Conversion of the lignocellulosic biomass to bioethanol contributes to the reduction of greenhouse gas emissions, enhancement of energy security, utilization of waste materials, and the promotion of sustainable agricultural practices. In this study, we report the effect of combining ball milling followed by liquid hot water (LHW) pretreatment of corn stover to lower the amount of enzyme required while also greatly increasing the recovery of xylose in fermentable form compared to either pretreatment alone. Short-duration ball milling for 60 min reduces the particle size of corn stover to 37.3 μm; however, the glucose only increased to 47% compared to 32% for unpretreated corn stover. In contrast, liquid hot water pretreatment alone can achieve increasing enzyme hydrolysis yields of cellulose from 49% to 93% as the pretreatment severity factor is increased from 3.24 to 4.41. However, the xylose yield decreased to 36% due to the fact that a considerable part of the xylose was degraded into furfural and humins. Surprisingly, the combination of mild ball milling (30 min) followed by mild liquid hot water pretreatment (190 °C, 15 min) could achieve both high glucose (83%) and xylose (72%) yields for a total sugar yield of 79%, theoretically. Thus, combining ball milling with liquid hot water pretreatment allows for milder conditions for both processes that lead to enhanced cellulose conversion without sacrificing xylose to degradation, which hinders enzymatic hydrolysis. Lignocellulosic biomass is a low-cost and environmentally-friendly resource that can be used to produce biofuels such as bioethanol and biogas, which are the leading candidates for the partial substitution of fossil fuels. However, the main challenge of using lignocellulosic materials for biofuel production is the low accessibility to cellulose for hydrolysis of enzymes and microorganisms, which can be overcome by pretreatment. Biological and chemical pretreatments have their own disadvantages, which could be reduced by combining the two methods. In this article, we review biological–chemical combined pretreatment strategies for biogas and bioethanol production. The synergy of fungal/enzyme–NaOH pretreatment is the only biological–chemical combination studied for biogas production and has proven to be effective. The use of enzyme, which is relatively expensive, has the advantage of hydrolysis efficiency compared to fungi. Nonetheless, there is vast scope for research and development of other chemical–biological combinations for biogas production. With respect to ethanol production, fungal–organosolv combination is widely studied and can achieve a maximum of 82% theoretical yield. Order of pretreatment is also important, as fungi may reduce the accessibility of cellulose made available by prior chemical strategies and suppress lignin degradation. The biofuel yield of similarly pretreated biomass can vary depending on the downstream process. The effects of CH3COOH and Na2SO3 pretreatment on the structural properties and hydrolyzability of fast-growing Paulownia elongate were investigated. Acetic acid increased cellulose's crystallinity and hydrolyzability when combined with alkaline sodium sulfite and sodium hydroxide. The cellulose content increased by 21%, the lignin content decreased by 6%, and the product showed better enzymatic digestibility. With a cellulase dose of 30 FPU/g DM, after 72 h hydrolysis, the hydrolysis yields of glucose and xylose were 78% and 83%, respectively, which were 51% and 69% higher than those of untreated materials. When the enzyme dosage was 20 FPU/g DM, after 72 h hydrolysis, the hydrolysis yields of glucose and xylose were 74% and 79%, respectively. The high hydrolyzability, low enzyme loading, and high hydrolysis yield demonstrate the potential of the proposed system for producing platform sugars from fast-growing Paulownia elongate. In this study the effect of nitrogen (N) supplementation in the fungal pretreatment of willow sawdust (WSD) via the white rot fungus Abortiporus biennis (A. biennis) was studied in terms of the fractionation of lignocellulosic biomass and biochemical methane potential (BMP). Thus, different external nitrogen sources (yeast extract (YE), urea (UR), and ammonium nitrate (AN)) at different ratios (N/C of 1/250 and 1/50) were added and the effect of the above parameters on the chemical composition of WSD during solid-state fermentation (SSF) experiments with A. biennis was assessed and compared to the experiment of fungal pretreatment without N supplementation (N/C was 1/500, control experiment). The results indicated that the addition of an external nitrogen source did not