Forming limit curves (FLC) at high temperatures under plane-stress states are beneficial for guiding the process parameter design of hot forming superalloy sheets. Constrained by the high deformation temperatures, the establishment of FLCs of superalloy sheets at high temperatures is extremely difficult. This paper conducts an experimental and modelling research to establish the physical-based model with the accurate prediction of hot flow behavior as well as micro grain sizes, using which, the forming limit curves can be predicted. First, the uniaxial tensile tests under different temperatures and strain rates and electron backscattered scattering detection (EBSD) observations of typical conditions were conducted to obtain the hot flow behavior and grain evolution. Second, the dome test at room temperature was performed, and the corresponding simulation was further carried out to obtain the forming limit at room temperature under plane-stress states. Using the above results, a plane-stress constitutive model for GH3128 superalloy was established enabling plane-stress test results under typical hot conditions to be accurately predicted. The study of drug–target interaction plays an important role in the process of drug development. The subject of DTI forecasting has advanced significantly in the last several years, yielding numerous significant research findings and methodologies. Heterogeneous data sources provide richer information and comprehensive perspectives for drug–target interaction prediction, so many existing methods rely on heterogeneous networks, and graph embedding technology becomes an important technology to extract information from heterogeneous networks. These approaches, however, are less concerned with potential noisy information in heterogeneous networks and more focused on the extent of information extraction in those networks. Based on this, a potential DTI predictive network model called FBRWPC is proposed in this paper. It uses a fine-grained similarity selection program to first integrate similarity on similar networks and then a bidirectional random walk graph embedding learning method with restart to obtain an updated drug target interaction matrix. Through the use of similarity selection and fine-grained selection similarity integration, the framework can effectively filter out the noise present in heterogeneous networks and enhance the model's prediction performance. The experimental findings demonstrate that, even after being split up into four distinct types of data sets, FBRWPC can still retain great prediction performance, a sign of the model's resilience and good generalization. Aiming at the prediction model of bearing performance degradation based on recurrent neural network (RNN) and its variants ignores the feature spatial correlation, and cannot effectively handle long time series data, this paper proposes a rolling bearing performance degradation prediction model based on temporal graph convolutional neural network (T-GCN). For non-stationary and nonlinear characteristics of vibration signals, this paper introduces a rolling bearing feature evaluation method based on multiscale dispersion entropy (MDE) to better characterize time series. To effectively solve the spatial correlation problem between samples and features, this paper uses the topological structure of a path graph to build a graph model and combines gated recurrent unit (GRU) and graph convolutional neural network (GCN) to build a T-GCN prediction model. Finally, this article established a rolling bearing fault prediction experimental platform and validated it using the University of Cincinnati public dataset. The experiment shows that compared with GRU, GCN, and LSTM models, the RMSE and the MAE evaluation indicators based on the T-GCN model have decreased by 6 % to 28 % and 11 % to 28 %, respectively, which suggests that the T-GCN model has a higher prediction accuracy and a better model fitting goodness. Trichoderma harzianum T4 is a soil fungus that plays an important role in the biological control of plant diseases. The aim of this study was to functionally characterize the β-1,6-glucanase gene Neg1 in T. harzianum T4 and to investigate the effect of its overexpression on biocontrol traits, especially antagonism against pathogenic fungi. We found that overexpression of Neg1 did not affect growth of T. harzianum but enhanced sporulation of T. harzianum T4 cultures. Generally, spores are closely related to the defense ability of defense fungi and can assist their proliferation and improve their colonization ability. Secondly, overexpression of Neg1 also increased the secretion level of various hydrolytic enzymes and enhanced the antagonistic ability against phytopathogenic fungi of Fusarium spp. The results suggest that Neg1 is a key gene for improving the biocontrol effect of T. harzianum T4, which contributes to a better understanding of the mechanism of action of T. harzianum T4 as a fungal biocontrol agent. Existing studies have suggested concurrent correlations between working memory and linguistic skills for children with intellectual disabilities; however, evidence for the longitudinal relationship is sparse. This study collected longitudinal data on working memory and linguistic skills from 103 Chinese school-aged children with intellectual disabilities. At the first time of testing, children were 6 to 16 years old and completed measurements of working memory and receptive vocabulary. The same measurements were administered 1 and 2 years later. Children's working memory was assessed by backward digit span and starting position selection, and receptive vocabulary was measured using the Peabody Picture Vocabulary test. The random intercept cross-lagged panel model results demonstrated that working memory scores significantly predicted changes in receptive vocabulary for children with intellectual disabilities over time, but the reverse pattern was not evident. To some extent, our findings suggested that interventions targeting boosting working memory may subsequently enhance receptive vocabulary in children with intellectual disabilities. Purpose: Breast cancer (BC) is the most prevalent malignant tumor worldwide among women, with the highest incidence rate. The mechanisms underlying nucleotide metabolism on biological functions in BC remain incompletely elucidated. Materials and Methods: We harnessed differentially expressed nucleotide metabolism-related genes from The Cancer Genome Atlas-BRCA, constructing a prognostic risk model through univariate Cox regression and LASSO regression analyses. A validation set and the GSE7390 dataset were used to validate the risk model. Clinical relevance, survival and prognosis, immune infiltration, functional enrichment, and drug sensitivity analyses were conducted. Results: Our findings identified four signature genes (DCTPP1, IFNG, SLC27A2, and MYH3) as nucleotide metabolism-related prognostic genes. Subsequently, patients were stratified into high- and low-risk groups, revealing the risk model's independence as a prognostic factor. Nomogram calibration underscored superior prediction accuracy. Gene Set Variation Analysis (GSVA) uncovered activated pathways in low-risk cohorts and mobilized pathways in high-risk cohorts. Distinctions in immune cells were noted between risk cohorts. Subsequent experiments validated that reducing SLC27A2 expression in BC cell lines or using the SLC27A2 inhibitor, Lipofermata, effectively inhibited tumor growth. Conclusions: We pinpointed four nucleotide metabolism-related prognostic genes, demonstrating promising accuracy as a risk prediction tool for patients with BC. As the center of the development of power industry, wind-photovoltaic (PV)-shared energy storage project is the key tool for achieving energy