The article aims to tease out the relationship between, on the one hand, changing rhetorical strategies for dealing with 'postwar-tabooed' antisemitism in the Austrian parliament and, on the other, shifts in democratic culture - that is, the expression of democratic equality in the publicly sayable. Starting from the theoretical assumptions that parliament symbolises democracy tout court and that parliamentarism is a 'rhetorical condition of democracy' (Kari Palonen), we seek to explore the nexus between parliamentary rhetoric and democracy in depth. We do so, first, by identifying the successive postwar rhetorical strategies for dealing with antisemitism in their (historical) political context and, second, by delineating how those strategies mark shifting boundaries of the sayable in relation to antisemitism in Austrian postwar parliamentary rhetoric. Third, we show how those strategies and shifts signify transformations of Austrian democratic culture and democracy and that this process has a gendered dimension. Methodologically, we draw on a multidisciplinary mix of qualitative approaches, combining discourse and rhetoric analysis, specialised approaches to the analysis of parliamentary debate, and Conceptual History. Conditions perturbing protein homeostasis are known to induce cellular stress responses in prokaryotes and eukaryotes. Here we show for the first time that expression and assembly of a functional type IV secretion (T4S) machinery elicit extracytoplasmic and cytoplasmic stress responses in Escherichia coli. After induction of T4S genes by a nutritional upshift and assembly of functional DNA transporters encoded by plasmid R1-16, host cells activated the CpxAR envelope stress signaling system, as revealed by induction or repression of downstream targets of the CpxR response regulator. Furthermore, we observed elevated transcript levels of cytoplasmic stress genes, such as groESL, with a concomitant increase of [[sigma].sup.32] protein levels in cells expressing T4S genes. A traA null mutant of plasmid R1-16, which lacks the functional gene encoding the major pilus protein pilin, showed distinctly reduced stress responses. These results corroborated our conclusion that the activation of bacterial stress networks was dependent on the presence of functional T4S machinery. Additionally, we detected increased transcription from the rpo[Hp.sub.1] promoter in the presence of an active T4S system. Stimulation of rpo[Hp.sub.1] was dependent on the presence of CpxR, suggesting a hitherto undocumented link between CpxAR and [[sigma].sup.32]-regulated stress networks.

Specialized lytic transglycosylases are muramidases capable of locally degrading the peptidoglycan meshwork of Gram-negative bacteria. Specialized lytic transglycosylase genes are present in clusters encoding diverse macromolecular transport systems. This paper reports the analysis of selected members of the specialized lytic transglycosylase family from type III and type IV secretion systems. These proteins were analysed in vivo by assaying their ability to complement the DNA transfer defect of the conjugative F-like plasmid R1-16 lacking a functional P19 protein, the specialized lytic transglycosylase of this type IV secretion system. Heterologous complementation was accomplished using IpgF from the plasmid-encoded type III secretion system of Shigella sonnei and TrbN from the type IV secretion system of the conjugative plasmid RP4. In contrast, neither VirB1 proteins (Agrobacterium tumefaciens, Brucella suis) nor lagB (Salmonella enterica) could functionally replace P19. In vitro, IpgF, lagB, both VirB1 proteins, HP0523 (Helicobacter pylon) and P19 displayed peptidoglycanase activity in zymogram analyses. Using an established test system and a newly developed assay it was shown that IpgF degraded peptidoglycan in solution. IpgF was active only after removal of the chaperonin GroEL, which co-purified with IpgF and inhibited its enzymic activity. A mutant IpgF protein in which the predicted catalytic amino acid, Glu42, was replaced by Gln, was completely inactive. IpgF-catalysed peptidoglycan degradation was optimal at pH 6 and was inhibited by the lytic transglycosylase inhibitors hexa-N-acetylchitohexaose and bulgecin A.

Microorganisms mainly from the domain of bacteria dominate life in the terrestrial subsurface up to a depth of several kilometers. Microbial life in that habitat is dependent on the availability of water and nutrients of biotic or abiotic origin. To produce biomass, bacterial primary producers are capable to use simple building blocks such as CO2, H2, CH4, reduced iron and sulfur, nitrate, phosphate as well as other salts and trace elements. Here, we describe the microbial content of several biofilm samples taken during the construction phase of a railway tunnel between the provinces of Styria and Carinthia (Koralm Tunnel). We could identify bacterial cells using light microscopy and used 16S rDNA based microbial profiling to characterize the bacterial community structure. Our results clearly show that the massive biofilms in tunnel drainage water were produced by a bacterial consortium typical for the deep continental subsurface biosphere. In many cases the majority of the consortium could be assigned to autotrophs, methanotrophs and methylotrophs. [ABSTRACT FROM AUTHOR]

