Cognitive Arousal, frequently elicited by environmental stressors that exceed personal coping resources, manifests in measurable physiological markers, notably in galvanic skin responses. This effect is prominent in cognitive tasks such as composition, where fluctuations in these biomarkers correlate with individual expressiveness. It is crucial to understand the nexus between cognitive arousal and expressiveness. However, there has not been a concrete study that investigates this inter-relation concurrently. Addressing this, we introduce an innovative methodology for simultaneous monitoring of these elements. Our strategy employs Bayesian analysis in a multi-state filtering format to dissect psychomotor performance (captured through typing speed), galvanic skin response or skin conductance (SC), and heart rate variability (HRV). This integrative analysis facilitates the quantification of expressive behavior and arousal states. At the core, we deploy a state-space model connecting one latent psychological arousal condition to neural activities impacting sweating (inferred through SC responses) and another latent state to expressive behavior during typing. These states are concurrently evaluated with model parameters using an expectation-maximization algorithms approach. Assessments using both computer-simulated data and experimental data substantiate the validity of our approach. Outcomes display distinguishable latent state patterns in expressive typing and arousal across different computer software used in office management, offering profound implications for Human-Computer Interaction (HCI) and productivity analysis. The comparative studies of aging have established a negative correlation between Gompertz postnatal growth constant and maximum lifespan across mammalian species, but the underlying physiological mechanism remains unclear. This study shows that the Gompertz growth constant can be decomposed into two energetic components, mass-specific metabolic rate and the energetic cost of biosynthesis, and that after controlling the former as a confounder, the negative correlation between growth constant and lifespan still exists due to a 100-fold variation in the latter, revealing that the energetic cost of biosynthesis is a link between growth and longevity in mammals. Previously, the energetic cost of biosynthesis has been thought to be a constant across species and therefore was not considered a contributor to the variation in any life history traits, such as growth and lifespan. This study employs a recently proposed model based on energy conservation to explain the physiological effect of the variation in this energetic cost on the aging process and illustrates its role in linking growth and lifespan. The conventional life history theory suggested a tradeoff between growth and somatic maintenance, but the findings in this study suggest that allocating more energy to biosynthesis may enhance the somatic maintenance and extend lifespan and, hence, reveal a more complex nature of the tradeoff. Amaranth species are C4 plants that are rich in betalains, and they are tolerant to salinity stress. A small family of plant-specific TCP transcription factors are involved in the response to salt stress. However, it has not been investigated whether amaranth TCP1 is involved in salt stress. We elucidated that the growth and physiology of amaranth were affected by salt concentrations of 50–200 mmol·L−1 NaCl. The data showed that shoot and root growth was inhibited at 200 mmol·L−1, while it was promoted at 50 mmol·L−1. Meanwhile, the plants also showed physiological responses, which indicated salt-induced injuries and adaptation to the salt stress. Moreover, AtrTCP1 promoted Arabidopsis seed germination. The germination rate of wild-type (WT) and 35S::AtrTCP1-GUS Arabidopsis seeds reached around 92% by the seventh day and 94.5% by the second day under normal conditions, respectively. With 150 mmol·L−1 NaCl treatment, the germination rate of the WT and 35S::AtrTCP1-GUS plant seeds was 27.0% by the seventh day and 93.0% by the fourth day, respectively. Under salt stress, the transformed 35S::AtrTCP1 plants bloomed when they grew 21.8 leaves after 16.2 days of treatment, which was earlier than the WT plants. The transformed Arabidopsis plants flowered early to resist salt stress. Heart rate variability (HRV), a measure of the fluctuations in the intervals between consecutive heartbeats, is an indicator of changes in the autonomic nervous system. A chronic reduction in HRV has been repeatedly linked to clinical depression. However, the chronological and mechanistic aspects of this relationship, between the neural, physiological, and psychopathological levels, remain unclear. In this review we present evidence by which changes in HRV might precede the onset of depression. We describe several pathways that can facilitate this relationship. First, we examine a theoretical model of the impact of autonomic imbalance on HRV and its role in contributing to mood dysregulation and depression. We then highlight brain regions that are regulating both HRV and emotion, suggesting these neural regions, and the Insula in particular, as potential mediators of this relationship. We also present additional possible mediating mechanisms involving the immune system and inflammation processes. Lastly, we support this model by showing evidence that modification of HRV with biofeedback leads to an improvement in some symptoms of depression. The possibility that changes in HRV precede the onset of depression is critical to put to the test, not only because it could provide insights into the mechanisms of the illness but also because it may offer a predictive and diagnostic physiological marker for depression. Simple Summary: Delphinids are top predators and display long lifespans, making them susceptible to accumulating harmful environmental contaminants within their tissues, which may lead to potential adverse effects on their physiology and overall health. This study aimed to investigate exposure to phthalates in aquarium-based bottlenose dolphins and killer whales and explore potential correlations with individual physiological responses while accounting for sex, age, and reproductive stage. We found that all individuals were exposed to phthalates, including newborn dolphins. Phthalates were also correlated with aldosterone concentrations in killer whales, suggesting that this class of contaminants may exert disruptive effects on the endocrine system and metabolism of dolphins. This study could help us better understand the physiological mechanisms and effects of phthalate exposure in delphinids, which could lead to stricter contaminant discharge regulations, improving both human and aquatic/marine health. There is growing concern about the potential adverse health effects of phthalates (PAEs) on human health and the environment due to their extensive use as plasticizers and additives in commercial and consumer products. In this study, we assessed PAE concentrations in serum samples from aquarium-based delphinids (Tursiops truncatus, n = 36; Orcinus orca, n = 42) from California, Florida, and Texas, USA. To better understand the physiological effects of phthalates on delphinids, we also explored potential correlations between phthalates and the biomarkers aldosterone, cortisol, corticosterone, hydrogen peroxide, and malondialdehyde while accounting for sex, age, and reproductive stage. All PAEs were detected in at least one of the individuals. ΣPAE ranges were 5.995–2743 ng·mL−1 in bottlenose dolphins and 5.372–88,675 ng·mL−1 in killer whales. Both species displayed higher mean concentrations of DEP and DEHP. PAEs were detected in newborn delphinids, indicating transference via placenta and/or lactation. Linear mixed model results indicated significant correlations between aldosterone, month, location, status, and ΣPAEs in killer whales, suggesting that aldosterone concentrations are likely affected by the cumulative effects of these variables. This study expands on the knowledge of delphinid physiological responses to PAEs and may influence management and conservation decisions on contamination discharge regulations near these species. Simple Summary: Galloanseres are one of the most widely distributed groups of birds, which occupy different ecological niches and exhibit different evolutionary trends. Mitochondria are widely used in phylogenetic analysis and ecological research due to unique genetic mechanisms and significant physiological functions. Adapting to different environments requires meeting varying energy demands, which is closely related to mitochondria. In this study, we constructed two complete mitogenomes of Aythya baeri and Aythya marila. The phylogenetic and divergence time analysis based on the mitochondrial PCGs were conducted to elucidate the evolutionary process of species within Galloanseres. Additionally, the analysis of selective pressures between the two sister clades, Galliformes and Anseriformes, revealed that different evolutionary directions have shaped distinct evolutionary patterns of mitochondrial genes. The two existing clades of Galloanseres, orders Galliformes (landfowl) and Anseriformes (waterfowl), exhibit dramatically different evolutionary trends. Mitochondria serve as primary sites for energy production in organisms, and numerous studies have revealed their role in