This article presents results of an investigation that explored the school pathways of young Argentine unskilled workers without a secondary degree. Different factors that contributed to their not finishing school, and the manner in which educational institutions provided (or not) continuous support, were analyzed. Ten in-depth interviews with young individuals were conducted using a narrative approach. The results reflect how personal, family, work, social, and institutional factors converged to their discontinuous school pathways. Reflections are made from the field of guidance and counseling on the institutional role and how they can help students to build meaningful and sustainable lives.

In this work, we present the preliminary data we have obtained in the Neolithic flint quarry of Pozarrate, currently under archaeological works. We want to put forward an update about the sedimentary fillings of one of the crescent-shaped dump quarries (6000–5600 BP). It is located at the Sierra de Araico-Cucho (Treviño, northern Spain). In this region, Tertiary carbonated terrains which host important silicifications called Treviño flint (Tarriño, A. (2006). El sílex en la cuenca vasco-cantábrica y Pirineo Navarro. Caracterización y su aprovechamiento en la Prehistoria. Monografía 21, Museo Nacional y Centro de Investigación de Altamira. Madrid: Ministerio de Cultura) are outcropped. The procurement of these silicifications by prehistoric populations was motivated by the good quality of this raw material for knapping. Currently, it constitutes one of the few Neolithic known flint mines of the Iberian Peninsula, understood as places of exploitation of flint with landscape modifications. Some instruments related to prehistoric quarry works have been recovered, such as picks, maces, and hammers made of flint, dolerite, and deer antler.

Background: Ae. aegypti mosquitoes are considered a global threat to public health due to its ability to transmit arboviruses such as yellow fever, dengue, Zika and Chikungunya to humans. The lack of effective arboviral vaccines and etiological treatments make vector control strategies fundamental in interrupting the transmission cycle of these pathogens. This study evaluated Ae. aegypti mosquito populations pre- and post-intervention period with disseminating stations of the larvicide pyriproxyfen to understand its potential influence on the genetic structure and population diversity of these vectors. Methodology/Principal findings: This study was conducted in Manacapuru city, Amazonas, Brazil, where 1,000 pyriproxyfen dissemination stations were deployed and monitored from FEB/2014 to FEB/2015 (pre-intervention) and AUG/2015 to JAN/2016 (post-intervention). Low-coverage whole genome sequencing of 36 individuals was performed, revealing significant stratification between pre- and post-intervention groups (pairwise FST estimate of 0.1126; p-value < 0.033). Tajima's D estimates were -3.25 and -3.07 (both p-value < 0.01) for pre- and post-intervention groups, respectively. Molecular diversity estimates (Theta(S) and Theta(Pi)) also showed divergences between pre- and post-intervention groups. PCA and K-means analysis showed clustering for SNP frequency matrix and SNP genotype matrix, respectively, being both mainly represented by the first principal component. PCA and K-means clustering also showed significant results that corroborate the impact of pyriproxyfen intervention on genetic structure populations of Ae. aegypti mosquitoes. Conclusions/Significance: The results revealed a bottleneck effect and reduced mosquito populations during intervention, followed by reintroduction from adjacent and unaffected populations by this vector. We highlighted that low-coverage whole genome sequencing can contribute to genetic and structure population data, and also generate important information to aid in genomic and epidemiological surveillance. Author summary: The Aedes aegypti mosquito is a global public health threat, transmitting diseases like yellow fever, dengue, zika and Chikungunya, requiring several vector control strategies. Traditional methods such as active search and treatment of accessible breeding sites by control agents can be supplemented using PPF dissemination stations. The tactic uses adult mosquitoes themselves to disseminate the larvicide adhered to their bodies, between treated and untreated breeding sites, which are usually cryptic and inaccessible. This tactic can easily be incorporated into integrated vector management strategies, reducing the emergence of adult mosquitoes and, consequently, arboviruses transmission. However, its impact on Ae. aegypti genetic and population structure remain unknown. The PPF intervention caused a bottleneck in the genetic structure, an event in which a population undergoes a drastic reduction in population density, in this case due to the pressure caused by the larvicide, followed by reintroduction of adult mosquitoes from nearby areas not exposed to PPF. This study uses a cost-effective low-coverage whole genome sequencing method to analyze Ae. aegypti genetic population structure based on mitochondrial genome, comparing pre- and post-intervention periods with pyriproxyfen. This approach can reveal both short and long-term effects of this control strategy in the vector genome dynamics. These findings have direct implications for planning control activities, enhancing genomic surveillance and epidemiological surveillance, contributing to global efforts against vector-borne diseases. Aedes aegypti and Aedes albopictus are responsible for transmitting major human arboviruses such as Dengue, Zika, and Chikungunya, posing a global threat to public health. The lack of etiological treatments and efficient vaccines makes vector control strategies essential for reducing vector population density and interrupting the pathogen transmission cycle. This study evaluated the impact of long-term pyriproxyfen exposure on the genetic structure and diversity of Ae. aegypti and Ae. albopictus mosquito populations. The study was conducted in Manaus, Amazonas, Brazil, where pyriproxyfen dissemination stations have been monitored since 2014 up to the present day. Double digest restriction-site associated DNA sequencing was performed, revealing that despite significant local population reductions by dissemination stations with pyriproxyfen in various locations in Brazil, focal intervention has no significant impact on the population stratification of these vectors in urban scenarios. The genetic structuring level of Ae. aegypti suggests it is more stratified and directly affected by pyriproxyfen intervention, while for Ae. albopictus exhibits a more homogeneous and less structured population. The results suggest that although slight differences are observed among mosquito subpopulations, intervention focused on neighborhoods in a capital city is not efficient in terms of genetic structuring, indicating that larger-scale pyriproxyfen interventions should be considered for more effective urban mosquito control. Prior research has established the anti-apoptotic effects in insect cell cultures of Bombyx mori (B. mori) hemolymph, as well as the heightened production yields of recombinant proteins facilitated by baculovirus vectors in insect cells cultivated in media supplemented with this hemolymph. In this study, we investigated the hemolymph of another Lepidoptera species, Trichoplusia ni (T. ni), and observed similar beneficial effects in insect cells cultivated in media supplemented with this natural substance. We observed enhancements in both production yield (approximately 1.5 times higher) and late-stage cell viabilities post-infection (30–40% higher). Storage-protein 2 from B. mori (SP2Bm) has previously been identified as one of the abundant hemolymph proteins potentially responsible for the beneficial effects observed after the use of B. mori hemolymph-supplemented cell culture media. By employing a dual baculovirus vector that co-expresses the SP2Bm protein alongside the GFP protein, we achieved a threefold increase in reporter protein production compared to a baculovirus vector expressing GFP alone. This study underscores the potential of hemolymph proteins sourced from various Lepidoptera species as biotechnological tools to augment baculovirus vector productivities, whether utilized as natural supplements in cell culture media or as hemolymph-derived recombinant proteins co-expressed by baculovirus vectors. In the presence of different nucleophilic Knoevenagel competitors, cyclic and acyclic ketones have been shown to undergo highly chemoselective aldol reactions with aldehydes. In doing so, the substrate breadth for this emerging methodology has been significantly broadened. The method is also no longer beholden to proline-based catalyst templates, e.g., commercially available O-t-Bu-L-threonine is advantageous for acyclic ketones. The key insight was to exploit water-based mediums under conventional (in-water) and non-conventional (deep eutectic solvents) conditions. With few exceptions, high aldol-to-Knoevenagel chemoselectivity (>10:1) and good product profiles (yield, dr, and ee) were observed, but only in DESs (deep eutectic solvents) in conjunction with ball milling did short reaction times occur.
