Most current statistics courses include some instruction relevant to causal inference. Whether this instruction is incorporated as material on randomized experiments or as an interpretation of associations measured by correlation or regression coefficients, the way in which this material is presented may have important implications for understanding causal inference fundamentals. Although the connection between study design and the ability to infer causality is often described well, the link between the language used to describe study results and causal attribution typically is not well defined. The current study investigates this relationship experimentally using a sample of students in a statistics course at a large western university in the United States. It also provides (non-experimental) evidence about the association between statistics instruction and the ability to understand appropriate causal attribution. The results from our experimental vignette study suggest that the wording of study findings impacts causal attribution by the reader, and, perhaps more surprisingly, that this variation in level of causal attribution across different wording conditions seems to pale in comparison to the variation across study contexts. More research, however, is needed to better understand how to tailor statistics instruction to make students sufficiently wary of unwarranted causal interpretation.

"Lesson study" has garnered considerable attention from educational researchers and practitioners as a promising method for improving instruction. At its core, lesson study reflects a collaborative inquiry process, grounded in cycles of planning, enacting, and reflecting, that promotes teacher learning through active engagement in lesson refinement. In this article, we leverage lesson study as an exemplar case to foreground an examination and application of theory-based evaluation. We overview a novel evaluation framework contextualized to this multidimensional instructional program and draw out the contributions of theory-based evaluation to the study of program effects. To ground the description of this framework, we summarize representative findings from our ongoing evaluation of a lesson study-based instructional program and discuss processes for selecting, using, and managing evidence sources as well as determining causal strands supporting program outcomes. We conclude by discussing lessons learned and implications for theory-based evaluation of complex programs.

Light scattering induced by contamination and defects on optical components can quickly limit the component’s performance. Therefore, imperfection analysis and budgeting are mandatory - but also challenging tasks. On the other hand, imperfections can be elegantly characterized using efficient, robust and non-contact light scattering techniques. This will be demonstrated in this contribution for area covering measurement approaches using laboratory instruments with highest sensitivity as well as elaborated sensor systems that are best suited for extended freeform surfaces. Moreover, the measurement results are used to derive practical imperfection scattering data and models that serve as input to model and predict the imperfection induced scattering on optical system level.

Adoption of Maker programs entails deep cultural and structural changes within schools. In this case study, we interviewed a principal and seven faculty members in a high school in the United States, after the first year of implementing making-centered curricula. We report how faculty members responded to the reform, their motivations and beliefs, and the concomitant shifts in power and status. We found that educators are required to make non-trivial adaptations to their skills, instructional approaches, and pedagogical beliefs, and that successful adaptation may lead them to gain status, resources, and support within the school. Those are gained on account of technical expertise and educators' efforts to promote the vision of the reform. The extent to which faculty members adapt to a reform, accommodate and support others in their process of adapting, or resist it, may determine whether the reform is successful or not. As such, school leaders face the challenge of encouraging faculty to buy into such reforms. The case study provides a unique perspective on Maker-centered reforms and outlines important implications for administrators seeking to implement similar programs.

Abstract Background X-linked adrenoleukodystrophy (ALD) is a neurometabolic disorder caused by pathogenic variants in ABCD1 resulting very long-chain fatty acids (VLCFA) accumulation in plasma and tissues. Males can present with various clinical manifestations, including adrenal insufficiency, spinal cord disease, and leukodystrophy. Female patients typically develop spinal cord disease and peripheral neuropathy. Predicting the clinical outcome of an individual patient remains impossible due to the lack of genotype-phenotype correlation and predictive biomarkers. Methods The availability of a large prospective cohort of well-characterized patients and associated biobank samples allowed us to investigate the relationship between lipidome and disease severity in ALD. We performed a lipidomic analysis of plasma samples from 24 healthy controls, 92 male and 65 female ALD patients. Results Here we show that VLCFA are incorporated into different lipid classes, including lysophosphatidylcholines, phosphatidylcholines, triglycerides, and sphingomyelins. Our results show a strong association between higher levels of VLCFA-containing lipids and the presence of leukodystrophy, adrenal insufficiency, and severe spinal cord disease in male ALD patients. In female ALD patients, VLCFA-lipid levels correlate with X-inactivation patterns in blood mononuclear cells, and higher levels are associated with more severe disease manifestations. Finally, hematopoietic stem cell transplantation significantly reduces, but does not normalize, plasma C26:0-lysophosphatidylcholine levels in male ALD patients. Our findings are supported by the concordance of C26:0-lysophosphatidylcholine and total VLCFA analysis with the lipidomics results. Conclusions This study reveals the profound impact of ALD on the lipidome and provides potential biomarkers for predicting clinical outcomes in ALD patients.

