Objective: To screen genes associated with poor prognosis of hepatocellular carcinoma (HCC) and to explore the clinical significance of these genes., Methods: The proper expression profile data of HCC was obtained from the Gene Expression Omnibus (GEO) database, and the differentially expressed genes (DEGs) were identified by differential expression analysis. The DAVID and String database were used for function enrichment analysis and to construct the protein-protein interaction (PPI) network respectively. The Cancer Genome Atlas (TCGA) database and the Cox Proportional Hazard Model were used for prognosis analysis of the DEGs., Results: A eligible human HCC data set (GSE84402) met the requirements. A total of 1141 differentially expressed genes were identified, including 720 up-regulated and 421 down-regulated genes. The results of function enrichment analysis and PPI network performed that CDK1、CDC6、CCNA2、CHEK1、CENPE 、PIK3R1、RACGAP1、BIRC5、KIF11 and CYP2B6 were prognosis key genes. And the prognosis analysis showed that the expressions of CDC6、PIK3R1、KIF11 and RACGAP1 were increased, and the expression of CENPE was decreased, which was closely related to prognosis of HCC., Conclusion: CDC6、CENPE、PIK3R1、KIF11 and RACGAP1 may be closely related to poor prognosis of HCC, and can be used as molecular biomarkers for future research of HCC prognosis.

Objective: To investigate the prognosis-related miRNA histological features and clinical significance of lung adenocarcinoma., Methods: Using The Cancer Genome Atlas (TCGA) data, the miRNA expression profile data of human lung adenocarcinoma were searched for differential analysis, and the prognosis-related miRNAs were screened by Cox risk regression model. The targeted miRNAs were predicted by mirwalk analysis platform, KEGG functional enrichment analysis, and finally, predict the function of prognosis-related miRNAs., Results: A total of 46 differential miRNAs in lung adenocarcinoma were screened, including 19 up-regulated and 27 down-regulated. Six prognostic-related miRNAs were screened by Cox survival analysis, namely hsa-mir-21, hsa-mir-142, hsa-mir-200a high expression, hsa-mir-101, hsa-let-7c, hsa-mir-378e low expression, hsa-mir-21 and hsa-mir-378e were associated with poor prognosis in patients with lung adenocarcinoma, and the survival time was shortened significantly ( P <0.05, AUC=0.618). KEGG analysis showed that the above prognosis-related miRNA targeting regulatory genes were related with immune response pathways, miRNA and cancer pathways, metabolic pathways and so on., Conclusions: Hsa-mir-21 and hsa-mir-378e are associated with poor prognosis of lung adenocarcinoma, and may be used as a molecular marker for prognosis of lung adenocarcinoma after further clinical verification.

The present paper discusses how to monitor and analyze the relative change in muscle oxygen content in quadriceps tissue, and measures and records the change in blood lactate acid concentration, blood volume and heart rate when eight players who are good at middle-distance races perform grade incremental intensity exercise on cycle ergometer by using noninvasive tissue oximeter with near infrared spectroscopy produced by China independently. The results show that muscle oxygen content has a close relationship (p < 0.01)with exercise load, blood lactic acid, blood volume and heart rate. When determined muscle oxygen content and blood lactate acid concentration was determined for many times to the same person, the test proved regular falling and rising. There was no significant changes when analyzed each set of the data was analyzed through horizontal comparison. It verifies we can judge the subjects's endurable exercise intensity and the upward inflection point of blood lactic acid corresponding to the decreasing inflection point of blood lactate acid concentration & muscle oxygen content according to the muscle oxygen content change of skeletal muscle while exercising. This paper shows NIRS research status and present situation in sports field through investigation, and analyzes the main trouble and research tendency in the future. By understanding NIRS technology gradually, the authors can realize that the muscle oxygen content which measured by noninvasive tissue oximeter using near infrared spectroscopy produced by China independently is a sensitive, nondestructive, up-to-date and reliable index, it has irreplaceable advantages when compared with traditional invasive, excised and fussy test methods.

Objective: To observe the changes of serum testosterone after allotransplantation of Leydig cells in rats., Methods: Leydig cells were isolated from the testes of SD rats with the Percoll technique and serum testosterone of the receptors was determined once a month for 3 successive times., Results: After allotransplantation of Leydig cells, the serum testosterone level of the receptors increased gradually, significantly higher at 3 months than that of the normal rats younger than 2 months old., Conclusion: Allotransplantation of Leydig cells has a promising application value in the treatment of male primary hypogonadism.

Objective: To study the chemical constituents of unsaponifiable matter from the seed oil of Trichosanthes hupehensis., Method: The fatty oil from the seeds of T. hupehensis was extracted with petroleum ether. The saponification was carried out with potassium hydroxide. The unsaponifiable matter was isolated and purified by silica gel column chromatography. Their structures were elucidated by means of MS, IR, UV, and 1H-NMR., Result: Karounidiol, isokarounidiol, 5-dehydrokarounidiol, 7-oxodihydrokarounidiol, stigmast-7-en-3beta-ol, stigmast-7, 22-dien-3beta-ol, 10alpha-Cucurbitadienol, beta-sitosterol, stigmast-7, 22-dien-3beta-O-beta-D-glucoside were elucidated., Conclusion: All of these compounds were found in this plant for the first time.

Aim: To study the regulation of immune function of rat Leydig cells by vasoactive intestinal peptide (VIP)., Methods: Rat Leydig cells were seperated and infected by UU, with or without the incubation with VIP. The expression of FasL on Leydig cells in different group was analysed by flow cytometry(FCM). The mRNA expression of IL-1, IL-6, TGF-beta and FasL of Leydig cells in different group was identified by RT-PCR. SD rats were infected by UU with or without the injection of VIP, and the testis tissnes of each group were har vested and observed under transmission electron microscope., Results: When testis was infected by UU, VIP up-regulated the mRNA expression of IL-1, IL-6, and TGF-beta, and down-regulated the expression of FasL in vitro. In addition, there was significant difference in testis morphous of rats from different group., Conclusion: VIP could regulate the expression pattern of IL-1, IL-6, TGF-beta and FasL by Leydig cells, which may contribute to maintain immune privilege of the testis. VIP could regulate rat Leydig cell immune function.

Aim: To study the influence of FasL overexpression in transgenic mice on the Sertoli cell immune response to testicular infection., Methods: Ureaplasma urealyticum (UU) was directly injected into bladders of FasL transgenic mice and wild-type mice respectively, to mimic an ascending infection pathway. At week 1, 2 and 3 after injection, respectively, the mice were put to death to observe the pathological alterations in testis section. And at the same time the differences of FasL, TGF-beta, IL-1alpha and IL-6 expressions on Sertoli cells were compared by immunohistochemical staining between wild mice and transgenic mice before and after infection, respectively. The high-purified Sertoli cells were isolated from the testis tissue of wild-type mice, comparing apoptotic capability of Fas(+) Jurkat cells mediated by FasL(+) Sertoli cells of wild control and wild UU-infected groups., Results: The pathological changes of testis tissue from transgenic mice were more serious as compared with wild-type mice and the model of cytokines secreted by sertoli cells was distinctive between the two kinds of mice. The UU-infected Sertoli cells increased Fas(+) Jurkat cell apoptosis., Conclusion: FasL overexpression can influence the cytokine's secretion in the process of anti-infection immunity and further affects the immune balance of testis locality. Not all FasL over expression is benefit to body's anti-infection immune response.
