Objective: To compare the effectiveness of long and short proximal femoral nail anti-rotation (PFNA) in the treatment of type A2.3 intertrochanteric fracture of femur (IFF)., Methods: The clinical data of 54 patients with type A2.3 IFF admitted between January 2020 and December 2022 were retrospectively analyzed. According to the length of PFNA nail used in the operation, they were divided into long nail group (PFNA nail length>240 mm, 24 cases) and short nail group (PFNA nail length≤240 mm, 30 cases). There was no significant difference in baseline data such as gender, age, fracture side, body mass index, and time from fracture to operation between the two groups ( P >0.05). The operation time, intraoperative blood loss, intraoperative fluoroscopy frequency, intraoperative reduction quality score, fracture healing, and complications of the two groups were recorded and compared. Harris score was used to evaluate the hip function of patients at 1 year after operation. According to the relationship between the fracture line of type A2.3 IFF and the lesser trochanter, the two groups of patients were divided into type Ⅰ(the fracture line extends to the level of the lesser trochanter), type Ⅱ(the fracture line extends to less than 2 cm below the lesser trochanter), and type Ⅲ (the fracture line extends to more than 2 cm below the lesser trochanter), and the postoperative stability and internal fixator loosening of each subtype were evaluated., Results: The operation time, intraoperative blood loss, and intraoperative fluoroscopy frequency in short nail group were significantly less than those in long nail group ( P <0.05). There was no significant difference in the intraoperative reduction quality score between the two groups ( P >0.05). Patients in both groups were followed up 12-18 months, with an average of 13.5 months. The postoperative stability score of short nail group was significantly lower than that of long nail group ( P <0.05). The Harris score in the long nail group was significantly higher than that in the short nail group at 1 year after operation ( P <0.05), but there was no significant difference in Harris score grading between the two groups ( P >0.05). Complications occurred in 3 cases of the long nail group (including 1 case of coxa varus caused by external nail entry point and 2 cases of loose internal fixator), and 7 cases of the short nail group (including 1 case of coxa varus caused by external nail entry point and 6 cases of loose internal fixator). Neither group had any anterior femoral arch damage, there was no significant difference in the incidence of complications between the two groups ( P >0.05). The number of type Ⅲ patients was relatively small and not included in the statistics; there was no significant difference in the postoperative stability score and the incidence of internal fixator loosening between the long and short nail groups in type Ⅰ patients ( P >0.05). In type Ⅱ patients, the postoperative stability score and the incidence of internal fixation loosening in the long nail group were significantly better than those in the short nail group ( P <0.05)., Conclusion: Long PFNA fixation for type A2.3 IFF has longer operation time and more intraoperative blood loss, but the overall stability of fracture is better after operation. For type A2.3 IFF with fracture line extending to less than 2 cm below the lesser trochanter, long PFNA is used for fixation, although the surgical trauma is large, but the postoperative stability is better than that of short PFNA; for type A2.3 IFF with fracture line extending to the lesser trochanter, there is no significant difference in postoperative stability between long and short PFNAs.

Phosgene is not only a dangerous asphyxiating chemical warfare agent, but also an important chemical raw material, which is widely used in chemical production. According to statistics, there are more than 1 000 phosgene production enterprises in China, with an annual production volume of more than 3 million tons and hundreds of thousands of employees. Therefore, once the leakage accident occurs during production, storage and transportation, it often causes a large number of casualties. In the past 20 years, phosgene poisoning accidents in China have occurred from time to time, and due to the weak irritation, high density, and high concentration of phosgene at the scene of the accident, it often results in acute high-concentration inhalation of the exposed, triggering acute lung injury (ALI), and is very likely to progress to acute respiratory distress syndrome (ARDS), with a mortality rate up to 40%-50%. In view of the characteristics of sudden, mass, concealed, rapid and highly fatal phosgene, and the mechanism of its toxicity and pathogenicity is still not clear, there is no effective treatment and standardized guidance for the sudden group phosgene poisoning. In order to improve the efficiency of clinical treatment and reduce the mortality, this paper has summarized the pathophysiological mechanism of phosgene poisoning, clinical manifestations, on-site treatment, research progress, and innovative clinical therapies by combining the extensive basic research on phosgene over the years with the abundant experience in the on-site treatment of sudden mass phosgene poisoning. This consensus aims to provide guidance for the clinical rescue and treatment of patients with sudden mass phosgene poisoning, and to improve the level of treatment.

Objective: To investigate the evolution and clinical significance of HER2 low expression status in HER2 negative patients in primary and recurrent/metastatic breast cancers. Methods: The data and archived sections of 259 breast cancer patients with recurrence/metastasis and HER2-negative primary foci were collected from January 2015 to January 2022 at the Fourth Hospital of Hebei Medical University, and the HER2 status of primary and recurrence/metastasis foci was determined by immunohistochemistry (IHC), among which IHC 2+patients were subject to fluorescence in situ hybridization (FISH). The HER2 status was classified as HER2-0 group; patients with IHC 1+, IHC 2+and no FISH amplification were classified as HER2 low expression group; and patients with IHC 3+, IHC 2+and FISH amplified were classified as HER2-positive group. The changes of HER2 status in patients with HER2 low expression in primary versus recurrent/metastatic breast cancer foci were compared, and their clinicopathologic characteristics and prognosis were analyzed. Results: The overall concordance rate between primary and recurrent/metastatic HER2 status in breast cancer was 60.6% (157/259, κ=0.178). A total of 102 patients (102/259, 39.4%) had inconsistent primary and recurrent/metastatic HER2 status; 37 patients (37/259, 14.3%) had HER2-0 at the primary foci and HER2-low expression at the recurrent/metastatic; and 56 patients (56/259, 21.6%) had HER2-low expression in the primary foci and HER2-0 in the recurrent/metastatic. The recurrent/metastatic foci became low-expressing compared with the recurrent/metastatic foci which remained HER2-0 patients, with longer overall survival time, higher ER and PR positivity, lower Ki-67 positivity index, and lower tumor histological grade; all with statistically significant differences (all P <0.05). In the primary HER2-low group, patients with recurrent/metastatic foci became HER2-0 while those with recurrent/metastatic foci remained low expression; there were no statistically significant differences in clinicopathological features and overall survival time (all P >0.05). Conclusions: Unstable HER2 status in patients with HER2-0 and low expression in primary versus recurrent/metastatic breast cancer foci, and HER2-0 in the primary foci but low HER2 expression status in recurrence/metastasis is associated with favourable prognosis, and testing HER2 status in recurrence/metastasis can provide more treatment options for such patients.

