Amazonian soils are heterogeneous. However, few studies have been carried out in the Amazon, mainly because of its considerable size, which complicates the collection of data and the ability to plan for the sustainable use of natural resources. In this study, the physical, chemical, and mineralogical attributes of soils in the state of Pará, Brazil, were characterized by examining the particle size, fertility, silicon extracted by sodium hydroxide, iron, and aluminum, and manganese extracted by sulfuric acid, sodium citrate-bicarbonate-dithionite, and ammonium oxalate + oxalic acid. A descriptive analysis, multivariate principal component analysis, and cluster analysis were performed. The soils had low concentrations of bioavailable P, Ca2+, Mg2+, and K and had high concentrations of Al3+, Si, and Al oxide in the Cambisols. Concentrations of Fe and Mn oxides were higher in both the Cambisols and Nitosols, which are rich in oxidic minerals. The multivariate analysis indicated an association between the organic carbon content and pH, P, Ca, Mg, and K concentrations. An additional association was observed between clay, potential acidity, and the Fe and Al oxide concentrations.

Abstract Background Forensic analysis heavily relies on DNA analysis techniques, notably autosomal Single Nucleotide Polymorphisms (SNPs), to expedite the identification of unknown suspects through genomic database searches. However, the uniqueness of an individual’s genome sequence designates it as Personal Identifiable Information (PII), subjecting it to stringent privacy regulations that can impede data access and analysis, as well as restrict the parties allowed to handle the data. Homomorphic Encryption (HE) emerges as a promising solution, enabling the execution of complex functions on encrypted data without the need for decryption. HE not only permits the processing of PII as soon as it is collected and encrypted, such as at a crime scene, but also expands the potential for data processing by multiple entities and artificial intelligence services. Methods This study introduces HE-based privacy-preserving methods for SNP DNA analysis, offering a means to compute kinship scores for a set of genome queries while meticulously preserving data privacy. We present three distinct approaches, including one unsupervised and two supervised methods, all of which demonstrated exceptional performance in the iDASH 2023 Track 1 competition. Results Our HE-based methods can rapidly predict 400 kinship scores from an encrypted database containing 2000 entries within seconds, capitalizing on advanced technologies like Intel AVX vector extensions, Intel HEXL, and Microsoft SEAL HE libraries. Crucially, all three methods achieve remarkable accuracy levels (ranging from 96% to 100%), as evaluated by the auROC score metric, while maintaining robust 128-bit security. These findings underscore the transformative potential of HE in both safeguarding genomic data privacy and streamlining precise DNA analysis. Conclusions Results demonstrate that HE-based solutions can be computationally practical to protect genomic privacy during screening of candidate matches for further genealogy analysis in Forensic Genetic Genealogy (FGG).

Madariaga virus (MADV) and Venezuelan equine encephalitis virus (VEEV) are emerging arboviruses affecting rural and remote areas of Latin America. However, clinical and epidemiologic reports are limited, and outbreaks are occurring at an increasing frequency. We addressed the data gap by analyzing all available clinical and epidemiologic data of MADV and VEEV infections recorded since 1961 in Panama. A total of 168 human alphavirus encephalitis cases were detected in Panama during 1961‒2023. We described the clinical signs and symptoms and epidemiologic characteristics of those cases, and also explored signs and symptoms as potential predictors of encephalitic alphavirus infection compared with those of other arbovirus infections occurring in the region. Our results highlight the challenges for the clinical diagnosis of alphavirus disease in endemic regions with overlapping circulation of multiple arboviruses.

Western equine encephalitis virus (WEEV) is a mosquitoborne virus that reemerged in December 2023 in Argentina and Uruguay, causing a major outbreak. We investigated the outbreak using epidemiologic, entomological, and genomic analyses, focusing on WEEV circulation near the Argentina‒Uruguay border in Rio Grande do Sul state, Brazil. During November 2023‒April 2024, the outbreak in Argentina and Uruguay resulted in 217 human cases, 12 of which were fatal, and 2,548 equine cases. We determined cases on the basis of laboratory and clinical epidemiologic criteria. We characterized 3 fatal equine cases caused by a novel WEEV lineage identified through a nearly complete coding sequence analysis, which we propose as lineage C. Our findings highlight the importance of continued surveillance and equine vaccination to control future WEEV outbreaks in South America.

Objective Adolescence marks a time of transition where teenagers are learning to advocate for themselves. In those with underlying chronic conditions such as adolescent dysautonomia, improving communication between clinic visits may improve connection with their health care provider which may aide management. Our aims were as follows: (1) to implement a text message platform to increase communication between adolescent patients and health care provider (HCP); (2) to evaluate its effect on quality of life (QoL), symptom burden, and patient engagement; and (3) to determine patient satisfaction with the platform. Methods Participants (age 12–18) with access to a personal mobile phone were recruited from a pediatric dysautonomia clinic. A weekly automated text message asking “How are you?” was sent to participants (WelTel Inc.). Responses were triaged to HCP and responded to within 48 hours. Results Twenty-six participants with median (interquartile range) age of 16.8 (15.7–17.4) years completed the study. Duration of the text messaging intervention was 33 (26.8–37.3) weeks. A total of 896 automated weekly messages were sent, which resulted in 206 (23%) care conversations. Participants found texting useful (96%) and produced feelings of connection to their HCP (92%). There was no change in overall QoL or symptom burden ( p > 0.05). Conclusion A text message platform was successfully implemented in adolescents seen in our Dysautonomia Clinic. Patients were engaged, satisfied with the platform, and felt connected to their HCP despite no changes in QoL or symptom burden.

This paper proposes a two-stage converter that can black start an isolated AC Microgrid with a Fuel Cell (FC) as the primary energy source. The first stage is connected to the FC and employs a Three-Leg Interleaved Boost DC/DC Converter (IBC), while the second is a Three-Phase Voltage Source Converter (VSC). The DC/DC stage utilizes a Cascade Voltage Control (CVC) to mitigate voltage fluctuations in the DC-link caused by the variability of the FC voltage. For the DC/AC stage, three distinct grid-forming (GFM) strategies are implemented with two of them with multi-loop cascaded structure and one with a single-loop structure. The power circuit of the system is simulated using the Real-Time Simulator (RTS) HIL 602+ from Typhoon-HIL, with the control strategies embedded on the Digital Signal Processor (DSP) TMS320F28379D - F28379D LaunchPad from Texas Instruments (TI). The performance of the cases are verified through CHIL simulations for a balanced and unbalanced inductive load steps. The results demonstrate that for both tests the GFM single loop structure presents smoother transients and shorter recovery times. Additionally, for the unbalanced loads, all the cases present similar results for the DC variables with more pronounced differences at the AC side.

