Cilj ovog rada bio je istražiti enzimatsku reakciju kataliziranu sintazom 2-sukcinil-5- enolpiruvil-6-hidroksi-3-cikloheksen-1-karboksilne kiseline uz kratkolančani α-ketoglutarat kao donorski supstrat, akrilonitril kao akceptorski supstrat, tiamin difosfat kao koenzim, te magnezijev sulfat kao kofaktor. Istraživan je utjecaj različitih vrijednosti temperature, pH i koncentracije svih reakcijskih komponenti na brzinu reakcije. Iz prikupljenih eksperimentalnih podataka su procijenjeni kinetički parametri Michaelis-Menteničinog modela koji je kasnije validiran eksperimentom u kotlastom reaktoru. The aim of this work was to investigate the enzymatic reaction catalyzed by 2-succinyl-5- enolpyruvyl-6-hydroxy-3-cyclohexene-1-carboxylate synthase with short-chain α- ketoglutarate as the donor substrate, acrylonitrile as the acceptor substrate, thiamine diphosphate as the coenzyme, and magnesium sulfate as the cofactor. The effect of different temperatures, pH values and concentrations of all reaction compounds on reaction rate has been investigated. From the gathered experimental data kinetic parameters of the Michaelis-Menten model type have been estimated. The mathematical model that was developed has been experimentally validated.
