Lipid extraction from microalgae was studied more specifically considering the effects of the extraction method and solvent mixture on cell walls. Chlorella vulgaris and Nannochloropsis oculata were maintained in Bold Basal Medium (BBM) and, Dunaliella salina and Dunaliella tertiolecta in Guillard "F/2" medium, sterile with continuous aeration at 18 ºC at 80 µmol photons m−2 s−1. The objective of the study was to evaluate and compare the lipid content and fatty acid profiles of 4 microalgae species (freshwater and marine species) using two lipid extraction methods, Soxhlet and ultrasonication. Biomass production was greatest in the stationary phase for all microalgae, with D. salina exhibiting the highest production with 0.40–0.42 g L−1. Biomass productivity for freshwater microalgae was higher in the stationary phase (0.0360–0.0407 g L−1 day−1) and in the exponential phase for marine microalgae (0.057–0.064 g L−1 day−1). The highest percentage of lipids in marine microalgae was between 38–54% by weight in the stationary phase, with D. tertiolecta; in comparison, the freshwater microalgae reached between 22–23% by weight in the exponential phase, for the species N. oculata. The highest number of fatty acids observed in marine microalgae was by Soxhlet extraction in D. tertiolecta in both growth phases. The highest percentage of any fatty acid with this method was palmitic acid (61%) in the stationary phase of C vulgaris. The ultrasonication extraction method presented the highest percentages in the stationary phase of N. oculata and D. tertiolecta, palmitic acid (C16:0) showing the highest percentages (87 and 51%) in the exponential phase in both marine microalgae. Purpose: Smaller lipid droplet morphology and GLUT 4 protein expression have been associated with greater muscle oxidative capacity and glucose uptake, respectively. The main purpose of this study was to determine the effect of an acute long-duration exercise bout on skeletal muscle lipid droplet morphology, GLUT4, perilipin 3, and perilipin 5 expressions. Methods: Twenty healthy men (age 24.0 ± 1.0 years, BMI 23.6 ± 0.4 kg/m2) were recruited for the study. The participants were subjected to an acute bout of exercise on a cycle ergometer at 50% VO2max until they reached a total energy expenditure of 650 kcal. The study was conducted after an overnight fast. Vastus lateralis muscle biopsies were obtained before and immediately after exercise for immunohistochemical analysis to determine lipid, perilipin 3, perilipin 5, and GLUT4 protein contents while GLUT 4 mRNA was quantified using RT-qPCR. Results: Lipid droplet size decreased whereas total intramyocellular lipid content tended to reduce (p = 0.07) after an acute bout of endurance exercise. The density of smaller lipid droplets in the peripheral sarcoplasmic region significantly increased (0.584 ± 0.04 to 0.638 ± 0.08 AU; p = 0.01) while larger lipid droplets significantly decreased (p < 0.05). GLUT4 mRNA tended to increase (p = 0.05). There were no significant changes in GLUT 4, perilipin 3, and perilipin 5 protein levels. Investigation of the lipid and osmolyte composition in conidia of the xerohalophile Aspergillus penicilloides grown on media with high NaCl and glycerol concentrations is important for the understanding of the functions of these compounds in the adaptation of the extremophile. Conidia of the fungus grown on a medium with glycerol contained up to 15% of the dry mass of carbohydrates and polyols (CaP) of the cytosol, with predominance of glycerol (60% of the total) in their composition, while on a medium with salt, the proportion of glycerol did not exceed 20% and the amount of CaP was 10%. On both media, the proportion (20%) and amount (2.5%) of trehalose were the same; the proportion of polyols was thus about 80%, although their composition was different, indicating a possible synergy between trehalose and polyols in adaptation. Both variants of conidia were characterized by a low content of both membrane and storage lipids represented mainly by triacylglycerides. Phosphatidylcholines and sterols were the predominant membrane lipids of the fungal conidia, regardless of the composition of the medium, which indicated high stability and conservatism of the lipid bilayer. The constancy of the composition of membrane and storage lipids and their low content, the high level of CaP, and significant differences in their composition indicate the key role of osmolytes in the adaptation of fungal conidia to various environmental conditions. Paclitaxel (PTX) is a hydrophobic small-molecule cancer drug that loads into the membrane (tail) region of lipid carriers such as liposomes and micelles. The development of improved lipid-based carriers of PTX is an important objective to generate chemotherapeutics with fewer side effects. The lipids 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE) and glyceryl monooleate (GMO) show propensity for fusion with other lipid membranes, which has led to their use in lipid vectors of nucleic acids. We hypothesized that DOPE and GMO could enhance PTX delivery to cells through a similar membrane fusion mechanism. As an important measure of drug carrier performance, we evaluated PTX solubility in cationic liposomes containing GMO or DOPE. Solubility was determined by time-dependent kinetic phase diagrams generated from direct observations of PTX crystal formation using differential-interference-contrast optical microscopy. Remarkably, PTX was much less soluble in these liposomes than in control cationic liposomes containing univalent cationic lipid 1,2-dioleoyl-3-trimethylammonium-propane (DOTAP) and 1,2-dioleoyl-sn-glycero-3-phosphatidylcholine (DOPC), which are not fusogenic. In particular, PTX was not substantially soluble in GMO-based cationic liposomes. The fusogenicity of DOPE and GMO is related to the negative spontaneous curvature of membranes containing these lipids, which drives formation of nonlamellar self-assembled phases (inverted hexagonal or gyroid cubic). To determine whether PTX solubility is governed by lipid membrane structure or by local intermolecular interactions, we used synchrotron small-angle X-ray scattering. To increase the signal/noise ratio, we used DNA to condense the lipid formulations into lipoplex pellets. The results suggest that local intermolecular interactions are of greater importance and that the negative spontaneous curvature-inducing lipids DOPE and GMO are not suitable components of liposomal carriers for PTX delivery. The microalga, Dunaliella salina, is a treasure of fine chemicals, carotenoids, and lipids of industrial importance. The present investigation focused on the maximization of lipid content in the context of biofuel. Mixotrophic cultures growing in different combinations of sodium bicarbonate (NaHCO3) and glucose (Glu) or sodium acetate (Na-Ace) were monitored for essential growth parameters such as photosynthesis, dry biomass, and Chl a content. The maximum photosynthetic quantum yield (Fv/Fm) of all the screened mixotrophic cultures along with control ranged between 0.63 and 0.72, indicating the cells were in a good physiological state. The highest biomass productivity (33.46 ± 0.58 mg/L/d) was recorded in 1.0 mM NaHCO3 + 1.0 mM Glu-containing cultures. However, the highest lipid content (56% DCW) was recorded in 1.0 mM NaHCO3 + 0.5 mM Na-Ace-containing cultures. Interestingly, a 1.7-fold higher starch content (72.3 μg/mL) was recorded in 0.25 mM NaHCO3 + 0.5 mM Na-Ace over 1.0 mM NaHCO3 + 0.5 mM Na-Ace-containing cultures. Thus, data indicated that the higher concentration of NaHCO3 along with Na-Ace favoured lipid biosynthesis, while the lower concentration of NaHCO3 with Na-Ace favoured starch synthesis. Thus, substrate provision determined the desired products. Lipids obtained from D. salina at stage II (nitrogen-deficient condition) possessed ideal fuel properties according to international standards of EN 14214 and ASTM D6751. Therefore, the present work explored the threshold level of bicarbonate along with Na-Ace which was able to maintain good physiological status and produced higher lipids with better biodiesel properties. Microalgal-biofilm culture has the advantages of less water requirement and higher lipid productivity. For efficient lipid production in an aerial phase condition, it