IL-12p35 is common to IL-35 and IL-12, which have opposing effects on inflammation. Here the authors show that the IL-12p35 subunit induces regulatory B cells and can be used therapeutically to limit autoimmune uveitis in mice.

Joint roughness coefficient (JRC) is a major factor that affects the mechanical properties of rock joints. Statistical methods that are used to calculate the JRC increasingly depend on a sampling interval (Δx). The variation rules of fitting parameters a, b, and b/a at different Δx values were analyzed on the basis of the relationship between the JRC and statistical parameter Z2. The relationship between the fitting parameters a and b was deduced in accordance with the ten standard profiles proposed by Barton. Empirical formulas for the JRC, Z2, and Δx were also established. The estimation accuracy of the JRC was the highest in the analysis of Δx values within 0.1–5.0 mm. JRC tests were conducted through inverse value comparative analysis. Results showed that the outcome calculated using the general formula and the JRC inverse values demonstrate improved agreement and verify the rationality of the general formula. The proposed formula can perform rapid and simple JRC calculation within the Δx range of 0.1–5.0 mm using Z2, thereby indicating favorable application prospects.

Interferon regulatory factor-4 (IRF4) and IRF8 regulate differentiation, growth and functions of lymphoid and myeloid cells. Targeted deletion of irf8 in T cells (CD4-IRF8KO) has been shown to exacerbate colitis and experimental autoimmune uveitis (EAU), a mouse model of human uveitis. We therefore generated mice lacking irf4 in T cells (CD4-IRF4KO) and investigated whether expression of IRF4 by T cells is also required for regulating T cells that suppress autoimmune diseases. Surprisingly, we found that CD4-IRF4KO mice are resistant to EAU. Suppression of EAU derived in part from inhibiting pathogenic responses of Th17 cells while inducing expansion of regulatory lymphocytes that secrete IL-10 and/or IL-35 in the eye and peripheral lymphoid tissues. Furthermore, CD4-IRF4KO T cells exhibit alterations in cell metabolism and are defective in the expression of two Ikaros zinc-finger (IKZF) transcription factors (Ikaros, Aiolos) that are required for lymphocyte differentiation, metabolism and cell-fate decisions. Thus, synergistic effects of IRF4 and IkZFs might induce metabolic reprogramming of differentiating lymphocytes and thereby dynamically regulate relative abundance of T and B lymphocyte subsets that mediate immunopathogenic mechanisms during uveitis. Moreover, the diametrically opposite effects of IRF4 and IRF8 during EAU suggests that intrinsic function of IRF4 in T cells might be activating proinflammatory responses while IRF8 promotes expansion of immune-suppressive mechanisms.

It is very important to identify fires in substations. Traditional fire alarm systems have problems such as low sensitivity and slow response. This article introduces the working principle of a fire alarm system based on artificial intelligence. And this article also designs different smoke particle sampling schemes for different areas in the substation, such as functional rooms/cable trenches, to ensure the correct operation of the system combined with the actual conditions of each area. Resulting that the fire alarm system based on artificial intelligence can detect the fire at the early stage and alarm in time.

At present, a variety of online monitoring technologies for substation equipment including winding temperature by optical fiber, however, the accuracy and stability show great differences among them. This paper focuses on the advancement, feasibility and economics of various new technologies, combined with the construction and operation experience of wisdom substations, in order to select mature, reliable, and effective new online monitoring technologies that reduce the amount of equipment operation and maintenance from a number of new technologies. In the end, the article gives suggestions on the application of various new online monitoring technologies.