Objective: To investigate the genetic and genomic profiling of juvenile myelomonocytic leukemia (JMML) and factors affecting its survival rate. Methods: Clinical characteristics, cytogenetics, molecular biology results and survival status of children with 27 JMML cases admitted to the Hematology Department of Children's Hospital, Capital Institute of Pediatrics from December 2012 to December 2021 were analyzed retrospectively, and the outcomes of the children were followed up. Kaplan-Meier method was used for survival analysis. Univariate analysis was used for analyzing factors affecting the overall survival (OS) rates of patients who received hematopoietic stem cell transplantation (HSCT). Log-Rank test was used for comparison of survival curves. Results: Among 27 JMML cases, there were 11 males and 16 females. The age of disease onset was 28 (11,52) months. There are 20 cases of normal karyotype, 4 cases of monosomy 7, 1 case of trisomy 8,1 case of 11q23 rearrangement and 1 case of complex karyotype. A total of 39 somatic mutations were detected.Those involved in RAS signal pathway were the highest (64%(25/39)), among which PTPN11 mutation was the most frequent (44% (11/25)). A total of 17 cases (63%) received HSCT, 8 cases (30%) did not receive HSCT, and 2 cases (7%) lost follow-up. For children receiving transplantation, the follow-up time after transplantation was 47 (11,57) months. The 1-year OS rate of high-risk transplantation group (17 cases) and high-risk non transplantation group (6 cases) was (88±8)% and (50±20)% respectively, with a statistically significant difference (χ 2 =5.01, P =0.025). The 5-year OS rate of the high-risk transplantation group was (75±11)%. The survival time of those who relapsed or progressed to acute myeloid leukemia after transplantation was significantly shorter than that of those who did not relapse (χ 2 =6.80, P =0.009). The OS rate of patients with or without PTPN11 mutation was (81±12) % and (67±19)% respectively (χ 2 =0.85, P =0.356). Conclusions: The main pathogenesis involved in JMML is gene mutation related to RAS signaling pathway, and the most common driver gene of mutation is PTPN11. Allogeneic HSCT can significantly improve the survival rate of high-risk JMML patients. The recurrence or progression after transplantation was related to poor prognosis.

This research and development of MenB meningococcal vaccines includes two technical routes: outer membrane vesicle (OMV) vaccines and recombinant protein vaccines. This article intends to review the development, production and application of MenB meningococcal OMV vaccines in order to provide a reference for the development of MenB meningococcal OMV vaccine in China.

Objective: To investigate the efficacy and safety of haploidentical hematopoietic stem cell transplantation (haplo-HSCT) in combination of ATG and post-transplant cyclophosphamide (PTCy) -induced immune tolerance after transplantation in treatment of childhood myelodysplastic syndromes(MDS）., Methods: From July 2016 to November 2020, a total of 8 children with MDS receiving the haploidentical allo-HSCT combined with ATG and PTCy-induced immune tolerance after transplantation in our hospital were enrolled, whose clinical data were retrospected and analyzed., Results: Median age at diagnosis of the 8 children (1 male and 7 females) was 6.4 (range, 10 months to 15 years) years old. The median medical history of MDS was 2.7 years (range, 3 months to 8 years). Among the 8 patients, 7 cases were diagnosed with refractory cytopenia of childhood and one with refractory anemia with excess of blasts. The HSC donors were father, mother or brother of patients and HLA matching in 6-9/12 loci were identical. All the donors were healthy and didn't carry the same pathogenic genes as the recipients. The median age of donors was 36.4 (range, 25 to 49) years old. The median mononuclear cell (MNC) number of the graft was 19.8, ranging in (13.2-47.3)×10 8 /kg, and the median CD34 + cell number was 11.8×10 6 /kg, ranging in (5.0-18.3)×10 6 /kg. Graft-versus-host disease prophylactic regimen was started on day 3 and 4 after transplantation, in which cyclophosphamide (50 mg/kg·d) was administered by intravenous infusion. From day 5 after transplantation, low-dose tacrolimus was administered by intravenous infusion and mycophenolate mofetil was administered orally. The median time of neutrophil and platelet engraftment was 12.6 (rang, 11 to 15) days and 13.3 (rang, 11 to 18) days, respectively. All the patients achieved full donor chimerism on neutrophil engraftment after transplantation. The median follow-up time was 1 032 (rang, 747 to 1 536) days. Both overall survival rate and disease-free survival rate were 100%., Conclusion: Haplo-HSCT combined with ATG and PTCy-induced immune tolerance after transplantation is a safe and effective treatment for children with MDS.

Objective: To explore the genetic characteristics and evolution of hantavirus carried by rodents in port area of Ningde in Fujian province in the summer of 2020. Methods: Rodents were captured in the port area of Ningde, the RNA was extracted from rodent lung tissues and detected by using specific kit. The positive samples were used for whole-genome sequencing of the virus. Bioinformatics software was used for the analysis on the similarity and genetic variation of the sequences. Results: A total of 112 rodents were captured, including 5 Rattus norvegicus and 2 Rattus flavipectus , the positive rate of hantavirus was 6.25% (7/112). By virus gene sequencing, two hantavirus complete genome sequences were obtained (named as FJ35 and FJ36, GenBank accession numbers: MW449188-MW449193). The genetic analysis results showed that the hantavirus detected in positive samples were SEOV and shared 99% nucleotide similarity with hantavirus strains LZSF21 and JX20140581 isolated from Shandong province. Phylogenetic analysis using the maximum likelihood method showed that the hantavirus detected in positive samples belonged to S3 subtype, sharing the same subtype with hantavirus strains Z37 from Zhejiang province, LZSF21 from Shandong province, and zy27 and Gongzhuling 415 from northeastern China. Compared with FJ372, the amino acid variation of N259S was observed at sites 251-264 of nucleoprotein, which might be related to antigenicity. Another variation of Q81R was observed in glycoprotein compared with SEOV 80-39 segment of coded amino acid of international reference strain, which might also cause the change in antigenicity. Conclusion: The high positive rate of hantavirus in rodents in the port area of Ningde- would increase the risk of natural human infection and epidemic in local area. The hantavirus positive rodents in this focus might be from an endemic area in Shandong. It is necessary to strengthen the imported rodent control in the port area of Ningde. The virus detected in 2 positive samples belonged to SEOV subtype Ⅲ and shared high homologies of nucleotides and amino acid sequences with the hantavirus strains in surrounding area. However, some slight variations occurred in glycoprotein and nucleoprotein amino acid sequences, which might cause changes in its antigeniity.