is ideal to simultaneously induce microalgal biofilm growth and lipid accumulation. Biomass production and lipid accumulation in the aerial microalga Coccomyxa subellipsoidea KGU-D001 were investigated in the logarithmic growth phase. Initially, the alga was cultured in three different conditions: in liquid phase using Bold's basal medium (BBM), and in the aerial phase in the presence or absence of BBM. In aerial phase conditions with nutrients (i.e., BBM), biomass production was prioritized over cell proliferation until approximately the 7th day of culture. In the liquid phase conditions, the ratio of total organic carbon to total nitrogen (C/N) in cells was almost constant during the culture period. However, in both aerial phase conditions, the C/N ratio increased during the culture period, more so in the absence of nutrients. The total fatty acid content also increased throughout the culture period. Then, algal cells were cultured for a 17-day culture; there would be a growth period of 10 days followed by a lipid accumulation period of 7 days. In such cultures in the presence of BBM, the biomass and lipid productivities (388 and 94.2 mg/m2/day) were both significantly higher in the aerial phase than in the liquid phase during the lipid production period. Spiritual well-being is considered a significant factor in helping to manage chronic diseases and cope with the disease process. This descriptive-correlational study aimed to investigate the relationship between spiritual well-being, diabetes burden, self-management, and among 300 outpatients with type 2 diabetes in Turkey. A significant relationship was found between the diabetes burden and self-management levels and the spiritual well-being of patients with diabetes (p < 0.005). Multiple linear regression analyses found that a high diabetes burden (ß = −0.106) decreased well-being, and high self-management increased well-being (ß = 0.415). Additionally, the results revealed that marital status, household members, performing daily life activities alone, hospitalization due to complications, diabetes burden, self-management, glycemic control, and blood lipid parameters explained 29% of the total variance in the spiritual well-being level. Accordingly, the present study recommended that health professionals should consider spiritual well-being to support disease management with a holistic approach to diabetes patients. Background: Dyslipidemia is a crucial risk factor for atherosclerotic cardiovascular disease. However, there is limited data available on the differences in the prevalence of dyslipidemia between rural and urban populations in India. The aim of the present investigation is to describe the prevalence of dyslipidemia and the risk factors associated with adverse lipid profiles among adults residing in rural, sub-urban, and urban areas of India, and to assess its association with diabetes and hypertension. Methods: We enrolled adults over the age of 20 who lived in three distinct demographic areas in South India: rural, sub-urban, and urban. Data on demographics, lifestyle, disease history, family history, body weight, height, waist circumference, blood pressure, and clinical characteristics were collected for this study. We used chi-square tests and multivariable logistic regression to analyze demographic prevalence and risk factors related to lipid abnormalities among study participants. Results: This study enrolled 2976 randomly selected participants from rural, sub-urban, and urban communities in Tamil Nadu, India. Of these, 865 (29.1%) were rural residents, 1030 (34.6%) were sub-urban residents, and 1081 (36.3%) were urban residents. About 80% of women who lived in the suburban area had higher rates of low-HDL cholesterol. Compared to sub-urban (29.9%, 49%, and 21.1%) and rural (33.4%, 43.4%, and 24.1%) populations, urban populations had higher prevalence rates of hypercholesterolemia, hypertriglyceridemia, and elevated LDL-C (37.3%, 52.5%, and 38.6%), respectively. Men were more likely than women to develop dyslipidemia before the age of 40, but after that age, men showed a reduced risk of dyslipidemia than women, except for low HDL cholesterol. Age group, gender, current drinker, overweight, obesity, diabetes, and hypertension showed a significant association (p < 0.05) with dyslipidemia. Conclusions: The study found that more than 85% of the sub-urban and urban population had dyslipidemia (at least one lipid abnormality) compared to rural residents (78.5%). Cationic membrane-active toxins are the most abundant group of proteins in the venom of snakes and insects. Cationic proteins such as cobra venom cytotoxin and bee venom melittin are known for their pharmacological reactions including anticancer and antimicrobial effects which arise from the toxin-induced alteration in the dynamics and structure of plasma membranes and membranes of organelles. It has been established that these cationic toxins trigger the formation of non-bilayer lipid phase transitions in artificial and native mitochondrial membranes. Remarkably, the toxin-induced formation of non-bilayer lipid phase increases at certain conditions mitochondrial ATP synthase activity. This observation opens an intriguing avenue for using cationic toxins in the development of novel drugs for the treatment of cellular energy deficiency caused by aging and diseases. This observation also warrants a thorough investigation of the molecular mechanism(s) of lipid phase polymorphisms triggered by cationic proteins. This article presents a review on the application of powerful biophysical methods such as resonance spectroscopy (31P-, 1H-, 2H-nuclear magnetic resonance, and electron paramagnetic resonance), luminescence, and differential scanning microcalorimetry in studies of non-bilayer lipid phase transitions triggered by cationic proteins in artificial and biological membranes. A phenomenon of the triggered by cationic proteins the non-bilayer lipid phase transitions occurring within 10−2–10−11 s is discussed in the context of potential pharmacological applications of cationic proteins. Next to the ATP dimer is an inverted micelle made of cardiolipin that serves as a vehicle for the transport of H+ ions from the intra-crista space to the matrix. It is proposed that such inverted micelles are triggered by the high density of H+ ions and the cationic proteins rich in lysine residue which compete with the conserved lysine residues of the ATP synthase rotor for binding to cardiolipin in the inner mitochondrial membrane and perturb the bilayer lipid packing of cristae. Lipids are complex organic molecules that fulfill energy demands and sometimes act as signaling molecules. They are mostly found in membranes, thus playing an important role in membrane trafficking and protecting the cell from external dangers. Based on the composition of the lipids, their fluidity and charge, their interaction with embedded proteins vary greatly. Bacteria can hijack host lipids to satisfy their energy needs or to conceal themselves from host cells. Intracellular bacteria continuously exploit host, from their entry into host cells utilizing host lipid machinery to exiting through the cells. This acquisition of lipids from host cells helps in their disguise mechanism. The current review explores various mechanisms employed by the intracellular bacteria to manipulate and acquire host lipids. It discusses their role in manipulating host membranes and the subsequence impact on the host cells. Modulating these lipids in macrophages not only serve the purpose of the pathogen but also modulates the macrophage energy metabolism and functional state. Additionally, we have explored the intricate pathogenic relationship and the potential prospects of using this knowledge in lipid-based therapeutics to disrupt pathogen dominance. Microalgae have been recognized as promising competitors for bioenergy production, because of their high lipid content and quick biomass development. In this work, freshwater microalgae Scenedesmus dimorphus was cultivated in a synthetic wastewater media with the object of investigating the bioenergy potential by physicochemical characterization. Batch experiments were performed photoautotrophically for biomass generation supplemented with different levels of CO2 concentration. Maximum biomass production