Based on observations obtained with the Nordic Optical Telescope (NOT) we investigate the spectral variability of the Herbig Ae star RR Tau. This star belongs to the UX Ori family, characterized by very deep fadings caused by the screening of the star with opaque fragments (clouds) of the protoplanetary discs. At the moments of such minima one observes strong spectral variability due to the fact that the dust cloud occults, for an observer, not only the star but also a part of the region where the emission spectrum originates. We calculated a series of obscuration models to interpret the observed variability of the H α line parameters. We consider two main obscuration scenarios: (1) the dust screen rises vertically above the circumstellar disc, and (2) the screen intersects the line-of-sight moving azimuthally with the disc. In both cases, the model of the emission region consists of a compact magnetosphere and a magnetocentrifugal disc wind. Comparison with observations shows that the first scenario explains well the variability of the radiation flux, the equivalent width, as well as the asymmetry of the H α line during eclipses, while the second scenario explains them only partly. This permits us to suggest that in the case of RR Tau, the main causes of the eclipses are either a structured disc wind, or the charged dust lifted along the field lines of the poloidal component of the magnetic field of the circumstellar disc. International audience; Multi-isotope fingerprinting (sulphur, oxygen and strontium isotopes) has been tested to study the provenances of medieval and Renaissance French and Swedish alabaster works of art. Isotope signatures of historical English, French and Spanish alabaster source quarries or areas are revealed to be highly specific, with a strong intra-group homogeneity and strong inter-group contrasts, especially for Sr and S isotopes. The chosen combination of isotope tracers is a good basis for forensic work on alabaster provenance, allowing verification of hypotheses about historical trade routes as well as identification of fakes and their origin. The applied analytical techniques of continuous flow isotope ratio mass spectrometry (CF-IRMS) and thermal ionization mass spectrometry (TIMS) only require micro-samples in the low-milligram range, thus minimizing the impact on the works of art.

A catalogue of molecular aberrations that cause ovarian cancer is critical for developing and deploying therapies that will improve patients’ lives. The Cancer Genome Atlas project has analysed messenger RNA expression, microRNA expression, promoter methylation and DNA copy number in 489 high-grade serous ovarian adenocarcinomas and the DNA sequences of exons from coding genes in 316 of these tumours. Here we report that high-grade serous ovarian cancer is characterized by TP53 mutations in almost all tumours (96%); low prevalence but statistically recurrent somatic mutations in nine further genes including NF1, BRCA1, BRCA2, RB1 and CDK12; 113 significant focal DNA copy number aberrations; and promoter methylation events involving 168 genes. Analyses delineated four ovarian cancer transcriptional subtypes, three microRNA subtypes, four promoter methylation subtypes and a transcriptional signature associated with survival duration, and shed new light on the impact that tumours with BRCA1/2 (BRCA1 or BRCA2) and CCNE1 aberrations have on survival. 25. Type I collagen alpha1 Sp1 polymorphism and the risk of cruciate ligament ruptures or shoulder dislocations.

- Abstract
BACKGROUND: Cruciate ligament ruptures and shoulder dislocations are often caused by trauma, but predisposing intrinsic factors might also influence the risk. These injuries are more common in those with a previously injured sibling, an observation that might indicate a genetic predisposition. It is well known that polymorphisms in the collagen I gene are associated not only with osteoporosis and osteoporotic fracture risk, but also with osteoarthritis. HYPOTHESIS: Because collagen I is abundant in ligaments and tendons, the authors hypothesized that collagen I alpha1 Sp1 polymorphism also was related to the occurrence of cruciate ligament ruptures and shoulder dislocations. STUDY DESIGN: Case-control study; Level of evidence, 3. METHODS: A total of 358 patients and 325 randomly selected population-based female controls were included in the study. Of the cases, 233 had a cruciate ligament rupture and 126 had had a shoulder dislocation. Age-adjusted odds ratios (ORs) with 95% confidence intervals (CIs) estimated by unconditional logistic regression were used as measures of association. RESULTS: Compared with the homozygous SS category, the heterozygous participants displayed a similar risk (OR, 1.06; 95% CI, 0.76-1.49), whereas the ss genotype was underrepresented in the injured population compared with the controls (OR, 0.15; 95% CI, 0.03-0.68). This latter estimate was similar for both cruciate ligament ruptures and shoulder dislocations, and was furthermore not modified by general joint laxity. CONCLUSION: The authors found a substantially decreased risk of these injuries associated with collagen type I alpha1 Sp1 polymorphism. 28. Attachment in a group of adult international adoptees.

