY, based on a combined data sample of (4.408 +/- 0.042) x 10(4) events. The Dalitz plot density is parametrised by a product of the P-wave phase space and a polynomial expansion in the normalised polar Dalitz plot variables Z and phi. For the first time, a deviation from pure P-wave phase space is observed with a significance of 4.1 sigma. The deviation is parametrised by a linear term 1+2 alpha Z, with alpha determined to be +0.147 +/- 0.036, consistent with the expectations of rho-meson-type final-state interactions of the P-wave pion pairs. Abstract: The exclusive charmonium production process in (P) over barp annihilation with an associated pi 0 meson (p) over barp -> J/psi pi(0) is studied in the framework of QCD collinear factorization. The feasibility of measuring this reaction through the J/psi -> e(+) e(-) decay channel with the AntiProton ANnihilation at DArmstadt ((P) over bar ANDA) experiment is investigated. Simulations on signal reconstruction efficiency as well as the background rejection from various sources including the (P) over barp -> pi(+)pi(-)pi(0) and (p) over barp -> J/psi pi(0)pi(0) reactions are performed with PANDAROOT, the simulation and analysis software framework of the (P) over bar ANDA experiment. It is shown that the measurement can be done at (P) over bar ANDA with significant constraining power under the assumption of an integrated luminosity attainable in four to five months of data taking at the maximum design luminosity. Abstract: Charge symmetry breaking (CSB) observables are a suitable experimental tool to examine effects induced by quark masses on the nuclear level. Previous high precision data from TRIUMF and IUCF are currently used to develop a consistent description of CSB within the framework of chiral perturbation theory. In this work the experimental studies on the reaction dd -> He-4 pi(0) have been extended towards higher excess energies in order to provide information on the contribution of p-waves in the final state. For this, an exclusive measurement has been carried out at a beam momentum of p(d) = 1.2GeV/c using the WASA-at-COSY facility. The total cross section amounts to sigma(tot) =(118 +/- 18(stat) +/- 13(sys) +/- 8(ext)) p band first data on the differential cross section are consistent with s-wave pion production. more...

Abstract: Differential and total cross sections for the pp -> ppK+K- reaction have been measured at a proton beam energy of 2.83 GeV using the COSY-ANKE magnetic spectrometer. Detailed model descriptions fitted to a variety of one-dimensional distributions permit the separation of the pp -> pp phi cross section from that of non-phi production. The differential spectra show that higher partial waves represent the majority of the pp -> pp phi total cross section at an excess energy of 76 MeV, whose energy dependence would then seem to require some s-wave phi-p enhancement near threshold. The non-phi data can be described in terms of the combined effects of two-body final state interactions using the same effective scattering parameters determined from lower energy data. more...

Abstract: We report on a high-statistics measurement of the basic double pionic fusion reaction $pn \to d\pi^0\pi^0$ over the energy region of the so-called ABC effect, a pronounced low-mass enhancement in the $\pi\pi$-invariant mass spectrum. The measurements were performed with the WASA detector setup at COSY. The data reveal the ABC effect to be associated with a Lorentzian shaped energy dependence in the integral cross section. The observables are consistent with a resonance with $I(J^P) =0(3^+)$ in both $pn$ and $\Delta\Delta$ systems. Necessary further tests of the resonance interpretation are discussed. more...

Abstract: The differential and total cross sections for the d+p->3He+eta reaction have been measured at COSY-ANKE at excess energies of 19.5, 39.4, and 59.4 MeV over the full angular range. The results are in line with trends apparent from the detailed near-threshold studies and also agree with those from CELSIUS, though the present data have higher precision. While at 19.5 MeV the results can be described in terms of s- and p-wave production, by 59.4 MeV higher partial waves are required. Including the 19.5 MeV point together with the near-threshold data in a global s- and p-wave fit gives a poorer overall description of the data though the position of the pole in the 3He scattering amplitude, corresponding to the quasi-bound or virtual state, is hardly changed., Comment: 4 pages, 4 figures, 1 table more...

