The goal of this research was to demonstrate the effectiveness of optimization techniques and test the possibility of using ABS plastic to optimize the existing crane hooks that use these techniques. Based on the optimization results, verification of the modified hook, and structural stability, this optimization reduces the weight of the crane hook while maintaining durability. This study utilized 3D modeling software to optimize the design of an existing hook, ensuring both sufficient strength and weight reduction. The research analyzes the results, concentrating on the stress that falls within the material strength limit. The research begins by utilizing a maximum of 1 kg of ABS plastic and a 5 kg liftable load. Using topology techniques, the study proposed five models that meet technical requirements. As a result, the proposed structure is highly economical and optimized in terms of materials and time, while still ensuring working performance. This work utilizes 3D modeling software for optimization design. It was effective to combine 3D printing manufacturing technology with size considerations. This article serves as a reference for future research and hook design.

This paper presents a Blockchain-based framework for providing Blockchain services for purposes of stability in terms of consensus protocol infrastructure and governance mechanisms and accessible auxiliary services suitable for the vast majority of current business needs, including fundamental factors such as digital identity with autonomous identity, building solutions to ensure transaction privacy with zero-knowledge proofs, and other services related to digital assets. The proposed framework helps promote digital transformation for businesses, especially small and medium enterprises with limited resources and costs, to apply Blockchain technology to their business models, increasing competitive advantages and assisting the companies in focusing on business logic while still using Blockchain technology in their functions.

Dynamic Wireless Charging (DWC) systems for electric vehicles (EVs) are being studied and developed for wide applications. To ensure a long life for lithium-ion batteries, Constant Current (CC) charging is required. However, the equivalent load of the battery changes during CC charging, which reduces the system's efficiency. To solve that problem, this paper proposes a new control method that combines CC charging and improves transfer efficiency using only an active rectifier on the secondary side of the DWC system. Moreover, this study also proposes a method to estimate the coupling coefficient through the parameters measured on the secondary side without the need for wireless communication between the two sides. A model of a 1.5 kW DWC system with a transfer distance of 150 mm was built in a laboratory to verify the accuracy of the proposed method. The results showed that the charging current reached the required value, and the maximum system efficiency was 85%.

In this paper, we formulate and prove the so-called $p$-adic non-commutative analytic subgroup theorem. This result is seen as the $p$-adic analogue of a recent theorem given by Yafaev in [11].

The fusion of low-cost sensor networks with air quality stations has become prominent, offering a cost-effective approach to gathering fine-scaled spatial data. However, effective integration of diverse data sources while maintaining reliable information remains challenging. This paper presents an extended clustering method based on the Girvan-Newman algorithm to identify spatially correlated clusters of sensors and nearby observatories. The proposed approach enables localized monitoring within each cluster by partitioning the network into communities, optimizing resource allocation and reducing redundancy. Through our simulations with real-world data collected from the state-run air quality monitoring stations and the low-cost sensor network in Sydney’s suburbs, we demonstrate the effectiveness of this approach in enhancing localized monitoring compared to other clustering methods, namely K-Means Clustering, Density-Based Spatial Clustering of Applications with Noise (DBSCAN) and Agglomerative Clustering. Experimental results illustrate the potential for this method to facilitate comprehensive and high-resolution air quality monitoring systems, advocating the advantages of integrating low-cost sensor networks with conventional monitoring infrastructure.

Osteocytes play an important role in the development and progression of tumour-associated bone disease. Here the authors report an interaction between malignant plasma cells and osteocytes in multiple myeloma and show that the osteocyte-expressed major histocompatibility complex class II transactivator (CIITA) contributes to myeloma-induced bone lesions.

