The multifunctional active smart biomass film was prepared by incorporating chitosan-adsorbed laurate esterified starch curcumin Pickering emulsion into the starch film matrix, with nano-cellulose serving as reinforcing agents. The mechanical and functional properties of the film were studied, and the film was used to monitor the freshness of pork. The results demonstrated a relatively uniform distribution of curcumin and Pickering emulsion droplets within the film matrix. Furthermore, the thermal stability was minimally impacted by the introduction of curcumin Pickering emulsion, while the tensile strength and tensile strain of the film were increased, and both its hydrophobicity and antioxidant properties were improved. The free radical scavenging rate reached 56.01 %, with sustained high antioxidant capacity even after 8 days. Additionally, the presence of curcumin provided the film with pH indicating ability and delayed pork spoilage. Lignin, a natural pol2ymer with a complex structure that is difficult to separate, is prone to C-C bond condensation during the separation process. To reduce the condensation of lignin, here, a novel method is proposed for separating the components by using a combination of maleic acid (MA)/ozone (O 3 ) to co-treat wheat straw. The removal of lignin, glucan, and xylan was 38.07 ± 0.2%, 31.44 ± 0.1%, and 71.98 ± 0.1%, respectively, under the conditions of ball-milling of wheat straw for 6 h, reaction temperature of 60 °C, and O 3 holding time of 9 min. Lignin-rich solutions were collected to extract the dissolved lignin (DL) after washing the treated samples. The DL obtained under MA/O 3 conditions had a carboxyl group (-COOH) content of 2.96 mmol/g. The carboxyl group of MA underwent esterification with the hydroxyl group (-OH) at the γ position of lignin and O 3 reacted on the positions of the lignin side chain or the phenolic ring, resulting in a break in the side chain and the opening of the phenolic ring to introduce the carboxyl group. The 2D-HSQC-NMR results revealed that the phenolic ring-opening reaction of lignin in the presence of O 3 was essentially free of β-β and β-5 condensation bonds.

Single-atom catalysts (SACs) with active metals dispersed atomically have shown great potential in heterogeneous catalysis due to the high atomic utilization and superior selectivity/stability. Synthesis of SACs using carbon-neutral biomass and its components as the feedstocks provides a promising strategy to realize the sustainable and cost-effective SACs preparation as well as the valorization of underused biomass resources. Herein, we begin by describing the general background and status quo of carbon-based SACs derived from biomass. A detailed enumeration of the common biomass feedstocks (e.g., lignin, cellulose, chitosan, etc.) for the SACs preparation is then offered. The interactions between metal atoms and biomass-derived carbon carriers are summarized to give general rules on how to stabilize the atomic metal centers and rationalize porous carbon structures. Furthermore, the widespread adoption of catalysts in diverse domains (e.g., chemocatalysis, electrocatalysis and photocatalysis, etc.) is comprehensively introduced. The structure-property relationships and the underlying catalytic mechanisms are also addressed, including the influences of metal sites on the activity and stability, and the impact of the unique structure of single-atom centers modulated by metal/biomass feedstocks interactions on catalytic activity and selectivity. Finally, we end this review with a look into the remaining challenges and future perspectives of biomass-based SACs. Current DES pretreatment is often performed under relatively severe conditions with high temperature, long time, and high DES usage. This work studied a short-time diol DES (deep eutectic solvent) pretreatment under mild conditions to fractionate the bamboo, facilitate enzymatic hydrolysis, and obtain high-quality lignin. At an optimized condition of 130 °C for only 10 min, lignin and xylan removal reached 61.34 % and 84.15 %, with residual glucan showing a ~90 % enzymatic hydrolysis yield. Equally important, the dissolved lignin could be readily recovered with 97.51 % yield, exhibiting 96.65 % β-O-4 preservation. The fractionation and lignin protection mechanisms were unveiled by XRD, FTIR, cellulose-DP, 2D HSQC NMR, 31 P NMR and GPC analysis. Three-dimensional (3D) printing, known as revolutionary and disruptive innovation in manufacturing technology, supports great opportunities to rapidly construct a wide range of tailored object geometries. Cellulose, hemicellulose, and lignin as the three most common natural polymers and main components of plant resources, possess great economical potential for bio-based products due to their attractive advantages. The integration of 3D printing technology involved with cellulose, hemicellulose and lignin as the major bio-based feedstock for high-performance 3D printed products has received great concern in the R&D areas. In this review, the aim is to shed light on a cutting-edge review on the most recent progress based on cellulose, hemicellulose and lignin, as well as their derivatives as multi-components of bio-feedstock for 3D printing, in which the applications, roles and functions of the plant-derived biomass for 