Abstract: The total cross section for the dd -> 4He K+ K- reaction has been measured at a beam momentum of 3.7 GeV/c, corresponding to an excess energy of 39 MeV, which is the maximum possible at the Cooler Synchrotron COSY-J\"{u}lich. A deuterium cluster-jet target and the ANKE forward magnetic spectrometer, placed inside the storage ring, have been employed in this investigation. Abstract: Measurements of the pn -> p_spec dK^+K^- reaction, where p_spec is a spectator proton, have been undertaken at the Cooler Synchrotron COSY-Juelich by detecting a fast deuteron in coincidence with a K^+K^- pair in the ANKE facility. Although the proton beam energy was fixed, the moving target neutron allowed values of the non-resonant quasi-free pn -> dK^+K^- total cross section to be deduced up to an excess energy epsilon ~ 100 MeV. Evidence is found for the effects of K^-d and KKbar final state interactions. Coincident dK^+ pairs were detected with the ANKE spectrometer, and events with a missing K^0-bar invariant-mass subsequently identified. Abstract: We investigate the reactions p n -> d omega and p n -> d phi close to threshold and at higher energies. Near threshold we calculate the S-wave amplitudes within the framework of the two-step model which is described by a triangle graph with pi-mesons in the intermediate state and find a ratio of the S-wave amplitudes squared of R =|A(phi)|^2/|A(omega)|^2 = (4-8) x 10^{-3}. Any significant enhancement of the experimental value of R(phi/omega) over this prediction can be interpreted as a possible contribution of the intrinsic s - anti s component in the nucleon-wave function. We present arguments that there is a strong resonance effect in the omega N channel close to threshold. Abstract: Regulation of apoptosis is crucial for tissue homeostasis under normal development and environmental stress. In Drosophila, cell death occurs in different developmental processes including embryogenesis. Here, we report that two members of the miR-2 seed family of microRNAs, miR-6 and miR-11, function together to limit the level of apoptosis during Drosophila embryonic development. Mutants lacking both miR-6 and miR-11 show embryonic lethality and defects in the central nervous system (CNS). We provide evidence that miR-6/11 functions through regulation of the proapoptotic genes, reaper (rpr), head involution defective (hid), grim and sickle (skl). Upregulation of these proapoptotic genes is responsible for the elevated apoptosis and the CNS defects in the mutants. Abstract: Gene regulatory mechanisms that specify subtype identity of central complex (CX) neurons are the subject of intense investigation. The CX is a compartment within the brain common to all insect species and functions as a 'command center' that directs motor actions. It is made up of several thousand neurons, with more than 60 morphologically distinct identities. Accordingly, transcriptional programs must effect the specification of at least as many neuronal subtypes. We demonstrate a role for the transcription factor Shaking hands (Skh) in the specification of embryonic CX neurons in Tribolium. The developmental dynamics of skh expression are characteristic of terminal selectors of subtype identity. In the embryonic brain, skh expression is restricted to a subset of neurons, many of which survive to adulthood and contribute to the mature CX. skh expression is maintained throughout the lifetime in at least some CX neurons. skh knockdown results in axon outgrowth defects, thus preventing the formation of an embryonic CX primordium. Abstract: Importance: Timely and complete disclosure of medical device defects is necessary to manage patient care safely and effectively., Objectives: To determine if the manufacturer's recommendations following the recall of a medical device were timely and complete, the follow-up information and data provided to patients and physicians were adequate for managing patient care, and the actions taken by the US Food and Drug Administration (FDA) regarding the recall were appropriate., Design, Setting, and Participants: This single-center retrospective case series included 90 of 448 patients who were implanted with a cardiac resynchronization therapy pacemaker at the Minneapolis Heart Institute from May 2003 through January 2011; this pacemaker was recalled in November 2015. In addition, returned product reports submitted by the manufacturer to the FDA via the Manufacturer and User Facility Device Experience (MAUDE) database from January 2008 through December 2018 were analyzed., Main Outcomes and Measures: Clinical outcomes were