We discuss the p -adic Weierstrass zeta functions associated with elliptic curves defined over the field of algebraic numbers and linear relations for their values in the p -adic domain. Abstract Background Therapeutic resistance occurs in most patients with multiple myeloma (MM). One of the key mechanisms for MM drug resistance comes from the interaction between MM cells and adipocytes that inhibits drug-induced apoptosis in MM cells; MM cells reprogram adipocytes to morph into different characterizations, including exosomes, which are important for tumor-stroma cellular communication. However, the mechanism by which exosomes mediate the cellular machinery of the vicious cycle between MM cells and adipocytes remains unclear. Methods Adipocytes were either isolated from bone marrow aspirates of healthy donors or MM patients or derived from mesenchymal stem cells. Co-culturing normal adipocytes with MM cells was used to generate MM-associated adipocytes. Exosomes were collected from the culture medium of adipocytes. Annexin V-binding and TUNEL assays were performed to assess MM cell apoptosis. Methyltransferase activity assay and dot blotting were used to access the m6A methylation activity of methyltransferase like 7A (METTL7A). RIP, MeRIP-seq, and RNA–protein pull down for assessing the interaction between long non-cording RNAs (LncRNAs) and RNA binding proteins were performed. Adipocyte-specific enhancer of zeste homolog 2 (EZH2) knockout mice and MM-xenografted mice were used for evaluating MM therapeutic response in vivo. Results Exosomes collected from MM patient adipocytes protect MM cells from chemotherapy-induced apoptosis. Two LncRNAs in particular, LOC606724 and SNHG1, are significantly upregulated in MM cells after exposure to adipocyte exosomes. The raised LncRNA levels in MM cells are positively correlated to worse outcomes in patients, indicating their clinical relevancy in MM. The functional roles of adipocyte exosomal LOC606724 or SNHG1 in inhibition of MM cell apoptosis are determined by knockdown in adipocytes or overexpression in MM cells. We discovered the interactions between LncRNAs and RNA binding proteins and identified methyltransferase like 7A (METTL7A) as an RNA methyltransferase. MM cells promote LncRNA package into adipocyte exosomes through METTL7A-mediated LncRNA m6A methylation. Exposure of adipocytes to MM cells enhances METTL7A activity in m6A methylation through EZH2-mediated protein methylation. Conclusion This study elucidates an unexplored mechanism of how adipocyte-rich microenvironment exacerbates MM therapeutic resistance and indicates a potential strategy to improve therapeutic efficacy by blocking this vicious exosome-mediated cycle.