3D printing are also highlighted. In this study, a series of conductive composite wires were successfully prepared by combining dispersions of multi-wall carbon nanotubes (MWCNTs) and TEMPO-oxidized cellulose nanofibers (TOCNFs) with different MWCNTs contents into a dispersion of partially deacetylated α-chitin nanofibers (α-DECHNs) followed with a drying process. The TOCNFs/MWCNTs/α-DECHNs composite wires were prepared by extruding the negatively charged TOCNFs/MWCNTs dispersion into the positively charged α-DECHNs dispersion. The contact of the positively charged α-DECHNs and the negatively charged TOCNFs/MWCNTs triggers the electrostatic interaction (heterocoagulation) resulting in wire-shaped conductive composites. The SEM analysis indicates this conductive composite material has a wire-like shape with a rough but tight surface. The properties of samples were characterized by a zeta potential analyzer (Zetasizer Nano), a four-probe, an electrochemical workstation, a Fourier transform infrared spectroscopy (FTIR), an X-ray diffractometer (XRD), and a thermogravimetric analyzer (TG). Besides, the conductivity and the AC impedance of TOCNFs/MWCNTs/α-DECHNs composite wires with different MWCNTs contents were also analyzed. The conductivity of the composite wire increases from 9.98 × 10 - 6  S∙cm - 1 as the MWCNTs content raises from 3.0 wt% to 14.0 wt%. When the MWCNTs content reaches 14.0 wt%, the prepared composite wire can light up LED at a voltage of 5 V, indicating the great potential of this biomass-based conductive composite in conductive material application.- 3  S∙cm - 1 as the MWCNTs content raises from 3.0 wt% to 14.0 wt%. Biomass can be converted to various types of products in biological and metabolic processes. For an in-depth understanding of biomass conversion, quantitative and qualitative information of products in these conversion processes are essential. Here we introduce analytical techniques including high-performance liquid chromatography (HPLC), gas chromatography (GC), gas chromatography-mass spectrometry (GC-MS), and nuclear magnetic resonance (NMR) for biomass-based products characterization in biological and metabolic processes.

The lignin pyrolysis products generated by biomass combustion make an essential contribution to the formation of secondary organic aerosols (SOAs). The ozone-initiated oxidation of guaiacol, syringol and creosol, major constituents of biomass burning, were investigated theoretically by using the density functional theory (DFT) method at the MPWB1K/6-311+G(3df,2p)//MPWB1K/6-31+G(d,p) level. Six primary addition reaction pathways and further decomposition routes with corresponding thermodynamic values were proposed. The Criegee intermediates can be excited by small molecules, such as NO x , H 2 O in the atmosphere, and would further proceed via self-decomposition or isomerization. The most predominant product for ozonation of guaiacol is the monomethyl muconate (P1). At 295 K and atmospheric pressure, the rate constant is 1.10 × 10 -19 cm 3 molecule -1 s -1 , which is lies a factor of 4 smaller than the previous experimental study. The branching ratios of the six channels are calculated based on corresponding rate coefficient. The present work mainly provides a more comprehensive and detailed theoretical research on the ozonation of methoxyphenol, which aspires to offer novel insights and reference for future experimental and theoretical work and control techniques of SOAs caused by lignin pyrolysis products.

Main Conclusion: Common duckweed Lemna minor was cultivated in 50% D 2 O to produce biomass with 50-60% deuterium incorporation containing cellulose with degree of polymerization close (85%) to that of H 2 O-grown controls. The small aquatic plant duckweed, particularly the genus Lemna, widely used for toxicity testing, has been proposed as a potential source of biomass for conversion into biofuels as well as a platform for production of pharmaceuticals and specialty chemicals. Ability to produce deuterium-substituted duckweed can potentially extend the range of useful products as well as assist process improvement. Cultivation of these plants under deuterating conditions was previously been reported to require addition of kinetin to induce growth and was hampered by anomalies in cellular morphology and protein metabolism. Here, we report the production of biomass with 50-60% deuterium incorporation by long-term photoheterotrophic growth of common duckweed Lemna minor in 50% D 2 O with 0.5% glucose. L. minor grown in 50% D 2 O without addition of kinetin exhibited a lag phase twice that of H 2 O-grown controls, before start of log phase growth at 40% of control rates. Compared to continuous white fluorescent light, growth rates increased fivefold for H 2 O and twofold for 50% D 2 O when plants were illuminated at higher intensity with a metal halide lamp and a diurnal cycle of 12-h light/12-h dark. Deuterium incorporation was determined by a combination of 1 H and 2 H nuclear magnetic resonance (NMR) to be 40-60%. The cellulose from the deuterated plants had an average-number degree of polymerization (DP n ) and polydispersity index (PDI) close to that of H 2 O-grown controls, while Klason lignin content was reduced. The only major gross morphological change noted was root inhibition.