Summary: Background: Chikungunya virus (CHIKV) continues to cause explosive epidemics in Brazil. We investigated its transmission dynamics in Salvador, Brazil, to understand the factors driving its reemergence and spread. Methods: In this epidemiological study, we analyzed by census tracts the chikungunya cases reported in Salvador during the 2019–2020 epidemics. We used SaTScan software to identify spatiotemporal clusters and assessed how census tract characteristics (socioeconomic, environmental, and prior chikungunya occurrence) influenced chikungunya incidence through a Bayesian spatial model using Integrated Laplace Approximation (INLA). Findings: Citywide, 19,129 cases (mean age: 40.2, range: 0–112; male: 41.8%, female: 58.0%, non-binary: 0.2%) were reported between 2016 and 2020, with a significant increase in 2019 and 2020 (4549 and 13,071 cases, respectively). We found nine spatiotemporal clusters in 2019 and seven in 2020, with 17.2% (387 of 2252) overlap of census tracts between the two years. The chikungunya incidence by census tract was negatively correlated with income and vegetation but positively correlated with land surface temperature. The census tract level incidence in 2020 exhibited a non-linear correlation with the 2019 incidence; up to a certain level, the 2020 risk increased as the 2019 incidence increased, but when the 2019 incidence was extreme, the 2020 risk was reduced. Interpretation: These findings suggest that CHIKV transmission is localized, even during what appeared to be a citywide epidemic, creating high-risk pockets within the city. Socioeconomic factors, environmental conditions, and prior chikungunya incidence, probably reflecting herd immunity, all influence case incidence. The semi-arid region of Ceará, located in Northeast Brazil, is typically characterized by low rainfall, high temperatures, and sparse vegetation – conditions generally associated with reduced soil erosion rates. However, the region's fragile soils, which are shallow, nutrient-poor, and particularly susceptible to degradation, exacerbate the erosion. This degradation poses a significant threat to soil fertility, agricultural productivity, and environmental sustainability in the area. The primary objective of this study was to quantify soil loss in Ceará and to challenge the prevailing assumption that semi-arid regions inherently experience minimal erosion due to limited rainfall. The Intensity of Erosion and Outflow (IntErO) model was used, in conjunction with remote sensing data and geographic information system (GIS) technologies. Through this approach, we estimated an average soil loss rate of 9.7 Mg ha⁻¹ yr⁻¹. The results indicate that 38% of Ceará’s land area experiences soil loss rates exceeding tolerance limits, particularly in regions lacking vegetation, as well as in agricultural lands and pastures. These elevated erosion rates are largely attributed to the area’s poor soil properties, occasional intense rainfall events, and unsustainable land management practices. The findings underscore the urgent need for improved management of soil erosion processes to address these challenges. Implementing targeted soil conservation strategies and policies that promote sustainable land use is essential for mitigating water erosion, safeguarding long-term agricultural productivity, and enhancing environmental and socioeconomic sustainability in semi-arid regions.

We conducted a cross-sectional serosurvey for chikungunya virus (CHIKV) exposure in fruit bats in Senegal during 2020–2023. We found that 13.3% (89/671) of bats had CHIKV IgG; highest prevalence was in Eidolon helvum (18.3%, 15/82) and Epomophorus gambianus (13.7%, 63/461) bats. Our results suggest these bats are naturally exposed to CHIKV.

We detected Mayaro virus (MAYV) in 3.4% (28/822) of febrile patients tested during 2018–2021 from Roraima State, Brazil. We also isolated MAYV strains and confirmed that these cases were caused by genotype D. Improved surveillance is needed to better determine the burden of MAYV in the Amazon Region.

Fermentation is an oxygen-free biological process that produces hydrogen, a clean, renewable energy source with the potential to power a low-carbon economy. Bibliometric analysis is crucial in academic research to evaluate scientific production, identify trends and contributors, and map the development of a field, providing valuable information to guide researchers and promote scientific innovation. This review provides an advanced bibliometric analysis and a future perspective on fermentation for hydrogen production. By searching WoS, we evaluated and refined 62,087 articles to 4493 articles. This allowed us to identify the most important journals, countries, institutions, and authors in the field. In addition, the ten most cited articles and the dominant research areas were identified. A keyword analysis revealed five research clusters that illustrate where research is progressing. The outlook indicates that a deeper understanding of microbiology and support from energy policy will drive the development of hydrogen from fermentation.

Abstract Hepatic glycogen storage diseases (GSD) are characterized by recurrent episodes of hypoglycemia, and anemia has been recognized as a frequent complication of these disorders.This was a convenience cross-sectional study to evaluate hepcidin and IL-6 concentrations in patients with hepatic GSD and their association with anemia and other parameters of iron metabolism.Levels of hepcidin, IL-6, and markers of iron metabolism were measured in 32 patients receiving uncooked cornstarch therapy for GSD (GSD Ia= 18; Ib= 7; III= 3; IXa= 3; IXb= 1; median age 9.5 years). IL-6 concentrations were compared to those of 8 individuals heterozygous for GSD. Nine patients were anemic and five patients had hepatic adenomas. IL-6 levels were higher in patients than in heterozygotes. Eight patients had hyperferritinemia, and one had elevated transferrin saturation as well. Hepcidin correlated positively with ferritin levels. IL-6 correlated with hemoglobin, iron, transferrin, and transferrin saturation. There was no correlation between hepcidin and IL-6 levels. Patients with GSD Ib had the highest IL-6 levels.Anemia is a common finding in hepatic GSD, especially in GSD Ib, the type of GSD associated with the highest IL-6 levels. These findings suggest that inflammation is strongly associated with development of anemia in GSD Ib.

Abstract This study evaluated the digestibility of whole mango (Mangifera indica) meal (WMM) and determined the growth performance, intestinal enzyme activity, and metabolic and hematologic responses of tambaqui (Colossoma macropomum) juveniles fed diets containing different proportions of corn meal (CM) substitution by WMM. Fish fed with graded levels of WMM (0 (control), 80, 160, 240, and 320 g kg diet-1), replacing part of the dietary CM. The apparent digestibility coefficients of WMM were above 96%. Diets with WMM did not affect growth performance or intestinal enzyme activity. However, they showed a positive linear effect on plasma glucose, amino acids, and albumin levels and a negative linear effect on hepatic aspartate aminotransferase activity and hepatic glycogen, plasma cholesterol, and hemoglobin levels. Increased erythrocyte values and decreased plasma triglyceride levels were verified in fish fed 80 and 160 g WMM kg diet-1. In conclusion, the WMM may be a viable alternative to the tambaqui juveniles’ diet, and WMM could replace up to 16% of CM without harming the growth and health of tambaqui juveniles.