was in the limit of 373 mg/L to 587 mg/L under different CO2 feed concentrations. Cultivation of microalgae along with CO2 supplementation found a substantial impact on buildup of lipid in microalgae biomass. Maximum 29% and 32% lipid content were achieved for 2% and 6% CO2 enrichment respectively, under nitrogen-depleted conditions. Fourier transform infrared spectroscopy, thermogravimetric analysis, and carbon, hydrogen, nitrogen, sulfur elemental analyzer instrumental tools were used to perform physicochemical characterization on biomass in order to determine its proximate analysis, ultimate analysis, higher heating value (HHV), and macromolecular composition, The higher heating value (HHV) of Scenedesmus dimorphus biomass achieved 33.26 MJ/kg with almost zero sulfur content. This HHV value is significantly higher than that of other microalgae sp. Moreover, this HHV value is also even higher than that of bituminous coal (24.3 MJ/kg) and other types of lignocellulosic biomass. Hydrothermal liquefaction (HTL) is a green technology for biocrude production from algae. The global reaction kinetic, component additivity model (CAM), and response surface methodology (RSM) of HTL from high-lipid microalgae (Aurantiochytrium sp.) were evaluated. Lipids, proteins, and carbohydrates of the microalgae cell reacted at different rates and produced the aqueous phase, bio-oil, and gas phase in the kinetic model. Performing global kinetic modelling, understanding the reactions, and the performance of algae for biofuel production by HTL were the main ideas and novelty behind this work. Also, the predictive ability of bio-oil yield was examined by kinetic, CAM, and RSM approaches. MATLAB was utilized to solve the system of ordinary differential equations and calculate the optimum values for Arrhenius kinetic parameters by minimizing the least-square differences between model and experimental yields. R-squared of 0.997 showed that the global kinetic modelling provides a great prediction of bio-oil yields. Activation energies were calculated at 32.31, 38.50, and 46.99 kJ/mol for the conversion of lipids, proteins, and carbohydrates to bio-oil. Furthermore, a maximum bio-oil yield of 53.43 wt% was predicted at 800 K in 2 min. Also, the result of bio-oil prediction by global kinetic modelling compared to CAM and RSM equations. In comparison with the CAM model, the RSM equations showed better adaption with the bio-oil yield at 350 °C and 40 min. Introduction: Sleep patterns are more frequently interrupted in patients with Parkinson's disease (PD), and it is still unclear whether genetic factors are involved in PD-related sleep disorders. In this study, we hypothesize that PD-associated genetic risk affects lipid metabolism, which in turn contributes to different types of sleep disorders. Methods: We used a non-targeted lipidomics to explore the lipid composition of cerebrospinal fluid (CSF) exosomes derived from patients with PD carrying phospholipase A2 Group VI (PLA2G6) and sphingomyelin phosphodiesterase 1 (SMPD1) mutations. Results: PLA2G6 mutations (c.1966C > G, Leu656Val; c.2077C > G, Leu693Val; c.1791delC, His597fx69) significantly increase the exosomal content of glycerophospholipids and lysophospholipids, specifically phosphatidylcholine (PC) and lysophosphatidylcholine (LPC). Exosome surface presence of melatomin receptor 1A (MTNR1A) was detectable only in patients with PLA2G6 mutations. We have further shown that, in patients with PD carrying PLA2G6 mutations, sleep latency was significantly longer compared to those carrying WT PLA2G6, and we speculate that functional PLA2G6 mutations lead to structural changes and lipid deregulation of exosomes, which in turn alters exosomal cargo and affects PD-related sleep disorders. In SMPD1, G508R variant-carrying patients with PD abundance of sphingomyelins was significantly higher and had significantly shorter rapid eye movement sleep. Conclusions: Our study demonstrated that the disturbed composition and function of CSF-derived exosome lipidome during the pathological stage of PD may affect different types of sleep disorder in PD. Energy and environmental issues related to conventional fossil-derived products and fuels have led researchers to focus on alternative, more environmentally-friendly processes, such as the production of microbial oils from renewable feedstocks or even pollutants as sustainable sources of biofuels, allowing to progressively move away from the use of fossil fuels. Among the oleaginous yeasts, Yarrowia lipolytica is a highly promising cell factory and microbial oil producer because of its high capacity to accumulate lipids for subsequent biofuel production. Y. lipolytica also stands out for its ability to assimilate various carbon sources, even at low cost, reaching lipid concentrations of at least 30% by weight with non-genetically modified strains, and even much higher values with engineered organisms. Among others, fatty acids have attracted recent interest as substrates for their lower cost and possible production from pollutants compared to sugars. This review pays special attention to some of those emerging carbon sources, i.e., carboxylic acids and even greenhouse gases. Besides, another focus is to provide detailed up to date information on the main characteristics and factors that most influence the fermentation process of this yeast, with the ultimate aim of optimising the bioconversion process and the synthesis of useful metabolites. Besides, the reader will find comprehensive information on the industrial applicability of the synthesised lipids, in addition to the production of biofuels. The extreme weather conditions of Transbaikalia cause phytostress in plants. This results in the formation of protective systems and mobilization that involves certain metabolic changes acting as survival means, including an increase in the content of lipids and lignin. Metabolism undergoes modifications determined by the pace and depth of changes that can occur in it without disturbing the balance between individual functions; as a result, the unity of the plant and the environment is not disturbed. Cryoarid conditions cause specific morphological and chemical adaptations in plants. It was found that the total content of lipids and lignin in the studied soils of Transbaikalia is higher, while the content of proteins is lower compared to similar soils in European Russia. Purpose of Review: Review updates for the association of HDL-cholesterol with atherosclerotic cardiovascular disease (ASCVD) and discuss the approach to incorporating HDL-cholesterol within risk assessment. Recent Findings: There is a U-shaped relationship between HDL-cholesterol and ASCVD. Both low HDL-cholesterol (< 40 mg/dL in men, < 50 mg/dL in women) and very-high HDL-cholesterol (≥ 80 mg/dL in men) are associated with a higher risk of all-cause and ASCVD mortality, independent from traditional risk factors. There has been inconsistency for the association between very-high HDL-cholesterol and mortality outcomes in women. It is uncertain whether HDL-cholesterol is a causal ASCVD risk factor, especially due to mixed results from Mendelian randomization studies and the collinearity of HDL-cholesterol with established risk factors, lifestyle behaviors, and socioeconomic status. Summary: HDL-cholesterol is a risk factor or risk enhancer in primary prevention and high-risk condition in secondary prevention when either low (men and women) or very-high (men). The calcifying cells of corals are responsible for skeleton formation, a process that is the basis for reef building. Their cell biology is therefore of primary interest, but current knowledge is limited because direct in vivo investigation of this cell type is challenging. Studies at the growing edge of laterally extending coral colonies allow for direct observations of actively calcifying cells in living samples. In the current study, we used this approach to study lipid droplets, which are considered storage organelles that fuel cells for physiological processes. Using the fluorescent lipid probe Nile Red and in vivo confocal microscopy, we observed lipid droplets in calcifying cells of the coral Stylophora pistillata and we suggest that they play a key role in coral calcification. Antibiotics are central to modern medicine, and yet