With the expansion of ICP-MS application into the field of bioanalysis, there is an urgent need for novel element tags today. Here, we report the design of a dual-element Ir-Eu tag, opening the door to simultaneous fluorescent imaging and ICP-MS quantification. The ratio of 153 Eu/ 193 Ir may serve as a precision control of the labeling process, allowing internal validation of the quantitative results obtained. As for SIRPα and its host cell analysis exemplified here, the Ir-Eu tag demonstrated superior figures of ICP-MS quantification with the LOD (3σ) down to 0.5 ( 153 Eu) and 1.1 ( 193 Ir) pM SIRPα and 220 ( 153 Eu) and 830 ( 193 Ir) RAW264.7 cells more than 130 times more sensitive compared with the LOD (3σ) of 65.2 pM SIRPα at 612 nm using fluorometry. Not limited to these demonstrations, we believe that the design ideas of the dual Ir-Eu tags should be applicable to various cases of bioanalysis when dual optical profiling and ICP-MS quantification are indispensable., (© 2023. Orbitrap-based charge detection mass spectrometry utilizes single-molecule sensitivity to enable mass analysis of even highly heterogeneous, high-mass macromolecular assemblies. For contemporary Orbitrap instruments, the accessible ion detection (recording) times are maximally ~1-2 s. Here by modifying a data acquisition method on an Orbitrap ultrahigh mass range mass spectrometer, we trapped and monitored individual (single) ions for up to 25 s, resulting in a corresponding and huge improvement in signal-to-noise ratio (×5 compared with 1 s), mass resolution (×25) and accuracy in charge and mass determination of Orbitrap-based charge detection mass spectrometry., (© 2024. The identification of geographical origin is an important factor in evaluating the authenticity of honey. However, at present, there are few studies concerning the honey of Apis cerana Fabricius (A. cerana, Asiatic honeybee). To identify geographical origin, we used two common methods (multi-physicochemical parameters and phenolic chromatographic fingerprints) but achieved only poor identification. To compensate for this shortcoming, we established an ICP-MS-based ionomics method using 18 elements in 27 A. cerana honey samples from three different areas in Shaanxi Province, China. Multivariate analysis showed that significant differences in contents can be used to discriminate the geographical origin of A. cerana honey. The method was further validated using an independent test set of 11 samples with 90.91% accuracy, demonstrating its potential for the identification and prediction of the geographical origin of honey., (Copyright © 2021 Elsevier Ltd. Studies on complex biological phenomena often combine two or more imaging techniques to collect high-quality comprehensive data directly in situ , preserving the biological context. Mass spectrometry imaging (MSI) and vibrational spectroscopy imaging (VSI) complement each other in terms of spatial resolution and molecular information. In the past decade, several combinations of such multimodal strategies arose in research fields as diverse as microbiology, cancer, and forensics, overcoming many challenges toward the unification of these techniques. Here we focus on presenting the advantages and challenges of multimodal imaging from the point of view of studying biological samples as well as giving a perspective on the upcoming trends regarding this topic. The latest efforts in the field are discussed, highlighting the purpose of the technique for clinical applications.