The growing presence of power ramps, typically caused by the intermittency of renewable energy sources (RESs), may ultimately threaten the stability and reliability of the power grid. In the context of power smoothing algorithms such as Moving Average, Ramp Rate and First-Order Low-Pass Filter have been widely used in reference generation for Energy Storage Systems (ESSs). In this scenario, this paper analyzes typical metrics used for evaluating power smoothing techniques and comments on their limitations. Validation of this analysis is conducted using PV generation data sourced from the National Renewable Energy Laboratory (NREL). The results highlight the need to develop new metrics for a fairier comparison. Finally, this work also sets a concrete path for the evolution of said metrics.

This study reports the observation of Elaphria agrotina (Guenée, 1852) (Lepidoptera, Noctuidae) attacking sugarcane (Saccharum spp.). These specimens were collected in a sugarcane field under renovation while feeding on bait traps made from sugarcane stalks. Larvae were also detected within soil layers up to 6 cm deep, near to crop residues and rhizomes. Notably, they were found on sugarcane plants displaying 'dead heart' symptoms. The presence of this species in bait of sugarcane stalks raises concerns about potential secondary damage to the crop, which is already susceptible to other pests, and the possibility of it causing infestations in healthy plants.

Abstract The implementation of restrictive measures during the pandemic drastically changed the routine and habits of the population. In the academic sphere, the sudden closure of universities difficulted any advance planning. The objective of this study was to verify the effects of the pandemic on the routine, physical and mental health of students at a public university in southern Brazil. This is a cross-sectional study that applied a questionnaire from May to August 2020. The target population comprised 528 undergraduates. The questionnaire addressed questions about sociodemographic characteristics, physical and mental health. A response was obtained from 406 participants. During the quarantine period evaluated, the main results were: 66.2% reported worsening of anxiety symptoms; 57.6% declared that their physical health was “reasonable” to “bad”. As for the quality of sleep there was an increase in 98.8% in the responses “bad sleep quality” and “very bad sleep quality”; increase in 26.6% for presence bruxism; 12.5% for orofacial pain, 3.8% for headache. It is concluded that there were several negative effects, among these effects, it was observed higher levels of anxiety, worsening physical health and quality of sleep, increase in the prevalence of bruxism, as well as the presence of orofacial pain and headache.

ABSTRACTIntroduction Fungal infections are caused by a broad range of pathogenic fungi that are found worldwide with different geographic distributions, incidences, and mortality rates. Considering that there are relatively few approved medications available for combating fungal diseases and no vaccine formulation commercially available, multiple groups are searching for new antifungal drugs, examining drugs for repurposing and developing antifungal vaccines, in order to control deaths, sequels, and the spread of these complex infections.Areas covered This review provides a summary of advances in fungal vaccine studies and the different approaches under development, such as subunit vaccines, whole organism vaccines, and DNA vaccines, as well as studies that optimize the use of adjuvants. We conducted a literature search of the PubMed with terms: fungal vaccines and genus of fungal pathogens (Cryptococcus spp. Candida spp. Coccidioides spp. Aspergillus spp. Sporothrix spp. Histoplasma spp. Paracoccidioides spp. Pneumocystis spp. and the Mucorales order), a total of 177 articles were collected from database.Expert opinion Problems regarding the immune response development in an immunocompromised organism, the similarity between fungal and mammalian cells, and the lack of attention by health organizations to fungal infections are closely related to the fact that, at present, there are no fungal vaccines available for clinical use.

Abstract Tuning the electronic properties of transition metals using pyrophosphate (P2O7) ligand moieties can be a promising approach to improving the electrochemical performance of water electrolyzers and supercapacitors, although such a material’s configuration is rarely exposed. Herein, we grow NiP2O7, CoP2O7, and FeP2O7 nanoparticles on conductive Ni-foam using a hydrothermal procedure. The results indicated that, among all the prepared samples, FeP2O7 exhibited outstanding oxygen evolution reaction and hydrogen evolution reaction with the least overpotential of 220 and 241 mV to draw a current density of 10 mA/cm2. Theoretical studies indicate that the optimal electronic coupling of the Fe site with pyrophosphate enhances the overall electronic properties of FeP2O7, thereby enhancing its electrochemical performance in water splitting. Further investigation of these materials found that NiP2O7 had the highest specific capacitance and remarkable cycle stability due to its high crystallinity as compared to FeP2O7, having a higher percentage composition of Ni on the Ni-foam, which allows more Ni to convert into its oxidation states and come back to its original oxidation state during supercapacitor testing. This work shows how to use pyrophosphate moieties to fabricate non-noble metal-based electrode materials to achieve good performance in electrocatalytic splitting water and supercapacitors.

Abstract Eco-friendly and cost-effective catalysts with multiple active sites, large surface area, high stability and catalytic activity are highly desired for efficient water splitting as a sustainable green energy source. Within this line, a facile synthetic approach based on solventless thermolysis was employed for the simple and tunable synthesis of Ni1−xMgxFe2O4 (0 ≤ x ≤ 1) nanosheets. The characterization of nanosheets (via p-XRD, EDX, SEM, TEM, HRTEM, and SAED) revealed that the pristine ferrites (NiFe2O4 and MgFe2O4), and their solid solutions maintain the same cubic symmetry throughout the composition regulation. Elucidation of the electrochemical performance of the nanoferrite solid solutions showed that by tuning the local chemical environment of Ni in NiFe2O4 via Mg substitution, the intrinsic catalytic activity was enhanced. Evidently, the optimized Ni0.4Mg0.6Fe2O4 catalyst showed drastically enhanced HER activity with a much lower overpotential of 121 mV compared to the pristine NiFe2O4 catalyst. Moreover, Ni0.2Mg0.8Fe2O4 catalyst exhibited the best OER performance with a low overpotential of 284 mV at 10 mA/cm2 in 1 M KOH. This enhanced electrocatalytic activity could be due to improved electronic conductivity caused by the partial substitution of Ni2+ by Mg2+ in the NiFe2O4 matrix as well as the synergistic effect in the Mg-substituted NiFe2O4. Our results suggest a feasible route for developing earth-abundant metal oxide-based electrocatalysts for future water electrolysis applications.