Objective: To explore the long-term efficacy of laparoscopic-assisted anorectoplasty and conventional anorectoplasty in the treatment of children with high and middle anal atresia. Methods: A retrospective cohort study was used. Inclusion criteria: (1) children with high and middle anal atresia; (2) complicated with rectourethral or rectovesical fistula; (3) complete follow-up data. Exclusion criteria: (1) complicated with 21-trisomy; (2) cerebral palsy and other mentaldisabilities; (3) Currarino syndrome; (4) FG syndrome. Clinical data of 88 patients with middle and high anal atresia, who complicated with rectourethral fistula or rectovesical fistula, and underwent anoplasty at Department of Pediatric Surgery, the First Affiliated Hospital of Zhengzhou University from January 2009 to June 2014 were enrolled in the study and analyzed. There were 24 cases with middle atresia and 64 cases with high atresia. All the cases were divided into 2 groups based on the operative method: laparoscopic group (laparoscopic-assisted anorectoplasty, 49 cases), pena group (posterior sagittal anorectoplasty, 39 cases). The demographic features of two groups were comparable. There were no statistically significant differences in gender, age, body mass, classification of anomaly types and sacral ratio (all P >0.05). Student t test and Chi square tests were used to compare the surgical conditions (operative time, postoperative hospital stay and complications), anal function (Kelly score), constipation (Krickenbeck constipation score) and anorectal pressure. Results: Children of both groups all completed operation ssuccessfully. There were no statistically significant differences between laparoscopic group and pena group in the operative time [(120±31) minutes vs. (112±23) minutes, t =1.343, P =0.091] and postoperative hospital stay [(7.1±2.3) days vs. (10.7±3.3) days, t =6.021, P =1.000]. Complications were more common in the pena group [16.3% (8/49) vs. 35.9% (14/39), χ(2)=4.436, P =0.035]. The main complications in laparoscopic group were anal prolapse (8.2%, 4/49) and anal stenosis (6.2%, 3/49), while in pena group were anal stenosis (12.8%, 5/39) and perioperative perianal skin erosion (10.3%, 4/39). As for the anal function, the degree of feces, defecation control and sphincter contractility, the single scoring differences of Kelly scoring system were not statistically significant between the two groups, but the proportion of good function in the laparoscopic group was higher than that in the pena group [67.3% (8/49) vs. 38.5% (15/39), χ(2)=7.308, P =0.007]. Constipation occurred in 6 (12.2%) patients in the laparoscopic group, of whom 5 were improved by diet regulation and 1 required laxatives, while 9 (23.1%) patients developed constipation in the pena group, of whom 4 were improved by diet regulation and 5 required long-term laxatives. The difference of constipation ratio was not statistically significant (χ(2)=1.802, P= 0.180). There were no cases of Krickenbeck constipation grade 3. Compared to the pena group, the laparoscopic group had higher anal resting pressure [(33.35±9.69) mmHg vs. (27.68±10.74) mmHg, t= 2.599, P =0.011], higher dilating pressure [(9.00±5.61) mmHg vs.(6.51±3.24) mmHg, t= 2.462, P =0.016], higher maximal squeeze pressure [(65.80±17.23) mmHg vs. (56.74±18.93) mmHg, t =2.389, P =0.019] and longer maximal contraction time [(21.16±5.02) seconds vs. (18.44±7.24) seconds, t =2.079, P =0.041]. The rectal resting pressure [(5.36±3.00) mmHg vs. (4.61±3.93) mmHg, t =1.015, P =0.312] was not statistically significantly different. Conclusions: Compared with posterior sagittal anorectoplasty, laparoscopic-assisted anorectoplasty in the treatment of high and middle anal atresia has better long-term efficacy with less perioperative complications.

Objective: To investigate the tumor necrosis factor receptor superfamily 1B gene (TNFRSF1B) polymorphism in relation to the outcomes of hepatitis C virus (HCV) infection. Methods: One thousand six hundred and forty-five cases without HCV infection, 545 cases with HCV clearance, and 783 cases with chronic HCV infection were enrolled. TaqMan probe method was used to investigate genotype rs1061622 (T > G) and rs1061624 (G > A). Two single nucleotide polymorphisms (SNPs) sites were genotyped and haplotypes were constructed to evaluate their relation with the outcome of HCV infection. Results: Logistic regression analysis showed that there was no relation to the two SNPs with HCV infection susceptibility and chronicity ( P > 0.05). Haplotype analysis showed that carrier TA had an increased susceptibility to HCV infection [adjusted odds ratio ( OR ) = 1.15, 95% confidence interval ( CI ): 1.01 to 1.30, P = 0.038)]. Carrier TA and GG haplotypes were conducive to chronic HCV infection (adjusted OR = 1.28, 95% CI : 1.08 to 1.53, P = 0.006; OR = 1.31, 95% CI : 1.03 to 1.66, P = 0.026). Conclusion: The combinational effects of rs1061622 and rs1061624 in TNFRSF1B gene may increase the risk of HCV chronicity and infection.

Objective: To explore the relationship between the tumor necrosis factor receptor superfamily members 11A (TNFRSF11A) and 11B (TNFRSF11B) gene polymorphisms and the outcome of hepatitis C virus (HCV) infection. Methods: In this case-control study, 749 cases of persistent HCV infection, 494 cases of spontaneous clearance and 1 486 control subjects were included from 2008 to 2016. TaqMan-MGB probe method was used to detect the genotype of TNFRSF11A rs1805034 and TNFRSF11B rs2073617. The genotypes distribution of the two single nucleotide polymorphisms (SNP) were analyzed in different populations. Results: Co-dominant model showed that individuals carrying the rs2073617 CC genotype were prone to have chronic HCV infection, compared with individuals carrying the rs2073617 TT genotype ( OR =1.517, 95 %CI : 1.055-2.181, P =0.024). Recessive model results showed that individuals carrying rs2073617 CC genotype were more likely to develop chronic HCV infection compared with individuals carrying rs2073617 TT or TC genotype ( OR =1.435, 95 %CI : 1.033-1.996, P =0.032). Additive model showed that the risk for chronic HCV infection increased with the increase of the number of rs2073617 C alleles ( OR =1.204, 95 %CI : 1.013-1.431, P =0.035). Conclusion: The genetic polymorphism of TNFRSF11B rs2073617 might be related with the chronicity of HCV infection.

Objective: To assess the application of probe-based confocal laser endomicroscopy (pCLE) in diagnosis of gastric carcinoma and precancerous lesions., Methods: Patients underwent pCLE in the First Affiliated Hospital of Zhejiang University School of Medicine during December 2013 and November 2014 and in the First Affiliated Hospital of Zhejiang Chinese Medical University during January 2014 and December 2017 were enrolled. The consistency between pCLE diagnosis and pathological diagnosis of gastric lesions, including atrophic gastritis, gastric intestinal metaplasia, low-grade intraepithelial neoplasia and high-grade intraepithelial neoplasia (including gastric carcinoma) was analyzed., Results: Totally 154 gastric lesions from 119 patients were detected by pCLE. Using pathological diagnosis as gold standard, the sensitivity, specificity, coincidence rate and κ value of pCLE diagnosis for atrophic gastritis were 94.34%, 91.09%, 92.21%and 0.83; those indicators for gastric intestinal metaplasia were 84.47%, 92.16%, 87.01% and 0.72. The coincidence rate and κ value of pCLE diagnosis of complete gastric intestinal metaplasia were 0.75 and 0.49; for incomplete gastric intestinal metaplasia were 0.79 and 0.48, respectively. The sensitivity, specificity, coincidence rate and κ value of pCLE diagnosis for low-grade intraepithelial neoplasia were 85.29%, 87.50%, 87.01%and 0.66; those for high-grade intraepithelial neoplasia (including gastric carcinoma) were 95.83%, 97.17%, 96.75%and 0.92., Conclusions: pCLE can be used for diagnosis of gastric carcinoma and pericancerous lesions and also for typing of gastric intestinal metaplasia.

