Glucocorticoids mediate immunosuppression Immunosuppression is a prominent feature of cancer development, restraining antitumor effector T cell responses and limiting efficacy of therapeutic interventions. Immunosuppression can be mediated by multiple mechanisms, [...], Suppression of antitumor immunity is a prominent feature of the tumor microenvironment. In this issue of the JCI, Taves, Otsuka, and authors show that glucocorticoids (GCs), which are potent immunosuppressive hormones mainly produced by the adrenals, can be reconverted from their inactive form to active metabolites via the 11[beta]-hydroxysteroid dehydrogenase type 1 (11[beta]-HSD1) enzyme expressed by murine tumor cell lines. In the tumor microenvironment, GCs acted on CD[4.sup.+] regulatory T cells to enhance their immunosuppressive function and promote tumor growth. The findings suggest that targeting GC recycling as a strategy for modulating tumor immunosuppression has the potential to improve therapeutic efficacy of immune checkpoint blockade.

Abstract

Introduction During chronic viral infection and cancer, tumor antigen-specific (TA-specific) [CD8.sup.+] T cells become dysfunctional and upregulate multiple inhibitory receptors, including programmed cell death 1/programmed death ligand 1 (PD-1/PD-L1) and [...], T cell immunoglobulin mucin domain-containing protein 3 (Tim-3) negatively regulates innate and adaptive immunity in cancer. To identify the mechanisms of Tim-3 in cancer immunity, we evaluated the effects of Tim-3 blockade in human and mouse melanoma. Here, we show that human programmed cell death 1-positive ([PD-1.sup.+]) [Tim-3.sup.+][CD8.sup.+] tumor-infiltrating lymphocytes (TILs) upregulate phosphatidylserine (PS), a receptor for Tim-3, and acquire cell surface myeloid markers from antigen-presenting cells (APCs) through transfer of membrane fragments called trogocytosis. Tim-3 blockade acted on [Tim-3.sup.+] APCs in a PS-dependent fashion to disrupt the trogocytosis of activated tumor antigen-specific [CD8.sup.+] T cells and [PD-1.sup.+][Tim-3.sup.+] [CD8.sup.+] TILs isolated from patients with melanoma. Tim-3 and PD-1 blockades cooperated to disrupt trogocytosis of [CD8.sup.+] TILs in 2 melanoma mouse models, decreasing tumor burden and prolonging survival. Deleting Tim-3 in dendritic cells but not in [CD8.sup.+] T cells impeded the trogocytosis of [CD8.sup.+] TILs in vivo. Trogocytosed [CD8.sup.+] T cells presented tumor peptide-major histocompatibility complexes and became the target of fratricide T cell killing, which was reversed by Tim-3 blockade. Our findings have uncovered a mechanism Tim-3 uses to limit antitumor immunity.

Abstract

LAG-3, TIM-3, and TIGIT comprise the next generation of immune checkpoint receptors being harnessed in the clinic. Although initially studied for their roles in restraining T cell responses, intense investigation over the last several years has started to pinpoint the unique functions of these molecules in other immune cell types. Understanding the distinct processes that these receptors regulate across immune cells and tissues will inform the clinical development and application of therapies that either antagonize or agonize these receptors, as well as the profile of potential tissue toxicity associated with their targeting. Here, we discuss the distinct functions of LAG-3, TIM-3, and TIGIT, including their contributions to the regulation of immune cells beyond T cells, their roles in disease, and the implications for their targeting in the clinic. LAG-3, TIM-3, and TIGIT comprise the next generation of immune checkpoint receptors being harnessed in the clinic. In this review, Joller et al. review advances in our understanding of the specialized functions of LAG-3, TIM-3, and TIGIT, focusing on their roles in immune cells beyond T cells and how this knowledge can be harnessed for rational development of therapies that maximize efficacy and mitigate immune toxicity. Abstract