In this work, multivariate data analysis was employed to correlate variables of pretreatment process of lignocellulosic biomass. Principal component analysis and partial least square methods were performed to get the inner-relationship and data interpretation between the crystallinity and other parameters of mechanical refining-assisted sodium hydroxide pretreatment followed by enzymatic saccharification of corn stover. The PCA and PLS models showed that Sodium hydroxide dosage, mechanical refining treatment, lignin removal rate and crystallinity had close inner-related relationship with the efficiency of pretreatment and enzymolysis. Alkaline reaction and mechanical refining treatment had strong influence on the crystallinity. Multivariate data analysis revealed that pretreated corn stover samples with lower crystallinity were more easily hydrolyzed by enzyme and could get more final reducing sugar. Liquor recycle in lignocellulosic biomass fractionation with ethanol-water has been studied. Runs have been carried out in a 6 L tank reactor with liquor recirculation. The liquors obtained in six successive fractioning operations have been analyzed together with the solid phase remnant. Experimental results revealed that the number of re-uses reduces solids recovery (from 52.2 to 42.6%) and cellulose recovery (from 28.1 to 23.3%) with minor or no effect on the hemicelluloses and lignin removal. The more remarkable effect is an increase of the glucose yield (from 76.7 to 95.3% after enzymatic hydrolysis during 72 h). The accumulation of acetic acid in the spent liquors (until 1.3 g/L) seems to be responsible of the higher enzymatic hydrolysis yield, from 76.3 (first use) to 87.7% (fifth re-use). Background: Due to the non-toxicity, abundance and biodegradability, recently more and more attention has been focused on the exploration of hemicellulose as the potential substrate for the production of liquid fuels and other value-added chemicals and materials in different fields. This review aims to summarize the current knowledge on the promising application of nature hemicellulose and its derivative products including its degradation products, its new derivatives and hemicellulosebased medical biodegradable materials in the medical and pharmaceutical field, especially for inmmune regulation, bacteria inhibition, drug release, anti-caries, scaffold materials and anti-tumor., Methods: We searched the related papers about the medical and pharmaceutical application of hemicellulose and its derivative products, and summarized their preparation methods, properties and use effects., Results: Two hundred and twenty-seven papers were included in this review. Forty-seven papers introduced the extraction and application in immune regulation of nature hemicellulose, such as xylan, mannan, xyloglucan (XG) and β-glucan. Seventy-seven papers mentioned the preparation and application of degradation products of hemicellulose for adjusting intestinal function, maintaining blood glucose levels, enhancing the immunity and alleviating human fatigue fields such as xylooligosaccharides, xylitol, xylose, arabinose, etc. The preparation of hemicellulose derivatives were described in thirty-two papers such as hemicellulose esters, hemicellulose ethers and their effects on anticoagulants, adsorption of creatinine, the addition of immune cells and the inhibition of harmful bacteria. Finally, the preparations of hemicellulose-based materials such as hydrogels and membrane for the field of drug release, cell immobilization, cancer therapy and wound dressings were presented using fifty-five papers., Conclusion: The structure of hemicellulose-based products has the significant impact on properties and the use effect for the immunity, and treating various diseases of human. When lignocellulosic biomass was used for acetone-butanol-ethanol (ABE) fermentation, several lignocellulose-derived inhibitors, which are toxic to Clostridium acetobutylicum, were generated during acid hydrolysis process and seriously hindered the industrialization of lignocellulosic butanol. In this study, an engineered strain 824(proABC) with significantly improved tolerance to multiple lignocellulose-derived inhibitors (formic acid and phenolic compounds) was constructed by strengthening the proline biosynthesis. The engineered strain exhibited more effective synthesis ability of proline and scavenging ability of reactive oxygen species (ROS). Consequently, the butanol produced by 824(proABC) was 1-, 2.4- or 3.4-fold higher than that of the wild type strain when using the undetoxified hydrolysate of soybean straw, rice straw or corn straw as the substrate, respectively. Therefore, enhancing the proline biosynthesis can be used as an effective strategy to improve the tolerance of C. acetobutylicum to multiple lignocellulose-derived inhibitors, and 824(proABC) has great potential to produce butanol from undetoxified lignocellulosic hydrolysates., (Copyright © 2018 Elsevier Ltd. The pertinent issue of resources