The professional certification of engineering education is an important measure in training the engineering talents in new era. Its education concept happens to coincide with the goal of ''curriculum ideological and political education (CIPE)'' of specialized courses. Under the background of engineering certification of food science and engineering, this paper summarized the curriculum characteristics of Food Machinery and Equipment, explored the ideological and political elements of the curriculum, and analyzed the relationship between the objectives of curriculum and CIPE based on the Outcomes-based Education (OBE) concept. Besides, the strategies for the integration of CIPE with the professional certification system of engineering education were discussed. The evaluation and continuous improvement plan for CIPE were also set up based on the OBE concept. This study would provide basis and supports for further improving the level of CIPE in specialized courses, and implementing the fundamental task of morality cultivation.

In order to effectively avoid the possible drawbacks of water seal inhibiting gas resolution and reducing coal seam permeability, based on the existing air jet research foundation, theoretical analysis and laboratory experiments were used to analyze and study the punching characteristics of high-pressure air jets and the law of punching and breaking coal under the combination of jet parameters. The results show that, among the three factors affecting the experimental results, the jet pressure has the greatest influence, the impact target distance is second, and the nozzle size has the least influence on the experimental results; the optimal combination of jet parameters in the experimental range is 12 MPa, 30 mm impact target distance and 3.5 Mach number nozzle size. The effect of jet punching increases with the increase of jet pressure, when the jet pressure reaches 10 MPa, the specimen penetrates, and when it is further increased to 12 MPa, the specimen is broken; there is an optimal impact target distance for jet punching, and the jet punching effect is the best under this target distance, which is generally 6-7 times the nozzle diameter; the nozzle size has little effect on the effect of punching experiments, and the punching effect tends to be constant.

ObjectivesA crankshaft is a key component of marine diesel engines. In order to monitor crankshaft stress and ensure the safe and reliable operation of ships, a crankshaft stress monitoring method based on digital twin technology is proposed.MethodsBased on the digital twin concept, a framework for the intelligent operation and maintenance of marine diesel engines is put forward. Taking a certain type of in-line 6-cylinder marine medium-speed diesel engine as the research object, a digital crankshaft health condition assessment system is developed on the basis of available data from real engines. First, based on an RBF neural network, the cylinder head vibration acceleration signal is used to identify in-cylinder pressure, then the load of the crank pin is calculated. Based on the finite element method, the key positions which affect the lifecycle of the crankshaft are obtained after analyzing its stress and fatigue. A BP neural network is used to evaluate the stress of the crankshaft after a reduced-order model of forces and stresses is used to determine real-time performance. A BP neural network is then used to evaluate the stress of the crankshaft.ResultsThe prediction errors of the cylinder pressure and crankshaft stress of the proposed model are both less than 5%, and the stress prediction time is shortened to the second level, realizing the real-time/quasi-real-time update of the twin model.ConclusionsThe results of this study show that it can be used as a new reference for the real-time monitoring and intelligent operation and maintenance of diesel engine components.

In this paper, the talent training goal of the major of food science and engineering and the concept of OBE were combined to evaluate the achievement degree of curriculum objectives of food machinery and equipment. The corresponding relationship between curriculum objectives and graduation requirements was determined, the teaching content and assessment method were formulated. The evaluation method and evaluation system of the achievement degree of curriculum objectives were designed. Afterward, the system was used to evaluate the achievement degree of curriculum objectives and the continuous improvement measures were suggested, so as to achieve the improvement and enhancement of curriculum teaching quality. It could effectively improve the quality of talent training for the major of food science and engineering.

A method of heat storage and temperature control for refuge chamber with phase change energy storage wall is put forward, that is, coupled temperature control method of mine air pressure, phase change cold storage for energy storage wall under conventional condition, and wall phase change heat absorption when mine accident occurs, the diatomaceous earth-based phase change material was used as the energy storage aggregate additive to prepare composite cement mortar, which was then laid on the wall of the refuge chamber to achieve temperature control effect. The test results show that when the amount of energy storage aggregate is 15% and the amount of activated carbon is 6%, the prepared composite cement mortar can be adapted to the energy storage wall structure of the refuge chamber and improve the indoor thermal environment of the refuge chamber.