Asymmetrical flow field-flow fractionation (AF4) hyphenated with inductively coupled plasma-mass spectrometry (ICP-MS) has been widely used to characterize metal containing particles. This study demonstrates the advantages of coupling AF4 with ICP-time-of-flight mass spectrometry (ICP-TOFMS) in standard and single particle modes to determine size distribution, elemental composition, and number concentration of composite particles. The coupled system was used to characterize two complex particle mixtures. The first mixture consisted of particles extracted from micro-alloyed steels with two size populations of different elemental composition. The second mixture consisted of particles extracted from soil spiked with various engineered nanoparticles (ENPs). The equivalent hydrodynamic sizes of individual micro-alloyed steel particles were up to 6 times larger than the sizes determined by single particle (sp)-ICP-TOFMS. The larger AF4 sizes were attributed to the presence of a surface coating, which is not reflected in the core size determined by sp-ICP-TOFMS. Two particle populations could not be separated by AF4 due to their broad size distributions but were resolved by sp-ICP-TOFMS using their unique elemental signatures. Multi-angle light scattering and ICP-TOFMS signals of soil suspensions increased with the spiked ENP concentrations. However, only after conducting full element screening and single particle fingerprinting by ICP-TOFMS could this increase be attributed to enhanced extraction efficiency of natural particles and the risk for false conclusions be eliminated. Essential and nonessential element concentrations in human blood provide important information on the nutritional status of individuals and can assist in the screening or diagnosis of certain disorders and their association with other causative factors. A simple and sensitive method, suitable for use with small sample volumes, for quantification of multiple trace element concentrations in whole blood and plasma has been developed using inductively coupled plasma-mass spectrometry. Method validation was performed using standard reference materials of whole blood and serum using varying sample treatments with nitric acid, water and hydrogen peroxide. The method was applied to quantify the trace element concentrations in whole blood and plasma samples (0.1 mL) from 50 adult blood donors in Queensland. The whole blood sample (5 mL) was collected in Vacutainer tubes with K2EDTA as anticoagulant. The developed method was able to quantify, in blood and plasma samples over a wide range of concentrations, several essential elements: cobalt, copper, zinc, iron, manganese and selenium; the nutritionally probably essential elements vanadium and strontium; and nonessential elements including lead, cadmium, arsenic, caesium, barium, thallium and uranium. Significant differences (P < 0.0001) were observed between whole blood and plasma concentrations for 13 elements; 5 of the measured elements, cobalt (0.49 vs. 0.36 μg/L), copper (1.0 vs. 0.75 mg/L), strontium (28 vs. 16 μg/L), barium (1.5 vs. 0.64 μg/L) and thallium (0.06 vs. 0.03 μg/L), had higher mean concentrations in plasma than in blood. Whole blood concentrations of nine trace elements were significantly correlated (P < 0.0001) with plasma concentrations. The distribution of the trace elements between human blood and plasma varied considerably for the different elements. One of the most important causes of the high mortality rate and low life expectancy of lung cancer is the detection at advanced stages. Thus, there is an urgent need for early diagnosis and the search of new selective biomarkers. Selenium is an important constituent of selenoproteins and a powerful antioxidant able to protect against cancer. In this work, the absolute quantification of selenium in selenoproteins and the total content in selenometabolites has been performed for the first time in serum from lung cancer patients (LC) and healthy controls (HC). To this end, a method for the simultaneous speciation of selenoproteins using size exclusion chromatography (SEC) and affinity chromatography (AF) with detection by ICP-QQQ-MS, and quantification by isotopic dilution (IDA) (SEC-AF-HPLC-SUID-ICP-QQQ-MS) was developed to determine the selenium concentration in eGPx, SEPP1 and SeAlb, as well as total selenometabolites, to find alterations that may serve as biomarkers of this disease. In the same way, a method based on anion-exchange chromatography coupled to ICP-QQQ-MS was developed to quantify selenometabolites (SeCys2, SeMeSeCys, SeMet, selenite and selenate) in the same LC and HC serum samples. The results showed that the averaged concentrations of selenium in eGPx, SeAlb and selenite were significantly higher in LC patients (LC (eGPx: 21.24 ± 0.77 ng g -1 ; SeAlb: 49.56 ± 3.16 ng g -1 and Se(IV): 6.20 ± 1.22 ng g -1 ) than in HC group (eGPx: 16.96 ± 0.53 ng g -1 ; SeAlb: 38.33 ± 2.66 ng g -1 and Se(IV): 3.56 ± 0.55 ng g -1 ). In addition, the ratios between selenoproteins and selenometabolites have been calculated for the first to study their potential use as LC biomarkers. A new chemical separation has been developed to isolate uranium (U) using two UTEVA columns to minimize iron and thorium interferences from high background area soil samples containing minerals like monazites and ilmenite. The separation method was successfully verified in some certified reference materials (CRMs), for example, JSd-2, JLk-1, JB-1 and JB-3. The same method was applied for purification of U in Fukushima soil samples affected by the Fukushima dai-ichi nuclear power station (FDNPS) accident. Precise and accurate measurement of 234 U/ 238 U and 235 U/ 238 U isotope ratios in chemically separated U were carried out using a multi-collector inductively coupled plasma mass spectrometer (MC-ICP-MS). In this mass spectrometric method, an array of two Faraday cups (10 11 Ω, 10 12 Ω resistor) and a Daly detector were simultaneously employed. The precision of U isotope ratios in an in-house standard was evaluated by replicate measurement. Relative standard deviation (RSD) of 234 U/ 238 U and 235 U/ 238 U were found to be 0.094% (2σ) and 0.590% (2σ), respectively. This method has been validated using a standard reference material SRM 4350B, sediment sample. The replicate measurements of 234 U/ 238 U in SRM shows 0.7% (RSD). This developed method is suitable for separation of U and its isotope ratio measurement in environmental samples.

