Objectives: To determine the antimicrobial activity of silver nano-particles(AgNPs) with tetracycline and ampicillin against multi-drug resistance (MDR) and extensively-drug resistance (XDR) Salmonella typhi. Methods: Cross sectional non-probability purposive study was conducted from September, 2021 to May, 2022 at Microbiology department PNS Shifa, Hospital Karachi. Blood cultures of patients suspicious of typhoid fever were collected and incubated in automated Bact/Alert system. Positive cultures were identified on blood and MacConkey and processed by API-10S, confirmed by serotyping (O9 antisera) (SSI Diagnostica's Salmonella). Antibiotic resistance was done by Kirby-Bauer disk diffusion (Sigma and Rich). MDR and XDR isolates were preserved in Brain Heart Infusion in a volume of 2ml in screw capped bottles at -70°C. Antimicrobial powders (ampicillin and tetracycline (Alfa Aesar) weighed by an electrical weighing balance (OHAUS) to take 1mg of antimicrobial drug. Absorbance spectra of serial concentrations of antibiotics (UV-Vis spectrophotometer (Mole-Qule-) AgNPs (10nm) (nanocomposix) + Antibiotic in (1:1 volume ratio). Conjugation of silver nanoparticles with tetracycline and ampicillin was done by FTIR (thermos scientificThermos ScientificNicolet 50). Results: Out of 77 isolates, 54 were resistant to ceftriaxone (XDR) and 23 sensitive to ceftriaxone (MDR). All isolates were susceptible to azithromycin and meropenem. Comparison of zone of inhibitions of ampicillin and Amp-AgNPsas and tetracycline with Tet-AgNPs was done. Minimal inhibitory concentration was also done to determine antimicrobial activity. Conclusion: Significant synergistic inhibitory effects against Salmonella Typhi isolates were obtained by combination of tetracycline with silver nano-particles even at low concentration. The emergence of antibiotic-resistant bacteria necessitates the development of novel, sustainable, and biocompatible antibacterial agents. This study addresses cytotoxicity and environmental concerns associated with traditional silver nanoparticles (AgNPs) by exploring lignin, a readily available and renewable biopolymer, as a platform for AgNPs. We present a novel one-pot synthesis method for lignin-based AgNPs (AgNPs@AL) nanocomposites, achieving rapid synthesis within 5 min. This method utilizes various organic solvents, demonstrating remarkable adaptability to a wide range of lignin-dissolving systems. Characterization reveals uniform AgNP size distribution and morphology influenced by the chosen solvent. This adaptability suggests the potential for incorporating lignin-loaded antibacterial drugs alongside AgNPs, enabling combined therapy in a single nanocomposite. Antibacterial assays demonstrate exceptional efficacy against both Gram-negative and Gram-positive bacteria, with gamma-valerolactone (GVL)-assisted synthesized AgNPs exhibiting the most potent effect. Mechanistic studies suggest a combination of factors contributes to the antibacterial activity, including direct membrane damage caused by AgNPs and sustained silver ion release, ultimately leading to bacterial cell death. This work presents a straightforward, adaptable, and rapid approach for synthesizing biocompatible AgNPs@AL nanocomposites with outstanding antibacterial activity. These findings offer a promising and sustainable alternative to traditional antibiotics, contributing to the fight against antibiotic resistance while minimizing environmental impact. The unearthing of various forms of broad and narrow-spectrum antibiotics have helped mankind combat various forms of microbic infections ranging from protozoal to bacterial. The applicability of antibiotics has been decisive in a wide range of frontline therapeutic dealings. An extensive use of antibiotics and certain genetic modifications in microbes has paved the way for a new issue to be answered by the scientific community, an antibiotic resistance. An unparallel infections triggered by multidrug-resistant organisms (MDRO) account to be a major cause of mortality and morbidity, today this situation demands the development of alternative strategies of cure and medicine by the scientific community. Study shows the amplification of the effect of antibiotics may be a possible route to address antibiotic resistivity. The assortment of various surface properties of nanoparticles (NPs) coupled with their ability to efficiently bind with bacterial surfaces leading to the rupture and lysis of cell deliberates potential strategies to manage infections caused by MDROs.The present study scrutinizes the amplification of the antibacterial activity of ciprofloxacin (CIP) against the bacterial strains of Staphylococcus aureus, Escherichia coli, Pseudomonas and Streptococcus mutans on coupling with synthesized copper, silver and zinc oxide NPs (ZnO NPs) by disc diffusion technique. The current study describes the efficacious synthesis and characterization of ZnONP, copper Nps (CuNPs) and silver NPs (AgNPs) followed by the induced amplification of the antibacterial activity of CIP by coupling it with synthesized metal/metal oxide (M/MO NP). The synthesized M/MO NPs were characterized using XRD and FE-SEM studies and TEM. The results indicated that among the ZnONP, AgNP, the Cu NPs potentiate the bactericidal efficacy of CIP antibiotics. The growing resistance of bacteria to antibiotics is one of the main public health problems nowadays. The influence of silver nanoparticle (AgNP) pretreatment of 220 cows with mastitis on the susceptibility of Staphylococcus epidermidis bacteria to 31 antibiotics was studied. The obtained results were compared with the previous results for Escherichia coli, Streptococcus dysgalactiae, and Staphylococcus aureus. For all four bacteria, an increase in susceptibility (9.5–21.2%) to 31 antibiotics after cow treatment with AgNPs was revealed, while after first-line antibiotic drug treatment as expected, the susceptibility decreased (11.3–27.3%). These effects were explained by (1) the increase in the contribution of isolates with efflux effect after antibiotic treatments and its decrease after AgNP treatment and (2) the changes in bacteria adhesion and anti-lysozyme activity after these treatments. The effect of the increasing antibacterial activity of antibiotics after AgNP treatment was the most pronounced in the case of E. coli and was minimal in the case of S. epidermidis. With AgNP treatment, the time of recovery decreased by 26.8–48.4% compared to the time of recovery after treatment with the first-line antibiotic drugs. For many years, combination therapies with two or more antimicrobial agents have gained interest to address the problem of emerging antibiotic resistance. Several studies have shown that the addition of AgNPs enhances the antimicrobial activity of antibiotics against resistant pathogens. However, the antibacterial activity of phytosynthesised silver nanoparticles (AgNPs) in combination with antibiotics has not been extensively studied. Therefore, this study aimed to investigate the antibacterial activity of the phytosynthesised AgNPs with different antibiotics against two common pathogenic bacteria, namely methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa. A green method was used to produce AgNPs, where Persicaria odorata extract acted as a reducing and stabilising agent in the aqueous synthesis process. The production of AgNPs was verified through various analytical techniques, including ultraviolet–visible spectroscopy, X-ray diffraction, Fourier transform infrared spectroscopy, high-resolution transmission electron microscopy and energy-dispersive X-ray. Notably, strong absorption spectrum corresponding to the surface plasmon resonance peaks of AgNPs was found at 436 nm, with particles ranging from 10 to 35 nm. The disc diffusion technique was used to investigate the antibacterial activity of AgNPs, both alone and in combination with antibiotics. Initially, the MRSA and P. aeruginosa showed 100 % resistance to antibiotics, especially ampicillin. However, the disc diffusion result exhibit a significant increase in the inhibition zone when ampicillin-AgNPs, cefoxitin-AgNPs and chloramphenicol-AgNPs were used in combination. In short, the findings of this study showed that AgNPs exhibit a promising synergy when used in combination with antibiotics, opening up possibilities for future combination therapies against antibiotic-resistant pathogens.