they are mainly the products of intra and inter-kingdom evolutionary warfare. To understand how nature evolves antibiotics around a common mechanism of action, we investigated the origins of an extremely valuable class of compounds, lipid II targeting glycopeptide antibiotics (GPAs, exemplified by teicoplanin and vancomycin), which are used as last resort for the treatment of antibiotic resistant bacterial infections. Using a molecule-centred approach and computational techniques, we first predicted the nonribosomal peptide synthetase assembly line of paleomycin, the ancestral parent of lipid II targeting GPAs. Subsequently, we employed synthetic biology techniques to produce the predicted peptide and validated its antibiotic activity. We revealed the structure of paleomycin, which enabled us to address how nature morphs a peptide antibiotic scaffold through evolution. In doing so, we obtained temporal snapshots of key selection domains in nonribosomal peptide synthesis during the biosynthetic journey from ancestral, teicoplanin-like GPAs to modern GPAs such as vancomycin. Our study demonstrates the synergy of computational techniques and synthetic biology approaches enabling us to journey back in time, trace the temporal evolution of antibiotics, and revive these ancestral molecules. It also reveals the optimisation strategies nature has applied to evolve modern GPAs, laying the foundation for future efforts to engineer this important class of antimicrobial agents. Lipid nanoparticles (LNPs) have been used as a carrier for messenger RNA (mRNA) vaccines. Surface properties of LNPs are important to the stability and function of mRNA vaccines. Polyethylene-glycol (PEG) is a functional lipid at the surface of LNPs that improves colloidal stability, increases circulation time, and impacts cellular uptake. In this study, we explore in-depth lipid composition at the surface of mRNA-LNPs using high-field nuclear magnetic resonance (NMR) spectroscopy. Our results provide a unique surface lipid profile of intact LNPs identifying PEG chains and partial ionizable lipids are present with quantification capability. The surface PEG density is determined to reveal the brush-like conformation on the surface of mRNA-LNPs. Furthermore, we implement a diffusion NMR strategy for routine testing of formulated drug products during drug development. The coronavirus disease 2019 (COVID-19), which affects multiple organs, is causing an unprecedented global public health crisis. Most COVID-19 patients recover gradually upon appropriate interventions. Viruses were reported to utilize the small extracellular vesicles (sEVs), containing a cell-specific cargo of proteins, lipids, and nucleic acids, to escape the attack from the host's immune system. This study aimed to examine the sEVs lipid profile of plasma of recovered COVID-19 patients (RCs). Plasma sEVs were separated from 83 RCs 3 months after discharge without underlying diseases, including 18 recovered asymptomatic patients (RAs), 32 recovered moderate patients (RMs), and 33 recovered severe and critical patients (RSs), and 19 healthy controls (HCs) by Total Exosome Isolation Kit. Lipids were extracted from sEVs and then subjected to targeted liquid chromatography-mass spectrometry. The size, concentration, and distribution of sEVs did not differ in RCs and HCs as validated by transmission electron microscopy, nanoparticle tracking analysis, and immunoblot analysis. Fifteen subclasses of 508 lipids were detected in plasma sEVs from HCs, RAs, RMs, and RSs, such as phosphatidylcholines (PCs) and diacylglycerols (DAGs), etc. Total lipid intensity displayed downregulation in RCs compared with HCs. The relative abundance of DAGs gradually dropped, whereas PCs, lysophosphatidylcholines, and sphingomyelins were higher in RCs relative to HCs, especially in RSs. 88 lipids out of 241 in sEVs of RCs were significantly different and a conspicuous increase was revealed with disease status. The sEVs lipids alternations were found to be significantly correlated with the clinical indices in RCs and HCs, suggesting that the impact of COVID-19 on lipid metabolism lingered for a long time. The lipid abnormalities bore an intimate link with glycerophospholipid metabolism and glycosylphosphatidylinositol anchor biosynthesis. Furthermore, the lipidomic analysis showed that RCs were at higher risk of developing diabetes and sustaining hepatic impairment. The abnormality of immunomodulation in RCs might still exist. Honokiol (HNK), one of the main active components of Magnolia officinalis, has a positive effect on non-alcoholic steatohepatitis (NASH). However, the effects of HNK on the composition of serum lipids and bile acids (BAs) and gut microbiota (GM) of NASH mice are still unknown.C57BL/6 mice were fed with methionine-choline deficiency (MCD) diet and gavaged with HNK (20 mg/kg/d) for 8 weeks, then the serum lipids and BAs were detected by LC–MS, the composition of ileum microflora and the mRNA expression of hepatic BAs homeostasis related genes were analyzed by 16S rDNA sequencing and RT-qPCR, respectively. HNK treatment decreased the degree of hepatic lipid drops, inflammatory cell infiltration and fibrosis. Meantime, the serum levels of 34 lipids and 4 BAs in MCD mice were significantly altered by HNK treatment, as well as the increased abundance of Ruminococcaceae, Caulobacteraceae and Brevundimonas, and the decreased abundance of Firmicutes and Dubosiella. Besides, HNK treatment increased the hepatic mRNA expression of Oatp1b2 in MCD mice. Partially defatted poppy seeds, a by-product of poppy oil cold pressing, could be an interesting dietary supplement for obesity management. The aim of this study was to compare the effects of dietary supplementation with a small amount of native or partially defatted poppy seeds on gastrointestinal function and lipid metabolism in rats fed a high-fat diet. The defatted poppy seeds had, among others, lower fat content and higher fibre and protein content than native poppy seeds. The rats fed with a high-fat diet were characterised by severe metabolic disorders, especially in the liver, and poppy seeds were unable to prevent them. However, depending on the seed form, dietary supplementation with poppy seeds differentially affected the microbial and endogenous lipid metabolism in rats. In the distal intestine, both dietary seed forms stimulated microbial acetate production, and the supplementation with partially defatted poppy seeds additionally inhibited isobutyrate and isovalerate formation, which indicates a reduction in putrefaction. Both dietary seed forms increased cholesterol accumulation in the liver. Only dietary supplementation with partially defatted poppy seeds attenuated visceral fat and hepatic triglyceride accumulations and lowered blood triglyceride concentrations, and at the transcriptional level, the inhibition of SREBP-1c, which upregulates genes responsible for de novo lipogenesis, was additionally observed in this organ. Previously, we observed curcumin improves aging-associated memory impairment in d-galactose (D-gal) and normal-aged (NA) mice. Evidence showed that multiple agents can be used in managing aging-induced memory dysfunction, drawn by the contribution of several pathways. Curcumin and Epigallocatechin 3 gallate (EGCG) combination substantially reduced the oxidative stress that commonly mediates aging. This study examined the combined effect of EGCG and curcumin on memory improvement in two recognized models, D-gal and normal-aged (NA) mice. The co-administration of EGCG and curcumin significantly (p < 0.05) increased retention time detected by passive avoidance (PA) and freezing response determined in contextual fear conditioning (CFC) compared to the discrete administration of EGCG or curcumin. Biochemical studies revealed that the combination of EGCG and curcumin remarkably ameliorated the levels (p < 0.05) of glutathione, superoxide dismutase, catalase, advanced oxidation protein products, nitric oxide, and lipid peroxidation compared to the monotherapy of EGCG or curcumin in mice hippocampi. The behavioral and biochemical studies revealed that the combination of EGCG and curcumin showed better improvement in rescuing aging-associated memory disorders in mice. Although the human immunodeficiency virus type 1 lipid envelope