Paper spray mass spectrometry (PS-MS) has evolved into a promising tool for monitoring reactions in thin films and microdroplets, known as reactive PS, alongside its established role in ambient and direct ionization. This study addresses the need for rapid, cost-effective methods to improve analyte identification in biofluids by leveraging reactive PS-MS in clinical chemistry environments. The technique has proven effective in derivatizing target analytes, altering hydrophobicity to enhance elution and ionization efficiency, and refining detection through thin-film reactions on paper, significantly expediting reaction rates by using amino acids (AAs) as model analytes. These molecules are prone to interacting with substrates like paper, impeding elution and detection. Additionally, highly abundant species in biofluids, such as lipids, often suppress AA ionization. This study employs the Schiff base (SB) reaction utilizing aromatic aldehydes for AA derivatization to optimize reaction conditions time, temperature, and catalyst presence and dramatically increasing the conversion ratio (CR) of formed SB. For instance, using leucine as a model AA, the CR surged from 57% at room temperature to 89% at 70 °C, with added pyridine during and after 7.5 min, displaying a 43% CR compared to the bulk reaction. Evaluation of various aromatic aldehydes as derivatization agents highlighted the importance of specific oxygen substituents for achieving higher conversion rates. Furthermore, diverse derivatization agents unveiled unique fragmentation pathways, aiding in-depth annotation of the target analyte. Successfully applied to quantify AAs in human and rat plasma, this reactive PS-MS approach showcases promising potential in efficiently detecting conventionally challenging compounds in PS-MS analysis.

Simultaneous detection of multiple amino acids (AAs) instead of individual AAs is inherently worthwhile for improving diagnostic accuracy in clinical applications. Here, a facile and reliable colorimetric microfluidic paper-based analytical device (μPAD) using carbon dots doped with transition metals (Cr 3+ , Mn 2+ , Fe 3+ , Co 2+ , Ni 2+ , Cu 2+ , and Zn 2+ ) has been provided to detect and discriminate 20 natural amino acids. To make the colourless metal-doped carbon dots suitable for colorimetric assays, they were mixed with ninhydrin to form a charge transfer complex. This optical tongue system, which was constructed by dropping mixtures of ninhydrin with a series of metal-doped carbon dots on a paper substrate in an array format, represented obvious but different colorimetric signatures for every examined amino acid. Since bovine serum albumin was used as a chiral selector reagent for synthesizing the CDs, the sensor device represented excellent selectivity to identify enantiomeric species of AAs. This is the first optical array device that can simultaneously discriminate AAs and several of their enantiomers. We employed various statistical and chemometric methods to analyze the digital data library collected by Image J software, including principal component analysis (PCA), linear discriminant analysis (LDA), and hierarchical cluster analysis (HCA). Twenty AAs could be well distinguished at various concentrations (10.00, 5.00, 2.50, and 1.25 mM). The colorimetric patterns were highly repeatable and were characteristic of individual AAs. Besides qualitative analysis, the designed μPAD-based optical tongue represented quantitative analysis ability, e.g. , for lysine in the concentration ranges of 0.005-20.0 mM with a detection limit of 1.0 × 10 -6 M and for arginine in the concentration range of 0.12-20.00 mM with a detection limit of 80.0 × 10 -6 M. In addition, the binary, ternary, and quaternary mixtures of AAs could also be well recognized with this sensor.

This work describes the development of a system that combines a derivatization protocol based on the Katritzky reaction with paper spray ionization mass spectrometry (PSI-MS) for the analysis of amino acid neurotransmitters in mouse brain tissues. The system is relatively simple, consisting of spraying the derivatization solution onto a mouse brain section mounted on a glass slide, applying a small volume of solvent to moisten the sample, pressing a triangular paper onto the sample surface to transfer the sample constituents to the paper surface, and using the paper as a substrate for PSI-MS analysis. The Katritzky reaction facilitated the ionization of the amino acids by reacting a pyrylium salt with the amino group of the analytes, forming very stable pyridinium cations, which greatly increased the sensitivity of the PSI-MS analysis. Most of the intensities of the amino acids modified by the Katritzky reaction were more than 10 times greater than the nonderivatized ones. The system was applied for the analysis of brain sections obtained from mice with Parkinson's disease, and the amino acids gamma-aminobutyric acid (GABA) and glycine (Gly), two compounds very well-known in studies of Parkinson's disease, were readily detected. The results suggest that the Katritzky reaction combined with PSI-MS might offer a significant advance in the knowledge on protocols that improve the sensitivity of detection of crucial biological compounds.

Amino acids are closely related to human health, and their rapid determination is important for the rapid diagnosis, timely treatment, and assessment of serious diseases. In this work, we propose a novel paper-based sample-processing device combined with isotope-dilution MS for the fast analysis of 11 amino acids from blood samples. By using an isoelectric focusing electrokinetic separation method, without the aid of carrier ampholytes and the addition of inhibitors, this approach uses only the characteristic of the isoelectric point of the target amino acids to achieve separation and purification from other coexisting components in the medium; it can meet the requirements for mass spectrometry detection. Driven by a DC voltage, a stable and sharp pH gradient (pH 3-10.5 over 5 mm) can be established in a glass-fiber paper-based fluidic channel with a MS-friendly electrolyte. Amphoteric species can be well separated from the complex blood matrix and concentrated into a narrow band in the channel within 2 min, which is 20 times faster than a commercial kit method. The method can be applied to both liquid and dry spot samples, and the cleaned sample band can be simply dissolved for direct IDMS detection in ESI MRM mode. This method is a promising strategy for the rapid MS-based detection of amino acids from serum without pre-separation via liquid chromatography.