The Upper Hunter Valley is a major coal mining area in New South Wales (NSW), Australia. Due to the ongoing increase in mining activities, PM10 (air-borne particles with an aerodynamic diameter less than 10 micrometres) pollution has become a major air quality concern in local communities. The present study was initiated to quantitatively examine the spatial and temporal variability of PM10 pollution in the region. An earlier paper of this study identified two air quality subregions in the valley. This paper aims to provide a holistic summarisation of the relationships between elevated PM10 pollution in two subregions and the local- and synoptic-scale meteorological conditions for spring and summer, when PM10 pollution is relatively high. A catalogue of twelve synoptic types and a set of six local meteorological patterns were quantitatively derived and linked to each other using the self-organising map (SOM) technique. The complex meteorology–air pollution relationships were visualised and interpreted on the SOM planes for two representative locations. It was found that the influence of local meteorological patterns differed significantly for mean PM10 levels vs. the occurrence of elevated pollution events and between air quality subregions. In contrast, synoptic types showed generally similar relationships with mean vs. elevated PM10 pollution in the valley. Two local meteorological patterns, the hot–dry–northwesterly wind conditions and the hot–dry–calm conditions, were found to be the most PM10 pollution conducive in the valley when combined with a set of synoptic counterparts. These synoptic types are featured with the influence of an eastward migrating continental high-pressure system and westerly troughs, or a ridge extending northwest towards coastal northern NSW or southern Queensland from the Tasman Sea. The Upper Hunter Valley is a major coal mining area containing approximately 40% of the currently identified total coal reserves in New South Wales (NSW), Australia. Due to the ongoing increase in mining activities, PM10 (airborne particles with an aerodynamic diameter of less than 10 micrometres) pollution has become a major air quality concern in local communities. This paper summarises the spatial and temporal variability modes of PM10 pollution in the region, based on long-term multi-site monitoring data and the application of the rotated principal component analysis (RPCA) and wavelet analysis techniques. RPCA identified two distinct air quality clusters/subregions in the valley: one in the west/northwest and the other in the southeast. Wavelet analysis revealed the annual cycle to be the most persistent temporal mode of PM10 variability in both subregions, with intermittent signals also observed at time scales of around 120, 30~90, and under 30 days. How these variation modes are related to the effects of local PM10 emissions and the influence of meteorology at different time scales deserves further attention in future work. The findings will be used in air quality reporting and forecasting in NSW. The methodology and results can also be useful for air quality research in similar regions elsewhere. Circular RNAs (circRNAs) are a class of single-stranded closed RNA molecules that are formed by precursor mRNA back-splicing or skipping events of thousands of genes in eukaryotes as covalently closed continuous loops. High-throughput sequencing and bioinformatics approaches have uncovered the broad expression of circRNAs across species. Their high stability, abundance, and evolutionary conservation among species points to their distinct properties and diverse cellular functions as efficient microRNAs and protein sponges; they also play important roles in modulating transcription and splicing. Additionally, most circRNAs are aberrantly expressed in pathological conditions and in a tissue-specific manner such as development and progression of cancer. Herein, we highlight the characteristics, functions, and mechanisms of action of circRNAs in cancer; we also provide an overview of recent progress in the circRNA field and future application of circRNAs as cancer biomarkers and novel therapeutic targets. Lung cancer is the leading cause of cancer-associated deaths worldwide. In particular, non-small-cell lung cancer (NSCLC) cells harboring epidermal growth factor receptor (EGFR) mutations are associated with resistance development of EGFR tyrosine kinase inhibitor (EGFR-TKI) treatment. Recent findings suggest that bone morphogenetic proteins (BMPs) and microRNAs (miRNAs) might act as oncogenes or tumor suppressors in the tumor microenvironment. In this study, for the first time, we identified the potential roles of BMPs and miRNAs involved in EGFR-TKI resistance by analyzing datasets from a pair of parental cells and NSCLC cells with acquired EGFR-TKI resistance. BMP4 was observed to be significantly overexpressed in the EGFR-TKI-resistant cells, and its mechanism of action was strongly associated with the induction of cancer cell energy metabolism through the modulation of Acyl-CoA synthetase long-chain family member 4. In addition, miR-139-5p was observed to be significantly downregulated in the resistant NSCLC cells. The combination of miR-139-5p and yuanhuadine, a naturally derived antitumor agent, synergistically suppressed BMP4 expression in the resistant cells. We further confirmed that LDN-193189, a small molecule BMP receptor 1 inhibitor, effectively inhibited tumor growth in a xenograft nude mouse model implanted with the EFGR-TKI-resistant cells. These findings suggest a novel role of BMP4-mediated tumorigenesis in the progression of acquired drug resistance in EGFR-mutant NSCLC cells. Epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs) are used clinically as target therapies for lung cancer patients, but the occurrence of acquired drug resistance limits their efficacy. Nicotinamide N-methyltransferase (NNMT), a cancer-associated metabolic enzyme, is commonly overexpressed in various human tumors. Emerging evidence also suggests a crucial loss of function of microRNAs (miRNAs) in modulating tumor progression in response to standard therapies. However, their precise roles in regulating the development of drug-resistant tumorigenesis are still poorly understood. Herein, we established EGFR-TKI-resistant non-small-cell lung cancer (NSCLC) models and observed a negative correlation between the expression levels of NNMT and miR-449a in tumor cells. Additionally, knockdown of NNMT suppressed p-Akt and tumorigenesis, while re-expression of miR-449a induced phosphatase and tensin homolog (PTEN), and inhibited tumor growth. Furthermore, yuanhuadine, an antitumor agent, significantly upregulated miR-449a levels while critically suppressing NNMT expression. These findings suggest a novel therapeutic approach for overcoming EGFR-TKI resistance to NSCLC treatment.