Objective: To explore the association between nuclear factor kappa-light-chain-enhancer of activated genetic polymorphisms in B cells ( NF-κB ) and the HCV susceptibility, among the Chinese population. Methods: A total of 1 679 participants were enrolled; including 503 drug users and 1 176 other participants at risk under the exposure for blood. By using the logistic regression analysis, related risk factors for HCV infection among subjects were analyzed. Two NF-κB pathway variants, NF-κB1 rs72696119 and REL rs13031237 were then genotyped by TaqMan assay method. Logistic regression analysis was performed to analyze the association between gene polymorphisms and the susceptibility on HCV. Results: Among the drug users, women ( OR =0.408, 95 %CI : 0.308-0.767) appeared to be associated with the decreased risk for HCV infection, while factors as drug injection ( OR =8.817, 95 %CI : 5.577-13.937) and the duration of drug-intake >5.5 years ( OR =2.891, 95 %CI : 1.824-4.583) were associated with the increased risk for HCV infection. Among the participants who had been exposed to blood, women ( OR =3.431, 95 %CI : 2.360-4.988) were associated with the increased risk for HCV infection, while the levels of education beyond elementary school ( OR =0.613, 95 %CI : 0.429-0.876) were associated with the decreased risk for HCV infection. Compared to the reference NF-κB1 rs72696119 CC genotype, the carriage of GG genotype was associated with an increased risk of susceptibility on HCV ( OR =1.412, 95 %CI : 1.035-1.927) among the total study population. Results from the interaction analysis showed that there was no interactive effects appeared between rs72696119 and route of infection, or between rs72696119 and gender among the total population under study (all P >0.05). Conclusion: polymorphism rs72696119 and related factors seemed associated with the susceptibility to HCV infection among high-risk Chinese populations.NF-κB1 polymorphism rs72696119 and related factors seemed associated with the susceptibility to HCV infection among high-risk Chinese populations.

Liver cancer is a common type of malignant tumor in digestive system. At present, computed tomography (CT) plays an important role in the diagnosis and treatment of liver cancer. Segmentation of tumor lesions based on CT is thus critical in clinical diagnosis and treatment. Due to the limitations of manual segmentation, such as inefficiency and subjectivity, the automatic and accurate segmentation based on advanced computational techniques is becoming more and more popular. In this review, we summarize the research progress of automatic segmentation of liver cancer lesions based on CT scans. By comparing and analyzing the results of experiments, this review evaluate various methods objectively, so that researchers in related fields can better understand the current research progress of liver cancer segmentation based on CT scans.

Objective: To investigate the effect of extensive retraction clefts (RC, >20% of tumor volume) on prognosis in invasive breast carcinoma of no specific type (IBC-NST). Methods: A total of 2 184 cases of IBC-NST diagnosed at the Fourth Hospital of Hebei Medical University from January 2006 to December 2008 were collected. All the cases were diagnosed according to the latest guideline and standard. After excluding cases of shrinkage due to tissue fixation, 483 cases with RC were identified, and the clinical and pathological features were retrospectively analyzed. Results: Among the 483 cases, the mean tumor size was 2.0 cm (range 0.8 to 4.8 cm). Two hundred and thirty-two cases were moderately differentiated (48.0%), 97 were well differentiated (20.1%), 154 were poorly differentiated (31.9%); 382 (79.1%) cases were of stages Ⅰ and Ⅱ. A total of 177 cases (36.7%) had lymphatic invasion; nodal metastasis were found in 202 cases (41.8%). Extensive RC was found in 237 of 483 cases (49.1%). Follow-up information was available in 407 patients, and 46 died of breast cancer with survival time from 37 to 103 months. Multivariate analysis of extensive RC showed that tumor size, histological grade and nodal metastasis were risk factors of patients with IBC-NST ( P <0.05). Lymphatic invasion and nodal metastasis were risk factors for extensive RCs in patients with IBC-NST ( P <0.05). There was a high probability of lymph node metastasis in patients of extensive RC without lymphatic invasion, and the difference was statistically significant ( P <0.05). Conclusions: Both lymphatic invasion and nodal metastasis are risk factors of extensive RC. The presence of extensive RC in IBC-NST patients is correlated with poor outcome. Tumors with lymphatic invasion are more likely to show extensive RC.

Objective: This study aims to investigate the effect of neurotrophin 3 (NT-3) on the osteogenic differentiation of human dental follicle cells (hDFCs)., Methods: hDFCs were isolated and cultured in vitro. Immunocytochemical staining was used to identify the origin of hDFCs. The effects of different NT-3 concentrations on hDFCs proliferation were detected by using CCK-8 assay. The alkaline phosphatase (ALP) activities and mRNA expression levels of bone morphogenetic protein-2 (BMP-2) and osteocalcin (OCN) were determined to investigate the effects of NT-3 on hDFCs osteogenesis. The difference in the number of mineralized nodules was detected using alizarin red staining., Results: Vimentin and cytokeratin staining results showed that hDFCs originated from the mesenchymal cells. NT-3 exerted no evident effect on hDFCs proliferation. The ALP activity and the BMP-2 and OCN mRNA expression levels of hDFCs were significantly improved under treatment with different NT-3 concentrations (25, 50, and 100 ng·mL ⁻¹) compared with those in the control group. BMP-2 and OCN mRNA relative expression levels of hDFCs reached the highest when the NT-3 concentration was 100 ng·mL ⁻¹. The number of mineralized nodules reached the maximum when the hDFCs were treated with 50 and 100 ng·mL ⁻¹ NT-3., Conclusions: Appropriate mass concentration of NT-3 can promote the osteogenic differentiation of hDFCs.

Objective: To investigate the grading system for lymph vessel tumor emboli and its prognostic value in patients with invasive carcinomas of no special type (ICNST) of the breast. Methods: Clinical data of 466patients with ICNST were collected from January 2006 to December 2008 in the Fourth Hospital of Hebei Medical University. The expression levels of D2-40, estrogen receptor(ER), progesterone receptor(PR) and human epidermal growth factor receptor 2 (HER-2) were analyzed using immunohistochemical staining. Grades for lymph vessel tumoremboli were classified based on the number of mitotic and apoptotic figures in tumor cells under a high-power field. Correlation analysis was performed using Spearman rank correlation test. Kaplan-meier curves and Log-rank tests were used to analyze the survival rate. Multivariate Cox proportional hazard model was used to analyze the prognostic factors. Results: Among the 466 patients, grades for lymph vessel tumor emboli were categorized as follows: 280 cases were grade 0 (60.1%); 112 cases were grade 1 (24.0%); 58 cases were grade 2 (12.5%); 16 cases were grade 3 (3.4%). Correlation analyses showed that lymph vessel tumor emboli grading system was positively correlated with lymph node metastasis ( r =0.365, P <0.001). Kaplan-Meier univariant analysis showed that histological grading, lymph vessel tumor emboli grading system, lymph node metastasis, the expression levels of ER, PR and HER-2 and molecular typing were associated with prognosis of patients ( P <0.05 for all). Multivariate analysis of Cox proportional hazard model showed that lymph vessel tumor emboli grading system and lymph node metastasis were independent prognostic factors in patients with ICNST( P <0.05 for all). Conclusion: Grading system for lymph vessel tumor emboli canpredict the clinical outcome of patients with ICNST.