shortage arising from global climate change in the recent years has accentuated the importance of materials that are environmentally friendly. Despite the merits of current material like cellulose as the most abundant natural polysaccharide on earth, the incorporation of lignocellulosic biomass has the potential to value-add the recent development of cellulose-derivatives in drug delivery systems. Lignocellulosic biomass, with a hierarchical structure is comprised of cellulose, hemicellulose and lignin. As an excellent substrate that is renewable, biodegradable, biocompatible and chemically accessible for modified materials, lignocellulosic biomass sets forth a myriad of applications. To date, materials derived from lignocellulosic biomass have been extensively explored for new technological development and applications, such as biomedical, green electronics and energy products. In this review, chemical constituents of lignocellulosic biomass are first discussed before we critically examine the potential alternatives in the field of biomedical application. In addition, the pretreatment methods for extracting cellulose, hemicellulose and lignin from lignocellulosic biomass as well as their biological applications including drug delivery, biosensor, tissue engineering etc. are reviewed. Thermo-chemical pretreatments of biomass typically result in environmental impacts from water use and emission. The degradation byproducts in the resulting sugars can be inhibitory to the activities of enzymes and yeasts. The results of this study showed that combining existing commercial comminution technology can reduce total energy consumption with improved saccharification yield while eliminating chemical use. Impact mill was found to be the most efficient milling for size reduction of forest residual chips from ca. 2 mm to a specific value below 100 µm. The further micronization effectively disrupted the recalcitrance of the woody biomass and produced the highly saccharifiable substrates for downstream processing. In addition, extrusion can be integrated into a clean cellulosic sugar process for further fibrillation in place of the conventional mixing processing. A sequential two-stage pretreatment process comprising alkaline pre-extraction and alkaline hydrogen peroxide pretreatment (AHP) was investigated to convert bamboo carbohydrates into bioethanol. The results showed that mild alkaline pre-extraction using 8% (w/w) sodium hydroxide (NaOH) at 100°C for 180min followed by AHP pretreatment with 4% (w/w) hydrogen peroxide (H 2 O 2 ) was sufficient to generate a substrate that could be efficiently digested with low enzyme loadings. Moreover, alkali pre-extraction enabled the use of lower H 2 O 2 charges in AHP treatment. Two-stage pretreatment followed by enzymatic hydrolysis with only 9FPU/g cellulose led to the recovery of 87% of the original sugars in the raw feedstock. The use of the pentose-hexose fermenting Saccharomyces cerevisiae SR8u strain enabled the utilization of 95.7% sugars in the hydrolysate to reach 4.6%w/v ethanol titer. Chlorine dioxide (ClO 2 ) is a bleaching reagent used in paper industry. Two different types of pretreatment methods were investigated incorporating ClO 2 as a secondary reagent: (a) alkaline followed by ClO 2 treatment; (b) dilute-sulfuric acid followed ClO 2 treatment. In these methods, ClO 2 treatment has shown little effect on delignification. Scheme-a has shown a significant improvement in enzymatic digestibility of glucan far above that treated by ammonia alone. On the contrary, dilute-acid followed by ClO 2 treatment has shown negative effect on the enzymatic hydrolysis. The main factors affecting the enzymatic hydrolysis are the changes of the chemical structure of lignin and its distribution on the biomass surface. ClO 2 treatment significantly increases the carboxylic acid content and reduces phenolic groups of lignin, affecting hydrophobicity of lignin and the H-bond induced association between the enzyme and lignin. Aiming for the complete utilization of bamboo biomass, an integrated process which combines ethanol production with the recovery of silica and lignin was proposed. To reduce chemical charge required for the fractionation of silica and lignin from bamboo and improve the digestibility of the obtained substrate, a sequentially two-stage pretreatment process of autohydrolysis and alkaline extraction was carried out. From the view of enhancing enzymatic hydrolysis and recovery of silica and lignin, a two-stage treatment of autohydrolysis at 180°C for 90min followed by alkaline extraction at 100°C with 6% NaOH (based on pretreated chips) for 120min was optimized. About 93.7% of original silica and 75.7% of original lignin could be recovered from bamboo. Enzymatic hydrolysis and fermentation of carbohydrates showed that an overall sugar yield of 88.6% of original sugar content and an ethanol recovery of 0.467g/g sugar were achieved based on the proposed scheme., (Copyright © 2017 Elsevier Ltd. Pyrolysis was