transformation. This research seeks to construct a feasible model for investment appraisal of wind-PV-shared energy storage power stations by combining geographic information system (GIS) and multi-criteria decision-making (MCDM) method. Firstly, a comprehensive criteria system is established from the perspectives of orography, economy, resources, climate, and society, and the evaluation data is described using probabilistic linguistic term sets (PLTSs). Then, to avoid the weight deviation produced by the single weighting approach, a comprehensive weighting model including the best–worst method (BWM) and entropy weight method is provided to calculate the weights of criteria. Next, expert weights are calculated based on trust analysis. Finally, alternatives are ranked by the improved gained and lost dominance score (GLDS) method. To verify the validity of the model, an empirical investigation is carried out in Shanxi Province. The results show that the economy is the primary factor influencing the investment decision. Among all the projects approved by the government, alternative F4 located in Yanzhuang Town, Yuanping City is the best investment object. Furthermore, to illustrate the stability of the result, triple sensitivity analysis and comparative analysis are conducted in Shanxi Province. The progress of aquaculture heavily depends on the efficient utilization of diverse genetic resources to enhance production efficiency and maximize profitability. Single nucleotide polymorphisms (SNPs) have been widely used in the study of aquaculture genomics, genetics, and breeding research since they are the most prevalent molecular markers on the genome. Currently, a large number of SNP markers from cultured fish species are scattered in individual studies, making querying complicated and data reuse problematic. We compiled relevant SNP data from literature and public databases to create a fish SNP database, FishSNP (http://bioinfo.ihb.ac.cn/fishsnp), and also used a unified analysis pipeline to process raw data that the author of the literature did not perform SNP calling on to obtain SNPs with high reliability. This database presently contains 45,690,243 (45 million) nonredundant SNP data for 13 fish species, with 30,288,958 (30 million) of those being high-quality SNPs. An immune-related gene signature (IRGS) was established to better understand the molecular and immunologic characteristics of microsatellite instable (MSI) and microsatellite stable (MSS) endometrial carcinoma (EC), and provide potential immunotherapy directions for MSS patients. Top 20 immune-related hub genes were screened by weight gene coexpression network analysis (WGCNA), and an IRGS was further established through Cox regression analysis. The molecular and immune characteristics were clarified in IRGS high and low risk groups. Expression and MS status validation of the IRGS were conducted through quantitative real-time Polymerase Chain Reaction (rt-qPCR) and immunohistochemistry (IHC) analysis. The IRGS includes 2 oncogenes (AGTR1 and HTR3C) and 2 tumor suppressor genes (CD3E and SERPIND1). Patients in IRGS high-risk group were more with MSS status, higher tumor grade, later FIGO stage, serous histology and elder ages compared with IRGS low-risk group (P < 0.05). Besides, patients in MSS group were more FIGO stages II–IV (42.7% vs. 26%), serous histology (35.7% vs. 5.3%) and with higher IRGS risk score (1.51 ± 3.11 vs. 1.02 ± 0.67) (P < 0.05) than patients in MSI group. Furthermore, patients in IRGS high-risk group had higher tumor purity, more Macrophages M1 and Macrophages M2 infiltrating, higher proportion of Macrophages M2 and Dendritic cells activated, lower proportion of T cells regulatory (Tregs), lower tumor mutation burden (TMB). Correspondingly, subjects in IRGS low-risk group had higher immunphenoscores than IRGS high-risk group. The relative mRNA level of AGTR1 and HTR3C were gradually increase, while CD3E and SERPIND1 were reversed in rt-qPCR. Through IHC experiments, AGTR1(69.2% vs 30%, P = 0.074) and HTR3C (76.9% vs 30%, P = 0.024) had higher positive staining rates in ECs than non-ECs. While SERPIND1 (84.6% vs 20%, P = 0.003) and CD3E (61.5% vs 40%, P = 0.000) had higher positive staining rates in non-ECs. IRGS is a potential diagnostic and prognostic biomarker for EC. Parenting practices are assumed to have an important influence on predicting adolescents' prosocial behavior. However, the underlying mechanisms of the link between the critical parenting practice of parental behavioral control and adolescents' prosocial behavior have received relatively less attention. Adolescents' empathy and self-esteem are also identified as contributing to their prosocial behavior. This study investigated whether adolescents' empathy mediated the relationship between maternal behavioral control and their prosocial behavior and whether the mediation process was moderated by their self-esteem. Self-report questionnaires measuring these variables were administered to 2502 Mainland Chinese adolescents (M age = 13.79 years; range = 11‒18 years; 52% girls). After controlling for demographic variables, the results revealed that empathy partially mediated the link between maternal behavioral control and prosocial behavior. Furthermore, the effect of maternal behavioral control on empathy and empathy on prosocial behavior was both moderated by self-esteem. The Tibetan livestock sector is now ailing from many infectious ailments brought on by harmful microorganisms. Therefore, this research aimed to assess the probiotic potential and safety of Bacillus amyloliquefaciens isolated from yaks in the Tibet area to provide upper-edge strain resources for probiotics development. The four strains isolated from the intestine of yaks had been identified as Bacillus amyloliquefaciens after the 16S rRNA sequence. The ethanol, bile salt, and acid tolerance revealed that the isolates had significant tolerance levels. The antibiotics susceptibility assay showed that the strains were sensitive to commonly used antibiotics, while the antibacterial assay prevented the isolates from outperforming five harmful bacteria in terms of antibacterial potency. Moreover, it was evident that strain BA5 had the strongest activity to scavenge hydroxyl radical and reduce power. According to the animal experiment, no apparent pathological change was observed in intestinal tissue sections. Furthermore, the strain had a positive effect on promoting the development of jejunal villi referred to its safety. The elevation in the optic nerve sheath (ONS) pressure (ONSP) due to microgravity-induced headward fluid shift is the primary hypothesized contributor to SANS. This longitudinal study aims to quantify the axial plane of the optic nerve subarachnoid space area (ONSSA), which is filled with cerebrospinal fluid (CSF) and expands with elevated ONSP during and after head-down tilt (HDT) bed rest (BR). 