The development of ruthenium-based complexes for cancer treatment requires a variety of pharmacological studies, one of them being a drug's binding kinetics to serum proteins. In this work, speciation analysis was used to study kinetics of ruthenium-based drug candidates with human serum proteins. Two ruthenium (Ru) complexes, namely [(η 6 - p -cymene)Ru(1-hydroxypyridine-2(1 H )-thionato)Cl] ( 1 ) and [(η 6 - p -cymene)Ru(1-hydroxypyridine-2(1 H )-thionato)pta]PF 6 ( 2 ) (where pta = 1,3,5-triaza-7-phosphaadamantane), were selected. Before a kinetics study, their stability in relevant media was confirmed by nuclear magnetic resonance (NMR). Conjoint liquid chromatography (CLC) monolithic column, assembling convective interaction media (CIM) protein G and diethylamino (DEAE) disks, was used for separation of unbound Ru species from those bound to human serum transferrin (Tf), albumin (HSA) and immunoglobulins G (IgG). Eluted proteins were monitored by UV spectrometry (278 nm), while Ru species were quantified by post-column isotope dilution inductively coupled plasma mass spectrometry (ID-ICP-MS). Binding kinetics of chlorido ( 1 ) and pta complex ( 2 ) to serum proteins was followed from 5 min up to 48 h after incubation with human serum. Both Ru complexes interacted mainly with HSA. Complex ( 1 ) exhibited faster and more extensive interaction with HSA than complex ( 2 ). The equilibrium concentration for complex ( 1 ) was obtained 6 h after incubation, when about 70% of compound was bound to HSA, 5% was associated with IgG, whereas 25% remained unbound. In contrast, the rate of interaction of complex ( 2 ) with HSA was much slower and less extensive and the equilibrium concentration was obtained 24 h after incubation, when about 50% of complex ( 2 ) was bound to HSA and 50% remained unbound.

Many natural product chemists are working to identify a wide variety of novel secondary metabolites from natural materials and are eager to avoid repeatedly discovering known compounds. Here, we developed liquid chromatography/mass spectrometry (LC/MS) data-processing protocols for assessing high-throughput spectral data from natural sources and scoring the novelty of unknown metabolites from natural products. This approach automatically produces representative MS spectra (RMSs) corresponding to single secondary metabolites in natural sources. In this study, we used the RMSs of Agrimonia pilosa roots and aerial parts as models to reveal the structural similarities of their secondary metabolites and identify novel compounds, as well as isolation of three types of nine new compounds including three pilosanidin- and four pilosanol-type molecules and two 3-hydroxy-3-methylglutaryl (HMG)-conjugated chromones. Furthermore, we devised a new scoring system, the Fresh Compound Index (FCI), which grades the novelty of single secondary metabolites from a natural material using an in-house database constructed from 466 representative medicinal plants from East Asian countries. We expect that the FCIs of RMSs in a sample will help natural product chemists to discover other compounds of interest with similar chemical scaffolds or novel compounds and will provide insights relevant to the structural diversity and novelty of secondary metabolites in natural products.

With developments in multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS), applications of metal stable isotopes received increasing attentions in the studies of source and fate of heavy metals in the environment. In light of the rapid progresses in this emerging field, we attempted to review the recent findings comprehensively in a way that environmental scientists can easily read. This review started with an introduction of basic terminologies in isotope geochemistry, followed with detailed descriptions of instrumentation and analytical procedures, and finally focused on the cases of three typical metal stable isotopes (Ag, Hg and Zn) to illustrate how they were applied to address environmental issues. Additionally, future perspectives on the applicability, opportunities, and limitations of metal stable isotope techniques as novel approaches in advancing environmental chemistry were discussed., (Copyright © 2019 Elsevier Ltd. Inadequate micronutrient intake in childhood harms growth and development, and it is related to increased rates of morbidity and mortality. The aim of this study is to evaluate the dietary intake and prevalence of inadequate micronutrient intake in preschool children (1-4 years old) attending two-day care centers. To assess children's dietary micronutrient intake, 24-h duplicate diets (n = 64) were collected for one week-day, including everything the children ate and drank both at home and in kindergarten. Anthropometric measurements were carried out to evaluate the children's nutritional status. The micronutrients copper, iron, calcium, magnesium, selenium, zinc, potassium, sodium, and manganese were determined by inductively coupled plasma mass spectrometry or graphite furnace atomic absorption spectrometry. Calcium and selenium were found with high inadequate intake rates: 50% and 42%, respectively, for children aged 1-3 years old, and 93% and 90% for children aged 4 years. Potassium was consumed in very low amounts, 13% and 5% of children aged 1-3 and 4 years old, respectively, achieved the adequate intake for the nutrient. Sodium intakes were excessive: 23% of the 1-3-year old and 42% of the 4-year-old children, respectively, had an intake higher than the tolerable upper levels. Regarding the nutritional status, overweight and obesity prevalence was 17%. Therefore, considering the damaging health effects for children of micronutrient deficiency and overweight and obesity status, it is necessary that government authorities be aware and update public policies and educational programs in order to promote healthy eating habits in early childhood., (Copyright © 2019 Elsevier GmbH. Here we propose a single acid digestion (SAD) sample preparation method for ICP-MS analysis of animal serum samples to determine trace element contents. The method was evaluated in comparison with a commonly used procedure involving dilution of samples in an alkaline solution (AKD). In the SAD procedure, aliquots (1 mL) of bovine serum samples were treated at low temperature with a mixture of concentrated nitric acid and hydrogen peroxide. Trace elements (As, B, Ba, Cd, Co, Cr, Cu, Fe, Hg, Li, Mn, Mo, Ni, Pb, Sb, Se, Sr, U, and Zn) were directly determined by ICP-MS analysis of diluted solutions of samples. Both methods were sufficiently sensitive to enable quantification of most trace elements, with the exception of the AKD method for Cd, Hg and Pb. The quality of the data was verified by using certified reference material. Good results were obtained for the SAD procedure and all elements, but recoveries were unacceptable with the AKD procedure for Se (recovery: 57%), Cd (154%) and Fe (139%). Strong associations (R2>0.90, P = 0.000) between the data obtained by both methods were demonstrated for the elements considered. The proposed SAD sample preparation method produced satisfactory results for determining most toxic and essential trace elements targeted in monitoring studies., Competing Interests: The authors have declared that no competing interests exist.

