We conducted research to develop a safe and effective toxic bait to control the small Indian mongoose (Urva auropunctata), an invasive vertebrate predator impacting the survival of native species in Hawai'i and in other parts of the world. A preserved fish-based bait product was found to be highly palatable to mongooses in cage trials and subsequent formulations with diphacinone (0.005%) showed promise as an efficacious toxic bait for mongooses. This product is intended for future use to control mongooses in conservation and urban areas, and as a biosecurity tool at ports of entry to address accidental introductions into mongoose-free areas. Anticipated delivery of this toxic bait is in tamper-proof bait stations. We designed three prototype bait stations constructed with polyvinyl chloride (PVC) drainage pipes and evaluated their performance in enclosure trials with wild-caught mongooses and in field trials with free-ranging mongooses. A commercially available tamper-resistant rodenticide bait station was also used for comparison to the prototypes in our trials. The goal was to develop a bait station that is readily used by mongooses, allows for bait consumption in place, prevents removal of bait, and restricts access to nontarget animals. We used a non-toxic formulation of the fish-based bait product and monitored bait station use, visitation rates, bait uptake, and spillage. All four bait station types were used by mongooses in the laboratory, and one PVC bait station design and the commercial bait station exhibited multiple mongoose visitations with minimal bait spillage in the field. We did not record any significant nontarget species interactions with the bait during the field trials. The PVC bait station design and commercial bait station are approved methods of bait delivery in the subsequent field efficacy trials under an Experimental Use Permit for the upcoming registration of "Fish-based Bait for Mongooses" with the Environmental Protection Agency. A specific and sensitive liquid chromatography-tandem mass spectrometry method was developed and validated for the determination of the anticoagulant rodenticide diphacinone (DPN) in mouse and rat liver. Tissue samples were extracted with a mixture of water and acetonitrile containing ammonium hydroxide. The extracted sample was cleaned up with a combination of liquid-liquid partitioning and dispersive solid phase extraction. Chromatographic separation was achieved using a Waters X-Bridge BEH C-18 LC column (50 mm, 2.1 mm ID, 2.5 μm particle size) with detection on a triple quadrupole mass spectrometer in multiple reaction monitoring (MRM) mode. The monitored transition for DPN was m/z 339.0 → 167.0 for quantitation and 339.0 → 172.0 and 339.0 → 116.0 for confirmation. The linear range was 0.5 to 375 ng/mL. The average precision of DPN, represented by the relative standard deviation of the observed concentrations, was 7.2% (range = 0.97% - 20.4%) and the average accuracy, represented by the relative error, was 5.8% (range = 1.06% - 14.7%). The recovery of DPN fortified at 3 different levels averaged 106% in rat liver and 101% in mouse liver. The established method was successfully used to determine DPN residue levels in Polynesian rats (Rattus exulans) and mice (Mus musculus) fed two different formulated baits containing DPN. The observed residue levels were consistent with values observed in other rodent studies. However, the amount of bait consumed was lower for the novel baits evaluated in this study. Oral rabies vaccination is the principal strategy used to control rabies in wildlife. No oral rabies vaccine is licensed for small Indian mongooses (Herpestes auropunctatus). The Ontario Rabies Vaccine Bait (ONRAB) is a human adenovirus type-5 rabies glycoprotein recombinant vaccine licensed for rabies control in striped skunks (Mephitis mephitis) in Canada and is under experimental evaluation in the US. We evaluated varying doses of ONRAB vaccine by direct instillation into the oral cavity with three groups of 10 mongooses: Group 1 received 109.5 TCID50, group 2 received 108.8 TCID50, and group 3 received 108.5 TCID50 of vaccine. Six control mongooses were sham-vaccinated with culture media. We collected a serum sample prior to vaccination and on days 14 and 30 postvaccination (PV). We quantified the level of rabies virus neutralizing antibodies (RVNA) from mongoose sera and compared titers among vaccinated groups and time points PV, where values greater than or equal to 0.1 IU/mL were considered positive. On day 14 PV, 87% (26 of 30, 95% confidence interval 70–95%) of vaccinates had seroconverted, whereas all vaccinates demonstrated RVNA by day 30 PV. There was a marginal effect of vaccine dose on group means of log-transformed RVNA titers at day 14 PV (F=2.5, P=0.099), but not day 