Bacterial interactions are vital for adapting to changing environments, with quorum sensing (QS) systems playing a central role in coordinating behaviors through small signaling molecules. The RRNPPA family is the prevalent QS systems in Bacillota and mediating communication through secreted oligopeptides, which are processed into active pheromones by extracellular proteases. Notably, in several cases the propeptides show the presence of multiple putative pheromones within their sequences, which has been proposed as a mechanism to diversify peptide-receptor specificity and potentially facilitate new functions. However, neither the processes governing the maturation of propeptides containing multiple pheromones, nor their functional significance has been evaluated. Here, using 2 Rap systems from bacteriophages infecting Bacillus subtilis that exhibit different types of pheromone duplication in their propeptides, we investigate the maturation process and the molecular and functional activities of the produced pheromones. Our results reveal that distinct maturation processes generate multiple mature pheromones, which bind to receptors with varying affinities but produce identical structural and biological responses. These findings add additional layers in the complexity of QS communication and regulation, opening new possibilities for microbial social behaviors, highlighting the intricate nature of bacterial interactions and adaptation.

ABSTRACT Hatching dynamics is key to population structure of annual fishes in temporary wetlands. Given the risk of false starts due to unpredictable hydroperiods, annual fish may employ risk‐spreading strategies in their hatching patterns consistent with bet hedging (such as the partial hatching of their egg bank and variable number of hatching cues) after being exposed to floods. However, the extent to which annual fish resort to bet hedging (as well as their potential drivers) over a wet–dry cycle is poorly understood. We assessed the hatching patterns of a South American killifish (Matilebias cyaneus; Cyprinodontiformes: Rivulidae) collected at different hydrological phases of a temporary wetland to test for variation in bet hedging over a wet–dry cycle. We incubated wild eggs of M. cyaneus obtained from male–female pairings separately reared in tanks originating in the ‘early‐wet’, ‘drying’ and ‘late‐wet’ phases of a temporary wetland and examined hatching patterns across a sequence of hydration trials under common‐garden conditions. We found variation in (i) the number of hatched and unhatched eggs and (ii) the number of hatched eggs across hydration trials among clutches from each phase (while controlling for parentage). Our results show that M. cyaneus variably commit to bet‐hedging hatching strategies over a wet–dry cycle, and suggest that the environmental conditions at specific periods of the year and the parental age at which eggs are produced drive annual fish hatching patterns. ABSTRACT In Firmicutes, important processes such as competence development, sporulation, virulence, and biofilm formation are regulated by cytoplasmic quorum sensing (QS) receptors of the RRNPPA family using peptide-based communication. Although these systems regulate important processes in a variety of bacteria, their origin and diversification are poorly understood. Here, we integrate structural, genomic, and phylogenetic evidence to shed light on RRNPPA protein origin and diversification. The family is constituted by seven different subfamilies with different domain architectures and functions. Among these, three were found in Lactobacillales (Rgg, ComR, and PrgX) and four in Bacillales (AimR, NprR, PlcR, and Rap). The patterns of presence and the phylogeny of these proteins show that subfamilies diversified a long time ago, resulting in key structural and functional differences. The concordance between the distribution of subfamilies and the bacterial phylogeny was somewhat unexpected, since many of the subfamilies are very abundant in mobile genetic elements, such as phages, plasmids, and phage-plasmids. The existence of diverse propeptide architectures raises intriguing questions about their export and maturation. It also suggests the existence of diverse roles for the RRNPPA systems. Some systems encode multiple pheromones on the same propeptide or multiple similar propeptides, suggesting that they act as “chatterers.” Many others lack pheromone genes and may be “eavesdroppers.” Interestingly, AimR systems without associated propeptide genes were particularly abundant in chromosomal regions not classed as prophages, suggesting that either the bacterium or other mobile elements are eavesdropping on phage activity. IMPORTANCE Quorum sensing (QS) is a mechanism of bacterial communication, coordinating important decisions depending on bacterial population. QS regulates important processes not only in bacterial behavior but also in genetic mobile elements and host-guest interactions. In Firmicutes, the most important family of QS receptors is the RRNPPA family. Despite the importance of such systems in microbiology, we know little about RRNPPA origin and diversification. In this work, the combination of sequence analysis and structural biology allowed us to identify a very large number of novel systems but also to class of them in functional families and thereby study of their origin and functional diversification. Moreover, peptide pheromone analysis revealed new and intriguing mechanisms of communication, such as “eavesdropper” systems which only listen for the pheromone and “chatterers” that take control of the communication in their microenvironment.

