The conformation of a polyelectrolyte (PE) brush grafted to the inner surface of a long cylindrical mesopore was described within analytical Poisson–Boltzmann strong stretching approximation. The internal structure of the PE brush, including brush thickness and radial density profile of monomer units, and radial distribution of electrostatic potential were analyzed as functions of the pore radius, degree of polymerization, and grafting density of the brush-forming PE chains as well as ionic strength of the solution. It is demonstrated that narrowing of the pore leads to a non-monotonous variation of the brush thickness, which passes through a maximum when the brush thickness becomes equal to the pore radius. Variation in the salt concentration triggers conformational transition that leads to the opening or closing of the hollow (PE-free) channel in the pore center that potentially allows controlling of the pore-selective permeability for charged nanocolloidal particles (e.g., globular proteins or viruses). The predictions of the analytical theory were validated by numerical calculations using the Scheutjens–Fleer self-consistent field modeling method. These theoretical findings may be used for the design of highly selective smart mesoporous membranes with PE brush-functionalized pores for, e.g., protein separation and purification. We investigate the effect of polydispersity on the properties of curved linear brushes in good solvent and for molten brushes. To this end, we extend the strong stretching theory for polydisperse brushes to curved geometries and investigate the polymer chain end profiles, bending moduli and other properties for experimentally relevant polymer chain length distributions of the Schulz–Zimm type. We also investigate the properties of end exclusion zones that may appear in convex geometries under certain conditions and show that their position in the brush can be engineered by careful selection of the polymer length distribution. A large number of experimental studies have demonstrated that globular proteins can be absorbed from the solution by both polycationic and polyanionic brushes when the net charge of protein globules is of the same or of the opposite sign with respect to that of brush-forming polyelectrolyte chains. Here, we overview the results of experimental studies on interactions between globular proteins and polycationic or polyanionic brushes, and present a self-consistent field theoretical model that allows us to account for the asymmetry of interactions of protein-like nanocolloid particles comprising weak (pH-sensitive) cationic and anionic groups with a positively or negatively charged polyelectrolyte brush. The position-dependent insertion free energy and the net charge of the particle are calculated. The theoretical model predicts that if the numbers of cationic and anionic ionizable groups of the protein are approximately equal, then the interaction patterns for both cationic and anionic brushes at equal offset on the "wrong side" from the isoelectric point (IEP), i.e., when the particle and the brush charge are of the same sign, are similar. An essential asymmetry in interactions of particles with polycationic and polyanionic brushes is predicted when fractions of cationic and anionic groups differ significantly. That is, at a pH above IEP, the anionic brush better absorbs negatively charged particles with a larger fraction of ionizable cationic groups and vice versa. [ABSTRACT FROM AUTHOR]

This feature article is devoted to the evaluation of different techniques for producing colloidal polyelectrolyte brushes (CPEBs) based on cellulose nanofibers modified with grafted polyacrylates. The paper also reviews the potential applications of these CPEBs in designing electrode materials and as reinforcing additives. Additionally, we discuss our own perspectives on investigating composites with CPEBs. Herein, polyacrylic acid (PAA) was grafted onto the surface of cellulose nanofibers (CNFs) employing a "grafting from" approach. The effect of the PAA shell on the morphological structure of a composite with polypyrrole (PPy) was investigated. The performance of as-obtained CNF-PAA/PPy as organic electrode material for supercapacitors was examined. Furthermore, this research highlights the ability of CNF-PAA filler to act as an additional crosslinker forming a physical sub-network due to the hydrogen bond interaction inside chemically crosslinked polyacrylamide (PAAm) hydrogels. The enhancement of the mechanical properties of the material with a concomitant decrease in its swelling ratio compared to a pristine PAAm hydrogel was observed. The morphology of superstructures formed by bottlebrush block copolymers in the melt can be tuned by changing the side chain length or/and their grafting density at constant volume fractions of the blocks. This feature enables fabrication of microphase separated bulk structures and mesoporous materials thereof with spherical or cylindrical domains (precursors of the mesopores), with high porosity unattainable for materials produced from conventional linear block copolymers. These paradigms are proven by DPD simulations that allow constructing morphological phase diagrams of the melt of block copolymers comprising one linear and one bottlebrush block and comparing the simulation results to the predictions of the mean field analytical theory. While the binodal lines separating the stability regions of spherical and cylindrical domains predicted by the theory perfectly match the simulation results, the simulation indicates appearance of a gyroid phase around the theoretical binodal separating the stability ranges of cylinders and lamellae. Purpose: To replicate previous genome-wide association study identified sprint-related polymorphisms in 3 different cohorts of top-level sprinters and to further validate the