The rapid and unprecedented expansion of the global population highlights concerns about the sufficiency of food resources to sustain this growth. This study investigates and substantiates the feasibility of renewable food resources in order to meet the nutritional requirements of consumers. Three edible aquatic weeds, helencha (Enhydra fluctuans), malancha (Alternanthera philoxeroides), and kalmi (Ipomoea aquatica), were used to produce edible paper sheets. The composition of the raw aquatic weeds and paper sheet samples was analyzed, including the proximate composition, amino acid content, minerals and heavy metal contents, and bioactive compounds. The dried raw aquatic weeds and paper sheets showed similar proximate compositions, with carbohydrates being the highest component (50.38–64.63%), followed by crude protein (15.25–19.13%), ash (9.30–15.88%), and lipid (1.55–3.43%). The raw weeds and paper sheets were rich in essential minerals like Na, Ca, and Zn with contents ranging from 27.7 mg/100 g to 30.4 mg/100 g, 126.8 mg/100 g to 489.65 mg/100 g, and 4.5 mg/100 g to 16.3 mg/100 g, respectively. Acceptable levels of heavy metals, including Ni, Pb, and Cu, were found. The paper sheets contained seven essential and eight non-essential amino acids. Among the essential amino acids, the phenylalanine content was the highest at 2735.9 mg/100 g in E. fluctuans paper sheets, followed by methionine at 2377.29 mg/100 g in the raw E. fluctuans and histidine at 1972.6 mg/100 g in E. fluctuans paper sheets. A. philoxeroides sheets showed the highest total amino acid content (16,146.81 mg/100 g), while I. aquatica showed the lowest (13,118.67 mg/100 g). The aquatic weed paper sheets were rich in bioactive compounds, and the numbers in E. fluctuans, A. philoxeroides, and I. aquatica paper sheets were 31, 33, and 40, respectively. There were no significant changes in the nutritional content of the aquatic weeds in paper sheet form compared with the raw weeds, which suggests promising prospects for their production and consumption as a source of nutrition and bioactive compounds.

Proteinoids - thermal proteins - are produced by heating amino acids to their melting point and initiation of polymerisation to produce polymeric chains. Proteinoids swell in aqueous solution into hollow microspheres. The proteinoid microspheres produce endogenous burst of electrical potential spikes and change patterns of their electrical activity in response to illumination. The microspheres can interconnect by pores and tubes and form networks with a programmable growth. Introduction: Distinguishing and categorizing the origin of garlic are highly significant, considering its widespread use as a flavoring agent. With billions of dollars annually in global trade, garlic is frequently susceptible to fraudulent practices. Methodology: Paper spray ionization mass spectrometry (PS‐MS) was employed to quickly analyze garlic samples from distinct geographic origins: China and Brazil. The so‐generated PS‐MS data were treated with metabolomic multivariate approaches, and the garlic samples from these different geographic regions were easily discriminated. Results: Brazilian garlic was characterized to contain higher levels of amino acids, such as arginine, proline, and valine, and organosulfur compounds, such as allicin, alliin, and l‐γ‐glutamil‐S‐allyl‐l‐cysteine, compared to Chinese garlic. The PS‐MS data were treated employing multivariate approaches, typically used in the metabolomics field, and this protocol was promptly able to discern among both types of samples. Conclusion: Hence, this combined strategy holds promise not only as an effective tool for the authentication of the geographical origin of garlic but also as a powerful means for biomarker discovery. Mixed waste from office paper (MOW) is an excellent source of low-cost, high-quality fiber for the papermaking industry. The limitations in the application of conventional deinking to such paper are related to the negative environmental impacts that chemical substances can cause, which is why deinking in a neutral medium is receiving increasing attention. This study focuses on investigating the effect of the application of the amino acids Aspartate Glutamate and Asparagine, in the deinking of office paper, using a flotation column on a laboratory scale. The methodology involves the sample preparation, a pretreatment, flotation, the manufacture of paper, microscopy analysis, and finally the evaluation of optical properties. Different doses for each of the amino acids and of the mixture of the three were tested, at different conditions of temperature and pH in the pretreatment. The best optical characteristics were obtained when using the mixture of the three amino acids at 0.6% on a dry weight basis of the paper to be deinked, proposed in this study, with 90.8% whiteness. From scanning electron microscopy analysis, no superficial damage to fiber morphology is observed in any of the fiber samples after deinking. [ABSTRACT FROM AUTHOR]

Introduction and Objectives: Food allergy has several negative nutritional consequences and may persist beyond the first year of lives. This study aimed to assess the role of a complete oral amino acid-based supplement in the diet of children on cow's milk protein elimination diet because of food allergy., Materials and Methods: This study included two groups of children aged 1-5 years paired by age and socioeconomic status: (1) study group, on cow's milk protein elimination diet plus an oral amino acid-based supplement, and (2) control group, on cow's milk protein elimination diet. Sociodemographic, clinical, anthropometric, and dietary data were obtained through online interviews. Two 24-h dietary recalls were collected on nonconsecutive days. Both groups comprised mostly boys., Results: The study group presented lower values of body mass index. The frequency of feeding difficulties was similar between groups. The study group had a higher intake of energy, protein, carbohydrates, calcium, iron, zinc, phosphorus, magnesium, copper, selenium, vitamins D, E, B1, B2, B6, and B12, niacin, and folic acid compared to the control group. Agricultural plantations in Indonesia and Malaysia yield substantial waste, necessitating proper disposal to address environmental concerns. Yet, these wastes, rich in starch and lignocellulosic content, offer an opportunity for value-added product development, particularly amino acid production. Traditional methods often rely on costly commercial enzymes to convert biomass into fermentable sugars for amino acid production. An alternative, consolidated bioprocessing, enables the direct conversion of agricultural biomass into amino acids using selected microorganisms. This review provides a comprehensive assessment of the potential of agricultural biomass in Indonesia and Malaysia for amino acid production through consolidated bioprocessing. It explores suitable microorganisms and presents a case study on using Bacillus subtilis ATCC 6051 to produce 9.56 mg/mL of amino acids directly from pineapple plant stems. These findings contribute to the advancement of sustainable amino acid production methods using agricultural biomass especially in Indonesia and Malaysia through consolidated bioprocessing, reducing waste and enhancing environmental sustainability., (© 2024. The analysis of fingerprint chemical composition