has been reported to be enriched with host cell sphingomyelin and cholesterol, the molecular mechanism of the enrichment is not well understood. Viral Gag protein plays a central role in virus budding. Here, we report the interaction between Gag and host cell lipids using different quantitative and super-resolution microscopy techniques in combination with specific probes that bind endogenous sphingomyelin and cholesterol. Our results indicate that Gag in the inner leaflet of the plasma membrane colocalizes with the outer leaflet sphingomyelin-rich domains and cholesterol-rich domains, enlarges sphingomyelin-rich domains, and strongly restricts the mobility of sphingomyelin-rich domains. Moreover, Gag multimerization induces sphingomyelin-rich and cholesterol-rich lipid domains to be in close proximity in a curvature-dependent manner. Our study suggests that Gag binds, coalesces, and reorganizes pre-existing lipid domains during assembly. Quantitative microscopies and specific lipid probes revealed that multimerizing HIV-1 Gag in the plasma membrane inner leaflet brings the outer leaflet sphingomyelin-rich and cholesterol-rich lipid domains closer in a curvature-dependent manner. Lipid droplets (LDs) are dynamic lipid storage organelles that can sense and respond to changes in systemic energy balance. The size and number of LDs are controlled by complex and delicate mechanisms, among which, whether and which SNARE proteins mediate LD fusion, and the mechanisms governing this process remain poorly understood. Here we identified a SNARE complex, syntaxin 18 (STX18)–SNAP23–SEC22B, that is recruited to LDs to mediate LD fusion. STX18 targets LDs with its transmembrane domain spanning the phospholipid monolayer twice. STX18–SNAP23–SEC22B complex drives LD fusion in adiposome lipid mixing and content mixing in vitro assays. CIDEC/FSP27 directly binds STX18, SEC22B, and SNAP23, and promotes the lipid mixing of SNAREs-reconstituted adiposomes by promoting LD clustering. Knockdown of STX18 in mouse liver via AAV resulted in smaller liver and reduced LD size under high-fat diet conditions. Metabolic reprogramming is a hallmark of cancers, but pan-cancer level roles of lipid metabolism in cancer development are remains poorly understood. We investigated the possible roles of lipid metabolic genes (LMGs) in 14 cancer types. The results indicate that: (1) there is strong evidence for increased lipid metabolism in THCA and KICH. (2) Although the overall levels of lipid metabolic processes are down-regulated in some cancer types, fatty acid synthase activity and fatty acid elongation are moderately up-regulated in more than half of the cancer types. Cholesterol synthesis is up-regulated in five cancers including KICH, BLCA, COAD, BRCA, UCEC, and THCA. (3) The catabolism of cholesterols, triglycerides and fatty acids is repressed in most cancers, but a specific form of lipid degradation, lipophagy, is activated in THCA and KICH. (4) Lipid storage is enhanced in in kidney cancers and thyroid cancer. (5) Similarly to primary tumors, metastatic tumors tend to up-regulate biosynthetic processes of diverse lipids, but down-regulate lipid catabolic processes, except lipophagy. (6) The frequently mutated lipid metabolic genes are not key LMGs. (7) We established a LMG-based model for predicting cancer prognosis. This study aimed at investigating the association of acrylamide consumption with the incidence of type 2 diabetes (T2D) in adults. The 6022 subjects of the Tehran lipid and glucose study participants were selected. The acrylamide content of food items were summed and computed cumulatively across follow up surveys. Multivariable Cox proportional hazard regression analyses were performed to estimate the hazards ratio (HR) and 95% confidence interval (CI) of incident T2D. This study was done on men and women, respectively aged 41.5 ± 14.1 and 39.2 ± 13.0 years. The mean ± SD of dietary acrylamide intake was 57.0 ± 46.8 µg/day. Acrylamide intake was not associated with the incidence of T2D after adjusting for confounding variables. In women, a higher acrylamide intake was positively associated with T2D [HR (CI) for Q4: 1.13 (1.01–1.27), P trend: 0.03] after adjusting for confounding factors. Matrix-Assisted Laser Desorption Ionization Mass Spectrometry Imaging (MALDI-MSI) spatially resolves the chemical composition of tissues. Lipids are of particular interest, as they influence important biological processes in health and disease. However, the identification of lipids in MALDI-MSI remains a challenge due to the lack of chromatographic separation or untargeted tandem mass spectrometry. Recent studies have proposed the use of MALDI in-source fragmentation to infer structural information and aid identification. Here we present rMSIfragment, an open-source R package that exploits known adducts and fragmentation pathways to confidently annotate lipids in MALDI-MSI. The annotations are ranked using a novel score that demonstrates an area under the curve of 0.7 in ROC analyses using HPLC–MS and Target-Decoy validations. rMSIfragment applies to multiple MALDI-MSI sample types and experimental setups. Finally, we demonstrate that overlooking in-source fragments increases the number of incorrect annotations. Oleaginous yeast lipids have myriad of industrial applications that are gaining significant interest owing to shorter incubation, ability to use broad spectrum substrates, and quality lipids. However, the lipid content produced is low and need to enhance by optimization of varied parameters. In the present study, crude glycerol a by-product of biodiesel industry was supplemented to Trichosporon shinodae for lipid accumulation using central composite design (CCD) of response surface methodology (RSM). The developed quadratic model was found to be significant with the R2 value of 95.20% and adj. R2 value of 91.97%. An optimal lipid content of 49.85 ± 0.8% (w/w) was obtained using T. shinodae with 6.2% (v/v) inoculum volume, pH 3.6, C/N ratio 105, 1.52 (g/L) of MgSO4, and 4.55 mM FeSO4 in 120.72 h at 30 °C. Lipid composition from T. shinodae depicted the presence of linoleic acid (C18:2), oleic acid (C18:1), stearic acid (C18:0), palmitic acid (C18:0), myristic acid (C14:0), and lauric acids (C12:0), respectively. T. shinodae lipids have 61.1% (w/w) saturated fatty acids and unsaturated fatty acids proportion accord to 38.9% (w/w). Lipid composition of T. shinodae indicates that these lipids were suitable for synthesis of high value products like fuel additives, surfactants, detergents, and cleaning applications. Value-added compounds like alpha-linolenic acid, 1,3-propanediol (1,3-PDO), fatty acid methyl esters (FAME), keto-carotenoids, lactic acid, 2,3-butanediol, citric acid, lutein, polyunsaturated fatty acids (PUFAs), 1,3-dihydroxyacetone, oleic acid, propionic acid, and succinic acid have huge applications in textile, cosmetic, food, metallurgy, and pharmaceutical industries. These microbial-based lipids have various industrial and technology applications as printing inks, lubricants, coatings, polymers, solvents, leather processing, hydraulic fluids, surfactants, glycerin (glycerol), pesticide/herbicide adjuvants, and fuels. Biotechnological processes such as medium engineering and metabolic engineering can enhance lipid production in microbes to a certain limit. Integrated biorefinery concept was introduced for cost reduction, efficient utilisation of feedstocks, high yield, zero waste discharges, high productivity, and economically viable technology development. It integrates the conversion of biomass to energy and other high-value products. Nonalcoholic fatty liver disease (NAFLD) is characterized by an increase in hepatic lipid accumulation due to impaired lipid metabolism. Although a correlation was found between NAFLD and sphingosine-1-phosphate (S1P), the role of the sphingolipid remains controversial. The aim of this study was to investigate any involvement of S1P in steatosis using its analog FTY720P and HepG2 cells. Lipid accumulation was induced by incubating the cells in a mixture of oleic and palmitic acid, and was quantified using Oil Red O. The involvement of signaling mediators was studied using pharmacological inhibitors and western blot analysis. FTY720P increased lipid accumulation, but this increase wasn't