Significant populations in tropical and sub-tropical locations all over the world are severely impacted by a group of neglected tropical diseases called leishmaniases. This disease is caused by roughly 20 species of the protozoan parasite from the Leishmania genus. Disease prevention strategies that include early detection, vector control, treatment of affected individuals, and vaccination are all essential. The diagnosis is critical for selecting methods of therapy, preventing transmission of the disease, and minimizing symptoms so that the affected individual can have a better quality of life. Nevertheless, the diagnostic methods do eventually have limitations, and there is no established gold standard. Some disadvantages include the existence of cross-reactions with other species, and limited sensitivity and specificity, which are mostly determined by the type of antigen used to perform the tests. A viable alternative for a more precise diagnosis is the application of recombinant antigens, which have been generated using bioinformatics approaches and have shown increased diagnostic accuracy. This approach proves valuable as it spans from epitope selection to predicting the interactions within the antibody–antigen complex through docking analysis. As a result, identifying potential new antigens using bioinformatics resources becomes an effective technique since it may result in an earlier and more accurate diagnosis. Consequently, the primary aim of this review is to conduct a comprehensive overview of the most significant in silico tools developed over time, with a focus on evaluating their efficacy and exploring their potential applications in optimizing the selection of highly specific molecules for a more effective diagnosis of leishmaniasis.

The biodiversity of Brazil provides an excellent climate and favorable pollination conditions for Apis mellifera L., especially in the Eastern Amazon region, which boasts vast floral wealth, including an abundance of açaí (Euterpe oleracea) flowers and fruits. In the present study, seven types of honey were evaluated: three containing floral nectar from açaí (Açaí honey) collected in the Eastern Amazon region (Açaí honey from Breu Branco (AH1 and AH2) and Açaí honey from Santa Maria (AH3), both from the state of Pará, Brazil) and four honeys from different regions of Brazil (Aroeira honey from Minas Gerais, Cipó-Uva honey from Distrito Federal, Mangue honey from Pará, and Timbó honey from Rio Grande do Sul). The characteristics of these honeys were evaluated by examining their physicochemical properties, melissopalynological aspects, color, antioxidant potential, and their constituent compounds, which were confirmed through GC-MS analysis. Açaí floral nectar honeys presented physicochemical results similar to those of other honeys, aligning with Brazilian legislation norms, but differed in their high values of free acidity, apparent sugars, and lower reducing sugars, which are directly related to their botanical origin. These differences correlate with unique flavor and distinct aroma characteristics. Melissopalynological analysis confirmed the botanical origin of the honeys containing açaí floral nectar, which had a color range from amber to dark amber. The three açaí honeys demonstrated high antioxidant capacity and superior flavonoid and polyphenol content compared to other honeys, particularly the açaí honey from Breu Branco (AH1), which had four times the content to combat free radicals compared to the honey with the highest potential (Aroeira honey). GC-MS analysis confirmed the presence of antioxidant properties as well as potential anti-inflammatory, antibacterial, antimicrobial, and antitumor capabilities in açaí honeys, which have not yet been fully studied.

The process of photosynthesis involves the absorption of light energy by photosynthetic pigments found in the thylakoid membrane of chloroplasts in order to produce chemical energy. In the presence of photosynthetic inhibitors, plant development is affected mainly by the reduction of the electron chain, which leads to growth inhibition. In this context, chlorophyll a (Chl a) fluorescence is an important technique used to identify the effects of inhibitors on the photosynthetic apparatus employing the JIP-test, which correlates the Chl a fluorescence transient to biophysical parameters, providing valuable information about the efficiency of photosystems I and II. Natural products have been highlighted as photosynthesis inhibitors due to the continued use of synthetic herbicides, which leads to the development of invasive plants resistant to these pesticides, in addition to the toxicity caused to humans and the environment. Thus, this review describes the main applications of extracts and isolated secondary metabolites obtained from plants and microorganisms in the investigation of electron transport inhibition on photosystem II.

The interaction between stingless bees and fungi has garnered recent interest due to potential mutual benefits. In Brazil, Melipona scutellaris stands out ecologically and economically. We investigated its microbiota, focusing on yeast species in pot-honey, pot-pollen, and nest surfaces. Samples from the Atlantic Forest (Pernambuco state, Brazil) were analyzed through morphological and physiological methods and D1/D2 LSU rDNA sequence analysis. We identified 20 yeast species, comprising 15 ascomycetes and five basidiomycetes, representing 11 and five genera, respectively. All yeast species, except Blastobotrys meliponae, were reported for the first time in association with M. scutellaris. Honey exhibited the highest species richness. In this study, the species Blastobotrys meliponae, Meyerozyma caribbica, Wickerhamomyces anomalus, and Moniliella carnis were exclusively isolated from honey. The understanding of the ecological association between stingless bees and yeasts, as revealed in our research, can significantly aid in insect conservation programs. We present the first report of yeasts associated with M. scutellaris substrates living in the Brazilian Atlantic Forest.

ABSTRACT To meet environmental standards and sustainability policies, diesel-biodiesel-ethanol blends have been investigated as partial replacements for diesel. This study aimed to evaluate the emissions and performance of a diesel engine using a blend of biodiesel and ethanol. Four blends of biodiesel (7, 10, 15, and 20%) and four blends of ethanol (0, 1, 3, and 5%) were used. The power remained stable with the addition of biodiesel but decreased with the addition of ethanol for all blends. The power was reduced from 76 to 74 kW when the ethanol content increased from 0 to 5%. The addition of 1% ethanol increased the maximum torque linearly from 372 to 378 kW. A slight increase in the specific consumption was observed with the use of biodiesel and ethanol in the blend. An increase in the ethanol fraction from 0 to 5% reduced nitrogen oxide emissions, especially at high loads. Ethanol caused a decrease of up to 42% in nitrogen oxide. At high loads, a reduction in carbon monoxide emissions was observed with an increase in the blends of ethanol and biodiesel. Blends of biodiesel and ethanol with concentrations of 20% biodiesel and 5% ethanol are alternatives to diesel in agricultural tractor engines.

Abstract High infestations of epibiont ciliates on vertebrates or invertebrates are normally related to aquaculture tanks or similar environments, and the importance of this relationship in natural habitats is often disregarded. Here, we describe the first record of high infestation of ciliates on Aegla serrana in South America and conduct a brief morphological and phylogenetic characterization of these ciliates. Our findings confirm that cases of high infestation of ciliates on metazoans can indeed occur in natural environments.

