To address problems including underestimation of heavy precipitation, rapid decay of synoptic systems and low computational efficiency in operational forecast of CMA-GFS V3.3, some key technologies related to physics and dynamics of the model are developed and applied.A suite of graupel-related microphysical processes is adopted in the cloud microphysics scheme to improve the forecast performance of heavy precipitation. These processes include graupel colliding with cloud water, ice crystals and snow, automatic conversions of ice crystals to graupel and snow to graupel, melting process of graupel to raindrop and sublimation process of graupel. In addition, the evaporation rate of cloud and rainwater is restricted, which can increase the liquid water content in warm areas and improve precipitation efficiency.In the convection parameterization scheme, the role of the sub-cloud environmental relative humidity to convection triggers is considered, and the unreasonable occurrence of convections in dry environment is suppressed. Also, the sensitivity of the entrainment rate of the convective updraft to the relative humidity outside the cloud is enhanced to weaken the convections in dry environment. At the same time, the quasi-equilibrium closure scheme is optimized to improve the accuracy in calculating cloud-base mass flux which is related to the convection intensity.To solve the problem of the mass loss in long time integration, a mass conservation correction method is introduced to the model dynamic framework. The method is developed to ensure the mass conservation by adjusting mass in each grid box according to different weight coefficients which are determined by the change of total atmospheric mass of the current time step relative to the previous step.In terms of computational efficiency, the two-dimensional reference profile algorithm is developed. Without losing calculation accuracy, the model integration time step is extended from 240 s to 300 s using the new profile instead of the original three-dimensional reference profile. Meanwhile, the PCSI method is adopted instead of the GCR method, which reduces the time consuming of solving Helmholtz equation. In addition, the radiation scheme and predictor-corrector algorithms are also optimized to improve the computational efficiency.Through the application of the above key technologies, the forecasting skills for weather pattern and precipitation of CMA-GFS are significantly improved. And its computational efficiency is increased by about 1/3, which meets operational time requirement for the model with 0.125° horizontal resolution. Based on the improved model and the research achievements in other aspects of the forecast system, CMA-GFS is upgraded to V4.0 with a significantly improved comprehensive performance.

Renal anemia is a prevalent complication of chronic kidney disease （CKD）， which severely affects the prognosis of patients. The pathogenesis of renal anemia is complicated， and the core of which is erythropoietin deficiency and iron metabolism disorder. For years， renal anemia has been mainly treated based on erythropoiesis-stimulating and iron agents. Blood transfusion is used as a remedial treatment. Other therapeutic interventions include alleviating malnutrition， supplementing folic acid and vitamin B12， adequate dialysis and controlling hyperparathyroidism， etc.， which yield low efficacy. In recent years， clinical application of hypoxia-inducible factor prolyl hydroxylase inhibitors has brought another innovation in the treatment of renal anemia， and some potential approaches for the treatment of renal anemia have been reported. In this article， recent research progresses upon the treatment of renal anemia in CKD patients were reviewed， aiming provide new ideas for clinical treatment of this disease.

Objective To investigate the protective effect of idebenone (IDBN) on hippocampal neuron injury in epileptic rats and its mechanism. Methods Forty-eight Wistar rats were randomly divided into normal control group, IDBN prevention group (prevention group), epilepsy group, IDBN 25, 50 and 100 mg groups. Behavioral changes of rats in different treatment groups before and after treatment with IDBN were observed. The activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) in hippocampal were detected. The ultrastructural changes of hippocampal neurons and mitochondria were observed. Results The expression levels of SOD, GSH-Px and MDA in different treatment groups were significantly different (P = 0.000, for all). The activity of SOD and GSH-Px was the highest and the content of MDA was the lowest in IDBN 100 mg group. The activity of SOD and GSH-Px in IDBN 100 mg group were higher than those in epilepsy group (P = 0.000, 0.000), IDBN 25 mg group (P = 0.000, 0.000) and 50 mg group (P = 0.004, 0.005), while the content of MDA was lower than that in epilepsy group (P = 0.000), IDBN 25 mg group (P = 0.000) and 50 mg group (P = 0.002). Compared with normal control group and prevention group, hippocampal neurons in epilepsy group showed different degrees of damage, which was relieved after treatment with IDBN and gradually decreased with the increase of dose. In epilepsy group, the mitochondrial structure of hippocampal neurons was damaged obviously, with deformation and swelling, and some mitochondria were vacuolated. After treatment with IDBN, the damage was relieved, and the 100 mg group had the least damage. Conclusions IDBN can protect hippocampal neuron structure and function by inhibiting oxidative stress injury in epileptic rats.

In order to improve the wear resistance of plunger and the service life of plunger assembly for low-speed marine diesel engine, TiN and DLC coatings were prepared on plunger by ion-plating technology and multiple arc magnetron coupling coating technology respectively. SEM, optical profiler and XRD were used to characterize the micro-morphology, surface roughness and phase composition of TiN and DLC coatings, and nano indentation instrument was employed to detect the nano-hardness and elastic modulus of such coatings. In addtion, the scratch and indentation methods were used to test the binding force of the coatings. And the friction coefficients of such coatings in the air and under the environment of heavy diesel oil were investigated through the reciprocating wear test. Then, the wear degrees of those coatings were qualitatively evaluated with the help of a optical microscope. Furthermore, a bench test was adopted to evaluate the actual wear situation of the coated plunger. Results showed that the crystals of two coatings grew well, and their structures were continuous and dense, and there was no stratification, cracking or peeling phenomenon. The DLC coating was relatively smooth and the roughness R;was about 0.10 μm, while the TiN coating R;was 0.16 μm. The nano-hardness, elastic modulus and poisson ratio of DLC coating were higher than those of the TiN coating. No matter in dry friction or heavy oil friction,the friction coefficient of TiN coating was higher than that of DLC coating,and the wear resistance of TiN coating was lower than that of DLC coating. After the bench test,relatively obvious parallel grooving wear marks appeared on the plunger with TiN coating,and the overall wear was relatively serious,while the wear marks on the plunger with DLC coating were narrow and shallow that not easy to find,further proving that the wear resistance of DLC was superior.