maintained in the presence of inhibitors of S1PR3, Gq, SREBP, mTOR, PI3K, and PPARγ indicating their involvement in the process. The results revealed that FTY720P binds to S1PR3 which activates sequentially Gq, PI3K, and mTOR leading to an increase in SREBP expression and PPARγ activation. Here we used cryo-electron microscopy (cryo-EM), double electron-electron resonance spectroscopy (DEER), and molecular dynamics (MD) simulations, to capture and characterize ATP- and substrate-bound inward-facing (IF) and occluded (OC) conformational states of the heterodimeric ATP binding cassette (ABC) multidrug exporter BmrCD in lipid nanodiscs. Supported by DEER analysis, the structures reveal that ATP-powered isomerization entails changes in the relative symmetry of the BmrC and BmrD subunits that propagates from the transmembrane domain to the nucleotide binding domain. The structures uncover asymmetric substrate and Mg2+ binding which we hypothesize are required for triggering ATP hydrolysis preferentially in one of the nucleotide-binding sites. MD simulations demonstrate that multiple lipid molecules differentially bind the IF versus the OC conformation thus establishing that lipid interactions modulate BmrCD energy landscape. Our findings are framed in a model that highlights the role of asymmetric conformations in the ATP-coupled transport with general implications to the mechanism of ABC transporters. Bisphenol A (BPA), a typical environmental endocrine disruptor, is an "obesogen" that can induce lipid accumulation in the liver. Highly similar in structure to BPA, bisphenol F (BPF) is becoming the dominant BPA substitute on the market, which attracts more and more attention due to its potential adverse effects. Recently, BPF exposure is found to cause non-alcoholic fatty liver disease (NAFLD)-like changes; however, the underlying toxic effects remain poorly understood. Therefore, in the current study, we focused on BPF-mediated lipid homeostasis, especially the alterations of lipid components and metabolism. In human serum, the BPF levels in healthy controls and NAFLD patients were assessed by ELISA, and BPF-induced disturbance of lipid metabolism was evaluated in mouse model via non-targeted lipomic methods with ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry. It suggested that BPF exposure was positively correlated with NAFLD severity and triglyceride level in patients. Based on the relationships, lipid metabolites were assessed in mouse livers between control and BPF-treated group, and it revealed that twenty-six lipid metabolites (including phospholipids, sphingolipids, and glycerides) were significantly changed in mouse livers. Phosphatidylcholine, phosphatidylethanolamine, and diglyceryl ester levels were lower than those in the control mice; hexose ceramide content in sphingolipids markedly increased in BPF-treated mouse livers. Noteworthily, the glycerophospholipid metabolic pathway was found to be the most pronounced in BPF-induced disturbance of lipid metabolism. Therefore, the current study, for the first time, is deciphering the BPF-induced lipid metabolic disturbance, which may provide novel intervention strategies for BPF-induced NAFLD-like changes. Fisetin (FST) is a naturally occurring flavonol that has recently emerged as a bioactive phytochemical with an impressive array of biological activities. To the author knowledge, boosting the activity of FST against severe acute pancreatitis (SAP) through a nanostructured delivery system (Nanophytomedicine) has not been achieved before. Thereupon, FST-loaded lipid polymer hybrid nanoparticles (FST-loaded LPHNPs) were prepared through conjoined ultrasonication and double emulsion (w/o/w) techniques. Comprehensive in vitro and in vivo evaluations were conducted. The optimized nanoparticle formula displayed a high entrapment efficiency % of 61.76 ± 1.254%, high loading capacity % of 32.18 ± 0.734, low particle size of 125.39 ± 0.924 nm, low particle size distribution of 0.357 ± 0.012, high zeta potential of + 30.16 ± 1.416 mV, and high mucoadhesive strength of 35.64 ± 0.548%. In addition, it exhibited a sustained in vitro release pattern of FST. In the in vivo study, oral pre-treatment of FST-loaded LPHNPs protected against l-arginine induced SAP and multiple organ injuries in rats compared to both FST alone and plain LPHNPs, as well as the untreated group, proven by both biochemical studies, that included both amylase and lipase activities, and histochemical studies of pancreas, liver, kidney and lungs. Therefore, the study could conclude the potential efficacy of the novel phytopharmaceutical delivery system of FST as a prophylactic regimen for SAP and consequently, associated multiple organ injuries. Creation of bioactive molecules for treatment of cardiovascular diseases based on natural peptides is the focus of intensive experimental research. In the recent years, it has been established that C-terminal fragments of apelin, an endogenous ligand of the APJ receptor, reduce metabolic and functional disorders in experimental heart damage. The review presents literature data and generalized results of our own experiments on the effect of apelin-13, [Pyr]apelin-13, apelin-12, and their chemically modified analogues on the heart under normal and pathophysiological conditions in vitro and in vivo. It has been shown that the spectrum of action of apelin peptides on the damaged myocardium includes decrease in the death of cardiomyocytes from necrosis, reduction of damage to cardiomyocyte membranes, improvement in myocardial metabolic state, and decrease in formation of reactive oxygen species and lipid peroxidation products. The mechanisms of protective action of these peptides associated with activation of the APJ receptor and manifestation of antioxidant properties are discussed. The data presented in the review show promise of the molecular design of APJ receptor peptide agonists, which can serve as the basis for the development of cardioprotectors that affect the processes of free radical oxidation and metabolic adaptation. Ocean-related global changes have altered phytoplankton community structure, especially the diatom-dinoflagellate competition, which further influences ecosystem structure and functions. The pivotal ecological roles of diatoms and dinoflagellates are strongly related with their biochemical composition, but quantitative comparisons of biochemical changes between diatoms and dinoflagellates under variable environments are still limited. We investigated responses of lipid biomarkers (sterols and fatty acids (FAs)) to different temperatures (12, 18 and 24 °C), nitrogen and phosphorus concentrations and their molar ratios (N:P ratios) of 10:1, 24:1 and 63:1 in marine diatom Phaeodactylum tricornutum and dinoflagellate Prorocentrum minimum. Over these wide ranges of temperature and nutrient conditions, sterol and FA profiles were relatively stable in the two species. For carbon-normalized contents of major lipid biomarkers, warming caused non-significant changes in the diatoms but an increase (up to 153%) in the dinoflagellates. Eutrophication caused an overall decrease (up to 53%) in major lipid biomarker contents in the diatoms but an overall increase (up to 77%) in the dinoflagellates. In contrast, imbalanced N:P ratios caused an overall increase (up to 64%) in major lipid biomarker contents in the diatoms but an overall decrease (up to 53%) in the dinoflagellates. Taken together, our results showed that the contents of major sterols and polyunsaturated FAs would change non-significantly in the diatom P. tricornutum, while those in the dinoflagellate P. minimum would increase (ca. 9–48%) under future ocean warming, eutrophication and imbalanced N:P ratios. This study expands our knowledge on lipid-based indicators of phytoplankton under changing environments, which by systematically linking with several other aspects of food quality will help to understand ecological consequences of diatom-dinoflagellate community changes. Multiple myeloma (MM) is the second most common hematological malignancy worldwide, characterized by abnormal proliferation of malignant plasma cells within a tumor-permissive bone marrow microenvironment. Metabolic dysfunctions are emerging as key determinants in the pathobiology of MM. In this review, we highlight