is a meaningful way to excavate the multidimensional information of fingerprint, including the donor profiling information and the age of a fingerprint, which broadens the evidential values of fingerprint, especially for the partial and distorted fingerprint. But the research remains still in the pilot phases or is ongoing. Amino acids are the dominant organic substances in latent sweat fingerprint and influenced by many donor factors. Hence, their content reflects personal information of donors. Forensic science will be revolutionized if suspects can be individualized by their amino acid content. The diverse nature, distinct physicochemical properties, and ultra-micro levels of amino acids present in fingerprints make it hard to detect. A high sensitivity method for detecting and quantifying multiple amino acid components is required. UHPLC-QqQ MS/MS offers high sensitivity, high separation, simultaneous multicomponents detection, and no derivatization, making it an ideal method for detecting and analyzing amino acids in fingerprints. Therefore, in this study, we propose and validate an efficient UHPLC-QqQ MS/MS method for the extraction and analysis of 13 amino acids from fingerprint. We compared the results of amino acids of 10 different substrates and found that the inherent amino acids in most porous substrates would have been extracted along with the fingerprint amino acids, making them unsuitable for quantitative amino acid analysis. Instead, plastic sheets are ideal substrates for laboratory studies. Then, extensive experiments were conducted among 30 donors for multidimensional information analysis. The type of samples analyzed were eccrine-rich fingerprints. A Binary Logistic Regression (BLR) model was developed, and the female and male donors were successfully differentiated by amino acids in fingerprints. Two other mathematical models were also developed to verify the accuracy, and all three different mathematical models were able to identify donors of different genders with over 90% accuracy. This demonstrates that amino acids have the potential to provide more information for donors as metabolic markers. A one-step analysis method was developed for four types of amino acids using a microfluidic paper-based analytical device fabricated from chromatography filtration paper and laminate films. Aminoacyl-tRNA synthetase was used to detect each amino acid. The obtained laminated paper-based analytical device (LPAD) contained four enzymatic reaction areas. Colorimetric detection was performed based on the molybdenum blue reaction. A model method for the simple, easy, and simultaneous detection of several amino acid concentrations was suggested, in contrast to the conventional methods such as HPLC or LC–MS. The method provided a selective quantification at the ranges of 3.6–100 μM for tryptophan, 10.1–100 μM for glycine, 5.9–100 μM for histidine and 5.6–100 μM for lysine with a detection limit of 1.1 μM, 3.3 μM, 1.9 μM and 1.8 μM, respectively. Herein, a cost-effective and portable microfluidic paper-based sensor is proposed for the simultaneous and rapid detection of glucose, free amino acids, and vitamin C in fruit. The device was constructed by embedding a poly(carboxybetaine acrylamide) (pCBAA)-modified cellulose paper chip within a hydrophobic acrylic plate. We successfully showcased the capabilities of a filter paper-based microfluidic sensor for the detection of fruit nutrients using three distinct colorimetric analyses. Within a single paper chip, we simultaneously detected glucose, free amino acids, and vitamin C in the vivid hues of cyan blue, purple, and Turnbull's blue, respectively, in three distinctive detection zones. Notably, we employed more stable silver nanoparticles for glucose detection, replacing the traditional peroxidase approach. The detection limits for glucose reached a low level of 0.049 mmol/L. Meanwhile, the detection limits for free amino acids and vitamin C were found to be 0.236 mmol/L and 0.125 mmol/L, respectively. The feasibility of the proposed sensor was validated in 13 different practical fruit samples using spectrophotometry. Cellulose paper utilizes capillary action to process trace fluids in tiny channels, and combined with pCBAA, which has superior hydrophilicity and anti-pollution properties, it greatly improves the sensitivity and practicality of paper-based sensors. Background: Asiatic hard clam (Meretrix meretrix) is an underutilized bivalve resource. This study discusses dried clam powders prepared from this resource to enhance its utilization and improve nutritional security in protein-deficient populations. Dried clam powder was prepared from Asiatic hard clam and the effects of different pre-cooking methods (boil-dried clam powder, BDCP; steam-dried clam powder, SDCP; and microwave-dried clam powder, MDCP) on nutritional (proximate composition, amino acid profiling, mineral profiling, fatty acid profiling) and physicochemical qualities were investigated., Results: Different pre-cooking methods significantly influenced the characteristics of the clam powder. The MDCP sample showed the highest concentration of amino acids, polyunsaturated fatty acids, Na, K, Ca and Mg content compared to BDCP and SDCP. The boiling process led to a loss of nutritional quality in terms of amino acids and macrominerals. The MDCP displayed the highest solubility in water (30.10%) but its oil and water absorption characteristics were the lowest among all the samples. Boil-cooked clam powder displayed the highest oil binding (2.03 mL g -1 protein) capacity. Boiling and steaming processes resulted in malondialdehyde generation compared to microwaving. Different pre-cooking processes did not influence the colour attributes significantly, but the control sample prepared without pre-cooking (CCP) had a significantly lower L* value (32.34), resulting in a darker product. In vitro digestibility of the clam powder varied in the order MDCP > SDCP > BDCP > CCP., Conclusion: The study demonstrated that nutritionally rich protein powder can be prepared from Asiatic hard clam. The simultaneous determination is reported of arginine (Arg) and valine (Val) amino acids in plasma using flower-shaped μPADs and PMMA/paper hybrid microfluidic chip based on AuNPs capped with R-thiazolidine-4-carboxylic acid (THP). In this article, the evaluation procedure is based on the smartphone colorimetric detection mechanism that results from the aggregation of the THP-AuNPs with the addition of amino acids and visual color change from red to blue. Arg and Val were selectively determined with good reproducibility and an acceptable linearity range. The flower-shaped (μPADs) provides many advantages, including low cost, reasonable sensitivity, simple and fast performance, simultaneous detection, disposable use, and high sample throughput compared with conventional colorimetric method using cuvette cells. The ratios between the absorbance wavelength at (A650/A525) and (A685/A525) are linearly proportional to the concentration of Arg and Val. Under the optimum conditions, the calibration range in aqueous solutions is 0.0068–100.0 and 0.0056–75.0 µM with a limit of detection of 2.25 and 1.86 nM for Arg and Val at pH 7.0, respectively. In the case of μPADs, the calibration curves for Arg and Val showed good linearity in the concentration range 0.01–75.0 µM. The detection limits for the analytes were 3.51 nM and 3.44 nM for Arg and Val, respectively. In addition, a PMMA/paper hybrid microfluidic chip was successfully employed to determine Arg and Val in plasma samples with a relative error below 5%. [ABSTRACT FROM AUTHOR]