obtained results in functional studies. Methods: A total of 240 Japanese, 290 Russians, and 593 Brazilians were evaluated in a case-control approach. Of these, 267 were top-level sprint/power athletes. In addition, the relationship between selected polymorphisms and muscle fiber composition was evaluated in 203 Japanese and 287 Finnish individuals. Results: The G allele of the rs3213537 polymorphism was overrepresented in Japanese (odds ratio [OR]: 2.07, P =.024) and Russian (OR: 1.93, P =.027) sprinters compared with endurance athletes and was associated with an increased proportion of fast-twitch muscle fibers in Japanese (P =.02) and Finnish (P =.041) individuals. A meta-analysis of the data from 4 athlete cohorts confirmed that the presence of the G/G genotype rather than the G/A+A/A genotypes increased the OR of being a sprinter compared with controls (OR: 1.49, P =.01), endurance athletes (OR: 1.79, P =.001), or controls + endurance athletes (OR: 1.58, P =.002). Furthermore, male sprinters with the G/G genotype were found to have significantly faster personal times in the 100-m dash than those with G/A+A/A genotypes (10.50 [0.26] vs 10.76 [0.31], P =.014). Conclusion: The rs3213537 polymorphism found in the CPNE5 gene was identified as a highly replicable variant associated with sprinting ability and the increased proportion of fast-twitch muscle fibers, in which the homozygous genotype for the major allele (ie, the G/G genotype) is preferable for performance. Using a combination of self-consistent field theory with coarse-grained Brownian dynamics simulations, we studied mechanical unfolding of an amphiphilic molecular brush in a solvent that was poor for the side chains but good for the main chain of the brush. In striking contrast to earlier theoretical predictions [O. V. Borisov and A. Halperin, Europhys. Lett. 34, 657 (1996)] made for amphiphilic graft copolymers, we discovered that in a partially unfolded molecular brush, a globular core co-exists, at equilibrium, with a necklace of smaller sized intra-molecular micelles, each comprising a few side chains connected by stretched main chain strands (bridges). A progressive increase in the main chain end-to-end distance of the brush triggers a sequence of discrete events of generation of new intramolecular micelles, giving rise to a sawtooth pattern in the restoring force–deformation curve. Polymer brushes are attractive as surface coatings for a wide range of applications, from fundamental research to everyday life, and also play important roles in biological systems. How colloids (e.g., functional nanoparticles, proteins, viruses) bind and move across polymer brushes is an important yet under‐studied problem. A mean‐field theoretical approach is presented to analyze the binding and transport of colloids in planar polymer brushes. The theory explicitly considers the effect of solvent strength on brush conformation and of colloid‐polymer affinity on colloid binding and transport. The position‐dependent free energy of the colloid insertion into the polymer brush which controls the rate of colloid transport across the brush is derived. It is shown how the properties of the brush can be adjusted for brushes to be highly selective, effectively serving as tuneable gates with respect to colloid size and affinity to the brush‐forming polymer. The most important parameter regime simultaneously allowing for high brush permeability and selectivity corresponds to a condition when the repulsive and attractive contributions to the colloid insertion free energy nearly cancel. This theory should be useful to design sensing and purification devices with enhanced selectivity and to better understand mechanisms underpinning the functions of biological polymer brushes. We considered dispersions of cylindrical polyelectrolyte (PE) brushes with stiff backbones, and polymer-decorated nanorods with tunable solubility of the brush-forming PE chains that affected thermodynamic stability of the dispersions. We focused on thermo-induced and deionization-induced conformational transition that provokes loss of aggregative dispersion stability of nanorods decorated with weakly ionized polyions. Brushes formed by arm-tethered starlike polyelectrolytes may exhibit internal segregation into weakly and strongly extended populations (stratified two-layer structure) when strong ionic intermolecular repulsions induce stretching of the tethers up to the limit of their extensibility. We propose an approximate Poisson-Boltzmann theory for analysis of the structure of the stratified brush and compare it with results of numerical self-consistent field modeling. Both analytical and numerical models point to the formation of a narrow cloud of counterions (internal double electrical layer) localized inside a stratified brush at the boundary between the layers. We apply a coarse-grained self-consistent field Poisson-Boltzmann framework to study interaction between Bovine Serum Albumin (BSA) and a planar polyelectropyte brush. Both cases of negatively (polyanionic) and positively (polycationic) charged brushes are considered. Our theoretical model accounts for (1) re-ionization free energy of the amino acid residues upon protein insertion into the brush; (2) osmotic force repelling the protein globule from the brush; (3) hydrophobic interactions between non-polar areas on the globule surface and the brush-forming chains. We demonstrate that calculated position-dependent insertion free energy exhibits different patterns, corresponding to either thermodynamically favourable BSA absorption in the brush or thermodynamically or kinetically hindered absorption (expulsion) depending on the p H and ionic strength of the solution. The theory predicts that due to the re-ionization of BSA within the brush, a polyanionic brush can efficiently absorb BSA over a wider p H range on