Transient sensor malfunction, improper replacement measures, and large non‐Gaussian noise can cause data outliers in real‐world scenarios. In this article, an outlier‐robust zonotope set‐membership filter is proposed for uncertain nonlinear systems susceptible to outliers. In order to linearize nonlinear systems, a linearization method based on Stirling's interpolation formula is proposed. Then, an outlier detector utilizing the linearization error bounds, the prediction domain, and the output feasible domain is constructed. Based on the linearized system, a tight strip construction method is given to reduce the conservatism of the estimated enclosure. Moreover, a prediction‐correction‐backward reconstruction filter structure is designed to improve estimation accuracy and reduce conservatism. The proposed structure fuses historical data to attenuate the impact of uncertainty on the system. The robustness, guarantee, and convergence of the proposed filter are investigated. The promotion of urban mobility by integrating people-and-goods transportation has attracted increasing attention in recent years. Within this framework, diversified forms such as co-modality, freight on transit, and crowdshipping have been proposed, piloted or implemented. The success of the implementation and market penetration depends on not only the novelties of the concept but also the planning and operational efficiency. Thus, a comprehensive review focusing on the operation of integrated people-and-goods transportation systems and associated critical decisions and subproblems is performed. Different practical forms in which people and goods are transported in an integrated manner are identified. The critical decisions associated with each form and subproblem are discussed, along with corresponding models and solution approaches. Notably, because integrated transportation systems are in the early exploration stage at present, new forms are expected to emerge. Therefore, this paper proposes a general framework to realise the planning and operation of new forms in the future. The decisions and subproblems identified from existing forms are fed to the proposed general framework to identify two key research opportunities: to improve or extend existing research and to conduct pioneering research to fill the gaps in the frameworks for operating potential forms of integrated people-and-goods transportation. A novel breast ultrasound tomography system based on a circular array of capacitive micromechanical ultrasound transducers (CMUT) has broad application prospects. However, the images produced by this system are not suitable as input for the training phase of the super-resolution (SR) reconstruction algorithm. To solve the problem, this paper proposes an improved medical image super-resolution (MeSR) method based on the sparse domain. First, we use the simultaneous algebraic reconstruction technique (SART) with high imaging accuracy to reconstruct the image into a training image in a sparse domain model. Secondly, we denoise and enhance the contrast of the SART images to obtain improved detail images before training the dictionary. Then, we use the original detail image as the guide image to further process the improved detail image. Therefore, a high-precision dictionary was obtained during the testing phase and applied to filtered back projection SR reconstruction. We compared the proposed algorithm with previously reported algorithms in the Shepp Logan model and the model based on the CMUT background. The results showed significant improvements in peak signal-to-noise ratio, entropy, and average gradient compared to previously reported algorithms. The experimental results demonstrated that the proposed MeSR method can use noisy reconstructed images as input for the training phase of the SR algorithm and produce excellent visual effects. In the process of making mushrooms into vacuum-fried crisps, the resulting blanched broth (BB) and centrifuged broth (CB) are often discarded, thereby increasing the amount of wastewater and treatment costs. This study measured the proximate compositions, bioactive components, taste components, and minerals of freeze-dried BB and CB and then used functional indigestible dextrin (Fibersol-2) as a carrier to make these two broths into instant drinks. The solids of the BB and CB contained protein (16.88–19.21%), fat (0.01–0.23%), ash (12.89–13.50%), carbohydrate (67.28–70.00%), sugars and polyols (40.55–45.68%), free amino acids (6.58–6.69%), 5′-nucleotides (0.98–1.47%), and bioactive components, especially polysaccharides (4.53–7.45%), ergothioneine (both 0.19%), and total phenols (0.15–0.36%). The equivalent umami concentration of BB was 2.77-fold higher than that of the CB. Both BB and CB showed compositions and essential minerals that are rich in taste. Using a nine-point hedonic test, it was found that the solid contents of BB and CB in the instant drink affected the consumer's preference. The flavor and overall preference of instant drinks with 2.5% BB or CB were the best amongst consumers. Overall, the BB and CB were rich in nutrients and bioactive and taste components and could be developed as a functional food in the form of a drink. [ABSTRACT FROM AUTHOR]