36 healthy male volunteers (72 eyes) underwent a 90-day strict 6° HDT BR. Without obtaining the pre-HDT data, measurements were performed on days 30, 60, and 90 during HDT and at 6 recovery time points extended to 180-days (R + 180) in a supine position. Portable B-scan ultrasound was performed using the 12 MHz linear array probe binocularly. The measurements of the ONS and the calculation of the ONSSA were performed with ImageJ 1.51 analysis software by two experienced observers in a masked manner. Compared to R + 180, the ONSSA on HDT30, HDT60, and HDT90 exhibited a consistently significant distention of 0.44 mm2 (95% CI: 0.13 to 0.76 mm2, P = 0.001), 0.45 mm2 (95% CI: 0.15 to 0.75 mm2, P = 0.001), and 0.46 mm2 (95% CI: 0.15 to 0.76 mm2, P < 0.001), respectively, and recovered immediately after HDT on R + 2. Such small changes in the ONSSA were below the lateral resolution limit of ultrasound (0.4 mm) and may not be clinically relevant, possibly due to ONS hysteresis causing persistent ONS distension. Future research can explore advanced quantitative portable ultrasound-based techniques and establish comparisons containing the pre-HDT measurements to deepen our understanding of SANS. Trichoderma harzianum is a well-known biological control agent (BCA) that shows great potential in controlling many pathogenic fungi. To screen for genes associated with mycoparasitism, we sequenced and analyzed the transcriptome of T. harzianum T4 grown in dual culture with Colletotrichum musae. We analyzed differentially expressed genes (DEGs) of Trichoderma harzianum T4 in three different culture periods: before contact (BC), during contact (C) and after contact (AC). A total of 1453 genes were significantly differentially expressed compared to when T. harzianum T4 was cultured alone. During the three periods of double culture of T. harzianum T4 with C. musae, 74, 516, and 548 genes were up-regulated, respectively, and 11, 315, and 216 genes were down-regulated, respectively. The DEGs were screened using GO and KEGG enrichment analyses combined with differential expression multiples. Six gene categories related to mycoparasitism were screened: (a) pathogen recognition and signal transduction, (b) hydrolases, (c) ribosomal proteins and secreted proteins, (d) multidrug-resistant proteins and transporters, (e) heat shock proteins and detoxification, and (f) oxidative stress and antibiotics-related genes. The expression levels of 24 up-regulated genes during T. harzianum T4’s antagonistic interaction with C. musae were detected via real-time fluorescence quantitative PCR (RT-qPCR). This study provided information on the transcriptional expression of T. harzianum T4 against C. musae. These results may help us to further understand the mechanism of mycoparasitism, which can provide a potential molecular target for improving the biological control capacity of T. harzianum T4. Alpha-fetoprotein (AFP) is inextricably linked to various diseases, including liver cancer. Thus, detecting the content of AFP in biology has great significance in diagnosis, treatment, and intervention. Motivated by the urgent need for affordable and convenient electronic sensors in the analysis and detection of aqueous biological samples, we combined the solution-gated graphene transistor (SGGT) with the catalytic reaction of enzyme nanoprobes (HRP-AuNPs-Ab2) to accurately sense AFP. The SGGT immunosensor demonstrated high specificity and stability, excellent selectivity, and excessive linearity over a range of 4 ng/mL to 500 ng/mL, with the lower detection limit down to 1.03 ng/mL. Finally, clinical samples were successfully detected by the SGGT immunosensor, and the results were consistent with chemiluminescence methods that are popular in hospitals for detecting AFP. Astrophysical very high energy (VHE) gamma-rays (with energies >30 GeV) are believed to result almost exclusively from the interactions of populations of highly relativistic particles with ambient matter or photon fields. The study of these VHE photons therefore allows us to examine the processes of particle acceleration in the Universe, and the extreme environments in which they occur. Gamma-rays are also highly directional, making them useful for studying the origin, acceleration, propagation, and composition of cosmic rays. This study uses the Monte Carlo method to simulate the process of extensive air showers being initiated when primary incident γ -rays with energies ranging from 10 GeV to 5 TeV interact with the atmosphere. The ultimate goal is to investigate and analyze the properties and lateral density distributions of Cherenkov light at different altitudes. The simulation considers various energies and zenith angles. The results show that a hump in the lateral density distribution of Cherenkov light is a distinctive feature of a γ -initiated shower, with the hump becoming less prominent at higher energy levels and altitudes. These significant findings not only facilitate better fitting of experimental data obtained from actual extensive air showers but also assist with estimating readings from detectors, thus advancing our understanding of these processes. Endophytic fungi play an important role in the growth and development of traditional Chinese medicine plants. We isolated a strain of Acrocalymma vagum from the endophytic fungi of the traditional Chinese plants Paris. To accurately identify this endophytic fungal species of interest, we sequenced the mitochondrial genome of A. vagum, which is the first discovered mitochondrial genome in Massarineae. The A. vagum mitochondrial genome consists of a 35,079-bp closed circular DNA molecule containing 36 genes. Then, we compared the general sequence characteristics of A. vagum with those of Pleosporales, and the second structure of the 22 tRNAs was predicted. The phylogenetic relationship of A. vagum was constructed using two different data sets (protein-coding genes and amino acids). The phylogenetic tree shows that A. vagum is located at the root of Pleosporales. The analysis of introns shows that the number of introns increases with the increase in branch length. The results showed that monophyly was confirmed for all families in Pleosporales except for Pleosporaceae. A. vagum is an ancient species in the Pleosporales, and Pleosporaceae may require further revision. Two-dimensional (2D) XY ferromagnets have drawn pronounced interest in recent years, but the characteristic of easy-plane magnetization restricts their application in spintronics to some extent. Here, we propose a general strategy for constructing multiferroic van der Waals heterostructures, aiming to achieve electrical control over the magnetic anisotropy in 2D XY ferromagnets. The validity of this strategy is verified by the heterostructure composed of ferromagnetic VBi2Te4 and ferroelectric In2Se3 monolayers. By manipulating the polarized states of In2Se3, the VBi2Te4 can be reversibly transformed between 2D XY and Heisenberg ferromagnets, characterized by the switching of easy magnetization axis between in-plane and out-of-plane directions. More interestingly, accompanied by the changes in magnetic anisotropy, the VBi2Te4 also demonstrates a phase transition from a semiconductor to a half-metal state, which can be ascribed to the band alignment and interfacial charge transfer. The switchable magnetic and electronic properties enable the heterostructure to be utilized in nonvolatile memory and logic devices. Additionally, the half-metallicity and magnetocrystalline anisotropy energy of the heterostructure can be effectively tuned by biaxial strain. These findings not only pave the way for electrically nonvolatile control of 2D XY ferromagnet, but also facilitate the development of interfacial magnetoelectric physics and applications. The underlying mechanism of chronic hepatitis B virus (HBV) functional cure by interferon (IFN), especially in patients with low HBsAg and/or young ages, is still unresolved due to the lack of surrogate models. Here, we generate a type I interferon receptor humanized mouse (huIFNAR mouse) through a CRISPR/Cas9-based knock-in strategy. Then, we demonstrate that human IFN stimulates gene expression profiles in huIFNAR peripheral blood mononuclear cells (PBMCs) are similar to those in human PBMCs, supporting the representativeness of this mouse model for functionally analyzing human IFN in vivo. Next, we reveal the tissue-specific gene expression atlas across multiple organs in response to human IFN treatment; this pattern has not been reported in healthy humans in vivo. Finally, by using the AAV-HBV model, we test the antiviral effects of human interferon. Fifteen weeks of human PEG-IFNα2 treatment significantly