the "wrong side" of the isoelectric point (IEP) compared to a polycationic brush. The results of our theoretical analysis correlate with available experimental data and thus validate the developed model for prediction of the interaction patterns for various globular proteins with polyelectrolyte brushes. The design of nanoassemblies can be conveniently achieved by tuning the strength of the hydrophobic interactions of block copolymers in selective solvents. These block copolymer micelles form supramolecular aggregates, which have attracted great attention in the area of drug delivery and imaging in biomedicine due to their easy-to-tune properties and straightforward large-scale production. In the present work, we have investigated the micellization process of linear–dendritic block copolymers in order to elucidate the effect of branching on the micellar properties. We focus on block copolymers formed by linear hydrophobic blocks attached to either dendritic neutral or charged hydrophilic blocks. We have implemented a simple protocol for determining the equilibrium micellar size, which permits the study of linear–dendritic block copolymers in a wide range of block morphologies in an efficient and parallelizable manner. We have explored the impact of different topological and charge properties of the hydrophilic blocks on the equilibrium micellar properties and compared them to predictions from self-consistent field theory and scaling theory. We have found that, at higher degrees of branching in the corona and for short polymer chains, excluded volume interactions strongly influence the micellar aggregation as well as their effective charge. In this article, we used the numerical self-consistent field method of Scheutjens–Fleer to study the micellization of hybrid molecules consisting of one polylysine dendron with charged end groups and several linear hydrophobic tails attached to its root. The main attention was paid to spherical micelles and the determination of the range of parameters at which they can appear. A relationship has been established between the size and internal structure of the resulting spherical micelles and the length and number of hydrophobic tails, as well as the number of dendron generations. It is shown that the splitting of the same number of hydrophobic monomers from one long tail into several short tails leads to a decrease in the aggregation number and, accordingly, the number of terminal charges in micelles. At the same time, it was shown that the surface area per dendron does not depend on the number of hydrophobic monomers or tails in the hybrid molecule. The relationship between the structure of hybrid molecules and the electrostatic properties of the resulting micelles has also been studied. It is found that the charge distribution in the corona depends on the number of dendron generations G in the hybrid molecule. For a small number of generations (up to G = 3 ), a standard double electric layer is observed. For a larger number of generations ( G = 4 ), the charges of dendrons in the corona are divided into two populations: in the first population, the charges are in the spherical layer near the boundary between the micelle core and shell, and in the second population, the charges are near the periphery of the spherical shell. As a result, a part of the counterions is localized in the wide region between them. We present a mean-field theory for the solution properties of polyelectrolyte molecular brushes, comprising multiple ionically charged side chains tethered to the main chain (backbone). The power-law dependences for local and large-scale conformational properties, i.e., the brush thickness, extension of spacers between the grafts, and end-to-end distance for the main chain macromolecule, are derived as a function of architectural parameters (the lengths of the main chain and of the grafts, grafting density). We demonstrate that at high grafting density or/and a large fraction of charged monomer units in the grafts, the localization of counterions in the intra-molecular volume occurs and we specify the onset of this transition. We prove that such localization of the counterions is accompanied by full extension of the main chain of the polyelectrolyte molecular brush in salt-free solution. This stretching of the main chain is relaxed upon an increase in the salt concentration. The dependence of the macromolecular dimensions of the polyelectrolyte molecular brush on the salt concentration is derived, and multiple power-law exponents valid in different salt concentration ranges are predicted. We present results of MD and MC simulations of the equilibrium properties of swelling gels with comb-like or bottlebrush subchains and compare them to scaling-theory predictions. In accordance with theory, the simulation results demonstrate that swelling coefficient of the gel increases as a function of the polymerization degree of the main chains and exhibits a very weak maximum (or is virtually constant) as a function of the polymerization degree and grafting density of side chains. The bulk osmotic modulus passes through a shallow minimum as the polymerization degree of the side chains increases. This minimum is attributed to the onset of overlap of side chains belonging to different bottlebrush strands in the swollen gel. [ABSTRACT FROM AUTHOR]

