The current study aimed to appraise extracts of Cleome simplicifolia (Cambess.) Hook. f. and Thomson leaves for chemical integrant and biological activities. In this study, different extracts of leaves were estimated for qualitative phytochemical screening, identification of functional groups, mineral content, and phyto-compounds and assessed for in vitro antioxidant and in vivo acute toxicity and hepatoprotective activity antagonistic toward paracetamol-intoxicated Wister albino rats. The results of the qualitative phytochemical assessment of the leaf extracts (acetone, methanol, and distilled water) exhibited the occurrence of useful metabolites. A Fourier transform infrared analysis confirmed the occurrence of O-H, N-H, C=C, S=O, C-O, C-N, C-Cl, and C-Br at 3367.14, 2920.79, 2850.32, 1631.04, 1384.59, 1168.64, 1063.78, 824.78, and 615.25 cm−1 wavelengths, whereas energy-dispersive X-ray showed the existence of carbon, oxygen, magnesium, aluminum, silicon, phosphorus, sulfur, chlorine, potassium, and calcium elements in the leaf, respectively. Thereafter, a gas chromatography–mass spectroscopy analysis unveiled the diverse volatile compounds in the methanolic leaf extracts, namely n-Heptyl acrylate—18.87%, undecane—17.49%, 2-Propenoic acid, 3-(3,4-dimethoxyphenyl)-, (E)—11.40%, Neophytadiene—11.02%, n-Hexadecanoic acid—10.78%, Glafenin—10.09%, Decane—7.45%, Phytol—6.0%, Benzene, (1-methyldodecyl)—3.48%, and 4-Cyclohexyl-1-butanol—3.41%, respectively. An analysis of in vitro antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl assay of methanolic leaf extract revealed elevated levels of antioxidant ability at 20 (46.18%), 40 (53.83%), 60 (66.64%), 80 (74.03%), and 100 (85.05%) μg/mL. In addition, in vivo acute toxicity determination proved that the methanolic leaf extract was innocuous and caused no mortality at 72 mg, 78 mg, or 82 mg/kg b.wt. doses. The methanolic leaf extracts' in vivo hepatoprotective activity against paracetamol revealed significant efficacy at 50 and 100 mg/kg b.wt. via reduction of aspartate transaminase, alanine transaminase, alkaline phosphatase, bilirubin, and cholesterol (serum blood biochemical markers) followed by an enhancement in superoxide dismutase, catalase, and glutathione (liver antioxidants) with lipid peroxidation depletion compared with the normal group rats. From the investigated study, it was concluded that the C. simplicifolia leaf are a potential source for the isolation of biologically active phyto-compounds and have the ability to prevent liver damage by paracetamol induction, where the hepatic restoration ability is indexed to its in vivo and in vitro antioxidant ability, which might be the result of its chemical constituents. Natural metabolites from beneficial fungi were recognized for their potential to inhibit multidrug-resistant human and plant fungal pathogens. The present study describes the isolation, metabolite profiling, antibacterial, and antifungal, antioxidant, and anticancer activities of soil fungi. Among the 17 isolates, the AK-7 isolate was selected based on the primary screening. Further, the identification of isolate AK-7 was performed by 18S rRNA sequencing and identified as Penicillium limosum (with 99.90% similarity). Additionally, the ethyl acetate extract of the Penicillium limosum strain AK-7 (AK-7 extract) was characterized by Fourier Transform Infrared Spectroscopy (FTIR) and a Gas Chromatography-Mass Spectroscopy (GC-MS) analysis, and the results showed different functional groups and bioactive metabolites. Consequently, a secondary screening of antibacterial activity by the agar well diffusion method showed significant antibacterial activity against Gram-negative and Gram-positive bacterial pathogens. The AK-7 extract exhibited notable antifungal activity by a food poisoning method and showed maximum inhibition of 77.84 ± 1.62%, 56.42 ± 1.27%, and 37.96 ± 1.84% against Cercospora canescens, Fusarium sambucinum and Sclerotium rolfsii phytopathogens. Consequently, the AK-7 extract showed significant antioxidant activity against DPPH and ABTS•+ free radicals with IC50 values of 59.084 μg/mL and 73.36 μg/mL. Further, the anticancer activity of the AK-7 extract against the human ovarian teratocarcinoma (PA-1) cell line was tested by MTT and Annexin V flow cytometry. The results showed a dose-dependent reduction in cell viability and exhibited apoptosis with an IC50 value of 82.04 μg/mL. The study highlights the potential of the Penicillium limosum strain AK-7 as a source of active metabolites and natural antibacterial, antifungal, antioxidant, and anticancer agent, and it could be an excellent alternative for pharmaceutical and agricultural sectors. Marine actinomycetes represent a highly favorable source of bioactive compounds and have been the mainstay of much research in recent years. Recent reports have shown that marine Streptomyces sp. can produce compounds with diverse and potent biological activities. Therefore, the key objective of the study was to isolate and screen a potential actinomycete from marine ecosystems of Devbagh and Tilmati