Objective: S gene of hantavirus(HV) was expressed in insect cells by genetic engineering technology. The expression product of S gene was used as antigen to detect anti-HV specific antibody IgG in serum. Methods: Gene encoding NP of the strain HV-Z10 was amplified by PCR and then its eukaryotic expression system rBAC-Z10S-TN was constructed by using the routine genetic engineering method. SDS-PAGE was applied to measure the expression of rNP.Ion-exchange plus Ni-NTA-affinity chromatography was performed to purify the recombinant product. Indirect immuno-fluorescence assay (IFA) was used to determine the specific immune-reactivity of rNP. WB assay was established to detect the serum samples from 95 confirmed HFRS patients. Parameters related to the outcomes of detection were compared with the routine HV-IgG IFA method. Results: rBAC-Z10S-TN was able to express rNP with high efficiency. The purified rNP only showed a single protein fragment in the gel after SDS-PAGE. HV IgG could efficiently recognize rNP and hybridize with the recombinant protein. 97.67 % of the serum samples from the HFRS patients were positive confirmed by WB. Conclusions: We successfully constructed a high efficient prokaryotic expression system of NP encoding gene from hantavirus strain HV-Z10. WB assay which was established in this study could be used as a new serological test for HFRS diagnosis, thanks to the simplicity, safety, sensitivity and specificity of this method.

Objective: This study investigated the effects of salvianolic acid B (Sal B), a major bioactive component of the Chinese medicine salvia miltiorrhiza, on osteogenic differentiation of human periodontal ligament cells (hPDLCs)., Methods: Third passage PDLCs were used in this experiment. Methyl thiazolyl tetrazolium (MTT) method was employed to observe the effects of different Sal B concentrations on proliferation activity of hPDLCs. Alkaline phosphatase (ALP) activity and mineralization capability were measured, and mRNA expression of osteocalcin (OCN) was detected to investigate the effects of Sal B on osteogenesis of hPDLCs., Results: Sal B did not influence the viability of hPDLCs. The ALP activity and OCN mRNA expression levels of hPDLCs were both significantly improved (P<0.05) under treatment with different Sal B concentrations (0.5, 1, and 5 μmol·L⁻¹) compared with those in OIM group. Moreover, the number of mineralized nodules formed by hPDLCs were considerably higher under treatment with different Sal B concentrations (0.5, 1, and 5 μmol·L⁻¹) than that in the OIM group., Conclusions: Appropriate Sal B concentration can improve the osteogenic differentiation of hPDLCs.

Objective: To investigate the effect of bufalin on proliferation and apoptosis through ERK/RSK2 pathway in esophageal squamous cell carcinoma xenografts in nude mice., Methods: The subcutaneous xenograft model of esophageal cancer ECA109 cells in nude mice was established. The mice were divided into the model group, low-dose bufalin group, medium-dose bufalin group, high-dose bufalin group, PD98059 group and combination group to evaluate the effect of bufalin on the xenografts. The morphology of xenografts was observed by microscopy. The cell apoptosis index of xenografts was detected by TUNEL assay. The expression of ERK and RSK2 mRNA of human ECA109 cell transplantation tumor in nude mice was examined by real-time quantitative PCR. The protein levels of ERK, p-ERK, RSK2, p-RSK2, GSK3β, p-GSK3β, Bad and p-Bad in the xenografts were examined by Western blot and Immunohistochemistry., Results: The tumor size of nude mice in the model group, low-dose bufalin group (BL), medium -dose bufalin group (BM), high-dose bufalin group (BH), PD98059 group and combined therapy group (BP) was (1.758±0.181) cm(3,) (1.680±0.150) cm(3,) (1.285±0.134) cm(3,) (0.873±0.095) cm(3,) (0.815±0.108) cm(3) and (0.530±0.104) cm(3,) respectively. Histological examination showed that the xenografts of each group had varying degrees of necrosis, and the most extensive necrosis was observed in the BP group. The TUNEL assay showed that the cell apoptosis index of xenografts in the model, BL, BM, BH, PD98059 and BP groups was (6.0±0.6)%, (11.0±0.7)%, (19.1±0.9)%, (25.1±1.4)%, (20.0±1.2)% and (17.1±0.7)%, respectively, which is highest in the BH group. The real-time quantitative PCR results showed that the ΔCT values of ERK mRNA in the model, BL, BM, BH, PD98059 and BP groups were 0.270±0.084, 0.293±0.081, 0.596±0.224, 0.857±0.183, 0.868±0.187 and 1.313±0.282, respectively. The ΔCT values of RSK2 mRNA in the model, BL, BM, BH, PD98059 and BP groups were 0.340±0.062, 0.337±0.071, 0.642±0.226, 0.915±0.170, 0.923±0.176 and 1.413±0.269, respectively. The relative expression of ERK and RSK2 mRNA was gradually decreased. Western blot and immunohistochemistry results showed that the protein levels of ERK, RSK2 and Bad in each group were not significantly different (P>0.05). The protein levels of p-ERK in the model, BL, BM, BH, PD98059 and BP groups were 0.721±0.094, 0.695±0.095, 0.555±0.080, 0.388±0.052, 0.341±0.060, 0.235± 0.056, respectively. The median immunoreactivity scores of p-ERK in each group were 8, 8, 6, 4, 5 and 3. The protein levels of p-RSK2 in the model, BL, BM, BH, PD98059 and BP groups were 0.613±0.085, 0.612±0.084, 0.427±0.089, 0.305±0.056, 0.258±0.051, 0.158±0.058, respectively. The median immunoreactivity scores of p-RSK in each group were 8, 8, 5, 3, 3 and 1. The protein level of GSK3β in the model, BL, BM, BH, PD98059 and BP groups were increased gradually, while the protein level of p-GSK3β and p-Bad were decreased gradually., Conclusions: Bufalin exerts significant inhibitory effect on the esophageal squamous cell carcinoma xenogragts in nude mice. Bufalin may suppress the growth of xenogragts in nude mice by down-regulating the level of ERK and RSK2 phosphorylation, inhibit the proliferation of xenogragts via inactivating GSK3β and promote apoptosis through down-regulation of p-Bad.