The present study examines whether foreign adoptees differ from normative samples in adult attachment status, and explores within-group differences associated with (in)secure attachment status in adult adoptees. Forty adoptees, from a previous representative study of foreign adoptees in Sweden, were interviewed with the Adult Attachment Interview (AAI). In addition, they were questioned about issues related to identity and family history, and self-assessment instruments were used to assess self-esteem and mental health. Compared to norm groups, the adoptees did not differ significantly in attachment status. However, insecure attachment organization on the AAI was associated with late adoption and a desire to know more about one's biological roots. Late adoption, memories from the time before the adoption, divorce, lack of contact in the adoptive family with the child's origin, and a tendency in adoptees not to think about their biological background, were all associated with unresolved/disorganized status with respect to loss or abuse. There was a nonsignificant tendency for secure attachment status to be associated with higher mental health and self-esteem than insecure attachment status. [ABSTRACT FROM AUTHOR]

Mirtazapine (Mir) is a novel antidepressant, reported to raise extracellular noradrenaline (NA) through blockade of α
2 -autoreceptors and serotonin (5-HT) output via (1) indirect activation of facilitatory α1 -adrenoceptors on the cell bodies of ascending 5-HT neurones and (2) blockade of presynaptic release-modulating α2 -heteroreceptors on 5-HT terminals in the forebrain. To further assess the effect of Mir on NA/5-HT system interplay, including putative regional differences in the effects of the drug on 5-HT release in rat forebrain, we used in vivo microdialysis in anaesthetised rats. Probes were implanted in the dorsal hippocampus (DH) and frontal cortex (FCx), representing median and dorsal raphe 5-HT projection areas, respectively. In the DH, Mir (10 mg/kg s.c.) completely blocked the 5-HT release-suppressing action of the selective α2 -adrenoceptor agonist clonidine (0.1 mg/kg s.c.), but had no effect per se on the 5-HT output. Neither drug significantly changed the 5-HT levels in the FCx. Mir perfused locally (10 µM via reverse-dialysis) also failed to significantly elevate 5-HT output, and did not affect the clonidine response in either brain area. Thus, the data confirm the basic α2 -adrenoceptor-blocking properties of Mir, but are only partly concordant with previous studies reporting an increase of 5-HT output after Mir alone. Moreover, we find no elevation in 5-HT by the reference α2 -adrenoceptor antagonist idazoxan (0.3–1.0 mg/kg s.c.). The discrepancies encountered, and the potential ability of α2 -adrenoceptor antagonists in general to raise the output of 5-HT, are discussed with particular reference to methodological and other factors that may influence the experimental outcome (e.g., brain regional aspects, different α2 -adrenoceptor subtypes, potential differences in adrenoceptor tone under varying experimental conditions). This article briefly summarizes, within the context of a brief review of the relevant literature, the outcome of our recent rat microdialysis studies on (1) the relative importance of serotonin (5-HT)1A versus 5-HT1B autoreceptors in the mechanism of action of 5-HT reuptake blocking agents, including putative regional differences in this regard, and (2) autoreceptor responsiveness following chronic SSRI administration. First, our data are consistent with the primacy of 5-HT1A autoreceptors in restraining the elevation of 5-HT levels induced by SSRIs, whereas nerve terminal 5-HT1B autoreceptors appear to have an accessory role in this regard. Second, there is an important interplay between cell body and nerve terminal 5-HT autoreceptors, and recent findings suggest that this interplay may potentially be exploited to obtain regionally preferential effects on 5-HT neurotransmission in the central nervous system, even upon systemic drug administration. In particular, emerging data suggest that somatodendritic 5-HT1A autoreceptor- and nerve terminal 5-HT1B autoreceptor-mediated feedback may be relatively more important in the control of 5-HT output in dorsal raphe-frontal cortex and median raphe-dorsal hippocampus systems, respectively. Third, 5-HT autoreceptors evidently retain the capability to limit the 5-HT transmission-promoting effect of SSRIs after chronic treatment. Thus, although the responsiveness of these sites is probably somewhat reduced, residual autoreceptor capacity still remains an effective restraint on large increases in extracellular 5-HT, even after prolonged treatment. If a further increase in extracellular 5-HT is crucial to the remission of depression in patients responding only partially to prolonged administration of antidepressants, then sustained adjunctive treatment with autoreceptor-blocking drugs may consequently prove useful in the long term. - Abstract
The biliary excretion of the vasopressin analogue 1-deamino-8-D-arginine vasopressin (dDAVP) was determined in the pig after three administration routes, intrajugular venous, intraportal venous and intraduodenal. In all cases the biliary excretion was less than 1% of the administered dose. The plasma/bile concentration ratio was less than 1:1. A significant first-pass effect was found when the liver was exposed to a high intraportal dose of dDAVP. Possible uptake and degradation/biotransformation was evaluated by incubating [
3 H]dDAVP with liver tissue slices showing that [3 H]dDAVP was rapidly removed from the incubation medium. The following conclusions can be drawn from these experiments: 1) The intestinal mucosa constitutes the major barrier to intestinal absorption of dDAVP. 2) dDAVP is excreted in bile in small amounts. 3) Indirect evidence suggests that the dDAVP molecule is degraded/biotransformed in the liver at its C-terminus. The degradation of the vasopressin analogue dDAVP was studied by reversed phase high-performance liquid chromatography (HPLC) after incubations in pancreatic juice and intestinal mucosa homogenates. dDAVP remained stable in pancreatic juice for a period of 60 min. while the parent hormone arginine vasopressin (AVP) was completely degraded within 5 min. In intestinal mucosa homogenates dDAVP was degraded with half-lives of 9 min. (fast phase) and 161 min. (slow phase), about four times slower than AVP. By amino-acid analysis it was confirmed that the metabolite [Mpa
1 , Des-D-Arg8 -Gly9 NH2 ]-vasopressin was gradually produced. No other breakdown products were observed. These findings may be of value for the further development of more stable peptide analogues which may be effective upon oral administration. [ABSTRACT FROM AUTHOR]- Published