beaches, Karwar. Streptomyces sp. KS20 was characterized and the ethyl acetate extract (EtOAc-Ex) was screened for biomedical applications. Streptomyces sp. KS20 produced grayish-white aerial and pale-yellow substrate mycelia and revealed an ancestral relationship with Streptomyces violaceusniger. Optimum growth of the organism was recorded at 30 °C and pH 7.0. The metabolite profiling of EtOAc-Ex expressed the existence of several bioactive metabolites, whereas the functional groups were indicated by Fourier transform infrared (FTIR) spectroscopy. A considerable antioxidant activity was shown for EtOAc-Ex with IC50 of 92.56 μg/mL. In addition to this, Streptomyces sp. KS20 exhibited significant antimicrobial properties, particularly against Escherichia coli, where a zone of inhibition measuring 36 ± 0.83 mm and a minimum inhibitory concentration (MIC) of 3.12 µg/mL were observed. The EtOAc-Ex even revealed significant antimycobacterial potency with IC50 of 6.25 μg/mL. Finally, the antiproliferative potentiality of EtOAc-Ex against A549 and PC-3 cell lines revealed a constant decline in cell viability while raising the concentration of EtOAc-Ex from 12.5 to 200 μg/mL. The IC50 values were determined as 94.73 μg/mL and 121.12 μg/mL for A549 and PC-3 cell lines, respectively. Overall, the exploration of secondary metabolites from marine Streptomyces sp. KS20 represents an exciting area of further research with the potential to discover novel bioactive compounds that could be developed into therapeutics for various medical applications. Green nanoparticle (NPs) synthesis is eco-friendly, non-toxic, and the NPs have demonstrated improved biocompatibility for use in healthcare. This study evaluated the biogenic synthesis of AgNPs from the leaves of Cardamine hirsuta L. and their biological properties. The UV-Vis. spectra at 411 nm exhibited a distinct resonance spectrum for C-AgNPs produced from C. hirsuta L. FT-IR analysis exhibited the presence of functional groups of phyto-compounds of C. hirsuta responsible of silver salt reduction and capping agents of C-AgNPs. The microscopic-based study, such as HR-TEM analysis, showed that the particles were uniformly distributed, spherical, and ranged in size from 5.36 to 87.65 nm. EDX analysis confirmed a silver (Ag) content of 36.3% by weight, and XRD analysis exhibited the face-centred cubic (FCC) crystalline nature of C-AgNPs. DLS measured the mean particle size of 76.5 nm. The zeta potential was significant at −27.9 mV, and TGA analysis revealed that C-AgNPs had higher thermal stability. C-AgNPs demonstrated moderate antimicrobial activity against the tested pathogens. In addition, the anti-proliferative activity measured by the MTT assay on the Caco-2 cell line demonstrated decreased cell viability with increasing C-AgNPs dosage, with an IC50 concentration of 49.14 µg/mL. In addition, an Annexin-V/Propidium iodide flow cytometric study was utilized to evaluate the induction of apoptosis in cancer cells. Early and late apoptosis cell populations increased significantly compared to the untreated control. Over past decades, the green method of synthesizing metal nanoparticles has acquired more attentiveness by scientific consensus because of its industrial and biomedical applications. This study focuses on the anti-proliferative effectiveness of AgNPs synthesized from Rotheca serrata (L.) Steane & Mabb. flower bud extract against the PANC-1 cell line in vitro. Various analytical instruments were utilized to visualize the formation of RsFb-AgNPs, such as UV-Vis spectroscopy, FT-IR, SEM, EDS, TEM, XRD, Zeta potential, and DLS analysis. The biosynthesis of RsFb-AgNPs was observed by a change in color and UV-Vis spectroscopy (415 nm). The FT-IR spectra exhibited the existence of many functional groups. XRD confirmed the crystallinity of the AgNPs. Morphology and elemental mapping were assessed by SEM and EDS analysis. The TEM micrograph revealed spherical-shaped particles with sizes ranging from 12 to 40 nm. Zeta potential and DLS analysis were used to measure surface charge and particle size. Biological properties, including the antioxidant, antimicrobial, and anticancer properties of synthesized RsFb-AgNPs, exhibited dose-dependent activities. In DPPH assay, synthesized RsFb-AgNPs inhibited the scavenging of free radicals in a dose-dependent manner. In addition, the resultant RsFb-AgNPs displayed moderate antimicrobial activity against tested pathogens. Further, the anti-proliferative efficacy of biosynthesized RsFb-AgNPs was determined against the PANC-1 cell line using the MTT assay. The results revealed a dose-dependent decrease in viability of cancer cells with an IC50 value of 36.01 µg/mL. Flow cytometry was then used to confirm the apoptotic effects by double staining with annexin V/PI. In response to the pancreatic ductal adenocarinoma cell line, the results showed notable early and late apoptosis cell population percentages. The present study describes the synthesis of silver nanoparticles (AgNPs) using Passiflora vitifolia (P. vitifolia) leaf extract as reducing and capping agent. Characterization