biological evolution and ecological adaptation. We assembled the complete mitogenome sequences of two species of the genus Aythya within Anseriformes: Aythya baeri and Aythya marila. A phylogenetic tree was constructed for 142 species within Galloanseres, and their divergence times were inferred. The divergence between Galliformes and Anseriformes occurred ~79.62 million years ago (Mya), followed by rapid evolution and diversification after the Middle Miocene (~13.82 Mya). The analysis of selective pressure indicated that the mitochondrial protein-coding genes (PCGs) of Galloanseres species have predominantly undergone purifying selection. The free-ratio model revealed that the evolutionary rates of COX1 and COX3 were lower than those of the other PCGs, whereas ND2 and ND6 had faster evolutionary rates. The CmC model also indicated that most PCGs in Anseriformes exhibited stronger selective constraints. Members of the GPCR superfamily have a wide variety of physiological roles and are therefore valuable targets for developing effective medicines. However, within this superfamily are receptors that are less well characterized and remain orphans, including GPR84. This receptor is stimulated by ligands derived from dietary nutrients, specifically medium chain fatty acids (C9–14), and novel synthetic agonists. There are data demonstrating the role of GPR84 in inflammatory pathways, in addition to emerging data suggesting a key role for GPR84 as a nutrient‐sensing GPCR involved in metabolism by sensing energy load via nutrient exposure and subsequent signalling leading to modulation of food intake. Exploring GPR84 pharmacology, its localization and what drives its expression has revealed multiple roles for this receptor. Here, we will reflect on these various roles of GRP84 demonstrated thus far, primarily by exploring data from pre‐clinical and clinical studies in various physiological systems, with a specific focus on the gastrointestinal tract. Background: Thermal stress in subtropical regions is a major limiting factor in beef cattle production systems with around $369 million being lost annually due to reduced performance. Heat stress causes numerous physiological and behavioral disturbances including reduced feed intake and decreased production levels. Cattle utilize various physiological mechanisms such as sweating to regulate internal heat. Variation in these traits can help identify genetic variants that control sweat gland properties and subsequently allow for genetic selection of cattle with greater thermotolerance. Methods: This study used 2,401 Brangus cattle from two commercial ranches in Florida. Precise phenotypes that contribute to an animal's ability to manage heat stress were calculated from skin biopsies and included sweat gland area, sweat gland depth, and sweat gland length. All animals were genotyped with the Bovine GGP F250K, and BLUPF90 software was used to estimate genetic parameters and for Genome Wide Association Study. Results: Sweat gland phenotypes heritability ranged from 0.17 to 0.42 indicating a moderate amount of the phenotypic variation is due to genetics, allowing producers the ability to select for favorable sweat gland properties. A weighted single-step GWAS using sliding 10 kb windows identified multiple quantitative trait loci (QTLs) explaining a significant amount of genetic variation. QTLs located on BTA7 and BTA12 explained over 1.0% of genetic variance and overlap the ADGRV1 and CCDC168 genes, respectively. The variants identified in this study are implicated in processes related to immune function and cellular proliferation which could be relevant to heat management. Breed of Origin Alleles (BOA) were predicted using local ancestry in admixed populations (LAMP-LD), allowing for identification of markers' origin from either Brahman or Angus ancestry. A BOA GWAS was performed to identify regions inherited from particular ancestral breeds that might have a significant impact on sweat gland phenotypes. Conclusions: The results of the BOA GWAS indicate that both Brahman and Angus alleles contribute positively to sweat gland traits, as evidenced by favorable marker effects observed from both genetic backgrounds. U.S. service members maintain constant situational awareness (SA) due to training and experience operating in dynamic and complex environments. Work examining how military experience impacts SA during visual search of a complex naturalistic environment, is limited. Here, we compare Active Duty service members and Civilians' physiological behavior during a navigational visual search task in an open-world virtual environment (VE) while cognitive load was manipulated. We measured eye-tracking and electroencephalogram (EEG) outcomes from Active Duty (N = 21) and Civilians (N = 15) while they navigated a desktop VE at a self-regulated pace. Participants searched and counted targets (N = 15) presented among distractors, while cognitive load was manipulated with an auditory Math Task. Results showed Active Duty participants reported significantly greater/closer to the correct number of targets compared to Civilians. Overall, Active Duty participants scanned the VE with faster peak saccade velocities and greater average saccade magnitudes compared to Civilians. Convolutional Neural Network (CNN) response (EEG P-300) was significantly weighted more to initial fixations for the Active Duty group, showing reduced attentional resources on object refixations compared to Civilians. There were no group differences in fixation outcomes or overall CNN response when comparing targets versus distractor objects. When cognitive load was manipulated, only Civilians significantly decreased their average dwell time on each object and the Active Duty group had significantly fewer numbers of correct answers on the Math Task. Overall, the Active Duty group explored the VE with increased scanning speed and distance and reduced cognitive re-processing on objects, employing a different, perhaps expert, visual search strategy indicative of increased SA. The Active Duty group maintained SA in the main visual search task and did not appear to shift focus to the secondary Math Task. Background: Posterior Circulation Syndrome (PCS) presents a diagnostic challenge characterized by its variable and nonspecific symptoms. Timely and accurate diagnosis is crucial for improving patient outcomes. This study aims to enhance the early diagnosis of PCS by employing clinical and demographic data and machine learning. This approach targets a significant research gap in the field of stroke diagnosis and management. Methods: We collected and analyzed data from a large national Stroke Registry spanning from January 2014 to July 2022. The dataset included 15,859 adult patients admitted with a primary diagnosis of stroke. Five machine learning models were trained: XGBoost, Random Forest, Support Vector Machine, Classification and Regression Trees, and Logistic Regression. Multiple performance metrics, such as accuracy, precision, recall, F1-score, AUC, Matthew's correlation coefficient, log loss, and Brier score, were utilized to evaluate model performance. Results: The XGBoost model emerged as the top performer with an AUC of 0.81, accuracy of 0.79, precision of 0.5, recall of 0.62, and F1-score of 0.55. SHAP (SHapley Additive exPlanations) analysis identified key variables associated with PCS, including Body Mass Index, Random Blood Sugar, ataxia, dysarthria, and diastolic blood pressure and body temperature. These variables played a significant role in facilitating the early diagnosis of PCS, emphasizing their diagnostic value. Conclusion: This study pioneers the use of clinical data and machine learning models to facilitate the early diagnosis of PCS, filling a crucial gap in stroke research. Using simple clinical metrics such as BMI, RBS, ataxia, dysarthria, DBP, and body temperature will help clinicians diagnose PCS early. Despite limitations, such as data biases and regional specificity, our research contributes to advancing PCS understanding, potentially enhancing clinical decision-making and patient outcomes early in the patient's clinical journey. Interactions between animals and microbes are ubiquitous in nature and strongly impact animal physiology. These interactions are shaped by the host immune system, which responds to infections and contributes to tailor the associations with beneficial microorganisms. In many insects, beneficial symbiotic associations not only include gut commensals, but also intracellular bacteria, or endosymbionts. Endosymbionts are housed within specialized host cells, the bacteriocytes, and are transmitted vertically across host generations. Host–endosymbiont co-evolution shapes the endosymbiont genome and host immune system, which not only fights against microbial intruders, but also ensures the preservation of endosymbionts and the control of their load and location. The cereal weevil Sitophilus spp. is a remarkable model in which to study the evolutionary adaptation of the immune system to endosymbiosis owing to its binary association with a unique, relatively recently acquired nutritional endosymbiont, Sodalis pierantonius. This Gram-negative bacterium has not experienced the genome size shrinkage observed in long-term endosymbioses and has retained immunogenicity. We focus here on the sixteen antimicrobial peptides (AMPs) identified in the