Abstract Linear confinement with Casimir scaling of the string tension in confining gauge theories is a consequence of a certain property of the Polyakov loop related to random matrices. This mechanism does not depend on the details of the theories (neither the gauge group nor dimensions) and explains approximate Casimir scaling below string-breaking length. In this paper, we study 3d SU(2) pure Yang-Mills theory numerically and find the same random-matrix behavior for rectangular Wilson loops. We conjecture that this is a universal feature of strongly coupled confining gauge theories.

Abstract We present our investigations of SU(N) adjoint QCD in two dimensions with one Majorana fermion on the lattice. We determine the relevant parameter range for the simulations with Wilson fermions and present results for Polyakov loop, chiral condensate, and string tension. In the theory with massive fermions, all observables we checked show qualitative agreement between numerical lattice data and theory, while the massless limit is more subtle since chiral and non-invertible symmetry of the continuum theory are explicitly broken by lattice regularization. In thermal compactification, we observe N perturbative vacua for the holonomy potential at high-T with instanton events connecting them, and a unique vacuum at low-T. At finite-N, this is a cross-over and it turns to a phase transition at large-N thermodynamic limit. In circle compactification with periodic boundary conditions, we observe a unique center-symmetric minimum at any radius. In continuum, the instantons in the thermal case carry zero modes (for even N) and indeed, in the lattice simulations, we observe that chiral condensate is dominated by instanton centers, where zero modes are localized. We present lattice results on the issue of confinement vs. screening in the theory and comment on the roles of chiral symmetry and non-invertible symmetry.

Abstract We propose an orbifold lattice formulation of QCD suitable for quantum simulations. We show explicitly how to encode gauge degrees of freedom into qubits using noncompact variables, and how to write down a simple truncated Hamiltonian in the coordinate basis. We show that SU(3) gauge group variables and quarks in the fundamental representation can be implemented straightforwardly on qubits, for arbitrary truncation of the gauge manifold.

Abstract We present new investigations on the Adjoint Filtering Method (AFM), a proposal for filtering gauge configurations by using adjoint zero modes. This method relies on the existence of the Supersymmetric Zero Mode (SZM), whose density follows the gauge action density for classical configurations. We review how a similar construction on the lattice is implemented based on the overlap operator and test the method against smooth configurations showing a remarkable agreement with the expected densities even when pairs of fractionals instanton/anti-instantons are present and noise is added. Then we explore the application of the method to Monte Carlo generated configurations based on SU(2) gauge group. The tuning of the parameters and quantitative results are explicitly shown for a T 3 × R lattice. We show explicit examples comparing the AFM to the density obtained from the Gradient Flow. The agreement is remarkable for some specific configurations containing fractional instantons with the advantage that the AFM does not modify the underlying structures.

Neutron irradiation causes embrittlement of reactor pressure vessel (RPV) steels. Post-irradiation annealing is capable of partly or fully restoring the unembrittled condition. While annealing at high temperatures (e.g., 475°C) was successfully applied to extend the lifetime of operating VVER-440 reactors, the benefit of annealing at lower temperatures (e.g., 343°C–the maximum to which the primary cooling water can be heated) is a matter of debate. In this study, neutron-irradiated VVER-440 RPV base metal and weld were exposed to isothermal annealing at 343°C up to 2,000 h. Given the limited amount of material, the degree of recovery was estimated in terms of Vickers hardness, the ductile-brittle transition temperature derived from small punch tests, and the master curve reference temperature derived from fracture mechanics tests of mini samples. For the base metal, small-angle neutron scattering was applied to underpin the findings at the nm-scale. We have found significant partial recovery in both materials after annealing for 300 h or longer. The variations of the degree of recovery are critically discussed and put into the context of wet annealing.