33. Determination of Thyroxine Release in Vivo: A Simple, Specific and Sensitive Method in Mice.

When thyroid hormone secretion in vivo was studied by the release of
125 I from thyroid to blood after TSH in mice preloaded with the isotope, only 54.4 ± 2.0% (± SE) of plasma125 I was butanol extractable, indicating low specificity for thyroid hormones. To improve specificity plasma125 I was bound to anti-T4 rabbit antibodies and precipitated with goat antirabbit serum. The intraassay coefficient of variation was 2.5%, and anti-T4 bound radioactivity correlated well with that extractable with butanol (r = 0.95, P < 0.001). The effect of 40-1000 μU TSH intravenously in mice pretreated with 1 μg T3 3 times during 2 days to suppress endogenous TSH was evaluated 2 hrs after the injection of TSH by 1) conventional radioimmunoassays of total and free stable T4 in plasma (no preloading with125 I), 2) total blood125 I in mice preloaded with125 I, and 3) anti-T4 bound125 I in plasma in mice preloaded with125 I. Stable T4 increased with TSH doses of 70 μU or higher, but the slope was low and the relation between error variance and slope did not permit a useful bioassay. Total blood125 I responded significantly to 40 μU TSH and showed a favourable relation between error variance and slope, but the specificity may be questioned. Anti-T4 bound125 I in plasma also responded significantly to 40 μU TSH and showed a very good relation between error variance and slope. The new technique of measuring anti-T4 bound radioactivity seems to combine the specificity of radioimmunoassay with a precision and sensitivity well comparable to that of the conventional radioiodine release technique. - Abstract
We present a measurement of the photon structure function F in the reaction ee→eeX for Q in the range 0.2< Q<7 GeV, using 9,200 multihadron events obtained with the TPC/Two-Gamma detector at PEP. The data have been corrected for detector effects using a regularized unfolding procedure and are presented as a function of x and Q. The structure function shows scaling in the region 0.3< Q<1.6 GeV, x<0.3 and rises for higher Q. At Q=5.1 GeV the results are compared with QCD and, within the scheme of Antoniadis and Grunberg, rather conservative bounds for the QCD scale parameter of 133±50< $$\Lambda _{\overline {{\rm M}S} } $$ <268±98 MeV are obtained. A study of the final state structure shows that the rise of F is consistent with being entirely due to the pointlike component of the photon. [ABSTRACT FROM AUTHOR]

Three currently used fragmentation models are tested by studying the distribution of particles between jet axes in the 3-jet events of e e annihilation, using data collected by the Time Projection Chamber at PEP. These three models - the Lund String model, the Webber Cluster model and an Independent Fragmentation model (IF) - each implement different Lorentz-frame structures for the fragmentation process of quarks and gluons into hadrons. The Lund model provides a good description of the data, while the IF model does not. The Webber model, which is untuned, does not describe the absolute particle densities between jets, but correctly predicts the ratios of those densities, which are less sensitive to the tuning. We study long-range correlations between hadrons in opposite jets in e e annihilation events at $$\sqrt s $$ =29 GeV. We use the data to measure differences between quark and antiquark fragmentation functions into charged hadrons, and to estimate the variation of fragmentation properties among quark flavors. Inclusive production cross sections for photons and π's in e e annihilation at a center of mass energy of 29 GeV have been measured. The π production spectrum agrees with a corresponding measurement for π. The ratio of the π inclusive rate to the average for π is 0.92±0.14. The fractions of the total energy carried by photons and π's are 0.244±0.016 and 0.217±0.033, respectively. The fraction of total energy carried by all stable hadrons, prompt leptons and photons is determined to be 0.938±0.045, leaving 0.062±0.045 for neutrinos. [ABSTRACT FROM AUTHOR]