Background: Declining vaccination coverage (VC) and vaccine hesitancy among medical students are global challenges. These challenges reflect individual and logistical barriers to a sufficient adherence toward essential vaccines for healthcare professionals, as well as presenting a need for educational strategies during undergraduate training. Methods: This is a prospective study for evaluating VC rates, sociodemographic associations, and the vaccine confidence among medical students at a federal university in Brazil. The data collection included questionnaires and individual analyses of the participants’ vaccination records. Results: A total of 237 medical students from all six years of an undergraduate program participated, of whom 124 (52.3%) had a vaccination record. Although the majority considered the vaccines to be “Completely Safe” (86.9%), the VC rates for complete vaccination schedules were relatively low, ranging from 87.9% (hepatitis B vaccine) to 3.2% (meningococcal B vaccine), including the vaccines from the National Immunization Program (NIP) and the private sector. Higher VC rates were found to occur among students in the final years of their undergraduate studies, in those from families with higher monthly incomes, and those from private secondary schools. Conclusions: Given the low VC rates among medical students, other factors in addition to vaccine confidence may be determinants, thus highlighting the importance of reviewing policies for the inclusion of priority groups in the NIP and in implementing educational interventions during undergraduate training.

Summary: In the Americas, one decade following its emergence in 2013, chikungunya virus (CHIKV) continues to spread and cause epidemics across the region. To date, 3.7 million suspected and laboratory-confirmed chikungunya cases have been reported in 50 countries or territories in the Americas. Here, we outline the current status and epidemiological aspects of chikungunya in the Americas and discuss prospects for future research and public health strategies to combat CHIKV in the region.

ABSTRACT Sesame ( Sesamum indicum L.) oilseed needs to be studied in relation to the production of biodiesel and its application as an alternative to the soybean biodiesel used in Brazil. The aim of this study was to evaluate biodiesel blends (B0, B5, B10, B20, B40, B80, and B100) of sesame in the performance and emissions of engines. A few studies of sesame biodiesel in engines were found in the literature imparts novelty to the current study. The fuels were tested in a 5-kVA generator engine at loads ranging from 1000 to 6000 W. The results of the analysis showed that the increase in the sesame biodiesel blend caused a reduction in the specific fuel consumption (SFC), carbon monoxide (CO) emissions, and an increase in nitrogen oxide (NOx) emissions. The SFC of sesame biodiesel (B100) was 11% higher than that of diesel (B0). CO emissions increased 39% for diesel (B0) compared to sesame biodiesel (B100). However, B20 showed lower SFC than diesel and lower gas emissions than B80 and B100 blends. Therefore, sesame biodiesel, especially up to B20, is a viable alternative for the partial replacement of conventional diesel. Sesame biodiesel should be considered as a promising candidate for alternative fuels.

ABSTRACT Different mechanical configurations of diesel engines, when powered by biogas, can influence engine performance and efficiency. Consequently, this study aims to evaluate various generator engines to determine the optimal configuration for a distributed generation unit at a swine farm. In this study, we assess the biogas consumption, specific consumption, and efficiency of five biogas generator engines. Three engines (A, D, and E) were provided by Biogás Motores Estacionários, while the remaining two (B and C) were provided by Electro Hager models and MWM, respectively. The biogas used was produced in plug flow biodigesters (piston flow) by the treatment of swine waste. Engine A, provided by Biogás Motores Estacionários, exhibited specific consumption at a power of 30 kW (low load) similar to engines B and C. However, engines B and C displayed higher efficiencies at high loads compared to engines A, D, and E, attributed to their advanced fuel injection and control systems.

Antibiotic resistance challenges the treatment of bacterial biofilm-related infections, but the use of nanoparticles as a treatment is a promising strategy to overcome bacterial infections. This study applied nitrogen-doped titanium dioxide (N–TiO2) conjugated with folic acid (FA) on biofilm-forming resistant bacteria. The photocatalytic effect of TiO2 nanoparticles (NPs) was studied under ultraviolet (UV), visible light, and dark conditions at 60, 120, and 180 min against planktonic cells and biofilms of Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), and Pseudomonas aeruginosa. TiO2 NPs were in the anatase phase, spherical shaped with sizes of 10-13 nm, and effectively doped and conjugated with N and FA. The FA-conjugated nanoparticles (N–TiO2-FA and FA-TiO2) were shown to have a bactericidal effect on all bacteria between 60 and 180 min under UV and visible light conditions. Concerning biofilms, N–TiO2-FA was shown to have a highly disruptive effect on all bacterial biofilms under UV irradiation at 180 min. Meanwhile, the nanoparticles did not show DNA damaging potential and they had no cytostatic effect, indicating that these NPs are biocompatible. In sum, nanoparticle conjugation with FA promoted photocatalytic effectiveness, revealing the promise this nanomaterial holds as a biocompatible antimicrobial agent.

This study aimed to assess the impact of palm oil deodorizer distillate (POD) on the ruminal environment, including (i) microbial community, (ii) ruminal degradability, and (iii) apparent digestibility in sheep. The data used were derived from twenty rumen-cannulated sheep fed five isoproteic and isofiber diets based on elephant grass (Pennisetum purpureum Schum. cv. Roxo) silage supplemented with 0, 25, 50, 75, or 100 g kg−1 POD on a dry matter (DM) basis. Rumen fluid samples were collected three hours after feeding directly from the ventral sac of the rumen via a cannula and then subjected to DNA extraction, which was subsequently used for 16S rDNA amplification, followed by sequencing and diversity analysis. In this study, the microbial diversity was dominated by Bacteroidetes and Firmicutes, followed by Euryarchaetoa, Actinobacteria, and Tenericutes, in the ruminal environment, and was slightly modified when supplemented with the POD up to 100 g/kg (10%), leading to only a slight decrease in the diversity index. The ruminal degradability, ruminal fermentation parameters, and apparent digestibility were slightly compromised by the inclusion of up to 25 g of POD per kg of DM, and larger inclusions interfered with the ruminal degradability of fibrous fractions and the apparent digestibility of dry matter. This lipid supplement showed good results for feeding sheep and is an inexpensive and abundant alternative in the regional market.