Bone morphogenetic proteins (BMPs) are a diverse class of molecules with over 20 growth factor proteins that belong to the transforming growth factor-β (TGF-β) family and are highly associated with bone formation and disease development. Aberrant expression of various BMPs has been reported in several cancer tissues. Biological function studies have elicited the dual role of BMPs in both cancer development and suppression. Furthermore, a variety of BMP antagonists, ligands, and receptors have been shown to reduce or enhance tumorigenesis and metastasis. Knockout mouse models of BMP signaling components have also revealed that the suppression of BMP signaling impairs cancer metastasis. Herein, we highlight the basic clinical background and involvement of BMPs in modulating cancer progression and their dynamic interactions (e.g., with microRNAs) in the tumor microenvironment in addition to their mutations and roles in chemoprevention. We also suggest that BMPs should be considered a powerful putative therapeutic target in tumorigenesis and bone metastasis. Two small genera of the subfamily Braconinae, Angustibracon Quicke, 1987 and Pseudospinaria Enderlein, 1905, are reported for the first time from Vietnam. Two Angustibracon species, Angustibracon leptogaster Cameron and Angustibracon maculiabdominis Zhou & You and one Pseudospinaria species, Pseudospinaria attenuata (Westwood) are newly recorded for Vietnam's braconid fauna. Additionally, the species are redescribed and illustrated. An illustrated key to the two known Augustibracon species and a mapped distribution of species in Vietnam are also given. Using data from nationwide surveys of provincial institutions and private manufacturing small medium enterprises, this study provided the first evidence of the impact of provincial institution quality and firms’ participation in and intensity of corrupt activities on firm productivity in Vietnam. We found that the bribe intensity instead of whether firms bribed state officials or not (measured by a dummy variable) has a negative effect on firm productivity when the endogeneity of corruption and unobservable characteristics are controlled for. This finding contrasts to a popular belief about a paradox for East Asian countries where corruption is positively associated with firm growth.

Hypoxia inducible factor-1α (HIF-1α) is an essential regulator of the cellular response to low oxygen concentrations, activating a broad range of genes that provide adaptive responses to oxygen deprivation. HIF-1α is overexpressed in various cancers and therefore represents a considerable chemotherapeutic target. Salternamide A (SA), a novel small molecule that is isolated from a halophilic Streptomyces sp., is a potent cytotoxic agent against a variety of human cancer cell lines. However, the mechanisms by which SA inhibits tumor growth remain to be elucidated. In the present study, we demonstrate that SA efficiently inhibits the hypoxia-induced accumulation of HIF-1α in a time- and concentration-dependent manner in various human cancer cells. In addition, SA suppresses the upstream signaling of HIF-1α, such as PI3K/Akt/mTOR, p42/p44 MAPK, and STAT3 signaling under hypoxic conditions. Furthermore, we found that SA induces cell death by stimulating G2/M cell cycle arrest and apoptosis in human colorectal cancer cells. Taken together, SA was identified as a novel small molecule HIF-1α inhibitor from marine natural products and is potentially a leading candidate in the development of anticancer agents.

Along with its rapid urban development, Ho Chi Minh City (HCMC) in recent years has suffered a high concentration of air pollutants, especially fine particulate matters or PM2.5. A comprehensive study is required to evaluate the air quality conditions and their health impact in this city. Given the lack of adequate air quality monitoring data over a large area of the size of HCMC, an air quality modeling methodology is adopted to address the requirement. Here, by utilizing a corresponding emission inventory in combination with The Air Pollution Model-Chemical Transport Model (TAPM-CTM), the predicted concentration of air pollutants is first obtained for PM2.5, NOx, and SO2. Then by associating the pollutants exposed with the mortality rate from three causes, namely Ischemic Heart Disease (IHD), cardiopulmonary, and lung cancer, the impact of air pollution on human health is obtained for this purpose. Spatial distribution has shown a high amount of pollutants concentrated in the central city with a high density of combustion vehicles (motorcycles and automobiles). In addition, a significant amount of emissions can be observed from stevedoring and harbor activities, including ferries and cargo handling equipment located along the river. Other sources such as household activities also contribute to an even distribution of emission across the city. The results of air quality modeling showed that the annual average concentrations of NO2 were higher than the standard of Vietnam National Technical Regulation on Ambient Air Quality (QCVN 05: 2013 40 µg/m3) and World Health Organization (WHO) (40 µg/m3). The annual average concentrations of PM2.5 were 23 µg/m3 and were also much higher than the WHO (10 µg/m3) standard by about 2.3 times. In terms of public health impacts, PM2.5 was found to be responsible for about 1136 deaths, while the number of mortalities from exposure to NO2 and SO2 was 172 and 89 deaths, respectively. These figures demand some stringent measures from the authorities to potentially remedy the alarming situation of air pollution in HCM City.

