Hyperglycemia in diabetes induces increased endothelin-1 (ET-1) production in the retina, kidney, and heart that may lead to hemodynamic impairment, permeability alteration, and increased extracellular matrix (ECM) protein production. Chronically elevated blood glucose levels may cause oxidative stress in these target tissues of diabetic complications. Poly(ADP-ribose) polymerase (PARP) is a nuclear enzyme activated by DNA strand breaks due to oxidative stress. We investigated the role of PARP in regulating ET-1 expression and ET-1-induced abnormalities in the targets organs of diabetic complications. Male Sprague-Dawley rats were injected with streptozotocin to induce diabetes. Once diabetes was established, half of the diabetic rats were randomly chosen to receive PARP inhibitor 3-aminobenzamide for 4 months. In a second set of experiments, PARP-/- mice and their controls were fed for 2 months with either a normal rodent diet or a 30% galactose diet to induce a normoinsulinemic hyperhexosemic state. Tissues harvested at the conclusion of both experiments were then subjected to real-time RT-PCR analysis for mRNA expression and immunohistochemical assessment of oxidative stress. In both experiments, the hyperhexosemic state upregulated expression of ET-1 mRNA in the retina, kidney, and heart. Furthermore, upregulation of ET-1-dependent ECM transcripts, such as fibronectin and extradomain B-containing fibronectin, was noted in all tissues. These tissues also demonstrated oxidative stress, as evidenced by the presence of nuclei positive for 8-hydroxy-2′-deoxyguanosine. In contrast, inhibition of PARP, either through a chemical means in the diabetic rats or by genetic manipulation in the galactose-fed animals, prevented both oxidative stress and hyperhexosemia-induced upregulation of these genes. These results suggest that, in diabetes, oxidative stress and PARP activation may produce their effects through ET-1. Hence, blockade of such pathways may constitute potential adjuvant treatment modalities in chronic diabetic complications. Les taux de glucose sanguin chroniquement élevés peuvent causer un stress oxydatif dans ces tissus cibles des complications diabétiques. La poly(ADP-ribose) polymérase (PARP) est une enzyme nucléaire activée par les cassures des brins d’ADN provoquées par un stress oxydatif. Nous avons examiné le rôle de la PARP dans la régulation de l’expression de l’ET-1 et des altérations induites par l’ET-1 dans les organes cibles des complications diabétiques. Des rats Sprague-Dawley ont été rendus diabétiques par l’administration de streptozotocine. Une fois le diabète installé, la moitié des rats diabétiques ont été choisis aléatoirement pour recevoir l’inhibiteur de PARP, 3-aminobenzamide, pendant une période de quatre mois. Ces animaux et leurs témoins appariés par l’âge et le sexe ont ensuite été sacrifiés et leurs tissus cultivés. Dans une deuxième expérience, des souris PARP-/- et leurs témoins ont reçu une diète normale pour rongeurs ou une diète contenant 30 % de galactose pour induire un état normoinsulinémique hyperhexosémique. Ces animaux, après un traitement de 2 mois, ont été sacrifiés, et leurs tissus cultivés. Les tissus ont été soumis à une analyse par RT-PCR en temps réel pour quantifier l’expression de l’ARNm, et leur stress oxydatif a été évalué par immunohistochimie. Dans les deux expériences, l’état hyperhexosémique a augmenté l’expression de l’ARNm de l’ET-1 dans la rétine, le rein et le cœur. De plus une augmentation de l’expression des transcrits de la MEC dépendants de l’ET-1, comme la fibronectine et l’extradomaine B contenant la fibronectine, a été notée dans tous les tissus. Ces tissus ont aussi démontré un stress oxydatif, comme indiqué par la présence de noyaux positifs pour la 8-hydroxy-2′-désoxyguanosine. L’inhibition de PARP, par une méthode chimique chez les rats diabétiques ou par une manipulation génétique chez les animaux nourris au galactose, a prévenu le stress oxydatif et l’augmentation de ces gènes induite par l’hyperhexosémie. Diabetic cardiomyopathy structurally manifests as cardiomyocyte hypertrophy and interstitial fibrosis, the latter is due to an increased extracellular matrix (ECM) protein production. Poly (ADP-ribose) polymerase (PARP), a nuclear enzyme activated by DNA strand breaks, is involved in several pathogenetic processes in diabetes. In this study, we investigated the role of PARP activation in the production of fibronectin (FN) and its splice variant EDB+ FN, in the heart in diabetes. Diabetes was induced in male Sprague-Dawley rats by IP injection of 65mg/kg streptozotocin (STZ). One group of diabetic animals were treated with 30 mg/kg (IP) of PARP inhibitor 3-aminobenzamide (ABA). After 4 months of treatment, the rats were sacrificed and cardiac tissues were harvested. In a second series of experiments, we used PARP knockout mice and wild type (WT) mice. As PARP knockout mice are resistant to STZ, we induced a hyperhexosemic state by 30% galactose feeding. After two months of follow-up, the mice were sacrificed and the cardiac tissues were collected. The tissues were analyzed for mRNA expression of FN, EDB+ FN and transcriptional co-activator p300. The diabetic animals showed hyperglycemia and reduced body weight gain. The cardiac tissues from the diabetic rats demonstrated increased mRNA expression of FN, EDB+ FN and p300 genes. PARP blockade by ABA prevented such upregulation. Similarly, galactose feeding caused augmented production of FN, EDB+ FN and p300 mRNA in the heart of WT mice. Such upregulation were also prevented in the PARP knockout mice on galactose. The cardiac tissues of WT mice on galactose further showed presence of increased interstitial fibrosis. The data from this study suggests that, in diabetes, PARP activation may modulate increased FN and EDB+ FN production in the heart via a p300-dependent mechanism. A similar mechanism is also important in the heart of galactose-fed animals.