Autophagy is one of the underlying causes of resistance to many antitumor drugs, including cisplatin (DDP). The low‐density lipoprotein receptor (LDLR) is a regulator of ovarian cancer (OC) progression. However, whether LDLR regulates DDP resistance in OC via autophagy‐related pathways remains unclear. LDLR expression was measured by quantitative real‐time PCR, western blot (WB) and IHC staining. A Cell Counting Kit 8 assay was employed to evaluate DDP resistance and cell viability, and flow cytometry was used to assess apoptosis. WB analysis was employed to evaluate the expression of autophagy‐related proteins and PI3K/AKT/mTOR signaling pathway proteins. The autophagolysosomes and the fluorescence intensity of LC3 were observed by transmission electron microscopy and immunofluorescence staining, respectively. A xenograft tumor model was established to explore the role of LDLR in vivo. LDLR was highly expressed in OC cells, which was correlated with disease progression. In DDP‐resistant OC cells, high LDLR expression was related to DDP resistance and autophagy. Downregulation of LDLR repressed autophagy and growth in DDP‐resistant OC cell lines by activating the PI3K/AKT/mTOR pathway, and these effects were eliminated by an mTOR inhibitor. In addition, LDLR knockdown also reduced OC tumor growth by suppressing autophagy associated with the PI3K/AKT/mTOR pathway. LDLR promoted autophagy‐mediated DDP resistance in OC associated with the PI3K/AKT/mTOR pathway, indicating that LDLR might be a new target to prevent DDP resistance in OC patients. Benzyl butyl phthalate (BBP) is a chemical softener and plasticizer commonly used in toys, food packaging, wallpaper, detergents and shampoos. The estrogenic actions of BBP have detrimental effects on humans and animals. In this study, the specific influence of BBP on mouse oocyte maturation was investigated using in vivo and in vitro models. The experiment first verified that BBP exposure significantly affected the rate of oocyte exclusion of the first polar body, although it did not affect germinal vesicle breakdown (GVBD) through in vitro oocyte culture system. Results of in vitro fertilization show that BBP exposure affects blastocyst rate. Subsequently, the results obtained by immunofluorescence staining technology showed that oocyte spindle organization, chromosomal arrangement and the distribution of cortical actin were disrupted by BBP exposure, and led to the failure of oocyte meiotic maturation and the subsequent early embryo development. Singe‐cell transcriptome analysis found that BBP exposure altered the expression levels of 588 genes, most associated with mitochondria‐related oxidative stress. Further analysis demonstrated that the detrimental effects of BBP involved the disruption of mitochondrial function and oxidative stress‐induced early apoptosis. Nicotinamide mononucleotide (NMN) supplementation reduced the adverse effects of BBP. Collectively, these findings revealed a mechanism of BBP‐induced toxicity on female reproduction and showed that NMN provides an effective treatment for BBP actions. [ABSTRACT FROM AUTHOR]

In this study, a dual-polarimetric radar observation operator is established and modified for the Taiwan area for the purpose of model verification. A severe squall line case during the Southwest Monsoon Experiment Intensive Observing Period 8 (SoWMEX IOP#8) on 14 June 2008, is selected and examined. Because the operator is adopted from the use of the midlatitude region, sensitivity tests are performed to obtain the optimal setting of the operator in the subtropical region. To accurately capture the dynamic structure of the squall lines, the ensemble-based data assimilation system, which assimilates both radial wind and reflectivity data, is used to obtain the optimal analysis field on the mesoscale for evaluating the performance of model simulation. The characteristics of two microphysics schemes are investigated, and the results obtained using the schemes are compared with the S-band dual-polarimetric radar observations. The horizontal and vertical cross-sections show that the analyses resemble the observations. Both schemes can replicate the polarimetric parameter signature such as ZDR and KDP columns. When comparing model simulation with polarimetric parameters through the drawing of contour frequency by altitude diagrams (CFADs), the results reveal that the single moment microphysics scheme performs better than the double moment scheme in this case. However, the reflectivity field in the stratiform area is more accurately captured when using the double moment scheme. Furthermore, validation with polarimetric variables (ZH, ZDR and KDP) histograms shows underestimation of the KDP field in both schemes. Overall, this study indicates the benefit of assimilating radial wind and reflectivity data for the analyses of severe precipitation systems and the necessity of assimilating polarimetric parameters for the accuracy of microphysical processes, especially complex microphysics schemes in subtropical region.

According to the World Health Organization global status report on road safety, traffic accidents are the eighth leading cause of death in the world, and nearly one-fifth of the traffic accidents were cause by driver distractions. Inspired by the famous two-stream convolutional neural network (CNN) model, we propose a driver behavior analysis system using one spatial stream ConvNet to extract the spatial features and one temporal stream ConvNet to capture the driver’s motion information. Instead of using three-dimensional (3D) ConvNet, which would suffer from large parameters and the lack of a pre-trained model, two-dimensional (2D) ConvNet is used to construct the spatial and temporal ConvNet streams, and they were pre-trained by the large-scale ImageNet. In addition, in order to integrate different modalities, the feature-level fusion methodology was applied, and a fusion network was designed to integrate the spatial and temporal features for further classification. Moreover, a self-compiled dataset of 10 actions in the vehicle was established. According to the experimental results, the proposed system can increase the accuracy rate by nearly 30% compared to the two-stream CNN model with a score-level fusion.

