The Penicillium expansum produces a toxic secondary metabolite, patulin. The PatC gene encodes the MFS transport protein, and transports the patulin precursor substance to the extracellular space, ultimately forming PAT. This has high reference value in the prevention of patulin. In order to study the transport mechanism of the PatC, the spatial structure of the PatC was predicted by using bioinformatics methods, and the interaction site and possible mechanism of E-ascladiol and the PatC were analyzed by molecular docking and molecular dynamics simulation. The results showed that the gene encoded 546 amino acids, containing 14 transmembrane helix and MFS functional domains. The molecular docking results showed that the protein had 4 binding sites with the E-ascladiol, namely SER353, TYR336, PRO339, and PRO188. Molecular dynamics simulations of 200 ns were performed for the Wild protein complex system and the P188A mutant system. The results showed that the small molecule substrate and the PatC were tightly bound, and that the protein's flexibility changed strongly after forming a complex in the Pro188~Ser197aa and Gly231~Val241aa regions. It could be inferred that these two regions might have functional sites. By analyzing the parameter data of the P188A mutant system, it could be predicted that PRO188 was an important target of the PatC protein, which could provide a basis for subsequent molecular experiments. The results of the research could lay the foundation for exploring the transport mechanism of patulin, and provide new strategies for the prevention of apple rot.

Penicillium expansum, a common spoilage organism in postharvest fruits, can cause fruit decay and deterioration and endanger human health. It is of great significance to investigate the antimicrobial mechanism of the antimicrobial peptide from Paenibacillus ehimensis on P. expansum spores. The antimicrobial activity of the antimicrobial peptide against P. expansum spores was determined by using the two-fold dilution method as well as measuring the time-killing curve. Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were used to evaluate the effect of the antimicrobial peptide on the ultrastructure of P. expansum spores. The effects of the antimicrobial peptide on the cell membrane and reactive oxygen species (ROS) accumulation of P. expansum were analyzed by fluorescence probes. The results showed that the minimum inhibitory concentration (MIC) of the antimicrobial peptide against P. expansum spores was 3.5 AU/mL. The spore germination rate was significantly decreased by 28.30%, 84.57% and 100% by the antimicrobial peptide at concentrations of 0.5 MIC, 1 MIC and 2 MIC compared with the blank control (P < 0.05). After treatment with the antimicrobial peptide, the spores appeared seriously sunken, the intracellular contents were leaked out, and the morphology and structure were changed. The antimicrobial peptide damaged the cell wall of P. expansum, resulting in the leakage of alkaline phosphatase. The antimicrobial peptide depolarized the cell membrane potential in a dose-dependent manner, and increased the cell membrane permeability, leading to K+ leakage. The fluidity of the cell membrane was increased, which in turn resulted in a significant decrease in DPH fluorescence intensity (P < 0.05). The integrity of the cell membrane was damaged by the antimicrobial peptide, so the fluorescence intensity of SYTOX-Green and the contamination rate of PI were increased. Moreover, the antimicrobial peptide at 1 MIC and 2 MIC increased the fluorescence intensity of DCFH-DA significantly (P < 0.05) and resulted in ROS accumulation, which affected the physiology and metabolism of P. expansum spores. This study indicated that the target sites of the antimicrobial peptide against P. expansum spores were mainly the cell membrane and ROS metabolism.

Objective: To establish a rapid detection method for viable Penicillium expansum by propidium monoazide (PMA) combined with quantitative real-time polymerase chain reaction (qPCR). Methods: PMA-qPCR detection method for viable P. expansum was established, including the optimization of the treatment concentration, dark incubation and exposure time of PMA, the screening of specific primers of P. expansum, and the conduction of qPCR. In addition, the standard curve was constructed, which was applied to the detection of artificially contaminated apples samples. The reliability of this method was also evaluated by comparing with the plate counting method. Results: The optimal PMA treatment conditions were: 10 µg/mL for PMA concentration, 5 min for the dark incubation and 10 min for the exposure time. Among the 4 pairs of primers, Pexp-patF showed strong specificity for P. expansum, which could be used as a optimal primer for PMA-qPCR detection. The correlation coefficient of the established quantitative standard curve was 0.9948, and the detection limit of the method was 102.6 CFU/mL. There was no obvious difference between the detection result of this method and the plate counting method, and the viable P. expansum could be detected in the non-rotted part of apples. Conclusion: The established PMA-qPCR technique could be applied to the detection of P. expansum in apples, which would provide technical support for the prevention and control of P. expansum.

The solid wastes of Chinese materia dedica industrialization represented by Salvia miltiorrhiza residues have a strong small-molecule bio-recalcitrance in the process of high-value utilization of biotransformation. Highly tolerant strains were bred to break bio-recalcitrance of Salvia miltiorrhiza residues and produce high-value added cellulose, which has a significant significance for recycling and industrial utilization of solid waste. In this study, a strain of fungus, Penicillium expansum SZ13, was found with small-molecule antibacterial substance tanshinone contained in Salvia miltiorrhiza residues by a biological method. The optimal enzyme production process and peak period of SZ13 were determined. It was found that SZ13 could maintain peak enzyme production for 5 days by degrading residues under the conditions of temperature 35 ℃, rotation speed 180 r·min~(-1), 5% of residues addition, and 5% seed solution addition. Meanwhile, the ability of SZ13 to degrade the enzyme production of multiple types of residues was explored. The results showed a high enzyme activity and stable enzyme production of SZ13 in the process of degrading residues. SZ13 could efficiently utilize various types of Chinese medicine residues, such as Salvia miltiorrhiza residues, to realize the high-value utilization of cellulose in multiple types of residues.