reduces HBsAg and HBeAg and even achieves HBsAg seroconversion. We observe that activation of intrahepatic monocytes and effector memory CD8 T cells by human interferon may be critical for HBsAg suppression. Our huIFNAR mouse can authentically respond to human interferon stimulation, providing a platform to study interferon function in vivo. PEG-IFNα2 treatment successfully suppresses intrahepatic HBV replication and achieves HBsAg seroconversion. There is increasing evidence that treatment of hepatitis B with interferon alpha can be beneficial. Regional and persistent PM2.5 pollution seriously undermines the development of urban ecological civilizations and the advancement of high-quality economies. The producer service sector, an example of a typical knowledge-intensive service industry, plays an important role in advancing the manufacturing industry and fostering economic growth while concurrently improving urban environmental conditions. Based on panel data of prefecture-level cities in the Yellow River Basin from 2006 to 2019, this study constructed a Spatial Durbin Model and a mediation effect model to comprehensively explore the impact of producer services agglomeration on PM2.5 pollution. The main conclusions are as follows: (1) From 2006 to 2019, PM2.5 pollution in the study area exhibited an initial rise followed by a subsequent decline, with notable spatial heterogeneity. PM2.5 pollution in the lower reaches of the Yellow River was significantly higher than in the middle and upper reaches. In addition, the spatial pattern of producer services agglomeration showed distinct "core-edge" characteristics. (2) The agglomeration of producer services had a significant negative impact on local and adjacent PM2.5 pollution, and there was a more pronounced haze reduction effect in the case of specialized agglomerations of producer services and low-end producer services. (3) The agglomeration of producer services indirectly improved PM2.5 pollution by promoting technological innovation and optimizing industrial structure, with the latter playing a greater mediating effect. This study not only helps expand the theoretical and empirical research on producer services agglomeration but also offers valuable insights for pursuing a green transformation of the Yellow River Basin by optimizing industrial patterns through the producer services sector. Background: BMP7 has been shown to have neuroprotective effects and to alleviate demyelination. However, its role in trigeminal neuralgia (TN) has not been well investigated. The current study aims to determine whether BMP7 plays a role in demyelination, its effects on pain behaviors and mechanism of action in rats with TN. Methods: We used an infraorbital-nerve chronic-constriction injury (ION-CCI) to establish a rat model of TN. Adeno-associated viruses (AAVs) were injected into the rats to upregulate or downregulate BMP7. The mechanical withdrawal thresholds (MWT) of the injured rats were detected using Von Frey filaments. The changes in expression levels of BMP7 and oligodendrocyte (OL) markers were examined by western blotting, quantitative real-time PCR, immunofluorescence, and transmission electron microscopy. Results: The ION-CCI induced mechanical allodynia, demyelination, and loss of OLs with a reduction of BMP7. Short-hairpin RNA (shRNA)-BMP7 that inhibited BMP7 expression also caused mechanical allodynia, demyelination, and loss of OLs, and its mechanism may be OL apoptosis. Overexpressing BMP7 in the trigeminal spinal subnucleus caudalis(VC) with AAV-BMP7 relieved all three phenotypes induced by the CCI, and its mechanism may be alleviating OLs apoptosis. Two signal pathways associated with apoptosis, STAT3 and p65, were significantly downregulated in the VC after CCI and rescued by BMP7 overexpression. Conclusion: BMP7 can alleviate TN by reducing OLs apoptosis and subsequent demyelination. The mechanism behind this protection could be BMP7-mediated activation of the STAT3 and NF-κB/p65 signaling pathway and subsequent decrease in OL apoptosis. Highlights: Heterostructure engineering is proposed to construct CoTe2/ZnTe heterostructures with built-in electric field. Conductive and elastic Ti3C2Tx MXene is introduced to improve the conductivity and alleviate the volume change of CoTe2/ZnTe upon cycling. The resulting CoTe2/ZnTe/Ti3C2Tx (CZT) demonstrates outstanding rate capability (137.0 mAh g−1 at 10 A g−1) and cycling stability (175.3 mAh g−1 after 4000 cycles at 3.0 A g−1). Moreover, the CZT-based full cells demonstrate excellent energy density (220.2 Wh kg−1) and power density (837.2 W kg−1). Exploiting high-rate anode materials with fast K+ diffusion is intriguing for the development of advanced potassium-ion batteries (KIBs) but remains unrealized. Here, heterostructure engineering is proposed to construct the dual transition metal tellurides (CoTe2/ZnTe), which are anchored onto two-dimensional (2D) Ti3C2Tx MXene nanosheets. Various theoretical modeling and experimental findings reveal that heterostructure engineering can regulate the electronic structures of CoTe2/ZnTe interfaces, improving K+ diffusion and adsorption. In addition, the different work functions between CoTe2/ZnTe induce a robust built-in electric field at the CoTe2/ZnTe interface, providing a strong driving force to facilitate charge transport. Moreover, the conductive and elastic Ti3C2Tx can effectively promote electrode conductivity and alleviate the volume change of CoTe2/ZnTe heterostructures upon cycling. Owing to these merits, the resulting CoTe2/ZnTe/Ti3C2Tx (CZT) exhibit excellent rate capability (137.0 mAh g−1 at 10 A g−1) and cycling stability (175.3 mAh g−1 after 4000 cycles at 3.0 A g−1, with a high capacity retention of 89.4%). Objective: The incidence of hyperlipidemic acute pancreatitis (HLAP) has rapidly increased in recent years in China. Autophagy has been implicated in the inflammatory response of pancreatic cells in HLAP, but the molecular mechanisms remain unclear. Methods: In this study, the role of HIF-1α–PPARγ–mTORC1 pathway-mediated autophagy in the inflammatory response of pancreatic cells and the underlying molecular mechanism were investigated in a rat model of HLAP using immunohistochemistry, ELISA, electron microscopy, and western blot analysis. Results: The results revealed that autophagy was significantly increased and pancreatic injury was exacerbated in HLAP rats, and the inflammatory response was further exacerbated by treatment with rapamycin but relieved by treatment with 3-MA. Hyperlipidemia induced upregulation of HIF-1α and downregulation of PPARγ, which in turn led to an increase in autophagy and consequently exacerbation of the inflammatory response of pancreatic cells. Conclusions: HIF-1α–PPARγ–mTORC1 pathway-mediated autophagy plays a critical role in the inflammatory response of pancreatic cells in HLAP, and interference with the HIF-1α–PPARγ–mTOR pathway can serve as a new strategy for the prevention and treatment of HLAP. Excessive activation of pro-inflammatory (M1) microglia phenotypes after spinal cord injury (SCI) disrupts tissue repair and increases the risk of secondary SCI. We previously reported that adeno-associated virus (AAV) mediated delivery of bone morphogenetic protein 7 (BMP7) promotes functional recovery after SCI by reducing oligodendrocyte loss and demyelination; however, little is known about the early effects of BMP7 in ameliorating neuroinflammation in the acute SCI phase. Herein, we demonstrate that treatment with recombinant human BMP7 (rhBMP7) suppresses the viability of LPS-induced HMC3 microglia cells and increases the proportion with the M2 phenotype. Consistently, in a rat SCI model, rhBMP7 decreases the activation of microglia and promotes M2 polarization. After rhBMP7 administration, the STAT3 signaling