serious adverse clinical events that occurred before and after the November 2015 recall notifying physicians and patients that the device's battery could fail unexpectedly because of high internal impedance. Technical outcomes were signs and causes of failure., Results: Five of 90 patients observed during 2015 experienced syncope when their pacemakers stopped pacing owing to battery or wire connection defects prior to the recall. Of the 90 patients, 37 (41%) were men, and the median (interquartile range) age at implantation was 71.3 (66.1-78.2) years. Analysis of the MAUDE data revealed that battery failures prior to the recall were associated with serious adverse events that included 1 death, 1 cardiac arrest, 5 syncopal attacks, and 6 heart failure exacerbations; 3 additional prerecall syncopal events were caused by wire connection defects. The manufacturer and the FDA were aware of the battery and wire connection defects for 19 months before issuing the recall, yet the wire connection problem was not included in the advisory and physicians were not informed that interrogating the pacemaker could result in loss of pacing. The FDA classified the recall as class II rather than the more critical class I., Conclusions and Relevance: This case series study of patients implanted with a defective pacemaker found that the pacemaker recall was delayed and that subsequent communications did not include all critical information needed for safe and effective patient care. Abstract: The red flour beetle, Tribolium castaneum, is an emerging model system well suited to the study of embryonic brain development and evolution (see Chapters 11 and 13 ). Brain genesis is driven by specific gene products whose expression underlies a tight spatiotemporal control. Therefore, the analysis of gene expression in time and space provides valuable insights into the molecular mechanisms that govern brain development. Since Tribolium-specific antibodies are scarce, fluorescent RNA in situ hybridization is the method of choice to determine the dynamics of individual gene expression. We have modified common RNA in situ protocols to facilitate the concomitant detection of two gene-specific expression patterns (double fluorescent RNA in situ). In addition, we describe a procedure which combines fluorescent single RNA in situ and immunostaining with gene-specific antibodies. Conventional in situ using RNA probes that are complementary to mature mRNAs often produce diffuse signals. We demonstrate that RNA in situ probes complementary to intronic gene sequences facilitate single cell resolution because the fluorescent signal is restricted to the nucleus. Abstract: The genetic control of anterior brain development is highly conserved throughout animals. For instance, a conserved anterior gene regulatory network specifies the ancestral neuroendocrine center of animals and the apical organ of marine organisms. However, its contribution to the brain in non-marine animals has remained elusive. Here, we study the function of the Tc-foxQ2 forkhead transcription factor, a key regulator of the anterior gene regulatory network of insects. We characterized four distinct types of Tc-foxQ2 positive neural progenitor cells based on differential co-expression with Tc-six3 / optix , Tc-six4 , Tc-chx / vsx , Tc-nkx2.1 / scro , Tc-ey , Tc-rx and Tc-fez1 . An enhancer trap line built by genome editing marked Tc-foxQ2 positive neurons, which projected through the primary brain commissure and later through a subset of commissural fascicles. Eventually, they contributed to the central complex. Strikingly, in Tc-foxQ2 RNAi knock-down embryos the primary brain commissure did not split and subsequent development of midline brain structures stalled. Our work establishes foxQ2 as a key regulator of brain midline structures, which distinguish the protocerebrum from segmental ganglia. Abstract: MicroRNAs are abundant in animal genomes, yet little is known about their functions in vivo. Here, we report the production of 80 new Drosophila miRNA mutants by targeted homologous recombination. These mutants remove 104 miRNAs. Together with 15 previously reported mutants, this collection includes 95 mutants deleting 130 miRNAs. Collectively, these genes produce over 99% of all Drosophila miRNAs, measured by miRNA sequence reads. We present a survey of developmental and adult miRNA phenotypes. Over 80% of the mutants showed at least one phenotype using a p < 0.01 significance threshold. We observed a significant correlation between miRNA abundance and phenotypes related to survival and lifespan, but not to most