Metal speciation analysis of microsamples in the clinical, biological and forensic fields is important for elucidating the metals' toxicity, mobility and metabolic behaviors of metals. Such analysis may be achieved by nanoliter high-performance liquid chromatography (nanoHPLC) hyphenated with inductively coupled plasma mass spectrometry (ICP-MS). In this work, an in-column high-pressure nebulizer (ICHPN) was developed to enhance the sensitivity and the separation efficiency of the sheathless nanoHPLC-ICP-MS coupling. The ICHPN consists of two concentric fused-silica capillaries with tapered tips connected via a PEEK tee, where C 18 silica particles with a size of 5 μm were packed in the tapered inner capillary based on the keystone effect. Combining a heated single pass spray chamber with a makeup gas, the ICHPN was capable of independently optimizing the nebulization efficiency and the transport efficiency. The ICHPN offered high sensitivities, low detection limits and good precision at nanoflow rates. Compared with commercial nebulizers, the ICHPN fabrication was simple, rapid, reproducible and inexpensive. By implementing the ICHPN, we achieved rapid separation of four mercury species, i.e., Hg 2+ and methyl-, ethyl- and phenylmercuric chloride, within 8.0 min with good resolution (2.0-13.9). Detection limits of 0.044-0.13 μg L -1 were obtained with precisions of peak heights and areas ranging from 1.5 to 3.5% for a 50 μg L -1 standard solution. Good agreement between the determined and certified values of mercury species in a certified reference material of caprine blood (SRM 955c) together with good recoveries (93-101%) validated the accuracy of the method., (Copyright © 2018 Elsevier B.V. A sensitive, simple and rapid method for the simultaneous determination of eleven arsenic species has been developed. A high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) technique was used for the analysis of eleven arsenic species in less than 17 min. Different extraction solutions were explored and the recovery results using water and aqueous acidic solvents, aqueous organic solvents and enzymes showed that 40 mg protease with 0.75 mL 0.5% hydrochloric acid (v/v) as the extraction agent gave the best experimental results. Species separation with ammonium carbonate solution as the mobile phase was conducted on an anion-exchange chromatographic column using gradient elution. The column temperature was 20 °C and kinetic energy discrimination (KED) was employed to eliminate spectral interference. The use of KED mode effectively removed interference from 75 ArCl. The detection limit (L D ) of the method was in the range of 0.11-0.59 μg kg -1 . Repeatability values obtained for spiked real fish samples were in the range of 1.1%-7.6%. Accuracy was calculated based on the analysis of spiked real fish samples at five concentration levels. Obtained recoveries were 91%-106%. The validated method was used in a pilot study to analyze real samples of fish, the organic arsenic especially AsB was the major arsenic specie present in the analyzed samples, only trace amount of inorganic arsenic were detected. Background: Biofortification of food crops has been used to increase the intake of Se in the human diet, even though this may change the concentration of other elements and modify the nutritional properties of the enriched food. Selenium biofortification programs should include routine assessment of the overall mineral composition of enriched plants., Results: Laser ablation inductively coupled plasma mass spectrometry (LA ICP-MS) was used for the assessment of mineral composition of table olives. Olive trees were fertilized with sodium selenate before flowering. At harvest, the edible parts of drupes proved to be significantly enriched in Se, delivering 6.1 μg g -1 (39% of the RDA for five olives). Such enrichment was followed by significant changes in the concentrations of B, Mg, K, Cr, Mn, Fe and Cu in edible parts, which are discussed for their impact on food quality., Conclusion: The biofortification of olive plants has allowed the enrichment of fruits with selenium. Enrichment with selenium has caused an increase in the concentration of other elements, which can change the nutritional quality of the drupes. In this work we present a simple and cost-effective approach for the determination of selenium species in algae and yeast biomass, based on a combination of thin-layer chromatography (TLC) with diode laser thermal vaporization inductively coupled plasma mass spectrometry (DLTV ICP MS). Extraction of freeze-dried biomass was performed in 4M methanesulphonic acid and the selenium species were vaporized from cellulose TLC plates employing a continuous-wave infrared diode laser with power up to 4 W using a simple laboratory-built apparatus. Selenomethionine and selenocysteine were quantified with limits of detection 3 μg L -1 in a Se-enriched microalgae Chlorella vulgaris and yeast certified reference material SELM-1. Results delivered by TLC-DLTV ICP MS were consistent with those obtained by a routine coupling of high-performance liquid chromatography (HPLC) to ICP MS. Heparin, a highly sulfated glycosaminoglycan, has been used as a clinical anticoagulant over 80 years. Low molecular