In recent decades, the global intensification of marine heatwaves has impacted several ecosystems and species, including the endemic Mediterranean seagrass Posidonia oceanica. However, the scarcity of research in Eastern Mediterranean meadows, where historical and present thermal conditions differ from those of the Western Mediterranean, hampers our ability to draw comprehensive conclusions regarding the species' response to elevated sea temperatures. Here, we studied flowering patterns of P. oceanica meadows (3–15 m depth) of the Greek seas and assessed their potential association with marine heatwaves, while also examining the effects on plant growth associated with the transition from vegetative to sexual reproduction. To do so, we reconstructed flowering patterns from 2007 to 2021 across 58 meadows using lepidochronology and estimated summer marine heatwave metrics and categories using satellite sea surface temperature. The results revealed that flowering, while sporadic in most meadows (mean flowering frequency: 0.15), it was triggered by strong (Category II) summer marine heatwaves and a cumulative intensity exceeding 30°C days. Nevertheless, flowering density was low (mean flowering density: 11 inflorescences m−2) and not fuelled by frequency, intensity, or duration of marine heatwaves. Despite the potential long‐term benefits of sexual reproduction, annual rhizome production of flowering shoots decreased 1.5‐fold for at least 2 yr postflowering. This study uncovers the susceptibility of Eastern Mediterranean P. oceanica meadows, akin to their western counterparts, to marine heatwaves induced stress and its effects on reproductive strategies, raising concerns about the prospective plant growth and reproductive fitness to the escalating trend in marine heatwaves. In crocodiles, teratology alterations have been described in captive animals (pets, zoos, farms) such as Crocodylus niloticus and Gavialis gangeticus. The present study aimed to characterize congenital malformations of C. acutus from a farm in Lomas de Matunilla, Ballestas, Bolívar, Colombia. The analyzed eggs presented macroscopic malformations, with 42 different types of anomalies observed. Limb and tail malformations (29%) were the most common changes observed. Seagrass meadows provide important ecosystem services including carbon sequestration, coastal protection from erosion, and sustained biodiversity and fisheries thereby improving the wellbeing and livelihoods of coastal communities. The erosion of millenary deposits of intertwined roots and rhizomes of Posidonia results in the formation of three-dimensional structures named escarpments that constitute a biogenic reef habitat. However, the natural history of seagrass escarpments including their formation processes and their role as habitat for reef fauna and flora remains poorly understood. This research located and characterized Posidonia oceanica escarpments in Menorca (Balearic Islands) and compared structural complexity and fish assemblages among seagrass escarpments, seagrass meadows, rocky substrates and bare sand with emphasis on its role as habitat and shelter for typical rocky fish. Fish abundance and biomass were similar between seagrass escarpments and rocky substrates (P > 0.05), but significantly lower in seagrass meadows (P < 0.001). The large number of caves found along seagrass escarpments provide shelter to fish, including species only associated to rocky substrates. Seagrass meadows form a rather homogenous habitat within their canopy, but the presence of seagrass escarpments enhances habitat heterogeneity and structural complexity along with fish abundance and biomass at the seascape level. The ability to accurately measure organic carbon (OC) in marine sediments or soils is overall taken for granted in scientific communities, yet this seemingly mundane task remains a methodological challenge when the soil matrix contains calcium carbonate (CaCO3), creating inaccuracies in Blue Carbon estimates. Here, we compared five common methods combining acidification, combustion, and wet oxidation pre‐treatments for determination of OC in sediments and soils containing CaCO3 based on the analyses of artificial soil mixtures made of different OC and CaCO3 contents, and multiple soils from Australian seagrass cores. The results obtained showed that methods involving acidification pre‐treatment entailed −17 ± 0.2% (mean ± SE) underestimation of OC content (ranging from −8% to −26%), whereas the combustion‐based method was accurate for samples with high CaCO3 content but entailed 32–47% overestimation in samples with low CaCO3 content. The Heanes method (wet oxidation method) showed <5% deviation from the known OC content, but this method is not suitable for soil samples containing reduced iron, sulfur and potentially manganese compounds. The differences observed among methods have significant impacts on local, regional, and global Blue Carbon storage calculations. Summary: The response of Posidonia oceanica meadows to global warming of the Eastern Mediterranean Sea, where the increase in sea surface temperature (SST) is particularly severe, is poorly investigated.Here, we reconstructed the long‐term P. oceanica production in 60 meadows along the Greek Seas over two decades (1997–2018), using lepidochronology. We determined the effect of warming on production by