In conjugative elements such as integrating conjugative elements (ICEs) or conjugative plasmids (CPs) transcription of DNA transfer genes is a prerequisite for cells to become transfer competent, i.e., capable of delivering plasmid DNA via bacterial conjugation into new host bacteria. In the large family of F-like plasmids belonging to the MobF12A group, transcription of DNA transfer genes is tightly controlled and dependent on the activation of a single promoter, designated PY. Plasmid encoded TraJ and chromosomally encoded ArcA proteins are known activators, whereas the nucleoid associated protein heat-stable nucleoid structuring (H-NS) silences the PY promoter. To better understand the role of these proteins in PY promoter activation, we performed in vitro DNA binding studies using purified H-NS, ArcA, and TraJR1 (TraJ encoded by the conjugative resistance plasmid R1). All proteins could bind to R1PY DNA with high affinities; however, only ArcA was found to be highly sequence specific. DNase I footprinting studies revealed three H-NS binding sites, confirmed the binding site for ArcA, and suggested that TraJ contacts a dyad symmetry DNA sequence located between −51 and −38 in the R1PY promoter region. Moreover, TraJR1 and ArcA supplied together changed the H-NS specific protection pattern suggesting that these proteins are able to replace H-NS from R1PY regions proximal to the transcription start site. Our findings were corroborated by PY- lacZ reporter fusions with a series of site specific R1PY promoter mutations. Sequential changes of some critical DNA bases in the TraJ binding site (jbs) from plasmid R1 to plasmid F led to a remarkable specificity switch: The PY promoter became activatable by F encoded TraJ whereas TraJR1 lost its activation function. The R1PY mutagenesis approach also confirmed the requirement for the host-encoded response-regulator ArcA and indicated that the sequence context, especially in the −35 region is critical for PY regulation and function. This paper explores the relation between the use of anti-Semitic rhetoric in post-war Austrian parliamentary debate and the development of the consensusoriented, corporatist model of Austrian democracy, the "consociational model," between 1945 and 1955. Specifically, this paper examines the anti-Semitic stereotypes found in parliament, an arena where "the sayable" of official politics is defined, and whether such anti-Semitic stereotyping serves political-strategic purposes. The predominant pattern of exclusion proves to be the attribution of ambivalence, drawing on the repertoire of nationalist anti-Semitic stereotypes, depicting "emigrants" as "cowards," incapable of love for and defense of their countries. The analysis shows this pattern of exclusion is rooted in an ethnicized, homogeneous, and masculinist understanding of the people - recurrent in contemporary right-wing movements and parties. It follows the lines of Carl Schmitt's concept of the political, in which the distinction of "friend" and "enemy," and hence, the eradication of pluralism and ambivalence, is pivotal. This article analyses antisemitic elements in the Austrian print media discourse about the 2008-2010 economic crisis. The relevant discursive statements are examined in the light of a theoretical understanding of antisemitic discursive threads as found in the prevalent modes of presentation of the economic and financial crisis in the media. The first main finding is the broad avoidance of openly antisemitic stereotypes, with the exception of the Neue Kronen Zeitung. The second main finding is that structurally antisemitic discursive elements appear above all where (a) specific groups("high finance","financial sharks","speculators ") are singled out as the main culprits, (b) these are opposed to a homogeneously constructed"us"(the"Volk"), and (c) where the formers' greed is stressed and where they are accused of harming the people. Here we find nationalistic and latent antisemitic discourses, the stereotypical contrasting of finance and production, conspiracy theories and anti-Americanism closely interwoven. This article presents results from a qualitative analysis of religious and gender-specific ‘othering’ in Austrian and French media discourse on Turkey’s accession to the EU (2004–2006). A typology of arguments justifying inclusion and exclusion of Turkey from Europe or the EU is presented, and gender-specific othering is placed in the context of differing national discourses about Europe and diverging visions of secularisation and citizenship. Secondly, various topoi of orientalism are reconstructed which play a crucial role in both national corpora, and it is shown how various historically shaped discourses of alterity intersect and produce gendered images of cultural and religious otherness. Expression of DNA transfer (tra) genes of F-type conjugative plasmids is required for the assembly of a functional type IV secretion machinery and subsequent plasmid DNA transfer from donor to recipient cells. Transcription of tra genes depends on the activation of a single promoter, designated PY, by the plasmid encoded TraJ protein. We here determine plasmid specificity of TraJ proteins from various subgroups of F-like plasmids and find that plasmid R1 conjugation and PY promoter activation can be achieved only by its cognate activator and by TraJ of the Salmonella plasmid pSLT and not by F or R100 TraJ proteins. In addition, we characterize the PY promoter of plasmid R1. We show that TraJ binds to PY DNA in vivo and that H-NS acts as a silencer of the PY promoter. In the natural plasmid context, H-NS silences transfer gene expression and horizontal plasmid DNA transfer. In contrast to what was found for the F plasmid, lack of H-NS did not abolish the requirement for ArcA and TraJ to reach full tra gene expression and DNA transfer activity. We propose that, besides a passive de-silencing activity, both ArcA and TraJ play a direct role in synergistically stimulating tra operon transcription and subsequent DNA transfer.