weight heparins (LMWHs), heparins partially depolymerized using different processes, are widely used as clinical anticoagulants. Qualitative molecular weight (MW) and quantitative mass content analysis are two important factors that contribute to LMWH quality control. Size exclusion chromatography (SEC), relying on multiple angle laser scattering (MALS)/refractive index (RI) detectors, has been developed for accurate analysis of heparin MW in the absence of standards. However, the cations, which ion-pair with the anionic polysaccharide chains of heparin and LMWHs, had not been considered in previous reports. In this study, SEC with MALS/RI and inductively coupled plasma/mass spectrometry detectors were used in a comprehensive analytical approach taking both anionic polysaccharide and ion-paired cations heparin products. This approach was also applied to quantitative analysis of heparin and LMWHs. Full profiles of MWs and mass recoveries for three commercial heparin/LMWH products, heparin sodium, enoxaparin sodium and nadroparin calcium, were obtained and all showed higher MWs than previously reported. The new guidelines of the United States pharmacopeia (USP), European pharmacopeia (EP) and international conference on harmonization (ICH) regulating elemental impurities limits in pharmaceuticals signify the end of unspecific analysis of metals as outlined in USP 〈231〉. The new guidelines specify both daily doses and concentration/limits of elemental impurities in pharmaceutical final products, active pharmaceutical ingredients (API) and excipients. In chapter USP 〈233〉 method implementation, validation and quality control during the analytical process are described. We herein report the use of a stabilising matrix that overcomes low spike recovery problem encountered with Os and allows the determination of all USP required elemental impurities (As, Cd, Hg, Pb, V, Cr, Ni, Mo, Cu, Pt, Pd, Ru, Rh, Os and Ir) in a single analysis. The matrix was used in the validation of a method to determine elemental impurities in TP-6076 active pharmaceutical ingredient (API) by ICP-MS according to the procedures defined in USP〈233〉 and to GMP requirements. This validation will support the regulatory submission of TP-6076 which is a novel tetracycline analogue effective against the most urgent multidrug-resistant gram-negative bacteria. Rapid and sensitive monitoring of volatile organic compounds (VOCs) in aqueous samples is very important to human health and environmental protection. In this study, an on-line spray inlet proton transfer reaction mass spectrometry (SI-PTR-MS) method was developed for the rapid and sensitive monitoring of 6 different kinds of VOCs, namely acetonitrile, acetaldehyde, ethanol, acetone, aether, and methylbenzene, in aqueous samples. The response time, limit of detection (LOD), and repeatability of the SI-PTR-MS system were evaluated. The response of the SI-PTR-MS was quite rapid with response times of 31-88 s. The LODs for all these VOCs were below 10 μg/L. The LOD of methylbenzene was 0.9 μg/L, much lower than the maximum contaminant level (MCL) in drinking water. The repeatability of this method was evaluated with 5 replicate determinations. The relative standard deviations (RSDs) were in the range of 0.8-3.1%, indicating good repeatability. To evaluate the matrix effects, the SI-PTR-MS system was employed for on-line monitoring of these 6 VOCs in different aqueous matrices, including lake water, tap water, and waste water. The relative recoveries were in the range of 94.6-106.0% for the lake water, 96.3-105.6% for the tap water, and 95.6-102.9% for the waste water. The results indicate that the SI-PTR-MS method has important application values in the rapid and sensitive monitoring of VOCs in these aqueous samples. In addition, the effect of salt concentration on the extracting efficiency was evaluated. Knowledge of the surface composition of sea spray aerosols (SSA) is critical for understanding and predicting climate-relevant impacts. Offline microscopy and spectroscopy studies have shown that dry supermicron SSA tend to be spatially heterogeneous particles with sodium- and chloride-rich cores surrounded by organic enriched surface layers containing minor inorganic seawater components such as magnesium and calcium. At the same time, single-particle mass spectrometry reveals several different mass spectral ion patterns, suggesting that there may be a number of chemically distinct particle types. This study investigates factors controlling single particle mass spectra of nascent supermicron SSA. Depth profiling experiments conducted on SSA generated by a fritted bubbler and total ion intensity analysis of SSA generated by a marine aerosol reference tank were compared with observations of ambient SSA observed at two coastal locations. Analysis of SSA produced by utilizing controlled laboratory methods reveals that single-particle mass spectra with weak sodium ion signals can be produced by the desorption of the surface of typical dry SSA particles composed of salt cores and organic-rich coatings. Thus, this lab-based study for the first time unifies findings from offline and online measurements as well as lab and field studies of the SSA particle-mixing state.