30 PV. The eradication or control of invasive small Indian mongooses from islands likely requires toxic baiting when removal by trapping proves insufficient. The one toxic bait currently registered for mongooses in the United States has relatively low palatability and efficacy for mongooses. Developing and registering a new pesticide can be very expensive, while funding for developing toxicants for mongooses is limited. Once registered, use of a toxic bait may be hindered by other factors, such as public opposition to an inhumane toxicant, poorer efficacy than expected, or if the toxic bait is difficult for applicators to apply or store. Therefore, we conducted a product feasibility assessment comparing the registration and use potential of toxic baits for mongooses containing either bromethalin, diphacinone, para-aminopropiophenone (PAPP), or sodium nitrite (SN). We estimated that a diphacinone bait would be the cheapest and fastest to register, and more application methods may be allowed compared to the others. On the negative side, we ranked diphacinone as the least humane toxicant of the four, largely due to a prolonged time to death following exposure and onset of symptoms. However, this interval also increases the probability that the antidote can be administered following an accidental exposure. If an alternative toxicant is required, use of a bromethalin, PAPP, or SN bait would likely be limited to bait stations or burrow baiting due to primary risks to non-target species. A bromethalin bait would be the cheapest and fastest to register of the three, particularly if a bait that is already commercially available proved efficacious for mongoose. However, we ranked bromethalin lower than PAPP or SN for overall humaneness. A PAPP bait would be slow and the most expensive to register. An SN bait would be challenging to formulate into a palatable bait with a reasonable shelf life. Although we focused on the U.S., mongooses are invasive in many parts of the world and the regulatory and use requirements for pesticides in other countries are generally comparable. A wide array of sizes, ecosystems, cultures, and invasive wildlife are represented among inhabited islands. Here, six cases from the United States of America (US) are selected to illustrate the high diversity of invasive animal management issues and objectives. We outline the background, define the problems and management objectives. We identify the management approaches and discuss the results and influences as they specifically relate to inhabited islands. The examples are: (1) Gambian giant pouched rats on Grassy Key, Florida; (2) coqui frogs on Kaua’i, Hawai’i; (3) feral swine on Cayo Costa Island, Florida; (4) rodents and monitor lizards on Cocos Island, Guam; (5) black spiny-tailed iguanas (ctenosaurs) on Gasparilla Island, Florida; and (6) mongooses on Puerto Rico. The small Indian mongoose (Herpestes auropunctatus) is a reservoir of rabies virus (RABV) in Puerto Rico and comprises over 70% of animal rabies cases reported annually. The control of RABV circulation in wildlife reservoirs is typically accomplished by a strategy of oral rabies vaccination (ORV). Currently no wildlife ORV program exists in Puerto Rico. Research into oral rabies vaccines and various bait types for mongooses has been conducted with promising results. Monitoring the success of ORV relies on estimating bait uptake by target species, which typically involves evaluating a change in RABV neutralizing antibodies (RVNA) post vaccination. This strategy may be difficult to interpret in areas with an active wildlife ORV program or in areas where RABV is enzootic and background levels of RVNA are present in reservoir species. In such situations, a biomarker incorporated with the vaccine or the bait matrix may be useful. We offered 16 captive mongooses placebo ORV baits containing ethyl-iophenoxic acid (et-IPA) in concentrations of 0.4% and 1% inside the bait and 0.14% in the external bait matrix. We also offered 12 captive mongooses ORV baits containing methyl-iophenoxic acid (me-IPA) in concentrations of 0.035%, 0.07% and 0.14% in the external bait matrix. We collected a serum sample prior to bait offering and then weekly for up to eight weeks post offering. We extracted Iophenoxic acids from sera into acetonitrile and quantified using liquid chromatography/mass spectrometry. We analyzed sera for et-IPA or me-IPA by liquid chromatography-mass spectrometry. We found adequate marking ability for at least eight and four weeks for et- and me-IPA, respectively. Both IPA derivatives could be suitable for field evaluation of ORV bait uptake in mongooses. Due to the longevity of the marker in mongoose sera, care must be taken to not confound results by using the same IPA derivative during consecutive evaluations.
