A memory-based frequent-pattern discovering algorithm for large-scale corpus was presented.First,the origi-nal corpus was partitioned into several parts using appropriate dividing policy.Then each partition was processed inde-pendently to produce a temporary result,and the union of all temporary results is the final frequent-pattern set.The algo-rithm prunes a subtree once it is sure that none of the corresponding pattern will be frequent.Experiment shows that it takes no more than 1.6 gigabytes of memory to discover all patterns appearing more than 100 times for a 3.6 gigabytes news corpus,the average speed is 3.28 magabytes per second.

To recognize the hotspot information through great deal of Internet data,the hotspot information detection al-gorithm（HID） was designed.Based on segmented word joint,with a customized noise library and multi-level filtering strategies,HID was provided with the ability to detect hotspot information in large amount of Internet pages.Experiment on TDT international standard test corpus proved the efficiency of HID.

Objective: To investigate the difference in the therapeutic effect of plasma exchange and continuous renal replacement therapy (PE+CRRT) combined with chemotherapy in the treatment of children with severe Epstein-Barr virus-associated hemophagocytic lymphohistiocytosis (EBV-HLH) and non-EBV-HLH., Methods: The clinical data of 21 cases of all children with severe HLH treated by PE+CRRT combined with chemotherapy from January 2017 to January 2020 were collected and retrospectively analyzed. According to the presence of EBV infection, the children were divided into EBV + group and EBV - group. The differences of the observation indexes between the children in the two groups and the improvement of the observation indexes of each group before and after treatment were compared., Results: Among the 21 children, 14 were divided into the EBV + group and 7 were divided into the EBV - group. There was no difference in age, sex and the number of organ damage between the children in the two groups (P>0.05). Duration of PE+CRRT was longer in the EBV + group as compared with the EBV - group (P<0.05). Before treatment, the ANC in the EBV + group was lower than that in the EBV - group (P<0.05), and there was no significant difference in the other observation indexes between the two groups (P>0.05). After treatment, Hb, Fib, APTT, SF, ALT, AST, LDH, Alb, CHE, TBil and TBA of the children in the EBV + group were significantly improved as compared with those before treatment (P<0.05), but ANC, PLT, TG showed not improve (P>0.05); Fib, APTT, SF, LDH, Alb, and CHE in the EBV - group were significantly as improved compared with those before treatment (P<0.05), while the ANC, PLT, Hb, TG, ALT, AST, TBil, and TBA were not improved (P>0.05). After treatment, the differences of Fib and SF in the children between the EBV + group and the EBV - group were statistically significant (P<0.05), and there was no significant difference in the other observation indexes of the children between the two groups (P>0.05). Compared with the children before treatment, EBV-DNA in the EBV + group were decreased significantly in 2-4 weeks after treatment (P<0.05). After PE+CRRT combined with chemotherapy, the overall survival rate of the children with severe HLH was 66.7%, and there was no significant difference in overall survival rate between EBV + group and EBV - group (P>0.05)., Conclusion: PE+CRRT combined with chemotherapy can reduce serum ferritin quickly, then improve organ function, and increase the overall survival rate of severe HLH, and it is a good effect on children with severe EBV-HLH and non-EBV-HLH.

Objective: To explore the correlation between the rs4705342 gene mutation in the promoter region of the miR-143/ miR-145 cluster and the risk of PCa in the Chinese Han population., Methods: We enrolled in this study 156 cases of PCa and 188 cancer-free controls. Using TaqMan SNP genotyping assay, we detected the polymorphism of rs4705342 in the promoter region of the miR-143/ miR-145 gene, and performed a stratified analysis on the family history of cancer, body mass index (BMI), age, smoking status, and alcohol consumption of the subjects., Results: The proportion of family history of cancer was significantly higher in the case group than in the control (P = 0.01). No statistically significant differences, however, were found between the two groups in age (P = 0.32), BMI (P = 0.79), smoking status (P = 0.47), and alcohol consumption (P = 0.34), nor in the distribution of the TT, CT and CC genotypes (P = 0.07) except in that of the CC and TT/CT combined genotypes (P = 0.01). There were statistically significant differences in the distribution of CC and TT/CT combined genotypes between the non-smoking subgroups, (P = 0.02) and alcohol drinking subgroups (P = 0.03), but not between the clinical stages of PCa (P = 0.81) or the levels of PSA (P = 0.39)., Conclusions: The rs4705342 gene mutation in the promoter region of the miR-143/ miR-145 cluster is significantly associated with the pathogenesis of PCa in the Chinese Han population.