demonstrated to investigate the thermal decomposition characteristics and potential of lignin-rich cotton by-products cotton exocarp (CE) and spent mushroom substrate consisted of cotton by-products (MSC) for biorefineries. The chemical component and structure alteration of CE and MSC was found to affect their thermochemical behaviors. The bio-oil yield from CE was 58.13wt% while the maximum yield from MSC was 45.01% at 600°C. The phenolic compounds obtained from CE and MSC were 33.9% and 39.2%, respectively. The yield of acetic acid from MSC between 400 and 600°C was about 30-38% lower than that from CE, which suggests the high quality of bio-oil was obtained. The effect of fluid dynamic conditions on enzymatic hydrolysis of acid pretreated corn stover (PCS) has been assessed. Runs were performed in stirred tanks at several stirrer speed values, under typical conditions of temperature (50°C), pH (4.8) and solid charge (20% w/w). A complex mixture of cellulases, xylanases and mannanases was employed for PCS saccharification. At low stirring speeds (<150rpm), estimated mass transfer coefficients and rates, when compared to chemical hydrolysis rates, lead to results that clearly show low mass transfer rates, being this phenomenon the controlling step of the overall process rate. However, for stirrer speed from 300rpm upwards, the overall process rate is controlled by hydrolysis reactions. Lignocellulosic materials are among the most promising alternative energy resources that can be utilized to produce cellulosic ethanol. However, the physical and chemical structure of lignocellulosic materials forms strong native recalcitrance and results in relatively low yield of ethanol from raw lignocellulosic materials. An appropriate pretreatment method is required to overcome this recalcitrance. For decades various pretreatment processes have been developed to improve the digestibility of lignocellulosic biomass. Each pretreatment process has a different specificity on altering the physical and chemical structure of lignocellulosic materials. In this paper, the chemical structure of lignocellulosic biomass and factors likely affect the digestibility of lignocellulosic materials are discussed, and then an overview about the most important pretreatment processes available are provided. This study presents high temperature water-only continuous flowthrough pretreatment coupled with nuclear magnetic resonance (NMR) as a promising analytical tool to examine the plant cell wall, to understand its recalcitrance (i.e., cell wall resistance to deconstruction), and to probe the chemistry occurring during batch pretreatment of biomass. (13)C-enriched corn stover stems were pretreated at 170°C for 60min with a hot-water flow rate of 20mL/min to control fractionation of the cell wall. This approach helped elucidate the nature of plant cell wall chemical recalcitrance and biomass pretreatment chemistry by tracking cell wall fragmentation as a function of time. The presence of lignin in lignocellulosic biomass is correlated with its enzymatic digestibility. Their correlation and mechanism have been investigated widely but have not been elucidated clearly. In this study, hydrophilic sulfonated lignin and hydrophobic kraft lignin were introduced into the enzymatic hydrolysis process to investigate their effects on the enzymatic digestibility of different pretreated lignocellulose. The influence of lignin addition on the enzymatic digestibility varied with both introduced lignin type and the pretreatment methods of substrates. Slight enhancement of enzymatic hydrolysis was observed for all substrates by adding kraft lignin. The addition of sulfonated lignin could effectively improve the enzymatic digestibility of green liquor and acidic bisulfite pretreated materials, but had little effect on sulfite-formaldehyde pretreated samples. Various economic and environmental sustainability concerns as well as consumer preference for bio-based products from natural sources have paved the way for the development and expansion of biorefining technologies. These involve the conversion of renewable biomass feedstock to fuels and chemicals using biological systems as alternatives to petroleum-based products. Filamentous fungi possess an expansive portfolio of products including the multifunctional organic acids itaconic, fumaric, and malic acids that have wide-ranging current applications and potentially addressable markets as platform chemicals. However, current bioprocessing technologies for the production of these compounds are mostly based on submerged fermentation, which necessitates physicochemical pretreatment and hydrolysis of lignocellulose biomass to soluble fermentable sugars in liquid media. This review will focus on current research work on fungal production of itaconic, fumaric, and malic acids and perspectives on the potential application of solid-state fungal cultivation techniques for the consolidated hydrolysis and organic acid fermentation of lignocellulosic biomass.

