Abstract

Atypical chronic myeloid leukemia (aCML) is a clonal disorder belonging to the myelodysplastic/myeloproliferative syndromes. About 13% of aCML cases carry somatic mutations in ETNK1 gene, encoding for H243Y, N244S, and G245V substitutions. We previously showed that ETNK1 mutations cause a decreased catalytic activity of the enzyme. Despite this evidence however, their oncogenic role remained largely unexplained. Since ETNK1 activity is essential for the synthesis of phosphatidylethanolamine (PE) and given that PE is one of the most abundant phospholipids in the inner membrane of mitochondria, we focused our attention on mitochondrial activity. In order to characterize the oncogenic effect of ETNK1 variants we generated CRISPR/Cas9 clones carrying heterozygous N244S mutation and homozygous ETNK1 deletion (KO cells) on the 293 Flp-In™ cell-line. Both N244S and KO cells showed a significant increase in mitochondrial activity (1.78 and 2.13 fold increase, respectively; p= 0.0096 and p=0.0050) compared to WT, as assessed by MitoTracker™ Red. In line with this finding, electron microscopy revealed a significant modification in mitochondria morphology for N244S and KO cells, changing from an elongated, tubular shape to a round, swollen one. Abstract

Studies on resource utilization by carnivores are essential to assess the role of the species in the ecosystem. These studies help us to understand the mechanisms that influence vertebrate community structure and the relationships between predators and prey. The aims of this study were to compare and describe the food habits of the sympatric Pampas fox (Lycalopex gymnocercus) and crab‐eating fox (Cerdocyon thous) in the Brazilian Pampa, and to evaluate the food niche overlap between them, and infer aspects of habitat preferences on the basis of their prey. For the diet analyses, 25 stomach contents of Pampas foxes and 33 of crab‐eating foxes were collected. We identified 80 prey items (49 for the Pampas fox and 63 for the crab‐eating fox) and grouped them into eight main categories. Mammals and invertebrates were the most frequent items in the diets of both canids, followed by fruits. The trophic niche breadth of the Pampas fox (Bsta = 0.56) was larger than that of the crab‐eating fox (Bsta = 0.47). The diet overlap between them was Ojk = 0.58. However, a PERMANOVA test showed that diets did not differ between species or season. Our results showed that both L. gymnocercus and C. thous had generalist diets, with a great diversity of prey, but based on similar main food items. Items that contributed the most to the diet of the Pampas fox were insects (Gryllotalpidae and Scarabaeidae) and cavies (Cavia aperea) and those that contributed the most to the diet of the crab‐eating fox were fruits of Syagrus romanzoffiana, beetles (Scarabaeidae), amphibians (Leptodactylus spp.) and cavies. This is the first study to describe and compare the food habits of these two species, occurring in sympatry in the Brazilian Pampa. Interspecific competition for food is a critical factor that determines species coexistence in carnivores. In this study we compare and describe the food habits of two sympatric foxes in southern Neotropics, the Pampas fox (Lycalopex gymnocercus) and the crab‐eating fox (Cerdocyon thous). Our results showed that both species had generalist diets, with a great diversity of prey, but based on similar main food items. The foraging strategies of these canids could be associated with the exploration of different microhabitats. This plasticity may be important for the survival of these species under the constant loss of their natural environments, persecution, and other sources of mortality. [ABSTRACT FROM AUTHOR]

Abstract

BACKGROUND: Antineoplastic drugs, such as cisplatin (CDDP), induce disabling peripheral neuropathies, representing a hindrance to effective cancer treatments. The exact pathogenesis of CDDP-induced neuropathy is not yet understood, and the dysregulation of gene expression has been proposed. Valproate (VPA) is an antiepileptic drug recently discovered to remodel gene expression, with hypothetically putative neuroprotective effects. MATERIALS AND METHODS: VPA was tested in both, in vitro and in vivo models of CDDP-neurotoxicity. RESULTS: VPA administered in combination with CDDP promoted dorsal root ganglia (DRG) neurons survival. Moreover, this treatment induced in Wistar rats an improvement of body weight, sensory nerve conduction velocity, and DRG morphometric analysis. In contrast, VPA was not able to rescue CDDP pre-treated rats. CONCLUSION: When used in combination with CDDP, VPA displays a protective action against neuropathy, in our models, suggesting possible future clinical applications.