Abstract We explain the microscopic origin of linear confinement potential with the Casimir scaling in generic confining gauge theories. In the low-temperature regime of confining gauge theories such as QCD, Polyakov lines are slowly varying Haar random modulo exponentially small corrections with respect to the inverse temperature, as shown by one of the authors (M. H.) and Watanabe. With exact Haar randomness, computation of the two-point correlator of Polyakov loops reduces to the problem of random walk on group manifold. Linear confinement potential with approximate Casimir scaling except at short distances follows naturally from slowly varying Haar randomness. With exponentially small corrections to Haar randomness, string breaking and loss of Casimir scaling at long distance follow. Hence we obtain the Casimir scaling which is only approximate and holds only at intermediate distance, which is precisely needed to explain the results of lattice simulations. For (1 + 1)-dimensional theories, there is a simplification that admits the Casimir scaling at short distances as well.

Many teacher education models involve reflecting on teaching practice for the sake of improving it. Such reflection must be carefully structured to help practitioners identify and act upon significant opportunities for improvement (SOIs). Learning from SOIs requires the cognitive activities of noticing students' mathematical thinking and its connection to instructional practice, along with an affective disposition to view sub-optimal teaching practices as learning opportunities. We draw upon existing literature and theory related to the notion of developing positive error cultures to identify design principles for helping teachers learn from their own sub-optimal practices rather than becoming discouraged by them. The design principles include experience-based learning, low-stakes settings, collaboration, process reflection, and exploration of disagreements. We then describe a mathematics teacher education environment incorporating the design principles. Examples of pre-service teachers' work within the environment are analysed for possible patterns of learning from SOIs within a positive error culture. Based on these examples, a four-quadrant model to characterise teachers' learning from SOIs is proposed. The four quadrants describe various outcomes related to recognising and resolving SOIs.

We present an exploratory study on the use of ontologies for student assessment during a focus group with teachers in student-centred learning environments. The work is framed by a theoretical argument about coherence between instructional values and assessment systems, and how this may be achieved through a "community of judgment" with teachers at the centre. The purpose of the study was to understand the process and perceived usefulness of collaborative ontology development, at a narrative and conceptual level without grades or detailed probabilistic relationships. Findings of two kinds are discussed. First, we identified a set of framework-building moves used by teachers during ontology-based reasoning. Second, we present evidence that participants perceived the ontology development process to be useful and empowering in sharpening their ability to communicate nuanced assessment arguments.

Alternative splicing (AS) is an important molecular biological mechanism regulated by complex mechanisms involving a plethora of cis and trans-acting elements. Furthermore, AS is tissue specific and altered in various pathologies, including infectious, inflammatory, and neoplastic diseases. Recently developed immuno-oncological therapies include monoclonal antibodies (mAbs) and chimeric antigen receptor (CAR) T cells targeting, among others, immune checkpoint (ICP) molecules. Despite therapeutic successes have been demonstrated, only a limited number of patients showed long-term benefit from these therapies with tumor entity-related differential response rates were observed. Interestingly, splice variants of common immunotherapeutic targets generated by AS are able to completely escape and/or reduce the efficacy of mAb- and/or CAR-based tumor immunotherapies. Therefore, the analyses of splicing patterns of targeted molecules in tumor specimens prior to therapy might help correct stratification, thereby increasing therapy success by antibody panel selection and antibody dosages. In addition, the expression of certain splicing factors has been linked with the patients’ outcome, thereby highlighting their putative prognostic potential. Outstanding questions are addressed to translate the findings into clinical application. This review article provides an overview of the role of AS in (tumor) diseases, its molecular mechanisms, clinical relevance, and therapy response.