This paper proposes a method to control the heat transfer from the surrounding rock to the roadway, that is, a new type of thermal insulation mortar is laid on the wall of the roadway surrounding rock, and the optimal proportion of three additives in the thermal insulation mortar is determined through orthogonal experiments, and the basic experimental measurement of the thermal insulation mortar under the optimal proportion is carried out. The experimental results show that with the increase of the three admixtures, the thermal conductivity of thermal insulation mortar decreases significantly, and the compressive strength decreases. When the content of fly ash ceramsite is 96%, the content of basalt fiber is 16%, and the content of expanded vermiculite is 16%, the performance of the specimen is the best.

Diabetes is a chronic metabolic disease caused by multiple factors, and it is an epidemic worldwide in the 21st century. The intestinal flora of diabetic patients is imbalanced, with reduced antioxidant capacity and low-grade inflammation. Relevant studies have shown that probiotics, as a kind of living microorganisms that are beneficial to the health of the intestine, can relieve the symptoms of diabetes by balancing the intestinal flora, reducing the body's oxidative damage and regulating the body's immune factors. This article combs the two commonly used methods for screening hypoglycemic strains with α-glucosidase and DPP-IV as targets, and evaluates the hypoglycemic effect of probiotics through in vivo experiments. At the same time, it further summarizes the mechanism of probiotics in alleviating type 2 diabetes from five aspects such as intestinal flora, immune response, oxidative stress, short-chain fatty acids and intestinal barrier, in order to provide a certain reference for related research on type 2 diabetes.

In view of the complicated operation of the current crack detection method of hydraulic support, a weak magnetic nondestructive detection method for cracks in the top beam of hydraulic support based on geomagnetic field excitation is proposed. Firstly, the stress field of the crack in the top beam of hydraulic support is analyzed by using COMSOL Multiphysics simulation software. Secondly, based on the principle of weak magnetic nondestructive testing, the geomagnetic field is loaded in the simulation space. The superimposed field formed by the magnetized material with crack defects and the geomagnetic field is obtained and analyzed. The results show the following points. ① There is stress concentration phenomenon in the top beam defect of hydraulic support. The lower the stress is, the greater the stress is. The crack expands from the outside to the inside to both sides. The failure risk of the top beam can be effectively reduced by finding the surface crack in time. ② When the length and depth of the crack defect are fixed, the horizontal distance between the valley and the peak and the peak value of the magnetic flux density mode curve of different width cracks increase with the increase of the width. The valley value decreases first and then increases with the increase of the width. The amplitude of the flux density mode increases with the crack width. The variation rate of the difference between the adjacent maximum valley and peak of the flux density mode decreases with the crack width. The change amplitude of magnetic flux density is positively correlated with the change of crack width. ③ When the length and width of the crack defect are fixed, the valley value of the magnetic flux density mode curve of different depth cracks has little difference. The left peak value increases with the increase of the width. The right peak value decreases with the increase of the width. With the increase of the crack depth, the change amplitude of the flux density mode increases. The change rate of the difference between the adjacent maximum valley and peak of the flux density mode is almost constant. The change amplitude of magnetic flux density is positively correlated with the change of crack depth. The magnetic flux density is more sensitive to the change of crack width than to the change of crack depth. ④ When the length, width and depth of the crack defect are fixed, the change in the direction of the crack does not affect the judgment of crack defects.

In order to clarify the influence of the distance of the air pressure cylinder and the ratio of the pumping and air pressure on the cooling of the driving face, a 1∶1 proportional physical model of 7302 track along the tunnel face of Zhaolou Coal Mine was established, and the air duct was compressed using AnsysCFD software. AnsysCFD software was used to numerically simulate the influence of air flow parameters on tunneling working face under the conditions of distance Lf (set as 2.5, 5, 7.5, and 10 m respectively), ventilation rate and pressure rate ratio B(set as 1, 1.5, and 2 respectively) between the air outlet of pressure pipe and the end face of heading face. The results show that when Lf is greater than 5 m, the wind flow in the working area can be maintained above 0.3 m/s. With the increase of Lf, the change of the vortex field near the tunneling head increases. When Lf=5 m and B=1.5, the average temperature of the roadway in the working area is 27 °C, and the long-drainage and short-pressure mixed ventilation system can achieve the best cooling effect. After applying the numerical simulation results to the optimization of the ventilation system of Zhaolou Coal Mine 7302 driving face, the wind speed and wind temperature were measured at the measuring points on multiple cross sections in the face. The average relative error between the measured wind speed and the numerical simulation results was 10.07%. The average relative error between temperature and numerical simulation results is 1.60%, and the numerical simulation results are more accurate.