other phenotypes. miRNA cluster mutants were no more likely than single miRNA mutants to produce significant phenotypes. Abstract: We sequenced Drosophila head RNA to identify a small set of miRNAs that undergo robust circadian cycling. We concentrated on a cluster of six miRNAs, mir-959-964, all of which peak at about ZT12 or lights off. The cluster pri-miRNA is transcribed under bona fide circadian transcriptional control, and all six mature miRNAs have short half-lives, a requirement for cycling. A viable Gal4 knockin strain localizes prominent cluster miRNA expression to the adult head fat body. Analysis of cluster knockout and overexpression strains indicates that innate immunity, metabolism, and feeding behavior are under cluster miRNA regulation. Manipulation of food intake also affects the levels and timing of cluster miRNA transcription with no more than minor effects on the core circadian oscillator. Abstract: Trichomes are cytoplasmic extrusions of epidermal cells. The molecular mechanisms that govern the differentiation of trichome-producing cells are conserved across species as distantly related as mice and flies. Several signaling pathways converge onto the regulation of a conserved target gene, shavenbaby (svb, ovo), which, in turn, stimulates trichome formation. The Drosophila ventral epidermis consists of the segmental alternation of two cell types that produce either naked cuticle or trichomes called denticles. The binary choice to produce naked cuticle or denticles is affected by the transcriptional regulation of svb, which is sufficient to cell-autonomously direct denticle formation. The expression of svb is regulated by the opposing gradients of two signaling molecules--the epidermal growth factor receptor (Egfr) ligand Spitz (Spi), which activates svb expression, and Wingless (Wg), which represses it. It has remained unclear how these opposing signals are integrated to establish a distinct domain of svb expression. We show that the expression of the high mobility group (HMG)-domain protein SoxNeuro (SoxN) is activated by Spi, and repressed by Wg, signaling. SoxN is necessary and sufficient to cell-autonomously direct the expression of svb. The closely related protein Dichaete is co-regulated with SoxN and has a partially redundant function in the activation of svb expression. In addition, we show that SoxN and Dichaete function upstream of Wg and antagonize Wg pathway activity. Abstract: The Drosophila melanogaster ventral nerve cord derives from neural progenitor cells called neuroblasts. Individual neuroblasts have unique gene expression profiles and give rise to distinct clones of neurons and glia. The specification of neuroblast identity provides a cell intrinsic mechanism which ultimately results in the generation of progeny which are different from each other. Segment polarity genes have a dual function in early neurogenesis: within distinct regions of the neuroectoderm, they are required both for neuroblast formation and for the specification of neuroblast identity. Previous studies of segment polarity gene function largely focused on neuroblasts that arise within the posterior part of the segment. Here we show that the segment polarity gene midline is required for neuroblast formation in the anterior-most part of the segment. Moreover, midline contributes to the specification of anterior neuroblast identity by negatively regulating the expression of Wingless and positively regulating the expression of Mirror. In the posterior-most part of the segment, midline and its paralog, H15, have partially redundant functions in the regulation of the NB marker Eagle. Abstract: Background: Segmentation of the Drosophila embryo is a classic paradigm for pattern formation during development. The Wnt-1 homolog Wingless (Wg) is a key player in the establishment of a segmentally reiterated pattern of cell type specification. The intrasegmental polarity of this pattern depends on the precise positioning of the Wg signaling source anterior to the Engrailed (En)/Hedgehog (Hh) domain. Proper polarity of epidermal segments requires an asymmetric response to the bidirectional Hh signal: wg is activated in cells anterior to the Hh signaling source and is restricted from cells posterior to this signaling source., Results: Here we report that Midline (Mid) and H15, two highly related T box proteins representing the orthologs of zebrafish hrT and mouse Tbx20, are novel negative regulators of wg transcription and act to break the symmetry of Hh signaling. Loss of mid and H15 results in the symmetric outcome of Hh signaling: the establishment