Rational design of unnatural amino acid building blocks capable of stabilizing predictable secondary structures similar to protein fragments is pivotal for foldamer chemistry/catalysis. Here, we introduce novel β-amino acid building blocks: [1S,2R,4R]exoCDA and [1S,2S,4R]endoCDA, derived from the abundantly available R(+)-camphor, which is traditionally known for its medicinal value. Further, we demonstrate that the homooligomers of exoCDA adopt 6-strand conformation, which switches to a robust 10/12-helix simply by inserting flexible β-hGly spacer at alternate positions (1 : 1 β-hGly/exoCDA heterooligomers), as evident by DFT-calculations, solution-state NMR spectroscopy and X-ray crystallography. To the best of our knowledge, this is the first example of crystalline-state structure of left-handed 10/12-mixed helix, that is free from the conventional approach of employing β-amino acids of either alternate chirality or alternate β 2 /β 3 substitutions, to access the 10/12-helix. The results also show that the homooligomers of heterochiral exoCDA don't adopt helical fold, instead exhibit banana-shaped strands, whereas the homodimers of the other diastereomer endoCDA, nucleate 8-membered turns. Furthermore, the homo-exoCDA and hetero-[β-hGly-exoCDA] oligomers are found to exhibit self-association properties with distinct morphological features. This study investigated the intricate interplay between Crimean-Congo hemorrhagic fever virus (CCHFV) infection and alterations in amino acid metabolism. Our primary aim is to elucidate the impact of Crimean-Congo hemorrhagic fever (CCHF) on specific amino acid concentrations and identify potential metabolic markers associated with viral infection. One hundred ninety individuals participated in this study, comprising 115 CCHF patients, 30 CCHF negative patients, and 45 healthy controls. Liquid chromatography-tandem mass spectrometry techniques were employed to quantify amino acid concentrations. The amino acid metabolic profiles in CCHF patients exhibit substantial distinctions from those in the control group. Patients highlight distinct metabolic reprogramming, notably characterized by arginine, histidine, taurine, glutamic acid, and glutamine metabolism shifts. These changes have been associated with the underlying molecular mechanisms of the disease. Noncanonical amino acids (ncAAs) containing tertiary alcohols are valuable as precursors of natural products and active pharmaceutical ingredients. However, the assembly of such ncAA scaffolds from simple material by C-C bond formation remains a challenging task due to the presence of multiple stereocenters and large steric hindrance. In this study, we present a novel solution to this problem through highly selective enzymatic decarboxylative aldol addition. This method allows for the streamlined assembly of multifunctionalized ncAAs with γ-tertiary alcohols from readily available materials, such as L -aspartatic acid and isatins, vicinal diones and keto esters. The modularity of electrophiles furnished four classes of ncAAs with decent efficiency as well as excellent site and stereocontrol. Chirality is ubiquitous in nature, and homochirality is manifested in many biomolecules. Although β-double helices are rare in peptides and proteins, they consist of alternating L- and D-amino acids. No peptide double helices with homochiral amino acids have been observed. Here, we report chiral β-double helices constructed from γ-peptides consisting of alternating achiral (E)-α,β-unsaturated 4,4-dimethyl γ-amino acids and chiral (E)-α,β-unsaturated γ-amino acids in both single crystals and in solution. The two independent strands of the same peptide intertwine to form a β-double helix structure, and it is stabilized by inter-strand hydrogen bonds. The peptides with chiral (E)-α,β-unsaturated γ-amino acids derived from α-L-amino acids adopt a (P)-β-double helix, whereas peptides consisting of (E)-α,β-unsaturated γ-amino acids derived from α-D-amino acids adopt an (M)-β-double helix conformation. The circular dichroism (CD) signature of the (P) and (M)-β-double helices and the stability of these peptides at higher temperatures were examined. Furthermore, ion transport studies suggested that these peptides transport ions across membranes. Even though the structural analogy suggests that these new β-double helices are structurally different from those of the α-peptide β-double helices, they retain ion transport activity. The bone is a large and complex organ (12-15% of body weight) consisting of specialized connective tissues (bone matrix and bone marrow), whereas joints are composed of cartilage, tendons, ligaments, synovial joint capsules and membranes, and a synovial joint cavity filled with synovial fluid. Maintaining healthy bones and joints is a dynamic and complex process, as bone deposition (formation of new bone materials) and resorption (breakdown of the bone matrix to release calcium and phosphorus) are the continuous processes to determine bone balance. Bones are required for locomotion, protection of internal organs, and have endocrine functions to maintain mineral homeostasis. Joints are responsible for resisting mechanical stress/trauma, aiding in locomotion, and supporting the overall musculoskeletal system. Amino acids have multiple regulatory, compositional, metabolic, and functional roles in maintaining the health of bones and joints. Their disorders are prevalent in mammals and significantly reduce the quality of life. These abnormalities in companion animals, specifically cats and dogs, commonly lead to elective euthanasia due to the poor quality of life. Multiple disorders of bones and joints result from genetic predisposition and are heritable, but other factors such as nutrition, growth rate, trauma, and physical activity affect how the disorder manifests. Treatments for cats and dogs are primarily to slow the progression of these disorders and assist in pain management. Therapeutic supplements such as Cosequin and formulated diets rich in amino acids are used commonly as treatments for companion animals to reduce pain and slow the progression of those diseases. Also, amino acids (e.g., taurine, arginine, glycine, proline, and 4-hydroxyproline), and glucosamine reduce inflammation and pain in animals with bone and joint disorders. This work presents a pseudo-operational study on thermal paper for the detection of latent fingermarks with vacuum metal deposition (VMD) and amino acid sensitive reagents (1,2-indanedione and ninhydrin) on 120 thermal receipts. Each receipt was cut in half and separated into two processing groups. Each group was processed by one of two sequential treatment processes. Both sides of all receipts (thermal and nonthermal side) were treated. Fingermarks developed by each process were counted and imaged. Both sequences started with the use of UV imaging. Process A then continued with VMD using gold and zinc (VMDAu/Zn), then VMD using silver (VMDAg), followed by two amino acid sensitive reagents (1,2 indanedione and ninhydrin). Process B used only the amino acid sensitive reagents. Process A increased the detection rate on the thermal