pathway was activated in LPS-induced HMC3 cells and microglia in spinal cord lesions. Furthermore, the levels of TNF-α and IL-1β were significantly decreased in cell culture supernatants, lesion sites of injured spinal cords, and cerebrospinal fluid circulation after rhBMP7 administration, thus reducing neuron loss in the injured spinal cord and promoting functional recovery after SCI. Dual topological insulator (DTI), which simultaneously hosts topological insulator (TI) and topological crystalline insulator (TCI) phases, has attracted extensive attention since it has a better robustness of topological nature and broad application prospects in spintronics. However, the realization of DTI phase in two-dimensional (2D) system is extremely scarce. By first-principles calculations, we predict that the 2D rectangular bismuth (R—Bi) bilayer is a novel DTI, featured by ℤ2 topological invariant ℤ2 = 1, mirror Chern number CM = −1, and metallic edge states within the bulk band gap. More interestingly, the TCI phase in bilayer is protected by horizontal glide mirror symmetries, rather than the usual mirror symmetry. The bulk band gap can be effectively tuned by vertical electric field and strain. Besides, the electric field can trigger the transition between TI and metallic phases for the bilayer, accompanied by the annihilation of TCI phase. On this basis, a topological field effect transistor is proposed, which can rapidly manipulate spin and charge carriers via electric field. The KBr(110) surface is demonstrated as an ideal substrate for the deposition of bilayer. Nanocellulose-based antibacterial materials, which hold excellent biocompatibility and dispersion stability, are attractive candidates to replace traditional antibacterial materials in blocking the spread of bacteria. However, environmentally polluting and complex preparation processes hinder the development of antibacterial nanocellulose. Herein, we propose a simple and green strategy for extracting natural antibacterial cellulose-based nanofibers (P-CNFs) from pine cones by directly thermo-oxidizing natural pine cones with H2O2. The obtained nanofibers possess strong antibacterial properties against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) by residual lignin and rosin. Furthermore, the minimum inhibitory concentration (MIC) of E. coli and S. aureus was as low as 1.5 and 2 mg mL−1, respectively. More surprisingly, benefiting from the residual lignin and rosin, P-CNFs could be well dispersed in ethanol. P-CNF/ethanol suspension could be regarded as a disinfectant with long residual action, which could resist the secondary infection of bacteria. The green preparation and excellent properties of P-CNFs made them viable candidates as environmentally friendly antibacterial materials. Space-filling designs are widely used in computer experiments. They are frequently evaluated by the orthogonality and distance-related criteria. Rotating orthogonal arrays is an appealing approach to constructing orthogonal space-filling designs. An important issue that has been rarely addressed in the literature is the design selection for the initial orthogonal arrays. This paper studies the maximin L2-distance properties of orthogonal designs generated by rotating two-level orthogonal arrays under three criteria. We provide theoretical justifications for the rotation method from a maximin distance perspective and further propose to select initial orthogonal arrays by the minimum G2-aberration criterion. New infinite families of orthogonal or 3-orthogonal U-type designs, which also perform well under the maximin distance criterion, are obtained and tabulated. Effective protection against X-ray is the premise of utilizing the X-ray, thus it is critical to develop novel X-ray shielding materials with both low density and high X-ray attenuation efficiency. As the even distribution of high-Z element components is of great significance for increasing the attenuation efficiency of X-ray shielding materials, in this study, the microfiber membrane (MFM), a type of synthetic leather featuring hierarchical structure was chosen to provide large surface area for the dispersion of rare earth (RE) element. Meanwhile, plant polyphenol was utilized to achieve the stable loading and uniform dispersion of the Ce or Er into MFM. Benefiting from the assistance of polyphenol and hierarchical structure of MFM, the even dispersion of RE element was successfully realized. The resultant shielding materials displayed approximately 10% superior X-ray attenuation efficiency compared to that without polyphenol, and an averagely 9% increment in X-ray attenuation efficiency than that without hierarchical structure. Moreover, the obtained composite with a thickness of 2.8 mm displayed superior X-ray shielding performance compared to 0.25 mm lead sheet in 16–83 keV and retained an ultralow density of 1.4 g cm–3. To identify the distribution pattern of macrofaunal assemblages of the Dafeng intertidal flats in response to hydrodynamic and sediment dynamic processes in the northern Jiangsu coast, East China, macrofauna sampling and hydrodynamic observations were carried out simultaneously across the mud flat, mixed mud-sand flat, and silt-sand flat of the intertidal zone in June 2018. Results show that there was a clear zonal distribution pattern of the macrofaunal communities, as is controlled by local hydrological and sedimentary environments. Principal component analysis (PCA) revealed three types of intertidal area in terms of hydrological and surficial sediment parameters. Similarly, three distinct groups of the macrofaunal communities, i.e., mud flat, mix mud-sand, and silt-sand groups, were recognized at similarity level of 24% based on the CLUSTER analysis in similarity profile (SIMPROF) test. Correlation analysis upon best variables stepwise search (BVSTEP) indicated the importance of the hydrodynamics (e.g., water temperature and salinity, tidal duration, flow speed, suspended sediment concentration, and wave height) in the differentiation of macrofaunal communities with different taxonomic classes over the intertidal zone. Therefore, macrofaunal assemblages, similar to hydrology and surficial sediment, have a unique zonation pattern. Small-sized deposit feeders adapt better to low energy environments, thus dominated the upper part of the intertidal flat, whilst the heavy and large-sized filter feeders and deposit feeders were dominant over the middle and lower parts. A single feature cannot show the operational state of a bearing during its entire life cycle. Therefore, a rolling bearing performance deterioration prediction method based on an SAE and the TCN-attention model is proposed. The SAE method is used to fuse the time-domain indicator and the frequency-domain indicator to construct the performance degradation characteristic indicator. The evaluation indices are used to comprehensively evaluate multiple performance degradation indices, and the fused feature indices together, to filter out the features that have a good overall performance. Attention is added to the TCN model, and the output state weight of the TCN model is calculated through a scoring function to increase the important information weight and the prediction accuracy. The appropriate network structure and parameter configuration are determined, and the rolling bearing performance degradation prediction model is established. A validation is performed using publicly available datasets from the University of Cincinnati and XJTU-SY. The results show that the method is more sensitive to the critical information part of the long time series than the other models. At the same time, the average absolute error and the root mean square error are minimized, the accuracy of the rolling bearing performance degradation