Objective: To investigate the efficacy of bundle treatment on patients with moderate or severe acute respiratory distress syndrome (ARDS)., Methods: A multicenter prospective observational study comparing the result of historical treatment strategy and bundle treatment was conducted. According to the new Berlin standard of definition, 73 patients with moderate or severe ARDS due to pulmonary factors, age from 18 to 65 years, admitted to Department of Critical Care Medicine of Taian Central Hospital and Handan Central Hospital were enrolled. Thirty-three patients admitted during September 2012 to May 2014 (prospective observation period) were enrolled as the bundle treatment group. Forty patients with matched disease history admitted from January 2010 to August 2012 were enrolled as the control group. The patients in bundle treatment group received bundle treatment based on the treatment strategy of primary diseases. Bundle treatment included restrictive fluid management, respiratory support, high-dose ambroxol combined with Xuebijing injection, prevention of ventilation associated pneumonia (VAP), individualized sedation plan, installation of continuous blood purification treatment for critical patients. A special team was organized to ensure the successful implementation of all bundle measures. The acute physiology and chronic health evaluation II (APACHEII) score, oxygenation index, duration of mechanical ventilation, the length of intensive care unit (ICU) stay, incidence of VAP, and 28-day mortality 5 days after treatment were compared between two groups., Results: There were no significant differences in basic characteristics of patients between the two groups, including gender, age, etiology, severity, etc. (all P>0.05) with comparability. Compared with the control group, there was no significant difference in APACHE II score 5 days after treatment in bundle treatment group (15.1±2.8 vs. 16.2±3.0, t=1.618, P=0.110). Compared with control group, oxygenation index in bundle treatment group was significantly improved [mmHg (1 mmHg=0.133 kPa): 135.4±34.5 vs. 117.1±34.2, t=-2.273, P=0.026), the duration of mechanical ventilation was obviously reduced (days: 8.70±2.50 vs. 10.10±2.67, t=2.308, P=0.024), incidence of VAP was significantly lower [18.2% (6/33) vs. 32.5% (13/40), χ2=5.027, P=0.025], and 28-day mortality rate was obviously lowered [24.2% (8/33) vs. 37.5% (15/40), χ2=4.372, P=0.037], the length of ICU stay shown no statistical difference (days: 10.40±1.94 vs. 11.30±2.34, t=1.620, P=0.110)., Conclusions: Implementation of bundle treatment can significantly shorten the duration of mechanical ventilation, reduce the incidence of VAP, and improve the prognosis of patients.

Objective: To evaluate the effect of a new schistosomiasis control strategy based on the infection source control in four-lake regions of Hubei Province., Methods: The new strategy based on the infection source control included the safe water supply, feces harmless treatment, replacing cattle with machine, captive livestock, chemotherapy, Oncomelania hupensis snail control in susceptible areas, hardening ditches, etc, and the new control strategy was implemented for 7 years in 9 counties (cities or districts) of the four-lake regions. The schistosomiasis situations were investigated before and after the intervention of the new strategy., Results: The coverage rates of feces harmless toilets (three format toilets), biogas digesters, and safe water supply were 42.00%, 23.16% and 93.76%, respectively. The number of ditch hardening was 1 960; the area of eliminating snails was 1378.42 hm2; the number of persons who received the health education was 3,524,818 accounting for 92.17% of the total population; the number of person-time of taking active protection measures was 516,636,963. The average annual decline rates of schistosome infection were 24.42% and 38.38% in residents and cattle, respectively. The endemic villages decreased by 4.77%, the villages with snails decreased by 7.05%, and the villages with snails accounted between 63.19% and 66.21% of the total endemic villages. The area with snails decreased by 90.35%, and the advance schistosomiasis patients decreased by 5.55%, and the fatality rate of advanced schistosomiasis patients decreased by 1.24%. No acute schistosomiasis patients occurred. Type One and Two endemic (heavy endemic) villages decreased to zero; Type Three endemic (moderate endemic) villages decreased by 10.22%; but Type Four endemic (mild endemic) villages increased by 66.38%; and Type Five endemic (no endemic for 5 years) villages decreased by 0.22%. The standards of schistosomiasis transmission controlled have achieved on schedule., Conclusion: The implementation of the new schistosomiasis control strategy based on the infection source control in four-lake regions of Hubei Province can effectively control schistosomiasis.

Objective: To investigate the mRNA expression and promoter methylation status of p73 gene in the peripheral blood of children with Wilms' tumor (WT), and their relationship., Methods: Forty-five children with WT were selected as the case group, and 15 sex- and age- matched children (without malignancies) who visited the hospital for physical examination or other reasons were selected as the control group. Peripheral blood was collected from both groups. Real-time quantitative PCR and methylation-specific PCR were used to determine the mRNA expression level and promoter methylation status of p73 gene. Their relationship with clinicopathological features and the effect of promoter methylation on mRNA expression of p73 gene were analyzed in the case group., Results: The relative quantity (RQ) of p73 mRNA in the case group was significantly higher than in the control group (3.2 ± 0.9 vs 1.6 ± 1.1; P<0.01). The positive rate of p73 gene promoter methylation in the case group was significantly lower than in the control group (20% vs 73%; P<0.01). In the case group, the RQ of p73 mRNA was significantly higher in children with methylated p73 gene promoter than in those with unmethylated p73 gene promoter (P<0.01). In children with methylated p73 gene promoter, the RQ of p73 mRNA was significantly higher in the case group than in the control group (P<0.01). In children with unmethylated p73 gene promoter, there was no significant difference in RQ of p73 mRNA between the case and control groups (P=0.810)., Conclusions: Aberrant promoter methylation of p73 gene in peripheral blood is one of the gene expression regulations in children with WT, and it is related to the onset and development of WT. The p73 gene may play a role as oncogene in WT patients with p73 gene promoter methylation and mRNA overexpression is associated with promoter methylation status of p73 gene.

Objective: Based on the antitumor function of the delta-tocotrienol, shed light on the relationship between the proliferation inhibition effect of delta-tocotrienol on human colon cancer cells SW620 with Wnt signal pathway., Methods: To adopt the MTT method, found the effects of diverse doses of delta-tocotrienol on colon cancer cells SW620. Detected the expressions level of the factors related to Wnt signal pathway via Western bloting and immunocytochemistry, to observe the influence of delta-tocotrienol on Wnt-1, beta-catenin, c-jun, cylin D1., Results: delta-tocotrienol has a significant proliferation inhibition effect on the colon cancer cells SW620,under delta-tocotrienol treated, SW620 cell inhibition rate is 70.43%. The IC50 is 15.18 micromol/L. The expression levels of wnt-1, beta-catenin c-jun and cylin D1 are downregulated (P < 0.05)., Conclusion: The proliferation inhibition effect of delta-tocotrienol on colon cancer cell may correlate with the ability of delta-tocotrienol to downregulate the expression levels of wnt-1, beta-catenin, c-jun, cylin D1 in the Wnt signal pathway.

Stratospheric ozone depletion results in an increased level of solar UV-B radiation (UV-B, 280-320 nm) reaching the earth surface. By the effect of UV-B radiation, various medicinal active ingredients changed because of the change of gene expression, enzyme activity and secondary metabolism, clinical effect is also changed. The research status of UV-B radiation and the accumulation of plant secondary metabolites in the past 10 years were summarized in this paper to supply reference for cultivation and exploitation of the medicinal plants.