side by approximately a factor of two and a half when compared to the VMD techniques. The VMD processes did not visualize all of the marks detected by UV; however, this was the case for the amino acid sensitive reagents. The use of VMD can be suitable for the detection of fingermarks on thermal paper in cases where the text needs to be retained; however, the use of 1,2-indanedione and ninhydrin, in sequence with VMD, will detect the highest number of marks. Selective structural modification of amino acids and peptides is a central strategy in organic chemistry, chemical biology but also in pharmacology and material science. In this context, the formation of tetrazole rings, known to possess significant therapeutic properties, would expand the chemical space of unnatural amino acids but has received less attention. In this study, we demonstrated that the classic unimolecular Wolff rearrangement of α-amino acid-derived diazoketones could be replaced by a faster intermolecular cycloaddition reaction with aryldiazonium salts under identical practical conditions. This strategy provides an efficient synthetic platform that could transform proteinogenic α-amino acids into a plethora of unprecedented tetrazole-decorated amino acid derivatives with preservation of the stereocenters. Density functional theory studies shed some light on the reaction mechanism and provided information regarding the origins of the chemo- and regioselectivity. Objective: Our goal was to determine the efficacy of the American Society for Parenteral and Enteral Nutrition's recommended carnitine dosage of 5 mg/kg/day in maintaining normal serum free carnitine and total acylcarnitine levels in preterm neonates receiving parenteral nutrition (PN)., Study Design: A retrospective cohort study was conducted on neonates born <30 weeks gestation and weighing <1250 g, comparing those who received carnitine supplementation to those without supplementation. Free carnitine and total acylcarnitine data were collected from routine newborn screens in the first days of life and on full enetral feeds. Univariate analysis was performed, and those factors that were significantly different between the two groups were adjusted for using mixed effects analysis., Results: There were 108 supplemented and 45 unsupplemented neonates in the study. At baseline, free carnitine (19.8 ± 3.3 vs 18.9 ± 3.7 µmol/L, P = 0.53) and total acylcarnitine (26.6 ± 5.1 vs 22.5 ± 7.1 µmol/L, P = 0.11) were similar between the two groups. At full enteral feeds, compared with unsupplemented group, supplemented infants had significantly higher free carnitine (27.1 ± 16.4 vs 17.1 ± 8.5 µmol/L, P < 0.001) and total acylcarnitine (30.3 ± 11.5 vs 20.2 ± 10.1 µmol/L, P < 0.001). None of the supplemented neonates developed biochemical carnitine deficiency as compared with 18% in the unsupplemented group (P < 0.001). Hypusine amino acid [N ε -(4-amino-2-hydroxybutyl)-lysine] was first isolated in 1971 from bovine brain extracts. Hypusine originates from a post-translational modification at the eukaryotic translation initiation factor 5A (eIF5A), a protein produced by archaebacteria and eukaryotes. The eIF5A protein is the only one described containing the hypusine residue, which is essential for its activity. Hypusine as a free amino acid is a consequence of proteolytic degradation of eIF5A. Herein, we showed, for the first time, evidence of biological activity for the free hypusine. C6 rat glioma cells were treated with hypusine, and different cellular parameters were evaluated. Hypusine treatment significantly reduced C6 cell proliferation and potently suppressed their clonogenic capacity without leading to apoptosis. Hypusine also decreased the Eif5A transcript content and the global protein synthesis profile that may occur due to negative feedback in response to high hypusine concentration, controlling the content of newly synthesized eIF5A, which can affect the translation process. Besides, hypusine treatment also altered cellular metabolism by changing the pathways for energy production, reducing cellular respiration coupled with oxidative phosphorylation, and increasing the anaerobic metabolism. These observed results and the relationship between eIF5A and tumor processes led us to test the combination of hypusine with the chemotherapeutic drug temozolomide. Combining temozolomide with hypusine reduced the MTT conversion to the same levels as those observed using double temozolomide dosage alone, demonstrating a synergetic action between the compounds. Thus, since 1971, this is the first study showing evidence of biological activity for hypusine not associated with being an essential component of the eiF5A protein. Thioamide peptides were synthesized in a straightforward one-pot process via the linkage of diverse natural amino acids in the presence of thiolphosphonate and trichlorosilane, wherein carbonyl groups were replaced with thiono compounds with minimal racemization. Experimental and computational mechanistic studies demonstrated that the trichlorosilane enables the activation of carboxylic acids via intense interactions with the Si-O bond, followed by coupling of the carboxylic acids with thiolphosphonate to obtain the key intermediate S-acyl dithiophosphate. Silyl-activated quadrangular metathesis transition states afforded the thioamide peptides. A number of goose breeds are raised commercially in China. However, the data on the slaughter performance of the goose breeds and the nutritional value of their meats lack a thorough comparative analysis. In this systematic review, the slaughter performance of the goose breeds and nutritional value of their meats were comparatively analyzed to provide an overview of the characteristics of the goose breeds raised commercially in China. Fifteen goose breeds were selected from 27 research articles published up to January 2022 on the slaughter performance of the goose breeds raised commercially in China and their nutrient composition after literature searching, literature screening, variety selection, and data collation. The slaughter indexes of the goose breeds and the basic nutrient composition, amino acid composition, and fatty acid composition of the meats of the goose breeds were standardized using min-max normalization and compared. The results suggest that the slaughter indexes and nutritional indicators of the meats of Yangzhou white goose, Xupu goose, Landaise geese, and Sichuan white goose are more balanced than those of the meats of the other goose breeds. Background: Nutrition therapy and administration of albumin preparations are common in postsurgical patients. However, the effects of these interventions on albumin metabolism are unclear. We elucidated the effect of postoperative albumin and/or parenteral nutrition administration on it., Methods: Sprague-Dawley rats underwent surgery involving intestinal rubbing followed by intestinal exposure. Subsequently, they were administered experimental solutions for 48 h, their blood samples were collected at 24 and 48 h, and livers were excised at 48 h. Based on experimental solutions, rats were divided into five groups: non-surgical (Non-surg); glucose and electrolyte solution (GE); amino acid, glucose, and electrolyte solution (AGE); GE + rat serum albumin (Alb) (GE + Alb); and AGE + Alb. Their plasma albumin concentrations; albumin fractional synthesis rate (ALB FSR); mercaptoalbumin/total albumin ratio (MA ratio); and messenger RNA (mRNA) expressions of albumin and hepatocyte nuclear factor-1 (HNF-1) in the liver were measured., Results: The GE and AGE groups showed significant decline in albumin concentrations. ALB FSR was significantly enhanced in the AGE group compared with the GE group. The mRNA expression of albumin was similar to ALB FSR in all groups and that of HNF-1 was significantly decreased in the GE + Alb and AGE + Alb groups compared with the Non-surg group. Purpose of Review: An increase in the plant-based characteristics of the diet is now recommended for human and planetary health. There is growing evidence that plant protein (PP) intake has beneficial effects on cardiometabolic risk. However, proteins are not consumed isolated and the protein package (lipid species, fiber, vitamins, phytochemicals, etc) may contribute, besides the protein effects per se, to explain the beneficial effects associated with PP-rich diets., Recent Findings: Recent studies have shown the potential of nutrimetabolomics to apprehend the complexity of both the human metabolism and the dietary habits, by providing signatures associated to the consumption of PP-rich diets. Those signatures comprised an important proportion of metabolites that were representative of the protein package, including specific amino acids (branched-chain amino acids and their derivates, glycine, lysine), but also lipid species (lysophosphatidylcholine, phosphatidylcholine, plasmalogens) and polyphenol metabolites (catechin sulfate, conjugated valerolactones and phenolic acids)., Summary: Further studies are needed to go deeper in the identification of all metabolites making part of the specific metabolomic signatures, associated to the large range of protein package constituents and their effects on the endogenous metabolism, rather than to the protein fraction itself. Organic matter (OM) and nutrient inputs generated by human activities promote the development of eutrophication. Amino acids (AAs) are an integral part of OM, and studying their patterns will provide new insights into organic matter dynamics in lakes. Four seasonal field campaigns in eutrophic Lake Taihu and a 14-day phytoplankton degradation experiment were carried out to determine the variability and bioavailability of amino acids. The quality and quantity of AAs varied among different seasons and lake types. The concentrations of particulate (PAA) and dissolved (DAA) AA were 14.67 ± 13.25 μM (carbon- and nitrogen-normalized PAA yields: PAA-C%, 23.8 ± 13.5%; PAA-N%, 22.8 ± 2.1%) and 2.95 ± 1.05 μM (carbon- and nitrogen-normalized DAA yields: DAA-C%, 3.7 ± 1.1%; DAA-N%, 12.6 ± 11.2%) in Lake Taihu, respectively. PAA and DAA showed high mean values in the algal-dominated northern area in summer. Glutamic acid (Glu), alanine (Ala), aspartic acid (Asp) and glycine (Gly) contributed to nearly 50% of PAA, while Gly, accounting for 17-24%, was the main component of DAA. During a 14-day phytoplankton degradation period, 98% of Chl a, 63% of POC and 92% of PAA were removed in the dark treatment, and a more obvious downward trend was recorded than in the light treatment. Principal component analysis of the indices of PAA indicated that Glu, arginine (Arg) and histidine (His) were dominant on day 0, while Gly and lysine (Lys) were dominant on day 14. There were notable serine + threonine (Ser + Thr [mol%]) and aspartic acid/glycine ratio (Asp/Gly [mol%]) end-member divisions among different organic matter sources. DAA had higher Ser + Thr [mol%] and lower Asp/Gly [mol%] values than PAA. The amino acid degradation index (DI) of PAA and DAA was 0.97 ± 0.28 and - 1.04 ± 0.43, respectively. The fresh DAA from the algae degradation incubation also had DI values similar to those of field DAA in the northern algae-dominated lake region. This study reports the synthesis of functional graphene quantum dots (GQDs) through the thermal pyrolysis of D-penicillamine (DPA) and citric acid towards water treatment. For the first time, a novel type of highly fluorescent probe for selective Cu2+ detection among 26 types of metal ions which considerably enhance the fluorescence intensity of GQD. Furthermore, the recognition effects can be strengthen addition of amino acids owing to raise the electron density of the system and boos the accumulation profile of the sensing molecules. This simple fluorescent probe was applied for facile metal ions recognition in human urine samples, as well as environmental fluids. The proposed DPA-GQDs supported amino acids respond to Cu2+, Hg2+, and Fe3+, with high sensitivity. The intensity of the fluorescence histogram of this probe significantly diminished in exposure to metal ions such as, Cu(II), Hg(II), and Fe(III). Moreover, in our work, microfluidic paper-based (µPADs) was fabricated through a facile and a cost-effective method. Cu2+ can selectively be recognized by GQD-DPA on the paper-based sensors by naked-eye. The functionalized DPA-GQDs is an excellent alternative to previously reported fluorescent probes for sensing, bio-labeling, and other biological usages in aqueous solution. Its nitrogen and oxygen-rich groups can coordinate with Cu2+ and cause to an apparent fluorescence quenching by the non-radiative electron-transmission. The technique presented in this paper is economical and simple in design, and provides a "mix-and-sense" protocol with no need to chemical- or dye-modifications. Our proposed Cu2+ sensing probe exhibits linear response in the concentrations ranging from 0.001 ppm to 5 ppm, with a lower limit of quantitation (LLOQ) of 0.1 ppm in both environmental fluids, and human urine samples. The enhanced color uniformity, the low instrumental needs of the stamp, and disposability of μPADs enable the application of the suggested paper-based device for the commercial diagnostics biosensor. Ynamides, a class of novel coupling reagents for peptide synthesis, facilitated peptide bond formation in a one-pot, two-step manner with α-acyloxyenamide active esters of amino acids as stable intermediates. Ynamide-mediated peptide synthesis proceeded by a reaction mechanism that is completely different from that of conventional coupling reagents and exhibited superiority in addressing the issue of racemization/epimerization during peptide bond formation. Herein, we present a systematic mechanistic analysis, including kinetics and Brønsted-type structure-reactivity studies and density functional theory calculations, providing unprecedented mechanistic insight into ynamide-mediated peptide bond formation. Here, a new and inexpensive electrochemical paper-based analytical device (ePAD) containing a pencil-drawn three-electrode setup, in which the working electrode was modified with graphene nanoribbons (GNRs), is presented for the first time. Under optimal conditions, the GNRs-ePAD provided higher peak currents (ca. 91 %), lower electron transfer resistance (ca. 32-fold) and greater electroactive area (more than twice) when compared to the unmodified ePAD. This significant improvement can be attributed to the increase in the number of active sites available on the electrode surface due to the high conductivity of GNRs. In addition, the ePAD showed good reproducibility and acceptable repeatability, with relative standard deviations (RSDs) ≤ 8.4 % for peak currents and peak potentials. As proof