Bacterial two-component systems are composed of a sensor histidine kinase (HK) and an effector response regulator and upon signal detection, the HK autophosphorylates a conserved His residue. Here the authors structurally and functionally characterise two HKs, HK853–RR468 and EnvZ–OmpR, and find that the rotamer of the phosphorylatable catalytic His is not influenced by the environmental pH, ruling out an earlier proposed pH-gated model.

We describe Argolebias adrianae, a new species of killifish from a small temporary wetland in the Paraná Forest ecoregion with no regular or predictable temporal pattern of water availability. This habitat is in the Lower Iguazú River Basin, known for its high fish endemism, but until now, only two species of Rivulidae were reported from it, but from the Araucarian Forest ecoregion. The genus Argolebias was previously only known from the lower portions of the Paraguay, Paraná, and Uruguay basins and middle Paraná. The new species is distinguished from all congeners by its unique coloration, which includes a conspicuously dark grey anterior third portion of the dorsal fin and the absence of iridescent spots on the basal half of the pectoral fin in live adult males, as well as dark grey spots on the anterocentral portion of the flanks of females. Our phylogenetic analysis shows A. adrianae to be closely related to Argolebias guarani from the adjacent Middle Paraná basin. We also provide data on the ecology, ontogeny of coloration, and chorion ornamentation of this species. Our findings have important implications for understanding the biogeography, ecology, and evolution of mechanisms that enable organisms to thrive in highly stochastic environments like this one. Killifishes (Cyprinodontiformes) are a group of fish that include a high proportion of small-bodied species living in seasonal aquatic habitats, with narrow geographical distributions and high human impact. They are among the most vulnerable vertebrates in the Neotropical region, with nearly half of the species in threat categories. Herein, we propose a new phylogenetic hypothesis of the Rivulidae genus Austrolebias , based on 10 genes (six nuclear and four mitochondrial) and 191 morphological characters, including 90% of the total valid species of this genus. An updated definition and diagnosis of the existing subgenera of Austrolebias is provided, and these are erected to genera. Also, four new genera are erected to accommodate the taxonomy of the group to the current phylogenetic hypothesis. Additionally, we describe Argolebias guarani sp. nov. , from a seasonal pond in the Middle Paraná River basin, which is diagnosed by a unique colour pattern. This addition reinforces this area as a biodiversity hotspot of endemicity and highlights the importance of this region for conservation. This paper provides evidence on how a school is able to offer a quality education to all students, while taking into account the diversity of their personal and contextual characteristics. From the analysis of the processes, it can be concluded that the key factor for the achievement of inclusive education lies in the good practices of teachers, and that the study of these practices should be implemented as a professional development project. The data presented here are part of a broader research project, and show the perceptions of teachers and families from four selected schools, focusing on the characteristics of good practices of coordination between the family and the school. These findings can contribute to greater educational attention being paid to students with Specific Needs of Educational Support (SENS). Within the framework of qualitative research, a multiple case study was carried out, using interviews as the main instrument for data collection. After an in-depth analysis of the data, some conclusions are drawn to show the educational community the educational actions that define professional and institutional development, which would be capable of effectively serving diverse students, therefore making the inclusive school a reality. Specimens of Imparfinis were recently collected in north‐western Argentina from the Bermejo River basin (Salta and Jujuy Provinces), del Valle River (Salta Province) and Horcones River (Santiago del Estero Province). An integrative approach to taxonomy, combining a detailed morphological study and molecular phylogenetic analyses, was applied to determine the species identity of these specimens. A principal components analysis of morphological data clustered the specimens from north‐western Argentina and from the Amazon basin, indicating a close morphological resemblance. Also, a molecular phylogenetic analysis showed populations of I. guttatus from Argentina and Peru forming a clade. According to the conducted haplotype network analysis these populations are distinct in two mutations. Thus, in the absence of morphological or molecular data indicating the contrary, the combined method supports the identity of the specimens from the tributaries of the Paraguay River in Argentina as I. guttatus, whose type locality is in the upper Beni River basin in Bolivia. This contribution is also the first record for this species from Argentina. The disjunct distribution of I. guttatus provides new evidence reinforcing the hypothesis for the origin of the Paraguayan ichthyofauna. Saflufenacil is an herbicide that is leachable in soil and has the potential to contaminate