Objective: To analyze the correlation between environmental factors and the lipophilic and hydrophilic constituents of Polygalae Radix., Method: The contents of lipophilic constituent were determined by GC-MS and hydrophilic constituents by HPLC. Geographical factors were collected by on-site inspection and climate factors by the local meteorological data. The relationship between the content of the lipophilic and hydrophilic constituents and the factors were analyzed by SPSS 18.0., Result: There was linear relationship between the content of lipophilic constituent and climate factors such as average temperature of July, average temperature of January. There was also linear relationship between hydrophilic constituents and climate factors such as annual average temperature, latitude, annual average rainfall., Conclusion: The main climate factors that affect liposoluble constituent content were average temperature of July, average temperature of January, and the main climate factors that affect hydrophilic constituent content were annual average temperature, latitude, annual average rainfall. The study would contribute to the quality evaluation and suitability of origin of Polygalae Radix.

Aimed to understand the causes of the boundary formation of Quercus wutaishanica forest, four sites were chosen along a precipitation and thermal gradient from Qinling Mountain to Loess Plateau of northern Shaanxi Province, i. e. , north slope of Qinling Mountain (Jiwozi) and southern (Chenjiashan), central (Huaishuzhuang), and northern site (Xiasiwan) of Loess Plateau, with the gas exchange, chlorophyll fluorescence parameters, stomatal density, and stomatal size of Q. wutaishanica leaves measured. From the south to the north along the precipitation and thermal gradient, weather varied from cool and humid to warm and semi-arid, and the maximum net photosynthetic rate (Pn max) and light saturation point (Lsp) of Q. wutaishanica leaves reduced obviously from 13.34 to 6.99 micromol CO2 x m(-2) x s(-1) and from 1489.9 to 1055.6 micromol x m(-2) x s(-1), dark respiration rate (Rd) and light compensation point (Lcp) increased evidently from 0.313 to 1.080 micromol CO2 x m(-2) x s(-1) and from 7.49 to 31.96 micromol x m(-2) x s(-1), the minimum fluorescence (F0) and coefficient of non-photochemical quenching (NPQ) displayed an increasing trend, and actual photochemical quantum yield of PS II (phiPS II), apparent electron transport rates (ETR), and coefficient of photochemical quenching (qP) reduced obviously. These results demonstrated that the photosynthetic capacity of Q. wutaishanica declined gradually with decreasing precipitation, but the stomatal density and stomatal size showed no obvious difference among the study sites except for the southern site of Loess Plateau. It was speculated that the depressed net photosynthetic rate of Q. wutaishanica leaves induced by the declined Phi PS II and ETR Could be one of the main causes for the formation of northern boundary of Q. wutaishanica forest.

The effects of enhanced ultraviolet-B (UV-B) radiation on chemical compositions of radix, stem and leaf of Scutellaria baicalensis Georgi were studied via FTIR Results showed that alcohols and phenolic substances in the radix and leaf of S. baicalensis Georgi were increased by UV-B radiation as well as the content of flavonoids and flavonoids glycosides. The terpenoids content in radix and protein content in leaf were increased by supplementary UV-B radiation. Contrary to the circumstance in radix and leaf, contents of alcohols and phenolic, flavonoids, unsaturated fat and lactone in stem were decreased by UV-B radiation. These results indicated that there is a trade-off in distribution of the metabolites in radix, stem and leaf of different parts. More information could be revealed for the result of FTIR spectral analysis via FTIR second derivative spectrum, especially the information of the combination between flavonoids and glycosides. The effects of UV-B radiation on chemical composition in radix, stem and leaf of S. baicalensis Georgi could be examined simply and rapidly via the method of FTIR. Thus, it can be used as a prior method before the further analysis of chemical composition.

Objective: To obtain polyploidy Scutellaria barbata., Methods: Explants of stem segment of Scutellaria barbata were treated with oryzalin at various concentration and time duration in vitro., Results: Comparing with the characteristics of diploid, mutant plants had significant changes in plant morphology inducing compact plant, darker leaf color, smaller leaf index, thicken stem, significantly larger stomata, guard cells and chloroplasts in stomata guard cells and significantly lower stomata density. Mutant plants were plant chimera of diploid and tetraploid assessed by chromosome counting and flow cytometry., Conclusion: The technique could be used for chromosome doubling in Scutellaria barbata.

The effects of ultraviolet-B (UV-B) radiation on protein structure in photosystem II (PS II) was studied using Fourier transform infrared spectroscopy. The results demonstrated that irradiation of the plant with enhanced UV-B caused the change in the structures of protein in PSII. The intensity of alpha-helix, beta-sheet was increased, while the intensity of beta-turn was decreased. The coupling of those structures with surrounding became weaker with UV-B radiation. Otherwise, the UV-B influenced the structure of tyrosine residue and made the polarity of tyrosine decrease. The changes caused by UV-B radiation could alter the combination between Mn cluster and pigments with protein inevitably. It would lead to a series of changes, such as the process of light transmission and conversion, electron transfer and oxygen evolution.

Taking the guerrilla type clonal plant Kingdonia uniflora at different altitudes in Taibai Mountain National Forest Park of Shaanxi Province, China as test material, the C, N, P, K and Mg contents in different vegetative organs of its ramet were measured. The results showed that ramet age, altitude, and their interaction had no significant effects on the total C content in ramet leaf, rhizome, and root (P > 0.05), but different effects on the total N, total P, K and Mg contents in these vegetative organs, suggesting that ramet could regulate the allocation of nutritive elements in its vegetative organs through physiological metabolism. At high altitude (3000-3100 m), ramet age had significant effects on the K content in all vegetative organs and the N content in rhizome and root (P < 0.05). All the test nutrient contents were significantly correlated with each other at all altitudes, except at middle altitude (2800-2900 m) where the ramet K content had no significant correlations with its N, P, and Mg contents. It was suggested that physiological plasticity contributed more to the performances of clonal plants at high altitude than at low altitude. Powerful physiological plasticity could help K. uniflora to effectively utilize heterogeneous resources, realize population expansion, and open up new habitat at high altitude. It was likely that the physiological plasticity of K. uniflora responding to heterogeneous habitats was formed in its long-term evolvement.

The study on the effects of 8.40 kJ x m(-2) UV-B radiation, 700 micromol x mol(-1) CO2 concentration, and their combination on tomato (Lycopersicon esculeutum ) growth and its fruit quality showed that under enhanced UV-B radiation, the plant height, fresh weight, dry weight, photosynthesis rate, water use efficiency, and contents of total chlorophyll, chlorophyll a, chlorophyll b, soluble protein, ascorbate acid (Vitamin C) and lycopene were decreased, which resulted in the deterioration of tomato fruit quality. Doubled CO2 concentration alone accelerated tomato growth, and accordingly, ameliorated its fruit quality. Under the combined effects of enhanced UV-B radiation and doubled CO2 concentration, tomato growth and its fruit quality were not significantly different from the control. It was suggested that the positive effects of doubled CO2 could lighten and counteract the negative effects of UV-B radiation under combined treatment of enhanced UV-B radiation and doubled CO2 concentration.