The Sigatoka disease complex (SDC), caused by Mycosphaerella fijiensis (Mf) and M. musicola (Mm), comprises the most destructive fungal leaf streak and spot diseases of commercial banana crops worldwide. In Brazil, the site-specific succinate dehydrogenase inhibitor (SDHI) fungicides labeled for SDC management since 2014 present a high risk for the emergence of resistance if deployed intensively and solo. Our study determined the levels of sensitivity to boscalid and fluxapyroxad in four populations of the SDC pathogens sampled in 2020 from three distinct geographical regions under contrasting fungicide programs. Resistance, defined as EC50 values exceeding 20 µg mL−1, was prevalent at 59.7% for fluxapyroxad and 94.0% for boscalid. Only 1.5% of isolates exhibited sensitivity to both fungicides. We also assessed the changes in the corresponding fungicide target protein-encoding genes (SdhB, C, and D). None of the target site alterations detected were associated with reduced sensitivity. A second SdhC paralog was also analyzed, but target alterations were not found. However, MDR (multidrug resistance) was detected in a selection of isolates. Further monitoring for Sdh target mutations will be important, but an important role for other resistance mechanisms such as the presence of additional Sdh paralogs and MDR cannot be ruled out. These results highlight the importance of implementing sound anti-resistance management strategies when SDHI fungicides are deployed for the management of SDC.

The relationship between phosphorus (P) availability and water restriction was explored in this study, focusing on its impact on phosphorus use efficiency (PUE) and water use efficiency (WUE) in various bromegrass (Bromus spp.) genotypes. Under controlled conditions, five bromegrass genotypes, as well as one ryegrass (Lolium perenne) cultivar, were compared by subjecting them to two P levels and two watering regimes. It was determined that combining water and phosphorus limitations led to reduced plant productivity. Initially, the ryegrass outperformed the bromegrass, but this result declined over time, while bromegrass exhibited consistent stability. Notably, under P and water stress, enhanced root development was observed in bromegrass compared to that in ryegrass. Distinct patterns of PUE and WUE allowed for the categorization of bromegrass genotypes into three groups. Genotype 3457 emerged as the most efficient, scoring 20 out of 24, while Pro 94-49 A achieved a score of only 10 out of 24. This study suggests that the drought resilience of bromegrass may be linked to increased root growth during the early vegetative stages, which potentially facilitates improved P acquisition. However, further validation through long-term field experiments is needed. The insights from this study are potentially valuable for use in shaping plant breeding programs by revealing the plant adaptation mechanisms for both P and water absorption.

Abstract Obesity represents a global health problem and is characterized by metabolic dysfunctions and a low-grade chronic inflammatory state, which can increase the risk of comorbidities, such as atherosclerosis, diabetes and insulin resistance. Here we tested the hypothesis that the genetic deletion of metabotropic glutamate receptor 5 (mGluR5) may rescue metabolic and inflammatory features present in BACHD mice, a mouse model of Huntington’s disease (HD) with an obese phenotype. For that, we crossed BACHD and mGluR5 knockout mice (mGluR5−/−) in order to obtain the following groups: Wild type (WT), mGluR5−/−, BACHD and BACHD/mGluR5−/− (double mutant mice). Our results showed that the double mutant mice present decreased body weight as compared to BACHD mice in all tested ages and reduced visceral adiposity as compared to BACHD at 6 months of age. Additionally, 12-month-old double mutant mice present increased adipose tissue levels of adiponectin, decreased leptin levels, and increased IL-10/TNF ratio as compared to BACHD mice. Taken together, our preliminary data propose that the absence of mGluR5 reduce weight gain and visceral adiposity in BACHD mice, along with a decrease in the inflammatory state in the visceral adipose tissue (VAT), which may indicate that mGluR5 may play a role in adiposity modulation.

Several studies have shown the development of electrochemical biosensors based on enzymes immobilized in metal–organic frameworks (MOFs). Although enzymes have unique properties, such as efficiency, selectivity, and environmental sustainability, when immobilized, these properties are improved, presenting significant potential for several biotechnological applications. Using MOFs as matrices for enzyme immobilization has been considered a promising strategy due to their many advantages compared to other supporting materials, such as larger surface areas, higher porosity rates, and better stability. Biosensors are analytical tools that use a bioactive element and a transducer for the detection/quantification of biochemical substances in the most varied applications and areas, in particular, food, agriculture, pharmaceutical, and medical. This review will present novel insights on the construction of biosensors with materials based on MOFs. Herein, we have been highlighted the use of MOF for biosensing for biomedical, food safety, and environmental monitoring areas. Additionally, different methods by which immobilizations are performed in MOFs and their main advantages and disadvantages are presented.

Agroforestry systems (AFSs) have gained recognition as a land use strategy to address food security and climate change. They involve intentionally cultivating trees alongside crops and/or animals. AFSs cover approximately 5% of the global forest area and promote sustainable soil conservation, including soil organic carbon (C) sequestration (CSEQ). In some areas of Chile, AFSs are used to preserve the ecological value of native forests. This study evaluates the effects of two AFSs, namely, an agroforest for fodder production (AGROFRST) and Silvopastoral (SPS), within a degraded native forest (Nothofagus obliqua sp.). The evaluation focuses on their impact on CSEQ capacity and soil quality (SQ), using soil quality indexes (SQIs) derived from 30 soil quality indicators (SINDs) related to physical, chemical, and microbiological properties at two depths (0–5 and 5–20 cm). The results for the total depth analyzed (0–20 cm) indicate an average CSEQ of 6.88 and 4.83 Mg C yr−1 and a global SQI of 37.8% and 31.0% for AGROFRST and SPS, respectively. Among the thirteen SINDs that demonstrated significant differences (p < 0.05), five were associated with the considered depths (P+, Ca2+, S, ECEC, and AlSAT), three differed between AGROFRST and SPS (BD, NH4+, NO3−), while SOC, K+, and Mg2+ varied across all conditions (e.g., combinations of systems and depths), and β-GLU and NMIN differed in a single condition. However, almost all 30 SINDs analyzed showed higher values at the 0–5 cm depth, indicating the positive effects of soil organic matter (SOM)/SOC additions. Significant interactions (Pearson’s correlation) revealed that SOC correlated with most SINDs (e.g., N, NH4+, P+, K+, Ca2+, Mg2+, S, ECEC, NMIN). These findings suggest that both AGROFRST and SPS systems have similar capabilities in restoring the ecological value of native Nothofagus forests while providing conditions for productive and complementary use. This sustainable option offers opportunities for cattle production alongside ecological restoration efforts and provides a possible strategy to generate public policies related to the ecosystem services of agroforestry systems.

