Abstract The Ryukyu Islands of Japan are a biodiversity hotspot due to geographical and historical factors. Tricyrtis formosana is a perennial herbaceous plant that commonly found in Taiwan. But only a few populations have been identified in a limited habitat on Iriomote Island, while populations of unknown origin occur near human settlements in an area on the main island of Okinawa. To better understand these populations of the phylogenetic uniqueness and intrinsic vulnerability, we conducted comparative analyses including (1) phylogeny and population structure with MIG-seq data, (2) photosynthesis-related traits of plants grown under common conditions and (3) transcriptome analysis to detect deleterious variations. Results revealed that T. formosana was split into two clades by the congeners and that Iriomote and Okinawa populations independently derived from ancestral Taiwanese populations in each clade. Photosynthetic efficiency was lowest in the Iriomote population, followed by Okinawa and Taiwan. Transcriptome analysis showed that the Iriomote population accumulated more deleterious variations, suggesting intrinsic vulnerability. These results indicate that each T. formosana population in Japan is phylogenetically unique and has been independently dispersed from Taiwan, and that the Iriomote population presents a high conservation difficulty with a unique photosynthesis-related characteristic and a larger amount of deleterious variations.

Hydrangea marunoi Tagane & S. Fujii, from the Kimotsuki Mountains in the Ohsumi Peninsula, southern Japan, is described and illustrated. It is morphologically similar to H. alternifolia in having three-petaloid calyx lobes in marginal flowers, but is distinguished by the larger stamen number, and longer styles and seeds. Multiplex ISSR genotyping by sequencing (MIG-seq) demonstrated that the new species is monophyletic and closely related to H. amamiohsimensis and H. moellendorffii rather than H. alternifolia.

The relative importance of the mechanisms underlying species radiation remains unclear. Here, the authors combine reference genome assembly and population genetics analyses to show that neutral forces have contributed to the radiation of the most species-rich tree genus Syzygium.

As biodiversity loss continues, there is an urgent need to develop efficient conservation measures to protect diversity with limited conservation resources. Conservation targets have generally been selected based on their population size, but more detailed assessments clarifying the phylogenetic genetic status, history, and phylogenetic uniqueness of rare species is crucial to set more appropriate and effective conservation measures. In Japan, the Act on Conservation of Endangered Species of Wild Fauna and Flora designated endangered plants with high conservation priority, but >40% of these species also grow overseas. We conducted comparative analyses based on ddRADseq and MIG-seq to evaluate the population conservation status and value of Vaccinium emarginatum and Elatostema platyphyllum which are growing across national borders at the eastern edge of their species distribution range. The analyses revealed contrasting conservation status between the two species; the Japanese population of V. emarginatum had lower genetic diversity at the individual level and phylogenetically differentiated from Taiwanese populations, while that of E. platyphyllum had higher diversity at the individual level and is a relatively recent migrant with little phylogenetical differentiation from Taiwanese populations. The two species, which share the common feature of being critically rare in Japan, showed contrasting genetic/phylogenetic characteristics. This study provided useful information for appropriate conservation measures based on species’ phylogenetic traits and genetic diversity.

Macrosolen bidoupensis Tagane & V.S.Dang, sp. nov. (Loranthaceae) is newly described from Bidoup Nui Ba National Park in Lam Dong Province, southern Vietnam. The new species is characterized by small broadly elliptic to circular leaves, sessile to short petioles, slightly cordate to rounded leaf bases, 4–5 pairs of lateral veins and a basally green corolla tube. An illustration, a summary of DNA barcoding of the plastid genes rbcL and matK, and a key to the species of Macrosolen in Vietnam are provided.

A new species, Garcinia hopii H.Toyama & V.S.Dang is described from Bidoup Nui Ba National Park, southern Vietnam. This species is similar to Garcinia hendersoniana Whitmore but differs from that species in having larger leaves, clustered pistillate flowers, a greater number of sterile anthers and a larger stigma of young fruits. A description, preliminary conservation assessment, illustration, photographs and DNA barcodes of the new species are provided, as well as an updated key to Garcinia sect. Hebradendron in Indochina.

Two new species of genus Helicteres (Malvaceae, Helicteroideae), Helicteres Taynguyenenis and Helicteres dakmilensis from the Central Highlands of Vietnam are described and illustrated. H. taynguyenensis is morphologically similar to Helicteres angustifolia widely distributed in mainland SE Asia to China, Japan and Australia, but differs from the latter by having shorter petioles, larger calyx and petals, and longer androgynophores. Helicteres dakmilensis is closely related to Helicteres poilanei endemic to Vietnam, but distinguished by shorter petioles, fewer secondary veins, persistent stipules and bracts, larger calyx and fruits. The IUCN conservation status, vernacular name, illustrations, photographs, and comparison with the closest allies are provided for each of the new species. [ABSTRACT FROM AUTHOR]

A new species, Lasianthus bidoupensis V.S. Dang & Naiki (Rubiaceae) is described and illustrated from Bidoup Nui Ba National Park, Lam Dong Province, southern Vietnam. It resembles L. yaharae which was recently discovered from Mt. Hon Ba and L. dalatensis which is endemic to Dalat, southern Vietnam. However, L. bidoupensis differs from them by having larger leaves and bracts, longer petioles, stipules, calyx and corolla tubes, and deep-red mature fruits. [ABSTRACT FROM AUTHOR]

A new species of Hamamelidaceae, Eustigma honbaense H.Toyama, Tagane & V.S.Dang, sp. nov., is described from Hon Ba Nature Reserve, Vietnam. This species is similar to Eustigma oblongifolium Gardner & Champ., but differs from it in having entire leaves, longer infructescences, capsules with a longer apical part and seeds with a larger hilum. A new species, Aporosa tetragona Tagane & V. S. Dang, sp. nov., is described and illustrated from Mt. Hon Ba located in the Khanh Hoa Province, South Vietnam. This species is characterized by tetragonal pistillate flowers and fruits, which are clearly distinguishable from the other previously known species of the genus. The morphology and phylogeny based on rbcL and matK of this species indicated that the new species belongs to section Appendiculatae Pax & K. Hoffm. [ABSTRACT FROM AUTHOR]