prediction is high, and the model has a strong robustness and generalization abilities. This study establishes an economic financial support model for the improvement of environmental quality through the development rural revitalization based on the extended Cobb–Douglas production function. Using statistical data from Shaanxi Province, China, from 2004 to 2019, a vector autoregressive (VAR) model is used to empirically analyze the development effect of financial support for rural revitalization and to give the focus points and optimization paths for financial support for environmental quality, rural revitalization and sustainable development. The research results show that financial support plays an active and long-term role in improving environmental quality and promoting rural revitalization and sustainable development. Specifically, the effect of financial instruments in supporting rural revitalization and sustainable development is continuous. Clonal hematopoiesis is a prevalent disease associated with all-cause death. Not only because it can be a precancerous lesion of blood system diseases but also has a strong association with cardiovascular disease. A narrow term, clonal hematopoiesis of indeterminate potential (CHIP), was proposed by Steensma et al. [1] to describe individuals with detectable somatic clonal mutations in their genes in blood or bone marrow but without a diagnosis of hematological disease or unexplained cytopenia. Recently, studies have suggested that CHIP is associated with adverse cardiovascular disease progression, particularly in patients with ten-eleven translocation 2 (TET2) mutations or DNA methyltransferase 3 alpha (DNMT3A) mutations. Age is the most crucial factor which is associated with increased CHIP prevalence. The underlying mechanisms appear to be related to inflammatory status. However, new evidence suggests that genetic factors, lifestyle and environmental factors such as smoking, obesity, and diet also play essential roles in developing CHIP. More research needs to be done on the potential genetic mechanisms driving CHIP and the environmental factors that modulate CHIP risk. This review summarizes the latest research on CHIP, discusses in detail the strong association between clonal hematopoiesis and accelerated cardiovascular disease, and rationalizes the intervention of CHIP in combination with existing evidence, which may be beneficial for future treatment. SARS-CoV-2 Omicron variants feature highly mutated spike proteins with extraordinary abilities in evading antibodies isolated earlier in the pandemic. Investigation of memory B cells from patients primarily with breakthrough infections with the Delta variant enables isolation of a number of neutralizing antibodies cross-reactive to heterologous variants of concern (VOCs) including Omicron variants (BA.1-BA.4). Structural studies identify altered complementarity determining region (CDR) amino acids and highly unusual heavy chain CDR2 insertions respectively in two representative cross-neutralizing antibodies—YB9-258 and YB13-292. These features are putatively introduced by somatic hypermutation and they are heavily involved in epitope recognition to broaden neutralization breadth. Previously, insertions/deletions were rarely reported for antiviral antibodies except for those induced by HIV-1 chronic infections. These data provide molecular mechanisms for cross-neutralization of heterologous SARS-CoV-2 variants by antibodies isolated from Delta variant infected patients with implications for future vaccination strategy. With the acceleration of urbanization, domestic waste has become one of the most inevitable factors threatening the environment and human health. Waste classification is of great significance and value for improving urban environmental quality and promoting human well-being. Based on the theory of planned behavior, we added external and socio-economic factors to systematically examine how they affect residents' waste classification behavior (WCB). We collected 661 valid data through a questionnaire survey conducted in Jinan, a pilot city for waste classification in China. Key driving factors were identified by combining binary logistic regression and the principal component analysis. The results showed that the elderly, women, and people with higher education are more likely to participate in waste classification. Attitude, collaborative governance, and institutional pressure positively affect WCB, while subjective norm and infrastructure have a negative effect. Knowledge mastery and degree of publicity are positively and significantly related to WCB, but other perceived behavioral control sub-variables negatively affect WCB. Based on the results and status of waste classification in Jinan, we propose the multi-agent linkage governance pattern from various dimensions to explore a powerful guiding incentive that can enhance WCB and provide a reference for waste management policymakers. The rupture of atherosclerosis (AS) vulnerable plaque is the major cause of fatal arterial thrombosis related diseases. The lipid dysbolism and pro-inflammatory microenvironment are considered two main driving factors for plaque rupture. Herein, a "two-pronged" nanosystem LCS-Se/Res was designed to efficiently stabilize AS plaques, by precisely regulating lipid metabolism and inflammatory microenvironment. LCS-Se/Res was constructed with the core of resveratrol (Res) loaded selenium (Se) nanoparticles (NPs), and the shell (LCS) of targeting peptide (LSIPPKA) modified chitosan (CS). This system was stable in blood circulation. When it arrived at the plaque site, LCS-Se/Res could actively recognize the highly expressed lectin-like oxidized low-density lipoprotein receptor 1 (LOX-1) receptor and accumulate there. Thereafter, Res was released from LCS-Se nanocarriers in situ by responding to acidic plaque microenvironment. On the one hand, Res increased autophagy flux in damaged endothelial cells by activating cAMP-PKA-AMPK-SIRT1 signaling pathway to promote lipid degradation. Interestingly, LCS-Se also promoted cholesterol efflux to improve lipid metabolism. On the other hand, fixed-point separated Res and LCS-Se synergistically improved plaque inflammatory microenvironment by reversing macrophage phenotype (M1 to M2) and alleviating plaque oxidative stress. Pharmacodynamics result proved that the plaque vulnerability index (VI) decreased from 1.39 ± 0.282 to 0.108 ± 0.02 after LCS-Se/Res treatment. In China, exploring the relationship between human capital and green economy in the context of economic transformation can promote the construction of an ecological civilization and high-quality economic development. This paper is dedicated to assessing the impact of human capital stock, human capital structure, and the difference between the two on China's green economy. Using the super-efficiency slack-based measure (SBM) model, this paper evaluated the green total factor productivity (GTFP) of 30 Chinese provinces to assess the greenness of its economy and analyzed its spatiotemporal evolution from 2000 to 2017. A spatial error model (SEM) was established to explore the impact of human capital on green economy. The results show that (1) the GTFP of China increased continuously from 0.219 to 0.457, showing a regional spatial differentiation of "eastern region > central region > western region." The spatial gaps of the green economy in the east-west and north-south directions are narrowing. (2) Moran's I index and Moran's I scatter plot indicate a significant spatial correlation between human capital and China's green economy. The local spatial correlation between human capital and green economy is mainly characterized by "high-high" and "low-low" types of agglomeration. (3) The effects of human capital stock and