In recent years, with the promotion of global renewable energy, the introduction of energy storage batteries has become more and more important. Redox flow battery has attracted more and more attention due to their environmental friendliness, safe and reliable operation. This work studies the effect of different magnetic field intensities and the duration of magnetic field addition on the mass transfer and electrochemical reaction process of iron-vanadium ions in deep eutectic solvent (DES). At the same time, the effect of magnetic field on the polarization performance of DES-based redox flow battery is also studied. The experimental results show that the iron and vanadium DES electrolytes increase their redox peak current density under the action of adding magnetic field. The test of the polarization performance of the DES-based redox flow battery shows that the addition of magnetic field can reduce the area specific resistance (ASR) of the battery and improve the polarization performance of the battery.

This study proposes an integrated deep network consisting of a detection and identification module for person search. Person search is a very challenging problem because of the large appearance variation caused by occlusion, background clutter, pose variations, etc., and it is still an active research issue in the academic and industrial fields. Although various studies have been proposed, following the protocols of the person re-identification (ReID) benchmarks, most existing works take cropped pedestrian images either from manual labelling or a perfect detection assumption. However, for person search, manual processing is unavailable in practical applications, thereby causing a gap between the ReID problem setting and practical applications. One fact is also ignored: an imperfect auto-detected bounding box or misalignment is inevitable. We design herein a framework for the practical surveillance scenarios in which the scene images are captured. For person search, detection is a necessary step before ReID, and previous studies have shown that the precision of detection results has an influence on person ReID. The detection module based on the Faster R-CNN is used to detect persons in a scene image. For identifying and extracting discriminative features, a multi-class CNN network is trained with the auto-detected bounding boxes from the detection module, instead of the manually cropped data. The distance metric is then learned from the discriminative features output by the identification module. According to the experimental results of the test performed in the scene images, the multi-class CNN network for the identification module can provide a 62.7% accuracy rate, which is higher than that for the two-class CNN network.

Creeping flashover easily occurs at the interface between oil and pressboard in transformer and thus results in outage of power transmission system. Investigations have shown that creeping flashover characteristics at oil/pressboard interface can be improved by the addition of TiO2 nanoparticles, but the mechanism is still not thoroughly known. In this work, creeping flashover performance at nanofluid/pressboard interface modified by different sizes of nanoparticles were studied and the mechanism was presented as well. Nanofluids with the same concentration but with different sizes of TiO2 nanoparticles were prepared, and pressboards impregnated with them were prepared as well. After that, their creeping flashover characteristics were measured and compared. Nanoparticle’s size affected the creeping flashover performance along oil/pressboard greatly under both AC and lightning impulse voltages. The highest creeping flashover voltage can be enhanced by as high as 12.2% and 32.0% respectively. The underlying electric field distribution and charge transportation behaviors were analyzed to demonstrate the influence of nanoparticle’s size. By the addition of nanoparticles with a smaller size, the dielectric constant of nanofluid was increased closer to that of the pressboard, thus they were matched better. Moreover, charge was easier to dissipate from the oil/pressboard interface and electric field distortion at the interface was consequently reduced. Therefore, the electric field was more like a uniform field and the forward development of flashover was more difficult, leading to a better performance of creeping flashover of oil-impregnated pressboard.

Multiple sclerosis (MS) is an inflammatory demyelinating disease in which cytokines produced by immune cells that infiltrate the brain and spinal cord play a central role. We show here that the IL-12p35, the alpha subunit of IL-12 or IL-35 cytokine, might be an effective biologic for suppressing neuroinflammatory responses and ameliorating the pathology of experimental autoimmune encephalomyelitis (EAE), the mouse model of human MS. We further show that IL-12p35 conferred protection from neuropathy by inhibiting the expansion of pathogenic Th17 and Th1 cells and inhibiting trafficking of inflammatory cells into the brain and spinal cord. In addition, in vitro exposure of encephalitogenic cells to IL-12p35 suppressed their capacity to induce EAE by adoptive transfer. Importantly, the IL-12p35-mediated expansion of Treg and Breg cells and its amelioration of EAE correlated with inhibition of cytokine-induced activation of STAT1/STAT3 pathways. Moreover, IL-12p35 inhibited lymphocyte proliferation by suppressing the expressions of cell-cycle regulatory proteins. Taken together, these results suggest that IL-12p35 can be exploited as a novel biologic for treating central nervous system autoimmune diseases and offers the promise of ex vivo production of large amounts of Tregs and Bregs for immunotherapy.