In order to effectively resolve the conflicts between environmental protection,output and benefit during grain cleaning,the vibration screening,specific gravity air separation,cyclone separation,pulse dust removal,closed-loop air circulation and other dust removal technologies are integrated.Efficient dust-free cleaning screens are developed.To achieve effective impurity removal,the problems of excessive dust concentration in grain depots and enterprises’ grain processing and cleaning sites are aimed to be solved.Field tests on the grain depots show that the average impurity content of the grain can be controlled under 0.7% after cleaning,and the dust suppression effect is as follows:dust concentration at 5 m of the operation range lower than 30 mg/m³,15 m lower than 20 mg/m³ while 25 m lower than 10 mg/m³.Therefore,the cleaning effect and dust removal effect are better than common cleaning mechine.

Under the background of poor plantation forests, shortage of timber resources, and rich bamboo resources in China, as a kind of biomass engineering material, laminated veneer lumber (LVL) has the advantage of efficiently utilizing the existing resources. Due to its excellent mechanical properties, it can realize the standardization and serialization of the production of other characteristics, and has shown a broad application prospect. Combined with the development of China's wood-based panel industry, this paper outlined the origin and development of LVL, focused on analyzing the structural characteristics of LVL from the perspective of composite mechanics, and introduced the characteristics of veneer grading and LVL billeting. Moreover, the two types of LVL reinforcement methods, namely, phenolic resin impregnation and composite of fiber materials were summarized. Finally, the evaluation standards of LVL at home and abroad were compared and discussed, and a number of suggestions were then put forward based on the current situation of the development of China's wood construction industry and the problems existing in the application process of LVL. The experiment was to investigate effect of adding complex plant essential oils (oregano essential oil, cinnamaldehyde, eucalyptus oil) to diet of broilers on growth performance, immune function and antioxidant capacity. A total of 120 one-day-old healthy broilers with similar body weight were selected and randomly divided into four groups, with three replicates in each group and 10 broilers in each replicate. The control group broilers were fed with basic diet, while low, medium and high dose groups were supplemented with 100, 200, and 300 mg/kg of composite plant essential oil in basic diet, respectively. Broilers were raised in three stages: 1~21, 22~42 and 43~60 d, the experiment lasted for 60 d. The results showed that ADG of broilers in medium and high dose groups was significantly higher than that in control group and low dose group on 1~21, 22~42 and 1~42 d (P<0.05), and F/G was significantly lower than other two groups (P<0.05). The ADFI of broilers in low, medium and high dose groups on 43~60 d was significantly higher than that of control group (P<0.05). Titer of newcastle disease antibody and content of serum IgG, IgA and lgM in medium and high dose groups of broilers were significantly higher than those in control group and low dose group (P<0.05). T-AOC and T-SOD in serum of broilers in low, medium and high dose groups were significantly higher than those in control group (P<0.05), and content of MDA was significantly lower than that in control group (P<0.05). The experiment indicates that adding complex plant essential oils to diet can improve growth performance, enhance immune function and antioxidant capacity of broilers with an appropriate dosage of 200 mg/kg. The reclamation time was examined by combining field investigation and remote sensing image analysis in the Linze desert oasis area in the middle of the Hexi Corridor. Characteristics of soil organic carbon storage (SOCD) in cropland (0-20 cm) with different reclamation backgrounds were compared to understand the evolution trend of SOCD during reclamation. The SOCD of crop¬land (0-20 cm) varied from 2.41 to 32.97 t • hm-2, with an average of 17.22 t • hm 2. The ave¬rage SOCD of cropland in saline-alkali land, Gobi and sandy land background was 19.36, 16.10, and 15.93 t • hm-2, respectively. The SOCD under three different reclamation conditions showed an increasing trend after cultivation. The cropland in sandy land and the Gobi background slowly increased after 20 years reclamation. The cropland in saline-alkali background showed a slowdown after 25 years reclamation. The SOC sequestration rates of the cropland under sandy land, Gobi and saline-alkali land background were 0.424, 0.485 and 0.811 t • hm-2 •a-1, respectively. The SOCD were positively correlated with total nitrogen, total phosphorus, available nitrogen, and available phosphorus content, but had no significant correlation with available potassium and soil pLL In conclusion, the SOC sequestration rate in the saline-alkali land background was significantly higher than that in Gobi and sandy land background. Lowever, SOCD were at a relatively low level after 30 years reclamation. NI provides a very outstanding solution in the parameter measurement and assessment. Based on NI-PXI platform, a platform for rotation speed measurement and debugging of the motor axis was developed under laboratory conditions. According to Holzer and photoelectric two kind principles, combined with the PXI-6251 card in the pulse counting and voltage measurement mode, a rotational speed measurement and debugging system was developed based on LabVIEW. The system realizes the speed dynamic measurement, real-time data acquisition and preservation, and also can achieve the performance test and evaluation of the two type speed sensors. The paper is an exploratory study on the travel intentions of mainland Chinese residents to Japan after Diaoyu Island political crisis. Due to the superior location and excellent service, Japan has become one of the most popular outbound tourism destinations for Mainland Chinese tourists in the past 10 years. However, since the complicated and subtle relations between China and Japan, the tourism industry of Japan is evidently sensitive and vulnerable to various crisis. Under such circumstances, it is necessary to investigate the decision tree model research as well as compare domestic and overseas researches at the very beginning of the study. On the basis of literature review and in-depth interview with tourism operators via a three rounds of pre-test and revision, a self-completion questionnaire was developed and used to measure domestic residents' attitude and travel intentions after the Diaoyu Island political crisis. Data was collected through surveys in cities of different sizes from both developed and undeveloped areas in eastern and western China, such as Shanghai and cities in Fujian, Guizhou, Hubei, Jiangsu and Sichuan provinces. All respondents have abilities to travel to Japan economically and potentials to travel. Altogether 800 questionnaires were distributed and 756 were returned. Descriptive analysis was used to analyze the travel intention characteristics of domestic residents to Japan utilizing SPSS21.0. Four vital realities were found from the analysis. First, seeking entertainment opportunities can be the main motivation for Chinese residents travelling to Japan. Second, the family-based tour ought to be considered as the main trip mode. Third, the primary channels for tourism information dissemination include travel agencies and tourism e-commercial websites. Fourth, four to six-day staying in Japan could be proved as a suitable choice for domestic residents who prefer travelling during the national holidays and paid vacations. The CHAID decision tree model is reasonably used to analyze the differences of the intentions to travel among potential tourists in China. Based on the decision tree analysis, most citizens would choose to "cancel the travel plan" (29.0%), followed by "replace tourist destination" and "postpone the travel", yet only 19.6% would "travel as scheduled" in the light of the post impacts of Diaoyu Island political crisis. There could be no doubt that the vast majority of citizens in China lay emphasis on the macro political environment when making a travel decision. Four dominant nodes were generated in tree simulation, namely respective age, number of times to Japan, monthly income and marital status. This study has made a detailed analysis on the causes, and provides each group with tailored marketing measures eventually. Thereby, mixed measures should be implemented, including short-term and long-term plans, to recover the tourism market effectively. From the perspective of crisis decision-making and crisis marketing, this study segments the mainland Chinese citizens in order to disclose the heterogeneity and clustering features of them. The results of the study may set up a systematic foundation for better positioning potential tourists by furnishing an exploratory method for crisis management and crisis marketing. With the sandy farmland in the marginal oasis in middle reaches of Heihe River Basin, Northwest China as test object, this paper studied soil NO3--N accumulation and leaching under effects of different farmland use type and winter irrigation. The results showed that the mean NO3--N concentration in 0-300 cm soil profile in different farmlands ranged from 1.27 mg∙kg-1 to 83.60 mg∙kg-1. Soil NO3--N concentration was higher in 0-40 cm and 135-300 cm layers, but lower in 40-135 cm layer. Greenhouse vegetable field had a significantly higher soil NO3--N concentration than the other farmland use types. The accumulated amount of soil NO3--N decreased in the order of greenhouse vegetable field > tomato field > cotton field>seed maize field>maize-wheat rotation field>maize-wheat stripe intercropping field > alfalfa field > jujube plantation. The NO3--N accumulation in 0-300 cm soil profile in greenhouse vegetable filed reached 2171.45 kg∙hm-2, which would be a serious menace to groundwater quality, followed by tomato field and cotton field. Lesser accumulation of soil NO3--N was found in seed maize field, maize-wheat intercropping field, maize-wheat rotation field, alfalfa field, and jujube plantation, but its pollution potential would not be neglected. After winter irrigation, soil NO3--N concentration decreased in 0-80 cm layer but increased in 80-300 cm layer, indicating that winter irrigation caused NO3--N leaching into deeper soil depth. The leached amount of soil NO3--N to deeper layers increased with increasing amount of winter irrigation. To mitigate soil NO3--N leaching and groundwater contamination, a comprehensive consideration should be made on the rational arrangement of farmland use type, proper decrease of planting N-accumulated crops, and reasonable winter irrigation. In order to research the problem of gas in severe-outburst and low-permeability single coal seam, by applying the fracture mechanics and elastic theory, a technology about space mesh hydraulic fracturing by drilling through strata for outburst prevention in severe-outburst and low-permeability single coal seam was presented. A model was established to deduce the value of initiation pressure, and a new device exporting flowrates and pressure was produced. The industrial experiments were implemented with SF6 tracer investigating the fracture network. The results show that the technology can largely increase the permeability of the seam, improve the comprehensive gas drainage rate by 1.69-1.83 times, expand the coal seam 0. 581%, and reduce the electromagnetic radiation prediction index 35%-45%. Compared with the small areas, this method can eliminate the outburst by increasing permeability with hydraulic fracturing in large areas. [ABSTRACT FROM AUTHOR]