the metabolic features of MM, showing how alterations in various lipid pathways, mainly involving fatty acids, cholesterol and sphingolipids, affect the growth, survival and drug responsiveness of MM cells, as well as their cross-talk with other cellular components of the tumor microenvironment. These findings will provide a new path to understanding the mechanisms underlying how lipid vulnerabilities may arise and affect the phenotype of malignant plasma cells, highlighting novel druggable pathways with a significant impact on the management of MM. We studied diverse prenylated intrinsically disordered regions (PIDRs) of Ras and Rho family small GTPases using long timescale atomistic molecular dynamics simulations in an asymmetric model membrane of phosphatidylcholine (PC) and phosphatidylserine (PS) lipids. Here we show that conformational plasticity is a key determinant of lipid sorting by polybasic PIDRs and provide evidence for lipid sorting based on both headgroup and acyl chain structures. We further show that conformational ensemble-based lipid recognition is generalizable to all polybasic PIDRs, and that the sequence outside the polybasic domain (PBD) modulates the conformational plasticity, bilayer adsorption, and interactions of PIDRs with membrane lipids. Specifically, we find that palmitoylation, the ratio of basic to acidic residues, and the hydrophobic content of the sequence outside the PBD significantly impact the diversity of conformational substates and hence the extent of conformation-dependent lipid interactions. We thus propose that the PBD is required but not sufficient for the full realization of lipid sorting by prenylated PBD-containing membrane anchors, and that the membrane anchor is not only responsible for high affinity membrane binding but also directs the protein to the right target membrane where it participates in lipid sorting. Lipids of air-dried leaves of the hemi-halophyte plant Limonium gmelinii (Willd.) Kuntze (Plumbaginaceae) growing on the dry bed of the Aral Sea were studied. The leaves contained neutral lipids (NL, 0.84% of the mass), glycolipids (GL, 1.78), and phospholipids (PhL, 0.74). The compositions of the lipophilic constituents of the NL and hydrocarbons were determined. Fatty acids of NL were dominated by 18:2n6 (53.45%); of GL and PhL, 16:0 (40.14 and 53.98%, respectively). Inhomogeneity in single molecule electron transfer at the surface of lipid in a single vesicle has been explored by single molecule spectroscopic technique. In our study we took Di-methyl aniline (DMA), as the electron donor (D) and three different organic dyes as acceptor. These dyes are C153, C480 and C152 and they reside in different regions in the vesicle depending upon their preference of residence. For each probe, we found fluctuations in the single-molecule fluorescence decay, which are attributed to the variation in the reactivity of interfacial electron transfer. We found a non-exponential auto-correlation fluctuation of the intensity of the probe, which is ascribed to the kinetic disorder in the rate of electron transfer. We have also shown the power law distribution of the dark state (off time), which obeys the levy's statistics. We found a shift in lifetime distribution for the probe (C153) from 3.9 ns to 3.5 ns. This observed quenching is due to the dynamic electron transfer. We observed the kinetic disorderness in the electron transfer reaction for each dye. This source of fluctuation in electron transfer rate may be ascribed to the inherent fluctuation, occurring on the time scale of ~ 1.1 ms (for C153) of the vesicle, containing lipids. Sphingolipids play crucial roles in cellular membranes, myelin stability, and signalling responses to physiological cues and stress. Among them, sphingosine 1-phosphate (S1P) has been recognized as a relevant biomarker for neurodegenerative diseases, and its analogue FTY-720 has been approved by the FDA for the treatment of relapsing–remitting multiple sclerosis. Focusing on these targets, we here report three novel polymeric capture phases for the selective extraction of the natural biomarker and its analogue drug. To enhance analytical performance, we employed different synthetic approaches using a cationic monomer and a hydrophobic copolymer of styrene-DVB. Results have demonstrated high affinity of the sorbents towards S1P and fingolimod phosphate (FTY-720-P, FP). This evidence proved that lipids containing phosphate diester moiety in their structures did not constitute obstacles for the interaction of phosphate monoester lipids when loaded into an SPE cartridge. In this study, it is aimed to design a smart drug delivery system for the diagnosis and treatment of cancer cells. Magnetic-solid lipid nanoparticles were synthesized as a drug delivery system and azathioprine (AZA) was loaded into this system. This newly created system was radiolabeled using technetium-99 m with a radiolabeling yield of 90.3%. Drug loading into the SLMNP was found as 86%. Purpose of Review: Stroke is the second leading cause of death and disability-adjusted life years worldwide, and the global lifetime risk of stroke is rising. Moreover, patients with a prior stroke are at high risk of recurrent events. We aimed at reviewing the evidence supporting aggressive secondary prevention strategies for lipid-lowering treatment in this population. Recent Findings: Statins are the key players in such aggressive management; however, stroke survivors remain at significant residual risk suggesting the need for both better implementation of statin use as well as additional lipid lowering therapies. Newer drugs have become available and represent important tools in the management of patients with prior ischemic stroke. The role of lipid lowering treatment in hemorrhagic stroke is more controversial, given epidemiological data linking low lipid levels with increased risk of first and recurrent events. Summary: Aggressive secondary prevention strategies, including lipid lowering treatments, have proven to mitigate the risk of recurrent events in post-stroke patients. Underlying mechanisms of the inverse relationship between moderate alcohol consumption and cardiometabolic disorders are unclear. Modification by types of alcoholic beverages consumed and drinking pattern remains understudied. We aimed to provide insight into the mechanisms by examining 14 insulinemic/glycemic, inflammatory and lipid markers. We used cross-sectional data from 15,436 women in the Nurses' Health Study, 19,318 women in the Nurses' Health Study II, and 6872 men in the Health Professionals Follow-up Study. Multivariable linear regression was used to estimate the percentage differences in biomarker concentrations according to alcohol intakes. The average alcohol intake in the combined cohort was 3.3 servings/week. We found a 1 serving/d increment in alcohol intake (14 g ethanol, 44 ml liquor or 355 ml beer or 118 ml wine per day) was associated with a 0.6% lower level of HbA1c, 1.7–3.6% lower proinflammatory markers and 4.2% higher adiponectin, as well as 7.1% higher HDL-cholesterol and 2.1% lower triglyceride with a significant linear trend. Wine, especially red wine, was associated with lower inflammation in particular. Beer had weaker favorable to null associations with blood lipids and adiponectin. Liquor was associated with higher C-peptide and interleukin-6, yet equally associated with lower HbA1c and higher HDL-cholesterol as other beverages. Drinking 3 days or more per week was related to a better biomarker profile than nonregular drinking independent of intake levels. Drinking appeared to have similar associations irrespective whether done with meals or not. Our data indicated moderate alcohol intake, especially if consumed from wine and done regularly, was associated with favorable profiles of insulinemic/glycemic and inflammatory markers and blood lipids. Objectives: This study aimed to explore the joint effect of body mass index (BMI) and serum lipids levels on incident dementia. Methods: We prospectively followed up with 1,627 dementia-free community residents aged ≥60 for 5.7 years on average. At baseline, weight, and height were measured, and total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were detected in serum. Demographic characteristics were collected through