Objective: To describe the brief survey of alcohol intake and the incidence of alcohol liver disease in Zhejiang province., Methods: 18,237 requested persons aged over 18 years were selected by multi-stage stratified cluster sampling in Zhejiang province. Questionnaire about alcohol consumption, hepatic ultrasonic scan and detection of hepatic enzymes and markers of HBV and HCV were carried out. Daily alcohol intake more than 40g (including equal to 40g/d) was essential for the diagnosis of alcoholic liver disease., Results: Among the 18,237 persons (male 12,042, female 6195), the average daily alcohol intake was (17.7 +/- 27.2) g. The incidence of alcoholic liver disease in Zhejiang province was 4.34% (male 6.36%, female 0.36%) in the whole population. Four subtypes were separated as alcoholic cirrhosis, alcoholic fat liver, alcoholic hepatitis and mild alcoholic injury in liver with the corresponding incidence of 0.68%, 0.94%, 1.51% and 1.21% separately., Conclusion: Alcoholic liver disease is found to be a common disease in Zhejiang province, indicating an urgent need for the public education on alcohol abuse and the treatment on related health problems

The effect of enhanced UV-B radiation (280-315 nm, approximating a 15% ozone layer reduction) on competitive interaction between spring wheat (Triticum aestivum) and wild oat (Avena fatua) was examined in the field. The density-dependent mortality of both wheat and wild oat did not exhibit a significant difference between control and UV-B treatments. A relatively high degree of competitive stress enhanced the effect of UV-B stress on biomass reduction. Under UV-B enhancement, the relative competitive status of wheat in terms of total biomass and aboveground biomass increased, but decreased when based upon grain production. Shifts in competitive balance occurred with significant changes in total biomass, especially when plants grew at higher densities in monocultures and mixtures. There were different effects in direction and intensity of UV-B radiation on competition balance of the other three species pairs, but in general, the effect of UV-B was in favor of monocotyledonous. The results implied that total degree of competitive stress, especially interspecific competition, might be crucial to assess the effect of UV-B enhancement on agriculture ecosystem properly.

The chromosomal mutation and mitosis of the wheat (Triticum aestivum, cv. 93-4736) were observed under the condition of the enhanced ultraviolet-B radiation. The results showed that the number of cells in mitosis of the UV-B treatments was 68.09% of the control, indicating that the UV-B treatment significantly inhibited division of root tip cells in wheat. Using t-test at P < or = 0.05, there was a significant difference between the control and the treatments. The enhanced UV-B significantly increased aberration frequency of chromosomes and cells (t = 9.51, n = 3, P < 0.05). Several types of chromosomal aberration such as lagging chromosomes of chromosomes bridge, fragments, nuclear aberration occurred in both of control and UV-B treatment. However, the frequency of lagging chromosome and fragments of chromosome was increased significantly by enhanced UV-B radiation (P < 0.05). Furthermore, the UV-B treatment caused several new types of chromosomal aberrations, such as three-bundle, four-bundle and six-bundle. In our opinion, these new types of chromosomal aberrations should be called 'partition-bundle division'. Under the condition of UV-B treatment, the amount of aberrations only accounted for 2.84% of the total during the interphase. In the period, nuclear abnormality appeared mainly in forms of micronucleolus and polycaryon. In the metaphase, there mainly exist fragment chromosome, which accounted for 26.67% of the total aberrations, being secondary to lagging chromosomes. The amount of chromosomal abnormality in the anaphase and telophase was the biggest, which included the types of bridge, lagging chromosomes, etc. Among the types, the lagging chromosomes were dominant, accounting for 32.84% of the total. In this study, what was especially emphasized was the UV-B induced abnormal mitosis that was first found. In the anaphase and telophase of the treated cells, chromosomes were divided into 3 bundles, 4 bundles, or 6 bundles, respectively accounting for 8.77%, 15.43%, and 8.02% of the total abnormal cells. Three-bundle division was nonsymmetrical. The number of chromosomes in each of the three bundles was not completely equal. Four-bundle division mainly symmetrized at direction of axis, the number of each of the two bundles on the same axis was basically the same, but was different from that on another axis. Four-bundle division was conducted in two patterns of division. With respect to the much more special six-bundle division, chromosomes were regularly divided into 3 groups, symmetrizing each other. As viewed from the number of chromosomes in each bundle, it was smaller than that in normal somatic cells. In the abnormal division of cells, cell plate or cell wall between the two divided bundles was not found, leading to 'multi-bundle bodies' occurrence by the end of cell division. Therefore, the abnormal division of the somatic cells should be called 'the multi-bundle division' or 'the partition bundle division'.

Regression analysis was used to study the relationship between four diversity indices and 19 soil factors at three different soil layers in ecotone between desert and oasis in Fukang, Xinjiang. The results indicated that soil pH, total salt content, soil organic matter, Cl-, K+ + Na+, Mg2+, total nitrogen, and available phosphorus were correlative to species diversity significantly (P < 0.05). The relationships between pH and diversity and between organic matter and diversity were best fitted in form of binomial model, and it meant that the highest species diversity occurred in middle of the gradient. The relationships between diversity and other 6 indices were in form of negative linear regression, showing that the contents of total salt, Cl-, K+ + Na+, Mg2+ were greater, and the species diversity indices were smaller. And the regression of total nitrogen and available phosphorus with evenness were similar to the former, that is, with the contents of soil total nitrogen and available phosphorus increasing, evenness indices decline correspondingly. The results of regression analysis between amount of plants and soil moisture and salt showed that plant abundance was influenced by soil water and salt significantly (P < 0.01).

To investigate the changes of the metabolism of rat myocardiac cells cultured in vitro under simulated weightlessness and gamma-irradiation, rat myocardiac cells were rotated on a horizontal clinostat, and irradiated with 2Gy 60Co gamma-ray. The results showed: (1) after irradiation, the LDH activity and SOD concentrations showed a significant increase, and the MDA concentration also increased. (2) After irradiation, LDH activity increased 60% on the 3rd day, and SOD concentration almost doubled on the first day. (2) MDA concentration also increased but it reduced to normal after the 3rd day. (3) Simple rotation on the clinostat showed no effect on the parameter, but there was a synergistic effect when it was combined with gamma-irradiation.

In order to evaluate the effect of anisodaminum and dexamethasone to the microcurculation, TNF, LPO and pathology in MODS, we first established MODS model using cecem ligature and perfuration of rabbit then divided it into four groups: anisodaminum, dexamethasone, combined anisodaminum and dexamethasone and control. After used the corresponding drugs we observed the change of microcirculation, TNF, LPO and organ pathology. The result showed that microcirculation improved, the serum level of TNF and LPO decreased, and the pathomorphological changes lessensed. It is suggested that the combined use of anisodaminum and dexamethasone may be a new way for treating MODS.

A special rotator designed for this experiment was used to simulate the gravitational conditions. 3 groups of osteoblast cells were cultured under simulated hypogravity, normal (1 g) or hypergravity acceleratively (3 g) for 7 d. Changes of cell shape and speed of cell division were observed. The results showed that cells cultured under simulated hypogravity became round and cells divided slowly as compared with 1g group. While cells cultured under 3 g divided more actively and many growth spots appeared in the culture bottle.