Sitophilus oryzae genome and their expression patterns in different tissues, along host development or upon immune challenges, to address their potential functions in the defensive response and endosymbiosis homeostasis along the insect life cycle. Oxygen (O2) binds to hemoglobin (Hb) in the lungs and is then released (dissociated) in the tissues. The Bohr effect is a physiological mechanism that governs the affinity of Hb for O2 based on pH, where a lower pH results in a lower Hb-O2 affinity and higher Hb-O2 dissociation. Hb-O2 affinity and dissociation are crucial for maintaining aerobic metabolism in cells and tissues. Despite its vital role in human physiology, Hb-O2 dissociation measurement is underutilized in basic research and in clinical laboratories, primarily due to the technical complexity and limited throughput of existing methods. We present a rapid Hb-O2 dissociation measurement approach by leveraging the Bohr effect and detecting the optical shift in the Soret band that corresponds to the light absorption by the heme group in Hb. This new method reduces Hb-O2 dissociation measurement time from hours to minutes. We show that Hb deoxygenation can be accelerated chemically at the optimal pH of 6.9. We show that time and pH-controlled deoxygenation of Hb results in rapid and distinct conformational changes in its tertiary structure. These molecular conformational changes are manifested as significant, detectable shifts in Hb's optical absorption spectrum, particularly in the characteristic Soret band (414 nm). We extensively validated the method by testing human blood samples containing normal Hb and Hb variants. We show that rapid Hb-O2 dissociation can be used to screen for and detect Hb-O2 affinity disorders and to evaluate the function and efficacy of Hb-modifying therapies. The ubiquity of optical absorption spectrophotometers positions this approach as an accessible, rapid, and accurate Hb-O2 dissociation measurement method for basic research and clinical use. We anticipate this method's broad adoption will democratize the diagnosis and prognosis of Hb disorders, such as sickle cell disease. Further, this method has the potential to transform the research and development of new targeted and genome-editing-based therapies that aim to modify or improve Hb-O2 affinity. The interaction of root hemi-parasite (sandalwood) with its hosts is crucial for establishing successful plantations under abiotic stresses. In the present study, we explored the best possible host for sandalwood along with its effect on sandalwood physiology in terms of water and nutrients. Interactive effects of host species (Alternanthera sp., Azadirachta indica, Dalbergia sissoo, Melia dubia, and Aquilaria malaccensis) with sandalwood were observed under eight treatments {100% best available water (BAW); 100% BAW + nutrient medium; 50% water deficit; 50% water deficit + nutrient medium; 100% saline water (ECiw 8ds/m); 100% saline water (ECiw 8ds/m) + nutrient medium; 50% water deficit + saline water (ECiw 8ds/m); and 50% water deficit + saline water (ECiw 8ds/m) + nutrient medium}. A significant change in morpho-physiological traits of sandalwood roots was observed under different stress conditions, which were slightly improved through external supply of nutrient medium. Dalbergia sissoo (Shisham) and Melia dubia (Dek) seemed to be the best host plants providing better environment for sandalwood growth and development, i.e., higher plant height (59.7 and 53.68 cm) and collar diameter (3.24 and 3.07 mm) under stresses by maintaining water and ionic balance. Root length is an important parameter that was reduced by 27.58%, 19.22%, and 36.3% under water deficit, salinity, and combined stress of water deficit and salinity. Sandalwood grown with D. sissoo and M. dubia maintained the lowest Ψw (− 1.38 MPa) and Ψs (− 1.47 and − 1.48 MPa), respectively. In addition, sandalwood cultivated with D. sissoo and A. indica had higher accumulation of soluble proteins (0.48 and 0.42 mg/g) and soluble sugars (98.56 and 91.04 mg/g) in their roots. Results also showed that sandalwood roots had higher K+/Na+ with compatible host, i.e., with A. indica (1.85) and D. sissoo (1.83) than other studied hosts. It was also observed that sandalwood plants could not grow and survive alone under stress conditions even with application of nutrient medium. Based on the morphological traits, it was observed that sandalwood grown with hosts, Dalbergia sissoo and Melia dubia, was able to tolerate stress conditions better than other studied hosts. Stillbirth is a major cause of perinatal death in India. Deciphering the cause of stillbirth will help in formulating prevention strategies to bring down the stillbirth rate. Our objective is to explore the antepartum and intrapartum causes and risk factors of stillbirths. Materials and methods: This was a prospective observational study done under WHO-SEARO project. This study included all stillbirths occurring in Safdarjung Hospital from August 2015 to December 2018. Verbal autopsies were done, and thorough antenatal records were documented and analyzed. Results: Out of 1,09,578 deliveries, 2,689 were stillbirths (2.4%). Among these, 1,583 (58.9%) were unsupervised and unbooked pregnancies, whereas 1,106 (41.1%) pregnancies were supervised and booked. Significantly lower percentage of patients in unbooked pregnancies had iron–folic acid intake; significantly higher percentage of mothers had anemia, hypertension, diabetes, and antepartum hemorrhage compared with booked pregnancies. Among antepartum causes, hypertension (23.4%), abruption (18.5%), and infection (3.4%) were the leading causes for stillbirths. Intrapartum causes constituted 39% of total stillbirths; a higher percentage was in the unbooked pregnancies. Approximately, 89% of patients with stillbirth had delayed seeking of healthcare facility. Conclusion: The most common cause of stillbirth was hypertensive disorders of pregnancy followed by abruptio placentae in this cohort. Various maternal-related factors are significantly higher in unbooked pregnancies. Reducing these factors might reduce the prevalence of stillbirth. [ABSTRACT FROM AUTHOR]

Premise: Previous studies have suggested a trade‐off between trichome density (Dt) and stomatal density (Ds) due to shared cell precursors. We clarified how, when, and why this developmental trade‐off may be overcome across species. Methods: We derived equations to determine the developmental basis for Dt and Ds in trichome and stomatal indices (it and is) and the sizes of epidermal pavement cells (e), trichome bases (t), and stomata (s) and quantified the importance of these determinants of Dt and Ds for 78 California species. We compiled 17 previous studies of Dt–Ds relationships to determine the commonness of Dt–Ds associations. We modeled the consequences of different Dt–Ds associations for plant carbon balance. Results: Our analyses showed that higher Dt was determined by higher it and lower e, and higher Ds by higher is and lower e. Across California species, positive Dt–Ds coordination arose due to it–is coordination and impacts of the variation in e. A Dt–Ds trade‐off was found in only 30% of studies. Heuristic modeling showed that species sets would have the highest carbon balance with a positive or negative relationship or decoupling of Dt and Ds, depending on environmental conditions. Conclusions: Shared precursor cells of trichomes and stomata do not limit higher numbers of both cell types or drive a general Dt–Ds trade‐off across species. This developmental flexibility across diverse species enables different Dt–Ds associations according to environmental pressures. Normally, the sleep–wake cycle is synchronized with the body's circadian rhythm, which in turn is synchronized with the 24-h period of sunlight. Circadian changes in key hormones, particularly cortisol, also tend to follow the body's circadian rhythm, masking their involvement in controlling the transition between sleep and wakefulness. In our work, we study the behavior of an extended mathematical model that describes the dynamics of both switching between sleep and wakefulness and the process of cortisol production, including their mutual influence. The main goal of our work is to evaluate the behavior of such an extended model in the light of the phenomenon of spontaneous internal desynchronization (SID). Our results indicate that the influence of the cortisol subsystem on the switch between sleep and wakefulness depends significantly on the strength of this feedback. In particular, the increased contribution of cortisol leads to desynchronization between the sleep–wake cycle and the circadian rhythm. With random deviations in the speed of the homeostatic process, the action of cortisol, on the one hand, increases the predictability of the behavior of physiological markers of the sleep–wake cycle, and on the other hand, can lead to the appearance of "excesses of activity"—days with early awakening and late falling asleep. Hard limestone substrates, which are extensively distributed, are believed to exacerbate drought and increase the difficulty of restoration in vulnerable karst regions. Fissures in such substrates may alleviate the negative effect of drought on plants, but the underlying mechanisms remain poorly understood. In a two-way factorial block design, the growth and photosynthesis of 2-year-old Phoebe