In this work, 28 cholinium ionic liquids (ILs), most of which are good solvents for dissolving lignin, were used for rice straw pretreatment to improve subsequent enzymatic hydrolysis. The anion exerted a significant effect on the pretreatment effectiveness of the IL as well as the subsequent enzymatic hydrolysis efficiency of rice straw residues. The presence of the basic group(s) in the anion significantly enhanced the IL pretreatment effectiveness, while the carboxyl, hydroxyl and aromatic groups had a negative impact on IL delignification. Except for amino acid-based ILs, the delignification abilities of the ILs are linearly and positively correlated with the pKa values of the conjugate acids of the anions. Of the ILs tested, amino acid-based ILs, especially basic amino acid-based ILs, are the most effective pretreatment solvents. Satisfactory reducing sugar yields (81% for glucose and 26% for xylose) were obtained in the enzymatic hydrolysis of rice straw pretreated by cholinium argininate ([Ch][Arg]) under a pretty mild pretreatment severity (60°C, 6 h). Anaerobic ammonium oxidation (anammox) has been proved to be a promising nitrogen removal method for treating ammonium-rich wastewater. However, because of the low-growth rate of anammox bacteria, maintenance of a sufficient amount of anammox biomass in reactor became a key factor in application. Gel immobilization is an efficient method to prevent biomass from being washed out and to promote hyper-concentrated cultures. This study focused on a nitrogen removal process by anammox enrichment culture immobilized in polyvinyl alcohol and sodium alginate (PVA-SA) gel beads. The rapid startup of reactor demonstrated that gel entrapment was supposed to be a highly effective technique for immobilizing anammox bacteria. The anammox bacteria present in the enrichment were identified to be Jettenia-like species (>98%). Moreover, the effect of hydraulic retention time (HRT), pH, and temperature on immobilized anammox processes were investigated. The effect of pH and temperature on the anammox process was evidently weakened in PVA-SA immobilized gel beads, however, the effect of HRT on the anammox reaction was enhanced. Therefore, a stable operated reactor could be obtained in an anaerobic sequencing batch reactor, which proved gel immobilization was an excellent method to maintain the biomass in anammox reactor for application.

Four fractions of one commercial sodium lignosulfonate (SXP) with different molecular weight (MW) and anionic polymers were studied to reduce non-productive adsorption of cellulase on bound lignin in a lignocellulosic substrate. SXP with higher MW had stronger blocking effect on non-productive adsorption of a commercial Trichoderma reesi cellulase cocktail (CTec2) on lignin measured by quartz crystal microgravimetry with dissipation monitoring. Linear anionic aromatic polymers have strong blocking effect, but they would also reduce CTec2 adsorption on cellulose to decrease the enzymatic activity. The copolymer of lignin and polyethylene glycol (AL-PEG1000) has strong enhancement in enzymatic hydrolysis of lignocelluloses, because it not only improves the cellulase activity to cellulose, but also blocks the non-productive cellulase adsorption on lignin. Apart from improving the cellulase activity to cellulose, the enhancements of enzymatic hydrolysis of lignocellulose by adding AL-PEG1000 and SXPs are the result of the decreased cellulase non-productive adsorption on lignin., (Copyright © 2013 Elsevier Ltd. To further investigate the effect of dilute acid pretreatment (DAP) and steam explosion pretreatment (SE) on the change in cellulose accessibility, several techniques were applied including a Simons' stain (SS) technique along with several NMR methods (i.e., NMR cryoporometry, (1)H spin-lattice (T1) and (1)H spin-spin (T2) relaxometry, and diffusometry). These methods were utilized to probe biomass porosity and thus assess cellulose accessibility on untreated and pretreated Populus. In general, these techniques indicate that pretreated Populus has larger pore size distributions and specific surface area (SSA) when compared to an untreated sample. The SS method revealed that DAP is more effective than SE in terms of the SSA increase, and that DAP increases SSA as a function of pretreatment severity. Chemical and surface characteristics of sulfite-pretreated royal palm sheath (RPS) fibers and parenchyma cells were investigated in order to study cell-type-dependent biomass hydrolysis by cellulase. Size, chemical composition, cellulose crystallinity and the exposure of cellulose microfibrils in pretreated RPS biomass affected the enzymatic accessibility and digestibility of different cell-type substrates.