Objective: To investigate the overview of thrombosis in myeloproliferative neoplasms(MPN) patients, and to explore the risk factors of thrombosis at diagnosis and during follow-up., Methods: The clinical data of 388 MPN patients treated in our hospital were collected. The patients were followed up by outpatient and phone. The risk factors of thrombosis were analyzed by statistical methods., Results: Among 388 MPN patients, 161 patients (41.49%) showed thromboses at diagnosis or during follow-up. Among them, 92.55% were arterial thromboses, 146 cases (96.27%) were complicated with thromboses at diagnosis, and 36 cases (11.46%) showed newly thromboses or progression of previous thromboses among the 314 received full follow-up patients. Age (P＜0.001, HR:1.033, 95%CI:1.016-1.051), JAK2V617F mutation (P=0.037, HR:1.72, 95%CI: 1.033-2.862), hypertension (P＜0.001, HR:2.639, 95%CI:1.659-4.197) and hyperlipidemia (P＜0.001, HR:2.659, 95%CI:1.626-4.347) were the independent risk factors affecting thrombosis at diagnosis of the patients. During the follow-up, age (P=0.016, HR:1.032, 95%CI: 1.006-1.059) and previous thrombosis history (P=0.019, HR:2.194, 95%CI: 1.135-4.242) were the independent risk factors affecting the progression of thrombosis at different sites or on the basis of the previous thrombosis in the patients., Conclusion: Patients with advanced age, JAK2V617F mutation or complicated with hypertension and hyperlipidemia shows a higher risk of thrombosis at diagnosis, while the patients with advanced age or previous thrombosis history shows a higher risk of progression of thrombosis during the follow-up.

Objective: To analyze the factors influencing the mobilization of autologous peripheral blood stem cells (auto-PBSCs) in patients with lymphoma and multiple myeloma, and provide reference for optimizing the autologous stem cell mobilization regimen., Methods: Clinical data of 33 multiple myeloma and lymphoma patients received auto-PBSCs mobilization in our center from January 2015 to December 2018 were collected, the correlation of mobilization failure rate with gender, age, courses of chemotherapy before mobilization, does of recombinant human granulocyte colony stimulating factor (rhG-CSF), type of disease, and chemotherapy regimen were retrospectively analyzed., Results: Type of disease and course of pre-mobilization chemotherapy could affect the mobilization failure rate (P<0.05). The mobilization failure rate of lymphoma patients was 42.1%, which was significantly higher than 7.1% of multiple myeloma patients (P=0.026). The mobilization failure rate was higher in the group with chemotherapy courses≥5 before mobilization (P=0.016). Age, gender, dose of rhG-CSF, and chemotherapy regimen had no significant correlation with mobilization failure rate (P>0.05)., Conclusion: Multi-course chemotherapy before collection and lymphoma patients are poor factors negatively impacting on auto-PBSCs mobilization.