of concept, the GNRs-ePAD was employed for analysis of 5-HTP in pharmaceuticals. As far as we know, this is the first report of the use of ePADs for determination of 5-HTP. The electroanalytical quantification was carried out using square-wave voltammetry (SWV). A linear range from 25 to 1000 μmol L−1 and a limit of detection of 7.6 μmol L−1 were obtained. The sensitivity achieved with the GNRs-ePAD was almost 5-fold greater than that obtained with the bare electrode. The results achieved during the analysis of pharmaceutical products showed good precision (RSD < 3 %) and great concordance with the values found by UV–vis spectrophotometry, utilized for comparison (relative error was < 2 %). None interference or matrix effects were observed during measurements. These results demonstrate promising potential for the GNRs-ePAD proposed here. This device shows itself as an interesting, efficient and low-cost analytical platform for analysis of electroactive species, being specially interesting for places with limited resources. The glycosylative modification of peptides could improve the pharmacological properties of peptide drugs and deliver them efficiently to the target sites. Compared with O-/N-glycosides, C-glycosides exhibit more metabolic stability. We here disclose the first example of visible-light-promoted and Cu-catalyzed stereoselective C-glycosylation. The mild reaction conditions are compatible with various carbohydrate substrates, as demonstrated with a series of monosaccharides and a disaccharide, and are amenable to the synthesis of a wide variety of C-glycoamino acids and C-glycopeptidomimetics with good yields and excellent stereoselectivities. A hemicellulose-first strategy of acetic acid pretreatment was applied for the protein-rich paper mulberry biowaste (PMB) to produce amino acid-rich xylo-oligosaccharide (XOS). In addition, the pretreated PMB was further used to produce fermentable monosaccharides by enzymatic hydrolysis. Results showed a yield of 18.2% (based on xylan in raw PMB) was achieved for XOS in hydrolyzate under the optimal pretreatment conditions (168 °C, 68 min, 1% acetic acid). Besides, it's found that the obtained hydrolyzate contains 14.3% (based on amino acids in raw PMB) amino acids degraded from the protein in PMB. Furthermore, an additional enzymatic hydrolysis process with endo-xylanase was used to upgrade the XOS with DP of 2–6 in the hydrolysate, which could increase XOS yield to 29.8%. Amino acid contents were unchanged after post-enzymatic hydrolysis. Besides, a glucose yield of 51.7% (11.5 g/L glucose) could be achieved for the pretreated PMB after enzymatic hydrolysis. This work serves as proof of concept for creating higher-value XOS from biorefinery processes considering protein-enriched low-value biomass feedstocks. A direct fluorimetric method, employing μicro-analytical paper-based devices (μ-PADs) for the selective determination of histidine (HIS) is described. The suggested method exploits the fluorescence emission of histidine after its rapid reaction with o -phthalaldehyde (OPA) at a basic medium (pH = 10) on the surface of a paper device with the application of a UV lamp at 354 nm. The devices are inexpensive and are composed of chromatographic paper and wax barriers. The analytical protocol is easily applicable with minimal technical expertise and without the need of expensive experimental apparatus. The user has to add a test sample, illuminate the device with a UV lamp, and read the fluorescence of the sensing area using a simple imaging device such as a cell-phone camera. The method is free from common interferences likely to affect the measurement of histidine and is selective among all other amino acids. This analytical procedure was optimized and validated, paying special attention to its intended application. The detection limits are as low as 1.8 μM with very satisfactory precision ranging from 6.4% (intra-day) to 8.9% (inter-day). Random urine samples from adult volunteers (n = 5) were successfully analyzed and HIS content ranged between 260 and 1114 μmol L−1 with percentage recoveries in the range of 78.2 and 124.6%. Measurement of ileal amino acids (AA) bioavailability is recommended to evaluate protein quality. A dual isotope tracer method, based on plasma isotopic enrichment ratios, has been proposed to determine true digestibility in humans. In a pilot study, we aimed to evaluate whether this method could be implemented in rats to determine AA bioavailability based on isotopic enrichment ratios measured in cecal digesta or plasma samples. Goat milk proteins were intrinsically labeled with 15 N and 2 H. Wistar rats were fed a meal containing the doubly labeled goat whey proteins and a tracer dose of 13 C-spirulina. Blood samples were collected 0, 1 h and 3 h after meal ingestion from the tail vein. The rats were euthanized 4 h (n = 6) or 6 h (n = 6) after meal to collect plasma and intestinal contents. True orocecal protein digestibility and AA bioavailability were assessed by means of 15 N and 2 H enrichment in cecum content and compared with absorption indexes determined at the plasma or cecum level using isotopic ratios. Plasma kinetics of isotopic enrichment could not be completed due to the limited quantity of plasma obtained with sequential blood collection. However, the absorption indexes determined from cecal 15 N or 2 H/ 13 C ratios gave coherent values with true orocecal AA bioavailability. This dual isotope approach with measurements of isotopic ratios in digestive content could be an interesting strategy to determine true AA bioavailability in ileal digesta of rats., (© 2022. Cardiovascular diseases (CVD) are a significant and growing burden on global health services, and it is now accepted that impairment of vascular function represents a major preliminary step in the development of CVD. There is considerable interest in identifying both causal factors of impaired vascular function, as well as related nutritional factors that may lower the risk of developing CVD, and food-derived bioactive peptides and amino acids have emerged as one such area. Dairy foods contain two groups of proteins, whey proteins and caseins, which represent a rich source of bioactive peptides that are released during food processing and/or digestion. These peptides have a number of physiological activities including the potential to reduce blood pressure. Research, including acute and longer-term randomised controlled trials, animal models and in vitro models has demonstrated the potential impact of dairy proteins on vascular function. The purpose of this paper is to narratively review the evidence, primarily from randomised controlled trials, examining the effects of whey proteins, their peptides and amino acids on vascular function and related issues including blood pressure. In addition, it will explore the potential underlying mechanisms responsible for these effects. It concludes that there is increasing evidence that whey proteins, and notably the bioactive peptides and amino acids released during their digestion, can have beneficial effects on aspects of vascular function and thus contribute to CVD risk reduction. In the current study, we have utilized semi-enclosed, leak-proof, microfluidic paper-based analytical devices (μPAD's) modified with isatin conjugated chitosan as specific colorimetric reagent for the detection of