Abstract Background We report a genomic surveillance of SARS-CoV-2 lineages circulating in Paraná, southern Brazil, from March 2020 to April 2021. Our analysis, based on 333 genomes, revealed that the first variants detected in the state of Paraná in March 2020 were the B.1.1.33 and B.1.1.28 variants. The variants B.1.1.28 and B.1.1.33 were predominant throughout 2020 until the introduction of the variant P.2 in August 2020 and a variant of concern (VOC), Gamma (P.1), in January 2021. The VOC Gamma, a ramification of the B.1.1.28 lineage first detected in Manaus (northern Brazil), has grown rapidly since December 2020 and was thought to be responsible for the deadly second wave of COVID-19 throughout Brazil. Methods The 333 genomic sequences of SARS-CoV-2 from March 2020 to April 2021 were generated as part of the genomic surveillance carried out by Fiocruz in Brazil Genomahcov Fiocruz. SARS-CoV-2 sequencing was performed using representative samples from all geographic areas of Paraná. Phylogenetic analyses were performed using the 333 genomes also included other SARS-CoV-2 genomes from the state of Paraná and other states in Brazil that were deposited in the GISAID. In addition, the time-scaled phylogenetic tree was constructed with up to 3 random sequences of the Gamma variant from each state in Brazil in each month of 2021. In this analysis we also added the sequences identified as the B.1.1.28 lineage of the Amazonas state and and the Gamma-like-II (P.1-like-II) lineage identified in different regions of Brazil. Results Phylogenetic analyses of the SARS-CoV-2 genomes that were previously classified as the VOC Gamma lineage by WHO/PANGO showed that some genomes from February to April 2021 branched in a monophyletic clade and that these samples grouped together with genomes recently described with the lineage Gamma-like-II. Additionally, a new mutation (E661D) in the spike (S) protein has been identified in nearly 10% of the genomes classified as the VOC Gamma from Paraná in March and April 2021.Finally, we analyzed the correlation between the lineage and the Gamma variant frequency, age group (patients younger or older than 60 years old) and the clinical data of 86 cases from the state of Paraná. Conclusions Our results provided a reliable picture of the evolution of the SARS-CoV-2 pandemic in the state of Paraná characterized by the dominance of the Gamma strain, as well as a high frequencies of the Gamma-like-II lineage and the S:E661D mutation. Epidemiological and genomic surveillance efforts should be continued to unveil the biological relevance of the novel mutations detected in the VOC Gamma in Paraná.

The study of the concentration dynamics of a pollutant substance in a fluid is a classic problem of fluid mechanics given by the transport equation $u_{t}+cu_{x}=0$, where $u=u(x,t)$ denotes the pollutant concentration along a horizontal pipe of a fixed cross-section in the positive $x$ direction at he time $t>0$ and $c$ represents the fluid propagation velocity. In view of that, the velocity of propagation of the fluid is a physical quantity, obtained, generally in an approximate form, which makes such quantity uncertain. In this paper, we propose to obtain the concentration when the constant $c$ represents the fuzzy set. The concentration was obtained by using the Zadeh's Extension Principle. Through the concentration obtained, we analyze the influence of uncertainty on the fluid propagation velocity in the concentration dynamics and explore possible practial applications in case-studies of engineering, environmental and soil sciences.

Abstract We consider transport properties of a hybrid device composed by a quantum dot placed between normal and superconducting reservoirs, and coupled to a Majorana nanowire: a topological superconducting segment hosting Majorana bound states (MBSs) at the opposite ends. It is demonstrated that if highly nonlocal and nonoverlapping MBSs are formed in the system, the zero-bias Andreev conductance through the dot exhibits characteristic isoconductance profiles with the shape depending on the spin asymmetry of the coupling between the dot and the topological superconductor. Otherwise, for overlapping MBSs with less degree of nonlocality, the conductance is insensitive to the spin polarization and the isoconductance signatures disappear. This allows to propose an alternative experimental protocol for probing the nonlocality of the MBSs in Majorana nanowires.

FMOC-Cl is a derivatizing agent widely used both in N-protection reactions of α-amino acids as well as in quantification and detection of amino acids and related molecules using HPLC-PDA or fluorescence analysis. In this work we studied the reaction of FMOC-Cl with gabapentin. However, it was observed that the degradation of gabapentin-N-FMOC and FMOC-Cl to FMOC-OH occur simultaneously in presence of the basic reaction medium which is specific for gabapentin derivatization. The degradation reaction is accelerated in temperatures above 45 °C and also by the time of contact of the reactant and product with basic medium. In this study, gabapentin derivatization reactions were shown to be cleaner and more efficient when an FMOC-Cl/gabapentin ratio of 2:1 was used, at 25 °C over a period of 15 minutes.

Abstract Palygorskite (Pal) is a natural clay mineral with fibrous morphology and high surface area. Depending on the geological origin, it presents impurities, such as quartz and carbonates, which can harm some of its properties. Therefore, this work seeks to define a viable methodology for the purification of a Brazilian Pal. Two types of mixing processes (sonication and milling) and two types of dispersing agents (sodium silicate and sodium polyacrylate) were investigated. In addition, a subsequent acid activation with hydrogen peroxide and sulfuric acid was performed for complete purification. The viability of the purification of Pal was confirmed by X-ray diffraction, X-ray fluorescence, and thermogravimetric analyses. The sonication mixture process and the use of sodium polyacrylate as a dispersing agent were more effective. In addition, BET analysis showed an increase in the surface area of Pal, and scanning electron microscopy confirmed the permanence of its fibrous morphology after the purification steps.

Transplacental transmission of Zika virus (ZIKV) during early pregnancy may lead to several neurological alterations, known as congenital Zika syndrome. We have previously shown that intravaginal infection of immunocompetent dams with ZIKV during the early stage of pregnancy triggers neuroinflammation in fetuses, characterized by increased brain expression of inflammatory factors, including IL-6, IL-18, CCL2, CXCL1, and CXCL10. The present study sought to understand the long-term consequences of these early cerebral alterations. For that, pregnant FVB/NJ immunocompetent females were infected intravaginally on the gestational day (GD) 4.5 and experiments were performed when offspring reached between 4 to 5 months of age. The exposure to ZIKV promoted significant neuronal cell loss detected in the CA1 region of the hippocampus of adult mice. However, only females showed reduced expression levels of brain-derived neurotrophic factor (BDNF), postsynaptic density protein 95 (PSD95), and syntaxin-1A, which are important genes related to neuronal function and synaptic plasticity. Moreover, the protein levels of the pre- and postsynaptic markers, SNAP25 and PSD95, respectively, were decreased exclusively in ZIKV-exposed female mice, indicating that ZIKV exposure in utero induces synaptic loss. Additionally, only females exposed to ZIKV showed risk-taking behavior and hippocampal-dependent spatial memory deficit. Together, these results demonstrate that intravaginal infection of mice on GD4.5 induces neurological alterations in the offspring detectable in their adulthood and that female mice, rather than male mice, are more susceptible to ZIKV-induced neurobehavioral alterations. Summary Statement In utero exposure to ZIKV leads to decreased number of neurons in adult mice. Female mice exposed to ZIKV in utero exhibit lower levels of BDNF, a decrease in synaptic markers, memory deficits, and risk-taking behavior during adulthood.