Focusing on typical mildly saline soil, S 1 (EC, 0.62 dS·m -1 ), and moderately saline soil, S 2 (EC, 1.17 dS·m -1 ), in Hetao Irrigation District of Inner Mongolia, the response of nitrification and denitrification process to salinization degree and the proportion of organic and inorganic nitrogen application were studied. The experimental treatments consisted of(1) no nitrogen(CK), (2) only inorganic nitrogen(U 1 ), and(3) organic nitrogen(U 3 O 1 , U 1 O 1 , U 1 O 3 , and O 1 ) compared to 25%, 50%, 75%, and 100% inorganic nitrogen. The results showed that increasing salinity reduced the soil nitrification potential and increased the soil denitrification capacity. The soil nitrification potential of the S 1 soil was 28.81%-69.67% higher than that of the S 2 soil, while the denitrification capacity was reduced by 17.16%-88.91%. With an increase in salinity, the AOB abundance and nitrification contribution rate were reduced, but the AOA abundance and nitrification contribution rate were increased. Furthermore, an increase in salinity increased the abundance of nirK and nirS bacteria, and increased N 2 O/(N 2 O+N 2 ) production, but reduced the abundance of nosZ . In the S 1 soil, the nitrification potential and denitrification capacity of U 1 O 1 were highest, increasing by as much as 18.59% and 15.87%, respectively, compared to the U 1 treatment. In the S 2 soil, the difference in the soil nitrification potential between the various fertilization treatments was not significant, and the denitrification capacity of the O 1 treatment was highest. The S 1 and S 2 saline soils treated with U 1 O 1 and O 1 , respectively, had higher AOB gene abundance and nitrification contribution rates, increased nirS and nosZ gene abundances, and significantly reduced N 2 O/(N 2 O+N 2 ) product ratios. Our findings suggested that mildly saline soils(120 kg·hm -2 urea+120 kg·hm -2 organic fertilizer) and moderately saline soils(240 kg·hm -2 organic fertilizer) are more conducive to soil nitrification and denitrification processes compared to soils to which inorganic nitrogen is applied alone.

Objective: To investigate the difference of clinical characteristics between young patients(age≤40 years old) and middle-older patients(age>40 years old) with the myeloproliferative neoplasms（MPN）., Methods: The clinical data (gene mutations， peripheral blood routine examinations， imaging examination and past history) of 269 MPN patients was collected and analyzed., Results: In essential thrombocythemia (ET) group, the proportion of triple-negative type in young patients was higher than that in middle-older group, while the peripheral white blood cell（WBC） and platelets（PLT） counts in the first visit were lower. In polycythemia vera (PV) group, the total detection rate of JAK2V617F (80.65%) was lower than that of other research reports. Young patients with PV showed the lower JAK2V617F rate and lower WBC count, compared with the middle-older aged patients. Both CALR and MPL mutations were not found in PV patients. There was only 1 primary myelofibrosis (PMF) patient aged <40 years old. 91.67% of the patients merged splenomegaly and this rate was higher than that of ET or PV patients. It was found that there were a diagnosed familial MPN family and an undiagnosed family， and the youngest patient was only 8 years old. The second-generation gene sequencing detection for them was not carried out., Conclusion: Age is an important reference index in the assessment of risks. The MPN patients with different age and types show much difference in gene mutations, peripheral blood cell counts, thrombotic events and sizes of spleen. The onset ages of patients with familial MPN trends to be generational younger.

A rapid analysis method based on ultraviolet-visual(UV-Vis) spectroscopy, near infrared(NIR) spectroscopy and multivariable data analysis was established for quality evaluation of Shengxuebao Mixture. The contents of eight active ingredients of Shengxuebao Mixture including albiflorin, paeoniflorin, 2, 3, 5, 4'-tetra-hydroxy-stilbene-2-O-β-D-glucopyranoside, specnuezhenide,ecliptasaponin D, emodin, calycosin-7-glucoside and astragaloside Ⅳ were simultaneously detected by using this method. HPLC-UV-MS was used as a reference method for determining the contents of these ingredients. Partial least squares(PLS) analysis was implemented as a linear method for multivariate models calibrated between UV spectrum/NIR spectrum and contents of 8 ingredients. Finally, the performance of the model was evaluated by 24 batches of test samples. The results showed that both UV-Vis and NIR models gave a good calibration ability with an R~2 value above 0.9, and the prediction ability was also satisfactory, with an R~2 value higher than 0.83 for UV-Vis model and higher than 0.79 for NIR model. The overall results demonstrate that the established method is accurate, robust and fast, therefore, it can be used for rapid quality evaluation of Shengxuebao Mixture.

Objective: To explore the role of bone marrow niche in the chemotherapy resistance of patient with acute myeloid leukemia (AML), and to investigate the effects of the MSCs on the apoptosis of HL-60 cell and its underlying mechanisms., Methods: MSCs were derived from the bone marrow of newly diagnosed AML patients (AML-MSCs) and health donors(MSCs) were co-cultured with HL-60 cells respectively. The apoptosis of HL-60 cells in the presence/absence of MSCs and/or Daunorubicin were determined by flow cytometry with Annexin V/PI double staining. In addition, the morphological features of HL-60 cells were observed by Wright-Giemsa staining, and the ratio of blasts and differentiated cells were counted. Furthermore, the expressions of apoptosis-related factors including Caspase-3, Caspase-8,Caspase-9 and Survivin were detected by Western blot., Results: The flow cytometry showed that there was no significant change in apoptosis of HL-60 cells co-cultured with MSC derived from healthy donors or AML patients. After adding Daunorubicin into different cultural systems, the apoptotic rates of HL-60, HL-60 co-cultured with normal MSCs and HL-60 co-cultured with AML-MSCs were (49.57±7.44)%, (30.72±4.05)% and (22.99±4.08)%, respectively, which showed that normal MSCs and AML-MSCs could remarkably supress Daunorubicin-induced HL-60 apoptosis, however, there was no statistically significant difference of apoptosis between HL-60 co-cultured with normal MSCs and HL-60 co-cultured with AML-MSCs. Wright-Giemsa staining showed that most of the HL-60 cells co-cultured with AML-MSCs were primitive, and cell differentiation was unusual. In AML-MSCs co-cultured group, the cell apoptosis and differentiation caused by DNR was significant decreased, and most of HL-60 cells were initial. Western blot showed that the cleavage activity of Caspase-3 of HL-60 in AML-MSCs and normal MSCs co-cultured group was decreased, compared with HL-60 in single cultured group, moreover, the decrease was significantly in AML-MSC group. Additionally, the expression of survivin in AML-MSCs and normal MSCs co-cultured group was increased, compared with that in single cultured group, and increase was significant in AML-MSCs group., Conclusion: MSCs can suppress Daunorubicin-induced HL-60 apoptosis via inhibiting Caspase-3 and maintaining survivin level.