Recombinant tissue plasminogen activator (rt-PA) is a well-characterized glycoprotein with a great deal of published information on its structure, post-translational modifications, and O- and N-glycosylation. Most of the characterization was accomplished in the late 1980s. During the past 2 decades, however, mass spectrometry has made a quantum leap forward offering new capabilities in soft electrospray ionization, speed, resolution, and accuracy of mass measurements. From this point of view, it is worthwhile to revisit the characterization of familiar proteins, such as rt-PA, using the new capabilities of modern analytical technology. In this work, we applied LC-MS with state-of-the-art instrumentation to the characterization of glycoforms of rt-PA. This method takes advantage of accurate mass measurements along with a fast 'in-source' voltage switching for the detection of characteristic oxonium ions of saccharides. This method confirmed previously identified glycan structures based on existing knowledge of rt-PA glycans. In addition, we identified two novel glycan structures in rt-PA. A low level of [Asn.sup.142] N-glycosylation was detected at an atypical Asn-Xaa-Cys consensus motif. It was found to be modified predominantly by biantennary hybrid structures. This N-glycosylation site was confirmed using a recently developed electron-transfer dissociation (ETD) technique. Also using this method, we detected low levels of elongation of fucose-O-[Thr.sup.61] to di-, tri-, and tetrasaccharides, not previously observed in rt-PA. The results demonstrate that use of state-of-the-art analytical methods can reveal low-level, previously undetected modifications of well-characterized biopharmaceuticals. 10.1021/ac901498k

Translation errors in protein biosynthesis may result in low level amino acid misincorporation and contribute to product heterogeneity of recombinant protein therapeutics. We report the use of peptide map analysis by reversed-phase high-performance liquid chromatography and high-resolution mass spectrometry to detect and identify mistranslation events in recombinant monoclonal antibodies expressed in mammalian cell lines including Chinese hamster ovary (CHO) cells. Misincorporation of an asparagine residue at multiple serine positions was detected as earlier-eluting peptides with masses 27.01 Da higher than expected. The exact positions at which misincorporation occurred were identified by tandem mass spectrometry of the asparagine-containing variant peptides. The identified asparagine misineorporation sites correlated with the use of codon AGC but with none of the other five serine codons. The relative levels of misincorporation ranged from 0.01%-0.2% among multiple serine positions detected across three different antibodies by targeted analysis of expected and variant peptides. The low levels of misincorporation are consistent with published predictions for in vivo translation error rates. Our results demonstrate that state-of-the-art mass spectrometry with a combination of high sensitivity, accuracy, and dynamic range provides a new ability to discover and characterize low level protein variants that arise from mistranslation events. 10.1021/ac901541h

So far, more than 200 DNA polymorphisms associated with strength/power phenotypes have been identified majorly involving nonathletic populations. The aim of the present study was to investigate individually and in combination the association of 217 DNA polymorphisms previously identified as markers for strength/power phenotypes with elite strength athlete status. A case-control study involved 83 Russian professional strength athletes (53 weightlifters, 30 powerlifters), 209 Russian and 503 European controls. Genotyping was conducted using micro-array analysis. Twenty-eight DNA polymorphisms (located near or in ABHD17C , ACTG1 , ADCY3 , ADPGK , ANGPT2 , ARPP21 , BCDIN3D , CRTAC1 , DHODH , GBE1 , IGF1 , IL6 , ITPR1 , KIF1B , LRPPRC , MMS22L , MTHFR , NPIPB6 , PHACTR1 , PLEKHB1 , PPARG , PPARGC1A , R3HDM1 , RASGRF1 , RMC1 , SLC39A8 , TFAP2D , ZKSCAN5 genes) were identified to have an association with strength athlete status. Next, to assess the combined impact of all 28 DNA polymorphisms, all athletes were classified according to the number of "strength" alleles they possessed. All highly elite strength athletes were carriers of at least 22 (up to 34) "strength" alleles, whereas 27.8% of Russian controls had less than 22 "strength" alleles (p < 0.0001). The proportion of subjects with a high (≥26) number of "strength" alleles was significantly greater in highly elite strength athletes (84.8%) compared with less successful strength athletes (64.9%; odd ratio [OR] = 3.0, p = 0.042), Russian (26.3%; OR = 15.6, p < 0.0001) or European (37.8%; OR = 6.4, p < 0.0001) controls. The interaction of colloidal particles with a planar polymer brush immersed in a solvent of variable thermodynamic quality is studied by a numerical self-consistent field method combined with analytical mean-field theory. The effect of embedded particle on the distribution of polymer density in the brush is analyzed and the particle insertion free energy profiles are calculated for variable size and shape of the particles and sets of polymer–particle and polymer–solvent interaction parameters. In particular, both cases of repulsive and attractive interactions between particles and brush-forming chains are considered. It is demonstrated that for large particles the insertion free energy is dominated by repulsive (osmotic) contribution and is approximately proportional to the particle volume in accordance with earlier theoretical predictions [Halperin et al., Macromolecules, 2011, 44, 3622]. For the particles of smaller size or/and large shape asymmetry the adsorption or depletion of a polymer from the particle surface essentially contributes to the insertion free energy balance. As a result, depending on the set of polymer–solvent and polymer–particle interaction parameters and brush grafting density the insertion free energy profile may exhibit complex patterns, i.e., from a pure repulsive effective potential barrier to an attractive well. The results of our study allow for predicting equilibrium partitioning and controlling diffusive transport of (bio)nanocolloids across (bio)polymer brushes of arbitrary geometry including polymer-modified membranes or nanopores.