The high morbidity and mortality of non-small cell lung cancer (NSCLC) did influence the quality of life of tumor patients world-wide. There is an urgent need to develop new therapies that have high anti-tumor activity and low toxicity side effects. It is widely accepted that autophagy can play diverse roles in carcinogenesis, such as induces pro-death of lung cancer cells or helps the escape from cell death, making it become a proper anticancer target. It’s believed that various monomers of Chinese traditional medicine closely correlates to anti-NSCLC activities, and that even could affect the acquired multiple drug resistance (MDR). Furthermore, autophagy might be the underling mechanisms which could play a role as the candidate targets of natural active compounds. Recent studies of terpenoids, alkaloid, dietary polyphenols, saponins and other active ingredients that extracted from a large variety of herbs suggest that different monomer compounds could either regulate the activity of pro-death autophagy or influence the level of protective autophagy of NSCLC cells, thus changing their drug sensitivity and cell viability. This paper aims to give a systemic description of the latest advances about natural compounds and their derivatives that involved in tumorigenesis of NSCLC via inducing the autophagy.

Objective To investigate the imaging and clinicopathological features of pachygyria limited in the right temporo-parieto-occipital lobe and the key points of its diagnosis and treatment, in order to improve the recognition of this disease. Methods and Results A 2-year-old boy was admitted to hospital because of paroxysmal loss of consciousness and convulsion for 18 months with progressive aggravation. MRI showed malformations of cortical development in the right temporo-parieto-occipital lobe. Epileptic foci resection on the right temporo-parieto-occipital lobe was made. Histological examination after operation showed uneven thickening of gray matter, shrinking of white matter and disappearing cortical stratification, while a lot of dysmorphic neurons, balloon cells and scattered balloon cells in white matter appeared. Immunohistochemical staining revealed that dysmorphic neurons were positive for non-phosphorylated neurofilament protein SMI-32, microtubule-associated protein-2 (MAP-2) and vimentin (Vim) or neurofilament protein (NF). Both dysmorphic neurons and balloon cells expressed phosphorylated ribosomal S6 protein (RPS6), while the former was stronger than the latter. Balloon cells were not positive for MAP-2 or Vim. No disturbance of consciousness or limb twitches occurred in this patient during one-year follow-up. Conclusions Congenital pachygyria was cortical dysplasia caused by the early proliferation and migration disorder of brain, and should be distinguished with focal cortical dysplasia (FCD) type Ⅱ b and tuberous sclerosis complex (TSC). Lung cancer, of which approximately 85% are non-small cell lung cancer (NSCLC), is the most leading cause of cancer mortality throughout the world. Despite recent improvements in its treatment, it still remains a highly lethal disease. It is becoming increasingly clear that estrogen and estrogen receptors are involve in the pathogenesis and development of lung cancer. Estrogen receptor modulators and aromatase inhibitor may reverse the drug-resistance of platinum based chemotherapy in NSCLC patients and can enhance curative effect of epidermal growth factor receptor tyrosine kinase inhibitor. In this article the progress in the function of estrogen in NSCLC and the treatment based on the ER signaling pathways for NSCLC is reviewed.

Chemopreventive drugs including natural chemopreventive drugs and synthetic chemopreventive drugs, it not only can prevent cancer, can also play a role in tumor treatment. MicroRNAs (miRNAs) is a kind of short chains of non-coding RNA, regulating the expression of many genes through the way of degradation of mRNA or inhibitting mRNA translation. In recent years, more and more studies have shown that chemopreventive drugs through influence the expression of miRNAs and their target genes play a role in the prevention and treatment in a variety of tumors, and chemopreventive drugs on the experimental study of miRNAs and their target genes in tumor have demonstrated a good safety and efficacy. Effect on chemopreventive drugs-based microRNAs and their target genes into cancer cells will be expected as a new starting point for cancer research. The thesis expounds and analyzes between the natural chemopreventive drugs and synthetic chemopreventive drugs and miRNAs and their target genes in tumor research progress.