Objective: To explore the role of bone marrow microenvironment(niche) in the development of acute myeloid leukemia (AML) and the effect of AML patients-derived MSC on the proliferation, cell cycle and immuno-phenotypes of HL-60 cells., Methods: The MSC derived from bone marrow of patients with newly diagnosed AML were isolated and co-cultured with HL-60 cells. The effect of MSC on proliferation of HL-60 cells was detected by using 3H-TdR incorporation method, the cell cycle and immunophenotypes of HL-60 cells were detected by flow cytometry., Results: The results of 3H-TdR incorporation assay showed that both AML-MSCs and normal MSCs remarkably suppressed the HL-60 cell proliferation in a time- and dose-dependent manner. The results of cell cycle analysis demonstrated that AML MSCs and normal MSCs induced arrest of the HL-60 cells in G 0 /G 1 phase. The results of immunophenotyping revealed that MSCs suppressed the expression of CD11a and CD154 on the surface of HL-60 cells. Moreover, AML MSCs exhibited increased inhibitory effects than that of normal MSCs. However, no remarkable effect of MSCs on CD54 expressions of HL-60 cells was observed in the current study., Conclusion: AML-MSCs possess effects on HL-60 cell proliferation, cell cycle and immunophenotypes similiar to normal MSCs, but exhibited increased suppressive capacity on the expression of CD11a and CD154.

Objective: To study the influence of acute myeloid leukemia (AML) microenvironment on mesenchymal stem cells (MSCs)., Methods: MSCs were isolated from the bone marrow of newly diagnosed AML patients (AML-MSCs) and were cultured. The morphology of MSC was observed by inverted microscopy, the immunophenotypes of MSC were detected by flow cytometry, the proliferation ability of MSC was detected by using MTT method, the multi-differentation ability of MSC was assayed by osteogenic, lipogenic and chrondrogenic induction. The morphologic features, immunophenotypic characteristics, cell proliferation, and multipotential differentiation capability were compared between the MSC derived from normal healthy donors and AML patients., Results: AML-MSCs presented the morphological features similar to the normal MSCs. In addition, AML-MSCs highly expressed CD29, CD44, CD73, CD105 and HLA-ABC. Meanwhile, they were homogenously negative for CD14，CD31, CD34, CD45, CD80, CD86 and HLA-DR. Further-more, AML-MSCs showed cell proliferation ability similar to normal MSCs. Notably, AML-MSCs exerted increased osteogenic-differentiation capacity as compared with normal MSCs., Conclusion: AML-MSCs possess typical MSC phenotypes but displayed enhanced osteogenic-differentiation capacity.

In this study, the synthetic pathway of β-amyrin was constructed in the pre-constructed Saccharomyces cerevisiae chassis strain Y0 by introducing β-amyrin synthase from Glycyrrhiza uralensis, resulting strain Y1-C20-6, which successfully produced β-amyrin up to 5.97 mg·L~(-1). Then, the mevalonate pyrophosphate decarboxylase gene(ERG19), mevalonate kinase gene(ERG12), 3-hydroxy-3-methylglutaryl-CoA synthase gene(ERG13), phosphomevalonate kinase gene(ERG8) and IPP isomerase gene(IDI1)were overexpressed to promoted the metabolic fluxto the direction of β-amyrin synthesis for further improving β-amyrin production, resulting the strain Y2-C2-4 which produced β-amyrin of 10.3 mg·L~(-1)under the shake flask fermentation condition. This is 100% higher than that of strain Y1-C20-6, illustrating the positive effect of the metabolic engineering strategy applied in this study. The titer of β-amyrin was further improved up to 157.4 mg·L~(-1) in the fed-batch fermentation, which was almost 26 fold of that produced by strain Y1-C20-6. This study not only laid the foundation for the biosynthesis of β-amyrin but also provided a favorable chassis strain for elucidation of cytochrome oxidases and glycosyltransferases of β-amyrin-based triterpenoids.

The reclamation time was examined by combining field investigation and remote sensing image analysis in the Linze desert oasis area in the middle of the Hexi Corridor. Characteristics of soil organic carbon storage (SOCD) in cropland (0-20 cm) with different reclamation backgrounds were compared to understand the evolution trend of SOCD during reclamation. The SOCD of cropland (0-20 cm) varied from 2.41 to 32.97 t·hm -2 , with an average of 17.22 t·hm -2 . The ave-rage SOCD of cropland in saline-alkali land, Gobi and sandy land background was 19.36, 16.10, and 15.93 t·hm -2 , respectively. The SOCD under three different reclamation conditions showed an increasing trend after cultivation. The cropland in sandy land and the Gobi background slowly increased after 20 years reclamation. The cropland in saline-alkali background showed a slowdown after 25 years reclamation. The SOC sequestration rates of the cropland under sandy land, Gobi and saline-alkali land background were 0.424, 0.485 and 0.811 t·hm -2 ·a -1 , respectively. The SOCD were positively correlated with total nitrogen, total phosphorus, available nitrogen, and available phosphorus content, but had no significant correlation with available potassium and soil pH. In conclusion, the SOC sequestration rate in the saline-alkali land background was significantly higher than that in Gobi and sandy land background. However, SOCD were at a relatively low level after 30 years reclamation. Therefore, cropland management needs to be carried out for different reclamation backgrounds to improve land use efficiency and productivity in desert oasis.