human capital structure on green economy were both positive, with coefficients of 0.0005 and 0.1601, respectively, but the effects of human capital structure were not significant. (4) The results of regional regression show that the difference between the human capital impact coefficients on green economy in the eastern and midwestern regions is small. The impact of human capital stock and human capital structure on green economy is consistent with the national level results. It can be preliminarily concluded that the development of China's green economy relied more on improving the human capital stock than on the improvement of the human capital structure. Background: Extrinsic molecular mechanisms that regulate hematopoietic stem/progenitor cell (HSPC) aging are still poorly understood, and a potential protective medication needs to be explored. Materials and methods: The senescent parameters of hematopoietic cells and bone marrow stromal cells (BMSCs) including cell cycle analysis, senescence-associated SA-β-gal staining and signals, hematopoietic factors and cellular junction were analyzed in femur and tibia of rats. Furthermore, Sca-1+ HSPCs and BMSCs co-culture system was established to evaluate the direct effects of BMSC feeder layer to HSPCs. Oxidative DNA damage indicators in Sca-1+ HSCs and senescence-associated secretory phenotype (SASP) of BMSCs, gap junction intercellular communication between BMSCs, osteogenesis/adipogenisis differentiation balance of BMSCs were detected. Results: In the D-gal pre-administrated rats, ASP treatment rescued senescence of hematopoietic cells and BMSCs, reserved CFU-GEMM; also, ASP treatment attenuated stromal oxidative load, ameliorated SCF, CXCL12, and GM-CSF production, increased Connexin-43 (Cx43) expression. BMSCs and Sca-1+ HSPCs co-cultivation demonstrated that ASP treatment prevented oxidative DNA damage response in co-cultured Sca-1+ HSPCs induced by D-gal pre-administration of feeder layer and the underlying mechanism may be related to ASP ameliorating feeder layer dysfunction due to D-gal induced senescence via inhibiting secretion of IL-1, IL-6, TNF-α, and RANTES, enhancing Cx43-mediated intercellular communication, improving Runx2 expression whereas decreasing PPARγ expression in BMSCs. Single cell analysis is crucial for elucidating cellular diversity and heterogeneity as well as for medical diagnostics operating at the ultimate detection limit. Although superbly sensitive biosensors have been developed using the strongly enhanced evanescent fields provided by optical microcavities, real-time quantification of intracellular molecules remains challenging due to the extreme low quantity and limitations of the current techniques. Here, we introduce an active-mode optical microcavity sensing stage with enhanced sensitivity that operates via Förster resonant energy transferring (FRET) mechanism. The mutual effects of optical microcavity and FRET greatly enhances the sensing performance by four orders of magnitude compared to pure Whispering gallery mode (WGM) microcavity sensing system. We demonstrate distinct sensing mechanism of FRET-WGM from pure WGM. Predicted lasing wavelengths of both donor and acceptor by theoretical calculations are in perfect agreement with the experimental data. The proposed sensor enables quantitative molecular analysis at single cell resolution, and real-time monitoring of intracellular molecules over extended periods while maintaining the cell viability. By achieving high sensitivity at single cell level, our approach provides a path toward FRET-enhanced real-time quantitative analysis of intracellular molecules. Chlorpyrifos (CPF), a common organophosphorus pesticide, is extensively used in agricultural practices. However, we lack sound evidence for the linkage between soil microbial diversity, soil function, and plant biomass under the influence of CPF, which prevents us from assessing the actual impact of CPF on agricultural production. In this study, we used high-throughput sequencing to test the effects of CPF on soil microbial diversity, soil function, and cotton biomass in indoor pot experiments. The use of CPF leads to a significant reduction in cotton biomass until the concentration of CPF used reaches 15 mg kg−1, and the cotton biomass is no longer significantly reduced. Compared with the original soil, the alpha-diversity of bacteria, which was significantly linearly related to cotton biomass, was significantly decreased when the soil was treated with 15 mg kg−1 CPF. Affected by CPF, the overall soil microbial composition has changed significantly. Acidobacteria, Nitrospirae, Planctomycetes, and Actinobacteria were significantly regulated after CPF treatment. Correspondingly, key soil functions, including nitrogen metabolism and iron (III) transporter, have been significantly down-regulated. The reduction of nitrogen and Fe3+ should deprive the cotton of essential nutrients during the short crop cycle and thus affect cotton biomass. Our study provides experimental evidence that CPF affects cotton rhizosphere soil microbial diversity, the relative content of key bacterial genera, and soil function, which shows that it has an important impact on plant biomass, and provides a reference for studying the actual impact of CPF on the environment and agricultural production. Electrically controlled half-metallicity in antiferromagnets is of great significance for both fundamental research and practical application. Here, by constructing van der Waals heterostructures composed of two-dimensional (2D) A-type antiferromagnetic NiI2 bilayer (bi-NiI2) and ferroelectric In2Se3 with different thickness, we propose that the half-metallicity is realizable and switchable in the bi-NiI2 proximate to In2Se3 bilayer (bi-In2Se3). The polarization flipping of the bi-In2Se3 successfully drives transition between half-metal and semiconductor for the bi-NiI2. This intriguing phenomenon is attributed to the joint effect of polarization field-induced energy band shift and interfacial charge transfer. Besides, the easy magnetization axis of the bi-NiI2 is also dependent on the polarization direction of the bi-In2Se3. The half-metallicity and magnetic anisotropy energy of the bi-NiI2 in heterostructure can be effectively manipulated by strain. These findings provide not only a feasible strategy to achieve and control half-metallicity in 2D antiferromagnets, but also a promising candidate to design advanced nanodevices. Hematopoietic stem cells show biological manifestations of aging, diminished hematopoietic function and abnormal differentiation, which can lead to leukemia. It is therefore important to explore the mechanism underlying hematopoietic stem cell aging to develop strategies for delaying the process. Our evaluations revealed that the number of bone marrow hematopoietic cells (BMHCs) started to decrease significantly after 45 years of age, and the number of senescent BMHCs, as determined by senescence-associated beta-galactosidase staining, gradually increased with age. In addition, BMHCs from individuals over 45 years of age presented with notably reduced proliferative capacity, increased G1-phase cell cycle arrest, and significantly decreased generation of mixed colony forming units, which suggests that BMHCs enter senescence during middle age. Furthermore, we observed significantly lower antioxidant capacity and a significant increase in oxidative damage products, a gradual increase in the expression of senescence-associated proteins and genes, and a gradual decrease in the expression of cell cycle related proteins in BMHCs after middle age. Taken together, these findings offer both a theoretical and experimental basis for better understanding of the senescence progression of BMHCs and the optimal timing for anti-senescence