reconstructing the annual and maximum (i.e. August) SST, considering the role of other production drivers related to water quality (i.e. Chla, suspended particulate matter, Secchi depth).Grand mean (±SE) production across all sites and the study period was 48 ± 1.1 mg DW per shoot yr−1. Production over the last two decades followed a trajectory of decrease, which was related to the concurrent increase in annual SST and SSTaug. Annual SST > 20°C and SSTaug > 26.5°C was related to production decline (GAMM, P < 0.05), while the rest of the tested factors did not help explain the production pattern.Our results indicate a persistent and increasing threat for Eastern Mediterranean meadows, drawing attention to management authorities, highlighting the necessity of reducing local impacts to enhance the resilience of seagrass meadows to global change threats. Although seagrass meadows form a relatively homogenous habitat, escarpments, which form three‐dimensional structures and originate from the erosion of seagrass peat, can provide important habitat for reef fishes. Here, we compare fish assemblages and habitat structural complexity among seagrass Posidonia australis escarpments and canopies, as well as limestone reef habitats, to understand the role of seagrass escarpments as reef fish habitat in Shark Bay, Western Australia. The total number of fish species, fish biomass, and top predator biomass were significantly higher in seagrass escarpments and reef habitats than in seagrass canopies due to lower habitat structural complexity and thus becoming suitable habitats for predators and prey in the latter. Both seagrass escarpment and reef habitats host similar assemblages of top predators and carnivorous fishes, such as Epinephelus coioides, Microcanthus strigatus, and Choerodon schoenleinii, that were absent in seagrass canopies. Seagrass escarpments provide an alternative habitat for reef fishes comparable to rocky reefs, which are limited in Shark Bay. Caves and ledges within the escarpments support 13.4 Mg of fish and 3.6 Mg of top predator species of commercial interest within the Shark Bay World Heritage Site. Additional research is needed to further understand the ecological importance of seagrass escarpments in enhancing fish biomass and biodiversity, as reproduction grounds or refuge from predators, and to investigate the role of meadow edges in ecosystem function. Seagrass meadows composed of larger species are assumed to store larger sediment organic carbon (Corg) stocks, storing more Corg in their tissues and larger leaves promoting greater burial of seagrass and non‐seagrass Corg. However, the influence of species composition on sediment Corg stocks remains poorly understood mainly from challenges in isolating it from confounding factors. We assessed Corg stocks in seagrass biomass and sediment of four species compositions in a tropical Caribbean meadow. We hypothesized that larger species would lead to higher sediment Corg stocks, within a limited geomorphic setting and time frame. Seagrass biomass and surficial and sediment profiles were collected to measure seagrass morphometrics, δ13C and δ15N, dry bulk density, Corg and inorganic carbon (Cinorg) stocks, and grain size. Seagrass biomass Corg stocks ranged from 0.04 to 3.7 Mg ha−1, with higher biomass Corg stocks in compositions with larger species. Surficial sediment Corg and Cinorg stocks (to 1.5 cm) averaged 2.6 ± 0.6 Mg Corg ha−1 and 68.8 ± 14.6 Mg Cinorg ha−1, respectively, and did not vary among species compositions. Isotopic analyses revealed a ~ 50% contribution of seagrasses to surficial sediment Corg in compositions with larger species, compared to a contribution of ~ 35% for those of smaller species. Abstract Background The adoptive transfer of autologous tumor reactive lymphocytes can mediate significant tumor regression in some patients with refractory metastatic cancer. However, a significant obstacle for this promising therapy has been the availability of highly efficient methods to rapidly isolate and expand a variety of potentially rare tumor reactive lymphocytes from the natural repertoire of cancer patients. Methods We developed a novel in vitro T cell cloning methodology using high throughput quantitative RT-PCR (qPCR assay) as a rapid functional screen to detect and facilitate the limiting dilution cloning of a variety of low frequency T cells from bulk PBMC. In preclinical studies, this strategy was applied to the isolation and expansion of gp100 specific CD8+ T cell clones from the peripheral blood of melanoma patients. Results In optimization studies, the qPCR assay could detect the reactivity of 1 antigen specific T cell in 100,000 background cells. When applied to short term sensitized PBMC microcultures, this assay could detect T cell reactivity against a variety of known melanoma tumor epitopes. This screening was combined with early limiting dilution cloning to rapidly isolate gp100154–162 reactive CD8+ T cell clones. These clones were highly avid against peptide pulsed targets and melanoma tumor lines. They had an effector memory phenotype and showed significant proliferative capacity to reach cell numbers appropriate for adoptive transfer trials (~1010 cells). Conclusion This report describes a novel high efficiency strategy to clone tumor reactive T cells from peripheral blood for use in adoptive immunotherapy.