2-Methyltetrahydrofuran (MeTHF), a biomass-derived compound, is a promising medium for biocatalysis and organometallic reactions. The regioselective acylation of 8-chloroadenosine (8-Cl-Ado) and its analogs was carried out in MeTHF with immobilized Penicillium expansum lipase. The lipase displayed more than twofold higher catalytic activity and much better thermostability in MeTHF than in other organic solvents and co-solvent systems. The optimum reaction medium, enzyme dosage, molar ratio of viny ester to nucleoside and reaction temperature for the enzymatic acylation of 8-Cl-Ado were MeTHF, 25 U/mL, 7.5 and 35 °C, respectively, under which the desirable 5'-O-undecylenoyl-8-Cl-Ado was obtained with a yield of 95% and a regioselectivity of >99% in 3 h. In addition, the lipase catalyzed regioselective undecylenoylation of other purine nucleosides, producing 5'-undecylenic acid esters with moderate to high yields (63-94%) and excellent 5'-regioselectivities (94->99%). Woody biomass is renewable only if sustainable production is imposed. An optimum and sustainable biomass stand production rate is found to be one with the incremental growth rate at harvest equal to the average overall growth rate. Utilization of woody biomass leads to a sustainable economy. Woody biomass is comprised of at least four components: extractives, hemicellulose, lignin and cellulose. While extractives and hemicellulose are least resistant to chemical and thermal degradation, cellulose is most resistant to chemical, thermal, and biological attack. The difference or heterogeneity in reactivity leads to the recalcitrance of woody biomass at conversion. A selection of processes is presented together as a biorefinery based on incremental sequential deconstruction, fractionation/conversion of woody biomass to achieve efficient separation of major components. A preference is given to a biorefinery absent of pretreatment and detoxification process that produce waste byproducts. While numerous biorefinery approaches are known, a focused review on the integrated studies of water-based biorefinery processes is presented. Hot-water extraction is the first process step to extract value from woody biomass while improving the quality of the remaining solid material. This first step removes extractives and hemicellulose fractions from woody biomass. While extractives and hemicellulose are largely removed in the extraction liquor, cellulose and lignin largely remain in the residual woody structure. Xylo-oligomers, aromatics and acetic acid in the hardwood extract are the major components having the greatest potential value for development. Higher temperature and longer residence time lead to higher mass removal. While high temperature (>200°C) can lead to nearly total dissolution, the amount of sugars present in the extraction liquor decreases rapidly with temperature. Dilute acid hydrolysis of concentrated wood extracts renders the wood extract with monomeric sugars. At higher acid concentration and higher temperature the hydrolysis produced more xylose monomers in a comparatively shorter period of reaction time. Xylose is the most abundant monomeric sugar in the hydrolysate. The other comparatively small amounts of monomeric sugars include arabinose, glucose, rhamnose, mannose and galactose. Acetic acid, formic acid, furfural, HMF and other byproducts are inevitably generated during the acid hydrolysis process. Short reaction time is preferred for the hydrolysis of hot-water wood extracts. Acid hydrolysis presents a perfect opportunity for the removal or separation of aromatic materials from the wood extract/hydrolysate. The hot-water wood extract hydrolysate, after solid-removal, can be purified by Nano-membrane filtration to yield a fermentable sugar stream. A commercially available deuterated kale sample was analyzed for deuterium incorporation by ionic liquid solution (2)H and (1)H nuclear magnetic resonance (NMR). This protocol was found to effectively measure the percent deuterium incorporation at 33%, comparable to the 31% value determined by combustion. The solution NMR technique also suggested by a qualitative analysis that deuterium is preferentially incorporated into the carbohydrate components of the kale sample.