Abstract

The GAUDI database (Ground-based Asteroseismology Uniform Database Interface, http://sdc.laeff.esa.es/gaudi/) is a preparatory archive for the COROT (COnvection, ROtation and planetary Transits, http://www.astrsp-mrs.fr/projets/corot/) mission developed at LAEFF (Laboratory for Space Astrophysics and Theoretical Physics, http://www.laeff.esa.es). Its intention is to make the ground-based observations obtained in the preparation of the asteroseismology programme available in a simple and efficient way. It contains spectroscopic and photometric data together with inferred physical parameters for more than 1500 objects gathered since January 1998 in 6 years of observational campaigns. In this paper, the main functionalities and characteristics of the system are described. The observations have been collected at ESO-La Silla, Telescopio Nazionale Galileo, Observatoire de Haute-Provence, South African Astronomical Observatory, Tautenberg Observatory and Sierra Nevada Observatory., 12 pages, 3 figures (quality degraded). Abstract

BACKGROUND: Dairy milking is a demanding work task that has been associated with hand and wrist musculoskeletal disorders. Clinical approaches to identify the early effects of musculoskeletal disorders among dairy parlor workers' wrist have not been well defined. OBJECTIVES: The purpose of this pilot study was to develop a study protocol that would assist in the identification and quantification of hand and wrist disorders among dairy workers that perform tasks in the dairy parlor. Additionally, such a study protocol was needed to perform relatively rapid assessments of the wrist/hand on large samples of dairy workers. METHODS:Fourteen dairy parlorworkers were assessed for i) upper limb symptoms andwork history through questionnaire, ii) a physical examination of the upper limb and in particular wrists and iii) wrist ultrasonography. An additional 21 unexposed paired participants (the control group) also participated in the data collection. RESULTS: The study results identified two ultrasound acoustic windows characterized by the highest predictive value for alteration of the wrist's structure. Study results indicated an impairment of the distal median nerve in structure and mobility and impairment of the distal tendon of the muscle extensor carpi ulnaris that included dislocation and frank damage. CONCLUSIONS: This study identified at least two acoustic windows that should be assessed with ultrasound studies on larger groups and in prospective periodical health surveillance of dairy workers. The study confirmed the wrist was at risk for biomechanical stress among workers conducting milking tasks in the dairy parlors. [ABSTRACT FROM AUTHOR]

Abstract

The light curves of Cepheids and other variable stars in Field A of IC 1613, obtained with a CCD and no filter ($Wh$ photometry), have been analyzed. It is possible to separate first overtone from fundamental mode population I Cepheids taking into account the pulsation amplitude, the shape of the light curve and the period. The expected separation is verified in the period--luminosity $PL$ diagram. Light curve Fourier parameters have been compared with those of Magellanic Clouds and galactic Cepheids, in order to point out the effects of the very low metallicity of IC 1613 on the light curve shape. Population II Cepheids of IC 1613 can be discriminated from those of population I in the $PL$ diagram, and, taking into account their color, from other red or blue variables. Their $PL$ relation is consistent with that observed in globular clusters, nearby dwarf spheroidal galaxies and LMC. We have shown it is possible to apply the single-phase method for deriving standard photometry $PL$ relations for population I and II Cepheids; therefore with just one accurate $BVRI$ observation it is possible to use the population I Cepheids for distance determinations. Some unusual stars have been identified on the basis of periods, light curve shapes and colors; they appear to be pulsating stars laying on the extension of $PL$ relation of known anomalous Cepheids. A multisite campaign involving five observatories allowed us to solve the light curve of the Gamma Doradus star HD 224945. The multiperiodic content, the high frequency term at 3.00 c/d and the frequency spacing between the observed terms strongly support its pulsational nature. Abstract

Sequential adsorption of PdCl42− within weak polyelectrolyte layer-by-layer (LbL) self-assembled multilayer films with further electrochemical reduction to yield Pd0 nanoparticles (Pd-NPs) has been demonstrated. The electrocatalytic hydrogenation (ECH) of model molecules such as acetophenone and benzophenone on Pd-NPs of different sizes (6 to 35 nm) and bulk Pd crystal surface in hydroalcoholic acid solution has been investigated. Distribution of reaction products (secondary alcohols and alkanes) and faradaic yield was systematically investigated. While the polyelectrolyte multilayers act as nanoreactors by confining PdCl42− ions and preventing the formation of large crystals, their presence also alters the hydrogenation reaction and therefore heat treated surfaces showed only the effect of nanocrystal size on the reaction selectivity and faradaic yield. Abstract