of the synthesized AgNPs was done with the help of UV-Vis. spectroscopy, atomic force microscopy (AFM), fourier transform infrared microscopy (FTIR), zeta potential, thermo gravimetric analysis (TGA), scanning electron microscopy (SEM), transmission electron microscopy (TEM) and X-ray diffraction (XRD) analysis. The formation of AgNPs was confirmed by the presence of an absorption peak at 388 nm in UV-Vis. spectrum, poly-dispersed nature and size range from 39.06 to 58.24 nm was displayed by AFM and different functional groups were recognized by FTIR analysis. AgNPs showed stability at − 20.3 mV through zeta potential. SEM and TEM analysis revealed that AgNPs were spherical, poly-dispersed particles with minimum size of 11.61 nm and XRD analysis showed face-centered cubic and crystalline nature. The AgNPs demonstrated a notable antimicrobial efficacy against pathogenic microorganisms and showed a substantial antioxidant activity against DPPH free radicals. Further, AgNPs conveyed potent anticancer activity against murine macrophage (RAW-264.7) cell line with a significant IC 50 value of 57.63 µg/mL. Plumeria alba (P. alba) is a small laticiferous tree with promising medicinal properties. Green synthesis of nanoparticles is eco-friendly, cost-effective, and non-hazardous compared to chemical and physical synthesis methods. Current research aiming to synthesize silver nanoparticles (AgNPs) from the leaf extract of P. alba (P- AgNPs) has described its physiochemical and pharmacological properties in recognition of its therapeutic potential as an anticancer and antimicrobial agent. These biogenic synthesized P-AgNPs were physiochemically characterized by ultraviolet-visible spectroscopy, Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), transmission electron microscope (TEM), atomic force microscopy (AFM), X-ray diffractometry (XRD), and zeta potential analysis. Antimicrobial activity was investigated against Escherichia coli, Pseudomonas aeruginosa, Enterobacter aerogenes, Enterococcus faecalis, Bacillus subtilis, Streptococcus pneumoniae, Candida albicans, and Candida glabrata. Anticancer activity against glioblastoma U118 MG cancer lines was investigated using an MTT assay, and apoptosis activity was determined by flow cytometry. UV–visible spectroscopic analysis portrayed surface plasmon resonance at 403 nm of synthesized P-AgNPs, and FTIR suggested the presence of amines, alkanes, and phenol molecules that could be involved in reduction and capping processes during AgNPs formation. Synthesized particles were spherical in shape and poly-dispersed with an average particle size of 26.43 nm and a poly-dispersity index (PDI) of 0.25 with a zeta potential value of −24.6 mV, ensuring their stability. The lattice plane values confirm the crystalline nature as identified by XRD. These P-AgNPs exhibited potential antimicrobial activity against selected human pathogenic microbes. Additionally, the in vitro MTT assay results showed its effective anticancer activity against the glioma U118 MG cancer cell line with an IC50 value of 9.77 µg/mL AgNPs by initiating apoptosis as identified by a staining study with flow cytometric Annexin V–Fluorescein Isothiocyanate (FITC) and Propidium Iodide (PI). Thus, P. alba AgNPs can be recommended for further pharmacological and other biological research. Rhizospheric soil is the richest niche of different microbes that produce biologically active metabolites. The current study investigated the antimicrobial, antifungal and anticancer activities of ethyl acetate extract of the potent rhizospheric fungus Aspergillus niger AK6 (AK-6). A total of six fungal isolates were isolated, and isolate AK-6 was selected based on primary screening. Further, it exhibited moderate antimicrobial activity against pathogens such as Klebsiella pneumonia , Candida albicans , Escherichia coli , Shigella flexneri , Bacillus subtilis and Staphylococcus aureus . The morphological and molecular characterization (18S rRNA) confirmed that the isolate AK-6 belonged to Aspergillus niger. Further, AK-6 showed potent antifungal activity with 47.2%, 59.4% and 64.1% of inhibition against Sclerotium rolfsii , Cercospora canescens and Fusarium sambucinum phytopathogens. FT-IR analysis displayed different biological functional groups. Consequently, the GC-MS analysis displayed bioactive compounds, namely, n-didehydrohexacarboxyl-2,4,5-trimethylpiperazine (23.82%), dibutyl phthalate (14.65%), e-5-heptadecanol (8.98%), and 2,4-ditert-butylphenol (8.60%), among the total of 15 compounds isolated. Further, the anticancer activity of AK-6 was exhibited against the MCF-7 cell line of human breast adenocarcinoma with an IC 50 value of 102.01 μg/mL. Furthermore, flow cytometry depicted 17.3%, 26.43%, and 3.16% of early and late apoptosis and necrosis in the AK-6 extarct treated MCF-7 cell line, respectively. The results of the present analysis suggest that the isolated Aspergillus niger strain AK-6 extract has the potential to be explored as a promising antimicrobial, antifungal and anticancer drug for medical and agricultural applications.