The standard decoction of Chinese herbal decoction pieces is a standard reference substance to measure whether different dosage forms of Chinese medicine are basically consistent with those of clinical decoction,and provides new ideas and methods for effectively solving the problems of uneven quality in Chinese medicine dispensing granules. In this study,a systematic method for evaluating the quality of Scrophulariae Radix decoction was established from the perspective of " standard decoction",providing reference for the quality control of the Scrophulariae Radix dispensing granules. 15 batches of Scrophulariae Radix decoction pieces from different origins were collected,and 15 batches of standard decoctions were prepared according to the standardized process with water as solvent.Harpagide and harpagoside were used as quantitative detection indicators to determine the content,calculate the transfer rates and determine the extraction rate. The high performance liquid chromatography( HPLC) was used to establish a standard decoction fingerprint analysis method. The results showed that the transfer rates of harpagide and harpagoside in 15 batches of Scrophulariae Radix pieces standard decoction were( 70. 84±13. 39) % and( 48. 56±6. 40) % respectively; the extraction rate was( 57. 47±5. 89) %. Nine peaks were identified in the HPLC fingerprint,and the similarity was higher than 0. 97 between the fingerprints of 15 batches of standard decoction and the control fingerprint. In this study,the preparation process of standard decoction of Scrophulariae Radix pieces conformed to the traditional decoction preparation method. The sources of the samples were representative,and the established fingerprint method was stable and feasible,which can provide reference for the preparation and quality control of Scrophulariae Radix dispensing granules.

In this study, two school-aged children had an acute onset in spring and had the manifestations of fever, headache, vomiting, disturbance of consciousness, purpura and ecchymosis, and positive meningeal irritation sign. There were increases in peripheral white blood cells and neutrophils, but reductions in the hemoglobin level and platelet count in the two children. They had a significant increase in C-reactive protein. There were hundreds or thousands of white blood cells in the cerebrospinal fluid, mainly neutrophils. Increased protein contents but normal levels of glucose and chloride in the cerebrospinal fluid were found. Head CT scan showed multiple hematomas in the right cerebellum and both hemispheres in one child. Bone marrow cytology indicated infection in the bone marrow, and both blood culture and bone marrow culture showed methicillin-resistant Staphylococcus aureus (MRSA). Both patients had cardiac murmurs and progressive reductions in the hemoglobin level and platelet count during treatment, and echocardiography showed the formation of vegetation in the aortic valve. Therefore, the patients were diagnosed with infectious endocarditis (IE). Vancomycin was used as the anti-infective therapy based on the results of drug sensitivity test. One child was cured after 6 weeks, and the other child was withdrawn from the treatment and then died. Dynamic monitoring of cardiac murmurs should be performed for children with unexplained fever, and echocardiography should be performed in time to exclude IE. IE should also be considered for children with purulent meningitis and skin and mucosal bleeding which cannot be explained by the reduction in platelet count.