Background and objective Pulmonary fibrosis is a common pathological phenomenon in lung cancer patients after chemotherapy or radiotherapy. It is also a key hindrance to the transport of drugs to lung tissue. Peptide transporters have become a target of the rational design of peptides and peptide drugs. The aim of this study is to investigates the expression of peptide transporter 2 (PEPT2) mRNA in the lungs of rats with bleomycin (BLM)-induced pulmonary fibrosis. Methods Fifty healthy adult Sprague-Dawley rats were randomly divided into five groups. One group was untreated (control), the second group was injected with normal saline solution (NS), and the three remaining groups were treated with a single dose of bleomycin to induce pulmonary fibrosis (BLM). Rats from the NS group were killed by exsanguination on day 14. Rats from the BLM group were killed by exsanguination on days 7, 14, and 28. The lung samples were observed under light microscopy and the hydroxyproline concentration was determined. The expression levels of PEPT2 mRNA were measured by RT-PCR. Results The morphological study showed that collagenous fiber proliferated in the lungs of rats injected with BLM, indicating pulmonary fibrosis. This proliferation was apparent at 14 d post-injection and especially at 28 d post-injection. Hydroxyproline levels increased seven days post-injection compared with the control group and NS group, but there was no significant statistical difference (P>0.05). Hydroxyproline levels significantly increased (P0.05). Conclusion PEPT2 is a potential peptide drug target in the treatment of pulmonary fibrosis, although there were no significant changes of PEPT2 mRNA expression in the lungs of rats with bleomycin-induced pulmonary fibrosis.

Background and objective The therapeutic efficacy of late lung-cancer was very poor, and cytokine-induced killer cells (CIK) were paid more attention to treat non-small cell lung cancer (NSCLC). The aim of this study is to get insight into the role of bronchial arterial infusion bronchial arterial infusion (BAI) plus CIK about NSCLC by comparing therapeutic efficacy among BAI, traditional vein chemotherapy and BAI plus CIK, for late NSCLC. Methods A total of 120 patients were enrolled in this study, dividing randomly into three groups: bronchial arterial infusion (BAI), traditional vein chemotherapy and BAI plus CIK. Clinical effects and side effects were estimated after two period of therapy. Results The effective rate (CR+PR%) of combined group is higher than the traditional vein chemotherapy group (66.67%, n=39) and there are significant differences (χ2=4.721, P=0.03); The side effect of rate of BAI plus CIK group is significantly lower than the traditional vein chemotherapy group, and so did the non-bone marrow inhibition side effects (P < 0.05). The tumor progression rate (PD%) of bronchial arterial infusion (BAI) group is higher than combined group (χ2=4.287, P=0.038). There was no difference between the traditional vein chemotherapy group and combined group (χ2=0.082, P=0.775). Conclusion Bronchial Artery Infusion combined with cytokine-induced killer cells is an ideal, safety, effective comprehensive treatment method for late stage lung cancer.

Background and objective Patients with adenocarcinoma of the lung have disproportionately response to the epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI). The aim of this study is to analyze the difference of EGFR gene amplification in bronchioloalveolar carcinoma (BAC), adenocarcinma mixed subtype and conventional adenocarcinoma of the lung and provide some information to clinical therapies. Methods Lung cancer cases were collected and reviewed from the archives of the Department of Pathology, Chinese PLA General Hospital during the time period from 2004 to 2006. The definite diagnosis of BAC based on 2004 WHO classification of lung tumors was made by two pathologists. Fluorescence in situ hybridization (FISH) was performed to detect EGFR gene amplification in pure BAC, adenocarcinma mixed subtype and conventional adenocarcinoma. MS nail free adhesive was prepared with silane-modified polyether (MS) resin as basic polymer, nano-calcium carbonate as filler, plasticizer and thixotropic agent. The influence of the types of resin, plasticizer and thixotropic agent on the mechanical properties of MS sealants was discussed. The results show that the MS nail free adhesive has a good comprehensive properties when the basic polymer is STP-E3, the plasticizer is polyether polyol and the thixotropic agent is polyamide wax, with tensile shear strength of 1.8 MPa, surface drying time of 5 min, initial anti slip of 2 mm, 24 h vulcanization depth of 3.2 mm and extrudability of 100 mL/min, applicable in the field of home decoration. The two-component thermal conductive liquid silicone rubber was filled with the different types of hollow glass beads as the lower density filler. The influence of hollow glass beads and their content on the density, viscosity and thermal conductive of the material was investigated. Results indicate that the hollow glass microsphere can reduce the density of liquid silicone rubber. When adding the same dosage, the type A hollow glass microspheres with large particle size and low density have a great influence on the density of the sealant. The addition of hollow glass microspheres will increase the viscosity of the sealant, and the effect on the liquid silicone rubber with high thermal conductivity is more significant. Enzymatic catalysis is the main process for the trehalose production. In this study, we used high cell density cultivation of recombinant Escherichia coli to produce trehalose synthase and synthesized trehalose using maltose as a substrate. Firstly, according to conditions of high cell density culture of E. coli, we optimized the culture medium, fermentation conditions and induction conditions for producing trehalose synthase. Then, the scale- up experiments of fed- batch fermentation were conducted in 5 L and 50 L fermentors. Finally the fed- batch fermentation strategy- changed exponential- pH- stat feeding was carried out to produce trehalose synthase. The cell density and enzyme activity was 97 (OD600) and (24 000±350)U/mL, respectively, which greatly improved the production efficiency of trehalose. [ABSTRACT FROM AUTHOR]