Vegetated coastal ecosystems, including tidal marshes, mangroves and seagrass meadows, are being increasingly assessed in terms of their potential for carbon dioxide sequestration worldwide. However, there is a paucity of studies that have effectively estimated the accumulation rates of sediment organic carbon (Corg), also termed blue carbon, beyond the mere quantification of Corg stocks. Here, we discuss the use of the 210Pb dating technique to determine the rate of Corg accumulation in these habitats. We review the most widely used 210Pb dating models to assess their limitations in these ecosystems, often composed of heterogeneous sediments with varying inputs of organic material, that are disturbed by natural and anthropogenic processes resulting in sediment mixing and changes in sedimentation rates or erosion. Through a range of simulations, we consider the most relevant processes that impact the 210Pb records in vegetated coastal ecosystems and evaluate how anomalies in 210Pb specific activity profiles affect sediment and Corg accumulation rates. Our results show that the discrepancy in sediment and derived Corg accumulation rates between anomalous and ideal 210Pb profiles is within 20% if the process causing such anomalies is well understood. While these discrepancies might be acceptable for the determination of mean sediment and Corg accumulation rates over the last century, they may not always provide a reliable geochronology or historical reconstruction. Reliable estimates of Corg accumulation rates might be difficult at sites with slow sedimentation, intense mixing and/or that are affected by multiple sedimentary processes. Additional tracers or geochemical, ecological or historical data need to be used to validate the 210Pbderived results. Introduction Sclerosing Encapsulating Peritonitis (SEP) is a rare condition with an incidence of up to 3% and a mortality of up to 51% among peritoneal dialysis (PD) patients (Brown et al., Korte et al. and Kawanishi et al.). In the last ten years, the incidence of SEP in kidney transplant recipients has increased (Nakamoto, de Sousa et al. and Korte et al.). Presentation of case A 31-year old male with a 15 years history of PD and later kidney retransplantation was admitted to the emergency service after experiencing several weeks of diffuse abdominal pain which had escalated to include vomiting and diarrhea during the 24 h previous to admission. The patient underwent an exploratory laparotomy where severe peritoneal thickening was found, in addition to signs of chronic inflammation and blocked intestinal loops. Histopathologic findings were suggestive of sclerosing peritonitis. After two months of treatment in hospital, the patient presented an obstructed intestine, with a rigid and thickened peritoneum compromising all the intestinal loops. Discussion Despite being rare, SEP, represents a significant complication due to its high mortality and recurrence. It is insidious in its early stages and culminates in an intestinal obstruction (Fieren). Risk factors for its development in kidney transplant recipients include a history of prolonged treatment with PD and the use of calcineurin inhibitors as an immunosuppressive treatment (Korte et al.). Conclusion Given the increase in the incidence of SEP in kidney transplant recipients, the clinician should be alert to the presence of this complication. A greater number of multi-centre studies are required to identify the risk factors for SEP that are inherent in renal transplant recipients. [ABSTRACT FROM AUTHOR]

Objective: Unusual setting of medical care Background: En-bloc transplantation is a surgical procedure in which multiple organs are transplanted simultaneously. It has some similarities with multi-organ transplantation but offers certain advantages. This report highlights the experience of our interdisciplinary group regarding the treatment and follow-up of patients who received en-bloc transplantation, with the aim of encouraging the development of this surgical technique. Case Report: The first case is a 38-year-old patient with type 1 diabetes mellitus, liver cirrhosis, and chronic kidney failure who received an en-bloc transplant of the liver, pancreas, and kidney with no intraoperative complications. He had a prolonged hospital stay due to anemia and systemic inflammatory response syndrome, which were resolved successfully. At follow-up, he had no requirement for insulin or for dialysis, or for new interventions. The second case describes a 48-year-old patient with type 2 diabetes mellitus, renal failure, and liver cirrhosis who received an en-bloc transplant of the liver, pancreas, and kidney with no complications. During the postoperative period, the patient suffered a possible episode of acute tubular necrosis, which evolved towards improvement, with a tendency to normal metabolic and renal functioning, with no additional events. The patient is currently in follow-up and is insulin-independent. Conclusions: En-bloc transplantation is a safe procedure, which is technically simple and which achieves excellent results. This procedure is indicated in patients with end-stage renal disease, cirrhosis, and diabetes mellitus that is difficult to control. [ABSTRACT FROM AUTHOR]