questionnaires. Dementia was based on consensus diagnosis of neurologists and neuropsychologists using DSM-IV criteria. Additive Cox proportional model was used to assess the exposure-response relationship between BMI and serum lipid levels and dementia risk. Interactions and further classifications of BMI and serum lipid levels were further presented by bivariate surface models and decision-tree models. Results: The joint effects of TC with BMI, TG with BMI, and LDL-C with BMI on the risk of incident dementia shared a similar pattern, different from their independent exposure-response curves. The joint effect of HDL-C with BMI showed an S-surface but without statistical significance. Participants with TC<5.4 mmol/L and BMI<21 kg/m2 (Hazard Ratio(HR) 1.93, 95% Confidence Interval (CI) 1.05-3.53), TC<5.4 mmol/L and BMI≥21 kg/m2 (HR 1.73, 95% CI 1.09-2.72), and TC≥5.4 mmol/L and BMI<21 kg/m2 (HR 4.02, 95% CI 2.10-7.71) were identified to have the increased risk of incident dementia compared to those with TC≥5.4 mmol/L and BMI≥21 kg/ m2. Participants with TG<1.7 mmol/L and BMI<21 kg/m2 had an increased risk of incident dementia compared to those with TG≥1.7 mmol/L and BMI≥21 kg/m2 (HR 1.98, 95%CI 1.17-3.3). Participants with LDL-C≥3.3 mmol/L and BMI<21 kg/m2 were identified to have an increased risk of incident dementia compared to those with LDL-C≥3.3 mmol/L and BMI≥21 kg/m2 (HR 3.33, 95%CI 1.64-6.78). Conclusions: Our study showed that low BMI combined with low or high levels of serum lipids may increase the risk of dementia among older adults. In this study, two rice varieties (RS4 and GZ93) with different amylose and lipid contents were studied, and their starch was used to prepare starch-palmitic acid complexes. The RS4 samples showed a significantly higher lipid content in their flour, starch, and complex samples compared to GZ93. The static in vitro digestion highlighted that RS4 samples had significantly lower digestibility than the GZ93 samples. The C∞ of the starch-lipid complex samples was found to be 17.7% and 18.5% lower than that of the starch samples in GZ93 and RS4, respectively. The INFOGEST undigested fractions were subsequently used for in vitro colonic fermentation. Short-chain fatty acids (SCFAs) concentrations, mainly acetate, and propionate were significantly higher in starch-lipid complexes compared to native flour or starch samples. Starch-lipid complexes produced a distinctive microbial composition, which resulted in different gene functions, mainly related to pyruvate, fructose, and mannose metabolism. Using Model-based Integration of Metabolite Observations and Species Abundances 2 (MIMOSA2), SCFA production was predicted and associated with the gut microbiota. While disorders in lipid metabolism have been associated with aging and age-related diseases, how lipid metabolism is regulated during aging is poorly understood. Here, we characterize the Drosophila endoribonuclease CG2145, an ortholog of mammalian EndoU that we named Age-related lipid regulator (Arlr), as a regulator of lipid homeostasis during aging. In adult adipose tissues, Arlr is necessary for maintenance of lipid storage in lipid droplets (LDs) as flies age, a phenotype that can be rescued by either high-fat or high-glucose diet. Interestingly, RNA-seq of arlr mutant adipose tissues and RIP-seq suggest that Arlr affects lipid metabolism through the degradation of the mRNAs of lipolysis genes – a model further supported by the observation that knockdown of Lsd-1, regucalcin, yip2 or CG5162, which encode genes involved in lipolysis, rescue the LD defects of arlr mutants. In addition, we characterize DendoU as a functional paralog of Arlr and show that human ENDOU can rescue arlr mutants. Altogether, our study reveals a role of ENDOU-like endonucleases as negative regulator of lipolysis. Fat is generally considered to have a negative effect on longevity but lipids can be beneficial for longevity. Purpose: Dietary intake may have pronounced effects on circulating biomarker concentrations. Therefore, the aim was to provide a descriptive overview of serum metabolite concentrations in relation to time since last meal, focusing on amino acids, lipids, one-carbon metabolites, and biomarkers of vitamin status. Methods: We used baseline data from the observational community-based Hordaland Health Study, including 2960 participants aged 46–49 years and 2874 participants aged 70–74 years. A single blood draw was taken from each participant, and time since last meal varied. Estimated marginal geometric mean metabolite concentrations were plotted as a function of time since last meal, up to 7 h, adjusted for age, sex, and BMI. Results: We observed a common pattern for nearly all amino acids and one-carbon metabolites with highest concentrations during the first 3 h after dietary intake. Homocysteine and cysteine were lowest the 1st hour after a meal, while no patterns were observed for glutamate and glutamic acid. The concentrations of phylloquinone and triglycerides were highest 1 h after dietary intake. Thiamine and thiamine monophosphate concentrations were highest, while flavin mononucleotide concentrations were lowest within the first 2 h after a meal. No clear patterns emerged for the other fat-soluble vitamins, blood lipids, or B-vitamin biomarkers. Conclusion: Our findings suggest that distinguishing between "fasting" and "non-fasting" blood samples may be inadequate, and a more granular approach is warranted. The vegetative organs of common basil (Ocimum basilicum L.) grown in water culture at different levels of illumination (50, 100, and 150 μmol PAR quanta/(m2 s)) for 21 days were studied. In the work, the Purple Velvet basil variety, which is popular in agriculture and has intensely colored leaves was used. The biomass, water content, and composition of fatty acids (FA) of total lipids in the leaves and roots of plants and the content of malondialdehyde and chlorophylls (a, b) in the aerial parts of plants were determined. The sensitivity of the FA composition and morphophysiological parameters (biomass and chlorophyll content in leaves) of O. basilicum plants to the intensity of illumination was shown. The greatest effect was caused by the illumination intensity mode of 150 µmol/(m2 s). Species composition of O. basilicum FA under all lighting conditions was wider in root lipids; however, more pronounced changes in the qualitative composition of FAs were observed in leaves. Photodependent regulation of FA component composition of O. basilicum manifested itself in an increase in the proportion of unsaturated FAs, especially polyene ones, which led to an increase in the unsaturation index (UI) of esterified lipid FAs. The highest UI values were obtained with illumination of 150 µmol/(m2 s). At the same time, with an increase in the illumination intensity, a significant increase in the activity of ω-3 and ω-9-desaturases occurred, which indicates in favor of their light-dependent activation. Thus, increasing the intensity of lighting to certain values directly proportionally affects the physiological parameters of O. basilicum. The maximum indicators of productive growth and the implementation of adaptive mechanisms of green and underground parts of basil plants correspond to illumination of 150 µmol/(m2 s). Effects of 1 M NaCl on the lipid profile of detergent-resistant membranes in chloroplasts and mitochondria of salt-accumulating halophyte plants of family Amaranthaceae, Salicornia perennans Willd. and Suaeda salsa (L.) Pall. were studied. The composition of lipids and fatty acids in detergent-resistant membranes differed from total lipids of chloroplast and mitochondrial membranes by the abundance of cerebrosides and sterols. Under the given salinity level, a manyfold increase in the relative content of cerebrosides was noted in the composition of detergent-resistant chloroplast membranes of S. perennans. A similar salinity-related increase in cerebroside content was detected in detergent-resistant mitochondrial membranes of S. salsa. The opposite effect was observed for sterols: their relative content decreased under the action of salt. The results indicate that the detergent-resistant membranes are involved in the interactions of chloroplasts and mitochondria in the cell response of halophytes to salinity. [ABSTRACT FROM AUTHOR]