drug interventions in clinical practice. The existence of an unreasonable residual stress state in the ring blank forgings of thin-walled casings harms the dimensional stability and service life of the parts. Stress control of rolled rings is achieved by using a segmental mandrel bulging process, the effectiveness of which is closely related to the process parameters. The objective of this investigation was to enhance the effectiveness of the bulging in controlling the residual stresses during the manufacturing process. The residual stress evolution mechanism for the bulging process was analyzed. The residual stress results obtained from response surface experimental procedures were established to relate the empirical relationship between each process parameter and the stress response. A quadratic regression model based on an analysis of variance was developed, and the validity of its response parameters was verified. The interactions between dopamine hydrochloride (DH) and vitamin B6 hydrochloride (VH) in aqueous solution were investigated by means of cyclic voltammetry (CV), 1H NMR, density functional theory (DFT), quantum theory of atoms in molecules (QTAIM) and reduced density gradient (RDG). The results from CV and NMR experiments indicate the π–π stacking between the benzene ring of DH and the pyridine ring of VH, which is confirmed by theoretical calculations. Furthermore, six DH-VH complexes obtained by geometry optimization contain hydrogen bonds, π–π stacking and C–H–π interactions, and π–π stacking play a vital role in DH–VH–H2O system. Despite timely immunization programs, and efficacious vaccines conveying protection against SARS-CoV-2 infection, breakthrough infections in vaccinated individuals have been reported. The Delta variant of concern (VOC) outbreak in Guangzhou resulted in local transmission in vaccinated and non-vaccinated residents, providing a unique opportunity to study the protective effects of the inactivated vaccines in breakthrough infection. Here, we find that the 2-dose vaccinated group has similar peak viral titers and comparable speeds of viral RNA clearance to the non-vaccinated group but accelerated viral suppression in the middle course of the disease. We quantitatively demonstrate that peak viral pneumonia is significantly mitigated in the 2-dose vaccine group (median 0.298%) compared with the non-vaccinated (5.77%) and 1-dose vaccine (3.34%) groups. Pneumonia absorbance is approximately 6 days ahead in the 2-dose group (median 10 days) than in the non-vaccinated group (16 days) (p = 0.003). We also observe reduced cytokine inflammation and markedly undisturbed gene transcription profiles of peripheral blood mononuclear cells (PBMCs) in the 2-dose group. In short, our study demonstrates that prior vaccination substantially restrains pneumonia development, reduces cytokine storms, and facilitates clinical recovery. The purpose of this paper is to explore the lubrication of graphite of different morphology in Al-Si alloys, especially at high temperatures.In this work, aluminum-silicon alloys with different graphite particles are prepared by powder metallurgy, and the effect of graphite morphology on friction and wear behaviors are investigated under a hyperthermal environment. The volumetric wear loss results denote that Al-27Si with faked graphite composites perform superior wear resistance than Al-27Si and Al-27Si with particle graphite composites under all test temperatures points. Due to the weak bonding of particle graphite, Al-27Si with particle graphite has the most volume wear loss. When the temperature rises to 400 ℃, the friction coefficient of all the samples has a strong wave with big fluctuation because of aluminum matrix soften and vibration. In this work, to make up for the lack of performance at high temperatures, the Al-27Si alloy was prepared successfully, which makes it possible to evaluate the required performance of the engine. Uniform designs have been widely used in physical and computer experiments due to their robust performances. The level permutation method can efficiently construct uniform designs with both lower discrepancy and less aberration. However, the related existing literature has mostly discussed uniform fixed-level designs, the construction of uniform mixed-level designs has been quite few studied. In this paper, a novel level permutation method for constructing uniform mixed-level designs is proposed. Our main idea is to perform level permutations on a new class of designs, called minimum average discrepancy designs, rather than generalized minimum aberration designs as in the fixed-level case. Several theoretical results on the design optimality and construction are obtained. Numerical results suggest the good performance of the resulting designs under various popular discrepancies. Key messages: Genetic analysis revealed that CmCLV3 is a candidate gene for the variation in melon carpel number. Carpel number (CN) is an important trait in melon. Three-CN melon fruit is oval, while 5-CN melon fruit has a round or flat shape. Herein, a genetic analysis of a population in which the CN locus was segregated indicated that 3-CN is controlled by a major dominant effective gene. Bulked segregant analysis and initial linkage mapping placed the CN locus in a 6.67 Mb region on chromosome 12, and it was narrowed to 882.19 kb with molecular markers and recombinant plants. Fine mapping with a large F2 population containing 1026 individuals further narrowed the locus to an 83.98 kb region harboring five annotated genes. Gene structure alignment between the parental lines revealed MELO3C035640.2 (annotated as CLAVATA3, CmCLV3) as the best candidate gene for the CN trait. CmCLV3 was more highly expressed in 3- than 5-CN lines and specifically expressed in terminal buds rather than in young leaves, hypocotyls, and roots. The CmCLV3 coding region was cloned from eight 3- or 5-CN melon accessions, and a nonsynonymous SNP site was highly correlated with CN variation. This SNP site was also related to CN variations among 40 melon lines according to their resequencing data, causing a helix alteration in the CmCLV3 protein. Promoter region sequence alignment and activity analysis showed that, unlike in cucumber and tomato, CmCLV3 promoter variation and activity were not the main reasons for CN alteration. Licorice chalcone A (LCA) possesses a variety of pharmacological activities amid poor solubility in water which consequently restricts its clinical application. In this study, LCA micelles (LCA-M) and LCA micelles modified with TPGS (LCA-M-T) were prepared, wherein in vitro, in vivo, and hepatoprotection potential of the formulations were evaluated. LCA-M-T and LCA-M were prepared by thin-film dispersion method and characterized by a transmission electron microscope (TEM). The encapsulation efficiency, in vitro release, and stability of LCA-M-T and LCA-M were detected by HPLC and UV, respectively. The pharmacokinetics, tissue distribution studies, and pharmacodynamics research were also carried out after oral administration. The sizes of micellar particles were 34.02 ± 0.29 nm (LCA-M) and 62.46 ± 0.51 nm (LCA-M-T) with 0.223 ± 0.021 (LCA-M) and 0.343 ± 0.006 (LCA-M-T) polydispersities, − 35.06 ± 2.27 mV (LCA-M) and − 38.58 ± 1.69 mV (LCA-M-T) zeta potentials, and 95.2 ± 0.27% (LCA-M) and 90.34 ± 0.31% (LCA-M-T) encapsulation efficiencies. The drug loadings were 8.56 ± 0.27% (LCA-M) and 8.04 ± 0.19% (LCA-M-T). The two preparations released quickly and reached over 95% in the four-dissolution media (pH = 1.2 pH = 6.8 pH = 7 pH = 7.4). The preparations of LCA showed significant elevation of cumulative release compared with the free LCA and more importantly 119% (LCA-M) and 285% (LCA-M-T) increments in the relative oral bioavailability of the drug.
