The main purpose of this study was to compare the antioxidant and anticancer activities of lycopene samples with different ratios of Z-isomers. Lycopene samples containing 5%, 30%, and 55% Z-isomers were successfully prepared by using thermal treatment combined with anti-solvent crystallization. The in vitro bio-accessibility of lycopene was estimated by the determination of partition factor (PF) and the results showed that lycopene with 55% Z-isomers possessed the highest bio-accessibility. Moreover, DPPH and ABTS assays suggested that the antioxidant activity of lycopene increased with the Z-isomers content from 5% to 55%. However, lycopene inhibited the survival of human hepatocellular carcinoma cells (HepG2) in a dose and time-dependent manner. The highest inhibition of HepG2 cell lines was achieved by 55% Z-ratio of lycopene. The cell viability was 22.54% at 20 μg/mL after incubating for 24 h, the number of cells was significantly reduced and the morphology was shrunk. Furthermore, molecular docking was introduced to compare the binding ability between different lycopene isomers with Scavenger Receptor class B type I (SR-BI), and the results revealed that the affinity of (all-E)-lycopene with SR-BI was lower compared to 5Z-lycopene and 13Z-lycopene, providing the reasons for different bioavailability of the above-mentioned lycopene isomers. All the above results demonstrated that Z-isomers-rich lycopene could enhance bio-accessibility and biological functionality.

Due to the abuse of antibiotics, the sensitivity of patients to antibiotics is gradually reduced. This work develops a Fe3O4@SiO2@Au/PDA nanochain which shows an interesting magnetic-field-induced improvement of its photothermal antibacterial property. First, SiO2 was wrapped on Fe3O4 nanospheres assembled in a chain to form a Fe3O4@SiO2 nanocomposite with a chain-like nanostructure. Then, the magnetic Fe3O4@SiO2@Au/PDA nanochains were prepared using in situ redox-oxidization polymerization. Under the irradiation of an 808 nm NIR laser, the temperature rise of the Fe3O4@SiO2@Au/PDA nanochain dispersion was obvious, indicating that they possessed a good photothermal effect. Originating from the Fe3O4, the Fe3O4@SiO2@Au/PDA nanochain showed a typical soft magnetic behavior. Both the NIR and magnetic field affected the antimicrobial performance of the Fe3O4@SiO2@Au/PDA nanochains. Escherichia coli and Staphylococcus aureus were used as models to verify the antibacterial properties. The experimental results showed that the Fe3O4@SiO2@Au/PDA nanochains exhibited good antibacterial properties under photothermal conditions. After applying a magnetic field, the bactericidal effect was further significantly enhanced. The above results show that the material has a broad application prospect in inhibiting the growth of bacteria.

The purpose of this study was to investigate the effect of okra polysaccharides with similar molecular weight on intestinal ecology. After extraction, ethanol precipitation, deproteinization and purification, two polysaccharide fractions, named as water-extracted polysaccharides (WEP) and alkaline-extracted polysaccharides (AEP), were obtained. According to the results of high performance liquid chromatography, AEP contained mannose, rhamnose, galacturonic acid and galactose, while WEP was composed of galactose, rhamnose, fructose, arabinose, glucose, galacturonic acid and mannose. The compositions of intestinal microbiota were affected by the change of daily substrates. Meanwhile, the concentration of ammonia and gastrointestinal transit time decreased, the total short chain fatty acid (SCFA) levels, moisture of caecum content increased. These findings suggested that the effect of okra polysaccharides on intestinal microbiota was achieved by the output of detrimental compounds and the supply of nutrients. Besides, AEP supported a more pivotal role in manipulating gut microbiota than WEP.

Alzheimer’s disease (AD) is a progressive neurodegenerative brain disorder characterized by memory loss, language impairment, personality changes and intellectual decline. Taking into account the key pathological features of AD, such as low levels of acetylcholine, beta-amyloid (Aβ) aggregation, oxidative stress and dyshomeostasis of biometals, a new series of paeonol derivatives 5a–5d merging three different functions, i.e., antioxidant, anti-acetylcholinesterase (AChE) activity, metal chelating agents for AD treatment have been synthesized and characterized. Biological assays revealed that compared with paeonol (309.7 μM), 5a–5d had a lower DPPH IC50 value (142.8–191.6 μM). 5a–5d could significantly inhibit hydrogen peroxide-induced neuronal PC12 cell death assessed by MTT assay in the concentration range of 5–40 μM. AChE activity was effectively inhibited by 5a–5d, with IC50 values in the range of 0.61–7.04 μM. 5a–5d also exhibited good metal-chelating ability. All the above results suggested that paeonol derivatives may be promising multifunctional agents for AD treatment.

Fresh Echinacea purpurea plant is a rich source of caffeic acid derivatives, which actively participate in the regulation of immune function. Herein, we describe a novel process for the production of fresh Echinacea purpurea juice rich in caffeic acid derivatives. This novel process includes steam heating (100°C, 15 min) before squeezing the juice to increase the recovery of caffeic acid derivatives from the flower or leaf by 1.5 to 3.7 fold. The recovery was further enhanced by 11-20% using ultrasonication at 500 W for 30 min. The flower, leaf, and root juice of Echinacea purpurea could be preserved for 4 weeks (4°C) or 6 months (-18°C) following ultra-high-pressure pasteurization (300 MPa, 15 min). Under these conditions, more than 90% of active cichoric acid was retained. Furthermore, the frozen-stored Echinacea purpurea juice exhibited similar or better immunoenhancement properties when measured by organ index, ear swelling, and carbon clearance test in mice compared with that of the positive control group (P < 0.05). Our results suggest that a combination of steam, ultrasound, and ultra-high-pressure treatment could serve as a superior process for the preparation of functional Echinacea purpurea juice. An efficient separation method of scopolamine from Hindu Datura extracts was successfully developed by combining liquid–liquid extraction, macroporous resins, and crystallization. First, the extraction solution was successively performed with liquid–liquid extraction by acid water and carbon tetrachloride. Secondly, D151 resin was selected from six tested resins, and the separation parameters were optimized. Finally, scopolamine hydrobromide was obtained after crystallization at −20°C overnight. Through one run of above combined treatments, the content of scopolamine increased from 3.02% (w/w) to 99.12% (w/w). Development of oral mucositis represents a rate-limiting factor in radiation therapy for the treatment of head and neck, as well as other cancers. In this work, we investigated the treatment effect of ecdysterone (a steroid derived from the dry root ofAchyranthes bidentate) on radiation-induced oral mucositis, and examined possible underlying mechanisms. Male Sprague-Dawley rats were exposed to 20 Gy X-ray irradiation (single dose, cranial localization) to induce oral mucositis. Possible therapeutic effects of ecdysterone on radiation-induced oral mucositis were investigated by monitoring weights, direct observations, visual scoring method and evaluation of hematoxylin and eosin staining. Assessments of leukocyte common antigen and proliferating cell nuclear antigen staining were also performed in the damaged areas of tongues harvested after irradiation, and changes in signaling pathways were investigated using Western blotting. The development and progression of radiation-induced oral mucositis in this model was similar to that observed in clinic patients. Ecdysterone effectively improved radiation-induced oral mucositis as assessed by direct observation and histopathology, and also increased proliferation of matrix cells, since the Ras-Raf-ERK signal pathway was found to be activated by its use. It was concluded that orally administered ecdysterone accelerated the healing process in a rat model of radiation-induced oral mucositis by upregulating the Ras-Raf-ERK signal pathway.