groundwater, besides having moderate toxicity to aquatic organisms. Some macrophyte species may interfere with the availability of herbicides in water, increasing dissipation in this environment. Thus, the objective of this work was to evaluate the absorption and dissipation of 14C-saflufenacil in water by Egeria densa and Pistia stratiotes. Dissipation was performed with 14C-saflufenacil applied directly in water and quantified by liquid scintillation spectrometry (LSS). The evaluation times were 0, 3, 6, 24, 48, 72 and 96 h after application (HAA) for E. densa and 0, 12, 24, 36, 48, 60, 84 and 108 HAA for P. stratiotes. Absorption was analyzed through plant combustion in a biological oxidizer. The presence of the macrophytes increased the dissipation of 14C-saflufenacil in water. The half-life time (DT50) of the herbicide decreased by 82.6% in the presence of E. densa at 96 HAA. For P. stratiotes, the reduction in DT50 was 94.8% at 108 HAA. The absorption of 14C-saflufenacil was low for both macrophytes during the evaluated time. However, the macrophytes E. densa and P. stratiotes showed potential for the phytoremediation of water contaminated with saflufenacil. [ABSTRACT FROM AUTHOR]

The correct application of a new herbicide depends on knowledge concerning its behaviour within the cultivation system. The objective of this study was to evaluate the sorption–desorption process of aminocyclopyrachlor in soils with the addition of three aged organic materials from sugar cane and their transport via leaching. Sugar cane straw (12 t/ha), filter cake (90 t/ha) and vinasse (200 m3/ha) were added to a clayey soil 15, 30 and 60 days before herbicide application. Sorption and desorption were evaluated by the batch equilibrium method. For leaching assessments, the materials were applied to the soil surface. Sorption was relatively low in all treatments (Kd = 0.17–0.41 L/kg), although significantly higher in soil without organic material addition. A negative correlation between herbicide sorption and increased soil base saturation was observed, indicating competition for sorption sites. With the addition of vinasse, 71% of the herbicide reached the leachate, while <50% reached the leachate in the other treatments. Aminocyclopyrachlor availability was not reduced with organic material addition to the soil, which may be favourable for weed control. However, the presence of vinasse leads to the risk of leaching to deeper soil layers than the seed bank. [ABSTRACT FROM AUTHOR]

The objective of this study was to evaluate the sorption-desorption process and biodegradation of glyphosate in two tropical soils aged with biochar derived from eucalyptus. The biochar aging period was 30 d. There was little difference between the amounts of sorbed glyphosate in Ultisol (96.8, 96.8 and 96.4%) and Alfisol (97.1, 97.5 and 97.4%) soils that were unamended or amended with biochar aged for 0 or 30 d, respectively. Similar amounts of desorbed herbicide occurred in Ultisol (3.3, 3.3 and 3.4%) and Alfisol (4.1, 4.2 and 3.9%) soils, respectively. The degradation time half-life (DT50) of glyphosate in Ultisol unamended and initial amended were higher (38 and 36 d, respectively) than DT50 in the amended soil with 30 d of biochar aging (27 d); and in the Alfisol DT50 was higher in unamended soil (38 d), and similar in soil unamended at 0 and 30 d of biochar aging (21 and 26 d, respectively). The addition of biochar to two tropical soils or its aging did not have any effect on the sorption and desorption of glyphosate and its biodegradation in relation to the unamended soils, and it can did not affect the transport and persistence of this herbicide in soil. Quinclorac is an extremely persistent herbicide in the environment. However, information regarding the degradation of this herbicide in the soil is missing. Thus, the objective of this study was to evaluate the behavior of quinclorac in five tropical soils of contrasting textures by quantifying the extracted residue with formation of metabolites, bound residue and mineralization to 14CO2. The herbicide was applied at the dose of 15.1 Bq per biometric vial. For mineralization, 10 mL of NaOH were used, with weekly analyses of radioactivity in the liquid scintillation spectrometer, up to 240 days after application (DAA). Extractions were performed using NaOH and acetone. Thin Layer Chromatography (TLC) plates were used for the separation of the extracted residue in quinclorac and its metabolites. After the extraction, the soils were burned in a biological oxidizer to quantify the bound residue. Half-life (DT50) ranged from 57.7 to 266.5 days and five metabolites were found. The mineralized 14CO2 ranged from 12.5 to 25.6%, extracted residue from 104.3% to 23.3% and the bound residue from 3.6 to 49.5%. Quinclorac showed high persistence in the soil, which could cause carryover in subsequent crops and reach non-target organisms, but this depends on the physicochemical characteristics of each soil. Freshwater sulfide springs have extreme environmental conditions that only few vertebrate species can tolerate. These species often develop a series of morphological and molecular adaptations to cope with the challenges of life under the toxic and hypoxic conditions of sulfide springs. In this paper, we described a new fish species of the genus Jenynsia, Anablepidae, from a sulfide spring in Northwestern Argentina, the first in the family known from such extreme