In this study, co-electrodeposition was used to fabricate CuZnSn precursor, and followed by preheating and sulfurization to synthesize CuvZnSnSi absorber layer. The co-deposition potential E = -1.25 V was determined by cyclic voltammetry (CV). The effects of pH and electrolyte concentration on the composition and phases of the metal precursor were investigated. By optimization of the parameters, a CZT film with composition close to n ( Cu )/n (Zn + Sn ) ≈ 0.8, n (Zn)/n (Sn) ≈ 1.2 was deposited from an aqueous solution containing 8 mmol/L SnSO4 22 mmol/L CuSO4 ⋅ 5H2O, 22 mmol/L ZnSO4 ⋅ 7H2O, and 200 mmol/L Na3C6H5O7 ⋅ 2H2O with pH = 5. 7. After preheating at 300 °C, the metal precursor transformed into a solid solution of Cu5Zn8 and Cu6Sn5 alloys, which established the foundation for a pure CZTS formation. During sulfurization, annealing time and temperature had a significant impact on the structure and morphology of the film. With the increase in annealing time, the intensity of the characteristic peak of CZTS increased, the intensity of impurity phase peaks (Cu2-xS, SnS and Cu2Sn3) decreased, and CZTS was purified gradually. When the sulfurization time was up to 60 min, the film had a good structure and morphology? but the film was easy to decompose with longer annealing. Too low sulfurization temperature would lead to binary and ternary phases in the film? while too high temperature would result in the loss of Sn and Zn. When sulfurization temperature was 580 °C, the CZTS film had obvious grain profile and distinct grain boundaries, and was dense and uniform (around 2 µm thickness) with less voids and good adhesion to the Mo layer. To study the unipotency of free group generated by two elements, some combinations of pimitive elements are discussed based on the combinatorial group theory, and some new systems benefit to studing the unipotency will be proved. The article presents a study, which examines the use of combined penalization for selection of variables in joint mean and variance models.

Studies of voluntary behavior among Chinese urban residents have important implications to understanding the development of citizenship and the future development of philanthropy sector in China. This article, which is based on a nationwide survey, studies the relationship between human capital, i. e. education level, economic status and health conditions, and voluntary behavior in China. It is found that 37% of the respondents have participated in voluntary activities in the past year. Among them, the better educated and healthier respondents are more likely to have participated in these activities. The results are statistically significant. However, citizens' economic conditions appear to have a negative relationship with the likelihood of engaging in voluntary activities. Better-off citizens are less likely to participate in voluntary activities. This phenomenon may be caused by increased opportunity costs for the better-off citizens. It might also mean that, despite improved economic conditions, the sense of citizenship and social responsibility has yet to grow. [ABSTRACT FROM AUTHOR]

Angiotension converting enzyme (ACE) inhibitory peptides from high-temperature sesame meal protein hydrolyzed by subcritical water were separated and purified by sequential nanofiltration, ultrafiltration and liquid chromatography, and their structures were identified by liquid chromatography-mass spectrometry (LC-MS). The ACE inhibitory activity was verified using the synthetic oligopeptides. The absorption and metabolism characteristics of the oligopeptides were predicted, a three-dimensional quantitative structure-activity relationship (3D-QSAR) model was established, and molecular docking analysis was carried out. The peptide fraction with molecular mass less than 3 kDa had the strongest ACE inhibitory effect. After further purification, nine ACE inhibitory peptides were identified from peak 1. These peptides could not interfere with the normal physiological activities of the human body. The 3D-QSAR model for LFRAF was successfully established using the comparative molecular force field analysis (CoMFA) method. The positively charged amino acid residues and the introduced large groups on the side chain at the C-terminus of the ACE inhibitory peptides could improve their activity. LFRAF occupied the S2 and S1’ active pockets of the ACE peptides and bund to Zn2+ to inhibit ACE activity. These results indicate that it is feasible to use subcritical water technology to degrade high-temperature sesame meal proteins to prepare ACE inhibitory peptides.