Thirteen compounds were isolated from the ethyl acetate extract of the rhizomes of Cyperus rotundus(Xiangfu) by means of various chromatographic techniques(silica gel, Al2O3, Sephadex LH-20, MCI GEL CHP-20P and HPLC), and their structures were identified as cyperotundic acid(1),(4S, 5E, 10R)-7-oxo-trinoreudesm-5-en-4β-ol(2), 4-hydroxy-4, 7-dimethyl-1-tetralone(3), taraxerone(4), dammaradienyl acetate(5), zeorin(6), sarmentine(7), guineensine(8), pellitorine(9), caprolactam(10), liriodendrin(11), 3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-2-[4-(3-hydroxy-1-(E)-propenyl)-2,6-dimethoxyphenoxy]propyl-β-D-glucopyranoside(12)and 1-(3, 4-methylenedioxyphenyl)-1E-tetradecene(13) by extensive spectroscopic analyses(IR, MS, 1D-and 2D-NMR). To study the chemical constituents of Lepidium meyenii, the air-dried rhizome of L. meyenii was extracted with 70% EtOH. The extract was condensed to a small amount of volume and extracted with petroleum ether, EtOAc and n-BuOH, successively. The compounds were isolated and purified by column chromatography, and identified based on spectral analyses (1H-NMR, 13C-NMR, HRESIMS). Eighteen compounds were isolated from L. meyenii, including 7 alkaloids and 4 fatty acids and 7 other compounds. They were characterized as (3-hydroxybenzyl) carbamic acid(1), phenylmethanamine(2), N-benzylformamide (3), N-benzylacetamide (4), pyridin-4-ylmethanamine(5), n-(4-methoxybenzyl) aniline(6), uracil(7), succininc acid(8), decanedioic acid(9), n-hexa- decanoic acid methyl ester(10), heptanoic acid(11), solerole(12), pyromucic acid methyl ester(13), 5-hydroxymethyl-2-furancar- boxadehyde(14), 5-(methoxymethyl)-1H-pyrrole-2-carbaldehyde(15), 1,7-dihydroxy-2,3, 4-trimethoxyxanthone (16), 1,7-di- hydroxy-3,4- dimethoxy-xanthone(17), (+)-pinoresinol(18). Meanwhile, compounds 1-18 were obtained from L. neyenii for the first time.

Objective: To study the clinical and pathological characteristics, diagnoses and treatments of nonfunctioning parathyroid cysts., Methods: Six cases of nonfunctioning parathyroid cysts treated in Tongren Hospital during 2002 - 2009 were retrospectively analyzed. Nonfunctioning parathyroid cysts in the six patients were inadvertently found as neck masses by physical examination. The levels of serum calcium, phosphorus and parathyroid hormone were normal. Five cases of 6 patients with imaging suggested the existence of cystic mass in the back of inferior thyroid in 5 cases of the 6 patients., Results: Tumors in the 6 patients were removed surgically and diagnosed as parathyroid cysts with post-operative pathological examination. PTH (parathyroid hormone), CgA (chromogranin A), Syn (synaptophysin) expressions in the tumors were positive. No recurrence was found with follow-up of 2 - 9 years after operation., Conclusions: Surgical resection is most effective for the treatments of nonfunctioning parathyroid cysts and pathologic examination is required for the determined diagnosis of this disease. Fine needle aspiration can be helpful for the diagnosis before operation.

Objective: It is known that Notch signal is very important to vascular remodeling during the process of embryonic development, vessel repair and tumor growth, but there are few studies about pulmonary vascular remodeling in pulmonary hypertension. This study was to explore the effect of inhibiting Notch signal on pulmonary vascular remodeling induced by angiotensin II., Methods: Vessel strips taken from healthy Wistar rats were co-cultured with extrogenous angiotensin II and the potent smooth muscle cell proliferation stimulators for 7 days. Vascular wall thickness, proliferating cell nuclear antigen (PCNA) positive cell rate and caspase-3 positive cell rate were examined in vessel strips. Then some vessel strips were cultured with angiotensin II and γ-secretase inhibitor DAPT, a Notch signaling inhibitor for 7 days. The levels of Notch 1 to 4 receptor and HERP1/2 mRNA were ascertained by FQ-PCR., Results: Angiotensin II stimulation in the cultured normal pulmonary arteries resulted in an increase in the vascular medial thickness by nearly 50%, and a significant increase in the PCNA positive cell rate and a decrease in the caspase-3 positive cell rate. DAPT treatment did not result in the alterations of Notch 1 to 4 receptor levels, but decreased remarkably HERP1 and HERP2 mRNA expression. DAPT treatment also decreased angiotensin II-induced vascular medial thickness and PCNA positive cell rate and increased caspase-3 positive cell rate., Conclusions: Inhibiting Notch signal by γ-secretase inhibitor may lead to the suppression of pulmonary vascular remodeling induced by angiotensin II, suggesting that the inhibition of Notch signal pathway might be a novel strategy for the treatment of pulmonary hypertension.

