The time required to recover from cold exposure (chill coma recovery time) may represent an important metric of performance and has been linked to geographic distributions of diverse species. Chill coma recovery time (CCRT) has rarely been measured in bumble bees (genus Bombus) but may provide insights regarding recent changes in their distributions. We measured CCRT of Bombus vosnesenskii workers reared in common garden laboratory conditions from queens collected across altitude and latitude in the Western United States. We also compared CCRTs of male and female bumble bees because males are often overlooked in studies of bumble bee ecology and physiology and may differ in their ability to respond to cold temperatures. We found no relationship between CCRT and local climate at the queen collection sites, but CCRT varied significantly with sex and body mass. Because differences in the ability to recover from cold temperatures have been shown in wild-caught Bombus, we predict that variability in CCRT may be strongly influenced by plasticity. Bumble bees (Bombus) are vitally important pollinators of wild plants and agricultural crops worldwide. Fragmentary observations. however, have suggested population declines in several North American species. Despite rising concern over these observations in the United States, highlighted in a recent National Academy of Sciences report, a national assessment of the geographic scope and possible causal factors of bumble bee decline is lacking. Here, we report results of a 3-y interdisciplinary study of changing distributions, population genetic structure, and levels of pathogen infection in bumble bee populations across the United States. We compare current and historical distributions of eight species, compiling a database of >73,000 museum records for comparison with data from intensive nationwide surveys of >16,000 specimens. We show that the relative abundances of four species have declined by up to 96% and that their surveyed geographic ranges have contracted by 23- 87%, some within the last 20 y. We also show that declining populations have significantly higher infection levels of the microsporidian pathogen Nosema bombi and lower genetic diversity compared with co-occurring populations of the stable (nondeclining) species. Higher pathogen prevalence and reduced genetic diversity are, thus, realistic predictors of these alarming patterns of decline in North America, although cause and effect remain uncertain. To replace deceased colonies or to increase the colony numbers, beekeepers often purchase honey bees, Apis mellifera L., in a package, which is composed of 909-1,364 g (2-3 lb) of worker bees and a mated queen. Packages are typically produced in warm regions of the United States in spring and shipped throughout the United States to replace colonies that perished during winter. Although the package bee industry is effective in replacing colonies lost in winter, packages also can be an effective means of dispersing diseases, parasites, and undesirable stock to beekeepers throughout the United States. To evaluate the quality of packages, we examined 48 packages representing six lines of bees purchased in the spring 2006. We estimated levels of the parasitic mite Varroa destructor Anderson & Trueman and the percentage of drone (male) honey bees received in packages. We surveyed for presence of the tracheal honey bee mite, Acarapi.s woodi (Rennie), and a microsporidian parasite, Nosema spp., in the shipped bees. We found significant differences in both the mean Varroa mite perbee ratios (0.004-0.054) and the average percentage of drones (0.04-5.1%) in packages from different producers. We found significant differences in the number of Nosema-infected packages (0.0-75.0%) among the six lines. No packages contained detectable levels of A. woodi. Considering the observed variability among honey bee packages, beekeepers should be aware of the potential for pest and disease infestations and high drone levels in packages. [ABSTRACT FROM AUTHOR]