environment. Jenynsia sulfurica n. sp. is diagnosable by the lack of scales on the pre-pelvic area or the presence of a single row of scales, continuous or not, from the isthmus to the bases of the pelvic fins. Additionally, it presents a series of morphological and molecular characteristics that appear convergent with those seen in other fish species (e.g., Poeciliids) inhabiting sulfide springs. Most notably, J. sulfurica has an enlarged head and postorbital area compared to other fish of the genus and a prognathous lower jaw with a hypertrophied lip, thought to facilitate respiration at the air-water interface. Analyses of cox1 sequence showed that J. sulfurica has two unique mutations resulting in amino acid substitutions convergent to those seen in Poeciliids from sulfide springs and known to provide a physiological mechanism related to living in sulfide environments. A phylogenetic analysis, including molecular and morphological characters, placed J. sulfurica as sister taxa to J. alternimaculata, a species found in nearby, non-sulfide habitats directly connected to the sulfide springs. Thus, it can be inferred that the selection imposed by the presence of H2S has resulted in the divergence between these two species and has potentially served as a barrier to gene flow. Gymnogeophagus jaryi, new species, is described from Southern tributaries of the Middle Paraná basin in Misiones. It can be distinguished from all other members of the genus, except from G. australis and G. caaguazuensis, by the presence of a hyaline to grey anterior portion of the dorsal fin. Gymnogeophagus jaryi differs from G. caaguazuensis by a longer caudal peduncle, caudal fin not lyrate, central portion of scales on dorsal portion of trunk light iridescent blue and by white spots in soft portion of dorsal fin in adult males, and from G. australis by the light iridescent blue coloration of central portion of scales on the dorsal portion of trunk and tail, and by the lack of scales on the soft portion of the dorsal fin. Additionally, it can be diagnosed by the following unique combination of characters: 10–11 dorsal-fin branched rays, 27–30 E1 scales, absence of lips thickening, and, in males, by the possession of a hump in adults, caudal fin not lyrate, presence of large white spots forming transversal stripes distally and in anterior area of the dorsal fin’s soft portion, central area of scales on the dorsal portion of the trunk light iridescent blue, lack of scales on the base of the dorsal fin’s soft portion, absence of a conspicuous and oblique dark band from the eye to the anterior border of the head, anterior portion of dorsal fin hyaline to grey, scales of the midlateral spot each bearing a semicircular light blue blotch, head hump starting at the horizontal through the eyes, concave anterior profile in lateral view, base of unpaired fins yellow, and whitish hyaline spots on caudal fin. The new species, based on mtDNA phylogeny, is the sister species of G. caaguazuensis from the Paraguay basin and is closely related to G. australis. [ABSTRACT FROM AUTHOR]

A redescription of Corydoras undulatus Regan, 1912 is presented. The original description of C. undulatus is very succinct, as is its diagnosis, which is based only on external morphology. Additional information in the scientific literature on this species is scarce. Specimens from the distribution area of this species were analyzed; Paraná and Paraguay river basins in Argentina, Uruguay river basin in Argentina, Brazil and Uruguay, and the Laguna dos Patos system in Brazil. Morphological analysis, principal component analysis (PCA), meristic comparison and osteological description were performed. Corydoras undulatus can be distinguished from its congeners mainly by having the following combination of characters: mesethmoid short, with anterior tip short, smaller than 50% of the entire bone length; posterior margin of the pectoral-fin spine with nearly all serrations directed towards origin of spine; pectoral-fin spine with conical serrations; and its peculiar color pattern. The analysis of the material from the different basins did not indicate relevant morphological differences, suggesting that the species presents a wide distribution in La Plata and Laguna dos Patos drainages. The shared geographic distribution between these two systems is also present in other fish species. The current work presents data about the type locality, taxonomy, osteology, distribution and ontogenetic variation of color pattern in C. undulatus. Austrolebias wichi, new species, is herein described from seasonal ponds of the Bermejo river basin in the Western Chacoan district in northwestern Argentina. This species was found in a single pond, a paleochannel of the Bermejo River, which is seriously disturbed by soybean plantations surrounding it. Despite intensive sampling in the area, this species was only registered in this pond where it was relatively scarce. Therefore, we consider this species as critically endangered. This species is the sister species of A. patriciae in our phylogenetic analyses and is similar, in a general external aspect, to A. varzeae and A. carvalhoi. It can be distinguished among the species of Austrolebias by its unique color pattern in males. Additionally, from A. varzeae by presenting a supraorbital band equal or longer than the infraorbital band (vs. shorter) and from A. patriciae by the convex dorsal profile of head (vs. concave).