zhennan seedlings were investigated in two water availabilities (high versus low) and three stimulated fissure habitat groups (soil, soil-filled fissure and non-soil-filled fissure). Moreover, the fissure treatments included both small and big fissures. Compared to the soil group, the non-soil-filled fissure group had decreased the total biomass, root biomass, total root length, and the root length of fine roots in the soil layer at both water availabilities, but increased net photosynthetic rate (Pn) and retained stable water use efficiency (WUE) at low water availability. However, there were no significant differences between the soil-filled fissure group and soil group in the biomass accumulation and allocation as well as Pn. Nevertheless, the SF group decreased the root distribution in total and in the soil layer, and also increased WUE at low water availability. Across all treatments, fissure size had no effect on plant growth or photosynthesis. Karst fissures filled with soil can alleviate drought impacts on plant root growth, which involves adjusting root distribution strategies and increasing water use efficiency. These results suggest that rock fissures can be involved in long-term plant responses to drought stress and vegetation restoration in rocky mountain environments under global climate change. Parasites represent a ubiquitous threat for most organisms, requiring potential hosts to invest in a range of strategies to defend against infection—these include both behavioural and physiological mechanisms. Avoidance is an essential first line of defence, but this behaviour may show a trade-off with host investment in physiological immunity. Importantly, while environmental stressors can lead to elevated hormones in vertebrates, such as glucocorticoids, that can reduce physiological immunity in certain contexts, behavioural defences may also be compromised. Here, we investigate anti-parasite behaviour and immune responses against a trematode (flatworm) parasite by larval amphibians (tadpoles) exposed or not to a simulated general stressor in the form of exogenous corticosterone. Tadpoles that were highly active in the presence of the trematode infectious stage (cercariae) had lower infection loads, and parasite loads from tadpoles treated only with dechlorinated water were significantly lower than those exposed to corticosterone or the solvent control. However, treatment did not affect immunity as measured through white blood-cell profiles, and there was no relationship between the latter and anti-parasite behaviour. Our results suggest that a broad range of stressors could increase host susceptibility to infection through altered anti-parasite behaviours if they elevate endogenous glucocorticoids, irrespective of physiological immunity effects. How hosts defend themselves against parasitism in the context of multiple challenges represents an important topic for future research, particularly as the risk posed by infectious diseases is predicted to increase in response to ongoing environmental change. PKD1 (polycystin 1) and PKD2 (polycystin 2) are expressed in a variety of different cell types, including arterial smooth muscle and endothelial cells. PKD1 is a transmembrane domain protein with a large extracellular N‐terminus that is proposed to act as a mechanosensor and receptor. PKD2 is a member of the transient receptor potential (TRP) channel superfamily which is also termed TRPP1. Mutations in the genes which encode PKD1 and PKD2 lead to autosomal dominant polycystic kidney disease (ADPKD). ADPKD is one of the most prevalent monogenic disorders in humans and is associated with extrarenal and vascular complications, including hypertension. Recent studies have uncovered mechanisms of activation and physiological functions of PKD1 and PKD2 in arterial smooth muscle and endothelial cells. It has also been found that PKD function is altered in the vasculature during ADPKD and hypertension. We will summarize this work and discuss future possibilities for this area of research. The importance of issues associated with urban/commensal rats and mice (property damage, management costs, and health risks) press upon research on these animals. While the demography of commensal rodents is mostly studied, the need for understanding factors influencing their natural morbidity/mortality is also stressed. In this respect, more attention is expected to be paid to immunity, the physiological mechanism of defense against host survival threats (pathogens, parasites, diseases). Commensal rats and mice carry numerous pathogens that evoke diverse immune responses. The state of immunity in commensal house mice is studied in great detail, owing to the use of laboratory strains in biomedical research. Because commensal rats are, compared to mice, carriers of more zoonotic agents, rats' immunity is studied mainly in that context. Some of these zoonotic agents cause chronic, asymptomatic infections, which justified studies of immunological mechanisms of pathogen tolerance versus clearance regulation in rats. Occurrence of some infections in specific tissues/organs pressed upon analysis of local/regional immune responses and/or immunopathology. A survey of immunological activity/responses in commensal rats is given in this review, with mention of existing data in commensal mice. It should throw some light on the factors relevant to their morbidity and lifespan, supplementing the knowledge of commensal rodent ecology. [ABSTRACT FROM AUTHOR]

Pressure overload-induced hypertrophy compromises cardiac stretch-induced compliance (SIC) after acute volume overload (AVO). We hypothesized that SIC could be enhanced by physiological hypertrophy induced by pregnancy's chronic volume overload. This study evaluated SIC-cardiac adaptation in pregnant women with or without cardiovascular risk (CVR) factors. Thirty-seven women (1st trimester, 1stT) and a separate group of 31 (3rd trimester, 3rdT) women [healthy or with CVR factors (obesity and/or hypertension and/or with gestational diabetes)] underwent echocardiography determination of left ventricular end-diastolic volume (LVEDV) and E/e' before (T0), immediately after (T1), and 15 min after (T2; SIC) AVO induced by passive leg elevation. Blood samples for NT-proBNP quantification were collected before and after the AVO. Acute leg elevation significantly increased inferior vena cava diameter and stroke volume from T0 to T1 in both 1stT and 3rdT, confirming AVO. LVEDV and E/e' also increased immediately after AVO (T1) in both 1stT and 3rdT. SIC adaptation (T2, 15 min after AVO) significantly decreased E/e' in both trimesters, with additional expansion of LVEDV only in the 1stT. NT-pro-BNP increased slightly after AVO but only in the 1stT. CVR factors, but not parity or age, significantly impacted SIC cardiac adaptation. A distinct functional response to SIC was observed between 1stT and 3rdT, which was influenced by CVR factors. The LV of 3rdT pregnant women was hypertrophied, showing a structural limitation to dilate with AVO, whereas the lower LV filling pressure values suggest increased diastolic compliance. Polyploidization is hypothesized to improve the freezing resistance of plants in cold regions. However, adaptive strategies and key physiological mechanisms involved in the freezing resistant ability of polyploids remain unclear. In Actinidia chinensis (kiwifruits), the tetraploids and hexaploids occupy higher altitude habitats with colder climates than the diploids, providing a study system to investigate mechanisms responsible for differentiation in freezing resistance between cytotypes. We characterized environmental conditions of their natural distribution areas, and measured leaf‐level traits of cold damages and water relations at typical sites of each cytotype along an altitudinal gradient. Polyploids showed lower semi‐lethal temperature (LT50) than ice nucleation temperature (INT), reflecting a tolerance strategy to cope with freezing events in the plateau regions. More negative turgor loss points and larger cell elastic modulus of polyploids could help to alleviate damages from freezing‐induced cell dehydration, thus strengthening their tolerance to lower subzero temperatures (lower LT50). The increased supercooling capacity of polyploids (lower INT) might correlate with less extracellular ice formation due to lower osmotic potential at full turgor, apoplastic water fraction and tissue capacitance. Our study uncovers a greater cold tolerance in polyploid kiwifruits than diploids, and suggests the potential linkage between freezing tolerance and water relations. Background: The aim of this study was to examine variations in stress, metabolic, and physiological parameters of horses used in the traditional equestrian team sport of Kök‐Börü in relation to winning and losing outcomes. Material and methods: To accomplish this, blood samples were taken from horses on four different teams who participated in two separate games, both before and after game. These samples were used to measure levels of cortisol, ACTH, beta‐endorphin, adrenaline, noradrenaline, triiodothyronine (T3), and thyroxine (T4) via species‐specific commercial ELISA kits. The autoanalyzer tested biochemical and hematological parameters. The gathered data were then analyzed statistically based on the teams' winning or losing status. Results: The results suggested that winning teams