Nucleoside analogen zijn als therapeutische klasse reeds lang gek end voor hun antivirale werking. Onder hen vormen de acyclische nucleosi de fosfonaten een aparte subklasse waaronder een aantal sterk actieve an alogen met een breed spectrum aan antivirale activiteit. Als voorbeeld k unnen hier het 9-(S)-[3-hydroxy-2-(phosphono-methoxy)propyl]cytosine&nbs p;[(S)-HPMPC; cidofovir, Vistide] en het 9-[2-(phosphono-methoxy)ethyl]a denine [PMEA, adefovir, Viread] aangehaald worden. Deze worden resp ectievelijk gebruikt voor de behandeling van cytomegalovirus retinitis i n immunogecomprimeerde patiënten, en voor de behandeling van AIDS. In onze speurtocht naar krachtige en selectieve antivirale nucleoside fo sfonaten, werden de synthese en eigenschappen bestudeerd van analogen me t een supplementaire amine groep in de zijketen. Verschillende analogen werden bereid en geëvalueerd, maar slechts een beperkte activiteit kon a angetoond worden tegen het herpes simplex (HSV) en varicella zoster viru s (VZV). In een tweede deel van ons onderzoekswerk, werden dezelfde acycli sche nucleoside analogen aangewend als bouwstenen om kleine stukjes DNA analogen te maken. De aldus bekomen aminopropyl nucleïnezuren (APNAs) zo uden het meest eenvoudige alternatieve nucleïnezuur kunnen vormen. In he t verleden werd reeds de theorie geopperd dat RNA ribonucleïnezuur d e voorloper was van het DNA, dat nu voor alle levensvormen op aarde het centrale informatiesysteem vormt. Het DNA zou dus uit het RNA geëvolueer d zijn als informatiedrager. Maar vermits RNA zelf reeds een complex mol ecule is, wat zou dan de voorloper van RNA geweest zijn? Een recente publicatie suggereert dat een ander nucleïnezuur de dienst k an uitgemaakt hebben, namelijk het (L)-a-threofuranosyl analoog of TNA. In dit rapport werd aangetoond dat TNA strengen een stabiele dubbele hel ix kunnen vormen met zowel complementaire DNA als RNA strengen. Deze eig enschap wordt juist beschouwd als één van de noodzakelijke voorwaar den voor een systeem om als voorloper van RNA te kunnen functioneren. An derzijds is er recent ook heel wat belangstelling voor potentiële nieuwe antisense moleculen (dit is een strategie voor het blokkeren van de tra nsfer van informatie voor het aanmaken van proteïnen). Daarom werden ami nopropyl oligonucleotiden ontwikkeld (R en S-3 -APNA evenals R en S-2 -A PNA) door reductie van het aantal koolstofatomen van een NH-TNA nucleoti de structuur met terzelfdertijd een vermindering van het aantal chirale centra. Hierbij dienden een aantal nieuwe synthese schema s te worden on twikkeld. Fysicochemische studie van de nieuw gevormde structuren toonde echter aan dat na inbouw van enkele acyclische eenheden, de affiniteit voor DNA strengen snel achteruit gaat. Met behulp van UV absorptie studi es kon helaas aangetoond worden dat geen enkele interactie plaats grijpt , noch wat betreft de vorming van acyclische duplexen, noch wat betreft de kruisgewijze paring met DNA of RNA strengen. De afgelopen decennia werd veel onderzoek gedaan naar nucleïnezuur analo gen en hun monomere bouwstenen. Dit niet alleen omdat deze enzymatisch s tabiele nucleïnezuur analogen een belangrijke rol kunnen spelen in versc hillende domeinen zoals therapie (antisense en siRNA technologie, aptame ren, ribozymes, ..), diagnose (het gebruik van specifieke nucleïnezuur p robes) en biotechnolgie (PCR, sequentiebepaling), maar ook uit interesse naar het onstaan van het leven op aarde en meer concreet het onstaan va n de eerste moleculen die een rol spelen in het opslaan en de transmissi e van de genetische informatie. De suiker-gemodificeerde nucleïnezuur an alogen TNA (tetrose nucleïnezuur) en CeNA (cyclohexeen nucleïnezuur) vor mden het onderwerp van mijn doctoraatsonderzoek. TNA is, structureel, een eenvoudiger nucleïnezuur dan RNA, dat gesynthet iseerd kan worden uitgaande van natuurlijke beginproducten. Bovendien is TNA in staat te hybridiseren met DNA en RNA. Uitgaande van deze eigensc happen kan TNA beschouwd worden als een voorloper of alternatief van RNA bij het ontstaan van leven op aarde. In dit onderzoek werd aangetoond d at ribozymes (die beschouwd worden als katalysatoren in de eerste stadia van leven op aarde) die de gemodificeerde tetrose nucleosiden bevatten, hun catalytische activiteit bijna volledig verliezen. We kunnen daarom besluiten dat TNA structureel te sterk verschilt van RNA om het bestaan van een catalytisch actief RNA/TNA chimeer ribozyme toe te laten. Verder werd de enzymatische herkenning van TNA bouwstenen door polymerasen nag egaan. Hoewel threosyl nucleoside trifosfaten herkend worden door natuur lijke polymerasen is een enzymatische synthese van TNA beperkt tot twee opeenvolgende bouwstenen. Een tweede deel van dit onderzoek gaat dieper in op de enzymatische herk enning van cyclohexeen nucleoside trifosfaten. Dit onderzoek beoogt een enzymatische synthese van CeNA uitgaande van de trifosfaat bouwstenen. T ot nu toe is de aanmaak van CeNA oligomeren enkel mogelijk via een chemi sche synthese welke niet alleen omslachtig is, maar ook een overmaat aan reagentia vereist. Een enzymatische CeNA synthese zou een grote vooruit gang zijn en zou een eenvoudige productie van CeNA oligomeren toelaten, welke dan verder onderzocht kunnen worden voor hun rol in therapie, diag nose of biotechnolgie. CeNA is immers een stabiel nucleïnezuur analoog d at bovendien sterk hybridiseert met de natuurlijk nucleïnezuren DNA en R NA. Er werd aangetoond dat natuurlijke DNA polymerasen in staat zijn om tot zeven opeenvolgende cyclohexeen nucleoside trifosfaten in te bouwen in een DNA hybride. Deze synthese is echter niet efficient. Daarom werd met gestuurde evolutietechnieken getracht een polymerase te ontwikkelen dat beter geschikt is voor de incorporatie van cyclohexeen bouwstenen.&n bsp; Compartmentalized self replication werd gebruikt voor de selectie van polymerasen met aangepaste substraat specificitieit. Uitgaande van polymerase bibliotheken waarbij diversiteit is geïntroduceerd in de A-mo tief en de B-motief regio van het Taq DNA polymerase konden verschillend e mutanten geselecteerd worden met een verbeterde CeATP incorporatie cap aciteit (in vergelijking met Taq wild type).