ABSTRACT Flaviviruses are important human pathogens worldwide. Diagnostic testing for these viruses is difficult because many of the pathogens require specialized biocontainment. To address this issue, we generated 39 virus-like particle (VLP)- and nonstructural protein 1 (NS1)-secreting stable cell lines in HEK-293 cells of 13 different flaviviruses, including dengue, yellow fever, Japanese encephalitis, West Nile, St. Louis encephalitis, Zika, Rocio, Ilheus, Usutu, and Powassan viruses. Antigen secretion was stable for at least 10 cell passages, as measured by enzyme-linked immunosorbent assays and immunofluorescence assays. Thirty-five cell lines (90%) had stable antigen expression over 10 passages, with three of these cell lines (7%) increasing in antigen expression and one cell line (3%) decreasing in antigen expression. Antigen secretion in the HEK-293 cell lines was higher than in previously developed COS-1 cell line counterparts. These antigens can replace current antigens derived from live or inactivated virus for safer use in diagnostic testing. IMPORTANCE Serological diagnostic testing for flaviviral infections is hindered by the need for specialized biocontainment for preparation of reagents and assay implementation. The use of previously developed COS-1 cell lines secreting noninfectious recombinant viral antigen is limited due to diminished antigen secretion over time. Here, we describe the generation of 39 flaviviral virus-like particle (VLP)- and nonstructural protein 1 (NS1)-secreting stable cell lines in HEK-293 cells representing 13 medically important flaviviruses. Antigen production was more stable and statistically higher in these newly developed cell lines than in their COS-1 cell line counterparts. The use of these cell lines for production of flaviviral antigens will expand serological diagnostic testing of flaviviruses worldwide.

Surgical site infection risk continues to increase due to lack of efficacy in current standard of care drugs. New methods to treat or prevent antibiotic-resistant bacterial infections are needed. Multivalent Adhesion Molecules (MAM) are bacterial adhesins required for virulence. We developed a bacterial adhesion inhibitor using recombinant MAM fragment bound to polymer scaffold, mimicking MAM7 display on the bacterial surface. Here, we test MAM7 inhibitor efficacy to prevent Gram-positive and Gram-negative infections. Using a rodent model of surgical infection, incision sites were infected with antibiotic-resistant bioluminescent strains of Staphylococcus aureus or Pseudomonas aeruginosa. Infections were treated with MAM7 inhibitor or control suspension. Bacterial abundance was quantified for nine days post infection. Inflammatory responses and histology were characterized using fixed tissue sections. MAM7 inhibitor treatment decreased burden of S. aureus and P. aeruginosa below detection threshold. Bacterial load of groups treated with control were significantly higher than MAM7 inhibitor-treated groups. Treatment with inhibitor reduced colonization of clinically-relevant pathogens in an in vivo model of surgical infection. Use of MAM7 inhibitor to block initial adhesion of bacteria to tissue in surgical incisions may reduce infection rates, presenting a strategy to mitigate overuse of antibiotics to prevent surgical site infections.

Zika virus (ZIKV) infection during pregnancy is associated with a congenital syndrome. Although the virus can be detected in human placental tissue and sexual transmission has been verified, it is not clear how the virus reaches the fetus. Despite the emerging severity caused by ZIKV infection, no specific prophylactic and/or therapeutic treatment is available. The aim of the present study was to evaluate the effectiveness antiviral of nitazoxanide (NTZ) in two important congenital transmission targets: (i) a primary culture of human placental chorionic cells, and (ii) human cervical epithelial cells (C33-A) infected with Brazilian ZIKV strain. Initially, NTZ activity was screened in ZIKV infected Vero cells under different treatment regimens with non-toxic drug concentrations for 48 h. Antiviral effect was found only when the treatment was carried out after the viral inoculum. A strong effect against the dengue virus serotype 2 (DENV-2) was also observed suggesting the possibility of treating other Flaviviruses. Additionally, it was shown that the treatment did not reduce the production of infectious viruses in insect cells (C6/36) infected with ZIKV, indicating that the activity of this drug is also related to host factors. Importantly, we demonstrated that NTZ treatment in chorionic and cervical cells caused a reduction of infected cells in a dose-dependent manner and decreased viral loads in up to 2 logs. Pre-clinical in vitro testing evidenced excellent therapeutic response of infected chorionic and cervical cells and point to future NTZ activity investigation in ZIKV congenital transmission models with the perspective of possible repurposing of NTZ to treat Zika fever, especially in pregnant women.

Abstract Background Cancer trials often incorporate intensive imaging with Magnetic Resonance Imaging (MRI) and Positron Emission Tomography with Computerised Tomography (PET/CT), which can be physically and mentally exhausting for patients. This questionnaire study aimed to determine the aspects of imaging that affect a patient’s decision to participate in clinical trials in order to inform the design of future trials that utilise imaging. This should achieve greater patient compliance and improve the patient experience. Method A detailed questionnaire assessing patient expectation and acceptability of imaging within clinical trials was developed in collaboration with two patient representatives. The questionnaire addressed the influence of scan type, length, frequency, scheduling, invasiveness and staff support on acceptability of imaging. It was applied to three patient groups. Group 1 consisted of patients newly recruited to studies with imaging, Group 2 consisted of previous participants in studies with imaging and Group 3 consisted of patients having imaging for clinical care. Results One hundred ninety six patients completed the questionnaires (Group 1:47; Group 2: 50 and Group 3: 99). The use of ionising radiation and number of scans required were identified as negative influences on decision to participate by 25% of Group 3 but only by 6% of Groups 1 and 2. Scan duration >30mins was perceived as a negative factor for decision to participate by all Groups (12–22%). Good communication provided by researchers in terms of discussing the study before and after reading study materials was a key factor in influencing decision to participate (> 50% in Groups 1 and 2 and > 20% in Group 3). Conclusion Factors relating to imaging procedures within clinical trials that affect participation have been identified with communication around study materials as the key determinant. These data will be used to influence the development of future research protocols. Modification of imaging requirements within clinical trials will improve patient tolerance and acceptability and is likely to raise recruitment.