of wg domains anterior and posterior to the signaling source predominantly, but not exclusively, in odd-numbered segments. Accordingly, loss of mid and H15 produces defects that mimic a wg gain-of-function phenotype. Misexpression of mid represses wg and produces a weak/moderate wg loss-of-function phenocopy. Furthermore, we show that loss of mid and H15 results in an anterior expansion of the expression of serrate (ser) in every segment, representing a second instance of target gene repression downstream of Hh signaling in the establishment of segment polarity., Conclusions: The data we present here indicate that mid and H15 are important components in pattern formation in the ventral epidermis. Abstract: Sox proteins form a family of HMG-box transcription factors related to the mammalian testis determining factor SRY. Sox-mediated modulation of gene expression plays an important role in various developmental contexts. Drosophila SoxNeuro, a putative ortholog of the vertebrate Sox1, Sox2 and Sox3 proteins, is one of the earliest transcription factors to be expressed pan-neuroectodermally. We demonstrate that SoxNeuro is essential for the formation of the neural progenitor cells in central nervous system. We show that loss of function mutations of SoxNeuro are associated with a spatially restricted hypoplasia: neuroblast formation is severely affected in the lateral and intermediate regions of the central nervous system, whereas ventral neuroblast formation is almost normal. We present evidence that a requirement for SoxNeuro in ventral neuroblast formation is masked by a functional redundancy with Dichaete, a second Sox protein whose expression partially overlaps that of SoxNeuro. Genetic interactions of SoxNeuro and the dorsoventral patterning genes ventral nerve chord defective and intermediate neuroblasts defective underlie ventral and intermediate neuroblast formation. Finally, the expression of the Achaete-Scute gene complex suggests that SoxNeuro acts upstream and in parallel with the proneural genes. more...

Abstract: The AP-2 family of transcriptional regulator proteins has three members, alpha, beta and gamma. AP-2alpha and gamma are expressed in placenta and in the human trophoblast cell line JEG-3. AP-2 has been shown to regulate expression of the placental human chorionic gonado-tropin (hCG) alpha- and beta-subunit genes, however, previous work did not distinguish between the family members. Tryptic peptides of the AP-2 protein complexes purified from JEG-3 cells by oligo-affinity chromatography using the hCGalpha AP-2 site match the amino acid sequence of AP-2gamma. The fact that AP-2gamma is present at significant levels and binds the hCGalpha trophoblast-specific element suggests that AP-2gamma is at least part of the binding complex in vivo and plays a role in regulating hCG expression. We show that mutation of each of four AP-2 binding sites within the hCGbeta promoter decreases expression in transfection assays, demonstrating that all four sites are required for maximal expression in JEG-3 cells. Furthermore, we find differences in regulation of the family members: AP-2alpha mRNA levels increase in response to cAMP while AP-2gamma mRNA levels do not. The demonstrated importance of the AP-2 sites in controlling hCGalpha and beta expression and the likely involvement of more than one family member suggest that a balance in AP-2 proteins is involved in coordinate regulation of these genes. Abstract: Asymmetric cell division is a widespread mechanism in developing tissues that leads to the generation of cell diversity. In the embryonic central nervous system of Drosophila melanogaster, secondary precursor cells-ganglion mother cells (GMCs)-divide and produce postmitotic neurons that take on different cell fates. In this study, we show that binary fate decision of two pairs of sibling neurons is accomplished through the interplay of Notch (N) signaling and the intrinsic fate determinant Numb. We show that GMCs have apical-basal polarity and Numb localization and the orientation of division are coordinated to segregate Numb to only one sibling cell. The correct positioning of Numb and the proper orientation of division require Inscuteable (Insc). Loss of insc results in the generation of equivalent sibling cells. Our results provide evidence that sibling neuron fate decision is nonstochastic and normally depends on the presence of Numb in one of the two siblings. Moreover, our data suggest that the fate of some sibling neurons may be regulated by signals that do not require lateral interaction between the sibling cells. more...