had lower MID, red blood cell, HGB, RDW‐SD, HCT, platelet distribution width, and creatine kinase values post‐game in comparison to their pre‐game state. Conversely, mean corpuscular hemoglobin concentration (MCHC), mean corpuscular hemoglobin (MCH), and CREA values increased in the winning teams' post‐game. Additionally, horses in the winning teams showed a decrease in cortisol, beta‐endorphin, and ACTH levels post‐game but increased levels of adrenaline and T3. Considering the pre‐game values, it was found that GRA and Cl levels were lower in the winning teams. Before the game, adrenaline and T3 levels were higher in the winning teams. No significant difference was observed in post‐game hematological parameters between the teams. However, post‐game K, adrenaline, and noradrenaline levels were higher among the winning teams' horses, while cortisol and beta‐endorphin levels were heightened in horses from the losing side. Conclusion: In conclusion, significant differences were not observed in the distribution of hematological and biochemical parameters of horses following the Kök‐Börü games, regardless of the outcome. Mulberries are vital for the silk industry as the sole natural food for silkworms, but their quality and quantity can be greatly impacted by environmental factors, notably water shortages or droughts. In this study, the proline content and expression levels of the Pyrroline-5-carboxylate reductase (P5CR) gene in four recommended Thai mulberry varieties (Sakhonnakorn, Sakhonnakorn 85, Burirum 60, and Srisaket 84) and one standard drought tolerant variety (SRCM9809-34) were measured under drought stress. Additionally, physiological data and antioxidative enzymatic activities were also examined. The findings revealed that SRCM9809-34, a drought-tolerant variety, had the lowest proline content, followed by Sakhonnakorn 85, Burirum 60, while the highest proline content was observed in Srisaket 84. Although there was no correlation between the expression level of the P5CR gene and proline content, the overall trend in all varieties was the same: proline content increased after drought conditions. Regarding physiological responses, the wilting score showed similar results to proline content, with SRCM9809-34 having the lowest proline content and wilting score. Moreover, SRCM9809-34 exhibited the highest RWC, Pn and WUE values, as well as the lowest level of MDA and H2O2. Our results validated and indicated that SRCM9809-34 is a drought-tolerant variety. From this finding, among the four Thai mulberry varieties, Sakhonnakorn 85 exhibited the highest potential for drought tolerance, and this potential can be enhanced through crossbreeding with SRCM9809-34. Larsucosterol, a potent endogenous epigenetic regulator, has been reported to play a significant role in lipid metabolism, inflammatory responses, and cell survival. The administration of larsucosterol has demonstrated a reduction in lipid accumulation within hepatocytes and the attenuation of inflammatory responses induced by lipopolysaccharide (LPS) and TNFα in macrophages, alleviating LPS- and acetaminophen (ATMP)-induced multiple organ injury, and decreasing mortalities in animal models. Results from phase 1 and 2 clinical trials have shown that larsucosterol has potential as a biomedicine for the treatment of acute and chronic liver diseases. Recent evidence suggests that larsucosterol is a promising candidate for treating alcohol-associated hepatitis with positive results from a phase 2a clinical trial, and for metabolic dysfunction-associated steatohepatitis (MASH) from a phase 1b clinical trial. In this review, we present a culmination of our recent research efforts spanning two decades. We summarize the discovery, physiological and pharmacological mechanisms, and clinical applications of larsucosterol. Furthermore, we elucidate the pathophysiological pathways of metabolic dysfunction-associated steatotic liver diseases (MASLD), metabolic dysfunction-associated steatohepatitis (MASH), and acute liver injuries. A central focus of the review is the exploration of the therapeutic potential of larsucosterol in treating life-threatening conditions, including acetaminophen overdose, endotoxin shock, MASLD, MASH, hepatectomy, and alcoholic hepatitis. Telocytes (TCs) are CD34-positive interstitial cells that have long cytoplasmic projections, called telopodes; they have been identified in several organs and in various species. These cells establish a complex communication network between different stromal and epithelial cell types, and there is growing evidence that they play a key role in physiology and pathology. In many tissues, TC network impairment has been implicated in the onset and progression of pathological conditions, which makes the study of TCs of great interest for the development of novel therapies. In this review, we summarise the main methods involved in the characterisation of these cells as well as their inherent difficulties and then discuss the functional assays that are used to uncover the role of TCs in normal and pathological conditions, from the most traditional to the most recent. Furthermore, we provide future perspectives in the study of TCs, especially regarding the establishment of more precise markers, commercial lineages and means for drug delivery and genetic editing that directly target TCs. Body condition in individuals is intricately linked to behaviour, physiology, and immunity. This study investigated how seasonal factors, such as reproduction and moulting, influence the body condition of 21 tanager species in the Atlantic Forest. Utilising publicly available data, I employed phylogenetic generalised linear mixed models to dissect these influences. Notably, females with brood patches exhibited concurrent body and feather moult, suggesting simultaneous energy-costly constraints. Furthermore, females with brood patches displayed higher body condition, indicating proactive preparation or increased resource availability during breeding. Lastly, individuals moulting body feathers exhibited enhanced body condition, suggesting heightened foraging activity. Excessive extracellular matrix (ECM) deposition is a defining feature of cardiac fibrosis. Most notably, it is characterized by a significant change in the concentration and volume fraction of collagen I, a disproportionate deposition of collagen subtypes, and a disturbed ECM network arrangement, which directly affect the systolic and diastolic functions of the heart. Immune cells that reside within or infiltrate the myocardium, including macrophages, play important roles in fibroblast activation and consequent ECM remodeling. Through both direct and indirect connections to fibroblasts, monocyte-derived macrophages and resident cardiac macrophages play complex, bidirectional, regulatory roles in cardiac fibrosis. In this review, we discuss emerging interactions between fibroblasts and macrophages in physiology and pathologic conditions, providing insights for future research aimed at targeting macrophages to combat cardiac fibrosis. Extracellular vesicles (EVs) are membrane-bound particles released by cells to perform multitudes of biological functions. Owing to their significant implications in diseases, the pathophysiological role of EVs continues to be extensively studied, leading research to neglect the need to explore their role in normal physiology. Despite this, many identified physiological functions of EVs, including, but not limited to, tissue repair, early development and aging, are attributed to their modulatory role in various signaling pathways via intercellular communication. EVs are widely perceived as a potential therapeutic strategy for better prognosis, primarily through utilization as a mode of delivery vehicle. Moreover, disease-associated EVs serve as candidates for the targeted inhibition by pharmacological or genetic means. However, these attempts are often accompanied by major challenges, such as off-target effects, which may result in adverse phenotypes. This renders the clinical efficacy of EVs elusive, indicating that further understanding of the specific role of EVs in physiology may enhance their utility. This review highlights the essential role of EVs in maintaining cellular homeostasis under different physiological settings, and also discusses the various aspects that may potentially hinder the robust utility of EV-based therapeutics. Most individuals with Parkinson's disease experience cognitive decline. Mounting evidence suggests this is partially caused by cholinergic denervation due to α-synuclein pathology in the cholinergic basal forebrain. Alpha-synuclein deposition causes inflammation, which can be measured with free water fraction, a diffusion MRI-derived metric of extracellular water. Prior studies have shown an association between basal forebrain integrity and cognition, cholinergic levels and cognition, and basal forebrain volume and acetylcholine, but no study has directly investigated whether basal forebrain physiology mediates the relationship between acetylcholine and cognition in Parkinson's disease. We investigated the relationship between these variables in a cross-sectional analysis of 101 individuals with Parkinson's disease. Cholinergic levels were measured using fluorine-18 fluoroethoxybenzovesamicol (18F-FEOBV) PET imaging. Cholinergic innervation regions of interest included the