Concentration values of 24 elements (Al, As, Ba, Be, Ca, Cd, Co, Cr, Cu, Fe, Ge, Hg, Mn, Mo, Pb, Sb, Se, Sn, Sr, Ti, Tl, U, V, and Zn) were determined in 72 honey samples produced in Italy by inductively coupled plasma mass spectrometry (ICP-MS). Considering the recommended established heavy metal daily intakes for humans, in this perspective, an equilibrated and ordinary honey consumption should not be considered matter of concerns for human health, even if particular attention should be addressed if honey is consumed by children, due to different maximum daily heavy metal intakes. Chemometric analysis of the results obtained highlights heavy metal content differences in honey samples obtained from notoriously polluted zones, confirming then that honey can be considered a bio-indicator of environmental pollution. Finally, Pearson coefficients highlighted correlations among element contents in honey samples.

A reversed phase high performance liquid chromatography coupled to an inductively coupled plasma mass spectrometer (HPLC-ICP-MS) approach in combination with isotope dilution analysis (IDA) for the separation and parallel quantification of nanostructured and ionic silver (Ag) is presented. The main focus of this work was the determination of the ionic Ag concentration. For a sufficient stabilization of the ions without dissolving the nanoparticles (NPs), the eluent had to be initially optimized. The determined Ag ion concentration was in a good agreement with results obtained using ultrafiltration. Further, the mechanism of the NP separation in the HPLC column was investigated. Typical size exclusion effects were found by comparing results from columns with different pore sizes. Since the recovery rates decreased with increasing Ag NP size and large Ag NPs did not elute from the column, additional interactions of the particles with the stationary phase were assumed. Our results reveal that the presented method is not only applicable to Ag NPs, but also to gold and polystyrene NPs. Finally, IDA-HPLC-ICP-MS experiments in single particle mode were performed to determine the particle cut-off size. This review of differential mobility spectrometry focuses primarily on mass spectrometry coupling, starting with the history of the development of this technique in the Soviet Union. Fundamental principles of the separation process are covered, in addition to efforts related to design optimization and advancements in computer simulations. Recent advances in mass spectrometry and lasers have facilitated the development of novel experiments combining the benefits of both technologies. This minireview focuses on the coupling of visible/ultraviolet photons with mass spectrometry for analysis of peptide and protein three-dimensional structure. Practical aspects of instrument design and the relationship between experiment and theory are discussed. Experiments utilizing spectroscopy, action spectroscopy, excitation energy transfer, photodissociation, and photoactivated radical chemistry are described. The strengths and weaknesses of each approach are discussed in relation to the type of information typically obtained. A significant body of data suggests that under appropriate source conditions, kinetically trapped structures are observed in these experiments rather than true gas phase minima, suggesting retention of solution phase structural features is possible. Further refinement and exploration of these methods promises to accelerate protein structure discovery in the near future.