Voice-based interfaces provide new opportunities for firms to interact with consumers along the customer journey. The current work demonstrates across four studies that voice-based (as opposed to text-based) interfaces promote more flow-like user experiences, resulting in more positively-valenced service experiences, and ultimately more favorable behavioral firm outcomes (i.e., contract renewal, conversion rates, and consumer sentiment). Moreover, we also provide evidence for two important boundary conditions that reduce such flow-like user experiences in voice-based interfaces (i.e., semantic disfluency and the amount of conversational turns). The findings of this research highlight how fundamental theories of human communication can be harnessed to create more experiential service experiences with positive downstream consequences for consumers and firms. Abstract Type I interferon (IFN) signalling is tightly controlled. Upon recognition of DNA by cyclic GMP-AMP synthase (cGAS), stimulator of interferon genes (STING) translocates along the endoplasmic reticulum (ER)-Golgi axis to induce IFN signalling. Termination is achieved through autophagic degradation or recycling of STING by retrograde Golgi-to-ER transport. Here, we identify the GTPase ADP-ribosylation factor 1 (ARF1) as a crucial negative regulator of cGAS-STING signalling. Heterozygous ARF1 missense mutations cause a previously unrecognized type I interferonopathy associated with enhanced IFN-stimulated gene expression. Disease-associated, GTPase-defective ARF1 increases cGAS-STING dependent type I IFN signalling in cell lines and primary patient cells. Mechanistically, mutated ARF1 perturbs mitochondrial morphology, causing cGAS activation by aberrant mitochondrial DNA release, and leads to accumulation of active STING at the Golgi/ERGIC due to defective retrograde transport. Our data show an unexpected dual role of ARF1 in maintaining cGAS-STING homeostasis, through promotion of mitochondrial integrity and STING recycling.

Abstract The Arctic environment is transforming rapidly due to climate change. Aerosols’ abundance and physicochemical characteristics play a crucial, yet uncertain, role in these changes due to their influence on the surface energy budget through direct interaction with solar radiation and indirectly via cloud formation. Importantly, Arctic aerosol properties are also changing in response to climate change. Despite their importance, year-round measurements of their characteristics are sparse in the Arctic and often confined to lower latitudes at Arctic land-based stations and/or short high-latitude summertime campaigns. Here, we present unique aerosol microphysics and chemical composition datasets collected during the year-long Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition, in the central Arctic. These datasets, which include aerosol particle number concentrations, size distributions, cloud condensation nuclei concentrations, fluorescent aerosol concentrations and properties, and aerosol bulk chemical composition (black carbon, sulfate, nitrate, ammonium, chloride, and organics) will serve to improve our understanding of high-Arctic aerosol processes, with relevance towards improved modelling of the future Arctic (and global) climate.

Manufacturing management and operations place heavy emphasis on monitoring and improving production performance. This supervision is accomplished through strategies of manufacturing performance management, a set of measurements and methods used to monitor production conditions. Over the last 30 years, the most prevalent measurement of traditional performance management has been overall equipment effectiveness, a percentile summary metric of a machine’s utilization. The technologies encapsulated by Industry 4.0 have expanded the ability to gather, process, and store vast quantities of data, creating the opportunity to innovate on how performance is measured. A new method of managing manufacturing performance utilizing Industry 4.0 technologies has been proposed by McKinsey & Company (New York City, NY, USA), and software tools have been developed by PTC Inc. (Boston, MA, USA) to aid in performing what they both call digital performance management. To evaluate this new approach, the digital performance management tool was deployed on a Festo (Esslingen, Germany) Cyber-Physical Lab (FCPL), an educational mock production environment, and compared to a digitally enabled traditional performance management solution. Results from a multi-day production period displayed an increased level of detail in both the data presented to the user and the insights gained from the digital performance management solution as compared to the traditional approach. The time unit measurements presented by digital performance management paint a clear picture of what and where losses are occurring during production and the impact of those losses. This is contrasted by the single summary metric of a traditional performance management approach, which easily obfuscates the constituent data and requires further investigation to determine what and where production losses are occurring.

The Epstein–Barr virus (EBV) has a very high prevalence (>90% in adults), establishes a lifelong latency after primary infection, and exerts an oncogenic potential. This dsDNA virus encodes for various molecules, including microRNAs (miRs), which can be detected in the latent and lytic phases with different expression levels and affect, among others, immune evasion and malignant transformation. In this study, the different EBV miRs are quantified in EBV-positive lymphomas, and the impact on the host cell transcriptome of the most abundant EBV miRs will be analyzed using comparative RNA sequencing analyses. The EBV miRs ebv-miR-BART1, -BART4, -BART17, and -BHRF1-1 were most highly expressed, and their selective overexpression in EBV-negative human cells resulted in a large number of statistically significantly down- and up-regulated host cell genes. Functional analyses showed that these dysregulated target genes are involved in important cellular processes, including growth factor pathways such as WNT, EGF, FGF, and PDGF, as well as cellular processes such as apoptosis regulation and inflammation. Individual differences were observed between these four analyzed EBV miRs. In particular, ebv-miR-BHRF1-1 appears to be more important for malignant transformation and immune evasion than the other EBV miRs.