Objective: To discuss feasibility and clinical effects of minimally invasive percutaneous plate osteosynthesis in treating acetabulum anterior column fracture with intact true pelvic brim., Methods: From May 2013 to December 2015, 8 patients with acetabulum anterior column fracture with intact true pelvic brim were reviewed retrospectively. According to Judet-Letournel classification, all were simple unstable acetabulum anterior column fracture. Among them, there were 5 males and 3 females with an average age of 42.8 years old ranging from 22 to 63. The injury was caused by crush in 4 cases, smash of heavy object in 3 cases, and falling down in 1 case. The time from injury to operation was ranged from 5 to 19 days with an average of 9.5 days. Preoperative CT showed high anterior column fracture of acetabulum with intact true pelvic brim, the fracture separation was less than 1 cm. All the patients were treated with closed reduction and minimally invasive percutaneous plate osteosynthesis. The quality of fracture reduction, operation time, blood loss, fracture healing time, hip function and postoperative complications were observed and recorded., Results: All patients were followed up from 10 to 19 months with an average of 14.5 months. The quality of reduction was classified as anatomical in 4 patients, imperfect in 3, poor in 1 by Matta's score system. The operation time was 30 to 80 min, averaged 51.3 min;the blood loss was 50 to 120 ml, averaged 86.2 ml; fracture healing time was 10 to 19 weeks, averaged 13.3 weeks. At the latest follow-up, the hip function was evaluated by Merle D'Aubigne scoring system, 5 cases got excellent results, 2 cases in good, and 1 case in fair. No vascular nerve injury, wound infection, bleeding, deep vein thrombosis and other complications occurred in 8 patients., Conclusions: Minimally invasive percutaneous plate osteosynthesis in treating acetabulum anterior column fracture with intact true pelvic brim has advantages of less trauma, less bleeding, quick recovery and good curative effect, and it is a good surgical procedures for acetabulum anterior column fracture., Competing Interests: The authors of this article and the planning committee members and staff have no relevant financial relationships with commercial interests to disclose.

Objective: To investigate the relationship between NK cell count/activity and acute graft-versus-host disease (aGVHD) in patients receiving allogeneic hematopoietic stem cell transplantation (allo-HSCT)., Methods: A total of 26 patients who had undergone allo-HSCT from January to July 2015 were enrolled in this study. The NK cell count/activity in the peripheral blood of recipients on day 30 after allo-HSCT were monitored by using 4-color flow cytometry. The incidence of aGVHD in patients was evaluated by clinical manifestation combinating with related pathologic indicators, and the relationship between NK cell count/activity and aGVHD were analyzed., Results: In the aGVHD group and the no-aGVHD group, the NK cell count and activity on days 30 after allo-HSCT were 655±216 cells/µl vs 1169±372 cells/µl(P=0.002) and 7.3±3.6% vs 9.0±3.6% (P=0.008). In the II-IV grade aGVHD group and the 0-I grade aGVHD group, the NK cell count/activity were 617±220 cells/µl vs 1081±399 cells/µl (P=0.001) and 4.2±1.7% vs 8.3±3.5%(P=0.001). As compared with the 0-I grade aGVHD group, patients in the II-IV grade aGVHD group had higher relapse rate (57% vs 5%)(P=0.010) , lower 1-year progression-free survival(PFS) rate (43% vs 84%)(P=0.010)., Conclusion: NK cell count/activity on day 30 after allo-HSCT were closely relates with aGVHD, which may be a potential marker for aGVHD and can provide a new target for aGVHD therapy.

The accurate identification of traditional Chinese medicine (TCM) which collected from different producing areas is important for its quality control and clinical effects. In the present study, Fourier transform infrared spectroscopy (FTIR) combined with second derivative spectra were used to identify and analyze H. rhamnoides subsp. sinensis from different producing areas. The characteristic absorption peaks, including 2 925, 2 854, 1 743, 1 541 and 1 173 cm-1 belonging to fatty acids, flavonoids and saccharides appear in all 20 samples. But the absorption peak intensities and locations varied due to the different geographical regions. The results also showed that the absorption peaks at the range of 3 429～3 336 and 1 744 cm-1 were important characteristic absorption peaks which can identify H. rhamnoides subsp. sinensis from different producing areas. Also, absorption peaks at 1 030 and 1 516 cm-1 further confirmed the existence of flavonoids in all samples by comparing the second derivative infrared spectra in the range of 1 800～1 000 cm-1. However, the samples’ differences can be intuitively found around peaks 1 711, 1 476 cm-1 and ranges from 1 689～1 515 and 1 400～1 175 cm-1. The results demonstrated that FTIR was a simple, convenient, fast and intuitive approach to identify and analyze H. rhamnoides subsp. sinensis from different producing areas. This method provides foundations for the analysis of chemical compositions and quality control for the TCM.

This study is to establish an HPLC fingerprint and quantitative analysis of 3 components of Gyantse Seabuckthorn from different producing areas.The separation was developed on Shimadzu InertSustain C18column (4.6 mm × 250 mm,5 μm) by gradient elution with acetonitrile and 0.2% phosphoric acid water as mobile phase at a flow rate of 1.0 mL•min ⁻¹; the detection wavelength was set at 360 nm and column temperature was set at 30 ℃. The data calculation was performed with similarity evaluation system for chromatographic fingerprint of traditional Chinese medicine(Version 2004A).The fingerprints of 10 batches of Gyantse Seabuckthorn were carried out by similarity comparison, and 12 chromatographic peaks were extracted as the common peaks of fingerprint, of which three main active ingredients were successfully determined. This is the first established fingerprint and multi-component quantitative determination of Gyantse Seabuckthorn by using HPLC. Objective: To evaluate the micro-push-out bond strengths of prefabricated glass fiber posts with poly-dopamine functionalized to root dentin using resin cements, contrasted with silane treatment., Methods: In the study, 30 glass fiber posts were randomly divided into 3 groups (10 posts in each group) for different surface treatments. Group 1, treated with poly-dopa; Group 2, treated with silane coupling agent for 60s; Group 3, no surface treatment (Control group). The 30 extracted human, single-rooted teeth were endodontically treated and a 9 mm post space was prepared in each tooth with post drills provided by the manufacturer. Following post cementation, the specimens were stored in distilled water at 37 °C for 7 days. The micro-push-out bond strengths were tested using a universal testing machine (0.5 mm/min), and the failure modes were examined with a stereomicroscope. The data of the three groups were statistically analyzed using the one-way ANOVA test(α= 0.05)., Results: The bond strengths were (7.909 ± 1.987) MPa for Group 1, (5.906 ± 0.620) MPa for Group 2, and 4.678 ± 0.910 MPa for Group 3. The bond strength of poly-dopamine group was significantly higher than that of the silane group (P<0.05)., Conclusion: Contrasted with silane treatment, surface poly-dopamine functionalization was confirmed to be a more reliable method for improving the bond strength of resin luting agents to fiber posts.

