The distribution of gas and metals in the circumgalactic medium (CGM) plays a critical role in how galaxies evolve. The MUSE-ALMA Haloes survey combines MUSE, ALMA, and HST observations to constrain the properties of the multiphase gas in the CGM and the galaxies associated with the gas probed in absorption. In this paper, we analyse the properties of galaxies associated with 32 strong |${\rm H\, {\small I}}$| Ly-α absorbers at redshift 0.2 ≲ z ≲ 1.4. We detect 79 galaxies within ±500 kms−1 of the absorbers in our 19 MUSE fields. These associated galaxies are found at physical distances from 5.7 kpc and reach star formation rates as low as 0.1 M⊙ yr−1. The significant number of associated galaxies allows us to map their physical distribution on the Δ v and b plane. Building on previous studies, we examine the physical and nebular properties of these associated galaxies and find the following: (i) 27/32 absorbers have galaxy counterparts and more than 50 per cent of the absorbers have two or more associated galaxies, (ii) the |${\rm H\, {\small I}}$| column density of absorbers is anticorrelated with the impact parameter (scaled by virial radius) of the nearest galaxy as expected from simulations, (iii) the metallicity of associated galaxies is typically larger than the absorber metallicity, which decreases at larger impact parameters. It becomes clear that while strong |${\rm H\, {\small I}}$| absorbers are typically associated with more than a single galaxy, we can use them to statistically map the gas and metal distribution in the CGM. The gas cycling in the circumgalactic regions of galaxies is known to be multi-phase. The MUSE–ALMA Haloes survey gathers a large multi-wavelength observational sample of absorption and emission data with the goal to significantly advance our understanding of the physical properties of such CGM gas. A key component of the MUSE–ALMA Haloes survey is the multi-facility observational campaign conducted with VLT/MUSE, ALMA, and HST. MUSE–ALMA Haloes targets comprise 19 VLT/MUSE IFS quasar fields, including 32 z abs <0.85 strong absorbers with measured N (H  i) ≥1018 cm−2 from UV-spectroscopy. We additionally use a new complementary HST medium program to characterize the stellar content of the galaxies through a 40-orbit three-band UVIS and IR WFC3 imaging. Beyond the absorber-selected targets, we detect 3658 sources all fields combined, including 703 objects with spectroscopic redshifts. This galaxy-selected sample constitutes the main focus of the current paper. We have secured millimeter ALMA observations of some of the fields to probe the molecular gas properties of these objects. Here, we present the overall survey science goals, target selection, observational strategy, data processing and source identification of the full sample. Furthermore, we provide catalogues of magnitude measurements for all objects detected in VLT/MUSE, ALMA, and HST broad-band images and associated spectroscopic redshifts derived from VLT/MUSE observations. Together, this data set provides robust characterization of the neutral atomic gas, molecular gas and stars in the same objects resulting in the baryon census of condensed matter in complex galaxy structures. We simulate the flux emitted from galaxy haloes in order to quantify the brightness of the circumgalactic medium (CGM). We use dedicated zoom-in cosmological simulations with the hydrodynamical adaptive mesh refinement code ramses , which are evolved down to z  = 0 and reach a maximum spatial resolution of 380 h −1 pc and a gas mass resolution up to |$1.8\times 10^{5} \, h^{-1}\, \rm {M}_{\odot }$| in the densest regions. We compute the expected emission from the gas in the CGM using cloudy emissivity models for different lines (e.g. Lyα, C  iv , O  vi , C  vi , O  viii) considering UV background fluorescence, gravitational cooling and continuum emission. In the case of Lyα, we additionally consider the scattering of continuum photons. We compare our predictions to current observations and find them to be in good agreement at any redshift after adjusting the Lyα escape fraction. We combine our mock observations with instrument models for Faint Intergalactic Redshifted Emission Balloon-2 (FIREBall-2; UV balloon spectrograph) and HARMONI (visible and NIR IFU on the ELT) to predict CGM observations with either instrument and optimize target selections and observing strategies. Our results show that Lyα emission from the CGM at a redshift of 0.7 will be observable with FIREBall-2 for bright galaxies (NUV∼18 mag), while metal lines like O  vi and C  iv will remain challenging to detect. We now have mounting evidences that the circumgalactic medium (CGM) of galaxies is polluted with metals processed through stars. The fate of these metals is however still an open question and several findings indicate that they remain poorly mixed. A powerful tool to study the low-density gas of the CGM is offered by absorption lines in quasar spectra, although the information retrieved is limited to 1D along the sightline. We report the serendipitous discovery of two close-by bright z gal = 1.148 extended galaxies with a fortuitous intervening z abs = 1.067 foreground absorber. MUSE IFU observations spatially probes kpc-scales in absorption in the plane of the sky over a total area spanning ∼30 kpc−2. We identify two [O  ii ] emitters at z abs down to 21 kpc with SFR ∼ 2 M⊙ yr−1. We measure small fractional variations (<30 per cent) in the equivalent widths of Fe  ii and Mg  ii cold gas absorbers on coherence scales of 8 kpc but stronger variation on larger scales (25 kpc). We compute the corresponding cloud gas mass <2 × 109 M⊙. Our results indicate a good efficiency of the metal mixing on kpc-scales in the CGM of a typical z ∼ 1 galaxy. Objectives: Advanced ERCP techniques (AETs) for difficult biliary stones include peroral cholangioscopy (POC) with electrohydraulic/laser lithotripsy (EHL/LL), endoscopic papillary large balloon dilation (EPLBD) and mechanical lithotripsy (ML). We assess the efficacy of AETs. Methods: A retrospective query for AETs. Primary outcome: Complete duct clearance. Secondary outcome: Complete duct clearance by technique. Statistical Analysis version 9.3 (SAS Inc., Cary, NC). Results: From 1/00 to 10/12, 349 patients were identified of which 222 (80% had prior ERCPs) had AETs. 211 with sufficient follow-up underwent 295 ERCPs; 280 of which were AET’s. Index AETs: POC with EHL/LL (n = 46/211, 22%), ML (n = 84/211, 40%), EPLBD with mean balloon size of 11.5 ± 1.7 mm (n = 39/211, 18%) and combination AETs (n = 42/211, 20%). Stone characteristics: 76% had ≥1 stone, 81% extrahepatic and 32% had strictures. Number of stones (mean 2.5 and range 1-20) did not differ among groups. EPLBD had higher percentage (95%) of extrahepatic stones (p = .0003). The ‘Combination’ and ‘POC’ groups had larger stones (mean 17.7 mm ±6.4 and 16.8 mm ±6.1, respectively; p < .001). Complete clearance: 209/211 (99%) at index AET 167/211 (79%) or after mean of 2.5 ± 0.7 AETs in 42/211 (20%). Partial clearance: 2/211 (1%). Clearance at index AETs was higher with EPLBD (90%, p = .014). Adverse Events: 7/280 (2.5%). Conclusions: AETs achieved clearance in 99%. EPLBD had higher clearance at index AET likely owing to higher extrahepatic stones. Larger stones, but not number, were associated with increased combination AETs and total ERCPs. Background In patients with chronic pancreatitis, laser lithotripsy (LL) permits stone fragmentation and removal during ERCP with some advantages over extracorporeal shock-wave lithotripsy (ESWL) and surgery. Objectives To evaluate the technical success of LL in pancreatic duct (PD) stones. Design Retrospective cohort. Setting Four tertiary referral centers. Patients Patients undergoing endotherapy for PD stones. Interventions ERCP with per-oral pancreatoscopy (POP)-guided LL. Main Outcome Measurement Technical success was defined as complete stone clearance. Results Over 3 years, 28 patients (16 men, 51 years [mean age]) underwent a median of 1 (range, 1-4) POP-LL for PD stones. Baseline parameters included pain requiring hospitalization (n = 19, 68%), opiate use (n = 14, 50%), or weight loss (n = 11, 39%). Before POP-LL, 22 of 28 patients (79%) had a median of 1 (range, 1-5) ERCP, 9 of 28 (32%) underwent a median of 2 (range, 1-3) ESWL sessions, and 5 underwent a median of 1 (range, 1-3) POP-guided electrohydraulic lithotripsy with failed (n = 2) or partial (n = 3) fragmentation. A median of 2 (range, 1-3) stones sized 15 mm (range, 4-32 mm) were identified in the head (n = 9, 32%), neck (n = 3, 11%), body (n = 9, 32%), tail (n = 1, 4%), or multiple sites (n = 6, 21%). Technical success occurred in 22 patients (79%) with complete clearance. Partial clearance occurred in 3 (11%). Clinical success at a median of 13 (range, 1-25) months of follow-up was noted in 25 of 28 patients (89%) by improvement in pain (n = 25), decreased narcotic use (n = 25), or reduced hospitalizations (n = 19). Mild adverse events occurred in 8 of 28 (29%). Conclusions POP-LL is feasible at expert centers in patients with accessible stones. Loss of parkin function is responsible for the majority of autosomal recessive parkinsonism. Here, we show that parkin is not only a stress-protective, but also a stress-inducible protein. Both mitochondrial and endoplasmic reticulum (ER) stress induce an increase in parkin-specific mRNA and protein levels. The stress-induced upregulation of parkin is mediated by ATF4, a transcription factor of the unfolded protein response (UPR) that binds to a specific CREB/ATF site within the parkin promoter. Interestingly, c-Jun can bind to the same site, but acts as a transcriptional repressor of parkin gene expression. We also present evidence that mitochondrial damage can induce ER stress, leading to the activation of the UPR, and thereby to an upregulation of parkin expression. Vice versa, ER stress results in mitochondrial damage, which can be prevented by parkin. Notably, the activity of parkin to protect cells from stress-induced cell death is independent of the proteasome, indicating that proteasomal degradation of parkin substrates cannot explain the cytoprotective activity of parkin. Our study supports the notion that parkin has a role in the interorganellar crosstalk between the ER and mitochondria to promote cell survival under stress, suggesting that both ER and mitochondrial stress can contribute to the pathogenesis of Parkinson's disease. In vivo electroporation is used to study gene regulation and gene function in the freshwater polyp Hydra. Although this approach has been used successfully by several investigators, efficacy and handling continue to present a problem. Here we show technical aspects of in vivo electroporation for introducing fluorescent dyes, plasmid DNA and double stranded RNA into Hydra polyps. We describe the fundamentals of the electroporation delivery system, discuss recent studies where this approach has been used successfully, compare it to alternative transfection methods such as lipofection, and identify future directions. This review is a research update of recent literature related to childhood-onset schizophrenia (onset of psychotic symptoms by age 12 years). This subgroup of patients has attracted considerable research interest because patients with a childhood onset may represent a more homogeneous patient population in which to search for risk or etiologic factors. We examine data indicating that childhood-onset schizophrenia (COS) shares the same clinical and neurobiologic features as later-onset forms of the disorder. Compared with adults with schizophrenia, however, this subgroup of patients appears to have more severe premorbid neuro-developmental abnormalities, more cytogenetic anomalies, and potentially greater family histories of schizophrenia and associated spectrum disorders. While preliminary, these data indicate that a greater genetic vulnerability may be one of the underpinnings of COS. Future studies of this subgroup may provide important clues as to the genetic basis for schizophrenia and how gene products influence certain features of the disease, such as age of onset and mode of inheritance.