Introduction Liver angiosarcoma is a very uncommon tumour of mesenchymal origin, representing between 0.1–2% of all primary tumours of the liver, affecting mainly men in their sixth or seventh decade of life, with a high mortality in the first years (Chaudhary et al., 2015). Literature reports of its surgical treatment vary from a total or partial hepatectomy with or without liver transplant. Presentation of case A 37 year old male, with a 7 year history of a fatty liver, was found to have a 12 cm diameter tumour in a cirrhotic liver, during an abdominal Computed Tomography (CT) scan. Patient was asymptomatic with negative tumour markers, yet tumour liver biopsy revealed a Liver Angiosarcoma with positive immunohistochemistry for neoplastic cells CD31 and CD34. Patient was deemed candidate for a partial hepatectomy of the affected liver segments which was done without complications and no evidence of other tumour lesions was found during surgery. Patient continued oncologic management with ongoing chemotherapy. Discusion Liver Angiosarcoma, although rare, persists with a high mortality due to its aggressive nature. Never the less liver transplantation, although proven to be an effective treatment for many pathologies that culminate in liver failure, fails to improve patients’ survival and prognosis, when compared to partial hepatectomy as surgical management to for liver Angiosarcoma, Conclusion Partial hepatectomy as surgical management, followed by adjuvant therapy, for Liver Angiosarcoma continues to prove favourable results and prognosis compared to Liver Transplantation. Introduction One of the frequent complications suffered by patients with chronic renal failure is the lack of vascular access due to venous thrombosis. This means that the transplant surgeon must have a detailed knowledge of the intra-abdominal venous system, and other alternative surgeries, at the time of performing the renal graft implant, in order to ensure a good venous drainage. Presentation of the case This article provides a case report regarding a patient with no vascular access and with surgical difficulties at the time of the kidney transplant, in whom a renal-portal venous drainage was performed with very good outcome. Discusion Renal-portal venous drainage is a way to performe kidney transplant with good outcome. In Fundación Valle del Lili we have overcome the lack of vascular access in patients that need a renal transplant by new surgical technics that improve the patients quality of life and survival. Conclusion We can conclude that new surgical alternatives exist for those patients with chronic renal failure that have no vascular access. The emerging field of blue carbon science is seeking cost-effective ways to estimate the organic carbon content of soils that are bound by coastal vegetated ecosystems. Organic carbon (Corg) content in terrestrial soils and marine sediments has been correlated with mud content (i.e., silt and clay, particle sizes < 63 µm), however, empirical tests of this theory are lacking for coastal vegetated ecosystems. Here, we compiled data (n D1345) on the relationship between Corg and mud contents in seagrass ecosystems (79 cores) and adjacent bare sediments (21 cores) to address whether mud can be used to predict soil Corg content. We also combined these data with the δ13C signatures of the soil Corg to understand the sources of Corg stores. The results showed that mud is positively correlated with soil Corg content only when the contribution of seagrass-derived Corg to the sedimentary Corg pool is relatively low, such as in small and fast-growing meadows of the genera Zostera, Halodule and Halophila, and in bare sediments adjacent to seagrass ecosystems. In large and long-living seagrass meadows of the genera Posidonia and Amphibolis there was a lack of, or poor relationship between mud and soil Corg content, related to a higher contribution of seagrass-derived Corg to the sedimentary Corg pool in these meadows. The relatively high soil Corg contents with relatively low mud contents (e.g., mud-Corg saturation) in bare sediments and Zostera, Halodule and Halophila meadows was related to significant allochthonous inputs of terrestrial organic matter, while higher contribution of seagrass detritus in Amphibolis and Posidonia meadows disrupted the correlation expected between soil Corg and mud contents. This study shows that mud is not a universal proxy for blue carbon content in seagrass ecosystems, and therefore should not be applied generally across all seagrass habitats. Mud content can only be used as a proxy to estimate soil Corg content for scaling up purposes when opportunistic and/or low biomass seagrass species (i.e., Zostera, Halodule and Halophila) are present (explaining 34 to 91% of variability), and in bare sediments (explaining 78% of the variability). Biotic and abiotic factors influence the accumulation of organic carbon (Corg) in seagrass ecosystems. We surveyed Posidonia sinuosa meadows growing in different water depths to assess the variability in the sources, stocks and accumulation rates of Corg. We show that over the last 500 years, P. sinuosa meadows closer to the upper limit of distribution (at 2-4m depth) accumulated 3- to 4-fold higher Corg stocks (averaging 6.3 kg Corg m-2) at 3- to 4-fold higher rates (12.8 g Corg m-2 yr-1) compared to meadows closer to the deep limits