A multicentre, randomised, open trial with a 2 x 2 factorial design was conducted to compare the benefits and risks of two thrombolytic agents, streptokinase (SK, 1·5 MU infused intravenously over 30-60 min) and alteplase (tPA, 100 mg infused intravenously over 3 h) in patients with acute myocardial infarction admitted to coronary care units within 6 h from onset of symptoms. The patients were also randomised to receive heparin (12 500 U subcutaneously twice daily until discharge from hospital, starting 12 h after beginning the tPA or SK infusion) or usual therapy. All patients without specific contraindications were given atenolol (5-10 mg iv) and aspirin (300-325 mg a day). The end-point of the study was the combined estimate of death plus severe left ventricular damage. 12 490 patients were randomised to four treatment groups (SK alone, SK plus heparin, tPA alone, tPA plus heparin). No specific differences between the two thrombolytic agents were detected as regards the combined end-point (tPA 23·1%; SK 22·5%; relative risk 1·04, 95% Cl 0·95-1·13), nor after the addition of heparin to the aspirin treatment (hep 22·7%, no hep 22·9%; RR 0·99, 95% Cl 0·91-1·08). The outcome of patients allocated to the four treatment groups was similar with respect to baseline risk factors such as age, Killip class, hours from onset of symptoms, and site and type of infarct. The rates of major in-hospital cardiac complications (reinfarction, post-infarction angina) were also similar. The incidence of major bleeds was significantly higher in SK and heparin treated patients (respectively, tPA 0·5%, SK 1·0%, RR 0·57, 95% Cl 0·38-0·85; hep 1·0%, no hep 0·6%, RR 1·64, 95% Cl 1·09-2·45), whereas the overall incidence of stroke was similar in all groups. SK and tPA appear equally effective and safe for use in routine conditions of care, in all infarct patients who have no contraindications, with or without post-thrombolytic heparin treatment. The 8·8% hospital mortality of the study population (compared with approximately 13% in the control cohort of the GISSI-1 trial) indicates the beneficial impact of the proven acute treatments for AMI.

Abstract

Magnetic iron oxide nanoparticles (NPs) are considered for various diagnostic and therapeutic applications in brain including their use as contrast agent for magnetic resonance imaging. In delivery application, the critical step is the transport across cell layers and the internalization of NPs into specific cells, a process often limited by poor targeting specificity and low internalization efficiency. The development of the models of brain endothelial cells and choroidal plexus epithelial cells in culture has allowed us to investigate into these mechanisms. Our strategy is aimed at exploring different routes to the entrapment of iron oxide NPs in these brain related cells. Here we demonstrated that not only cells endowed with a good phagocytic activity like activated macrophages but also endothelial brain capillary and choroidal plexus epithelial cells do internalize iron oxide NPs. Our study of the intracellular trafficking of NPs by TEM, and confocal microscopy revealed that NPs are mainly internalized by the endocytic pathway. Iron oxide NPs were dispersed in water and coated with 3,4-dihydroxyl-L-phenylalanine (L-DOPA) using standard procedures. Magnetic lipid NPs were prepared by NANOVECTOR: water in oil in water (W/O/W) microemulsion process has been applied to directly coat different iron based NPs by lipid layer or to encapsulate them into Solid Lipid Nanoparticles (SLNs). By these coating/loading the colloidal stability was improved without strong alteration of the particle size distribution. Magnetic lipid NPs could be reconstituted after freeze drying without appreciable changes in stability. L-DOPA coated NPs are stable in PBS and in MEM (Modified Eagle Medium) medium. The magnetic properties of these NPs were not altered by the coating processes. We investigated the cellular uptake, cytotoxicity, and interaction of these NPs with rat brain capillary endothelial (REB4) and choroidal plexus epithelial (Z310) cells. By means of widefield, confocal microscopy and flow cytometry we studied the cell uptake of magnetic SLNs derivatized with a fluorescent reporter molecule and of L-DOPA-TRITC coated NPs. Inhibition of the caveolae-mediated pathway by preincubation with filipin and nystatin did not modify the cellular uptake of these NPs in both cell lines. Furthermore a mild decrease of the NPs cell uptake was obtained after chlorpromazine and NaN3 pretreatment, which interferes with clathrin and energy-dependent endocytosis, and cytochalasin and amiloride pretreatment which interfere with macropinocytosis. NPs particle size as such can strongly affect the efficiency of cellular uptake and the mode of endocytosis. Considering that our L-DOPA and magnetic SLNs display a medium hydrodynamic size of 120 nm with a polydispersity index of 0.3, we can assume that the cell uptake process of these NPs may develop, depending the particle size, both via clathrin mediated endocytosis and macropinocytosis and only to less extent via the pathway of caveolae-mediated endocytosis. Taken together these results let us to conclude that SLNs iron loaded and iron based L-DOPA coated NPs are internalized into brain endothelial and choroidal plexus epithelial cells and this might provide the first step of an intracellular trafficking to transport these NPs between blood and brain.