facilitate delignification, regardless of the type of nitrogen source and the ratio of N/C used. On the other hand, enhanced cellulose uptake was observed. Samples of the 28th day of cultivation, with and without N supplementation, were used for BMP tests, where a reduction in methane yield was observed, compared to the control experiment. In addition, a combination of fungal with alkali (20% NaOH w/w dry mass) pretreatment was performed in order to assess the effect of combined pretreatment on the lignocellulosic content and the BMP. Various pretreatment methods have been combined and employed for maximizing the solubilization of waste-activated sludge (WAS). However, the question "by changing the series of applied combined pretreatments (CPs), can the solubilization efficiency of WAS be affected?" has never been addressed. In this study, firstly, thermal (T), alkaline (A), and ultrasonic (U) pretreatments were individually applied at broad strengths (T = 80–120 °C for 30 min, A = pH 9–12, and U = 5–60 min at 300 W). Then, pretreatment conditions that caused similar solubilization (13.0%) (120 °C, pH 11, and 30 min for T, A, and U, respectively), were adopted for CP with reverse sequences of T&A, U&A, and T&U. A similar disintegration degree was observed in U→A and A→U, while a meaningful difference was found in T&A and T&U: T→A (28.3%), A→T (42.9%), T→U (22.9%), and U→T (27.1%). The difference in pretreatment series also affected the characteristics of soluble matters, which was analyzed by excitation emission matrix and molecular weight distribution. Due to these differences, the highest methane yield of 68.8% (based on (chemical oxygen demand) CODinput) was achieved at A→T, compared to T→A (62.3%). Our results suggested a simple strategy for increasing solubilization, at the same expense of energy, which might be beneficial in the following treatment process, such as dewatering and transportation. A combined ball milling and ethanol organosolv process is proposed for the pretreatment of three types of herbaceous biomass, giant miscanthus, corn stover, and wheat straw. The combined pretreatment was effective at both removing lignin and increasing the glucan content. After 120 min pretreatment, the glucan content increased to 63.09%, and 55.89% of the acid-insoluble lignin was removed from the giant miscanthus sample. The removal of cellulose, hemicellulose, and acetyl groups were correlated with the removal of lignin. The pretreatment of corn stover showed the highest removal of cellulose, but this was dependent on the removal of acid-insoluble lignin. The slope of the regression lines, which shows the correlation between the removal of lignin and cellulose, was lower than other correlations. The changes in biomass size were analyzed using size distribution graphs. With increasing pretreatment time, the particle size reduction improved in the three types of herbaceous biomass. Because of the combined physicochemical pretreatment, the enzymatic digestibility improved, and a maximum of 91% glucan digestibility was obtained from the pretreated corn stover when 30 FPU/g-glucan enzyme was added. A two-step pretreatment using NaOH and ozone was performed to improve the enzymatic hydrolysis, compositions and structural characteristics of corn stover. Comparison between the unpretreated and pretreated corn stover was also made to illustrate the mechanism of the combined pretreatment. A pretreatment with 2% (w/w) NaOH at 80 °C for 2 h followed by ozone treatment for 25 min with an initial pH 9 was found to be the optimal procedure and the maximum efficiency (91.73%) of cellulose enzymatic hydrolysis was achieved. Furthermore, microscopic observation of changes in the surface structure of the samples showed that holes were formed and lignin and hemicellulose were partially dissolved and removed. X-ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR) and Cross-Polarization Magic Angle Spinning Carbon-13 Nuclear Magnetic Resonance (CP/MAS 13C-NMR) were also used to characterize the chemical structural changes after the combined pretreatment. The results were as follows: part of the cellulose I structure was destroyed and then reformed into cellulose III, the cellulose crystal indices were also changed; a wider space between the crystal layer was observed; disruption of hydrogen bonds in cellulose and disruption of ester bonds in hemicellulose; cleavage of bonds linkage in lignin-carbohydrate complexes; removal of methoxy in lignin and hemicellulose. As a result, all these changes effectively reduced recalcitrance of corn stover and promoted subsequent enzymatic hydrolysis of cellulose. [ABSTRACT FROM AUTHOR]