In order to clarify the change of the corrosion properties of 2205 duplex stainless steel in artificial seawater after cold rolling，2205 duplex stainless steel was cold-rolled at different reduction rates (PCRR ＝20%-80%)，and then the sample was studied by metallographic observation and hardness measurement.The corrosion resistance of cold-rolled 2205 duplex stainless steel in artificial seawater (3.5%NaCl)was investigated using dynamic potential polarization curve and electrochemical impedance spectroscopy.Results showed that the hardness and corrosion resistance of 2205 stainless steel presented non-monotonic variation with the increase of PCRR.In the interval from 0 to 40%，the corrosion resistance and hardness increased with the increase of the reduction rate，and the self-corrosion potential increased from -309 mV to-269 mV.In the interval from 40%to 80%，the corrosion resistance decreased with the increase of the reduction rate，and the self-corrosion potential decreased from -269 mV to -322 mV.Moreover，the corrosion resistance was highest at the reduction rate of 40%，and the Brinell hardness of the sample with the reduction rate of 60%was the highest，about 374 HBW.Suitable cold rolling could result in high-density dislocation and structure refinement，which was beneficial to the corrosion resistance of 2205 duplex stainless steel.However，an excessive reduction rate would result in the precipitation of σ-phase which was detrimental to corrosion resistance.

“Lab on paper” is a paper-based microanalytical system with unique advantages such as high sensitivity, low cost, portability, and mass production, which breaks through the limitations of traditional detection technologies and shows great potential in the field of rapid on-site food detection. In this paper, the typical “labs on paper”, including chromatographic paper, chemical test chip, lateral flow assay (LFA), microfluidic paper-based analytical devices (μPADs), and synthetic biological paper, are systematically reviewed. Special emphasis is put on the application of “lab on paper” in the field of food detection. Finally, the advantages, challenges, and future prospects of paper-based analytic methods are discussed, which may provide theoretical support for better application of “lab on paper” in the field of food detection.

Objective This study combined public data analysis with biological experiment using the tissues of hepato-cellular carcinoma to explore the epigenetic regulation mechanism of EZH2 gene which is potentially related to the progression of hepato-cellular carcinoma. Methods Using the TCGA's public clinical data of hepato-cellular carcinoma, the relation of EZH2 gene expression with tumor progression as well as prognosis was analyzed. Subsequently, the effect of EZH2 inhibitor GSK343(10 μmol/L) on the proliferation of hepato-cellular carcinoma cell line HepG2 was further analyzed by ATP cell activity assay and crystal violet assay. Then, the differentially expressed genes between EZH2 high expression group and low expression group in TCGA samples were analyzed. After that, the ChIP-seq public data of ENCODE was used to analyze the histone modification at the transcription starting site of EZH2 downstream target gene. Finally, RT-qPCR was used to detect the gene expression of downstream target genes regulated by EZH2. Results EZH2 gene-expression was significantly correlated with the progression of hepato-cellular carcinoma from stage 1 to stage 3(P＜0.05). Survival analysis showed that the high expression of EZH2 gene was significantly correlated with the poor prognosis of hepatocellular carcinoma(P＜0.05). The EZH2 inhibitor could significantly inhibit the proliferation of hepatocellular carcinoma cell line HepG2 in vitro (P＜0.05). Histone modification H3K27me3 was enriched at the transcription start site of EZH2 binding gene. There was 163 genes significantly different in EZH2 high expression group and low expression group and had EZH2 binding sites at their transcription start sites. The experiment verified that ADRA1A was a downstream target gene of EZH2. Conclusions EZH2 gene may affect expression of its downstream target genes through epigenetic regulation in hepato-cellular carcinoma.