The applicability of the emerging non-destructive technique, proton transfer reaction mass spectrometry (PTR-MS), was explored for the first time in the quality control of saffron. Monitoring of volatile organic compounds (VOCs) was achieved using a minute sample (35 mg). Fresh saffron was stored under selected conditions (25 and 40 °C, aw=0.64) over a five weeks period to produce lower quality material, which was used to prepare mixtures with fresh saffron. Analysis showed that the VOCs fingerprint changed upon storage, and the concentration of initially dominant VOC safranal decreased progressively. Examination of calculated and recorded fingerprints for various admixtures showed that PTR-MS VOCs analysis, in combination with chemometrics, could be used to screen for the presence of lower quality saffron in a commercial product in a few minutes. Facing the lack of studies on characterization and quantification of cerium oxide nanoparticles (CeO2 NPs), whose consumption and release is greatly increasing, this work proposes a method for their sizing and quantification by Flow Field-flow Fractionation (FFFF) coupled to Inductively Coupled Plasma-Mass Spectrometry (ICP-MS). Two modalities of FFFF (Asymmetric Flow- and Hollow Fiber-Flow Field Flow Fractionation, AF4 and HF5, respectively) are compared, and their advantages and limitations discussed. Experimental conditions (carrier composition, pH, ionic strength, crossflow and carrier flow rates) are studied in detail in terms of NP separation, recovery, and repeatability. Size characterization of CeO2 NPs was addressed by different approaches. In the absence of feasible size standards of CeO2 NPs, suspensions of Ag, Au, and SiO2 NPs of known size were investigated. Ag and Au NPs failed to show a comparable behavior to that of the CeO2 NPs, whereas the use of SiO2 NPs provided size estimations in agreement to those predicted by the theory. The latter approach was thus used for characterizing the size of CeO2 NPs in a commercial suspension. Results were in adequate concordance with those achieved by transmission electron microscopy, X-ray diffraction and dynamic light scattering. The quantification of CeO2 NPs in the commercial suspension by AF4-ICP-MS required the use of a CeO2 NPs standards, since the use of ionic cerium resulted in low recoveries (99 ± 9% vs. 73 ± 7%, respectively). A limit of detection of 0.9 μg L(-1) CeO2 corresponding to a number concentration of 1.8 × 1012 L(-1) for NPs of 5 nm was achieved for an injection volume of 100 μL., (Copyright © 2016 Elsevier B.V. A residual gas analyzer (RGA) coupled with a high vacuum chamber is described for the non-invasive diagnosis of the Helicobacter pylori (H. pylori) infection through ¹³C-urea breath analysis. The present RGA-based mass spectrometry (MS) method is capable of measuring high-precision ¹³CO₂ isotope enrichments in exhaled breath samples from individuals harboring the H. pylori infection. The system exhibited 100% diagnostic sensitivity, and 93% specificity alongside positive and negative predictive values of 95% and 100%, respectively, compared with invasive endoscopy-based biopsy tests. A statistically sound diagnostic cut-off value for the presence of H. pylori was determined to be 3.0‰ using a receiver operating characteristic curve analysis. The diagnostic accuracy and validity of the results are also supported by optical off-axis integrated cavity output spectroscopy measurements. The δ¹³(DOB)C‰ values of both methods correlated well (R² = 0.9973 at 30 min). The RGA-based instrumental setup described here is simple, robust, easy-to-use and more portable and cost-effective compared to all other currently available detection methods, thus making it a new point-of-care medical diagnostic tool for the purpose of large-scale screening of the H. pylori infection in real time. The RGA-MS technique should have broad applicability for ¹³C-breath tests in a wide range of biomedical research and clinical diagnostics for many other diseases and metabolic disorders.

High-field asymmetric waveform ion mobility spectrometry (FAIMS) is an atmospheric pressure ion mobility technique that separates gas-phase ions by their behavior in strong and weak electric fields. FAIMS is easily interfaced with electrospray ionization and has been implemented as an additional separation mode between liquid chromatography (LC) and mass spectrometry (MS) in proteomic studies. FAIMS separation is orthogonal to both LC and MS and is used as a means of on-line fractionation to improve the detection of peptides in complex samples. FAIMS improves dynamic range and concomitantly the detection limits of ions by filtering out chemical noise. FAIMS can also be used to remove interfering ion species and to select peptide charge states optimal for identification by tandem MS. Here, the authors review recent developments in LC-FAIMS-MS and its application to MS-based proteomics.