This study presents a novel, efficient and environmentally friendly process for the cooking of corn stalk that uses active oxygen (O2 and H2O2) and a recoverable solid alkali (MgO). The structural changes on the surface of corn stalk before and after cooking were characterized by attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS) and atomic force microscopy (AFM) techniques. The results showed that lignin and extractives were effectively removed, especially those on the surface of corn stalk. Additionally, the changes included becoming fibrillar, the exposure of cellulose and hemi-cellulose and the pitting corrosion on the surface, etc. The results also showed that the removal reaction is from outside to inside, but the main reaction is possibly on the surface. Buddleja davidii is a unique biomass that has many attractive agroenergy features, especially its wide range of growth habitat. The anatomical characteristics of B. davidii were investigated before and after ethanol organosolv pretreatment (one of the leading pretreatment technologies) in order to further understand the alterations that occur to the cellular structure of the biomass which can then be correlated with its enzymatic digestibility. Results showed that the ethanol organosolv pretreatment of B. davidii selectively removes lignin from the middle lamella (ML), which does not significantly disrupt the crystalline structure of cellulose. The removal of ML lignin is a major factor in enhancing enzymatic cellulose-to-glucose hydrolysis. The pretreatment also causes cell deformation, resulting in cracks and breaks in the cell wall. These observations, together with characterization analysis of the cell wall polymer material, lend support to the hypothesis that the physical distribution of lignin in the biomass matrix is an important structural feature affecting biomass enzymatic digestibility., (© 2010 Wiley Periodicals, Inc.)

The conversion of biomass to chemicals and energy is imperative to sustaining our way of life as known to us today. Fossil chemical and energy sources are traditionally regarded as wastes from a distant past. Petroleum, natural gas, and coal are not being regenerated in a sustainable manner. However, biomass sources such as algae, grasses, bushes and forests are continuously being replenished. Woody biomass represents the most abundant and available biomass source. Woody biomass is a reliably sustainable source of chemicals and energy that could be replenished at a rate consistent with our needs. The biorefinery is a concept describing the collection of processes used to convert biomass to chemicals and energy. Woody biomass presents more challenges than cereal grains for conversion to platform chemicals due to its stereochemical structures. Woody biomass can be thought of as comprised of at least four components: extractives, hemicellulose, lignin and cellulose. Each of these four components has a different degree of resistance to chemical, thermal and biological degradation. The biorefinery concept proposed at ESF (State University of New York - College of Environmental Science and Forestry) aims at incremental sequential deconstruction, fractionation/conversion of woody biomass to achieve efficient separation of major components. The emphasis of this work is on the kinetics of hot-water extraction, filling the gap in the fundamental understanding, linking engineering developments, and completing the first step in the biorefinery processes. This first step removes extractives and hemicellulose fractions from woody biomass. While extractives and hemicellulose are largely removed in the extraction liquor, cellulose and lignin largely remain in the residual woody structure. Xylo-oligomers and acetic acid in the extract are the major components having the greatest potential value for development. Extraction/hydrolysis involves at least 16 general reactions that could be divided into four categories: adsorption of proton onto woody biomass, hydrolysis reactions on the woody biomass surface, dissolution of soluble substances into the extraction liquor, and hydrolysis and dehydration decomposition in the extraction liquor. The extraction/hydrolysis rates are significantly simplified when the reactivity of all the intermonomer bonds are regarded as identical within each macromolecule, and the overall reactivity are identical for all the extractable macromolecules on the surface. A pseudo-first order extraction rate expression has been derived based on concentrations in monomer units. The reaction rate constant is however lower at the beginning of the extraction than that towards the end of the extraction. Furthermore, the H-factor and/or severity factor can be applied to lump the effects of temperature and residence time on the extraction process, at least for short times. The conversion of biomass into chemicals and energy is essential in order to sustain our present way of life. Fossil fuels are currently the predominant energy source, but fossil deposits are limited and not renewable. Biomass is a reliable potential source of materials, chemicals and energy that can be replenished to keep pace with our needs. A biorefinery is a concept for the collection of processes used to convert biomass into materials, chemicals and energy. The biorefinery is a "catch and release" method for using carbon that is beneficial to both the environment and the economy. In this study, we discuss three elements of a wood-based biorefinery, as proposed by the SUNY College of Environmental Science and Forestry (ESF): hot-water extraction, hydrolysis, and membrane separation/concentration. Hemicelluloses are the most easily separable main component of woody biomass and thus form the bulk of the extracts obtained by hot-water extraction of woody biomass. Hot-water extraction is an important step in the processes of woody biomass and product generation, replacing alternative costly pre-treatment methods. The hydrolysis of hemicelluloses produces 5-carbon sugars (mainly xylose), 6-carbon sugars (mainly glucose and mannose), and acetic acid. The use of nano-filtration membranes is an efficient technology that can be employed to fractionate hot-water extracts and wood hydrolysate. The residual solid mass after hot-water extraction has a higher energy content and contains fewer easily degradable components. This allows for more efficient subsequent processing to convert cellulose and lignin into conventional products.