medial, lateral capsular and lateral perisylvian regions and the hippocampus. Brain volume and free water fraction were quantified using T1 and diffusion MRI, respectively. Cognitive measures included composites of attention/working memory, executive function, immediate memory and delayed memory. Data were entered into parallel mediation analyses with the cholinergic projection areas as predictors, cholinergic basal forebrain volume and free water fraction as mediators and each cognitive domain as outcomes. All mediation analyses controlled for age, years of education, levodopa equivalency dose and systolic blood pressure. The basal forebrain integrity metrics fully mediated the relationship between lateral capsular and lateral perisylvian acetylcholine and attention/working memory, and partially mediated the relationship between medial acetylcholine and attention/working memory. Basal forebrain integrity metrics fully mediated the relationship between medial, lateral capsular and lateral perisylvian acetylcholine and free water fraction. For all mediations in attention/working memory and executive function, the free water mediation was significant, while the volume mediation was not. The basal forebrain integrity metrics fully mediated the relationship between hippocampal acetylcholine and delayed memory and partially mediated the relationship between lateral capsular and lateral perisylvian acetylcholine and delayed memory. The volume mediation was significant for the hippocampal and lateral perisylvian models, while free water fraction was not. Free water fraction in the cholinergic basal forebrain mediated the relationship between acetylcholine and attention/working memory and executive function, while cholinergic basal forebrain volume mediated the relationship between acetylcholine in temporal regions in memory. These findings suggest that these two metrics reflect different stages of neurodegenerative processes and add additional evidence for a relationship between pathology in the basal forebrain, acetylcholine denervation and cognitive decline in Parkinson's disease. The cytoskeleton plays a pivotal role in maintaining the epithelial phenotype and is vital to several hallmark processes of cancer. Over the past decades, researchers have identified the epithelial protein lost in neoplasm (EPLIN, also known as LIMA1) as a key regulator of cytoskeletal dynamics, cytoskeletal organization, motility, as well as cell growth and metabolism. Dysregulation of EPLIN is implicated in various aspects of cancer progression, such as tumor growth, invasion, metastasis, and therapeutic resistance. Its altered expression levels or activity can disrupt cytoskeletal dynamics, leading to aberrant cell motility and invasiveness characteristic of malignant cells. Moreover, the involvement of EPLIN in cell growth and metabolism underscores its significance in orchestrating key processes essential for cancer cell survival and proliferation. This review provides a comprehensive exploration of the intricate roles of EPLIN across diverse cellular processes in both normal physiology and cancer pathogenesis. Cluster of differentiation 44 (CD44), a multi-functional cell surface receptor, has several variants and is ubiquitously expressed in various cells and tissues. CD44 is well known for its function in cell adhesion and is also involved in diverse cellular responses, such as proliferation, migration, differentiation, and activation. To date, CD44 has been extensively studied in the field of cancer biology and has been proposed as a marker for cancer stem cells. Recently, growing evidence suggests that CD44 is also relevant in non-cancer diseases. In liver disease, it has been shown that CD44 expression is significantly elevated and associated with pathogenesis by impacting cellular responses, such as metabolism, proliferation, differentiation, and activation, in different cells. However, the mechanisms underlying CD44's function in liver diseases other than liver cancer are still poorly understood. Hence, to help to expand our knowledge of the role of CD44 in liver disease and highlight the need for further research, this review provides evidence of CD44's effects on liver physiology and its involvement in the pathogenesis of liver disease, excluding cancer. Purpose: Soccer substitutes are exposed to periods of limited activity before entering match-play, likely negating benefits of active warm-ups. This study aimed to determine the effects of using a passive heat intervention following a pre-match, and half-time warm-up, on muscle and core temperature in soccer players during ambient (18 °C) and cold (2 °C) conditions. Methods: On four occasions, 8 male players, completed a pre-match warm-up, followed by 45 min of rest. Following this, participants completed a half-time re-warm-up followed by an additional 45 min of rest, simulating a full match for an unplaying substitute. During periods of rest, participants wore either standardised tracksuit bottoms (CON), or heated trousers (HEAT), over typical soccer attire. Results: Vastus lateralis temperature declined less in HEAT compared to CON following the 1st half in 2 °C (Δ − 4.39 ± 0.81 vs. − 6.21 ± 1.32 °C, P = 0.002) and 18 °C (Δ − 2.48 ± 0.71 vs. − 3.54 ± 0.88 °C, P = 0.003). These findings were also observed in the 2nd half for the 2 °C (Δ − 4.36 ± 1.03 vs. − 6.26 ± 1.04 °C, P = 0.002) and 18 °C (Δ − 2.85 ± 0.57 vs. − 4.06 ± 1 °C, P = 0.018) conditions. In addition, core temperature declined less in HEAT compared to CON following the 1st (Δ − 0.41 ± 0.25 vs. − 0.84 ± 0.41 °C, P = 0.037) and 2nd (Δ − 0.25 ± 0.33 vs. − 0.64 ± 0.34 °C, P = 0.028) halves of passive rest in 2 °C, with no differences in the 18 °C condition. Perceptual data confirmed that participants were more comfortable in HEAT vs. CON in 2 °C (P < 0.01). Endotoxin administration is commonly used to study the inflammatory response, and though traditionally given as a bolus injection, it can be administered as a continuous infusion over multiple hours. Several studies hypothesize that the latter better represents the prolonged and pronounced inflammation observed in conditions like sepsis. Yet very few experimental studies have administered endotoxin using both strategies, leaving significant gaps in determining the underlying mechanisms responsible for their differing immune responses. We used mathematical modelling to analyse cytokine data from two studies administering a 2 ng kg−1 dose of endotoxin, one as a bolus and the other as a continuous infusion over 4 h. Using our model, we simulated the dynamics of mean and subject‐specific cytokine responses as well as the response to long‐term endotoxin administration. Cytokine measurements revealed that the bolus injection led to significantly higher peaks for interleukin (IL)‐8, while IL‐10 reaches higher peaks during continuous administration. Moreover, the peak timing of all measured cytokines occurred later with continuous infusion. We identified three model parameters that significantly differed between the two administration methods. Monocyte activation of IL‐10 was greater during the continuous infusion, while tumour necrosis factor α$ {\alpha} $ and IL‐8 recovery rates were faster for the bolus injection. This suggests that a continuous infusion elicits a stronger, longer‐lasting systemic reaction through increased stimulation of monocyte anti‐inflammatory mediator production and decreased recovery of pro‐inflammatory catalysts. Furthermore, the continuous infusion model exhibited prolonged inflammation with recurrent peaks resolving within 2 days during long‐term (20–32 h) endotoxin administration. Understanding how the early stages of sexual signal diversification proceed is critically important because these microevolutionary dynamics directly shape species trajectories and impact macroevolutionary patterns. Unfortunately, studying this is challenging because signals involve complex interactions between behavior, morphology, and physiology, much of which can only be measured in real-time. In Hawaii, male Pacific field cricket song attracts both females and a deadly parasitoid fly. Over the past two decades, there has been a marked increase in signal variation in Hawaiian populations of these crickets, including novel male morphs with distinct mating songs. We capitalize on this rare opportunity to track changes in morph composition over time in a population with three novel morphs, investigating how mate and parasitoid attraction (components of sexual and natural selection) may shape signal evolution. We find dramatic fluctuation in morph proportions over the three years of the study, including the arrival and rapid increase of one novel morph. Natural and sexual selection pressures act differently among morphs, with some more attractive to mates and others more protected from parasitism. Collectively, our results suggest that differential protection from parasitism among morphs, rather than mate attraction, aligns with recent patterns of phenotypic change in the wild. OBJECTIVES: To use the ventricular pressure–volume relationship and time-varying elastance model to provide a foundation for understanding cardiovascular physiology and pathophysiology, interpreting advanced hemodynamic monitoring, and for illustrating the physiologic basis and hemodynamic effects of therapeutic interventions. We will build on this foundation