The novel magnetic bio-char(MBC), which was derived from sewage sludge in municipal wastewater treatment plant after pyrolysis and Fe2+/Fe3+ modification by loading nano-sized iron oxide particles.The MBC can be used for the adsorption of heavy metal ions in aqueous system remediation. The physical/chemical characteristics of MBC were revealed by VSM, SEM-EDS, XRD, FTIR and batch adsorption experiments. The results showed that the nano-sized γ-Fe2O3 particles grew on the surface of bio-char uniformly, and the saturation magnetization of MBC reached 13.53Am²/kg. In adsorption experiments, when the dosage of MBC was 1.25g/L, adsorption time was 24h, pH was 5.0, the adsorption capacity was 67.68mg/g which increased by 60.08% than that of bio-char. The Langmuir isothermal and pseudo-second-order model can describe adsorption process of magnetic bio-char well. The MBC derived from sewage sludge has significant adsorption capacity, which is easy to separate from aqueous system. This disposition can recycle solid waste to pollution treatment. Objective: To establish and evaluate a double Interferon-gamma(IFN-γ) antibody sandwich Enzyme-linked immunosorbent assay(ELISA)which was based on immunomagnetic beads for human IFN-γ detection. Methods: Magnetic nanoparticle were prepared through a hybrid miniemulsion method and coupled with hunman IFN-γ antibody as solid phase. IFN-γ Antibody was coupled to magnetic nanoparticle. ELISA based on immunomagnetic beads for human IFN-γ detection was established and evaluated.Then the standard curve was drawn and assay performance characteristics of the new method was evaluated. Results: Immunoparticles coated human IFN-γ antibody were obtained with a coupling efficiency of 54.5 %. Immunoparticles were used to establish the double IFN-γ antibody sandwich system with detection range of 0-1000 pg/m L, coefficiency of correlation of 0.9996, sensitivity of 23.2 pg/m L,functional sensitivity of 0 pg/m L, and intra- and inter-assay coefficients of variation(CVs) <8 %. The total test time was 2 hours.Conclusions: A sandwich ELISA was successfully established for quantitative IFN-γ detection based on immunomagnetic beads. [ABSTRACT FROM AUTHOR]

To develop a magnetic nanoparticle chemiluminescence immunoassay (CLIA) for the determination of type Ⅰ procollagen N-terminal peptide (PINP) in human serum, we expressed a recombinant PINP-α1 protein in Corynebacterium glutamicum and used it as an immunogen to immunize BALB/c mice. We obtained three hybridoma cell lines that stably secret antibody against PINP-α1 protein. After further pairing and screening, we chose a monoclonal antibody 8C12 coupled with biotin as the capture antibody, and a monoclonal antibody 1F11 labeled horseradish peroxidase as the detection antibody. The antibodies combined with the serum samples, forming a sandwich complex which was used to detect the concentration of PINP in serum. After optimizing the conditions, we determined that the best working concentration of the capture antibody and the detection antibody were 3 μg/mL, and the incubation time was 30 minutes. The quantitative assay had a detection range of 5-1 100 ng/mL, with recovery rates between 93%-107% and the minimum detection limit of 1.22 ng/mL achieved. The intra-and inter-assay precisions were lower than 10%. The correlation coefficient of PINP results between this CLIA method and the Roche electrochemiluminescence immunoassay system was 0.906 2. Therefore, this CLIA method is specific and can be used to quantitatively detect the content of PINP in serum, which has the potential to become an auxiliary approach for bone disease examination.