Objective: To screen and identify new specific antigen gene from a cDNA library of adult Clonorchis sinensis, and investigate the immunogenicity of the recombinant proteins., Methods: The lambdaZAP cDNA library of adult C. sinensis was immunoscreened with pooled sera of clonorchiasis patients. The positive clones were sequenced and analyzed. The sequence encoding the mature peptide was cloned into prokaryotic expression vector pET28b(+). The recombinant plasmid was transformed into E. coli BLR21 (DE3) or BLR21 (DE3) pLysS and followed by expression of the protein induced by IPTG. The recombinant protein was purified by His-bind-resin (Ni-NTA) affinity chromatography and identified by Western blotting. BALB/c mice were immunized with purified recombinant pET28b-Cs2 protein, and the sera from immunized mice were analyzed for specific antibodies by ELISA., Results: A total of 44 positive clones were isolated from the C. sinensis cDNA library. Three clones containing specific tandem repeats of PPMP amino acid sequence were named as C. sinensis PPMP antigen genes. The genes containing KPPMPGDRDA, QPPMPGGRDA were named as type PPMP I and type PPMP II antigens, respectively. Sequence analysis revealed that these PPMP genes were a novel specific C. sinensis antigen gene family. Two new genes, PPMP I Cs2 and PPMP II Cs3, were expressed in E. coli, and SDS-PAGE showed that the two recombinant proteins were about M(r) 22 000 and M(r) 39 000. The two soluble recombinant proteins were recognized by pooled sera of clonorchiasis patients. A high level of specific IgG against the recombinant proteins (maximum dilution 1 : 64 000) was produced in immunized mice., Conclusion: A novel PPMP gene family of C. sinensis has been identified, and its recombinant proteins show high immunogenicity.

Objective: To investigate the effect of polycythemia on hypoxia induced pulmonary hypertension and pulmonary vascular remodeling in rats., Methods: The healthy female Sprague-Dawley rats were randomly divided into 3 groups: normoxia control group (C group), hypoxia group (H group), hypoxia + different doses of human recombine hemopoietin (rEPO) group. All rats in hyoxia groups were exposed to hypoxia, 8 hours every day, for 21 days. The rEPO groups were injected sc with different doses of rEPO (300 U/kg, 600 U/kg, 900 U/kg, 1200 U/kg) thrice weekly. Blood samples were taken for the measurement of RBC, Hb, Hct, plasma EPO concentration, whole blood/plasma viscosities, the animals were then catheterized to record mean pulmonary arterial pressure (mPAP) and demised to calculate the ratio [RV/(LV+S)]. Percentage of vascular wall thickness and muscularization of non-muscular pulmonary arteriole were examined microscopically., Results: (1) As the dosage of exogenous rEPO increased, blood concentration of EPO increased correspondingly, as RBC, Hb, Hct and whole blood/plasma viscosities increased in various degrees. (2) There was positive correlation between whole blood viscosity and Hct at both high and low shears and linear correlation between mPAP and whole blood viscosity at high shear. (3) The degree of pulmonary hypertension, reflected by mPAP increased in accordance to rEPO dosage increment. However, the extent of pulmonary vascular remodeling alleviated somehow as the rEPO dose increased and so did right ventricular hypertrophy., Conclusion: Polycythemia induced by exogenous EPO increases the blood viscosity and the pulmonary vascular resistance, which contributes to the formation of hypoxia induced pulmonary hypertension.