This study established an HPLC fingerprint of Tibetan medicine Shaji Gao from different habitats and lay a foundation for Shaji Gao varieties identification and preparation process. The chromatographic condition was as follow: Agilent zorbax SB-C18 (4.6 mm x 250 mm, 5 μm) eluted with the mobile phases of acetonitrile and 0.4% phosphoric acid water in gradient mode. The flow rate was 1.0 mL x min(-1), and the detection wavelength was set at 360 nm. The fingerprints of 15 batches Shaji Gao were carried out by similarity comparation, 7 chromatographic peaks were extracted as the common peaks of fingerprint, 3 peaks were identified, which were quercetin, kaempferol and isorhamnetin. The similarity degrees of 14 batches of samples were above 0.9 and 1 batch of samples was below 0.9. This is the first established fingerprint of Shaji Gao by using HPLC. This method has good precision, stability and repeatability that it could provide basis for quality control and evaluation of Shaji Gao.

The 1H-NMR fingerprints of three different species tibetan medicine sea buckthorn were established by 1H-HMR metabolomics to find out different motablism which could provide a new method for the quality evaluation of sea buckthorn. The obtained free induction decay (FID) signal will be imported into MestReNova software and into divide segments. The data will be normalized and processed by principal component analysis and.partial least squares discriminant analysis to perform pattern recognition. The results showed that 25 metabolites belonging to different chemical types were detected from sea buckthorn,including flavonoids, triterpenoids, amino acids, carbohydrates, fatty acids, etc. PCA and PLS-DA analysis showed three different varietiest of sea buckthorn that can be clearly separated by the content of L-quebrachitol, malic acid and some unidentified sugars, which can be used as the differences metabolites of three species of sea buckthorn. 1H-NMR-based metabonomies method had a holistic characteristic with sample preparation and handling. The results of this study can offer an important reference for the species identification and quality control of sea buckthorn.

In irrigated agricultural ecosystems, the accumulation, distribution and transfer of nitrate nitrogen (NO(3-)-N) in soil profile and groundwater nitrate pollution were influenced by irrigation and fertilization, and were closely related to soil textural characteristics. In this study, a monitoring section with 10 groundwater observation wells along Heihe River flood land-old oasis croplands-newly cultivated sandy croplands-fixed sandy land outside oasis was established in Pingchuan desert-oasis in Linze county in the middle of Heihe river basin, and groundwater NO(3-)-N concentration was continuously monitored. Soil texture and NO(3-)-N concentration in the unsaturated zone at different landscape locations were determined. The NO(3-)-N transfer change in soil profile, nitrate leaching of soils with different texture and fertility levels in the 0-100 cm layer were analyzed. The results indicated that the vertical distribution of soil texture was sandy loam in the 0-130 cm depth, loam in the 130-190 cm and clay loam in the 190-300 cm for the old oasis croplands. For newly cultivated sandy croplands, sand content was more than 80% in each soil layer of the 0-300 cm profile, although a thin clay layer occurred in the 140-160 cm depth. The clay layer occurred 160 cm below the sand-fixing zone outside oasis. There were significant correlations between soil NO(3-)-N concentration and silt + clay content, and the order of significant degree was the natural soils of sandy lands > the newly cultivated sandy croplands > the old oasis croplands. The loss of N leaching was closely correlated to the silt + caly content in the 0-100 cm soil depth. The groundwater NO(3-)-N concentration varied from 1.01 to 5.17 mg · L(-1), with a mean value of 2.65 mg · L(-1) and from 6.6 to 29.5 mg · L(-1), with an average of 20.8 mg · L(-1) in the area of old oasis croplands and the newly cultivated croplands, respectively. The averaged groundwater NO(3-)-N concentration in the area of newly cultivated sandy croplands during the period of May and October, 2013 was 26.5 mg · L(-1), which was increased by 9.5 mg · L(-1) in comparison with the same period of 2012. There was a clear increasing trend in groundwater NO(3-)-N concentration in the sand-fixing zone outside oasis. The textural characteristics of soil unsaturated zone in the shallow groundwater distribution area was the key determining factor for controlling soil NO(3-)-N leaching and groundwater nitrate pollution. The newly cultivated sandy croplands were the nitrate vulnerable zones and high-risk areas of groundwater nitrate pollution. The implementation of cultivation pattern and irrigation and fertilization management that could effectively reduce groundwater NO(3-)-N pollution should be considered in the development of ecological agriculture.

Objective: To analyze CYP21A2 gene mutation in two families with 21-hydroxylase deficiency (21-OHD) and to explore the correlation between genotype and clinical phenotype., Methods: Two patients with 21-OHD and their families were investigated. CYP21A2 gene mutation was analyzed by PCR and direct sequencing., Results: The probands from family 1 and 2 have been respectively diagnosed with simple virilizing and non-classical 21-OHD. Both showed increased baseline serum 17hydroxyprogesterone, testosterone and adrenocorticotropic hormone (ACTH), but had no evidence of salt loss. Computer tomography revealed bilateral adrenal hyperplasia in both patients. After 1 year treatment, both had conceived successfully. DNA sequencing revealed that the proband of family 1 had compound heterozygous mutations for IVS2 13 A>G and Ile172Asn. Her father was heterozygous for Ile172Asn, whilst her mother and brother were heterozygous for IVS213A/C>G. In family 2, the proband was heterozygous for Arg341Trp and Gln318X. Her father, sister and nephew were heterozygous for Arg341Trp, whilst her mother was heterozygous for Gln318X. her brother and niece were non-affected. Carriers of single heterozygous mutations in both families had no clinical sign., Conclusion: In both families, the disease has been caused by compound heterozygous mutations, for which there has been a good genotype-phenotype agreement. Screening of CYP21A2 gene can facilitate both diagnosis and genetic counseling.