Performing arts computing environments have received little attention in the educational sphere; yet, they offer opportunities for learners to validate their efforts, ideas, and skills through showcasing their work in a public-facing performance. In this work, we explore an out-of-school dance and computing educational program run by the organization, STEM From Dance. The organizational mission is to create an equitable learning experience for young women of color to engage with computing while exposing them to STEM careers. Through an analysis of eleven interviews with youth participants, instructors, and the executive director, we examine how the social, cultural, and political dimensions of the learning environment facilitate identity work in computing and dance. Our findings point to three primary activities used by the organization to promote equity: (1) providing psychological safety through a supportive community environment, (2) meaningfully engaging with learners' social and cultural context through creative work with constructionist artifacts, and (3) actively promoting identity work as women of color in computing and STEM through both artifact work and community events. Applying the constructs of identity and psychological safety we explore the tensions and synergies of designing for equity in this performing arts and computing learning environment. We demonstrate how the seemingly contradictory elements of a high-stakes performance within a novice learning environment provides unique opportunities for supporting young women of color in computing, making them non-negotiable in the organization's efforts to promote equity and inclusion. Our work illustrates how attending closely to the sociocultural dimensions in a constructionist learning environment provides lenses for navigating equity, identity work, and support for inclusive computing.

Traditional robot navigation passively plans/replans to avoid any contact with obstacles in the scene. This limits the obtained solutions to the collision‐free space and leads to failures if the path to the goal is obstructed. In contrast, humans actively modify their environment by repositioning objects if it assists locomotion. This article aims to bring robots closer to such abilities by providing a framework to detect and clear movable obstacles to continue navigation. The approach leverages a multimodal robot skin that provides both local proximity and tactile feedback regarding physical interactions with the surroundings. This multimodal contact feedback is employed to adapt the robot's behavior when interacting with object surfaces and regulating applied forces. This enables the robot to remove bulky obstacles from its path and solves otherwise infeasible navigation problems. The system's ability is demonstrated in simulation and real‐world scenarios involving movable and nonmovable obstacles.

Digital Fabrication (DF) bezeichnet das Planen, Konstruieren und Herstellen von Gegenständen mithilfe von Software und computergesteuerten Werkzeugen. DF ist eng mit Making und Maker Education verbunden und bietet gleichzeitig viele inhaltliche Anknüpfungsmöglichkeiten für den Medien- und Informatikunterricht. Der Beitrag argumentiert für DF als Verbindungsstück zwischen Maker Education und Informatikunterricht, indem ein DF-Prozess, bestehend aus den drei Schritten Konstruktion, Transformation und Interpretation, eingeführt wird. Dieser Prozess wird sowohl aus der Anwender- als auch aus der fachinformatischen Perspektive betrachtet. Die herausgearbeiteten Anknüpfungspunkte werden anschliessend mit existierenden Kompetenzmodellen der Informatik verglichen. Das Resultat ist, dass mit der Anwendung von DF im Informatikunterricht eine breite Abdeckung der Kompetenzbereiche (ausgehend von den Bildungsstandards der GI) erreicht werden kann. Neben dieser wichtigen Voraussetzung ist für eine feste Integration von Making und DF in den Informatikunterricht vor allem die Fortbildung von Lehrkräften diesbezüglich von grosser Bedeutung, weshalb ein Seminarkonzept für Lehramtsstudierende erarbeitet wurde. Die ersten Learnings der Pilotierung zeigen, dass eine Integration von DF in den Informatikunterricht gelingen kann und zeigen offene Forschungsfragen auf. Insbesondere ist für eine nachhaltige und umfassende Verankerung von Making im Schulunterricht neben der inhaltlichen Passung auch die Integration von Projektmethoden und Schülerzentrierung in den Informatikunterricht notwendig.