Objective: To investigate the distribution of common food-borne parasitic diseases and dietary habits in Guangxi., Methods: In the survey from 2002 to 2005, questionnairing, fecal examination, ELISA or skin test were applied in the endemic areas of clonorchiasis, taeniasis, cysticercosis, trichinelliasis or paragonimiasis in Guangxi Zhuang Autonomous Region. 13 990 residents at 27 sites in 9 counties were examined for clonorchiasis, 22772 residents at 22 sites in 10 counties for taeniasis, 9 638 residents at 20 sites in 10 counties for trichinelliasis and 9 347 residents at 18 sites in 6 counties for paragonimiasis., Results: Among 109 counties, residents in 64 counties (59.8%) were found with local habit of eating raw meat, covering Han and other 6 ethnic groups (Zhuang, Yao, Miao, Maonan, Dong and Buyi) . Various animal food such as fish, pork, pork liver, swine blood, beef, ox blood, chicken blood, goat blood, crab, shrimp, clamworm, snail, oyster, and duck meat were consumed in raw with or without seasonings, or by ways of hot-pot, pickle, dipping in boiling water or wine-soaked. Food-borne diseases such as clonorchiasis, taeniasis, cysticercosis, trichinelliasis and paragonimiasis were prevalent in the Region. The survey showed that clonorchiasis is prevalent mainly in the river basins of Yongjiang, Zuojiang and Youjiang in the middle-west and southern part of the Region with a prevalence of 9.8%, taeniasis and cysticercosis in the north and middle-north such as Rongshui prevalence of 43.3% and 2.1% respectively, Bingyang (2.1% and 1.0%) and Tianlin (1.6% and 1.1%) counties. Sporadical cases of trichinelliasis were found in the counties of Debao, Baise, Luzhai and Nandan with positive rate 9.8%, 6.4%, 5.3% and 3.1% by serological test. Skin test on paragonimiasis among residents in sampled areas revealed a positive rate of 2.6% and the cases mainly distributed in Rongshui, Longsheng and Quanzhou counties., Conclusion: Consumption of raw or undercooked meat is popular in the local residents especially the ethnics, which is an important factor in the prevalence of food-borne parasitic diseases.

Objective: To describe the discovery of a residual foci of bancroftian filariasis in Fuchuan County where the disease was announced to have been eliminated, and reveal its epidemiologic feature., Methods: The investigation was carried out from August 2007 to March 2008 among residents in Changtang village where the first case of filariasis was found and the neighboring villages. They were screened with two thick blood smears. Immunochromatographic technology (ICT) was conducted for those going out but returned and those in surrounding areas. Vector mosquitoes were collected and dissected to find filaria larvae. Historical documents were reviewed and relevant people were interviewed., Results: In Changtang administrative village, 1052 residents were screened and 19 cases with microfilaremia were found in 2 natural villages, with a Mf-positive rate of 1.8% (5.1% in Gangshang and 1.4% in Yinshan respectively). No Mf-positive case was found in 4119 residents screened in other 3 villages. The average microfilaria density in the 19 cases was 17.37/60 +/- 1 blood. All the 19 cases belonged to 12 families, and 13 cases were relatives to each other, which showed a feature of spatial clustering and family clustering. More patients were identified in the age groups of 20-29 and 50-59, and 57.9% of them were older than 50 years. No larvae were found in 54 Culex pipiens fatigans dissected., Conclusion: The Changtang village is identified as a residual focus of bancroftian filariasis with a low, limited endemicity. More cases have been among the elderly with a low average microfilaremia.

To observe the graft-versus-host disease (GVHD) in rat-to-mouse model of bone marrow transplantation to build a GVHD model, BALB/c mice were conditioned with 8.5 Gy lethal total body irradiation and divided into two groups. One group of mice was infused with 4 x 10(7) bone marrow cells (BMC) from SD rats. The other group of mice was infused with 4 x 10(7) bone marrow cell and 2 x 10(7) spleen cells from SD rats. GVHD in mice of two groups were observed for 60 days. The results showed that mice in the group infused with only BMC mostly (80%) survived more than 60 days, but in the other group infused with mixed BMC and spleen cells, all mice died within 14 days and showed GVHD with pathologic evidence. In conclusion, to induce GVHD in rat-to-mouse bone marrow transplantation needs additional rat spleen cells.