Introduction T cell responses are controlled by multiple receptors: while costimulatory receptors ensure optimal T cell activation and proliferation to generate a protective immune response, coinhibitory or checkpoint receptors dampen [...], Coinhibitory receptors are critical for the maintenance of immune homeostasis. Upregulation of these receptors on effector T cells terminates T cell responses, while their expression on Tregs promotes their suppressor function. Understanding the function of coinhibitory receptors in effector T cells and Tregs is crucial, as therapies that target coinhibitory receptors are currently at the forefront of treatment strategies for cancer and other chronic diseases. T cell Ig and ITIM domain (TIGIT) is a recently identified coinhibitory receptor that is found on the surface of a variety of lymphoid cells, and its role in immune regulation is just beginning to be elucidated. We examined TIGIT-mediated immune regulation in different murine cancer models and determined that TIGIT marks the most dysfunctional subset of [CD8.sup.+] T cells in tumor tissue as well as tumortissue Tregs with a highly active and suppressive phenotype. We demonstrated that TIGIT signaling in Tregs directs their phenotype and that TIGIT primarily suppresses antitumor immunity via Tregs and not [CD8.sup.+] T cells. Moreover, [TIGIT.sup.+] Tregs upregulated expression of the coinhibitory receptor TIM-3 in tumor tissue, and TIM-3 and TIGIT synergized to suppress antitumor immune responses. Our findings provide mechanistic insight into how TIGIT regulates immune responses in chronic disease settings.

Dougan [sup.4] [...], CEACAM1 functions as a novel heterophilic ligand for TIM-3 and is necessary for TIM-3-mediated tolerance, which has marked consequences for inflammation, infection and cancer. Interaction of CEACAM1 and TIM-3 T cell regulation is mediated by a number of inhibitory cell surface molecules such as T-cell immunoglobulin domain and mucin domain-3 (TIM-3), a possible target for cancer immunotherapy that has been reported to act as an immune checkpoint in tumour-induced immune suppression. However, the tolerance inducing ligand for TIM-3 has yet to be uncovered, and TIM-3 may also possess activating functions under some circumstances. This study identifies carcinoembryonic antigen cell adhesion molecule 1 (CAECAM1) as the heterophilic ligand for murine TIM-3 that is required for TIM-3 to mediate tolerance and ensure its optimal maturation, cell surface expression and regulatory function. Co-blockade of CEACAM1 and TIM-3 leads to enhancement of anti-tumour immune responses, with improved elimination of tumours in mouse colorectal cancer models. T-cell immunoglobulin domain and mucin domain-3 (TIM-3, also known as HAVCR2) is an activation-induced inhibitory molecule involved in tolerance and shown to induce T-cell exhaustion in chronic viral infection and cancers.sup.1,2,3,4,5. Under some conditions, TIM-3 expression has also been shown to be stimulatory. Considering that TIM-3, like cytotoxic T lymphocyte antigen 4 (CTLA-4) and programmed death 1 (PD-1), is being targeted for cancer immunotherapy, it is important to identify the circumstances under which TIM-3 can inhibit and activate T-cell responses. Here we show that TIM-3 is co-expressed and forms a heterodimer with carcinoembryonic antigen cell adhesion molecule 1 (CEACAM1), another well-known molecule expressed on activated T cells and involved in T-cell inhibition.sup.6,7,8,9,10. Biochemical, biophysical and X-ray crystallography studies show that the membrane-distal immunoglobulin-variable (IgV)-like amino-terminal domain of each is crucial to these interactions. The presence of CEACAM1 endows TIM-3 with inhibitory function. CEACAM1 facilitates the maturation and cell surface expression of TIM-3 by forming a heterodimeric interaction in cis through the highly related membrane-distal N-terminal domains of each molecule. CEACAM1 and TIM-3 also bind in trans through their N-terminal domains. Both cis and trans interactions between CEACAM1 and TIM-3 determine the tolerance-inducing function of TIM-3. In a mouse adoptive transfer colitis model, CEACAM1-deficient T cells are hyper-inflammatory with reduced cell surface expression of TIM-3 and regulatory cytokines, and this is restored by T-cell-specific CEACAM1 expression. During chronic viral infection and in a tumour environment, CEACAM1 and TIM-3 mark exhausted T cells. Co-blockade of CEACAM1 and TIM-3 leads to enhancement of anti-tumour immune responses with improved elimination of tumours in mouse colorectal cancer models. Thus, CEACAM1 serves as a heterophilic ligand for TIM-3 that is required for its ability to mediate T-cell inhibition, and this interaction has a crucial role in regulating autoimmunity and anti-tumour immunity.

Abstract

Recent advances have re-defined a role for T cell factor 1 (Tcf1) that goes beyond T cell development and T memory formation and encompasses new functions in the regulation of T cell biology. Here, we discuss the multi-faceted and context-dependent role of Tcf1 in peripheral T cells, particularly during disease-induced inflammatory states such as autoimmunity, cancer, and chronic infections. Understanding how Tcf1 fine-tunes peripheral T cell biology holds the potential to tailor improved immune-targeted therapies.