by using a cardiovascular simulator to illustrate the application of these principles in the care of patients with severe sepsis, cardiogenic shock, and acute mechanical circulatory support. DATA SOURCES: Publications relevant to the discussion of the time-varying elastance model, cardiogenic shock, and sepsis were retrieved from MEDLINE. Rice is a staple food for more than half of the world's population. However, the pervasive problem of salinity is severely undermining rice production, especially in coastal and low-lying areas where soil salinization is widespread. This stress, exacerbated by climate change, necessitates the development of salt-tolerant rice varieties to ensure food security. In this study, an F2:3 population (n = 454) from a cross of KDML105 and its chromosome segment substitution line (CSSL) was used to identify genomic regions associated with salt tolerance at the seedling stage. Using the QTL-seq approach, a QTL significantly associated with salt tolerance was identified on chromosome 1. Annotation of candidate genes in this region revealed the potential regulators of salt tolerance, including MIKC-type MADS domain proteins, calmodulin-binding transcription factors, and NB-ARC domain-containing proteins. These and other identified genes provide insights into the genetic basis of salt tolerance. This study underscores the importance of using advanced genomics tools and CSSL populations in the study of complex traits such as salt tolerance in rice. Several candidate genes identified in this study could be used in further studies on molecular or physiological mechanisms related to the salt response and tolerance mechanism in rice. There has been a shift in tertiary education, whereby large portions of theory are now taught online, and the practical skills underpinned by this knowledge taught in clinical environments. It is unclear if this flipped approach is appropriate for the learning styles Exercise Science (ES) and Clinical Exercise Physiology (CEP) students. First- and second-year ES and CEP students at a South Australian metropolitan university were invited to participate between March 2021 and October 2022. Eligible participants completed the Kolb's Learning Style Inventory (KLSI) (Version 3.1) to identify learning styles. Descriptive statistics were calculated for the four KLSI learning orientations. Group scores for the learning orientations were used to identify group preference for abstractness over concreteness and action over reflection. A total of 69 students completed the KLSI fully. As a single cohort, Accommodators were the most common learning style, and there was a preference for concreteness over abstractness when gaining experience and a preference for action over reflection when transferring experience into knowledge. These findings indicate that Australian ES and CEP students have a unique learning style that may be suited to flipped classroom approaches, but only if they include opportunity to experience skill learning in a hands-on manner, while also allowing time for reflection. Friendships are developmentally significant peer relationships that meaningfully contribute to adolescent adjustment. Despite extensive evidence that friendships contribute to adolescents' psychological well‐being and mental health, less is known about the connections between adolescents' friendships and physical health outcomes. Therefore, the current review synthesizes a growing body of research examining associations between adolescent friendships and physical health. The findings reviewed provide evidence for links between the quantity, quality, and stability of adolescents' friendships and their corresponding subjective and physiological health. Consistency of findings varied as a function of friendship dimensions and health outcomes studied. In turn, we end the review with a discussion of conceptual and methodological consistencies and inconsistencies across the studies reviewed, a proposed agenda for future research, and a presentation of a novel process‐oriented model explaining how friendships may contribute to adolescent physical health. Ectopic liver regeneration in the spleen is a promising alternative to organ transplantation for treating liver failure. To accommodate transplanted liver cells, the splenic tissue must undergo structural changes to increase extracellular matrix content, demanding a safe and efficient approach for tissue remodelling. We synthesised sulphated hyaluronic acid (sHA) with an affinity for the latent complex of transforming growth factor-β (TGF-β) and cross-linked it into a gel network (sHA-X) via click chemistry. We injected this glycan into the spleens of mice to induce splenic tissue remodelling via supraphysiological activation of endogenous TGF-β. sHA-X efficiently bound to the abundant latent TGF-β in the spleen. It provided the molecular force to liberate the active TGF-β dimers from their latent complex, mimicking the 'bind-and-pull' mechanism required for physiological activation of TGF-β and reshaping the splenic tissue to support liver cell growth. Hepatocytes transplanted into the remodelled spleen developed into liver tissue with sufficient volume to rescue animals with a metabolic liver disorder (Fah -/- transgenic model) or following 90% hepatectomy, with no adverse effects observed and no additional drugs required. Our findings highlight the efficacy and translational potential of using sHA-X to remodel a specific organ by mechanically activating one single cytokine, representing a novel strategy for the design of biomaterials-based therapies for organ regeneration. Cell transplantation may provide a lifeline to millions of patients with end-stage liver diseases, but their severely damaged livers being unable to accommodate the transplanted cells is a crucial hurdle. Herein, we report an approach to restore liver functions in another organ – the spleen – by activating one single growth factor in situ. This approach, based on a chemically designed polysaccharide that can mechanically liberate the active transforming growth factor-β to an unusually high level, promotes the function of abundant allogenic liver cells in the spleen, rescuing animals from lethal models of liver diseases and showing a high potential for clinical translation. Background: Some people with multiple sclerosis (pwMS) avoid exercise due to overheating. Evidence from a variety of cooling treatments shows benefits for pwMS. Objective: Conduct a randomized controlled trial of antipyretic treatment before exercise in pwMS. Methods: Adults over age 18 diagnosed with relapsing–remitting MS reporting heat sensitivity during exercise were randomly assigned to one of six sequences counterbalancing aspirin, acetaminophen, placebo. At each of three study visits separated by ≥ one week, participants received 650-millograms of aspirin, acetaminophen, or placebo before completing a maximal exercise test. Primary outcomes were body temperature change and total time-to-exhaustion (TTE), secondary outcomes were physiological and patient-reported outcomes (PROs). Results: Sixty participants were enrolled and assigned to treatment sequence; 37 completed ≥ one study visit. After controlling for order effects, we found that body temperature increase was reduced after aspirin (+ 0.006 ± 0.32 degrees Fahrenheit, p < 0.001) and after acetaminophen (+ 0.31 ± 0.35; p = 0.004) compared to placebo (+ 0.68 ± 0.35). TTE after aspirin (331.6 ± 76.6 s) and acetaminophen (578.2 ± 82.1) did not differ significantly from placebo (551.0 ± 78.4; p's > 0.05). Aspirin benefited all secondary outcomes compared to placebo (all p's < 0.001); acetaminophen showed broadly consistent benefits. Conclusion: These results support antipyretic treatment as effective for reducing overheating during exercise in pwMS and failed to support antipyretics for increasing TTE in the context of a maximal exercise test. Lemna aequinoctialis Welw. is a widely spread species that has diverse physiological and molecular properties. Flower characteristics are important factors in deducing taxonomical status; however, owing to the rarity of flowering observations in Lemna, studying them has been a prolonged challenge. In this study, physiological and morphological analyses were conducted by inducing flowering, and molecular analysis was done based on the two chloroplast DNA loci (matK, atpF−atpH intergeneric spacer) of L. aequinoctialis sensu Landolt (1986) from 70 strains found in 70 localities in Japan, Korea, Thailand, and the US. In total, 752 flowering fronds from 13 strains were observed based on axenic conditions. Two different trends in flower organ development—protogyny and adichogamy—were detected in these strains. Their physiological traits were divided into two groups, showing different morphological features based on frond thickness, root cap, and anther sizes. Molecular analysis showed two lineages corresponding to two physiological groups. These were identified as L. aequinoctialis sensu Beppu et al. (1985) and L. aoukikusa Beppu et Murata based on the description of the nomenclature of L. aoukikusa. These were concluded as independent taxa and can be treated as different species. Targeted metabolomic analysis was conducted on juvenile Chinook salmon exposed for 10 days to wastewater effluent (WWE) from a large urban treatment plant. Exposures included five dilutions of WWE (20%, 