of distribution (at 6-8m depth; 1.8 kg Corg m-2 and 3.6 g Corg m-2 yr-1). In shallower meadows, Corg stocks were mostly derived from seagrass detritus (88% in average) compared to meadows closer to the deep limit of distribution (45% on average). In addition, soil accumulation rates and fine-grained sediment content (< 0.125 mm) in shallower meadows (2.0 mm yr-1 and 9 %, respectively) were approximately 2-fold higher than in deeper meadows (1.2 mm yr-1 and 5 %, respectively). The Corg stocks and accumulation rates accumulated over the last 500 years in bare sediments (0.6 kg Corg m-2 and 1.2 g Corg m-2 yr-1) were 3- to 11-fold lower than in P. sinuosa meadows, while fine-grained sediment content (1 %) and seagrass detritus contribution to the Corg pool (20 %) were 8- and 3-fold lower than in Posidonia meadows, respectively. The patterns found support the hypothesis that Corg storage in seagrass soils is influenced by interactions of biological (e.g., meadow productivity, cover and density), chemical (e.g., recalcitrance of Corg stocks) and physical (e.g., hydrodynamic energy and soil accumulation rates) factors within the meadow. Primary splenic angiosarcoma is an extremely rare and aggressive neoplasm of the vasculature. Uniformly, primary splenic angiosarcoma is a fatal disease despite early diagnosis and treatment. Only patients with localized disease amenable to surgical resection achieve long-term, disease-free survival. We present a review of the literature and report a case of a 3-year-old girl with metastatic primary splenic angiosarcoma who was offered aggressive surgical and medical treatment with curative intent despite her advanced presentation. Mesenchymal hamartoma is the second most frequent benign liver tumor in young children and usually occurs in the first two years of life. Currently, the gold standard for treatment is a complete surgical resection. We report a 17-month old patient with a giant mesenchymal hamartoma who, due to the impossibility of resection and the risk of malignant transformation, received liver transplantation from a living donor with a satisfactory outcome in the first year post-transplant. In this report, we show that liver transplantation is a viable therapeutic alternative for this pathology, in patients who have no possibility of receiving another type of treatment due to their clinical characteristics. The recent focus on carbon trading has intensified interest in ‘Blue Carbon’–carbon sequestered by coastal vegetated ecosystems, particularly seagrasses. Most information on seagrass carbon storage is derived from studies of a single species, Posidonia oceanica, from the Mediterranean Sea. We surveyed 17 Australian seagrass habitats to assess the variability in their sedimentary organic carbon (Corg) stocks. The habitats encompassed 10 species, in mono-specific or mixed meadows, depositional to exposed habitats and temperate to tropical habitats. There was an 18-fold difference in the Corg stock (1.09–20.14 mg Corg cm−3 for a temperate Posidonia sinuosa and a temperate, estuarine P. australis meadow, respectively). Integrated over the top 25 cm of sediment, this equated to an areal stock of 262–4833 g Corg m−2. For some species, there was an effect of water depth on the Corg stocks, with greater stocks in deeper sites; no differences were found among sub-tidal and inter-tidal habitats. The estimated carbon storage in Australian seagrass ecosystems, taking into account inter-habitat variability, was 155 Mt. At a 2014–15 fixed carbon price of A$25.40 t−1 and an estimated market price of $35 t−1 in 2020, the Corg stock in the top 25 cm of seagrass habitats has a potential value of $AUD 3.9–5.4 bill. The estimates of annual Corg accumulation by Australian seagrasses ranged from 0.093 to 6.15 Mt, with a most probable estimate of 0.93 Mt y−1 (10.1 t. km−2 y−1). These estimates, while large, were one-third of those that would be calculated if inter-habitat variability in carbon stocks were not taken into account. We conclude that there is an urgent need for more information on the variability in seagrass carbon stock and accumulation rates, and the factors driving this variability, in order to improve global estimates of seagrass Blue Carbon storage. Here, we present Lisen&Curate, a curation system that facilitates linking sentences or part of sentences (both considered sources) in articles with their corresponding curated objects, so that rich additional information of these objects is easily available to users. These sources are going to be offered both within RegulonDB and a new database, L-Regulon. To show the relevance of our work, two senior curators performed a curation of 31 articles on the regulation of transcription initiation of E. coli using Lisen&Curate. As a result, 194 objects were curated and 781 sources were recorded. We also found that these sources are useful to develop automatic approaches to detect objects in articles by observing word frequency patterns and by carrying out an open information extraction task. Sources may help to elaborate a controlled vocabulary of experimental methods. Finally, we discuss our ecosystem of interconnected applications, RegulonDB, L-Regulon, and Lisen&Curate, to facilitate the access to knowledge on regulation of transcription initiation in bacteria.