A better understanding of background tropospheric ozone delivers multiple benefits. Robust estimates of regional background ozone are required to understand the limits of anthropogenic emissions controlling ozone reduction. Long-term estimates of background ozone assist in characterising changes in atmospheric composition and can help quantify the influence of human activity on the atmosphere. Background tropospheric ozone measurements representative of continental air masses are scarce in Australia. Here, we use k-means clustering to identify a cluster of measurements from the long-term air quality monitoring station at Oakdale, NSW, which are likely to be representative of background air. The cluster is associated with NOx-limited air masses of continental origin. From this analysis, we estimate background ozone representative of Eastern Australia. We find recent (2017–2022) mean ozone mixing ratios of 28.5 ppb and identify a statistically significant (α = 0.05) trend in the mean of +1.8 (1.0–2.8) ppb/decade. Our methods demonstrate that some long-term monitoring stations within or near urban areas can provide suitable conditions and datasets for regional Global Atmosphere Watch monitoring. Aberrant activation of hepatocyte growth factor (HGF)/c-Met signaling pathway caused by gene amplification or mutation plays an important role in tumorigenesis. Therefore, c-Met is considered as an attractive target for cancer therapy and c-Met inhibitors have been developed with great interests. However, cancers treated with c-Met inhibitors inevitably develop resistance commonly caused by the activation of PI3K/Akt signal transduction pathway. Therefore, the combination of c-Met and PI3Kα inhibitors showed synergistic activities, especially, in c-Met hyperactivated and PIK3CA-mutated cells. In our previous study, we rationally designed and synthesized DFX117(6-(5-(2,4-difluorophenylsulfonamido)-6-methoxypyridin-3-yl)-N-(2-morpholinoethyl) imidazo[1,2-a]pyridine-3-carboxamide) as a novel PI3Kα selective inhibitor. Herein, the antitumor activity and underlying mechanisms of DFX117 against non-small cell lung cancer (NSCLC) cells were evaluated in both in vitro and in vivo animal models. Concurrent targeted c-Met and PI3Kα by DFX117 dose-dependent inhibited the cell growth of H1975 cells (PIK3CA mutation and c-Met amplification) and A549 cells (KRAS mutation). DFX117 subsequently induced G0/G1 cell cycle arrest and apoptosis. These data highlight the significant potential of DFX117 as a feasible and efficacious agent for the treatment of NSCLC patients.

Besides the critical functions in hemostasis, thrombosis and the wounding process, platelets have been increasingly identified as active players in various processes in tumorigenesis, including angiogenesis and metastasis. Once activated, platelets can release bioactive contents such as lipids, microRNAs, and growth factors into the bloodstream, subsequently enhancing the platelet⁻cancer interaction and stimulating cancer metastasis and angiogenesis. The mechanisms of treatment failure of chemotherapeutic drugs have been investigated to be associated with platelets. Therefore, understanding how platelets contribute to the tumor microenvironment may potentially identify strategies to suppress cancer angiogenesis, metastasis, and drug resistance. Herein, we present a review of recent investigations on the role of platelets in the tumor-microenvironment including angiogenesis, and metastasis, as well as targeting platelets for cancer treatment, especially in drug resistance.
