Objective: To investigate the association between serum vascular endothelial growth factor (VEGF)-B and insulin resistance in pre-diabetes and newly diagnosed type 2 diabetes mellitus (T2DM) patients. Methods: Fasting serum VEGF-B levels were measured using ELISA in 160 normal glucose tolerance (NGT), 142 pre-diabetes, and 117 newly diagnosed T2DM subjects, and their association with insulin secretion, insulin sensitivity, parameters of adiposity, glucose and lipid metabolism was studied. Results:Fasting serum VEGF-B levels did not differ among NGT (130.8 pg/mL [IQR 61.3-227.5]), pre-diabetes (146.7 pg/mL [84.1-214.9]), and T2DM patients (135.3 pg/mL [58.3-214.8]) (all P>0.05). Correlated analysis showed that no significant association was found with body mass index (BMI), waist-to-hip ratio (WHR), lipid profiles, insulin secretion and sensitivity, as measured by a 75-g oral glucose tolerance test. Conclusions: Fasting serum VEGF-B levels are not related to adiposity, lipid profiles, insulin secretion and sensitivity in humans. These findings demonstrated that VEGF-B may be dispensable for insulin resistance and type 2 diabetes in humans. [ABSTRACT FROM AUTHOR]
