The mechanisms of the P. oxalicum SL2-mediated microbial community on phosphorus solubilization and Pb stabilization were investigated through a 90-day soil experiment. In the treatments inoculated with P. oxalicum SL2, the amount of P. oxalicum SL2-GFP remained at 77.8%-138.6% of the initial inoculation amount after 90 days, and the available phosphorus (AP) content increased 21.7%-40.8% while EDTA-Pb decreased 29.9%-43.2% compared with CK treatment. SEM-EDS results showed that P. oxalicum SL2 changed the agglomeration degree of microaggregates and promoted the combination of Pb with C and O elements. These phenomena were enhanced when applied with Ca 3 (PO 4 ) 2 . Microbial community analysis showed that P. oxalicum SL2 improved soil microbial activity, in which the fungi absolute abundance increased about 15 times within 90 days. Correlation analyses and a partial least-squares path model showed that the activation of Penicillium , Ascobolus , Humicola , and Spizellomyces in a fungal community increased the content of oxalate and AP, which directly decreased EDTA-Pb content, while the change of Bacillus , Ramlibacter , Gemmatimonas , and Candidatus Solibacter in the bacterial community regulated Fe/Mn/S/N cycle-related functions, thus promoting the conversion of Pb to oxidizable state. Our findings highlight that P. oxalicum SL2 enhanced the microbial-induced phosphate precipitation process by activating soil microbial communities and regulating their ecological functions.

A hydroponic experiment combined with synchronous radiation X-ray fluorescence (SRXRF) analysis was designed to understand the influence of humic acid (HA) in tea plants under lead stress. The results showed that the quantitative relationship (QR) between humic acid and Pb is an important factor affecting the regulation of humic acid with respect to the accumulation of Pb in tea plants. Besides, excess humic acid might stimulate the accumulation of Pb in the root cell wall and transfer to the shoot organs through undifferentiated casparian band structure. This study could provide a theoretical basis for the scientific evaluation of the effect of humic acid on tea uptake and the accumulation of Pb and the practical application of humic acid in reducing Pb pollution in the field.

Photoreduction is a major obstacle for using the X-ray absorption near-edge structure (XANES) fingerprint to perform metal speciation at the molecular level in biological and environmental samples, especially for metalloproteins. In this study, soft X-ray induced photoreduction was observed in organic Cu(II) compounds during XANES measurements in a third-generation synchrotron source. Next Cu L(3)-edge, O K-edge, and C K-edge XANES spectroscopy, together with the scanning transmission X-ray microscopy (STXM), were used to probe the specific radiation damage processes of Cu acetate with similar local structures to Cu metalloproteins. Breakup of the Cu-Cu bond was hypothesized for the initial photoreduction of Cu acetate. The following radiation damage of Cu acetate produced CuO and an organic Cu(I) compound with a C═C bond, and the further photoreduction of the resulting CuO to Cu metal was also demonstrated. Our results indicated the importance of consideration of photoreduction during soft XANES measurements for the solid state compounds with high valence metals. Reducing the radiation dose to ~0.1 MGy effectively prevented the photoreduction of organic Cu(II) compounds during these measurements. In recent years, increasing concern has surrounded the consequences of improper electric and electronic waste (e-waste) disposal. In order to mitigate or remediate the potentially severe toxic effects of e-waste recycling on the environment, organisms, and humans, many contaminated sites must first be well-characterized. In this study, soil samples were taken from Taizhou city, one of the largest e-waste disposal centers in China, which was involved in recycling for nearly 30 years. The extracts of the samples were assayed for aryl hydrocarbon receptor (AhR)-mediated ethoxyresorufin-O-deethylase (EROD) induction in the rat hepatoma cell line H4IIE. Some of the target AhR agonists, including polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs), and polycyclic aromatic hydrocarbons (PAHs), were instrumentally analyzed as well. The cause-effect relationship and dose-response relationship between the chemical concentrations of AhR agonists and observed EROD activity were examined. The results showed that soil extracts could induce AhR activity significantly, and the chemically derived 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) equivalents (TEQcal) were perfectly correlated to bioassay-derived TCDD equivalents (TEQbio; R = 0.96, P < 0.001), which indicated that the known AhR agonists could account for the observed responses. Among different contributors, PCBs accounted for 87.2-98.2% and PCDD/Fs contributed 1.7-11.6% of TEQcal, while the contribution of PAHs could almost be neglected. Under these conditions, a quantitative dose-effect relationship between TEQ(PCB) and EROD activity could be evaluated, suggesting that the observed AhR effect was mainly caused by PCBs. Further source identification by congener profiles analysis showed that the crude dismantling of electric power devices and open burning of electric wires and printed circuit boards may be the main sources of these dioxin-like compounds. This study suggests that the combination of in vitro bioassay and chemical analysis is useful to screen, identify, and prioritize AhR agonists in soil from e-waste recycling areas.
