Abstract Nonsense‐mediated RNA decay (NMD) is an RNA‐based quality control mechanism that eliminates transcripts bearing premature translation termination codons (PTC). Approximately, one‐third of all inherited disorders and some forms of cancer are caused by nonsense or frame shift mutations that introduce PTCs, and NMD can modulate the clinical phenotype of these diseases. 5‐azacytidine is an analogue of the naturally occurring pyrimidine nucleoside cytidine, which is approved for the treatment of myelodysplastic syndrome and myeloid leukemia. Here, we reveal that 5‐azacytidine inhibits NMD in a dose‐dependent fashion specifically upregulating the expression of both PTC‐containing mutant and cellular NMD targets. Moreover, this activity of 5‐azacytidine depends on the induction of MYC expression, thus providing a link between the effect of this drug and one of the key cellular pathways that are known to affect NMD activity. Furthermore, the effective concentration of 5‐azacytidine in cells corresponds to drug levels used in patients, qualifying 5‐azacytidine as a candidate drug that could potentially be repurposed for the treatment of Mendelian and acquired genetic diseases that are caused by PTC mutations.

Rana temporaria is one of the most widespread Palearctic brown frogs. We aimed to clarify distribution pattern of two main genetic clades in the understudied Balkan peninsula by using 16SrRNA and MT-CYTB sequences, already widely applied in analyses of populations from other parts of Europe, while focusing on the broad area along the Morava river (central Balkans) as a known gap in the species distribution. Additionally, we were interested in revealing the extent of haplotype diversity within the main genetic clades in the Balkans, particularly around the supposed suture zone. The results revealed a suture zone between the Western and Eastern Clades in the central part of the Balkan Peninsula. This indicated the existence of a historical barrier between the Balkan Mountain Belt and geographically close mountains surrounding the Vlasina Plateau (Rhodope/Serbian–Macedonian Massif). The overall observed haplotype diversity in populations of R. temporaria from the Balkan Peninsula seems high. Harboring both main genetic clades of R. temporaria qualifies the Balkan Peninsula as another important center of species’ genetic diversity, as well as rich in unique haplotypes. This points out the necessity of applying conservation measures focused on the common European frog populations and habitats in this part of the species’ distribution area.

We monitored the population size of the agile frog (Rana dalmatina) and the common toad (Bufo bufo) from 2011 or 2012, respectively, to the year 2017 at a syntopic breeding site in the vicinity of Belgrade. Adult R. dalmatina population size had minor fluctuations during the years of study (from 351 to 108 frogs). On the contrary, the adult B. bufo population was widely fluctuating towards decline (from 1158 to 141 toads). In both species, population fluctuations were not significantly related to variation of meteorological parameters (air temperature, humidity, precipitation). Density dependence effects on population size were not detected in either species. Apart from possible effects of climate change, the indicated trend towards decline of the monitored B. bufo population could also be the outcome of common population fluctuations or of increasing anthropogenic impact (vicinity of settlement and agricultural land). More years of monitoring more than one population are required to obtain precise information. Nevertheless, our results seem to be coherent with other studies that recommend conservation action for this species.

Hydropower plants (HPPs) have a strong environmental impact on freshwater wetlands. Small diversion HPPs (SDHPPs) with 0.1–10 MW of installed power, redirect water from small mountainous rivers into several-kilometer-long tubes, disrupting complex dynamics of local aquatic food webs and their interactions with neighbor terrestrial food webs. It certainly affects local aquatic communities, but it is often neglected that two highly threatened vertebrate groups—amphibians and reptiles—which live in and around these wetlands, could be affected as well. In the Balkan peninsula, a part of Southeastern Europe, SDHPPs recently became very attractive and profitable for potential investors after they were proclaimed as eligible for subsidies from the national budget. As a result, in year 2020, the maximal projected number of SHPPs in the Balkans increased to 4,556. According to the literature data, ~28% of amphibian species in the Balkan Peninsula use small rivers and streams in the upper parts of watersheds as feeding, breeding and/or nursery habitats. Additionally, 38% of the total number of reptile species in Europe are registered in the hilly/mountainous areas of the peninsula, and 33% of these species strictly need humid habitats. The attempt of this mini-review is to present the facts which show that SHPPs and DSHPPs, in the way they are currently being installed, present harmful energy solution for the biodiversity of the mountain parts of Balkan peninsula, particularly for local amphibian and reptile populations which rely on lotic aquatic ecosystems and/or humid terrestrial habitats.