Abstract: Several bumble bee (Bombus) species in North America have undergone range reductions and rapid declines in relative abundance. Pathogens have been suggested as causal factors, however, baseline data on pathogen distributions in a large number of bumble bee species have not been available to test this hypothesis. In a nationwide survey of the US, nearly 10,000 specimens of 36 bumble bee species collected at 284 sites were evaluated for the presence and prevalence of two known Bombus pathogens, the microsporidium Nosema bombi and trypanosomes in the genus Crithidia. Prevalence of Crithidia was ⩽10% for all host species examined but was recorded from 21% of surveyed sites. Crithidia was isolated from 15 of the 36 Bombus species screened, and were most commonly recovered from Bombus bifarius, Bombus bimaculatus, Bombus impatiens and Bombus mixtus. Nosema bombi was isolated from 22 of the 36 US Bombus species collected. Only one species with more than 50 sampled bees, Bombus appositus, was free of the pathogen; whereas, prevalence was highest in Bombus occidentalis and Bombus pensylvanicus, two species that are reportedly undergoing population declines in North America. A variant of a tetranucleotide repeat in the internal transcribed spacer (ITS) of the N. bombi rRNA gene, thus far not reported from European isolates, was isolated from ten US Bombus hosts, appearing in varying ratios in different host species. Given the genetic similarity of the rRNA gene of N. bombi sampled in Europe and North America to date, the presence of a unique isolate in US bumble could reveal one or more native North American strains and indicate that N. bombi is enzootic across the Holarctic Region, exhibiting some genetic isolation. Bumble bees are integral pollinators of native and cultivated plant communities, but species are undergoing significant changes in range and abundance on a global scale. Climate change and land cover alteration are key drivers in pollinator declines; however, limited research has evaluated the cumulative effects of these factors on bumble bee assemblages. This study tests bumble bee assemblage (calculated as richness and abundance) responses to climate and land use by modeling species-specific habitat requirements, and assemblage-level responses across geographic regions. We integrated species richness, abundance, and distribution data for 18 bumble bee species with site-specific bioclimatic, landscape composition, and landscape configuration data to evaluate the effects of multiple environmental stressors on bumble bee assemblages throughout 433 agricultural fields in Florida, Indiana, Kansas, Kentucky, Maryland, South Carolina, Utah, Virginia, and West Virginia from 2018 to 2020. Distinct east versus west groupings emerged when evaluating species-specific habitat associations, prompting a detailed evaluation of bumble bee assemblages by geographic region. Maximum temperature of warmest month and precipitation of driest month had a positive impact on bumble bee assemblages in the Corn Belt/Appalachian/northeast, southeast, and northern plains regions, but a negative impact on the mountain region. Further, forest land cover surrounding agricultural fields was highlighted as supporting more rich and abundant bumble bee assemblages. Overall, climate and land use combine to drive bumble bee assemblages, but how those processes operate is idiosyncratic and spatially contingent across regions. From these findings, we suggested regionally specific management practices to best support rich and abundant bumble bee assemblages in agroecosystems. Bumble bees are ecologically and economically important insect pollinators. Three abundant and widespread species in western North America, Bombus bifarius , Bombus vancouverensis , and Bombus vosnesenskii , have been the focus of substantial research relating to diverse aspects of bumble bee ecology and evolutionary biology. We present de novo genome assemblies for each of the three species using hybrid assembly of Illumina and Oxford Nanopore Technologies sequences. All three assemblies are of high quality with large N50s (> 2.2 Mb), BUSCO scores indicating > 98% complete genes, and annotations producing 13,325 - 13,687 genes, comparing favorably with other bee genomes. Analysis of synteny against the most complete bumble bee genome, Bombus terrestris , reveals a high degree of collinearity. These genomes should provide a valuable resource for addressing questions relating to functional genomics and evolutionary biology in these species., (Copyright © 2020 Heraghty et al.)

Mermithid nematodes (Nematoda: Mermithida: Mermithidae) parasitize a wide range of both terrestrial and aquatic invertebrate hosts, yet are recorded in bumble bees (Insecta: Hymenoptera: Apidae: Bombus) only six times historically. Little is known about the specific identity of these parasites. In a single-season nationwide survey of internal parasites of 3646 bumble bees, we encountered six additional instances of mermithid parasitism in four bumble bee species and genetically characterized them using two regions of 18S to identify the specific host-parasite relationships. Three samples from the northeastern USA are morphologically and genetically identified as Mermis nigrescens, whereas three specimens collected from a single agricultural locality in the southeast USA fell into a clade with currently undescribed species. Nucleotide sequences of the V2-V6 region of 18S from the southeastern specimens were 2.6-3.0% divergent from one another, and 2.2-4.0% dissimilar to the nearest matches to available data. The dearth of available data prohibits positive identification of this parasite and its affinity for specific bumble bee hosts. By doubling the records of mermithid parasitism of bumble bee hosts and providing genetic data, this work will inform future investigations of this rare phenomenon.

We evaluated the performance of six named types of package honey bees, Apis mellifera L (Hymenoptera: Apidae), from four commercial producers. We examined the effects of levels of the parasitic mite Varroa destructor Anderson & Trueman, the endoparasitic mite Acarapis woodi (Rennie), the gut parasite Nosema (species not determined) in samples from bees in 48 packages, and levels of adult drones in the same packages on corresponding levels of those same traits in the fall in colonies that developed from those 48 packages. After package installation, we measured the rate of queen failure, the removal of freeze-killed brood (an assay to assess hygienic behavior), varroa-sensitive hygiene, and short-term weight gain in all colonies. We examined the correlations among these traits and the effect of initial package conditions and package-type on the expression of these traits. In general, differences among sources were not significant, except that we did observe significant differences in the proportion of mite infected worker brood in the fall. There was no significant difference in weight gain in colonies established from nosema-infected packages versus those established from noninfected packages. Freeze-killed hygienic behavior and varroa-sensitive hygienic behavior were positively correlated, suggesting that both traits could be selected simultaneously. Neither trait was correlated with colony weight gain, suggesting that both traits could be selected without compromising honey production.