proline. Proline is one of the globally accepted stress biomarker in plants and also one of the prominent amino acid present in wine and some processed food. Quantification of proline is regularly required in agriculture field, food and wine industries. Specific interaction of isatin with proline, uniform film forming ability of chitosan which results in uniform coloration and the presence of leak-proof layer which prevent the diffusion of colorimetric reagent deeper resulted in enhancement of color signal intensity at the reaction zone were utilized. Further, the images of the μPAD's were captured using smartphone with 3D printed imaging box which houses smartphone and μPAD's. This platform utilizes smartphone flash for uniform illumination and ensures constant positioning of μPAD's to capture images. This greatly enhances the sensitivity and accuracy of our platform. Compared to previously published highly sensitive multi-layer, paper-based platform for detection of proline, current method has enhanced detection range (∼7 fold) and has comparable limit of detection of 23.75 μM. Moreover, the developed μPAD's platform has reduced optimum reaction temperature and time compared to previous work. The developed paper based platform was utilized for evaluation of proline content in young Arabidopsis plants which are subjected to water stress for 5 days. The devised paper-based methods have the potential to be applicable for the on-site evaluation of various stresses in plants. Aquatic animals have particularly high requirements for dietary amino acids (AAs) for health, survival, growth, development, and reproduction. These nutrients are usually provided from ingested proteins and may also be derived from supplemental crystalline AA. AAs are the building blocks of protein (a major component of tissue growth) and, therefore, are the determinants of the growth performance and feed efficiency of farmed fish. Because protein is generally the most expensive ingredient in aqua feeds, much attention has been directed to ensure that dietary protein feedstuff is of high quality and cost-effective for feeding fish, crustaceans, and other aquatic animals worldwide. Due to the rapid development of aquaculture worldwide and a limited source of fishmeal (the traditionally sole or primary source of AAs for aquatic animals), alternative protein sources must be identified to feed aquatic animals. Plant-sourced feedstuffs for aquatic animals include soybean meal, extruded soybean meal, fermented soybean meal, soybean protein concentrates, soybean protein isolates, leaf meal, hydrolyzed plant protein, wheat, wheat hydrolyzed protein, canola meal, cottonseed meal, peanut meal, sunflower meal, peas, rice, dried brewers grains, and dried distillers grains. Animal-sourced feedstuffs include fishmeal, fish paste, bone meal, meat and bone meal, poultry by-product meal, chicken by-product meal, chicken visceral digest, spray-dried poultry plasma, spray-dried egg product, hydrolyzed feather meal, intestine-mucosa product, peptones, blood meal (bovine or poultry), whey powder with high protein content, cheese powder, and insect meal. Microbial sources of protein feedstuffs include yeast protein and single-cell microbial protein (e.g., algae); they have more balanced AA profiles than most plant proteins for animal feeding. Animal-sourced ingredients can be used as a single source of dietary protein or in complementary combinations with plant and microbial sources of proteins. The composition of amino acid and fatty acid has a vital function on meat quality and animal health. However, the underlying mechanism of amino acid and fatty acid metabolism in sheep during different grazing periods is still unclear. In this study, a total of 12 sheep were employed in different grazing periods. Our results showed that the composition of amino acids and fatty acids in muscle and adipose tissues was significantly altered between dry grass (DG) period and green grass (GG) period. Changes in the activities of the metabolism-related enzymes including BCKD, BCAT2, ACC, SCD, HSL, GSK3β, p-GSK3β, and FABP4 were observed in muscle and adipose during different grazing periods. In addition, the mRNA expression levels of ACC, FAS, SCD, HSL, LPL, and DGAT1 in muscle and adipose tissue were changed markedly in different grazing periods. Furthermore, the expression levels of mTOR and β-catenin/PPARγ/C/EBPα pathway-related proteins were predominantly altered in muscle and adipose among DG and GG. Taken together, all investigations simplified the process of amino acid and fatty acid metabolism disorders caused by different grazing periods, and the mTOR and β-catenin/PPARγ/C/EBPα play the essential role in this process, which provided an underlying mechanism of metabolism and meat quality., (© 2022 Japanese Society of Animal Science.)

Nonalcoholic fatty liver disease (NAFLD) is one of the most common liver diseases worldwide and is often associated with aspects of metabolic syndrome. Despite its prevalence and the importance of early diagnosis, there is a lack of robustly validated biomarkers for diagnosis, prognosis and monitoring of disease progression in response to a given treatment. In this Review, we provide an overview of the contribution of metabolomics and lipidomics in clinical studies to identify biomarkers associated with NAFLD and nonalcoholic steatohepatitis (NASH). In addition, we highlight the key metabolic pathways in NAFLD and NASH that have been identified by metabolomics and lipidomics approaches and could potentially be used as biomarkers for non-invasive diagnostic tests. Overall, the studies demonstrated alterations in amino acid metabolism and several aspects of lipid metabolism including circulating fatty acids, triglycerides, phospholipids and bile acids. [Display omitted] • High-efficiency (2 orders of magnitude), reproducible (RSD 2.5%), fast (within 2 min) parallel electric stacking PAD was created; • The wet real-time photographic colorimetry results of the parallel PAD system are insensitive to the changes of light conditions; • On-line colorimetric detection of pigments in drinks and off-line MS/MS detection of serum amino acids are used to demonstrate their applicability. A microfluidic paper-based analytical device (PAD) with both pre-concentration and parallel operation functions is of great promise for sensitive and high throughput detection. In this paper, a fast and multiplexed PAD was designed based on wax-screen-printing with glass fiber paper substrate and validated by image-based colorimetry. Driven by the same DC power supply, parallel electrokinetic stacking with the PAD of 8 channels can be completed within 2 min, and each channel can be loaded with different samples. Enrichment factor of 2 orders of magnitude was demonstrated with RSD below 2.5% between the chips and the channels of a chip. Reproducible and multiplexed stacking operations of this PAD provide a solid foundation for online calibration and sample detection on the same chip at the same time. Applications of this PAD was demonstrated by online colorimetric detection of food pigments from beverages and offline MS/MS detection of amino acids from a serum sample. Besides high-throughput and sensitive point-of-care test detection, this PAD method could also find applications for efficient pre-separation and pre-concentration of complex samples.