To improve the thermal insulation performance of thermal insulation concrete for mining and save energy, we developed an environmentally friendly thermal insulation concrete with better thermal insulation and performance using a recyclable green organic matter wheat straw fiber. Scanning electron microscopy of the wheat straw fiber revealed that it had a large number of closed microporous structures, which are good heat insulation materials. Different percentages of wheat straw fibers(0%, 2%, 4%, 6%, 8%, 10%) and cement quality were mixed into the thermal insulation mortar, and the thermal conductivity of the thermal insulation concrete gradually decreased. When maintained at 0.201 0-0.200 2 W/(m·K), the compressive strength first increased and then decreased, and the maximum compressive strength was 7.14 MPa. Adding 2% of wheat straw fiber can improve the thermal insulation performance and compressive strength. Compared with the heat-insulation concrete without straw fiber, the heat-insulation property and compressive strength are improved, and the heat-insulation property and compressive strength are increased 7.5% and 5.9% respectively.

The unsafe behavior of miners is the direct cause of coal mine accidents. In order to realize the control and management of miners’ unsafe behavior fundamentally, from the interaction between internal motivation and external management system, the miners’ safety motivation is divided into four parts by self-determination theory. The model of miners’ safety behavior selection is constructed, and the risk matrix diagram of safety color behavior is constructed by using the risk partition method of risk matrix. This paper classifies individual safety behaviors from two dimensions of safety motivation and safety behavior, and constructs a theoretical model of miners’ safety color behavior. Based on the safety color behavior model, corrective measures are proposed for the safety management of coal mine work behavior to provide effective countermeasures for coal mine safety behavior management.

Objective: To induce protective effect of co-immunization with S. japonicum triose-phosphate isomerase fused to heat shock protein 70 (SjCTPI-Hsp70) plasmid and interleukin-12 (IL-12) DNA vaccines against Schistosoma japonicum (Chinese strain) infection in water buffalo., Methods: Forty-five 8-10 months-old water buffalo from a nonendemic area were divided into three treatment groups each with fifteen buffalo: experimental group A (SjCTPI-Hsp70+IL-12, 300 microg), experimental group B (SjCTPI+IL-12, 300 microg), and control group C (pVAX+IL-12, 300 microg). All buffalo were immunized with a series of 3 intramuscular injections administered once every four weeks. Twenty-eight days postvaccination, water buffalo were percutaneously challenged with 1000 S. japonicum cercariae. Fecal examinations were conducted two days prior, one day prior, and on perfusion day, and the number of hatching miracidia and eggs per gram feces were recorded. Fifty-six days post-infection, the buffalo were sacrificed and perfused via the descending aorta. The recovered adult worms and eggs in liver tissue were counted., Results: Groups A and B showed a worm reduction rate of 51.2% and 41.5% (chi2=1.89, P>0.05)), female worm reduction of 48.9% and 44.7% (chi2=0.35,P>0.05), fecal egg reduction of 52.1% and 38.3% (chi2=3.84,P<0.05), a reduction of miracidia-hatching rate by 52.1% and 33.2% (chi2=7.30, P<0.01), and liver egg reduction of 61.5% and 42.0% (chi2=7.61 , P<0.01), respectively., Conclusion: Co-immunization with SjCTPI-Hsp70 and IL-12 DNA vaccines induces protective immunity against S. japonicum in water buffalo.

The degree of periportal fibrosis, hepatic parenchymatous fibrosis and the diameter of portal vein in fishermen from highly endemic area of schistosomiasis japonica in Dongting Lake region were measured. The results showed a significant correlation between the degree of periportal fibrosis and parenchymatous fibrosis and the portal venous diameter with a correlation coefficient of 0.375 and 0.332 respectively. The authors consider that the diameter of the portal vein can be used to assess the hepatic morbidity of patients.