Background: The study of iron metabolism is essential in nutritional sciences as iron deficiency is one of the most common nutritional deficiencies in humans and represents a serious health problem worldwide. The mouse is utilized as a unique and powerful model for the identification and characterization of genes involved in iron metabolism and for studying the pathogenesis of iron disorders. Thus, sophisticated and sensitive techniques have been developed to study iron metabolism in this animal model. In particular, iron absorption has been studied in mice by using the radioisotopes (55)Fe and (59)Fe in tied-off or dissected and everted duodenal segments. Nevertheless, several drawbacks discourage the extended use of these approaches., Methods and Results: Here, we report the use of the stable isotope (57)Fe to measure iron absorption in mice. We show that after oral administration of (57)Fe-containing solutions, it is possible to measure both duodenal iron retention and duodenal iron transfer to specific organs, using inductively coupled plasma mass spectrometry (ICP-MS). As (57)Fe is administered orally, no surgical operation is needed before the end of the experiment, thus allowing the measurement of iron absorption under physiologic conditions. Moreover, the use of ICP-MS for (57)Fe detection ensures high sensitivity and provides quantitative data. Finally, the use of a stable isotope enables the measurement of both iron absorption and histologic and/or biochemical analyses in the same animal., Conclusions: The use of (57)Fe to measure iron absorption in mice, therefore, represents an alternative to radioisotope-based methods, providing a new tool to extend our knowledge on the mechanism of iron absorption.

Vacuum ultraviolet (VUV) photon-induced ionization and fragmentation of N-methyl glycine (sarcosine) were investigated with infrared laser desorption/tunable synchrotron VUV photoionization mass spectrometry (IR LD/VUV PIMS) and theoretical calculations. Fragment-controllable mass spectra of sarcosine were measured at various photon energies. By tuning the photon energy, the fragments at m/z 44, 45, 43, 42, 30, and 60 were gradually detected. The ionization energy of the precursor was obtained by measuring the photoionization efficiency spectrum. Possible formation pathways of the fragment ions at m/z 44 (CH(3)NHCH(2)(+)), 45 (CH(3)NH(2)CH(2)(+)), 43 (CH(2)NHCH(2)(+)), 42 (CH(2)NCH(2)(+)), 30 (CH(2)NH(2)(+)), and 60 (CH(2)COOH(2)(+)) were discussed in detail with the help of calculations at the G3B3 and B3LYP/6-31++G(d,p) levels.

An algorithm is introduced to assess spectral quality for peptide CID spectra acquired by a quadrupole ion trap mass spectrometer. The method employs a quadratic discriminant function calibrated with manually classified 'bad' and 'good' quality spectra, producing a single 'spectral quality' score. Many spectra examined that do not have significant matches are assessed to have good spectral quality, indicating that advances in search methods may yield substantial improvements in results.

The rapid growth of mass spectrometry (MS)-based computer software applications has been fueled by the unprecedented need to capture and analyze MS data and provide the information necessary for decision-making. Shorter timelines and a significantly greater number of samples has resulted in a tremendous focus on streamlined approaches that provide scientists, managers, and executives the capability to readily obtain, or even request, the necessary information that leads to accelerated product development. The generation of analytical data using roboticized high-throughput hardware has produced a bottleneck since data can be generated faster than it can be analyzed. New techniques including MS/MS and accurate mass experiments are feasible only using computers to capture and manage the enormous amounts of data necessary to perform the experiments. Whatever the nature of the experiments conducted, the MS analysis strategy is to extract the appropriate information required for decision-making in as facile a manner as possible. We will review here a survey of the creation of commercial and laboratory specific reference databases and associated searching algorithms and also recent efforts to introduce advancedprocessing and analysis algorithms to the hands of the masses, specifically as an aid to structure elucidation.

The composition of propolis, a resinous hive product collected by honeybees from various plant sources, depends on various factors such as season and vegetation of the area. Based on standards (either isolated from Brazilian propolis or reported from propolis) including chromane, diterpenes and phenolic compounds, different Brazilian propolis were analysed by LC-MS in order to determine their chemical constituents. Dicaffeoylquinic acids were detected in almost all water extracts of Brazilian propolis, whereas diterpenes, flavonoids and prenylated phenolic compounds were found in their methanol extracts. Based on the identified chemical constituents and their biological activities, it was determined that the quality of Brazilian propolis could be directly related to the phenolic constituents. Moreover, Baccharis dracunculifolia was concluded to be an important source of Brazilian propolis. Propolis samples from Peru, China and the Netherlands were also studied.

Ants of different sexes and castes produce different odorous compounds. In Pheidole fallax Mayr, soldier ants produce an indole base, probably skatole, whereas minor workers produce a trial substance, Males of certain species of Lasius and Acanthomyops produce mixtures of terpenes and an indole base. These mixtures are discharged during mating pheromones. The terpene mixtures are qualitatively similar, but each species produces a blend of distinctive proportions. Citronellol and 2,6-dimethyl-5-hepten-1-ol have been identified in the mixtures by gas chromatography and mass spectrometry.