5.3%, 1.4%, 0.4%, and 0.1%) and a control with 7 replicates per treatment. Liver was extracted from fish and analyzed via liquid chromatography - mass spectrometry (LC-MS) for 361 endogenous metabolites, of which 185 were detected. Control-versus-treatment comparisons identified several metabolites that were associated with altered biochemical pathways observed for all treatments, including several that are important for energy generation and utilization. These altered pathways are crucial for fish health and may be an early indicator of potential adverse effects on growth, reproduction, behavior, and immune function. Juvenile ocean-type Chinook salmon spend several days to weeks in the nearshore estuary where they may encounter high concentrations of WWE contaminants. They are exposed to a wide range of potent pharmaceuticals, personal care products, and industrial compounds from WWE that have the potential to affect physiological homeostasis and disrupt their normal life cycle. Wastewater treatment plants release complex mixtures of chemicals into the aquatic environment as wastewater effluent (WWE); however, the effects of these mixtures are still poorly understood. Chinook salmon (Oncorhynchus tshawytscha) are a culturally important species in the Pacific Northwest and are a vital food resource for southern resident killer whales (Orcinus orca) that are listed as 'critically endangered' under the U.S. Endangered Species Act (ESA). Chinook populations have declined drastically in this region and have failed to show significant recovery despite regional-to-federal efforts, resulting in many populations being listed as threatened under the ESA. One source of stress to juvenile Chinook is chemical pollution from WWE during their outmigration along river corridors and residency in estuaries. In this study, we investigated effects of WWE on juvenile Chinook health in a ten-day exposure to dilutions of WWE from 0.1% to 20%. At the end of the exposure, we measured endpoints associated with endocrine disruption, brain function, osmoregulation, stress, and metabolism. Exposure to WWE significantly (a = 0.1 for all analyses) induced vitellogenesis, indicating endocrine system disruption. We saw significant reductions in plasma glucose, an indication of stress, and brain Na+/K+-ATPase (NKA) activity, an enzyme essential for neuronal signaling. Lastly, metabolism was affected as evidenced by altered total protein, cholesterol, and albumin in plasma, a drastic decrease in whole body lipid content, and a significant increase in visible liver anomalies. We compared contaminant concentrations in exposure water with effects concentrations from the literature for chemicals known to induce vitellogenin or inhibit brain NKA. For most contaminants, concentrations in exposure waters were several orders of magnitude below effects concentrations in the literature. The exception was estrogenic hormones, which were detected at similar concentrations in this study compared to concentrations in other studies that induced vitellogenin. Based on comparisons to the literature, contaminants measured in this study could not explain the inhibition of brain NKA; however, WWE mixtures contain many quantified and undetected compounds that are likely acting together to cause harmful effects in Chinook. Cilia are critical for detecting chemo-, photo-, and mechanosensory stimuli in the sensory systems. Most neurons in the central nervous system harbor cilia enriched in neuromodulatory receptors. Neuronal cilia are structurally and molecularly diverse, and play various roles in the central nervous system. The functionality of cilia is determined by their structure, the presence of receptors and ion channels, and their specific context. Neuronal cilia are found adjacent to chemical synapses and axonal segments, and can form axociliary synapses. Cilia are fascinating organelles that act as cellular antennae, sensing the cellular environment. Cilia gained significant attention in the late 1990s after their dysfunction was linked to genetic diseases known as ciliopathies. Since then, several breakthrough discoveries have uncovered the mechanisms underlying cilia biogenesis and function. Like most cells in the animal kingdom, neurons also harbor cilia, which are enriched in neuromodulatory receptors. Yet, how neuronal cilia modulate neuronal physiology and animal behavior remains poorly understood. Increasingly frequent and intense heatwaves threaten ecosystem health in a warming climate. However, plant responses to heatwaves are poorly understood. A key uncertainty concerns the intensification of transpiration when heatwaves suppress photosynthesis, known as transpiration–photosynthesis decoupling. Field observations of such decoupling are scarce, and the underlying physiological mechanisms remain elusive. Here, we use carbonyl sulphide (COS) as a leaf gas exchange tracer to examine potential mechanisms leading to transpiration–photosynthesis decoupling on a coast live oak in a southern California woodland in spring 2013. We found that heatwaves suppressed both photosynthesis and leaf COS uptake but increased transpiration or sustained it at non‐heatwave levels throughout the day. Despite statistically significant decoupling between transpiration and photosynthesis, stomatal sensitivity to environmental factors did not change during heatwaves. Instead, midday photosynthesis during heatwaves was restricted by internal diffusion, as indicated by the lower internal conductance to COS. Thus, increased evaporative demand and nonstomatal limitation to photosynthesis act jointly to decouple transpiration from photosynthesis without altering stomatal sensitivity. Decoupling offered limited potential cooling benefits, questioning its effectiveness for leaf thermoregulation in xeric ecosystems. We suggest that adding COS to leaf and ecosystem flux measurements helps elucidate diverse physiological mechanisms underlying transpiration–photosynthesis decoupling. The crustose coralline alga Lithophyllum stictiforme is an important reef builder, forming coralligenous concretions in the Mediterranean Sea. This algal species complex is composed of eight cryptic species (or clades). The objective of our study was to compare light-dependent physiological processes within and between the two most abundant cryptic species (C1 and C4) of L. stictiforme from the Bay of Marseille (France). Physiological rates of respiration, photosynthesis and calcification were measured using incubation chambers under various irradiance levels and in the dark during the summer period. We compared the physiology of the C1 and C4 cryptic species of L. stictiforme among three algal groups: C1 (the most common in the Marseille area) from 28 m depth, C4 from the same site and depth, and C1 from a deeper site at 45 m depth. We found both interspecific (between C1 and C4) and intraspecific (within C1) physiological differences. Photosynthetic parameters showed acclimation to light (or depth) with higher values of initial slope of photosynthetic light response curve (α) and lower values of saturating irradiance (Ek) and compensating irradiance (Ec) for C1 from 45 m than for C1 and C4 from 28 m depth. At the shallower depth, significant physiological differences were observed between C1 and C4 with a higher value of maximum rate of gross photosynthesis (Pg max) in C4, suggesting better ability to grow under high irradiance. Light and dark calcification rates differed only between C4 from 28 m and C1 from 45 m depth, being intermediate in C1 from 28 m depth. On a 24 h basis, diel net calcification rates were significantly higher in specimens from the shallower waters than from the deeper waters. Physiological differences suggest physiological plasticity within the regionally dominant cryptic species C1 and species-specific physiological traits between the cryptic species C1 and C4. Photosynthesis and calcification rates differ among Lithophyllum stictiforme cryptic species. The regionally dominant cryptic species exhibits light-dependent physiological plasticity. Intestinal stem cells (ISCs) of the fruit fly, Drosophila melanogaster, offer an excellent genetic model to explore homeostatic roles of ISCs in animal physiology. Among available genetic tools, the escargot (esg)-GAL4 driver, expressing the yeast transcription factor gene, GAL4, under control of the esg gene promoter, has contributed significantly to ISC studies. This driver facilitates activation of genes of interest in proximity to a GAL4-binding element, Upstream Activating Sequence, in ISCs and progenitor enteroblasts (EBs). While esg-GAL4 has been considered an ISC/EB-specific driver, recent studies have shown that esg-GAL4 is also active in other tissues, such as neurons and ovaries. Therefore, the ISC/EB specificity of esg-GAL4 is questionable. In this study, we reveal esg-GAL4 expression in the corpus allatum (CA), responsible for juvenile hormone (JH) production. When driving the oncogenic gene, RasV12, esg-GAL4 induces overgrowth in ISCs/EBs as reported, but also increases CA cell number and size. Consistent with this observation, animals alter expression of JH-response genes. Our data show that esg-GAL4-driven gene manipulation can systemically influence JH-mediated animal physiology, arguing for cautious use of esg-GAL4 as a "specific" ISC/EB driver to examine ISC/EB-mediated animal physiology.