In the present paper, a method of monitoring progressive damage of composite structures by non-uniform fiber Bragg grating (FBG) reflection spectrum is proposed. Due to the finite element analysis of corrugated composite skins specimens, the failure process under tensile load and corresponding critical failure loads of corrugated composite skin was predicated. Then, the non-uniform reflection spectrum of FBG sensor could be reconstructed and the corresponding relationship between layer failure order sequence of corrugated composite skin and FBG sensor reflection spectrums was acquired. A monitoring system based on FBG non-uniform reflection spectrum, which can be used to monitor progressive damage of corrugated composite skins, was built. The corrugated composite skins were stretched under this FBG non-uniform reflection spectrum monitoring system. The results indicate that real-time spectrums acquired by FBG non-uniform reflection spectrum monitoring system show the same trend with the reconstruction reflection spectrums. The maximum error between the corresponding failure and the predictive value is 8.6%, which proves the feasibility of using FBG sensor to monitor progressive damage of corrugated composite skin. In this method, the real-time changes in the FBG non-uniform reflection spectrum within the scope of failure were acquired through the way of monitoring and predicating, and at the same time, the progressive damage extent and layer failure sequence of corru- gated composite skin was estimated, and without destroying the structure of the specimen, the method is easy and simple to operate. The measurement and transmission section of the system are completely composed of optical fiber, which provides new ideas and experimental reference for the field of dynamic monitoring of smart skin.

Objective: The aim of the study was to investigate the influences of high-fat diet and methanogens inhibitor (bromochloromethane, BCM) on the gut microbiota and fat metabolism in mice., Methods: Thirty two female C57BL/6J mice were randomly divided into 4 groups: fed respectively by Low-fat diet, Low-fat diet + BCM, High-fat diet and High-fat diet + BCM. BCM was administrated through drinking water for six weeks. The body weight of mice was recorded every week. At the end of experiment, the blood was collected for serum biochemical analysis. Bacterial communities in cecal digesta were analyzed by denaturing gradient gel electrophoresis (DGGE) and real-time PCR. VFA (volatile fatty acid) concentrations in feces were determined by gas chromatography. Expressions of fat-related genes in liver were analyzed using real-time PCR., Results: DGGE analysis shows that samples from mice fed with high-fat diet gathered together, and separated with samples from low-fat diet group. However, both the fat level in diet and BCM treatment had no effect on the numbers of total bacteria, sulfate reducing bacteria and methanogens. The acetate proportion in feces was significantly decreased (P < 0.05) when fed high-fat diet to mice (P < 0.05). BCM treatment significantly increased the propionate proportion (P < 0.05). High-fat diet also significantly increased the levels of high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and total cholesterol (TG) in blood (P < 0.05). High-fat diet up-regulated the expression of TGH gene (P < 0.05)., Conclusion: High-fat diet affected cecal microbiota and fat metabolism of mice, whereas BCM treatment had little effect.

Objective: The Chinese Children's Leukemia Group (CCLG)-acute lymphoblastic leukemia (ALL) 08 protocol for childhood ALL was established in 2008. This study aims to evaluate the drug-related toxicities of CCLG-ALL 08 protocol in the treatment of childhood ALL., Methods: A total of 114 children with newly diagnosed ALL were treated with the CCLG-ALL 08 protocol. The protocol was divided into five phases: remission induction (VDLD), early reinforcement (CAM), consolidation therapy, delayed reinforcement (DIa & DIb) and maintenance treatment. Drug-related toxicities in each phase were evaluated according to the Common Terminology Criteria for Adverse Events version 4.0., Results: Toxicities were more frequent in phase VDLD than other treatment phases, including hepatotoxicity (87.7%), dental ulcer (20.2%), hyperglycemia (20.2%), prolonged activated partial thromboplastin time (21.1%) and decreased fibrinogen (34.2%), with the incidence rates of severe adverse events at 7%, 0, 1.3%, 0.8% and 2.7% respectively. The incidence of allergic reaction to L-ASP was significantly higher in phase DIa than in phase VDLD (28.0% vs 7.9%; P<0.01), and there were no longer any allergic reactions in 15 patients who received continuing treatment with pegaspargase instead. There was no severe arrhythmia, myocardial ischemia, decreased left ventricular function, osteonecrosis, myopathy, organ failure or treatment-related mortality., Conclusions: The drug-related toxicities of CCLG-ALL 08 protocol are common in phase VDLD, but they are mild and reversible. There is no treatment-related mortality. The CCLG-ALL 08 protocol for childhood ALL is safe.

The normal Raman (NR) and surface enhanced Raman spectroscopy (SERS) for Alternaria alternate, Colletotrichum musae, and fusarium suspensions were measured by a portable Raman spectrometer (785 nm). The result indicates that the silver colloid prepared by microwave method shows high enhancement of Raman scattering for the three kinds of pathogens. Comparing SERS spectrums of the three kinds of pathogens, there are some similarities. In these SERS spectrums, the peaks in the range of 500-1 000 cm(-1) are very weak. Additionally, the peaks in the range of 1 000-1 600 cm(-1) are strong. Meanwhile, the peak at 481 cm(-1) is the strongest. Although these spectrums are similar, there are some differences such as the distribution and shape of peaks. Therefore, the three kinds of pathogens can be discriminated quickly.

Objective: To evaluate the effects of using longer xenografts in conjunctions with the location of Adamkiewicz artery (AKA) on midterm outcomes of endovascular treatment for thoracic aortic dissection., Methods: From March 2005 to September 2011, 217 patients with type B dissection were recruited. There were 143 males and 74 females with a mean age of 65 ± 11 years. Among them, 43 patients were from Fifth Affiliated Hospital of Sun Yat-Sen University while another 174 patients from Affiliated Zhongshan Hospital of Fudan University. They were divided into 2 groups according to whether AKA was identified or not pre-operatively. Endovascular repairs were performed for all patients. Distal landing levels of xenografts were recorded. The thrombosis of false lumen and the complications of spinal cord injury and endoleak were analyzed., Results: AKA was detected in 121 (55.8%) patients (group A) but not in 96 (44.2%) patients (group B). According to the levels of AKA, the patients of group A obtained the stabilization of affected thoracic aorta over a longer distance. And the ratio of patients with distal landing levels at T8-T10 was significantly higher than in group B (59.5% vs 12.5%, χ² = 49.85, P < 0.01). Also, during the follow-up period of 7.3 months, the ratio of patients with total thrombosis of false lumen in group A was significantly higher than that in group B (32.1% vs 19.1%, χ² = 4.34, P < 0.05)., Conclusion: During the endovascular repair of thoracic aortic dissection, selecting a longer device may provide a better structural stability of affected aorta and promote false lumen thrombosis.