Objective: To investigate the relationship of the immune status and the intensity of infection or the severity of the hepatosplenic pathology among fishermen with schistosomiasis japonica in the Dongting Lake region., Methods: Inquiring and physical examination (IPE), stool examination, B-ultrasonography of the liver and spleen, flow cytometry, turbidimetry and ELISA were undertaken to acquire or determine the intensity of infection (EPG in stool), pathological change in the liver and spleen and the level of cellular and humoral immunity. Data were analyzed with SPSS 10.0 statistics software., Results: Compared with subjects from non-endemic area, the CD4+ T cells and the CD4+/CD8+ ratio in fishing population in the endemic area significantly decreased. The decrease of the CD4+/CD8+ ratio was more significant among population with positive stool exam and with the increase of EPG and/or severity of pathological change in the liver and spleen. Contrarily, the level of the total IgM and the anti-SEA IgG in serum from fishing population in the endemic area was significantly higher than those from non-endemic area. High level serum antibodies in those stool positives were remarkable with the increase of EPG and/or the severity of hepatosplenic pathological change. The total IgA increased considerably in the subjects with significant pathological change of the liver and spleen. A high total IgG was only detected in those stool positives., Conclusion: The immune status in fishermen with schistosomiasis in the Dongting Lake showed a suppressed cellular immunity and a hyper functioning humoral immune response. The imbalance of the immunity was related to the increase of the intensity of infection and the progress of the hepatosplenic pathology.

Objective: To introduce a new method for enlarging the survival area of an expanded flap., Methods: After the skin expander was inflated with enough injection, the first delaying was performed. In the operation, two incisions were made in the skin and subcutaneous tissue superficial to the expander capsule on both sides of the long axis of the expanded flap. After 10 to 14 days, the second delaying followed, in which one pedicle was divided to form a unilateral-pedicled, super-long random flap. When the flap was transferred to the recipient Since 2000,this technique has been used in 16 patients. All the area, the door site was closed directly., Results: flaps survived completely., Conclusions: The technique of twice delaying can enlarge the survival area of the expanded skin flap.

Objective: To clarify and evaluate the morbidity of schistosome infection and the effectiveness of chemotherapy among fishermen on East Dongting Lake., Methods: Information on water-contact, history of infection and of praziquantel (PZQ) treatment among fishermen was collected. Kato-Katz method and miracidium hatching test were applied to detect the pathogens in stool specimen. Serum antibodies against soluble egg antigen (SEA) were detected with ELISA and IHA. B-ultrasonic examination was used to determine the pathological changes of liver and spleen. Chemotherapy [PZQ 40 mg/(kg x d)] was given to the fishermen followed by a re-examination after a transmission season., Results: The first investigation (six months before chemotherapy) showed that among 268 people inquired, 90.7% were frequently or intermittently contacting water, 24.0% were treated with PZQ each year, 39.4% had never been treated in the recent five years. Stool positive rate was 68.1% (111/163) and the geometric mean eggs per gram feces (EPG) were 48.77. Males had a higher infection rate (76.0%) and intensity (62.97 EPG) compared with that of females (58.7% infection rate and 30.42 EPG). The highest positive rate (83.3%) was in the age group of 11 to 20 years old. The prevalence of those who frequently or intermittently contacted water and were never treated before was 76.3% (106/139) and 79.7% (51/64), respectively. Serological positive rate was 88.0% (IHA) or 78.7% (ELISA). B-ultrasound revealed 77.4% (82/106) of the fishermen showing pathological changes in liver and/or spleen due to schistosomiasis. 37.7% of the patients showed II-III stage liver fibrosis (male: 53.0%, female: 15%), 58.5% hepatomegaly and 19.8% splenomegaly. The second investigation (six months after chemotherapy with PZQ) showed a stool positive rate of 35.4% and an average EPG 36.13 in the treatment group which were considerably lower than 56.5% infection rate and 68.47 EPG in the group without treatment. In 39 patients treated, the reversion rate from egg positive to negative was 48.7%, pathological change in liver and spleen declined by 40.6%., Conclusion: The prevalence and morbidity of schistosomiasis in fishermen on Dongting Lake were high due to frequent exposure to the affected water, and chemotherapy can effectively reduce the prevalence, the intensity of infection and morbidity of the fishermen.

To utilize Pichia pastoris to produce S-adenosyl-L-methionine (SAM), an intracellular expression vector harboring S. cerevisiae SAM2 was transformed into GS115. A recombinant strain having 2 copies of expression cassette was obtained through G418 resistance screening. This strain had higher SAM synthetase activity and higher SAM production capacity than the original strain, when cultured in medium containing methanol and methionine. The carbon source and nitrogen source of medium was optimized. The results showed SAM production by this strain was closely related to carbon metabolism. With supplementation of 0.2% glycerol every day from the beginning of 3rd day, this strain produced 1.58 g/L SAM when cultured in a medium containing 0.75% L-methionine and optimized carbon and nitrogen source after 6 days.
